Monosodium glutamate (MSG), a sodium salt derived from glutamic acid, is widely used in commercial food products to improve taste, quality, and preservation. However, its consumption may have detrimental effects on male reproductive function. Nevertheless, plant extracts, such as Lepidium meyenii (Maca), Trigonella foenum-graecum (Fenugreek), Spirulina platensis (Spirulina), and Tribulus arabica (Tribulus), may ameliorate these adverse effects. To this effect, the phytochemical properties of Lepidium meyenii, Trigonella foenum-graecum, Spirulina platensis, and Tribulus arabica were assessed, and their potential impact on MSG-induced impairment of reproductive parameters was examined. The phytochemical composition (steroids, terpenes, phenols, flavonoids) of the plants was profiled through spectrophotometry and the antioxidant activity was assessed using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Thirty-six male Wistar rats were divided into six groups at random: a control group receiving distilled water, and five experimental groups (MSG, Maca, Fenugreek, Spirulina, and Tribulus) receiving 900 mg/kg/day of MSG dissolved in water for 45 days. Subsequently, the animals in the experimental groups were administered 500 mg/kg/day of the respective plant extract via oral gavage for an additional 35 days, while the MSG group continued to receive water only. Following the treatment period, the animals were sacrificed, and their reproductive tract organs were collected, weighed, and subjected to further analysis. Phytochemical analysis revealed the presence of diverse bioactive elements in the plant extracts, including phenolic and flavonoid compounds. Exposure to MSG negatively impacted total and progressive sperm motility, which was ameliorated by Lepidium meyenii treatment. Sperm morphology showed no significant differences among groups. Treatment of the phytochemical agents diminished histomorphometric alternations of the testicular length, germinal epithelium height, and number of cells in seminiferous tubules, which were caused by the initial administration of MSG. Testosterone and LH levels were reduced in the MSG group but improved in extract-treated groups. The study suggests Lepidium meyenii as a potential remedy for reproductive dysfunction. However, further investigation into its mechanisms and human safety and efficacy is warranted.

Oxide copper minerals are commonly extracted via acidic leaching, using acids such as H2SO4, HCl, or HNO3. These strong acids are the most widely used because of their high dissolution kinetics. However, their main concern is the high acid consumption because copper oxide deposits contain large amounts of acid-consuming gangue. This paper proposes using an alternative aqueous alkaline monosodium glutamate (MSG) system to leach copper oxide minerals. Tenorite (CuO) was used as the copper oxide mineral under study. The influence of process variables (such as temperature and glutamate concentration) and kinetics of this system on copper leaching from tenorite were studied. The results showed that temperature has a significant effect on copper dissolution rates. Increased temperature from 15 °C to 60 °C enhanced the copper extraction from 9.1% to 97.7% after 2 h. Leaching kinetics were analyzed using the shrinking core model (SCM) under various conditions, indicating that the leaching rate presented a mixed control. This method, however, fails to describe leaching for broad particle sizes due to its requirement for single-sized solid grains. This study demonstrated that a large particle size distribution in tenorite supported a successful extension of the SCM for leaching it from mixed glutamate solutions. The activation energy for the 15–60 °C temperature range was calculated to be 102.6 kJ/mol for the chemical control.

In most current farm operations, lactating sows need to overcome reproductive and environmental stresses that have resulted in poor sow production performance and piglet growth. Therefore, this study aimed to investigate the effects of in-feed supplementation of monosodium glutamate (MSG) in sows during late gestation lactation in regard to litter performance. The study subjects were 12 multi-parity sows (Landrace × Large White), farrowing sows with an average parity of four (three with three parities, seven with four parities, and two with five parities). They were randomly divided into the following two diet groups: the basal diet as a control (CON) group based on corn and soybean meal; and the basal diet + 2% MSG group. The experimental time ranged from 109 days before delivery to 21 days after delivery. There were six sows in each group, and each sow served as the experimental unit. There were no significant differences (p > 0.05) in body weight (BW), back fat (BF) thickness and estrus interval between sows supplemented with 2% MSG in their diets before and after farrowing and during weaning (p > 0.05). However, MSG-treated sows tended to increase BW loss at farrowing more than the CON group (p = 0.093) but lost less weight during lactation than the CON group (p = 0.019). There were no significant differences in the body condition scores (BCSs) and BF loss of the two groups of sows before and after farrowing and at weaning (p > 0.05). There was no significant difference in the weight of newborn piglets between the two groups of sows (p > 0.05). The weaning weight (p = 0.020) and average daily gain (ADG) (p = 0.045) of suckling piglets were higher in the MSG treated group compared to the CON group. The daily milk production of sows in the MSG treatment group was higher compared to the CON group (p = 0.045). The protein concentration of milk at week 3 (p = 0.060) and fat concentration of milk at week 5 (p = 0.095) of the MSG-supplemented sows tended to increase more than the CON group. In summary, the dietary inclusion of MSG supplementation had a beneficial effect on the late gestating sows and their piglet’s growth and milk production. Our research has shown that the addition of 2% MSG in late gestation and lactation diet would be beneficial for both sow and piglet production.

The prostate-specific membrane antigen (PSMA) has been targeted for PET imaging and radioligand therapy (RLT) in patients with prostate cancer. Xerostomia is a common side effect of RLT because of the high salivary gland uptake of PSMA radioligands. Here, we aimed to determine the impact of monosodium glutamate (MSG) administration on PSMA-radioligand biodistribution within healthy organs and tumor lesions by using 68Ga-PSMA-11 PET imaging. Methods: Sixteen men with prostate cancer were randomized (1:1) into oral ingestion and oral topical application ("swishing") arms. Each subject underwent 2 68Ga-PSMA-11 PET/CT scans within 14 d under baseline and MSG conditions. The salivary glands and whole-body tumor lesions were segmented using qPSMA software. We quantified tracer uptake via SUVmean and SUVmax and compared parameters within each patient. Results: For the oral ingestion arm, salivary gland SUVmean and SUVmax decreased on average from the control scan to the MSG scan by 45% ± 15% (P = 0.004) and 53% ± 11% (P < 0.001), respectively. Tumor lesion SUVmean and SUVmax also decreased by 38% (interquartile range, -67% to -33%) and -52% (interquartile range, -70% to -49%), respectively (P = 0.018). Swishing had no significant effect on 68Ga-PSMA-11 accumulation in normal organs or tumor lesions. Conclusion: Oral ingestion but not topical application of MSG reduced 68Ga-PSMA-11 uptake in salivary glands. Tumor uptake also declined; therefore, the clinical application of MSG is unlikely to be useful in the framework of RLT.

Modern enzyme biotechnology is a promising and rapidly developing field that requires the latest research on the conditions of enzyme biosynthesis. Optimizing the composition of the nutrient medium depending on the needs of microorganisms and physicochemical factors directly affect the increase in the efficiency of the biosynthesis of amylolytic enzymes, namely the biosynthetic capacity of the strain Streptomyces recifensis var. lyticus 2P-15. Modulation of the biosynthetic activity of strains producing amylolytic enzymes will allow to significantly increase their economic yield. Aim. The purpose of this work is to optimize the biosynthetic capacity of the strain Streptomyces recifensis var. lyticus 2P-15 in terms of the synthesis of amylolytic enzymes and the study of the dynamics of the influence of physical and chemical factors on optimization. Methods. The object of the study is the strain Streptomyces recifensis var. lyticus 2P-15, obtained by three-stage selection of the producer. The simplex method of selecting the composition of the environment was used for the research. The ratio of amylolytic activity to the level of biomass accumulation was taken as the biosynthetic capacity of the strain. A photocolometric method was used to determine amylolytic activity. The level of biomass accumulation was determined by the weight method. Results. It was established that as a result of optimizing the composition of the simplex nutrient medium by the method of mathematical modeling, the biosynthetic capacity increased by 3.63 compared to the control variant. It was also investigated that the optimal concentration of such a component of the nutrient medium as monosodium glutamate С5Н8NO4Na・H2O is 1.5%, which increases the amylolytic activity by 2.63 and increases the accumulation of biomass. Separately, it should be noted the obtained results of the study of the optimal concentrations of heavy metal ions added to the optimized version of the nutrient medium, which allow further research in this aspect to be continued and the use of Co, Mo, Cd ions in the composition of the nutrient medium. With the obtained results, there is an increase in amylolytic activity in the best response by 3.54. The obtained results have theoretical and practical significance for further research in the biotechnology of enzymes. Conclusions. The prospect of further research into the optimization of the biosynthesis of actinomycetes by the simplex method of other aspects of its regulation will be to increase the biosynthetic capacity of the studied strain, which will have a positive effect on the economic output of the production of amylolytic enzyme preparations by obtaining microbial synthesis.

