Nanoplastics induce transgenerational toxicity to aquatic organisms, but the specific pathways for the maternal transfer of nanoplastics remain unclear. Herein, laser scanning confocal microscopy (LSCM) observations identified the specific pathways on the maternal transfer of polystyrene (PS) nanoplastics (25 nm) in Daphnia magna . In vivo and in vitro experiments showed that PS nanoplastics could enter the brood chamber through its opening and then be internalized to eggs and embryos using LSCM imaging (pathway I). In addition, PS nanoplastics were observed in the oocytes of the ovary, demonstrating gut-ovary-oocyte transfer (pathway II). Furthermore, label-free hyperspectral imaging was used to detect the distribution of nanoplastics in the embryos and ovary of Daphnia , again confirming the maternal transfer of nanoplastics through the two pathways mentioned above. The contribution from pathway I (88%) was much higher than pathway II (12%) based on nanoflow cytometry quantification. In addition, maternal transfer in Daphnia depended on the particle size of PS nanoplastics, as demonstrated by using LSCM and hyperspectral imaging. Unlike 25 nm nanoplastics, 50 nm PS nanoplastics could enter the brood chamber and the eggs/embryos (pathway I), but were not detected in the ovary (pathway II); 100 nm PS nanoplastics were difficult to be internalized by eggs/embryos and could not enter the ovary either. These findings provide insight into the maternal transfer mechanisms of nanoplastics in Daphnia , and are critical for better understanding the transgenerational toxicity of aquatic organisms.

Mercury (Hg) is a highly toxic and widely distributed metal that is bioaccumulated in insectivorous mammals and may cause adverse effects on the reproductive system. Bats are considered excellent Hg bioindicators due to their wide distribution, life span, trophic position, metabolic rate and food intake. However, few studies have analysed Hg residues in bats, and to the best of our knowledge, no studies have been made in the Iberian Peninsula. The main aim of this study was to undertake the first ever assessment of Hg exposure in Schreiber's bent-winged bats inhabiting a natural cave in the southeast of Spain. The findings suggest that Schreiber's bent-winged bats in the sampling area are chronically exposed to low levels of Hg. The Hg concentrations found in different tissues (fur, kidney, liver, muscle and brain) were below the threshold levels associated with toxic effects in mammals. Non-gestating females showed Hg concentrations in the brain and muscle that doubled those found in gestating females. This could be due to Hg mobilization from the mother to the foetus in gestating females, although other factors could contribute to explain this result such as variations in hunting areas and the insect-prey consumed and/or different energetic needs and average food consumption during the breeding season. Hg levels were 1.7 times higher, although not significant, in foetus' brains than in the maternal brains, and Hg concentration in foetus' brain was significantly correlated with levels in the corresponding mothers' kidney. These results suggest that there could be an active mother-to-foetus transfer of Hg in bats, which would be of special relevance in a scenario of higher Hg exposure than that found in this study. However, further research is needed to support this view due to the limited number of samples analysed. Given the scarce ecotoxicological data available for bats and their protected status, we encourage further opportunistic studies using carcasses found in the field, the validation of non-destructive samples such as fur and guano for Hg monitoring, and new modelling approaches that will increase the data needed for proper ecological risk assessment in bat populations. Global climate changes are magnified in the Arctic and are having an especially dramatic effect on the spatial and temporal distribution and the thickness traits of sea ice. Decline of Arctic sea ice may lead to qualitative and/or quantitative changes in diet and reduced body condition (i.e. adipose tissue stores) of ice-associated apex predators such as polar bears ( Ursus maritimus ). This may further affect their tissue concentrations of lipophilic pollutants. We determined how variations in adipose tissue stores associated to both breeding status and spatial changes in sea ice conditions and diet influence concentrations and biotransformation of lipophilic persistent organic pollutants (POPs). We collected 112 blood and fat samples from female polar bears ( Ursus maritimus ) of different breeding status (alone, with cubs of the year, or with yearlings) during two seasons (April and September) in 2012 and 2013 at three locations of Svalbard, Norway, with contrasted sea ice conditions. We inferred diet from nitrogen and carbon stable isotope ratios in red blood cells and fatty acid composition in adipose tissue. Relative to diet, body condition, which was negatively related to sea ice extent at both temporal and spatial scales, was the most important predictor for concentrations of POPs in plasma and fat, whereas diet showed a minor influence. Additionally, fatter females were more efficient at biotransforming PCBs than were leaner ones. Breeding status influenced the concentrations of less lipophilic compounds such as β -hexachlorocyclohexane, which were lower in females with yearlings, probably due to excretion into milk and subsequent offloading to young. In conclusion, our results indicate that declining sea ice indirectly leads to increased concentrations of lipophilic pollutants in polar bears mediated through reduced feeding opportunities and declining body condition rather than changes in diet composition. Differences in element concentrations have been observed between embryonated and non-embryonated eggs of the Eurasian Reed Warbler Acrocephalus scirpaceus, presumably due to the resorption of calcium and other elements from the shell. To investigate this difference, we tested the hypotheses that (1) the distribution of elements in eggshells and egg contents, (2) the individual element-element relationships between shells and contents, and (3) the relationships among the elements in eggshells and egg contents vary between the two egg samples. Based on the levels of 11 elements (Cr, Cu, Ni, Cd, Pb, Mn, Fe, Co, Zn, Mg and Ca) in Reed Warbler eggs, our results confirmed the second and third hypotheses. Importantly, we found that embryonic development tended to blur the lines between two types of relationships: (1) between elements in eggshells and egg contents through an increase in the number of significant correlations, some of which may be enhanced by high levels of certain elements (Cd, Co, Pb, Zn are indicative of trace metal pollution in fish ponds); (2) among the various elements in eggshells and egg contents, as demonstrated by the differential grouping of elements in principal components analysis (PCA). The consistent positive direction of relationships for almost all elements in both the shells and contents of eggs is presumed to