Patients with asthma–COPD overlap (ACO) are at risk of rapid progression of disease and severe exacerbations. However, in some patients with ACO, a clear distinction from COPD is very difficult by using physiological testing techniques. This study aimed to apply a novel metabolomic approach to identify the metabolites in sera in order to distinguish ACO from COPD.Methods: In the study, blood samples were collected from patients with COPD, ACO, and healthy controls. Cholamine derivatization-ultrahigh performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF/MS) was used to investigate serum metabolites of eicosanoids.Results: A clear intergroup separation existed between the patients with ACO and those with COPD, while ACO tends to have higher serum metabolic levels of eicosanoids. A robust Orthogonal Projections to Latent Structures-Discriminant Analysis (OPLS-DA) model was found for discriminating between ACO and COPD (R2Y =0.81, Q2=0.79). In addition, there is a significant correlation between some metabolites and clinical indicators, such as hydroxyeicosatetraenoic acids (HETEs), hydroperoxyeicosatetraenoic acids (HPETEs) and FEV1/FVC. The higher values of area under the receiver operating characteristic curves (ROC) of HETEs, which were metabolized from HPETEs through lipoxygenase (LOX), indicated that they should be the potential biomarkers to distinguish ACO from COPD.Conclusion: Eicosanoids can clearly discriminate different biochemical metabolic profiles between ACO and COPD. Abstract

Rheumatoid arthritis (RA) is a chronic inflammatory condition characterized by articular synovitis that eventually leads to the destruction of cartilage and bone in the joints with resulting pain and disability. The current therapies for RA are divided into 4 categories: non‐steroidal anti‐inflammatory drugs (NSAIDs), glucocorticoids, nonbiological disease‐modifying anti‐rheumatic drugs (DMARDs), and biological DMARDs. Each drug grouping is beset with significant setbacks that not only include limited drug bioavailability and high clearance, but also varying degrees of drug toxicity to normal tissues. Recently, nanotechnology has provided a promising tool for the development of novel therapeutic and diagnostic systems in the area of malignant and inflammatory diseases. Among these, magnetic nanoparticles (MNPs) have provided an attractive carrier option for delivery of therapeutic agents. Armed with an extra magnetic probe, MNPs are capable of more accurately targeting the local lesion with avoidance of unpleasant systemic side effects. This review aims to provide an introduction to the applications of magnetic nanoparticles in RA, focusing on the latest advances, challenges, and opportunities for future development. Abstract

Ig is a Y‐shaped protein produced by plasma cells and exerts multiple functions in humoral immunity. There are five groups of Igs including IgA, IgD, IgE, IgG, and IgM, which differ in their heavy chain class. The primary function of Igs includes the neutralization of extrinsic pathogens, agglutination of foreign cells for phagocytosis, precipitation of soluble antigens in serum, and complement fixation. The B cells activated by antigen(s) can differentiate into antibody‐producing cells that are called plasma cells and usually matured in the germinal center (GC). Follicular T helper (Tfh) cells crosstalk with antigen‐presenting cells and play a crucial role in the development of the GC. Moreover, Tfh cells regulate trafficking through the GC to allow formative interaction with GC B cells that ultimately results in affinity maturation, B‐cell memory, and Ig class switching. The B7 family is a series of number of structurally related membrane proteins that bind with a specific receptor to deliver costimulatory or co‐inhibitory signals that regulate the activation of T cells in GC. Abstract

Applying the finite element method, the function of finite element analysis software AnsysWorkbench is powerful. Analysis 30 groups of different groove curvature radius coefficient model in extreme condition, obtained in applying extreme working conditions, stress and deformation data. Analysis results show that the inner groove curvature coefficient is 0.525, the outer ring raceway curvature coefficient of bearing is 0.52 when the slewing bearing shows maximum carrying capacity. Calculation, results provide analysis basis for slewing bearing design.

Abstract

To the Editor, Group 2 innate lymphoid cells (ILC2s) produce the canonical type 2 cytokines interleukin (IL)-5 and IL-13 in response to epithelial cytokines IL-25, IL-33, and thymic stromal lymphopoietin (TSLP).[1] ILC2s have been to be involved in the pathogenesis of asthma, allergic rhinitis (AR) and atopic dermatitis (AD).[2] We have reported the higher ILC2 levels in patients with asthma and patients with AR.[[3]] Recently, the cysteinyl leukotriene (CysLT) D4 (LTD4) and LTE4 were reported to induce Th2 cytokine production from ILC2s.[[5]] LTE4 is the stable and dominant CysLT in the pathogenesis of allergic conditions. We found higher prevalence of ILC2s (Lin SP - sp CRTH2 SP + sp CD127 SP + sp cells, Figure A) and the higher expression of CysLT1R in ILC2s (Figure B-C B ) b from patients with AR compared to healthy controls. It suggests that the activation of ILC2s to LTC4, LTD4 but not LTE4 in AR is related to the increased expression of CysLT1R in AR, at least in our study. [Extracted from the article]