The purpose of this study was to determine the potential of administration robusta coffee extract as an antioxidant on the renal histopathology in mice exposed to monosodium glutamate (MSG) orally. This study used 20 adult male rats with Balb C strain divided into 5 experimental groups, namely K + (MSG 0.12 mg / head and Vitamin C 6 mg/mice), K- (MSG 0.12 mg/mice and Sodium carboxymethyl cellulose (CMC Na) 0,1 ml/mice), P1 (MSG 0.12 mg/mice and Robusta coffee extract 0.1 mg/mice), P2 (MSG 0.12 mg/mice and Robusta coffee extract 0.2 mg/mice), and P3 (MSG 0.12 mg/mice and 0.4 mg Robusta coffee extract mg/mice). Excessive neuronal excitation by glutamate is a well-established cause of neurotoxicity, leading to severe impairment of brain function. Excitotoxicity is a key factor in numerous neurodegenerative conditions. In this study, we investigated the neuroprotective effects of Danshensu (DSS) against monosodium glutamate (MSG)-induced neurotoxicity in adult mice and their offspring. We randomly divided one hundred 8-week-old Kunming mice (equal number of males and females) into a control group and an experimental group. The experimental group was further subdivided into various treatment groups, including MSG gavage treatment, bwbw DSS treatment group 1 (bwbw DSS treatment group 2, a drug control group, and a normal control group (receiving an equal volume of physiological saline for ten consecutive days). Additionally, another one hundred healthy 8-week-old Kunming mice were similarly divided into groups and treated. These mice were paired randomly (one male and one female) and pregnant females were housed separately to obtain offspring. Subsequently, we conducted histological and behavioral analyses on adult mice and their offspring. MSG treatment induced significant cellular edema and hippocampal damage in both the treated mice and their offspring. However, varying doses of DSS effectively counteracted the neurotoxic effects of MSG, with no adverse impact on brain tissue structure or neural function in either adult mice or their offspring. Behavioral experiments further confirmed that DSS exerted a substantial protective effect against MSG-induced impairment of learning and memory in the treated adult mice and their offspring, in addition to mitigating central nervous system overexcitation and inhibiting exploratory behavior. In conclusion, DSS exerts significant protective effects against MSG-induced neurotoxicity in both adult mice and their offspring.

Monosodium glutamate (MSG) is one of the most extensively used flavour enhancers worldwide. Although it is widely regarded as a safe food additive with no recommended daily dosage, its over-consumption has been associated with notably pathophysiological events in various tissues and organs of the body. Previous studies have reported of the neuro- cardio- and hepato- toxic effects of its excessive exposure. Moreover, the food additive instigates metabolic dysfunction. It has been established that MSG damages male reproductive accessory organs like prostate glands and epididymis. In addition, it impairs serum enzymatic activities and serum levels of testosterone, gonadotropin-releasing hormone, luteinizing hormone and cholesterol. Reduced sperm count, sperm motility, sperm morphology, and sperm viability, imbalances in male reproductive hormones, alongside alteration in the histoarchitecture of the testes and other male reproductive tissues have also been connected with excessive exposure to MSG. Literature reports affirm the link between the over-consumption of MSG and reproductive organ weight and male sexual behaviour. This review article addresses the multi-systemic effects of exposure to MSG and the possible mechanism of action of the compound with a focus on the negative implications of the food additive on male reproductive functions and the possible role of natural antioxidants in male reproductive functions. carefully selected keywords were used during the literature search to gather credible and up-to-date information about the subject matter.

Vernonia amygdalina (VA) is popularly consumed as food and as medicine due to its nutritional and bioactive constituents. This study assessed the anti-genotoxic effect of aqueous leaf extract of VA against monosodium (MSG) –induced genotoxicity. Crude extraction and phytochemical analysis were done using standard methods. In silico studies was done using compounds in the extract against Bcl-2, NF-kB 50, DNA polymerase lambda, DNA ligase, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Twelve rats were divided into three groups with four rats in each group. Group I was fed on food and water, group II received MSG (4 g/kg) per body weight (pbw) intraperitoneally, group III received MSG (4 g/kg) pbw intraperitoneally followed by oral dose of VA leaf extract (250 mg/kg) per body weight. The number of the micronucleated red blood cells and white blood cells were determined from blood smears microscopically. Results showed that aqueous extract of VA contained in mg/100 g alkaloids (7.04 ± 0.16), saponins (3.91 ± 0.13), flavonoid (1.64 ± 0.16), phenol (3.40 ± 0.12) and tannins (0.07 ± 0.32). In silico studies revealed high binding interaction (ΔG > −8.6) of vernoniosides D and E with all the tested proteins. There was a reduction in the number of micronucleated cells, neutrophils and eosinophils of the treated group compared to the MSG group, while there was an increase in the lymphocyte count. The anti-genotoxic effects of VA leaf extract might be attributed to the synergistic interaction of the various bioactive components in the extract. VA could be a potential plant for the prevention of cancer and other diseases that attenuate the immune system.

Current study examined the protective impact of Rhodiola rosea (R. rosea) extract on hepatic toxicity markers caused by MSG. MSG induced significant increases in hepatic stress biomarkers. There were significant declines in GSH, catalase and SOD levels, which were accompanied by substantial elevations in IL-1β, IL-6, and TNF-α. There was hepatic cellular damage, and upregulation of caspase-3 and a decrease in Bcl-2 expression. The hepatotoxic impacts of MSG were normalized by Rhodiola supplementation. R. rosea downregulated caspase-3 and upregulated HO-1 and Nrf2 mRNA expression. R. rosea regulated the immunoreactivity of COX-2, TGF-β1, and NFkB genes associated with hepatic toxicity. R. rosea extract normalized histopathological changes induced by MSG and restored hepatic architecture. R. rosea extract exhibited anti-apoptotic, antioxidant, and anti-inflammatory properties. In conclusion, R. rosea extract is a promising medication against hepatic toxicity induced by MSG, which works by regulating the different signaling pathways of inflammation-, oxidative-stress-, and apoptosis-associated markers.

