Abstract

Introduction: Viral infections and reactivations still remain a cause of morbidity and mortality after hematopoietic stem cell transplantation due to immunodeficiency and immunosuppression. Transfer of unmanipulated donor-derived lymphocytes (DLI) represents a promising strategy for improving cellular immunity but carries the risk of graft versus host disease (GvHD). Depleting alloreactive naïve T cells (TN) from DLIs was implemented to reduce the risk of GvHD induction while preserving antiviral memory T-cell activity. Here, we compared two TN depletion strategies via CD45RA and CD62L expression and investigated the presence of antiviral memory T cells against human adenovirus (AdV) and Epstein-Barr virus (EBV) in the depleted fractions in relation to their functional and immunophenotypic characteristics. Methods: T-cell responses against ppEBV_EBNA1, ppEBV_Consensus and ppAdV_Hexon within TN-depleted (CD45RA−/CD62L−) and TN-enriched (CD45RA+/CD62L+) fractions were quantified by interferon-gamma (IFN-γ) ELISpot assay after short- and long-term in vitro stimulation. T-cell frequencies and immunophenotypic composition were assessed in all fractions by flow cytometry. Moreover, alloimmune T-cell responses were evaluated by mixed lymphocyte reaction. Results: According to differences in the phenotype composition, antigen-specific T-cell responses in CD45RA− fraction were up to 2 times higher than those in the CD62L− fraction, with the highest increase (up to 4-fold) observed after 7 days for ppEBV_EBNA1-specific T cells. The CD4+ effector memory T cells (TEM) were mainly responsible for EBV_EBNA1- and AdV_Hexon-specific T-cell responses, whereas the main functionally active T cells against ppEBV_Consensus were CD8+ central memory T cells (TCM) and TEM. Moreover, comparison of both depletion strategies indicated that alloreactivity in CD45RA− was lower than that in CD62L− fraction. Conclusion: Taken together, our results indicate that CD45RA depletion is a more suitable strategy for generating TN-depleted products consisting of memory T cells against ppEBV_EBNA1 and ppAdV_Hexon than CD62L in terms of depletion effectiveness, T-cell functionality and alloreactivity. To maximally exploit the beneficial effects mediated by antiviral memory T cells in TN-depleted products, depletion methods should be selected individually according to phenotype composition and CD4/CD8 antigen restriction. TN-depleted DLIs may improve the clinical outcome in terms of infections, GvHD, and disease relapse if selection of pathogen-specific donor T cells is not available. [ABSTRACT FROM AUTHOR]

Abstract

Arbuscular mycorrhizal fungi (AMF) establish a symbiotic association with the roots of 80% of terrestrial plants and form complex tree-shaped feeding structures called arbuscules in colonized root cells. This association with AMF not only provides more efficient uptake of nutrients for the plant, but also confers protection against pathogens and increased tolerance to environmental stress such as salt stress [1]. Rice (Oryza sativa), the most salt-sensitive crop species amongst cereals, has a productivity strongly reduced around the world due to soil salinity/salinization and increased sea level (in deltas). High Na+ concentrations (salt stress conditions) impairs K+ uptake and inhibits many K+-activated enzymes. Rice exhibits molecular mechanisms to alleviate salt stress such as maintaining a high cellular K+/Na+ ratio, e.g. by a more efficient K+ uptake, which was recently shown to occur upon root/AMF interaction [2]. Knowledge on the role of root/AMF interaction on K+/Na+ transport upon salt stress is sparse [2, 3]. The aim of the research is to understand the mechanisms by which AMF improve plant K+ nutrition upon salt stress, taking Rhizophagus irregularis (model AMF)-rice interaction as a model. We will investigate the mechanisms by which AMF mediate K+ uptake and translocation towards root cells. Rice loss-of-function mutants for each of the three major K+ uptake systems of the plant, OsAKT1, OsHAK5 and OsHAK1, were inoculated with R. irregularis and submitted to salt stress, and their K+ contents and K+/Na+ ratios were monitored to identify key player(s) for AMF-mediated improved rice K+ nutrition and salt tolerance.

