Abstract

In high dam construction projects in China, stilling basin design with an abrupt bottom drop is sometimes introduced to reduce the bottom velocity and pressure loads by generating A-type hydraulic jumps. Although the stilling basin design is not new, A-type hydraulic jumps have not been studied taking into account the air entrainment and evolution of internal air–water flow structures. This paper presents an experimental study of self-aerated A-type jumps in terms of bubble transport and free-surface fluctuations over the bottom drop. Four Froude numbers from 4.1 to 10.3 are tested for three drop heights, in addition to the flat-bottom case. Compared to the classic hydraulic jumps, A-jumps are observed with longer jump lengths and weaker free-surface fluctuations. The downward deflection of the jet-shear flow and formation of a bottom roller in the step cavity require a modification to the analytical expression of velocity and void fraction distributions. The relationship between the bubble diffusivity and jump spreading rate differs from that in classic hydraulic jumps, suggesting a faster expansion of the bubble diffusion layer than the turbulent shear flow downstream of the drop, especially for large drop heights. At large approach velocities, the reattachment of the deflected jet-shear flow to the lowered bed may cause a local rise in bubble counts downstream the bottom roller. Abstract

Beyond its potential for biofuel production, Pseudomonas putida’s capability to metabolize lignin and other lignocellulosic materials earmarks it as a pivotal candidate for engineering to yield diverse value-added chemicals, thereby challenging traditional petrochemical approaches. Recognizing the inherent environmental, economic, and societal advantages, amplifying role of P. putidain industrial applications becomes imperative. In this context, our study focused on characterizing a comprehensive set of promoters and ribosome binding site tailored for P. putida, spanning a broad spectrum of activities. By leveraging these genetic tools, we adeptly balanced the heterologous mevalonate (MVA) pathway flux within P. putida. As a culmination of our efforts, the optimal MVA-producing strains were identified, achieving a remarkable yield of 5 g/LMVA in a 5 L fed-batch fermenter, marking the highest reported yield in Pseudomonasto date. This research not only provides valuable genetic tools for future engineering studies with P. putida, but also accentuates P. putida’s potential in synthetic biology and its promise for sustainable chemical production.

Abstract

Prion diseases are a group of neurodegenerative diseases characterized by mitochondrial dysfunction and neuronal death. Mitophagy is a selective form of macroautophagy that clears injured mitochondria. Prohibitin 2 (PHB2) has been identified as a novel inner membrane mitophagy receptor that mediates mitophagy. However, the role of PHB2 in prion diseases remains unclear. In this study, we isolated primary cortical neurons from rats and used the neurotoxic prion peptide PrP106–126 as a cell model for prion diseases. We examined the role of PHB2 in PrP106–126-induced mitophagy using Western blotting and immunofluorescence microscopy and assessed the function of PHB2 in PrP106–126-induced neuronal death using the cell viability assay and the TUNEL assay. The results showed that PrP106–126 induced mitochondrial morphological abnormalities and mitophagy in primary cortical neurons. PHB2 was found to be indispensable for PrP106–126-induced mitophagy and was involved in the accumulation of PINK1 and recruitment of Parkin to mitochondria in primary neurons. Additionally, PHB2 depletion exacerbated neuronal cell death induced by PrP106–126, whereas the overexpression of PHB2 alleviated PrP106–126 neuronal toxicity. Taken together, this study demonstrated that PHB2 is indispensable for PINK1/Parkin-mediated mitophagy in PrP106–126-treated neurons and protects neurons against the neurotoxicity of the prion peptide. Abstract

Rubbers are extensively applied in chemical protective clothing (CPC) due to their eye-catching anti-penetration of chemicals. However, their impermeability, particularly that of natural rubber (NR), is unsatisfactory. In this work, we demonstrate the facile construction of Ti3C2Tx MXene/NR interface using a plant-scale and feasible method combining latex mixing, emulsion flocculation, and flat-plate vulcanisation. The above crafts achieved a homogeneous dispersion of Ti3C2Tx MXene in the NR matrix in a single layer, thereby constructing a strong interfacial interaction between Ti3C2Tx MXene and NR, which induced the formation of a robust three-dimensional (3D) network in the composite. The anti-swelling capacity of the 3D cross-linked network structure and the layered structure of Ti3C2Tx MXene effectively prolonged the permeation path of toxic chemicals. Compared with pure NR, the nanocomposite with 1 wt% of Ti3C2Tx MXene showed substantially enhanced breakthrough times of toluene, dichloromethane, and concentrated sulfuric acid (increased by 140%, 178.6%, and 92.5%, respectively). Furthermore, its tensile strength, elongation at break, and shore hardness increased by 7.847 MPa, 194%, and 12 HA, respectively. Taken together with the satisfactory anti-permeability, tensile strength, elongation at break, and shore hardness, the resulting Ti3C2Tx MXene/NR nanocomposites hold promise for application to long-term and high-strength CPC in the chemical industry and military fields. Abstract

The majority of water inrush accidents in coal mines are caused by mining engineering activities. To avoid water inrush accidents, the Time-lapse Electrical Resistivity Tomography (TLERT) is applied to monitor water migration in fractured zone. A great challenge for the application of TLERT monitoring is the huge and numerous time series data sets generated by monitoring systems, which are difficult to process manually. This research proposed a distributed fuzzy clustering algorithm based on kernel function estimation to analyze TLERT images automatically. The resistivity date can be classified with different cluster centroids. The fuzzy c-means algorithm was chosen to display resistivity change. The algorithm was validated using a floor water inrush model. The results indicate that the water migration in the fractured zone can be monitored automatically and the edge of the resistivity changing area can be shown clearly.