[Display omitted] The combination of chemo- and biocatalysts to perform one-pot synthetic route has presented great challenges for decades. Herein, glutamate oxidase (GLOX) and trimanganese tetraoxide (Mn 3 O 4) nanocrystals were combined for the first time by one-step biomineralization to construct a mimic multi-enzyme system (GLOX@Mn 3 O 4) for chemoenzymatic synthesis of α‑ketoglutaric acid (α‑KG). Mn 3 O 4 not only served as a support for the enzyme immobilization, but also contributed its catalytic activity to co-operate with natural enzymes for the cascade reactions. The as-synthesized chemo-enzyme catalysts with directly contacted catalytic sites of the enzyme and inorganic catalyst maximizes the substrate channeling effffects for in situ rapid decomposition of the oxidative intermediate, H 2 O 2 , during the enzymatic oxidation of sodium glutamate, thus relieving the inhibition of H 2 O 2 accumulation for GLOX. Benefiting from the excellent stability and reusability of GLOX@Mn 3 O 4 , a nearly 100% conversion (99.7%) of l -glutamate to α-KG was achieved, over 4.7 times higher than that of the free GLOX system (21.2%). Monosodium glutamate (MSG) is one of the most frequently used food additives that endanger public health. The antioxidant, hyperlipidemic, and cytoprotective properties of Lepidium sativum seeds (LSS) as a natural remedy can minimize the harmful effects of MSG. This study investigated the potential protective effect of LSS against MSG-induced hepatotoxicity in rats. Male albino Sprague Dawley rats (n = 24) were equally divided into four groups for 30 days: the control group (G1) received a basal diet without supplement, group (G2) was fed a basal diet + MSG (30 g/kg b.w.) as a model group, group (G3) was fed a basal diet + MSG (30 g/kg b.w.) + LSS (30 g/kg b.w.), and group (G4) was fed a basal diet + MSG (30 g/kg b.w.) + LSS (60 g/kg b.w.). LSS enhanced serum alkaline phosphatase activity as well as total cholesterol, triglyceride, and glucose levels. It can decrease peroxide content in serum lipids and inhibit glutathione reductase and superoxide dismutase in hepatic cells. The dietary supplementation with LSS provided cytoprotection by enhancing the histoarchitecture of the liver and decreasing the number of apoptotic cells. Due to their antioxidant and anti-apoptotic properties, LSS effectively protect against the hepatotoxicity of MSG. These findings are of the highest significance for drawing attention to incorporating LSS in our food industry and as a health treatment in traditional medicine to combat MSG-induced hepatic abnormalities. This systematic review analyzes monosodium glutamate (MSG) in the Alzheimer’s disease-like condition to enhance translational research. Our review seeks to understand how MSG affects the brain and causes degenerative disorders. Due to significant preclinical data linking glutamate toxicity to Alzheimer’s disease and the lack of a comprehensive review or meta-analysis, we initiated a study on MSG’s potential link. We searched PubMed, ScienceDirect, ProQuest, DOAJ, and Scopus for animal research and English language papers without time constraints. This study used the PRISMA-P framework and PICO technique to collect population, intervention or exposure, comparison, and result data. It was registered in PROSPERO as CRD42022371502. MSG affected mice’s exploratory behaviors and short-term working memory. The brain, hippocampus, and cerebellar tissue demonstrated neuronal injury-related histological and histomorphometric changes. A total of 70% of MSG-treated mice had poor nesting behavior. The treated mice also had more hyperphosphorylated tau protein in their cortical and hippocampus neurons. Glutamate and glutamine levels in the brain increased with MSG, and dose-dependent mixed horizontal locomotor, grooming, and anxiety responses reduced. MSG treatment significantly decreased phospho-CREB protein levels, supporting the idea that neurons were harmed, despite the increased CREB mRNA expression. High MSG doses drastically lower brain tissue and serum serotonin levels. In conclusion, MSG showed AD-like pathology, neuronal atrophy, and short-term memory impairment. The present study sought to analyze the possible changes that might be caused by MSG and soybean on the secretion of both the male and female reproductive hormones in wistar rats. Two hundred and ten wistar rats (105 female and 105 male rats) were used for this study. The 105 female rats were equally divided into three groups representing the various experimental durations of 2, 4, and 6 months. Each of these groups consisting of 35 rats were further divided equally into 7 subgroups each containing 5 rats. 3 out of the 7 subgroups were orally administered MSG, another 3 out of the 7 subgroups were orally administered soybean according to the established LD50 as follows: 1000 mg/kg b.w (low dose, LD), 2000 mg/kg b.w (medium dose, MD), 3000 mg/kg b.w (High dose, HD) daily, while the 7th group served as the control group and was given only water and normal rat chow. The 105 male rats were also grouped in same manner and orally administered the MSG and soybean. The levels of LH, FSH, progesterone (PRG), oestrogen (E2) and testosterone were analyzed by ELISA technique. Long period administration of MSG significantly (p<0.05) decreased the levels of LH, PRG, E2 and FSH in the female rats when compared with the control group. Consumption of soybean for a long period of time significantly (p<0.05) decreased the level of E2 and FSH in the female rats, while short period administration significantly (p<0.05) increased the PRG level and long period administration significantly (p<0.05) decreased the PRG level in comparison with the control. Long and short period consumption of MSG as well as soybean significantly (p<0.05) decreased the levels of testosterone and LH in the male rats, while only long period administration of both significantly (p<0.05) decreased the level of FSH in the male rats when compared with the control group. The consumption of both small and large amounts of MSG and soybean elicit reproductive hormonal imbalance irrespective of the gender through the alteration of the levels of FSH, LH, E2, testesterone and PRG. Two strains of γ-aminobutyric acid (GABA) producing bacteria, L. brevis Y1 and L. plantarum LM2, were microencapsulated in sodium alginate with two concentrations (1% and 2%) of monosodium glutamate (MSG) by using vibrating technology. The mix of both species was microencapsulated both in fresh and freeze-dried form. After 0, 1, 2, and 4 weeks of storage at 4 °C in quarter strength Ringer's solution, the microcapsules were subjected to cell viable counting and sub-cultured in MRS at 37° for 24 h. The MRS cultures were analyzed for the GABA content. The amount of GABA produced per CFU of MRS inoculum was then calculated. Only the 4-week-old microcapsules were used to inoculate a chocolate milk drink with the aim of obtaining a functionalized drink containing viable probiotic cells and GABA after a 1-week incubation at 4 °C. Therefore, the GABA production in chocolate milk per CFU of the probiotic culture after the incubation time was calculated. Results of the GABA analysis by liquid chromatography mass spectrometry of the MRS sub-cultures showed no significant difference (p > 0.05) in GABA yield between 1% and 2% MSG for the microcapsules containing fresh cells. On the contrary, a significant difference (p < 0.05) in productivity along the storage was registered. Microcapsules containing freeze-dried cells showed significant differences (p < 0.05) in GABA yield between 1% and 2% MSG only after 2 and 4 weeks of storage. A significant difference (p < 0.05) in GABA yield between the storage time was found only for the trials with 2% MSG for freeze-dried cells. The synthesis of GABA in chocolate milk significantly decreased (p < 0.05) only for fresh cells when comparing 2% with 1% MSG. In conclusion, a 1-month storage of microcapsules containing both culture forms, fresh and freeze-dried, did not affect GABA production. [ABSTRACT FROM AUTHOR]