Abstract

Potassium (K+) acquisition, translocation and cellular homeostasis are mediated by various membrane transport systems in all organisms. We identified and described an ion channel in the ectomycorrhizal fungus Hebeloma cylindrosporum (HcSKC) that harbors features of animal voltage-dependent Shaker-like K+ channels, and investigated its role in both free-living hyphae and symbiotic conditions. RNAi lines affected in the expression of HcSKC were produced and used for in vitro mycorrhizal assays with the maritime pine as host plant, under standard or low K+ conditions. The adaptation of H. cylindrosporum to the downregulation of HcSKC was analyzed by qRT-PCR analyses for other K+-related transport proteins: the transporters HcTrk1, HcTrk2, and HcHAK, and the ion channels HcTOK1, HcTOK2.1, and HcTOK2.2. Downregulated HcSKC transformants displayed greater K+ contents at standard K+ only. In such conditions, plants inoculated with these transgenic lines were impaired in K+ nutrition. Taken together, these results support the hypothesis that the reduced expression of HcSKC modifies the pool of fungal K+ available for the plant and/or affects its symbiotic transfer to the roots. Our study reveals that the maintenance of K+ transport in H. cylindrosporum, through the regulation of HcSKC expression, is required for the K+ nutrition of the host plant. [ABSTRACT FROM AUTHOR]

Abstract

Background and objective: Fixed airflow obstruction (FAO) in asthma occurs despite optimal inhaled treatment and no smoking history, and remains a significant problem, particularly with increasing age and duration of asthma. Increased lung compliance and loss of lung elastic recoil has been observed in older people with asthma, but their link to FAO has not been established. We determined the relationship between abnormal lung elasticity and airflow obstruction in asthma. Methods: Non‐smoking asthmatic subjects aged >40 years, treated with 2 months of high‐dose inhaled corticosteroid/long‐acting beta‐agonist (ICS/LABA), had FAO measured by spirometry, and respiratory system resistance at 5 Hz (Rrs5) and respiratory system reactance at 5 Hz (Xrs5) measured by forced oscillation technique. Lung compliance (K) and elastic recoil (B/A) were calculated from pressure–volume curves measured by an oesophageal balloon. Linear correlations between K and B/A, and forced expiratory volume in 1 s/forced vital capacity (FEV1/FVC), Rrs5 and Xrs5 were assessed. Results: Eighteen subjects (11 males; mean ± SD age: 64 ± 8 years, asthma duration: 39 ± 22 years) had moderate FAO measured by spirometry ((mean ± SD z‐score) post‐bronchodilator FEV1: −2.2 ± 0.5, FVC: −0.7 ± 1.0, FEV1/FVC: −2.6 ± 0.7) and by increased Rrs5 (median (IQR) z‐score) 2.7 (1.9 to 3.2) and decreased Xrs5: −4.1(−2.4 to −7.3). Lung compliance (K) was increased in 9 of 18 subjects and lung elastic recoil (B/A) reduced in 5 of 18 subjects. FEV1/FVC correlated negatively with K (rs = −0.60, P = 0.008) and Rrs5 correlated negatively with B/A (rs = −0.52, P = 0.026), independent of age. Xrs5 did not correlate with lung elasticity indices. Conclusion: Increased lung compliance and loss of elastic recoil relate to airflow obstruction in older non‐smoking asthmatic subjects, independent of ageing. Abstract