result from the initial parallel co-sequestration of all the elements which accompanies intensive calcium transfer from the female body into the incipient egg. Our study highlights the fact that only limited inferences can be drawn from the interactions of metals and calcium in eggshells and egg contents if the fertility status of the eggs is unknown. The maternal transfer of trace elements is a process by which offspring may accumulate trace elements from their maternal parent. Although maternal transfer has been assessed in many vertebrates, there is little understanding of this process in invertebrate species. This study investigated the maternal transfer of 13 trace elements (Ag, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se, and Zn) in Atlantic horseshoe crab (Limulus polyphemus) eggs and compared concentrations to those in adult leg and gill tissue. For the majority of individuals, all trace elements were transferred, with the exception of Cr, from the female to the eggs. The greatest concentrations on average transferred to egg tissue were Zn (140 µg/g), Cu (47.8 µg/g), and Fe (38.6 µg/g) for essential elements and As (10.9 µg/g) and Ag (1.23 µg/g) for nonessential elements. For elements that were maternally transferred, correlation analyses were run to assess if the concentration in the eggs were similar to that of adult tissue that is completely internalized (leg) or a boundary to the external environment (gill). Positive correlations between egg and leg tissue were found for As, Hg, Se, Mn, Pb, and Ni. Mercury, Mn, Ni, and Se were the only elements correlated between egg and gill tissue. Although, many trace elements were in low concentration in the eggs, we speculate that the higher transfer of essential elements is related to their potential benefit during early development versus nonessential trace elements, which are known to be toxic. IgZ is considered to be the last immunoglobulin discovered in vertebrate species. In this study, the structure of secreted form of blunt snout bream ( Megalabrama amblycephala ) IgZ (sIgZ) heavy chain gene is VH-Cζ1-Cζ2-Cζ3-Cζ4, in which Cζ4 provides the specificity to the IgZ isotype. The deduced amino acid sequence of sIgZ shows highest similarity with that of Ctenopharyngodon idella (79%). The ontogeny of sIgZ gene expression showed a V-shape change pattern: decreased initially from unfertilized egg stage to 16-cell embryos and increased significantly from blastula stage, which exhibited maternal transmission effects. Compared with the juvenile fish, sIgZ mRNA level was higher in head kidney, spleen, trunk kidney, liver, intestine and gill of adult fish. In both juvenile and adult fish, sIgZ mRNA was detected in intestine and gill. Aeromonas hydrophila challenge experiment showed that sIgZ transcription significantly increased in skin, gill and intestine, indicating a prominent mucosal immune response. The results of Western blot also verified the protein alterations of sIgZ in mucosal organs in M. amblycephal . Our studies indicate a prominent role of IgZ in mucosa-associated lymphoid tissue immunity and further support the specialized role of IgZ in teleost mucosal immune responses. [ABSTRACT FROM AUTHOR]

Mercury (Hg) is a ubiquitous environmental contaminant and potent neurotoxin. In aquatic environments, Hg can be transformed into methylmercury (MeHg), which bioaccumulates in aquatic food webs, including fish. Methylmercury has been shown to transfer from female fish to developing eggs; however, relatively little is known regarding the effects of maternally transferred MeHg on fish embryos. The present study evaluated the effects of maternally transferred MeHg on fathead minnow ( Pimephales promelas) embryos. Embryos were collected from adult fatheads exposed for 30 d to 1 of 3 diets spiked with MeHg: a control diet (0.02 ppm Hg dry wt), a low diet (0.87 ppm Hg dry wt), or a high diet (5.5 ppm Hg dry wt). No effects on spawning frequency, clutch size, or total egg output were observed. In embryos, Hg concentration was a function of female diet and the duration (number of days) of female exposure. Compared with controls, embryos from the low-diet treatment displayed altered embryonic movement patterns (hyperactivity) and decreased time to hatch. Embryos from the high-diet treatment had delayed hatching and increased mortality compared with the other treatments. Collectively, these results suggest that maternally transferred Hg may impact survival, behavior, and developmental milestones of the embryo-larval stages of fish. Biomagnification of methylmercury in aquatic systems can cause elevated tissue mercury (Hg) and physiological stress in top predators. Mercury is known to affect stress hormone levels in mammals, birds and fish. In this study, the effects of maternally-transferred methylmercury on the stress physiology of Northern Water Snake ( Nerodia sipedon) neonates were tested. Gravid females were dosed via force-fed capsules during late gestation with 0, 0.01, or 10 µg methylmercury per gram of body mass. Plasma corticosterone levels and leukocyte differentials were analyzed in baseline and confinement-stressed neonates from all dose levels. Neither Hg nor confinement stress had a significant effect on leukocyte differentials nor was Hg related to corticosterone levels. However, stress group neonates showed lower heterophil/lymphocyte ratios and this study was the first to show that neonate N. sipedon can upregulate CORT in response to stress. The tissue concentrations of dechlorane plus and its analogues were determined in ducks collected from several e-waste recycling villages of Taizhou, China. Compared with the published literature, the relatively high concentrations of these compounds were detected in ducks, indicating serious DP contamination. Since both the duck meat and eggs were important components for diet, this result reminded us of keeping a watchful eye on human dietary exposure to DP and its analogues in this study area. The wet-weight concentrations of DP and its analogues were significantly related to tissue lipid content ( p < 0.05 ), indicating that the lipid pools predominantly impacted the distribution of DPs in ducks. On the basis of lipid adjustment, the significantly lower levels in brain than those in liver and blood, displayed the occurrence of liver sequestration and blood–brain barrier to DP and its analogues in the duck ( p < 0.05). The maternal transfer of DP and Mirex was not obviously limited, and the transferring extent of Dec 602 was over one. The stereo-selected accumulation of two DP isomers occurred among tissues with preference to syn -DP in blood, and to anti -DP in brain. We conducted a detailed assessment of the maternal transfer of mercury and selenium to eggs in three bird species ( n = 107 parents and n = 339 eggs), and developed predictive equations linking contaminant concentrations in eggs to those in six tissues of the mother (blood, muscle, liver, kidney, breast feathers, and head feathers). Mercury concentrations in eggs were positively correlated with mercury concentrations in each of the