The effect of age and gender on taste function is rarely investigated. Therefore, we tried to study the effect of age and gender on taste function as evaluated by the Waterless Empirical Taste Test (WETT®). The WETT® consists of 40 strips that are coated with one of five tastants (sucrose, citric acid, sodium chloride, caffeine, or monosodium glutamate). Each tastant is prepared with four different concentrations. These 40 strips are interspersed with an additional 13 tasteless strips. To implement the WETT®, a strip was placed on the middle portion of the tongue. The subjects closed their mouth and tasted the strip. They then chose one of six answers (sweet, sour, salty, bitter, brothy, or no taste at all). If the answer was correct, one score was acquired. One-hundred-and-twenty healthy men and women were collected in this study. Among them, there were 40 subjects in each age group of 20–39 years, 40–59 years, and ≥60 years. The overall taste and individual tastant function decreased with age, particularly between subjects aged 20–39 years and those aged ≥60 years. The overall taste and individual tastant function were better in females than in males, especially for subjects aged older than 59 years. In the present study, the colour and C:N ratio of quinoa flours pigmented with M. purpureus supplemented with monosodium glutamate and sodium chloride were evaluated during 14 days of fermentation. The best values of L*, a* and b* corresponded to the eighth day. This research showed that the pigmented flour produced by solid state fermentation of quinoa by M. purpureus showing variations in the colour red along with the C:N ratio during the fermentation time, resulting in a product with good visual sensory attributes that can be used to develop new naturally pigmented products with potential functional characteristics. Monosodium glutamate (MSG) is the sodium salt of glutamic acid (GLA), used as a flavour enhancer. MSG is considered a controversial substance. It is incriminated in disturbing the antioxidant system, but also has beneficial effects, as GLA metabolism plays a crucial role in homeostasis. This study highlights which positive or negative aspects of MSG sub-chronic consumption are better reflected in subjects potentially affected by advanced age. Daily doses of MSG were administered to four groups of two-year-old Wistar rats for 90 days: (I) 185 mg/kg bw, (II) 1500 mg/kg bw, (III) 3000 mg/kg bw and (IV) 6000 mg/kg bw, compared to a MSG non-consumer group. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, direct and total bilirubin, total cholesterol, triglycerides, creatinine and urea levels were analysed; stomach, liver and kidney samples were subjected to histopathological analysis. Although, in most cases, there were no statistical differences, interesting aspects of the dose–effect relationship were observed. After MSG sub-chronic consumption, the positive aspects of GLA seem to be reflected better than the negative ones. The hormesis effect, with low-level reactive oxygen species' protective effects and GLA metabolism, may represent the hypothesis of a potential defence mechanism triggered by MSG sub-chronic consumption in ageing rats. A series of lanthanide coordination polymers, i.e., [LnIII4(oba)6(H2O)9]·3H2O, where LnIII corresponded to EuIII (I), GdIII (II), TbIII (III), H2oba—4,4'-oxybis(benzoic acid), were synthesized using the rapid and facile microwave technique. They were structurally characterized and evaluated for use as photoluminescent probes in monosodium glutamate (MSG) detection. Possible interferences of amino acids were examined. Based on the devised ratiometric parameters, I showed viable responses to MSG. Linear calibration plot could be constructed over 10–100 mM with sensitivity, limits of detection and quantification of 33.2% mM, 1.3, and 4 mM, respectively. The constructed calibration plot could be used to determine concentrations of standard MSG solutions providing the percentage recoveries of 94–103%. Background: Monosodium glutamate (MSG) is a popular food flavor enhancer, and a glutamate subset that induces different toxicities such as hepatotoxicity, neurotoxicity, reproductive toxicity, and nephrotoxicity. This study was conducted to assess the effects of MSG on the α7 and α4 nicotinic acetylcholine receptor (nACHR) protein subunits expression of adult rat testis and the safety role of vitamin C. Materials and Methods: For this experimental research, 24 rats were haphazardly grouped into four equal groups (n=6) and orally gavaged for 30 days as follows: control group (distilled water gavage), MSG group (3 g/kg/b.w/day), vitamin C group (150 mg/kg/b.w/day), and MSG+vitamin C group (3 g/kg/b.w/day+150 mg/kg/b.w/day, respectively) that rats of all groups on the 30th day were anesthetized, and the left testes were used for of α4 and α7 nACHR protein subunit evaluation by immunohistochemistry (IHC). Statistical computations were performed using Graph Pad Prism software. Results: The present study revealed a significant reduction in the expression and optical density (OD) of α7 nACHR and α4 nACHR in the seminiferous tubules and intertubular connective tissue in the MSG group compared to the control group. In the MSG+vitamin C group, the expression and OD of α7 nACHR and α4 nACHR increased in the seminiferous tubules and intertubular connective tissue but this improvement was not significant compared to the MSG group. Conclusion: MSG decreased the expression level of nACHR protein subunits, α7 and α4, in the seminiferous tubules and interstitial testicular tissue. Vitamin C in the MSG+vitamin C group could not significantly improve the expression of α7 and α4 nACHR subunits in testicular tissue. Secondary metabolites synthesized by the producer Streptomyces are widely used in the food, pharmaceutical, cosmetic, textile, and agricultural industries. These industries around the world are developing rapidly, as a result of which they need new engineering solutions that should increase the yield of the final product and optimize the production process. Understanding the dependence of the optimal correlation of the components of the nutrient medium on the increase in the biosynthesis of secondary metabolites by the producer Streptomyces will promote the development of these industries in economic and ecological aspects. In this study, we optimized the quantitative correlation of twelve (6+6) components of the nutrient fermentation medium for Streptomyces recifensis var. lyticus 2P-15. Foroptimization we used the simplex method of mathematical modeling of the optimization conditions of the biotechnological process. After optimization, a 6.36 times increase in the level of biosynthetic capacity was obtained compared to the control version of the medium. To determine the dynamics of biosynthesis, samples of culture fluid were taken. Sampling was carried out every 24 hours of cultivation, the dynamics of pH diapason, biomass accumulation in mg/ml and amylolytic activity in U/mL were determined. The correlation of amylolytic activity to the level of biomass accumulation was taken as the biosynthetic capacity of the strain. A photocolometric method based on the starch-iodine method was used to determine amylolytic activity. The volume of biomass accumulation was determined by a weighted method. As a result of the optimization, the composition of the nutrient medium was developed, in which the degree of synthesis of amylolytic enzymes increased by 6.11 times, and there was a significant increase in biomass accumulation, while the cost of the optimized medium was reduced by 1.5–2.0 times from the initial one. Positive dynamics were studied when new components were introduced into the environment, such as sodium glutamate and corn extract. Optimum concentrations of monosodium glutamate were established at 0.5% of the volume of the nutrient medium and corn extract at a concentration of 1%, respectively. The significance of the positive effect upon the introduction of heavy metal ions and some vitamins into the medium was also checked, the obtained results provide an opportunity for further research into these aspects of the composition of the nutrient medium. The advantage of biotechnological developments in matters of industrial enzymology is the opportunity to obtain not only raw materials for the pharmaceutical or other industries (where it will only acquire a final form), but also produce as a final product in a ready, convenient form. Introduction1: In the poultry industry, feed efficiency is of great importance in order to reduce the cost of feed by maximizing production efficiency. To achieve this result, the use of synthetic amino acids, such as the amino acids methionine, lysine, arginine, etc. in poultry nutrition can play an effective role in animal performance. Application of monosodium glutamate in animals cause to increased levels of triglycerides, total protein, cholesterol and blood glucose in rats. Addition of 1% monosodium glutamate in broiler diets increased feed intake compared to the control group and also in relation to weight gain at the level of 0.25 and 0.5% monosodium glutamate had a significant decrease compared to the control group. Therefore, considering the role and importance of laying hen performance during the production process and due to the very important role of monosodium glutamate in the occurrence of these changes, the effects of consumption of different levels of monosodium glutamate on performance, egg quality characteristics and blood parameters were investigated. Materials and Methods: In this study, 60 laying hens of "Hy-Line W36" strain at the age of 68 to 74 weeks in 4 treatments and 5 replications and 3 hens per replication were used. The experiment was conducted as a completely randomized design in four periods 14-day for 8 weeks. Experimental treatments included 0, 0.4, 0.8 and 1.2% levels of monosodium glutamate per kg of feed. To better benefit from the data, all measurement factors except body weight gain were sampled and evaluated at the end of every two weeks. Egg mass was also obtained by multiplying the percentage of daily egg production by the average weight of eggs produced on the same day. Egg quality traits including height and diameter of albumin and yolk, relative weight of yolk and shell and albumin, shell thickness, shape and specific gravity were measured. Blood parameters were selected from two cages in each cage every two weeks and blood samples were taken from their wing veins and the concentrations of glucose, cholesterol, triglyceride, HDL and VLDL were measured using a Pars azmon kit and a spectrophotometer. Experimental data were statistically analyzed using statistical software (2002) SAS 9.1. Significant differences between treatments were compared with Duncan's multiple range test at a significance level (P<0.05). Results and Discussion: The effect of adding treatments on the traits related to egg mass production and feed conversion ratio at the age of 72-74 weeks and feed consumption in all weeks of the experiment was significant. Today, several neurotransmitters have been identified to regulate feed intake, one of which is glutamate, which is most abundant in the central nervous system, which reduces feed intake in broilers. Feed intake is reduced by adding monosodium glutamate throughout the production period. Moreover of experimental treatments on Haugh unit, albumin and yolk height in the whole period was significant so that adding 0.4% of monosodium glutamate to the diet was able to increase the height of albumin and Haugh units and decrease yolk height compared to other treatments. Adding 0.8% monosodium glutamate increased the white diameter in the whole period compared to the control treatment and the treatment containing 0.4% monosodium glutamate. The data showed that adding 0.8% of monosodium glutamate to the diet could increase cholesterol, triglyceride and VLDL concentrations at 70-72 weeks of age compared to other treatments. Also, adding 1.2% of monosodium glutamate to the diet has been able to increase the concentration of HDL and VLDL at the age of 76-74 weeks. Conclusion: In general, it can be concluded that the addition of monosodium glutamate to the diet has no significant effect on the percentage of production, the relative weight of yolk, albumin and egg weight despite the significant effect on blood parameters that was observed also it did not have qualitative parameters of the shell. On the other hand, the addition of 0.4% monosodium glutamate to the diet reduced feed consumption in the entire production period. Also, this treatment was able to increase the height of the albumin and Haugh unit and decrease the height of the yolk compared to other treatments in the entire production period. Monosodium glutamate (MSG) is globally used to impart flavor in cooking and to improve palatability. This work assessed the protective effects of quercetin (QU) on alterations associated with exposure to MSG. Thirty female rats were equally sorted into five groups: G (I) received water and was considered the healthy control, G (II) received corn oil orally, G (III) received QU orally for 30 days, G (IV) received MSG as an oral solution (15 mg/kg body weight) for 30 days, while G (V) was given MSG and QU simultaneously. Alterations in the levels of (serum estrogen, lipid peroxidation, and glutathione), and the activity of (glutathione peroxidase, and superoxide dismutase), ovarian histological lesions, and ultrastructural changes induced by MSG administration were remarkably ameliorated on treatment with QU (14 mg/kg b. wt.). Monosodium glutamate (MSG) production has been attracting environmental concerns due to its high energy consumption and high pollutant discharge, especially to water and the air. The water footprint of MSG has been investigated in previous studies; however, the carbon emissions have not been evaluated. This study conducted a carbon footprint analysis on monosodium glutamate production, to identify the critical material and stage that contribute the most to the carbon footprint during MSG production. The results indicated that the carbon footprint of producing 1 t of MSG is 3.14 × 10³ kgCO2eq, and 32.92 % of which is from the extraction and refinement process. Indirect emissions from raw materials used in MSG production, such as caustic soda liquid and concentrated sulfuric acid, and on-site emissions from the prodsteam production are major carbon footprint contributors. In addition, the glutamate fermentation stage accounts for 23.35 % of the total carbon footprint of MSG production due to the use of steam and liquid ammonia. In terms of the hotspot of carbon footprint in MSG production, it is identified that the steam used is the largest contributor, accounting for 34.90 % of the total carbon footprint in MSG production, followed by coal (16.47 %). Based on the research results, several cleaner production suggestions, such as greener raw materials utilization, green supply chain construction, and industrial symbiosis network establishment are proposed to further reduce the overall carbon footprint of MSG production. Background: Excitotoxicity is a condition in which neurons are damaged/injured by the over-activation of glutamate receptors. Excitotoxins play a crucial part in the progression of several neurological diseases. Marsilea quadrifolia Linn (M. quadrifolia) is a very popular aquatic medicinal plant that has been utilised for a variety of therapeutic benefits since ancient times. Its chemical composition is diverse and includes phenolic compounds, tannins, saponins, flavonoids, steroids, terpenoids, alkaloids, carbohydrates and several others that possess antioxidant properties. Objective: The objective of the present study was to investigate the neuroprotective potential of M. quadrifolia against monosodium glutamate (MSG)-induced excitotoxicity in rats. Methods: A high-performance thin-layer chromatography (HPTLC) analysis of chloroform extract of M. quadrifolia (CEMQ) was conducted to identify the major constituents. Further, the in silico docking analysis was carried out on selected ligands. To confirm CEMQ's neuroprotective effects, the locomotor activity, nonspatial memory, and learning were assessed. Results and discussion: The present study confirmed that CMEQ contains quercetin and its derivatives in large. The in-silico findings indicated that quercetin has a better binding affinity (-7.9 kcal/mol) towards the protein target 5EWJ. Animals treated with MSG had 1) a greater reduction in the locomotor score and impairment in memory and learning 2) a greater increase in the blood levels of calcium and sodium and 3) neuronal disorganization, along with cerebral edema and neuronal degeneration in the brain tissues as compared to normal control animals. The changes were however, significantly improved in animals which received standard drug memantine (20 mg/kg) and CEMQ (200 and 400 mg/kg) as compared to the negative control. It is plausible that the changes seen with CEMQ may be attributed to the N-methyl-D-aspartate (NMDA) antagonistic properties. Conclusion: Overall, this study indicated that M. quadrifolia ameliorated MSGinduced neurotoxicity. Future investigations are required to explore the neuroprotective mechanism of M. quadrifolia and its active constituents, which will provide exciting insights in the therapeutic management of neurological disorders. Food contaminants are closely monitored to establish food safety profiles. Previously, we used the lobster cockroach to delineate the oxidative and antioxidant activities that characterize exposure to the organometallic cation (methylmercury; MeHg) sometimes found in fish and the common cooking seasoning (NaCl and MSG). Here, we further examined the mechanism of inflammatory response to MeHg, NaCl, and MSG, in an attempt to establish the crosstalk between redox and inflammatory signaling during heavy metal exposure in food. The insects were exposed to diets contaminated with MeHg, NaCl, and MSG for 21 days. Subsequently, mRNA from head homogenate was used to quantify the expression of a member of the Nox/Duox NADPH oxidases that produce reactive oxygen species (DUOX), as well as target genes of the JNK, TOLL, and UPD3/JAK/STAT pathways. MeHg exposure deregulated redox and inflammation-related genes, including upregulated DUOX in the MeHg group, upregulated Reaper, UPD3, and SOCS36E in the MeHg + NaCl group, and downregulated Reaper in the MeHg + MSG group. Both MeHg + NaCl and MeHg + MSG exposures upregulated PVF, EGR, and REL and downregulated FOXO levels. Our results suggest a role for the activation of the Nox/Duox NADPH oxidases, alongside JNK and Rel signaling during MeHg and MSG exposure, therefore offering insights for handling heavy metal poisoning and showcasing Nauphoeta cinerea as a viable model for understanding redox and inflammatory signaling. Background: Monosodium glutamate (MSG) has been identified as a trigger of abdominal pain in irritable bowel syndrome (IBS), but the mechanism is unknown. This study examined whether MSG causes visceral hypersensitivity using a water‐avoidance stress (WAS) mouse model of visceral pain. Methods: Mice were divided into four groups receiving treatment for 6 days: WAS + MSG gavage, WAS + saline gavage, sham‐WAS + MSG gavage, and sham‐WAS + saline gavage. The acute effects of intraluminal administration of 10 μM MSG on jejunal extrinsic afferent nerve sensitivity to distension (0–60 mmHg) were examined using ex vivo extracellular recordings. MSG was also applied directly to jejunal afferents from untreated mice. Glutamate concentration was measured in serum, and in the serosal compartment of Ussing chambers following apical administration. Key Results: Acute intraluminal MSG application increased distension responses of jejunal afferent nerves from mice exposed to WAS + MSG. This effect was mediated by wide dynamic range and high‐threshold units at both physiologic and noxious pressures (10–60 mmHg, p < 0.05). No effect of MSG was observed in the other groups, or when applied directly to the jejunal afferent nerves. Serum glutamate was increased in mice exposed to WAS + MSG compared to sham‐WAS + saline, and serosal glutamate increased using WAS tissue (p = 0.0433). Conclusions and Inferences: These findings demonstrate that repeated exposure to MSG in mice leads to sensitization of jejunal afferent nerves to acute ex vivo exposure to MSG. This study focuses on the effect of amino acid surfactant (sodium lauroyl glutamate [SLG]) on foam properties of a mixture of fluorocarbon (FS‐50) and hydrocarbon (APG0810) surfactants. The molecular interactions, foamability, and foam stability of the surfactant solutions are systematically studied. The results show that SLG has stronger ability to lower surface tension of water compared to APG0810, but worse that than FS‐50, and to adsorb onto gas/liquid interface in mixed solutions with FS‐50 compared to APG0810. The addition of SLG increased foamability of APG0810/FS‐50 mixed solution, and decelerated foam drainage and coarsening of individual APG0810, individual FS‐50, and mixture of APG0810/FS‐50 solutions. The mixture of SLG/FS‐50 exhibited the higher foamability and foam stability than the frequently used critical component of the new developed AFFF, the mixture of APG0810/FS‐50. Monosodium glutamate (MSG) and aspartame are the most food addaitives allover-the world. Prolonged xposure to the two additives results in several adverse effects on the differnet body organs. Recently, natural palnts and their fruits are considered as a new medical approach for alleviating of many diseases. In the curunt work we attempt to evaluate the possible ameliorative role of Opuntia ficus-indica Fruit against adverse effects of MSG and aspartame on the liver of mother's rats and their offspring. 36 pregnant rats were used in these studies, they were classified into 6 groups(six for each); control, MSG, aspartame, MSG+ Opuntia ficus-indica Fruit, Aspartame+ Opuntia ficus-indica Fruit and MSG+Aspartame + Opuntia ficus-indica Fruit. At the end of weaning (21 day postnatal), the mothers rats and and their pups were sacrificed, the blood was collected and the whole liver was removed to investigate the histopathological and biochemical changes induced in the liver. The obtained results revealed severe histopathological signs in the liver of MSG and aspartame supplemented mother's rats and their pups, these signs included dilated hepatic sinosoids, dialted central veins, pyknotic hepatocytes and presence of multiple Kupffer cells. Also, remarkable apoptosis appeared in the hepatocytes that indicated by strong expression of P53 and weak expression for Bcl-2. An obvious increase in the liver enzymes (ALT&AST) and caspase-3 but significant decrease in the levels of antioxidants (SOD&CAT) were observed in MSG and aspartame supplemented mother's rats and their pups. Suppelementaion of Opuntia ficusindica Fruit juice was successfully ameliorated the deleterious histopathological and biochemical alterations induced by MSG and aspartame. Abstract Monosodium glutamate (MSG) is used in food-additives, and the Food and Drug Administration has placed it under intense scrutiny following several reports that it causes glutamate neurotoxicity. Ashwagandha (ASH) roots are traditionally used for memory enhancement. This study aimed to evaluate the nootropic activity of ASH as well as its therapeutic anti-amnesic activity against MSG-induced hippocampal-dependent spatial memory impairment and hippocampal-NMDAR modulation. A total of 36 rats were divided equally into six groups (n = 6 in each group); the rats in the normal and negative groups were administered daily doses of normal saline and MSG (300mg/kg), respectively, for 21 days. Two nootropic groups were administered ASH at 300 and 500 mg/kg o.p., respectively, for 21 days. Two other treatment groups were administered daily doses of MSG 300mg/kg o.p. as well as 300mg/kg and 500mg/kg o.p. of ASH for 21 days. The rats’ spatial memory was assessed for five days using the MWM. Additionally, NMDAR were measured quantitatively by immunohistochemistry. We found that the rats in the nootropic groups showed significantly enhanced nootropic activity characterized by improved hippocampal-dependent spatial memory, as well as increases in the level of NMDAR in the Cornu Ammonis 1 region of their hippocampus. Moreover, we elucidated the therapeutic potential of ASH to protect against the depression of spatial memory caused by MSG-induced neurotoxicity. Further, we elucidated a strong correlation between NMDAR-positive cells in the hippocampus and enhancement of spatial learning induced by long-term administration of ASH as well as a strong correlation between NMDAR positive cells in the hippocampus and depression of spatial learning induced by long-term administration of ASH and MSG. Introduction: Monosodium Glutamate (MSG) as a flavour enhancer is still being widely used in a range of diet formulations. Several studies have recently revealed undesirable side effects of its long-term intake. The immune system is a potential target of toxicity following chemicals exposure. Thymus is a primary lymph organ which produces immunocompetent T cells. It enhances the maturation of lymphoid precursors into T cells to establish the T cell pool during life in human. Aim of the Work: To evaluate the toxic effect of MSG on the thymus of adult male albino rat and the possible protecting role of vitamin C when administered concomitantly. Materials and Methods: Forty adult male albino rats were used for this work and were divided randomly into four equal groups; Group 1 used as control, Group II received 100 mg/kg vitamin C, Group 3: received 3 g/kg MSG and Group 4 received both MSG and vitamin C concomitantly. Both drugs were given once daily orally. After 60 days the thymus glands were obtained and processed for histological, immunohistochemical using caspase 3, morphometric and statistical studies. Results: MSG administration resulted in marked decrease in thymic cellularity with significant increase in caspase 3 positive cells in association with ultrastructural changes in the thymocytes and epithelial reticular cells. Concomitant administration of vitamin C with MSG reduced the previous histological alteration with decrease in caspase 3 positive cells. Conclusion: Monosodium glutamate affects the histological structure of the thymus gland and its concomitant administration with vitamin C diminished its thymic toxicity. [ABSTRACT FROM AUTHOR]