Adenovirus (HAdV) infections confer a high risk of morbidity and mortality for immunocompromised patients after stem cell transplantation (SCT). Treatment with standard antiviral drugs is of limited efficacy and associated with a high rate of adverse effects. HAdV‐specific T cells are crucial for sustained viral elimination and the efficacy of adoptive T‐cell therapy with donor‐derived HAdV‐specific T cells has been reported by several investigators. Here, we report our experience with the transfer of HAdV‐specific T cells specific for penton, which was recently identified as an immunodominant target of T cells, and hexon in a 14‐year‐old boy after T‐cell‐depleted haploidentical SCT for myelodysplastic syndrome (MDS). He developed severe HAdV‐associated enteritis complicated by acute graft‐versus‐host disease (GvHD). The patient received ten infusions of allogeneic HAdV‐specific T cells manufactured from the haploidentical stem cell donor using the CliniMacs Interferon‐γ (IFN‐γ) cytokine capture and immunomagnetic selection. Initially, T cells were generated against the immunodominant target hexon and in subsequent transfers dual antigen‐specific T cells against hexon and penton were applied. T‐cell transfers were scheduled individually tailored to current immunosuppressive treatment. Each transfer was followed by reduction of HAdV load in peripheral blood and clinical improvement. Importantly, T‐cell responses to both penton and hexon pools emerged in patient blood after repetitive transfers. Unfortunately, the patient experienced bacterial sepsis, and in this context, severe GvHD requiring intensive immunosuppression followed by secondary progression of HAdV infection. The patient succumbed to multiorgan failure 283 days after SCT. This case demonstrates the feasibility of HAdV‐specific T‐cell transfer even in the presence of immunosuppressive treatment. Targeting of multiple immunodominant viral proteins may prove valuable in patients with complicated HAdV infections. [ABSTRACT FROM AUTHOR]

Abstract

Grapevine (Vitis vinifera L.), one of the most important fruit crops, is a model plant for studying the physiology of fleshy fruits. Here, we report on the characterization of a new grapevine Shaker-type K+ channel, VvK5.1. Phylogenetic analysis revealed that VvK5.1 belongs to the SKOR-like subfamily. Our functional characterization of VvK5.1 in Xenopus oocytes confirms that it is an outwardly rectifying K+ channel that displays strict K+ selectivity. Gene expression level analyses by real-time quantitative PCR showed that VvK5.1 expression was detected in berries, roots, and flowers. In contrast to its Arabidopsis thaliana counterpart that is involved in K+ secretion in the root pericycle, allowing root to shoot K+ translocation, VvK5.1 expression territory is greatly enlarged. Using in situ hybridization we showed that VvK5.1 is expressed in the phloem and perivascular cells of berries and in flower pistil. In the root, in addition to being expressed in the root pericycle like AtSKOR, a strong expression of VvK5.1 is detected in small cells facing the xylem that are involved in lateral root formation. This fine and selective expression pattern of VvK5.1 at the early stage of lateral root primordia supports a role for outward channels to switch on cell division initiation. [ABSTRACT FROM AUTHOR]

