2006/2007 In questo studio di tesi sono stati sviluppati ed ottimizzati nuovi supporti rigidi per la sintesi in fase solida in collaborazione con l’azienda Resindion srl. (Mitsubishi Chemical Corporation). Tali supporti, denominati Synbeads, sono stati poi caratterizzati dal punto di vista chimico e fisico ed applicati alla sintesi peptidica in fase solida. La parte iniziale dello studio si è concentrata sulla messa a punto del processo di polimerizzazione in modo da ottenere dei polimeri Synbeads caratterizzati da un grado di porosità ottimale e da una omogenea distribuzione del diametro particellare. Questi due parametri infatti influiscono notevolmente sull’applicabilità dei polimeri per sintesi su fase solida: - il grado di porosità, nei polimeri rigidi e che quindi non rigonfiano in solvente, deve garantire un buon trasferimento di massa dei reagenti e nello stesso tempo assicurare una buona resistenza dei polimeri stessi allo stress meccanico - la distribuzione particellare deve essere compresa in un ben definito range di diametro, in modo da permettere l’utilizzo dei Synbeads anche in sistemi automatizzati, senza che vi siano presente né particelle fini che potrebbero intasare i filtri, né particelle di dimensioni troppo grandi che potrebbero rendere difficoltoso il trasferimento delle stesse nei sistemi automatici. La successiva fase di ottimizzazione dei Synbeads si è concentrata su altri due parametri molto importanti, ossia la lunghezza dello spaziatore tra la matrice polimerica e il gruppo funzionale e la densità di gruppi funzionali presenti sui polimeri: - una distanza ottimale del gruppo funzionale dalla matrice del polimero ne garantisce la completa accessibilità chimica, evitando problematiche legate all’ingombro sterico - la densità di gruppi funzionali deve assicurare sia una buona capacità di carico (mmoli di gruppi funzionali per gdry di polimero) sia una completa accessibilità chimica di tutti i gruppi funzionali presenti sul polimero. In seguito a questo studio di ottimizzazione sono stati preparati i Synbeads con gruppo funzionale amminico (Synbeads A310). Partendo quindi dalle caratteristiche chimico-fisiche ottimizzate per la produzione dei Synbeads A310, sono stai messi a punto dei protocolli per la preparazione e la caratterizzazione dei altri Synbeads recanti gruppi funzionali diversi, ossia clorometilenici, bromometilenic, carbossilici ed idrossilici. Successivamente, i Synbeads A310 sono stati funzionalizzati con diversi linker che ne permettano applicazioni diverse nella sintesi in fase solida. Infatti, i Synbeads recanti i linker sono stati utilizzati per la preparazione da diversi Fmoc-AA-Wang-Synbeads e per la sintesi di un pentapeptide. Questo pentapeptide costituisce la sequenza dei primi cinque aminoacidi del Fuzeon®, un peptide che presenta attività inibitoria nei confronti dell’HIV. L’applicazione dei Synbeads è stata studiata anche per quanto riguarda il loro possibile utilizzo nella sintesi peptidica automatizzata con l’impiego di microonde, in particolare nel sistema Liberty CEM®. Grazie alle caratteristiche chimico-fisiche dei Synbeads e alla messa a punto di protocolli sintetici adatti a questi polimeri rigidi, si è potuto ottenere il prodotto desiderato con buone rese e un elevato grado di purezza. Nella parte finale di questo studio di tesi è stata indagata la distribuzione dei gruppi funzionali all’interno della matrice dei Synbeads. Combinando la tecnica ATR-FT IR con luce convenzionale e con luce di sincrotrone è possibile seguire la diffusione dei reagenti all’interno della matrice polimerica e verificare l’omogeneità della matrice stessa. Dopo aver messo a punto questa metodologia analitica, che permette un’indagine approfondita di ciò che avviene all’interno della matrice dei polimeri rigidi non trasparenti, un altro approccio analitico è stato sviluppato. Al fine di poter verificare in modo più rapido l’omogeneità della matrice polimerica, sezioni di Synbeads funzionalizzati con diverse concentrazioni di fluoresceina sono stati analizzati, permettendo in tal modo di verificare anche la distribuzione dei gruppi funzionali. Questo studio di dottorato quindi ha permesso di ottenere una nuova classe di polimeri rigidi per la sintesi su fase solida, i Synbeads, che hanno dimostrato di permettere l’ottenimento di ottimi risultati sia nella sintesi classica che in quella assistita da microonde. Nuovi approcci analitici sono stati studiati e applicati per verificare l’omogeneità della matrice polimerica e seguire i fenomeni di diffusione all’interno della matrice stessa. In tal modo, i Synbeads presentano un ottimo potenziale per un’applicazione su larga scala in processi industriali. XX Ciclo 1979