mother's internal tissues ( R 2 ≥ 0.95), but generally not with feathers. For each species, the proportion of mercury transferred to eggs decreased as mercury concentrations in the mother increased. At the same maternal mercury concentration, the proportion of mercury transferred to eggs differed among species, such that Forster's tern ( Sterna forsteri ) and black-necked stilt ( Himantopus mexicanus ) females transferred more methylmercury to their eggs than American avocet ( Recurvirostra americana ) females. Selenium concentrations in eggs also were correlated with selenium concentrations in the mother's liver ( R 2 = 0.87). Furthermore, mercury and selenium concentrations in tern eggs were positively correlated with those in the father ( R 2 = 0.84). Incubating male terns had 21% higher mercury concentrations in blood compared to incubating females at the same egg mercury concentration. Persistent organic pollutants (POPs), including brominated flame retardants (BFRs), perfluoroalkyl substances (PFAS) and metals, can accumulate in marine mammals and be transferred to offspring. In this study, we analyzed 64 lipophilic POPs, including four emerging BFRs, in the blubber, liver and muscle of 17 adult common minke whales (Balaenoptera acutorostrata) from the Barents Sea to investigate occurrence and tissue partitioning. In addition, the placental transfer concentration ratios of 14 PFAS and 17 metals were quantified in the muscle of nine female-fetus pairs to investigate placental transfer. Legacy lipophilic POPs were the dominating compound group in every tissue, and we observed generally lower levels compared to previous studies from 1992 to 2001. We detected the emerging BFRs hexabromobenzene (HBB) and pentabromotoluene (PBT), but in low levels compared to the legacy POPs. We detected nine PFAS, and levels of perfluorooctane sulfonate (PFOS) were higher than detected from the same population in 2011, whilst levels of Hg were comparable to 2011. Levels of lipophilic contaminants were higher in blubber compared to muscle and liver on both a wet weight and lipid adjusted basis, but tissue partitioning of the emerging BFRs could not be determined due to the high number of samples below the limit of detection. The highest muscle ΣPFAS levels were quantified in fetuses (23 ± 8.7 ng/g ww), followed by adult males (7.2 ± 2.0 ng/gg ww) and adult females (4.5 ± 1.1 ng/g ww), showing substantial placental transfer from mother to fetus. In contrast, Hg levels in the fetus were lower than the mother. Levels were under thresholds for risk of health effects in the whales. This study is the first to report occurrence and placental transfer of emerging contaminants in common minke whales from the Barents Sea, contributing valuable new data on pollutant levels in Arctic wildlife. Pesticides could induce long-term impacts on aquatic ecosystem via transgenerational toxicity. However, for many chiral pesticides, the potential enantioselectivity of transgenerational toxicity has yet to be fully understood. In this study, we used zebrafish as models to evaluate the maternal transfer risk of tebuconazole (TEB), which is a chiral triazole fungicide currently used worldwide and has been frequently detected in surface waters. After 28-day food exposure (20 and 400 ng/g) to the two enantiomers of TEB (S - and R -TEB) in adult female zebrafish (F0), increased malformation rate and decreased swimming speed were found in F1 larvae, with R -TEB showing higher impacts than S -enantiomer. Additionally, enantioselective effects on the secretion of thyroid hormones (THs) and expression of TH-related key genes along the hypothalamic-pituitary-thyroid (HPT) axis were found in both F0 and F1 after maternal exposure. Both the two enantiomers significantly disrupted the triiodothyronine (T3) and thyroxine (T4) contents in F0 with different degrees, whereas in F1, significant effects were only found in R -TEB groups with decreasing of both T3 and T4 contents. Most of the HPT axis related genes in F0 were upregulated by TEB and more sensitive to R -TEB than to S -TEB. In contrast, most of the genes in F1 were downregulated by both R - and S -TEB, especially the genes that are primarily responsible for thyroid development and growth (Nkx2–1), TH synthesis (NIS and TSHꞵ) and metabolism (Deio1). Mother-to-child transmission is a major route for infections in newborns. Vaccination in mothers to leverage the maternal immune system is a promising approach to vertically transfer protective immunity. During infectious disease outbreaks, such as the 2016 Zika virus (ZIKV) outbreak, rapid availability of vaccines can prove critical in reducing widespread disease burden. The recent successes of mRNA vaccines support their evaluation in pregnant animal models to justify their use in neonatal settings. Here we evaluated immunogenicity of self-amplifying replicon (repRNA) vaccines, delivered with our clinical-stage LION nanoparticle formulation, in pregnant rabbits using ZIKV and HIV-1 as model disease targets. We showed that LION/repRNA vaccines induced robust antigen-specific antibody responses in adult pregnant rabbits that passively transferred to newborn kits in utero. Using a matrixed study design, we further elucidate the effect of vaccination in kits on the presence of pre-existing maternal antibodies. Our findings showed that timing of maternal vaccination is critical in maximizing in utero antibody transfer, and subsequent vaccination in newborns maintained elevated antibody levels compared with no vaccination. The effects of sex and pregnancy on the bioaccumulation and tissue distribution of legacy and emerging per- and polyfluoroalkyl substances (PFASs) in Chinese water snakes were investigated. The bioaccumulation factor of PFASs showed a positive correlation with their protein-water partition coefficients (log K PW . PFAS levels in females were significantly lower than those in males. The chemical composition of pregnant females was significantly different from that of non-pregnant females and males. The maternal transfer efficiencies of perfluorooctane sulfonic acid were higher than those of other PFASs, and a positive correlation between the maternal transfer potential and log 3 . PFAS levels in females were significantly lower than those in males. The chemical composition of pregnant females was significantly different from that of non-pregnant females and males. The maternal transfer efficiencies of perfluorooctane sulfonic acid were higher than those of other PFASs, and a positive correlation between the maternal transfer potential and log K PW was observed for other PFASs. Tissues with high phospholipid content exhibited higher concentrations of ∑PFASs. Numerous physiological changes occurred in maternal organ systems during pregnancy, leading to the re-distribution of chemicals among different tissues. The change in tissue distribution of PFASs that are easily and not-so-easily maternally transferred was in the opposite direction. The extent of compound transfer from the liver to the egg determined tissue re-distribution during pregnancy.