»Ins Freie« entwirft Grundzüge einer philosophischen Ästhetik vor dem Hintergrund künstlerischer Praxis. Brennpunkt ist dabei ein Vergleich von künstlerischen und philosophischen Tätigkeiten, die als »Praktiken des Öffnens« beschrieben werden. Der Redesinn dieser Formulierung speist sich aus zwei Quellen. Zum einen spricht Theodor W. Adorno in seiner Negativen Dialektik vom Ziel, dem »Telos der Philosophie« als »das Offene und Ungedeckte«. In einer solchen Ausrichtung entzieht Denken sich unaufhörlich einem abschließenden Urteil und hinterfragt beständig die bestehenden Verhältnisse. Zum anderen wurde die Formulierung »Öffnen« von Untersuchungen inspiriert, die im Kontext von Performance-Kunst unternommen werden. Dort unterbrechen Experimentelle Live-Interventionen Handlungsmuster und Wahrnehmungserwartungen und produzieren Unvorhersehbarkeiten im Kommunikationsverlauf mit einem Publikum. Die Erfahrung solcher Impulse kann dazu motivieren, bestehende Identifikationen plötzlich in Zweifel zu ziehen. Sie kann bestehende Wahrnehmungs- und Denk-Gewohnheiten für eine andere Selbst- und Weltwahrnehmung öffnen. Woran kann man einen Öffnungs-vorgang erkennen? Im Kontext dieser Frage formuliert sich die Ausgangslage und der Grundgedanke der vorliegenden Arbeit: Zeitgenössische Kunst bildet nicht einfach ab, sondern hat sich in gewisser Weise darauf spezialisiert, gewohnte Wahrnehmungs- und Denkmuster zu unterbrechen. Der heutige Künstler scheint ein »Spezialist für Ent-selbstverständlichung« zu sein. In dieser Perspektivierung stellt sich »Ins Freie« der Herausforderung, die künstlerische Reflexion von verlaufsoffenen Prozessen beschreibbar zu machen. Die zugrunde liegenden systematischen Fragen lauten • Sind Philosophie und Kunst in ihren Tätigkeiten vergleichbar? • In welchen Medien bewegen sie sich? • Was heißt Reflexion in dem einen wie dem anderen Fall? • Gibt es in der Philosophie etwas einem »verlaufsoffenen Prozess« vergleichbares? • Welche Positionen von »Künstler und Philosoph« bieten sich im Hinblick auf einen Dialog an und warum? • Ist es überhaupt möglich, verlaufsoffene Prozesse im Zusammenhang künstlerischer und philosophischer Tätigkeiten zu beschreiben und welche Aspekte können dabei als Koordinaten dienen? Ausgehend von der anfänglichen Beobachtung, dass Kunst im musealen Kontext von einer zunehmenden Flut von kommentierenden Texten begleitet wird, entwickelt die vorliegende Arbeit einen philosophischen Vergleich von Werken des amerikanischen Künstlers Richard Serra mit Werken des französischen Philosophen Maurice Merleau-Ponty und exemplifiziert dabei die Annahme, dass man künstlerisch reflektieren kann, ohne einem Kunstwerk eine Aussage zu unterstellen.

Abstract

Ectomycorrhizal (ECM) symbiosis is defined as a mutual association between fungal symbionts and roots of woody plants. This symbiosis is strongly represented in temperate and boreal forests and is considered to be the most widespread means of increasing P acquisition by trees. Despite the importance of the ECM association, current knowledge about molecular mechanisms involved in membrane transport between the two partners is still limited. Therefore, the identification of the mechanisms essential for the symbiosis is a major issue. To better understand these mechanisms, we used a model association between the ECM basidiomycete Hebeloma cylindrosporum and the maritime pine Pinus pinaster. The fungal genome contains three transporters of Pi (HcPT1.1, HcPT1.2, and HcPT2), which have been already characterized as H+ Pi carriers. We hypothesized that one of these carriers could be responsible for both influx and efflux of Pi from the soil to the fungus and from the fungus to the plant. Our results suggest that HcPT2 would be the best candidate to fill both roles. An in vitro ectomycorrhizal system was used to investigate phosphate flux from H. cylindrosporum toward a liquid compartment containing pine roots. Interestingly, the expression pattern of HcPT2 was correlated to fungal phosphate efflux. Moreover, immuno-localization experiments showed that the protein is located (i) in the extraradical hyphae and the fungal sheath, compatible with Pi uptake, and (ii) within the Hartig net, which is the place of exchange between fungal and host cells. In order to study more specifically the role of HcPT2 at the fungus-plant interface, overexpressing and knock-down transgenic H. cylindrosporum strains were produced. Fungal strains with up regulated expression of HcPT2 increased phosphate efflux within the interaction medium, supporting a role of HcPT2 in phosphate transfer to the plant. In addition, the modification of HcPT2 expression strongly affects Pi nutrition of P. pinaster.