Structure, cation distribution, Mn oxidation states, and conductivity behavior of the Mn-substituted (up to 30% of Ti ions) Li4Ti5O12 have been investigated by the combined use of X-ray powder diffraction, electron paramagnetic resonance (EPR), 7Li MAS NMR, and impedance spectroscopy techniques. The spinel structure of the lithium titanate is preserved and the lattice parameter decreases with increasing the Mn content. Mn2+ ions progressively occupy the tetrahedral site up to an approximately constant value reached for 10% Mn-substituted samples. Mn3+ ions occupy both octahedral and tetrahedral sites, with a constant value on the tetrahedral one, independent of the total Mn amount; Mn4+ ions are not detected. The Mn2+ paramagnetic ions give rise to a through-space interaction with Li+ ions of both cationic sites, as evaluated by the area, proportional to the Mn2+ ions content, of a peak at ∼8 ppm observed in the 7Li NMR spectra for the substituted samples. The obtained cation distribution and the Mn valence states satisfactorily explain the decrease of conductivity observed in the Mn-doped samples with respect to the pure Li4Ti5O12

Aquest treball es va iniciar amb la intenció de millorar les propietats sensores d'alguns òxids metàl·lics tan a partir del control del creixement dels cristalls en el cas de capes fines, com en l'augment de la superfície específica en el cas dels materials granulars. En una primera s'analitzen les capes fines de SnO2 crescudes per esprai piròlisi, estudiant el creixement dels cristalls en funció d'un dels paràmetres tecnològics de dipòsit. En una segona part, es descriu la síntesi i caracterització de diversos òxids metàl·lics nanoestructurats. S'analitza també la incorporació d'additius catalítics a la matriu de l'òxid i la seva influència en la capacitat sensora. Pel que fa al creixement de capes fines de SnO2, hem pogut confirmar la influència dels paràmetres de dipòsit en el creixement dels cristalls. Els resultats obtinguts han permès relacionar la temperatura de piròlisi amb l'estructura granular de la capa obtinguda. Per a la segona part d'aquest treball, s'han sintetitzat amb èxit quatre estructures mesopòriques d'òxid de silici: SBA-15, SBA-16, FDU-5 i KIT-6. Entre elles, s'ha escollit la SBA-15 i la KIT-6, per tal d'utilitzar-les com a motlles rígids per a la síntesi de les rèpliques cristal·lines proposades.Les anàlisis de TEM mostren que, en tots els casos, les rèpliques del SBA-15 estan formades per nanopartícules orientades aleatòriament mentre que les rèpliques del KIT-6 mostren l'estructura cúbica bicontínua, especialment clara en el cas de l'òxid de tungstè, format per anells hexagonals monocristal·lins.Un cop obtingudes aquestes estructures, s'ha introduït coure i crom en diferents concentracions a les matrius d'òxid de tungstè per tal d'analitzar la seva resposta elèctrica com a sensor de NO2. Pel que fa a l'òxid d'indi, s'ha comprovat que el CaO forma carbonats en condicions atmosfèriques, donant lloc a una mostra formada per una matriu d'òxid d'indi amb petites quantitats d'òxid de calci i de carbonat de calci superficials.Pel que fa a l'ús d'additius catalítics, veiem que permeten modular la resposta elèctrica del material sensor. Fins ara, no hi havia cap sensor de WO3 d'aquestes característiques capaç de detectar concentracions tan petites de NO2 (inferiors a 100 ppb). Així doncs, aquests materials són uns bons candidats per a la detecció selectiva de petites concentracions de NO2 de manera ràpida i efectiva. Els resultats de test de l'òxid d'indi mostren que la presencia CaO a la matriu de l'òxid millora molt acusadament la sensibilitat al CO2. S'ha observat també que la resposta dels sensors disminueix al realitzar-se tests consecutius, fet que s'atribueix a un fenomen de deshidratació. No obstant, el fenomen de hidratació/deshidratació és reversible i la resposta es recupera completament després d'un breu tractament tèrmic en ambient atmosfèric. A partir des mesures de DRIFT s'ha vist que el CO2 interactua amb el CaCO3 per formar espècies de bicarbonats, les quals provoquen un canvi en la resistivitat del material. Pel que sabem, els valors de sensitivitat obtinguts són els més alts publicats fins ara per a sensors que contenen calci en la seva composició. Els resultats obtinguts posen de manifest que l'ús d'òxids semiconductors dopats amb CaO/carbonats com a sensors resistius per a la detecció de CO2 obre un gran ventall de possibilitats en el camp dels sensors de gasos.Finalment, s'ha sintetitzat amb èxit l'òxid doble de ceri i gadolini amb estructura mesopòrica. S'ha utilitzat SBA-15 com a motlle i nitrats com a precursors. El mètode de síntesi usat és doncs molts versàtil, ja que permet obtenir una àmplia gamma d'òxids nanoestructurats susceptibles de ser usats més enllà del camp dels sensors de gas. No tan sols és possible sintetitzar rèpliques acurades d'òxids simples (WO3, CeO2, In2O3) sinó també d'òxids dobles (Gd0.2Ce0.8O1.9)., Tin dioxide thin films grown by spray pyrolysis on Si substrates were studied. The evolution of the crystallographic orientation of these films with variation of the pyrolysis temperature (350-535 ºC) is reported. Some crystallographic models of SnO2 nanocrystals deposited at different pyrolysis temperatures are proposed. These models were tested by computer image simulation and compared with the experimental images. In order