Maternal transfer of metals can be an important exposure route for animals. The maternal transfer of Cr and its effects on reproduction in fish are still largely unknown. In this study, Japanese medaka were exposed to a sublethal Cr(VI) concentration for 6 days (acute) and for 3 months (chronic). Chromium accumulation in the gonads, maternal transfer of Cr, and effects of Cr on the reproduction, histopathology and expressions of antioxidants in the gonads were evaluated. Both acute and chronic exposures resulted in significant Cr accumulation in gonads, eggs and larvae. In chronic Cr-exposed fish, approximately 61% of the Cr accumulated in the ovary was depurated by spawning during the first 3 days after exposure, suggesting that maternal transfer is a very important pathway for accumulation in offspring. The chronic exposure caused decreases in body weight, standard length, gonad weight, gonad-somatic index (GSI) and fecundity. The last of these was most severely affected: the total number of broods and eggs per female decreased by 57.1% and 75.9%, respectively. Moreover, egg weight and fertilization rate were also reduced (by approximately 20%) following chronic Cr(VI) exposure. Histopathological analyses showed that the Cr exposure resulted in the onset of follicular atresia and a reduction in the number of mature oocytes, along with a reduction in abundance of mature spermatozoa in testes. The GSH/GSSG ratio was greatly elevated after chronic Cr(VI) exposure, implying that GSH played a role in scavenging the reactive oxygen species generated by the reduction of Cr(VI) inside cells. This study provides evidence for the maternal transfer of Cr, highlights the importance of spawning in Cr depuration from the ovary, and demonstrates that chronic Cr(VI) exposure has serious impacts on reproduction in the Japanese medaka. Reported concentrations of dioxin-like compounds accumulated in the European eel (Anguilla anguilla) were used to perform a risk assessment for eel larval survival, taking into account a modeled amplification of tissue concentrations with a factor of 1.33 during spawning migration. The calculated concentrations of dioxin-like compounds finally deposited in the eggs were compared with the internal effect concentrations for survival of early life stages of the European eel; these concentrations, by lack of experimental data, were estimated from a sensitivity distribution based on literature data by assuming that eel larvae are among the 10% most sensitive teleost fish species. Given concentrations of dioxin-like contaminants and assuming a relatively high sensitivity, it can be expected that larvae from eggs produced by eel from highly contaminated locations in Europe will experience increased mortality as a result of maternally transferred dioxin-like contaminants. As historical persistent organic pollutant concentrations in eel tissue were higher, this impact must have been stronger in the past. Potential effects of other compounds or effects on the migration, condition, and fertility of the parental animals were not taken into account. Cadmium (Cd) is one of the most toxic heavy metals in aquatic ecosystem which affects fish health and aquaculture. In the present study, we examined the bioaccumulation of Cd in the gonads of tilapia via dissolved and dietary routes. We evaluated the subchronic effects of Cd on the histology of gonads, steroid hormone levels and sex-related gene expressions in tilapia. In addition, we also studied maternal transfer of Cd. Our results indicated that Cd was accumulated significantly in both ovary and testis from both exposure routes. Histopathological analysis showed that Cd induced ovary and testis injuries. Estradiol levels were significantly increased in dissolved Cd exposed female fish. In addition, the Cd exposure displayed different effects on gene expressions in gonads. In females, the estrogen receptor (ERα) was stimulated in dissolved Cd-exposed fish at 70.32 and 143.78 μg/L for 30 days and in fish at 143.78 μg/L for 60 days. Vitellogenin expression was significantly down-regulated in the ovary of dissolved Cd-exposed fish. In testis, GR expression was elevated after 60 days of dissolved Cd and dietary exposure. Furthermore, Cd level was significantly higher in the eggs than that in the fry. Our results demonstrated that both dissolved and dietary Cd exposures affected gonad development by altering steroid hormone levels and sex-related gene expressions in tilapia. The European eel ( Anguilla anguilla ) is regarded as a critically endangered species. Scientists are in agreement that the “quality of spawners” is a vital factor for the survival of the species. This quality can be impaired by parasites, disease and pollution. Especially endocrine disrupting organic chemicals pose a potential threat to reproduction and development of offspring. To our knowledge, the findings in this publication for the first time describe maternal transfer of contaminants in eels. We analysed the concentrations of in total 53 polybrominated diphenyl ethers (PBDEs) and their halogenated substitutes in muscle, gonads and eggs of artificially matured European eels and in muscle and gonads of untreated European eels that were used for comparison. We found evidence that persistent organic pollutants such as PBDEs, as well as their brominated and chlorinated substitutes are redistributed from muscle tissue to gonads and eggs. Concentrations ranged from 0.001 ng g − 1 ww for sum Dechlorane metabolites (DPMA, aCL 10 DP, aCl 11 DP) to 2.1 ng g − 1 ww for TBA in eggs, 0.001 ng g − 1 ww for Dechlorane metabolites to 9.4 ng g − 1 ww for TBA in gonads and 0.002 ng g − 1 ww for Dechlorane metabolites to 54 ng g − 1 ww for TBA in muscle tissue. Average egg muscle ratios (EMRs) for compounds detectable in artificially matured eels from both Schlei Fjord and Ems River ranged from 0.01 for Dechlorane 602 (DDC-DBF) to 10.4 for PBEB. Strong correlations were found between flame retardant concentrations and lipid content in the analysed tissue types, as well as transfer rates and octanol–water partitioning coefficient, indicating that these parameters were the driving factors for the observed maternal transfer. Furthermore, indications were found, that TBP-DBPE, TBP-AE, BATE and TBA have a significant uptake from the surrounding water, rather than just food and might additionally be formed by metabolism or biotransformation processes. Dechloranes seem to be of increasing relevance as contaminants in eels and are transferred to eggs. A change of the isomer pattern in comparison to the technical product of Dechlorane Plus (DP) was observed indicating a redistribution of DP from muscle tissue to gonads during silvering with a preference of the syn-isomer. The highly bioaccumulative