Abstract

Extracellular proteins play crucial roles in the interaction between mycorrhizal fungi and their environment. Computational prediction and experimental detection allowed identification of 869 proteins constituting the exoproteome of Hebeloma cylindrosporum. Small secreted proteins (SSPs) and carbohydrate-active enzymes (CAZymes) were the two major classes of extracellular proteins. Twenty-eight per cent of the SSPs were secreted by free-living mycelia and five of the 10 most abundant extracellular proteins were SSPs. By contrast, 63-75% of enzymes involved in nutrient acquisition were secreted. A total of 150 extracellular protein-coding genes were differentially expressed between mycorrhizas and free-living mycelia. SSPs were the most affected. External environmental conditions also affected expression of 199 exoproteome genes in mycorrhizas. SSPs displayed different patterns of regulation in response to presence of a host plant or other environmental signals. Several of the genes most overexpressed in the presence of organic matter encoded oxidoreductases. Hebeloma cylindrosporum has not fully lost its ancestral saprotrophic capacities but rather adapted them not to harm its hosts and to use soil organic nitrogen. The complex and divergent patterns of regulation of SSPs in response to a symbiotic partner and/or organic matter suggest various roles in the biology of mycorrhizal fungi.

In forest ecosystems, the availability of phosphorus as inorganic P (Pi) limits tree growth. To overcome this deficiency, trees are associated with fungi through new organs called ectomycorrhizae (ECM). Despite the importance of phosphate nutrition, the mechanisms involved in the unidirectional transfer of P between the fungus and the plant are still unknown. Therefore, the identification of these mechanisms essential for symbiosis is a major issue. To understand better these mechanisms, we used a model association between the ECM basidiomycete Hebeloma cylindrosporum and the maritime Pine, Pinus pinaster. Immunolocalization showed that the protein is located in (i) the extraradical hyphae and the fungal sheath involved in Pi uptake and (ii) the Hartig net, ie the interface between fungal and host cells. We also produced overexpressing and knock-down transgenic H. cylindrosporum strains to study specifically the role of HcPT2 in (i) the P efflux from the fungus measured in vitro and (ii) the P nutrition of the host plant grown at low and high P levels. Finally, if our hypothesis is confirmed, the main question will be to define how this transporter is regulated to provide both influx and efflux of phosphate in ECM symbiosis.

We therefore investigated the impact of the ectomycorrhizal fungus Hebeloma cylindrosporum on the potassium nutrition of Pinus pinaster and examined the involvement of the fungal potassium transporter HcTrk1. HcTrk1 transcripts and proteins were localized in ectomycorrhizas using in situ hybridization and EGFP translational fusion constructs. Importantly, an overexpression strategy was performed on a H. cylindrosporum endogenous gene in order to dissect the role of this transporter. The potassium nutrition of mycorrhizal pine plants was significantly improved under potassium-limiting conditions. Fungal strains overexpressing HcTrk1 reduced the translocation of potassium and phosphorus from the roots to the shoots of inoculated plants in mycorrhizal experiments. Furthermore, expression of HcTrk1 and the phosphate transporter HcPT1.1 were reciprocally linked to the external inorganic phosphate and potassium availability. The development of these approaches provides a deeper insight into the role of ectomycorrhizal symbiosis on host plant K(+) nutrition and in particular, the K(+) transporter HcTrk1. The work augments our knowledge of the link between potassium and phosphorus nutrition via the mycorrhizal pathway.

International audience; Potassium (K+) is one of the most abundant element of soil composition but its very low availability limits plant growth and productivity of ecosystems. Because this cation participates in many biological processes, its constitutive uptake from soil solution is crucial for the plant cell machinery. Thus, the understanding of strategies responsible of K+ nutrition is a major issue in plant science. Mycorrhizal associations occurring between roots and hyphae of underground fungi improve hydro-mineral nutrition of the majority of terrestrial plants. The contribution of this mutualistic symbiosis to the enhancement of plant K+ nutrition is still underestimated and poorly studied so far. This mini-review examines the current knowledge about the impact of both arbuscular mycorrhizal and ectomycorrhizal symbioses on the transfer of K+ from the soil to the plants. A model summarizing plant and fungal transport systems identified and hypothetically involved in K+ transport is proposed. In addition, some data related to benefits for plants provided by the improvement of K+ nutrition thanks to mycorrhizal symbioses are presented.