to increase the surface area, mesoporous oxides have been synthesized. We have used mesoporous silica SBA-15 and KIT-6 as a template for the synthesis of different semiconductor oxides: CeO2 and WO3. SBA-15 and KIT-6 silica materials are used as templates for the synthesis of two different crystalline mesoporous WO3 replicas usable as NO2 gas sensors. Single-crystal hexagonal rings set up the atomic morphology of the WO3 KIT-6 replica, whereas the SBA-15 replica is composed of randomly oriented nanoparticles. The presence of chromium in the material results in a shorter response time and improved sensor response to the lowest NO2 concentrations tested. Electrical differences related to mesostructure are reduced as a result of additive introduction. . The introduction of copper as catalytic additive allowed improving both sensor response and response time. A mesoporous CaO-loaded In2O3 material has been synthesized and used as resistive gas sensor for the detection of CO2. A nanostructured In2O3 matrix has been obtained from the SBA-15 silica template. Additive presence does not distort the lattice of In2O3, which crystallizes in the Ia3 cubic space group. Pure In2O3 based sensors are low sensitive to CO2, whereas those containing the additive show an important response in the 300-5000 ppm range of gas concentrations. As seen by DRIFTS, the electrical response arises from the interaction between CO3 2- and CO2, yielding bicarbonates products. The reaction is water-assisted, so that hydration of the sensing material ensures sensor reliability whilst its dehydration would inhibit sensor response. Gadolinium doped ceria (GDC) particles have been synthesized by hard template route for the first time. The GDC particles show high crystallinity and nanometric. The synthesized nanometric powder is expected to be used in solid oxide fuel cells.

inositol Triphosphate is a second messenger that releases calcium stored in the endoplasmatic reticulum, providing several cell responses depending on the cell where the process is done. Adenophostin A and B are two potent glyconucleotides triphosphate that were isolated from a penicillium brevicompactum culture in 1993. They are the powerful agonist described up to now for the IP3 receptor, with affinities 10-100 higher than IP3.The replacement of a phosphate group by an isoster methylencarboxilate group in biological active molecules doesn't altered the recognition of their specific receptors, being more stable to the phosphatases hydrolisis. On the other hand, the introduction of a fluorine atom in a biological active molecule involves a significant change in their activity, enhancing the stability of the nearby positions of the molecule at the same time.This thesis is the beginning of a wide investigation project where the target is synthesize different analogues of Adenophostin A. In these analogues phosphate groups have been replaced systematically by isoster methylencarboxilate groups. On the other hand, another analogue with a fluorine atom in the 2" position would be synthesized in order to determine the function of the replaced essential hydroxil group and, at the same time, enhance the neighbour glycosidic bond. First of all, we have done three different synthesis towards the glucose 3,4-dialquilated derivative. We obtain the adenosine 2'-allyl derivative in six synthetic stages. After doing a study of the glycosidic reaction with three different glycosil donors (glucose fluoride, phosphite and tricloroacethimidate derivatives), we found that the best result was with the glycosil tricloroacethimidate with tert-buthyldimethylsilil as activator. The yield of this reaction weren't brilliant, so we decide to investigate a second synthetic way, where the functionalities of the target molecule were included in the glycosil donor and acceptor before the glycosilation reaction. Firstly, the glucopyranoside with two phosphate groups in the positions 3 and 4 was synthesized, being this a new interesting way to obtain adenophostin derivatives. But it was not possible to obtain the 2'-methylencarboxilate derivative of adenosine because of an unexpected lactonization. The Selectfluor treatment of different protected glycals lead to four glycosil 2-deoxy-2-fluoro-glucopyranoside donors with estereoselectivity and yields good. The glycosilation reaction was performed with a protected adenosine derivative as glycosil acceptor, which had been synthesized before. The best result was obtained with glycosil bromide and silver triflate and silver carbonate activation system. Subsequent deprotection steps led us to a very close synthetic precursor of the target molecule. This work has opened the synthetic route to the different target molecules and, nowadays, there is another doctoral thesis which is developing this project in the laboratory., El inostilo trifosfato (IP3) es un mensajero secundario, responsable de la liberación del calcio almacenado en el retículo endoplasmático, dando lugar a diferentes respuestas celulares en función de la célula en la