DDC-DBF was the most abundant Dechlorane in all fish of the comparison group even though it is not produced or imported in the EU. Lead (Pb) poisoning has caused significant mortality in waterfowl populations worldwide. In spite of having been banned since 2003, prevalence of Pb shot ingestion in mallards ( Anas platyrhynchos ) from the Ebro delta was still 15.5% in 2011–12. We collected mallard eggs from this area to study the effects of maternally transferred Pb on eggshell properties and on immune response and oxidative balance of ducklings. Eggshell Pb levels were positively correlated with Pb levels in the blood of ducklings. Ducklings with blood Pb levels above 180 ng mL −1 showed reduced body mass and died during the first week post hatching. Blood Pb levels positively correlated with humoral immune response, endogenous antioxidants and oxidative stress biomarkers, and negatively correlated with cellular immune response. Steller's eiders and spectacled eiders are sea duck species whose populations have declined significantly and infectious diseases could influence offspring survival. Therefore, the maternal transfer of immunoglobulin Y (IgY) into yolk was investigated in captive Steller's and spectacled eiders during the 2007–2013 breeding seasons. This project had two objectives: establish baseline IgY levels in Steller's and spectacled eider yolk under controlled captive conditions and evaluate the effect of year, laying date, egg fertility, egg incubation duration, individual hen, hen age and mass, and laying order to determine which variables influenced IgY levels. Average IgY concentrations were 0.03–0.48 mg ml −1 in Steller's eider yolk and 0.10–0.51 mg ml −1 in spectacled eider yolk. The year and individual hen influenced IgY concentration in Steller's and spectacled eider yolk. The laying date was negatively correlated with egg IgY levels for most Steller's eider hens, but laying order was positively correlated with egg IgY concentration for spectacled eiders. Concentrations of PCBs, organochlorine pesticides (OCPs), brominated flame retardants and a suite of relevant metabolites of these POPs, in all 175 different compounds, were determined in liver and plasma of traditionally hunted pilot whales (n = 14 males and n = 13 females of different age groups) from the Faroe Islands. The main objectives of this study were to determine differences in the presence and concentrations of the compounds in the liver and plasma, how they depend on developmental stage (calves, sub adults, and adult females), and to assess maternal transfer of the compounds to suckling calves. Generally, the lipid weight (lw) concentrations of quantified POPs in the liver and plasma of pilot whales were positively correlated, and lw concentrations of most POPs did not differ between these matrices. However, concentrations of some individual POPs differed significantly (p < 0.05) between plasma and liver; CB-153 (p = 0.044), CB-174 (p = 0.027) and BDE-47 (p = 0.017) were higher in plasma than in liver, whereas p,p′-DDE (p = 0.004) and HCB (p < 0.001) were higher in liver than in plasma. POP concentrations differed between age/gender groups with lower levels in adult females than in juveniles. The relative distribution of compounds also differed between the age groups, due to the influence of the maternal transfer of the compounds. The results indicated that larger, more hydrophobic POPs were transferred to the offspring less efficiently than smaller or less lipid soluble compounds. Very low levels of both OH- and/or MeSO 2 -PCB and PBDE metabolites were found in all age groups, with no significant (p > 0.05) differences between the groups, strongly suggesting a very low metabolic capacity for their formation in pilot whales. The lack of difference in the metabolite concentrations between the age groups also indicates less maternal transfer of these contaminant groups compared to the precursor compounds. The present study assessed the effects of maternal copper or cadmium exposure in a live-bearing fish. After a 10-d exposure to background levels (control) or 0.15 μM copper or cadmium, gravid females were transferred to clean water. Once a female gave birth, the authors analyzed her newborn offspring for lipid peroxidation, elemental composition (copper, cadmium, and calcium), and metal tolerance. The authors raised other offspring until sexual maturity and analyzed their growth rate, incidence of abnormalities, and sex ratio. Their earlier research, using the same species and exposure design, demonstrated that cadmium and copper were transferred from gravid females to their offspring. The present study showed that offspring of copper-exposed females had a reduced size at birth, developmental abnormalities, elevated tissue cadmium levels, and reduced tissue calcium levels. Offspring of cadmium-exposed females had elevated levels of lipid peroxidation, developmental abnormalities, and lower tissue levels of both copper and calcium. No effects were detected with respect to offsprings' growth rate or sex ratio. Offspring of metal-exposed fish had a reduced tolerance to the metal that their female parent had been exposed to, and the tolerance showed an inverse relationship to the tissue metal level in the offspring. The latter indicates that the reduced tolerance was the result of an increased body burden prior to the tolerance quantification. When females care for broods of mixed paternity, there is a threat of enhanced rivalry among offspring of different paternal lineages. This competition is against the best evolutionary interest of the female because she is equally related to all of her offspring and enhanced offspring competition would decrease her fitness. It was hypothesized earlier that mothers should therefore conceal the information about paternal origin in the recognition cues used by her offspring, for example by transferring the chemical cues used by her offspring for kin recognition. Here, we used a cross-fostering experiment between two closely related and sympatric earwig species, Forficula auricularia and F. decipiens, to demonstrate that F. auricularia nymphs tended by F. decipiens females differ in their cuticular hydrocarbon profiles from F. auricularia nymphs tended by F. auricularia females. The function of IgD in fish and mammals has not been fully understood since its discovery. In this study, we have isolated and characterized the cDNA that encodes membrane-bound form of the immunoglobulin D heavy chain gene (mIgD) of blunt snout bream (Megalobrama amblycephala) using RT-PCR and rapid amplification of cDNA ends (RACE). The full-length cDNA of mIgD consisted of 3313 bp, encoding a putative protein of 943 amino acids. The structure of blunt snout bream mIgD is VDJ-ì1-ä1-ä2-ä3-ä4-ä5-ä6-ä7-TM. Multiple