Aiming to optimize the residue analysis methods of thiadiazole derived pesticides in kiwi fruits, a method based on the modified QuEChERS method coupled with UPLC-MS/MS was developed. The target pesticides were transformed into thiadiazole by reacting with NaOH and adjusting the pH to 2 ~ 3 with HCl. Then thiadiazole was extracted with acetonitrile, salted out with NaCl, cleaned up with C18 and GCB, and detected with UPLC-MS/MS. The results showed that the correlation coefficient (r) was no less than 0.999 within the concentration range from 0.001 to 0.5 mg/L, which indicated a satisfactory linear relation between the areas and the concentrations of thiazole zinc(THIA), thiediazole copper(THIE), thiosen copper(THIO) and thiediazole(AMT). Meanwhile, the average recoveries of THIA, THIE, THIO and AMT were 81% ~ 93% with the maxium RSD of 9.3% at the spiked concentration of 0.01 to 1 mg/kg which indicated that the accuracy and precision of the method met the requirements of residue analysis. The limit of quantification (LOQ) was 0.01 mg/kg, showing the high sensitivity of the method. Compared with the exiting traditional methods, this new developed method has greater universality, higher sensitivity and more efficiency, and the sample preparation process is safer and environmental friendly, which is helpful to improve the residue risk monitoring levels of thiadiazole derived pesticides. Objective　To explore the effect of CYP2C19 gene polymorphism on clopidogrel plasma concentration, rate of platelet inhibition and safety. Methods　We screen the patients who took clopidogrel after PCI in our hospital, according to the inclusion and exclusion criteria. Blood samples were collected on the 6th day after clopidogrel administration. Clopidogrel blood concentration was determine by RP-HPLC. The CYP2C19 genotype was detected by non-amplified immune hybridization. The rate of platelet inhibition was evaluated by the thromboelastogram. The results were analyzed by SPSS 20.0 software. Results　A total of 87 patients were recruited, including 46 males and 41 females. Among them, 34 cases were fast metabolism. 38 cases were medium metabolism. 15 cases were slow metabolism. The result showed that there was no significant difference in drug concentration between fast and intermediate metabolism(P=0.667). There was a significant difference in drug concentration between slow metabolism and fast metabolism or medium metabolism(P<0.05). Analysis of variance and chi-square test showed that CYP2C19 gene polymorphism has a significant effect on clopidogrel platelet inhibition rate and safety (P<0.05). Conclusion　Guiding clopidogrel clinical medication based on CYP2C19 genotype alone does not necessarily achieve better therapeutic effects. CYP2C19 genotype detection and blood concentration monitoring can be combined to guide the clinical individualized administration of clopidogrel. [ABSTRACT FROM AUTHOR]

BACKGROUND: Previous studies in animals have explored the pathway of apoptosis in the endoplasmic reticulum after treatment with Shenshuai Yin for chronic renal failure. However, the mechanism of cell apoptosis through other pathways remains to be studied. OBJECTIVE: To investigate the regulatory mechanism of renal cell apoptosis of chronic renal failure rats through Fas/FasL pathway. METHODS: Sixty male Sprague-Dawley rats were randomly divided into a blank group, a model group, a Niaoduqing group and a Shenshuai Yin group. A rat model of chronic renal failure was established by intragastric administration of adenine suspension 200 mg/kg per day. Rats in the Shenshuai Yin group were given Shenshuai Yin 33.3 g/kg per day by gavage. Rats in the Niaoduqing group were given Niaoduqing granule 2.25 g/kg per day by gavage. Rats in the blank group and model group were given 0.9% sodium chloride injection 8 mL/kg by gavage. Treatments in each group were conducted once a day for 4 consecutive weeks. After treatment, serum creatinine and urea nitrogen levels were detected. Renal histopathological changes were observed by hematoxylin-eosin staining. Apoptosis in renal tissues was evaluated by TUNEL staining. The protein levels of Fas, FasL, Caspase8 and Caspase3 were detected by western blot. An approval by the Animal Ethics Committee of Liaoning University of Traditional Chinese Medicine was given with an approval No. 21000092017044. RESULTS AND CONCLUSION: Compared to the blank group, the levels of serum creatinine and urea nitrogen were significantly increased in the model, Niaoduqing and Shenshuai Yin groups (P < 0.01), the apoptotic index of renal cells was significantly increased (P < 0.01), and the protein levels of Fas, FasL, Caspase3 and Caspase8 were significantly increased (P < 0.01). The above indexes of Niaoduqing group and Shenshuai Yin group were all significantly improved as compared to the model group (P < 0.01), but there was no significant difference between the Niaoduqing group and Shenshuai Yin group (P > 0.05). To conclude, Shenshuai Yin can reduce renal cell apoptosis and improve renal function in chronic renal failure rats. The mechanism may be related to the inhibition of Fas/FasL mediated apoptosis pathway by Shenshuai Yin. [ABSTRACT FROM AUTHOR]