cual se dé el proceso. En 1993 se aislaron de un cultivo de penicillium brevicompactum dos potentes gliconucleósidos trifosfatados, la Adenofostina A y B. Éstos son los agonistas más potentes descritos hasta el momento para el receptor específico del IP3, con afinidades 10-100 veces superiores al mismo IP3.Se ha descrito que la substitución de un grupo fosfato por un grupo metilencarboxilato isóstero en moléculas con actividad biológica no modifica el reconocimiento por parte de sus receptores específicos, siendo más estables frente la hidrólisis por parte de las fosfatasas. Por otro lado, la introducción de un átomo de flúor en una molécula con actividad biológica supone un importante cambio en su actividad, reforzando al mismo tiempo la estabilidad de las posiciones vecinas, y con ello toda la molécula.La presente tesis se encuadra dentro de un proyecto de investigación más amplio, constituyendo el punto de partida del mismo. En este proyecto se pretende, en primer lugar, sintetizar diferentes análogos de la Adenofostina A en los que se ha substituido sistemáticamente los grupos fosfatos de la molécula original por grupos metilencarboxilatos isósteros, y por otro lado introducir un átomo de flúor en la posición 2" para estudiar el grupo hidroxilo esencial que substituye y al mismo tiempo reforzar el enlace glicosídico vecinal.En primer lugar se han realizado tres aproximaciones sintéticas diferentes para obtener el derivado 3,4-dialquilado de la glucosa.Se obtenió el 2'-alil derivado de la adenosina en seis etapas sintéticas, realizándose un estudio de la reacción de glicosilación con tres diferentes dadores de glicosilo (fluoruro, fosfito y tricloroacetimidato de glicosilo derivado de glucosa). El mejor resultado fue con el tricloroacetimidato de glicosilo y triflato de terc-butildimetilsililo cómo activador.Sin embargo, el rendimiento de esta reacción no era muy bueno, así que se decidió estudiar una segunda aproximación sintética, donde la funcionalización de la molécula final estuviera incluida en el dador y aceptor de glicosilo antes de la reacción de glicosilación. Por un lado se obtuvo el glucopiranósido con los dos grupos fosfato en las posiciones 3 y 4, abriendo así una nueva vía de obtención de derivados de la adenofostina que puede resultar muy interesante. Pero no fue viable la síntesis del derivado 2'-metilencarboxilato de la adenosina porque se daba una inesperada lactonización.El tratamiento con selectflúor de un glical protegido condujo a quatro diferentes dadores de glicosilo 2-desoxi-2-flúoro-glucopiranósidos con muy buena estereoselectividad y buenos rendimientos. Se ensayo la reacción de glicosilación con un derivado de la adenosina protegido como aceptor de glicosilo, sintetizado previamente, obteniéndose un muy buen resultado con bromuro de glicosilo y triflato de plata-carbonato de plata como sistema activador. La síntesis continuó con el tratamiento de grupos protectores llevando a la obtención del precursor sintético más inmediato de la molécula objetivo.El trabajo ha abierto las rutas sintéticas para obtener las diferentes moléculas objetivo y actualmente se está continuando la síntesis en el laboratorio dentro de otra tesis doctoral.

β-Keto enol ethers have been synthesized in very good yield in solvent-free conditions from differently substituted alcohols and β-diketones in the presence of Yb(OTf)3 as catalyst. The method is applicable to both cyclic and acyclic β-diketones with only slight differences in the experimental procedure.

O-Isopropylidene derivatives of sugars are readily prepared by using the Lewis acid and dehydrating agent triphenylphosphine polymer-bound/I 2 complex. This new method is characterized by smooth, non-equilibrating reaction conditions and a very clean, simple work-up, making it particularly suitable for O-isopropylidenation of sugars under mild conditions and with low environmental impact.

Viene descritta una procedura per la creazione di una funzione di trasformazione di un segnale vocale neutro in uno caratterizzato emotivamente. Questa funzione è stata sviluppata sulla base di un modello statistico, a mistura di funzioni gaussiane, dello spettro del segnale vocale. Sono utilizzati, come segnali di riferimento per l'allenamento del modello, due database di segnali vocali creati ad hoc: uno registrato da un parlatore, simulando l'emozione della collera, e uno neutro, con la stessa intonazione e durata dei fonemi, ottenuto con un sintetizzatore vocale per concatenazione di difoni, che utilizza la "voce" dello stesso parlatore. Il modello a mistura di gaussiane, addestrato sui coefficienti mel-cepstrali estratti dal segnale neutro, è utilizzato per dividere questo spazio acustico in classi fonetiche equivalenti e per calcolare, per ogni classe identificata, i parametri delle funzioni di conversione. Il metodo di trasformazione del segnale nel dominio delle frequenze ha fornito delle ottime prestazioni, come è stato dimostrato da un test percettivo in cui un segnale neutro convertito è stato riconosciuto come "arrabbiato".