The low fatization of high-fat foods is a significant trend that will impact the future developments of food products. Consumers have regarded health attributes as a critical indicator for purchasing food. In this study, enzyme-modified soy protein isolate, sea fish collagen, and ovalbumin were used to prepare the composite fat substitute for the protein matrix. This matrix was applied to the traditional surimi-based product Nemipterus virgatus fish sausage to replace the exogenous fat, and a new type of low-fat fish sausage was developed. This change is expected to reduce the exogenous fat in the traditional fish sausage without reducing the flavor and sensory quality of the original product. The results showed that taking the sensory evaluation and gel strength value of the product as indicators, the optimal ratio of compound fat substitute (enzyme-modified soy protein isolate:sea fish collagen:ovalbumin) was 2:1:3 when using the orthogonal test method for the first time. In the next step, with compound fat substitutes, exogenous fats and transglutaminase as the main factors, single factor and response surface method were used to explore the best formula of new low-fat Nemipterus virgatus fish sausage. The results showed that the best gel strength and sensory evaluation scores were obtained when the compound fat substitute, TGase, and exogenous fat were 0.59 g, 0.245 g, and 8.03 g, respectively. The optimal formulation of the low-fat Nemipterus virgatus fish sausage was obtained as follows: surimi, 67.52%; complex fat substitute, 0.66%; TGase, 0.28%; fat, 9.04%; starch, 6.75%; sugar, 3.94%; salt, 2.25%; monosodium glutamate, 0.23%; I&G, 0.34%; and water, 9%. Compared with the traditional fish sausage, the content of exogenous fat in the new, low-fat Nemipterus virgatus fish sausage was reduced by 54.8%. Meanwhile, the sensory score of fish sausage was increased by 21.79%, maintaining its good flavor and sensory quality. Twelve proline (Pro)/glutamic acid (Glu) dipeptides were synthesized to explore the molecular interaction mechanism between Pro/Glu dipeptides and umami receptors. Based on sensory evaluation, the homology modeling and molecular docking technique were used to assess the structure-activity relationship between Pro/Glu dipeptides and umami receptors, including taste receptor type 1 member 1 (T1R1), taste receptor type 3 member 3 (T1R3) and calcium-sensitive receptor (CaSR). Our results indicated that except for proline-serine (Pro-Ser), valine-proline (Val-Pro), and leucine-glutamic acid (Leu-Glu), the umami threshold of other dipeptides was lower than that of sodium glutamate (0.3 mg/mL). The umami threshold of γ-glutamic acid-methionine (γ-Glu-Met) and glycine-glutamic acid (Gly-Glu) was the lowest (0.07 mg/mL). Notably, Asp147, Thr149, Ser172, and Arg277 were the key binding sites of Pro-Glu dipeptides to T1R1, the key umami receptor of Glu dipeptides. Similarly, Glu45, Ser147, Val277, and His278 were the key binding sites to T1R3, whereas Ser147 was the key binding site between N-γ-Glu dipeptides and the T1R3 receptor. Leu173, Asn176, Gln179, Arg220, Ser244, and Asp275 were the key binding sites to CaSR, with Glu dipeptides had a higher affinity for binding to the CaSR receptor compared to Pro dipeptides. Binding of Pro/Glu dipeptides to receptors was mainly achieved through hydrogen bonds and hydrophobic interaction. During molecular docking, dipeptides with strong umami interactions were predominantly embedded in the depth of the receptor binding pocket, whereas dipeptides with weak umami interactions were located in the shallow position of the binding pocket, with some of their hydrophobic or hydrophilic regions being exposed to the surface of the receptors. This study aided in clarifying the interaction mechanism between Pro-Glu dipeptides and umami receptors, establishing a foundation for further study on the mechanism of action of umami peptides. In this study, the flavor compounds of ivory shell (Babylonia areolata) and their changes caused by ammonia and salinity stresses were studied. Ammonia stress improved the contents of free amino acids (FAAs), 5′-adenosine monophosphate (AMP), citric acid, and some mineral ions such as Na+, PO43−, and Cl−. The FAA contents decreased with increasing salinity, while the opposite results were observed in most inorganic ions (e.g., K+, Na+, Mg2+, Mn2+, PO43−, and Cl−). Hyposaline and hypersaline stresses increased the AMP and citric acid contents compared to the control group. The equivalent umami concentration (EUC) values were 3.53–5.14 g monosodium glutamate (MSG)/100 g of wet weight, and the differences in EUC values among treatments were mainly caused by AMP. Hexanal, butanoic acid, and 4-(dimethylamino)-3-hydroxy- and (E, E)-3,5-octadien-2-one were the top three volatile compounds, and their profiles were significantly affected when ivory shells were cultured under different ammonia and salinity conditions. Gamma-aminobutyric acid (GABA) is an important non-proteinogenic amino acid and a potent bioactive compound with many anti-hypertensive and anti-depressant activities. The bioconversion of GABA by glutamic acid decarboxylase (GAD) has been eagerly studied. Herein, novel pyridoxal-5-phosphate monohydrates (PLP)-dependent GAD, which is not quite similar to reporting, was cloned from Latilactobacillus curvatus and efficiently expressed in E. coli. The conveniently purified GAD (designated LcGAD10s) appeared as a single protein on SDS-PAGE with a molecular mass of 52.0 kDa. LcGAD10s exhibited a specific activity of 303.7 U/mg after purification by Ni–IDA affinity chromatography, with optimal activity at 55 °C and pH 5. LcGAD10s displayed excellent temperature (50 °C) and pH (4–8) stability which relative activity above 80% and 70%, respectively. The enzymatic activity was, respectively, increased and depressed by 130%, and 24% in the presence of Mn+ and Cu2+. Enzyme activity over 90% can be achieved by adding at least 25 mM of PLP. LcGAD10s was able to efficiently transform 15 g/L GABA with a single-factor optimized reaction of pH (5), temperature (50 °C), time (2 h), LcGAD10s dosage (0.4 U) and monosodium glutamate level (5 g/L). Additionally, LcGAD10s can be applied to a tofu fermentation system to achieve GABA conversion and achieved 14.9 mg/g of GABA conversion when added at 2 U/mL, which is higher than most of the commercial sufu and previous application reports, increasing its functional substances. Aim:Although monosodium glutamate is widely used in the food industry, it has effects in terms of teratogenicity, especially during pregnancy and embryogenesis. Our study analyses the effect on the development of the notochord by giving monosodium glutamate to chicken egg embryos.Methods:The incubation and embryonic development follow-up of our study were held in the experimental animals laboratory of University of Health Sciences Turkey, Istanbul Bakirkoy Prof. Dr. Mazhar Osman Mental Health and Neurological Diseases Training and Research Hospital, and the preparation and microscopy of the samples were carried out in the Pathology Department of the Cerrahpasa Faculty of Medicine. 120 fertile, pathogen-free eggs were incubated at 75% humidity and 37.4±0.2 °C until embryos reached Hamburger and Hamilton stages 9-10 (30th hour). Eggs were divided into four groups. Group 1 consisted of uninjected eggs, group 2 consisted of eggs injected with saline (10 nL 0.9 NaCl), group 3 consisted of eggs injected with 15 mg/kg monosodium glutamate (MSG); and group 4 consisted of eggs injected at regular intervals.Results:The correlation values between height, dilatation, autolysis, chest, and lumbar diameters of embryos in all groups were examined. The mean embryo length was 1.0017±0.36 cm. The mean chest diameter was 20.70±14.45 cm. The mean lumbar diameter was 10.31±14.34 cm. When the groups were compared in terms of embryo length, it was observed that the groups given MSG were significantly shorter than the control and SF groups (p=0.00).Conclusion:The amount of MSG taken with food is important because it can affect the organs. Depending on the amount consumed, MSG may adversely affect notochord development.