alignment and phylogenetic analyses indicated that blunt snout bream mIgD clusters with the homologues of cyprinid fish and that its highest identity is with that of C. idella (82%). The mIgD expression in early different developmental stages showed that the level of mIgD mRNA decreased dramatically from the unfertilized egg stage to the 32-cell stage, suggesting that mIgD mRNA was maternally transferred. As cell differentiation initially took place in the blastula stage, the mIgD expression increased significantly from the blastula stage to prelarva, which might be attributed to embryonic stem cell differentiation processes. Compared with juvenile fish, the expression and tissue distribution patterns of mIgD in adult individuals exhibited considerable variation. After the injection of Aeromonas hydrophila, mIgD expression was up-regulated in various tissues, reaching the peak expression at 5 d, 14 d or 21 d (depending on the tissue type). This study investigated the effect of maternal copper and maternal cadmium exposure on life history variables and population dynamics in a live-bearing fish species. Gravid females were exposed to copper, cadmium, or background metal levels (control); maternal transfer of the metals was previously demonstrated using the exact same design. Each female's first brood, born after the exposure, was subdivided into two groups. One group was raised in the laboratory, to assess time-to and size-at sexual maturity, reproductive output and other life history variables. Offspring from the other group were used to start four mesocosm populations for each treatment. These populations were sampled monthly, for about 18 months, to assess population dynamics. For the laboratory-reared fish, offspring of copper-exposed females reached sexual maturity at a smaller size than did offspring from the other treatments. Maternal copper exposure and maternal cadmium exposure both resulted in fewer broods and an increase in gestation time. No impacts were detected for brood size, inter-brood interval, time-to-sexual-maturity, or life span. In the greenhouse population study, no effect of maternal copper or cadmium exposure was evident for population parameters, other than that the relative abundance of juveniles and/or newborns was reduced in populations established with offspring of the exposed females. This study provided evidence that a short-term metal exposure of gravid females can negatively affect their offspring's life history variables and potentially influence population dynamics in a life-bearing fish species. Elasmobranchs can bioaccumulate considerable amounts of persistent organic pollutants (POPs) and utilize several reproductive strategies thereby influencing maternal transfer of contaminants. This study provides preliminary data on the POP transfer from pregnant females to offspring of three species (Atlantic stingrays, bonnethead, blacktip sharks) with different reproduction modes (aplacental, placental viviparity). Polychlorinated biphenyl (PCB) levels were generally higher than any other POPs. Stingrays and blacktip shark embryos contained the lowest POP concentrations while bonnetheads and the blacktip adult female had the highest concentrations. Results suggest that POPs are more readily transferred from the mother to the embryo compared to what is transferred to ova in stingrays. Statistically significant differences in levels of selected POPs were found between embryos from the left and right uterus within the same litter as well as between female and male embryos within the same litter for bonnetheads, but not for the blacktip sharks. Marine mammals are one of the groups of animals most affected by marine pollution including that by organic compounds which, besides bearing recognised harmful effects to adults, they may also affect foetuses through placental transfer. In this study we analysed samples of fin whale (Balaenoptera physalus) females and their foetuses collected in the western Iceland summer feeding grounds in 2018. Three different families of organic pollutants: organophosphate esters (OPEs); halogenated flame retardants (HFRs); and short chain chlorinated paraffins (SCCPs), were analysed and their placental transfer investigated. HFRs were detected in 87.5% of females and 100% of foetus samples with concentrations that ranged between nd-15.4 and 6.37–101 ng/g lipid weight (lw), respectively. OPEs were detected in all samples, both from females (85.8–567 ng/g lw) and foetuses (nq-1130 ng/g lw). SCCPs were detected in 87.5% of female samples and 100% of foetal samples with concentrations that ranged between nd-30.9 and nq-574 ng/g lw, respectively. For OPE compounds, a significant negative correlation was observed between the logarithm maternal transfer ratio and their log K ow, indicating that a high lipophilicity reduced placental transfer rate. Interestingly, the decabromodiphenyl ethane (DBDPE) was the compound with the highest log K ow but also the one that was transferred the most from mothers to foetuses, calling for in-depth research on this pollutant. These results constitute the first evidence of mother-calf transfer of plasticizers and flame retardants in fin whales. Further investigations are needed to determine their potential effects on this species and other groups of animals. Although many studies have documented the wide occurrence of polybrominated diphenyl ethers (PBDEs) in cetaceans, little evidence exists regarding the detrimental effects of PBDE exposure on calf death rates for free-ranging cetaceans. This study analyzed life-history-associated PBDE bioaccumulation patterns in 128 stranding Indo-Pacific humpback dolphin ( Sousa chinensis ) samples over an 18-year timespan from the Pearl River Estuary (PRE). In comparison to the records of PBDE levels in cetaceans worldwide, the median levels of PBDEs (median = 10600 ng g -1 lw, range = 721-50900 ng g -1 lw) in all samples were the highest to date. One-way analysis of variance (ANOVA) showed that adult males (median = 16100 ng g -1 lw, range = 4070-50900 ng g -1 lw) and calves (12000 ng g -1 lw, range = 1250-35300 ng g -1 lw) both had the highest levels of PBDEs compared to the rest of the age/sex groups ( p < 0.05). Concentrations of PBDEs in noncalves significantly decreased over the studied period, while those in calves had a slightly increasing trend, which may be due to different exposure routes via fish or milk, respectively. A significant and positive relationship was found between annual calf stranding death rates and body-length-adjusted PBDE concentrations in calves ( r = 0.62, p < 0.05), suggesting that maternal exposure of calves to elevated levels of PBDEs may have contributed to the high annual stranding death rates of calves in the last two decades.