We describe a new synthetic path for the preparation of isotopically labelled saturated odd-numbered fatty acids by a homologation procedure for the direct conversion of carboxylic acids into their 2,2-dideuterated homologues. This process can be used to obtain both odd- and even-numbered 2,2-dideuterated fatty acids starting from carboxylic acids. Furthermore, the reiteration of this process can also be used for the synthesis of polydeuterated fatty acids in which the heavy atoms are placed on adjacent carbon atoms or differently arranged in the molecule. [reaction: see text] An efficient route for the synthesis of orthogonally protected l-sugars has been opened up, starting from the heterocyclic homologating agent 1 and 2,3-O-isopropylidene-l-glyceraldehyde (2). Our synthetic path enables the synthesis of a 2,3-unsaturated-l-pyranoside, which can be suitably functionalized to afford the desired l-hexoses. In this paper, we report the synthesis of l-manno- and l-altro-pyranosides. Moreover, this strategy may be used to prepare all eight sugars and their derivatives in either enantiomeric form.

Alpha-melanocyte stimulating hormone (alpha-MSH) is an endogenous antiinflammatory peptide with antimicrobial properties. We recently found that a synthetic analogue, [dNal(2')-7, Phe-12]-alpha-MSH (6-13), was considerably more potent in killing Candida albicans, but the anti-cytokine potential of the molecule was not investigated. Because molecules that combine candidacidal and antiinflammatory properties could be very useful in clinical practice, we measured the anti-TNF-alpha potential of [dNal(2')-7, Phe-12]-alpha-MSH (6-13) and explored effects of amino acid deletions and substitutions on both anti-Candida and anti-TNF-alpha activities. The results show that anti-TNF-alpha properties of this candidacidal peptide are only marginally increased relative to the native sequence. Conversely, we found that a closely related candidacidal analogue, [dNal(2')-7, Pro-12]-alpha-MSH (6-13), had enhanced anti-TNF-alpha effects in vitro and in vivo. This peptide, and other melanocortins with a similar dual effect, could be very useful to eradicate infections and, concurrently, reduce inflammatory reactions.

Els alcaloides de Securinega són un grup de compostos policíclics de plantes de la família Euphorbiaceae. La majoria presenten una estructura tetracíclica amb un anell de -lactona ,-insaturada (anell D) i un sistema heterocíclic de 6-azabiciclo[3.2.1]octà (anells B i C). Es poden classificar en dos grups en funció de la grandària de l'anell A: els alcaloides tipus securinina, si l'anell A és una piperidina, i els alcaloides tipus norsecurinina, si l'anell A és una pirrolidina. Aquests alcaloides tenen una gran activitat biològica. Aquesta Tesi Doctoral centra el seu estudi en els alcaloides del tipus securinina.El primer objectiu d'aquesta Tesi fou continuar amb la síntesi cap als alcaloides tipus securinina emprant una cicloaddició 1,3-dipolar com a pas clau. Aquesta aproximació s'anomena Ruta A.La cicloaddició 1,3-dipolar de la nitrona cíclica 5 al compost carbonílic 7 donà 3 cicloadductes, essent 12 el majoritari amb un 70% de rendiment.De la reducció de 12 amb L-Selectride® s'aïllaren dos alcohols, 15, com a producte majoritari (74% de rendiment), i 16. La mesilació de 15 donà la isoxazolidina 17 amb un 89% de rendiment. De la reducció de l'enllaç N-O de 17, seguida de ciclació pemeté aïllar la indolizidina 18, on ja estan presents els anells A i B dels alcaloides.La desoxigenació de la posició 2 de 18 amb una reducció de Barton-McCombie donà 19 amb un 41% de rendiment, que es reduí amb LiAlH4 per obtenir 20 amb un 94% de rendiment. 20 es tractà amb o-NO2C6H4SeCN per donar 26 amb un 75% de rendiment. De la reacció de 26 amb H2O2 en presència de TFA s'aïllà 21 amb 71% de rendiment.Tots els intents d'oxidació de l'olefina 21 foren negatius.La formació de l'anell de butenolida fou impossible, donada la presència de l'amina terciària a l'estructura de la indolizidina. Decidírem canviar l'estratègia, utilitzant un substrat en què l'àtom de nitrogen estigués protegit com a carbamat.Aquesta segona ruta (Ruta B) tenia com a substrat de partida l'àcid 47. La reducció de 47 amb BH3·THF, seguida d'oxidació de Swern donà l'aldehid 49 amb un 77% de rendiment. De la reacció de 49 amb el reactiu de Grignard 51 s'aïllaren els alcohols 52, que es tractaren en condicions d'oxidació de Swern per donar la cetona 53, amb un 66% rendiment.Els grups funcionals necessaris per la RCM s'incorporaren per tractament de 53 amb bromur de vinilmagnesi i clorur d'acriloïl. Aquesta reacció donà els acrilats 56/57 amb una relació 6/1 i 57% de rendiment. La RCM de 56/57 s'efectuà utilitzant un 10% molar del