This systematic review analyzes monosodium glutamate (MSG) in the Alzheimer’s disease-like condition to enhance translational research. Our review seeks to understand how MSG affects the brain and causes degenerative disorders. Due to significant preclinical data linking glutamate toxicity to Alzheimer’s disease and the lack of a comprehensive review or meta-analysis, we initiated a study on MSG’s potential link. We searched PubMed, ScienceDirect, ProQuest, DOAJ, and Scopus for animal research and English language papers without time constraints. This study used the PRISMA-P framework and PICO technique to collect population, intervention or exposure, comparison, and result data. It was registered in PROSPERO as CRD42022371502. MSG affected mice’s exploratory behaviors and short-term working memory. The brain, hippocampus, and cerebellar tissue demonstrated neuronal injury-related histological and histomorphometric changes. A total of 70% of MSG-treated mice had poor nesting behavior. The treated mice also had more hyperphosphorylated tau protein in their cortical and hippocampus neurons. Glutamate and glutamine levels in the brain increased with MSG, and dose-dependent mixed horizontal locomotor, grooming, and anxiety responses reduced. MSG treatment significantly decreased phospho-CREB protein levels, supporting the idea that neurons were harmed, despite the increased CREB mRNA expression. High MSG doses drastically lower brain tissue and serum serotonin levels. In conclusion, MSG showed AD-like pathology, neuronal atrophy, and short-term memory impairment. Monosodium glutamate (MSG) is one of the most popular food additives in the world and is often ingested with commercially processed foods. It can be described as a sodium salt of glutamic acid with the IUPAC name - Sodium 2-aminopentanedioate and is ionized by water to produce free sodium ions and glutamic acid. MSG use has significantly increased over the past 30 years, its global demand stands huge at over three million metric tons which is worth over $4.5 billion. Asia was responsible for more than three quarter of world MSG consumption with the country China also leading in global consumption as well as production and export to other countries. Prior to year 2020, global demand for MSG increased by almost four percent each year with the highest significant increase in demand for MSG predicted to rise in Thailand, Indonesia, Vietnam and China, followed by Brazil and Nigeria. However, several researches featured in this review has identified MSG consumption as a major contributor to the development and progression of some metabolic disorders such as obesity, which is a risk factor for other metabolic syndromes like hypertension, diabetes mellitus and cancer initiation. The mechanism by which MSG induce obesity involves induction of hypothalamic lesion, hyperlipidemia, oxidative stress, leptin resistance and increased expression of peroxisome proliferator-activated receptors (PPARs) Gamma and Alpha. Similarly for induction of diabetes mellitus, MSG consumption resulted in decreased pancreatic beta cell mass, increased oxidative stress and metabolic rates, reduced glucose and insulin transport to adipose tissue and skeletal muscles, insulin insensitivity, reduced insulin receptors and induced severe hyperinsulinemia. Dietary salt, an active component of MSG is also found to be a major risk factor for high blood pressure (which may lead to hypertension). MSG is used to enhance the taste of tobacco, causing smokers to consume the product in excess and thereby increasing the risk of cancer development. Depending on the amount consumed, MSG has both positive and negative effects. Despite the controversy surrounding MSG's safety and its probable contribution to risk of development and progression of metabolic disorders, its global consumption is still very high. Therefore, this article will sensitize the public on the need for cautious use of MSG in foods and also aid regulatory agencies to further review the daily MSG consumption limit based on metabolic toxicities observed at the varied dosages reported in this review.