Killer whales ( Orcinus orca ) have been deemed one of the most contaminated cetacean species in the world. However, concentrations and potential health implications of selected 'contaminants of emerging concern' (CECs) and new persistent organic pollutants (POPs) in endangered Southern Resident and threatened Bigg's (Transient) killer whales in the Northeastern Pacific (NEP) have not yet been documented. Here, we quantify CECs [alkylphenols (APs), triclosan, methyl triclosan, and per- and polyfluoroalkyl substances (PFAS)] and new POPs [hexabromocyclododecane (HBCCD), PFOS, PFOA, and PFHxS] in skeletal muscle and liver samples of these sentinel species and investigate in utero transfer of these contaminants. Samples were collected from necropsied individuals from 2006 to 2018 and analyzed by LC-MS/MS or HRBC/HRMS. AP and PFAS contaminants were the most prevalent compounds; 4-nonylphenol (4NP) was the predominant AP (median 40.84 ng/g ww), and interestingly, 7:3-fluorotelomer carboxylic acid (7:3 FTCA) was the primary PFAS (median 66.35 ng/g ww). Maternal transfer ratios indicated 4NP as the most transferred contaminant from the dam to the fetus, with maternal transfer rates as high as 95.1%. Although too few killer whales have been screened for CECs and new POPs to infer the magnitude of contamination impact, these results raise concerns regarding pathological implications and potential impacts on fetal development and production of a viable neonate. This study outlines CEC and new POP concentrations in killer whales of the NEP and provides scientifically derived evidence to support and inform regulation to mitigate pollutant sources and contamination of Southern Resident killer whale critical habitat and other marine ecosystems.

Decabromodiphenyl ethane (DBDPE), a replacement for decabromodiphenyl ether (deca-BDE), was investigated in captive Chinese alligators from China. DBDPE was detected in adult tissues, neonates and eggs of Chinese alligators with concentrations ranging from 4.74-192, 0.24-1.94, and 0.01-0.51 ng g lipid weight, respectively. Compared to PBDEs and PCBs, DBDPE contamination was limited in Chinese alligators. Additionally, DBDPE concentrations in adult muscles were one to three orders of magnitude higher than those in neonates and eggs, suggesting the limited maternal transfer potential of DBDPE in Chinese alligators. A breeding population of the common toad Bufo bufo living in the vicinity of a Zn-Pb smelting works in Bukowno, Poland was studied for the presence of thallium. Tl concentration was measured in the bottom sediments of the spawning pond, in the laid eggs, in juveniles after metamorphosis, and in the selected tissues of the adult individuals. A very high concentration of Tl was detected in the spawn (13.97±8.90 mg/kg d.w.). In 50 % of the spawn samples, levels exceeded 20 mgTl/kg d.w. The issue of maternal transfer of thallium from females to oocytes is discussed. Due to a significant accumulation of thallium, spawn analysis can be used as a sensitive indicator of the presence of this element in the environment and may replace more invasive methods that involve the killing of adult animals. In those regions that are abundant in Zn-Pb ores, the spawn of amphibians may be a very important source of thallium contamination for predators. From among all tissues of the Bukowno adult toads, the livers have shown the highest accumulation of thallium (mean 3.98 mg/kg d.w. and maximum value—18.63). Problem In twin pregnancies, factors that influence total umbilical cord IgG concentration and IgG transfer ratio are not well known. Method Blood samples were prospectively collected from 57 twin pregnancies. Stepwise multivariate regression analysis was used to evaluate the association between total IgG levels in the umbilical cord blood and IgG transfer ratio according to serum IgG concentration, pregnancy chorionicity, the presence of abnormal umbilical artery pulsatility index, intrauterine growth restriction, gestational age at delivery ( GAD), birthweight, and placental weight. Results Umbilical cord IgG concentration showed a positive correlation with serum IgG concentration and GAD; levels were significantly lower in monochorionic compared with dichorionic pregnancies. IgG transfer ratio also increased with GAD but was inversely correlated with serum IgG concentration levels. Conclusion In twin pregnancies, besides serum IgG concentration and GAD, chorionicity also influences umbilical cord IgG concentration. Pacific herring embryos spawned in nearshore habitats may be exposed to toxic contaminants as they develop, from exogenous sources in spawning habitats and from maternal transfer. Determining baseline concentrations of these toxic contaminants is important for evaluating the health of this species, especially during this sensitive life stage. In this study we compared concentrations of polycyclic aromatic hydrocarbons, or PAHs, in naturally spawned herring embryos from five spawning areas across Puget Sound. The summed values of 31 PAH analytes (Σ 31 PAH) in early- to late-stage development embryos ranged from 1.1 to 140 ng/g, wet weight. Σ 31 PAH concentrations increased with development time in embryos from one spawning area where the greatest concentrations were observed, and the relative abundance of PAH chemicals in late-stage embryos was similar to those in nearby sediments, suggesting accumulation from local environmental sources. PAHs in both sediments and late-stage embryos appeared to exhibit a pyrogenic pattern. Although maternal transfer of PAHs appeared to be a negligible source to embryos in spawning areas with the greatest embryo PAH concentrations, maternal transfer may have been the dominant source in embryos from spawning areas where the lowest levels of embryo-PAHs occurred. Chronic embryo mortality has been reported in spawning habitats where we observed the greatest concentration of PAHs in embryos, and necrotic tissue in herring embryos from one such location was similar in description to phototoxic PAH necrosis reported elsewhere for embryonic zebrafish. We studied maternal transfer of an essential metal (copper) and a non-essential one (cadmium) in the live-bearing fishes Heterandria formosa and Gambusia affinis. The goals of this study were: (1) to determine whether metals are transferred from exposed females to their developing offspring; (2) to determine if this transfer differs between two fish species that differ in their degree of maternal provisioning during development; (3) to determine the duration of maternal metal transfer once females are no longer exposed; and (4) to determine whether copper and cadmium are transferred equivalently. We exposed gravid females to background levels (control) or 0.15 µM of metal for 10 days, and