catalitzador de Grubbs de segona generació 59, obtenint així les butenolides 60/61 amb un 78% yield i una relació diastereomèrica de 6/1.El sistema dioxolànic de 60/61 s'hidrolitzà amb DDQ i els aldehids resultants 62/63 es tractaren amb iodur de iodometilentrifenilfosfoni per obtenir les (Z)-iodoolefines 66 i 67 amb un 28% i 7% de rendiment, respectivament. La configuració absoluta de 66 s'assignà com a (2S,2'R) d'acord amb el producte final sintetitzat (vide infra).El tancament de l'anell C es realitzà en condicions de reacció de Heck, utilitzant [PdCl2(PPh3)2]. Així, la iodoolefina 66 donà el compost tricíclic 68 amb 78% de rendiment i la iodoolefina 66 donà el compost tricíclic 69, amb el mateix rendirment.La formació de l'últim anell s'efectuà mitjançant bromació al·lílica amb NBS i peròxid de benzoïl, desprotecció del carbamat amb TFA i tractament del cru resultant amb K2CO3. Quan 68 es tractà en aquestes condicions, s'obtingué la virosecurinina, (+)-1, però només amb un 20% d'excés enantiomèric., Securinega alkaloids are a group of polycyclic compounds elaborated by plants of the Euphorbiaceae family. Most of them present a tetracyclic structure formed by an ,-butenolide (ring D), a 6-azabicyclo[3.2.1]octane (rings B and C) and a piperidine or pyrrolidine (ring A). According to the size of this last heterocycle, are classified in two main groups, securinine- and norsecurinine-type alkaloids, respectively. These alkaloids have a high biological activity. The study of this Thesis is focused on securinine-type alkaloids.The first objective of this Thesis was to go on with a route to securinine-type alkaloids using a 1,3-dipolar cycloaddition as a key step. This route has been called Route A.The 1,3-dipolar cycloaddition of the cyclic nitrone 5 to the carbonylic compound 7 gave three cycloadducts, with 12 as main product in 70% yield.Reduction of 12 with L-Selectride® yielded two alcohols, 15, as main product in 74% yield, and 16. Mesylation of 15 afforded the isoxazolidine 17 in 89% yield. Reduction of N-O bond of 17, followed by intramolecular cyclisation gave indolizidine 18 in 91% yield, which already presents rings A and B of the alkaloids.The deoxigenation of position 2 of 18 was acomplished by a Barton-McCombie reduction and furnished 19 in 41% overall yield, which was reduced with LiAlH4 to afford 20 in 94% yield. This alcohol was treated with o-NO2C6H4SeCN to give the seleniumderivative 26 in 75% yield. The reaction of 26 with H2O2 in presence of TFA provided 21 in 71% overall yield. Oxidation reactions of the olefine 21 were studied, but all attempts were unsuccessful.Due to the presence of the terciary amine in the indolizidine structure, the formation of ring D was not possible. We decided then to change the strategy, using a starting material in which the nitrogen atom was already protected as a carbamate.The second route (Route B) of this Thesis had (S)-N-tert-butoxycarbonyl pipecolinic acid, 47, as a starting material. Reduction of 47 with BH3·THF, followed by Swern oxidation afforded the aldehyde 49 in 77% overall yield. The reaction of 49 with the Grignard derivative 51 furnished alcohols 52, which were submitted to Swern oxidation giving rise to the ketone 53 in 66% overall yield. The required moieties for the RCM reaction were incorporated by sequential treatment of 53 with vinylmagnesium bromide and acryloyl chloride in a one-pot procedure. This reaction afforded an inseparable mixture of the diastereomeric acrylates 56/57 in a 6/1 ratio in 57% overall yield. The RCM reaction of a mixture 6/1 of 56/57 was performed using the Grubbs second generation catalyst 59. The best results were obtained by addition of a 10% molar amount of catalyst. Thus, butenolides 60/61 were obtained in 78% yield as an inseparable mixture in a 6/1 ratio.The dioxolane system of a mixture 6/1 of 60/61 was hydrolyzed by treatement with DDQ and the resulting aldehydes 62/63 were treated with iodomethylenetriphenylphosphonium iodide to furnish the (Z)-iodoolefines 66 and 67 in 28% and 7% yield, respectively. The absolute configuration of isomer 66 was assigned as (2S,2'R) according to the synthesized natural product (vide infra). Closing of the ring C under Heck conditions was accomplished by reaction with [PdCl2(PPh3)2]. Thus, (Z)-iodoolefin 66 gave tricyclic compound 68 in 78% yield and (Z)-iodoolefin 67 afforded 69 in 78% yield.The formation of the last ring of the alkaloids was performed by allylic bromination with N-bromosuccinimide and benzoyl peroxide, removal of the carbamate group with TFA and treatement of the resulting crude material with K2CO3. When 68 was treated under these conditions, virosecurinine, (+)-1, was obtained, but only with a 20% enantiomeric excess.