The expression of canonical chemosensory receptors of the tongue, such as the heteromeric sweet taste (TAS1R2/TAS1R3) and umami taste (TAS1R1/TAS1R3) receptors, has been demonstrated in many extra-oral cells and tissues. Gene expression studies have revealed transcripts for all TAS1 and metabotropic glutamate (mGlu) receptors in different types of immune cells, where they are involved, for example, in the chemotaxis of human neutrophils and the protection of T cells from activation-induced cell death. Like other class-C G protein-coupling receptors (GPCRs), TAS1Rs and mGlu receptors form heteromers within their families. Since mGlu receptors and TAS1R1/TAS1R3 share the same ligand, monosodium glutamate (MSG), we hypothesized their hitherto unknown heteromerization across receptor families in leukocytes. Here we show, by means of immunocytochemistry and co-IP/Western analysis, that across class-C GPCR families, mGlu2 and TAS1R3 co-localize and heterodimerize in blood leukocytes. Expressing the recombinant receptors in HEK-293 cells, we validated their heterodimerization by bioluminescence resonance energy transfer. We demonstrate MSG-induced, mGlu2/TAS1R3 heteromer-dependent gain-of-function and pertussis toxin-sensitive signaling in luminescence assays. Notably, we show that mGlu2/TAS1R3 is necessary and sufficient for MSG-induced facilitation of N-formyl-methionyl-leucyl-phenylalanine-stimulated IL-8 secretion in neutrophils, using receptor-specific antagonists. In summary, our results demonstrate mGlu2/TAS1R3 heterodimerization in leukocytes, suggesting cellular function-tailored chemoreceptor combinations to modulate cellular immune responses. Gamma-aminobutyric acid (GABA) is a widely available non-protein amino acid whose physiological importance goes beyond its role as an inhibitory neurotransmitter in mammals. The GABA synthesis ability of ten strains of Lactiplantibacillus plantarum was screened. They produced GABA ranging from 48.19 ± 3.44 to 100.75 ± 1.63 mg/L at 24 h-cultivation. Among them, Lp. plantarum FRT7 showed the highest GABA production. Therefore, FRT7 was chosen for GABA yield optimization. A one-factor-at-a-time strategy analysis of the GABA yield of FRT7 was performed, including the culture temperature, incubation time, inoculum volume, initial pH, the initial amount of monosodium glutamate (MSG), and pyridoxal 5'-phosphate (PLP) concentration, based on which the response surface methodology (RSM) was performed. After being cultured in an MRS culture medium supplemented with 3% MSG and 2 mmol/L of PLP at 40 °C with an initial pH of 7.0 for 48 h, the GABA reached a maximum yield of 1158.6 ± 21.22 mg/L. The results showed the experimental value of the GABA yield was in good agreement with the predicted values. Furthermore, the results from the RSM also indicated that the initial MSG addition, PLP concentration, and incubation time were significant variables. These results suggest that Lp. plantarum FRT7 has the potential to be a health-beneficial probiotic with commercial capabilities. [ABSTRACT FROM AUTHOR]