then transferred them to clean water. We allowed females to give birth to up to three broods, and then quantified metal levels in offspring born at least 3 days after the transfer. We detected maternal metal transfer for both metals and in both species. Offspring metal levels decreased as females spent more time in clean water. Similarly, metal levels were lower in later broods than in earlier ones. Maternal metal transfer was higher in H. formosa than in G. affinis. Our results constitute the first report of maternal metal transfer in live-bearing fishes, and show that developing embryos acquire both essential and non-essential metals from their mothers in both species. The distribution of perfluoroalkyl substances (PFASs) and the isomers of perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) was investigated among various tissues (including muscle, gill, kidney, liver) and eggs, in aquatic organisms in Taihu Lake, China. Highest concentration of ΣPFASs was mostly found in liver (278–685 ng/g ww) and eggs (66.0–467 ng/g ww) while the lowest was in muscle (40.6–165 ng/g ww). n -PFOS was the predominant PFOS isomer in most of the tissues with a proportion of 46.3–96.5%. Ratios of PFAS concentrations in eggs to those in liver (E/L) increased positively with the protein-water partition coefficient. The E/L of PFOS isomers descended in the order: linear > monomethyl > diperfluoromethyl isomers. The liver/muscle and kidney/muscle ratios of n -PFOS were higher than branched isomers, suggesting that n -PFOS has higher binding affinity with hepatic proteins or branched isomers are preferentially excreted though liver and kidney. Immunoglobulins (Igs), which bind antigens with high specificity, are essential molecules in adaptive immune system of jawed vertebrates. In this study, cDNA encoding the secreted form of the immunoglobulin heavy chain of IgM (sIgM) was cloned from the mesonephros of blunt snout bream ( Megalabrama amblycephala ) using RT-PCR and rapid amplification of cDNA ends (RACE). The full-length cDNA of sIgM heavy chain gene has 1961 nucleotides encoding a putative protein of 569 amino acids, constant region shares high amino acid identity with that of Ctenopharyngodon idella (80%), Carassius auratus langsdorfii (65%) and Danio rerio (59%). Multiple protein sequence alignment revealed that blunt snout bream sIgM was clustered with the homologues of cyprinid fish and constructed one clade. Using quantitative real-time PCR (qRT-PCR) analysis, the level of sIgM mRNA was determined, with a V-shape change pattern: decreased initially from unfertilized egg stage to 4 cells stage and increased from 16 cells stage to prelarva. This sharp drop indicates that sIgM mRNA is maternally transferred, and was continuously degraded until 16 cells stage. The drastic rising in sIgM level from blastula stage to prelarva might be attributed to embryonic stem cell differentiation procedure. Compared with juvenile fish, the expression of sIgM was significantly higher in pronephros, liver, spleen, gill and muscle of adult fish. After the injection of Aeromonas hydrophila , the expression pattern of sIgM was found first down-regulated at 4 h, then up-regulated and reached the peak at 7 d and 21 d in mesonephros, spleen, liver and gill, respectively. Hen muscle, eggs, and newborn chick tissues (muscle and liver) were collected from an electronic waste recycling site in southern China. The authors examined the maternal transfer, potential metabolism, and tissue distribution of several halogenated flame retardants (HFRs) during egg formation and chicken embryo development. The pollutant composition changes significantly from hen muscle to eggs and from eggs to tissues of newborn chicks. Higher-halogenated chemicals, such as octa- to deca-polybrominated diphenyl ether (PBDE) congeners, deca-polybrominated biphenyl (PBB209), and dechlorane plus (DP), are less readily transferred to eggs compared with lower-halogenated chemicals. During embryo development, PBDEs are the most likely to be metabolized, whereas decabromodiphenyl ethane (DBDPE) is the least. The authors also observed selective maternal transfer of anti-DP and stereoselective metabolism of syn-DP during chicken embryo development. During tissue development, liver has greater affinity than the muscle for chemcials with a high log octanol-water partition coefficient, with the exception of DBDPE. The differences in metabolism potential of different chemicals in chicken embryos cause pollutant composition alterations. Halogenated flame retardant from maternal transfer and tissue distribution also exhibited chemical specificity, especially for DBDPE. Levels of DBDPE were elevated along with the full process from hen muscle to eggs and from eggs to chick tissues. Abstract: Advances have been made recently in assessing accumulation and effects of coal combustion residues (CCR). I provide a brief review of recent advancements, provide a tabulated summary of results of recent work, and put forth recommendations for future studies. One advancement is that mercury accumulation has begun to receive (limited) attention, whereas it had rarely been considered in the past. Additionally, some constituents of CCR have been shown to be accumulated by adults and transferred to offspring, sometimes compromising offspring health. Studies have demonstrated that amphibians, possessing complex life cycles, may accumulate and transfer some contaminants to terrestrial systems. Some study has been given to molecular and cellular effects of CCR exposure, although these studies have been limited to invertebrates. Population models have also been applied to CCR affected systems and have shown that CCR may affect animal populations under some conditions. In light of these advancements, there are several topics that require further assessment. First, more attention to Hg and its dynamics in CCR affected systems is warranted. Hg can be highly accumulative and toxic under some conditions and may interact with other components of CCR (notably Se), perhaps altering accumulation and effects of the contaminant mixtures. Second, further investigation of maternal transfer and effects of CCR contaminants need to be conducted. These studies could benefit from incorporation of quantitative models to project impacts on populations. Finally, more attention to the organic constituents of CCR (PAHs) is required, as a focus on inorganic compounds only may restrict our knowledge of contaminant dynamics and effects as a whole. While further studies will shed light on some chemical and biological nuances of exposure and effect, information available to date from numerous study sites implicates CCR as a bulk effluent that presents risks of bioaccumulation and effects on organisms in aquatic systems.