The iodio-induced cyclization of 2-alkenyl-1,3-dicarbonyl compounds with the mono- and disubstituted double bond occurred with good diastereoselectivity. A study of stereochemical aspects for different substituents on the allyl side chain was carried out. When the substituents were allyl groups, the trans isomers formed preferentially; in the case of aromatic substituents, the reaction lead instead to cis isomers.

[structure: see text] Novel suitably functionalized tetracyanoanthraquinodimethane (TCAQ) derivatives covalently linked to thiophene moieties have been synthesized. The thiophene-based monomers have been chemically polymerized and copolymerized to yield new and soluble donor-acceptor double-cable polymers. The absorption and emission data reveal that the optical properties can be finely tuned by modifying the ratio of monomers in the copolymerization process.

Aquesta tesi s'emmarca dins les investigacions realitzades en el Departament d'En-ginyeria Química de la Universitat Autònoma de Barcelona en col·laboració amb la Unitat de Química i Bioquímica de Proteïnes del CID-CSIC, en el camp de la síntesi enzimàtica de pèptids.El treball es centra en desenvolupar un procés per a la síntesi enzimàtica de l'octapèptid C-terminal de la CCK (CCK-8). L'interès més rellevant d'aquesta molècula és que presenta un ampli espectre d'activitat biològica i un elevat potencial terapèutic.- La introducció de la memòria s'ha distribuït en tres parts:- Diferents aspectes cabdals en la síntesi enzimàtica de pèptids.- Característiques i funcions de la CCK i CCK-8.- Un resum dels estudis previs realitzats sobre la síntesi enzimàtica del penta-pèp-tid C-terminal de la CCK-8, els quals han servit de punt de par-tida per aquest treball.- El capítol de resultats i discussió s'ha subdividit en diferents apartats.- Inicialment es presenten les investigacions de N-a protecció/desprotecció realitzades amb penicil·lina G acilasa (PGA) d'E. coli. Aquests estudis es van començar amb la N-a protecció de diferents derivats d'aminoàcids a escala analítica amb els grups protectors fenilacetil i mandelil (PhAc, Mand). Segui-da-ment aquestes reaccions es van realitzar a escala pre-pa-ra-tiva.En segon terme es presenta l'estudi de l'enllaç peptídic R-Gly-Trp-OBzl (dipèptid 4-5 de la CCK-8, on R: PhAc, Mand) amb l'objectiu d'investigar i comparar la influència dels grups protectors PhAc i Mand sobre la reactivitat del donador d'acil. Al mateix temps s'intenta optimitzar la reacció amb una finalitat de procés.Aquest estudi es tanca amb l'aplicació de la PGA en una reacció de despro-tecció del grup N-a protector. Concretament es tria desprotegir el grup PhAc del pentapèptid PhAc-Gly-Trp-Met-Asp(OBut)-Phe-NH2, etapa necessària per tal de poder abordar la síntesi de l'octapèptid.- En un segon bloc, s'inicia l'estudi de la síntesi del tripèptid N-terminal de la CCK-8 a escala analítica, a partir de les dues estratègies de síntesi possibles (Asp+Tyr)+Met i Asp+(Tyr+Met).- Seguidament es presenta un apartat dedicat a trobar unes bones condicions per a la síntesi de l'octapèptid i tot seguit s'introdueixen les modificacions necessàries per a l'obtenció de la PhAc-CCK-8.- L'últim bloc del capítol es destina a presentar el procés global per a la síntesi enzimàtica de la PhAc-CCK-8 a escala preparativa.Tenint com a base l'esquema de síntesi del pentapèptid dels estudis previs, es proposa un nou esquema amb la idea de definir un procés global per a la síntesi d'aquesta molècula. Finalment es du a terme la síntesi de la PhAc-CCK-8 a escala preparativa., This PhD dissertation is included in the research performed in the Chemical En-gi-neering Dept. of the UAB in collaboration with the Unitat de Química i Bio-quí-mica de Proteïnes of CSIC, in the field of enzymatic peptide synthesis.The main goal of this work is to develop a process for the enzymatic syn-thesis of the C-terminal octapeptide of CCK (CCK-8).The main interest of this molecule is its wide spectrum of biological activity and a great therapeutic po-tential.- The introductory chapter has consists of three parts:- Several capital aspects in the enzymatic peptide synthesis.- Features and functions of CCK and CCK-8.