This study aimed to evaluate the effect of replacing NaCl with KCl and monosodium glutamate in beef burgers. The effect of this reformulation on physicochemical properties, salt content, color, sensory aspect, and consumers' acceptance was mainly examined. The findings of this study indicated that all treatments did not significantly differ in terms of moisture, color, and cooking loss. The presence of KCl induced little impact on pH. Furthermore, the replacement of NaCl with KCl did not affect the perceived saltiness and bitterness of the products. On the other hand, concerning overall acceptance, burgers with monosodium glutamate were mostly preferred by consumers. It was concluded that lowering sodium content by partially replacing NaCl with KCl and monosodium glutamate is possible even in beef burgers. [ABSTRACT FROM AUTHOR]

We prove under certain conditions that the intersection of two ɸ - 2 -absorbing primary hyperideals is also a ɸ - 2 -absorbing primary hyperideal. Sunflower seeds are rich in protein and can be an excellent raw material for the production of umami peptides. In this study, sunflower seed meal, which was defatted at a low temperature, was taken as the raw material, and proteins were separated, followed by hydrolyzation for 4 h by flavourzyme® to obtain hydrolysates with strong umami intensity. These hydrolysates were deamidated using glutaminase to increase the umami intensity. The highest umami value of 11.48 was recorded for hydrolysates deamidated for 6 h, and the umami intensity was determined. The umami hydrolysates mixed with 8.92 mmol IMP + 8.02 mmol MSG showed the highest umami value of 25.21. Different concentrations of ethanol were used for further separation of hydrolysates, and the highest umami value of 13.54 was observed for 20% ethanol fraction. The results of this study provide utilization method for sunflower seed meal protein and a theoretical basis for the preparation of umami peptides. Practical Application: A large number of sunflower seed meals after oil production are used as feed for livestock and poultry. Sunflower seed meal is rich in protein, and umami amino acid composition in sunflower seed meal is up to 25%–30%, which is potentially an excellent raw material for the production of umami peptides. The umami flavor and synergistic effect of obtained hydrolysates, with MSG and IMP, were analyzed in the present study. A group of Bacillus sp. was extracted from monosodium glutamate wastewater. Lignocellulose/montmorillonite composite was selected as the carrier. Lignocellulose/montmorillonite composite immobilized Bacillus sp./calcium alginate microspheres were prepared by immobilized microorganism techniques. The microspheres were used to treat monosodium glutamate wastewater with significantly reduced ammonia nitrogen (NH3-N) and chemical oxygen demand (COD) concentrations. The optimum preparation conditions of microspheres in the treatment of NH3-N and COD of monosodium glutamate wastewater were studied. The concentration of sodium alginate was 2.0 wt%, lignocellulose/montmorillonite was 0.06 wt%, Bacillus sp. was 1.0 wt%, CaCl2 solution was 2.0 wt%, coagulation time was 12 h, and the removal capacities of NH3-N and COD were 44832 and 78345 mg/L, respectively. The surface structure, element content, functional group change, and crystal structure of the microspheres were characterized by SEM, EDS, and other methods. The results showed that the -COOH in lignocellulose/montmorillonite and the -OH in the Bacillus sp. form intermolecular hydrogen bonds. The Si–O and Al-O bonds in lignocellulose/montmorillonite reacted with sodium ions in sodium alginate. New crystal structures appear inside the material after crosslinking, and the microspheres was formed. Thus, the study has shown that the microspheres were successfully prepared and contributes to the treatment of NH3-N and COD in monosodium glutamate wastewater. This work can provide an interesting strategy for the removal of COD and NH3-N in industrial wastewater by reasonably combining bio-physicochemical processes. [ABSTRACT FROM AUTHOR]

γ-aminobutyric-acid (GABA) is a mental health-supporting substance that helps release anxiety and depression and improves memory. Lactiplantibacillus plantarum SKKL1, a GABA-producing bacterium, has been introduced to formulate a gut-brain axis product. However, growth and sugar consumption of L. plantarum SKKL1 in milk were ineffective. This obstacle was investigated by varying different types of milk, sugars, fermentation temperatures, and times. The results revealed that none of these parameters improved growth and bacterial metabolism in milk, except addition of soluble protein as found in yeast extract and malt extract. Although a protease deficiency of L. plantarum SKKL1 was discovered, it was not a primary barrier to cell propagation. Insight of this study showed clearly that soluble protein was an essential metabolic activator for growth, nutrient consumption, and protease synthesis, then stimulated lactic acid and GABA productions. While, milk casein and casein hydrolysate, a complex protein structure with low solubility, were not utilized by L. plantarum SKKL1. The novelty of this study is the first in-depth investigation to confirm a significant effect of soluble protein on enrich-GABA milk fermentation by L. plantarum SKKL1 as the sole starter without protease and monosodium glutamate addition.