- An abstract of previous studies on the enzymatic synthesis of the C-terminal pen-ta-peptide of CCK-8. This work was the starting point of the present one.- The results and discussion chapter has been subdivided into several sections.- Firstable is showed the N-a protection/deprotection studies performed with pe-ni-cillin G acylase (PGA) from E. coli. These studies started with N-a pro-tec--tion of different aminoacid derivatives at anlytical scale using phenylacetil and mandelil (PhAc, Mand) as protecting groups. After that this reactions were carried out at preparative scale.Secondly the study of peptidic bond R-Gly-Trp-OBzl (dipeptide 4-5 of CCK-8 sequence, where R: PhAc, Mand) is shown, having in mind to compare the influence of PhAc and Mand protecting groups on the acyl donor reactivity. At the same time we optimize the reaction from a process approach.This study is closed applying PGA in a deprotection reaction of the N-a group. In particular it has been chosen the deprotection of the pentapeptide PhAc-Gly-Trp-Met-Asp(OBut)-Phe-NH2, because this stage is needed to achieve the octapeptide synthesis.- A second block covers the N-terminal tripeptide synthesis of CCK-8 at analytical scale, from the only two synthetic strategies (Asp+Tyr)+Met and Asp+(Tyr+Met).- The next block is devoted to find the best reaction conditions for the octapeptide synthesis and, afterwards, the introduction of the modifications required to obtain the PhAc-CCK-8.- The last block is focused on the global process for the enzymatic synthesis of PhAc-CCK-8 at preparative scale.Taking into account previous pentapeptide synthesis studies, a novel scheme is proposed to define a global process for the syn-thesis of this molecule. The feasibility of this scheme has been tested at analytical scale. Afterwards the pentapeptide synthesis and PhAc depro-tection (using PGA as a catalyst) have been performed at gram scale.Afterwards, the N-terminal tripeptide synthesis process is studied, taking into consideration the conditions at analytical scale previously stated. Finally the synthesis of PhAc-CCK-8 at preparative scale has been achieved.

Negli ultimi anni, i polimeri biodegradabili sono stati oggetto di grande attenzione per il loro impatto sullo smaltimento dei rifiuti plastici. I poliesteri alifatici sono fra i composti più promettenti per la realizzazione di materiali biodegradabili. La velocità d'idrolisi enzimatica dei polimeri è in genere influenzata da diversi parametri come la massa molare, la mobilità segmentale, la struttura chimica, la stereochimica, l'area superficiale, la morfologia e la cristallinità. Sono stati sintetizzati dei poli(butilensuccinato-co-butilensebacato), P(BSu-co-BSe), ad alta massa molare e a diverse composizioni, mediante reazione di transesterificazione tra l'1,4-butandiolo ed i dimetilesteri dell'acido succinico e dell'acido sebacico allo stato fuso ed in presenza di Ti(BuO)4, con lo scopo di studiare le relazioni proprietà-struttura che influenzano la biodegradabilità dei copoliesteri sintetizzati. La struttura e le proprietà fisiche dei poliesteri sono state investigate mediante 1H-NMR, SEC, DSC, Diffrazione ai Raggi X (WAXD) e Viscosimetria. I film dei campioni sono stati sottoposti all'attacco enzimatico mediante lipasi da Rhizopus Arrhizus o mediante lipasi da Mucor Miehei e sono state confrontate le relative perdite in peso normalizzate. I risultati indicano che i valori di massa molare non hanno un'influenza significativa sul grado di biodegradazione per i film dei poliesteri studiati. L'indipendenza del grado di biodegradazione dalle variazioni della massa molare potrebbe indicare che le lipasi usate sono enzimi di tipo endo, attaccano cioé in maniera random i legami esterei all'interno della catena. Le attività enzimatiche delle lipasi usate risentono solo in parte delle variazioni dell'indice di cristallinità e l'influenza della cristallinità appare più evidente per i film di P(BSu-co-BSe), trattati con lipasi da Rhizopus Arrhizus. La struttura chimica delle unità monomeriche nella catena polimerica sembra invece influenzare in modo predominante l'attività delle lipasi da Rhizopus Arrhizus e da Mucor Miehei.
