Polynucleating oxygen donor ligands as precursors have been employed to construct three high-nuclearity heterometallic ethynide clusters. Compound 2 consists of a Cl@Ag12 ethynide cluster capped by two trinuclear organooxotin phosphonate clusters [(nBuSn)3(μ3-O)(tBuPO3)3(OMe)(OH)2]2-. Compound 3 consists of a Ag8 ethynide cluster fused with a nonanuclear oxo-bismuth phosphonate cluster [Bi9O6(tBuPO3)9(tBuPO3H)]4-, while compound 4 is composed of two Cl@Ag15 ethynide clusters bridged by a hexanuclear oxo/hydroxo-bismuth phosphonate cluster [Bi6O4(OH)4(tBuPO3)6(hfac)2(CF3COO)2]10-.

Specific groups in Asia, including the Chinese, are more susceptible to colorectal cancer (CRC). The best strategy for anticancer drug action is to induce cancer cell apoptosis and autophagy. Bufalin is a potent inducer of apoptosis in some human cancer cell lines, but bufalin has barely been evaluated in colorectal cancer cells as a potent autophagy inducing agent. The aim of this study was to investigate the roles and interactions of bufalin in autophagy and the effects of the drug on human colorectal cancer. Rationale: Sustained cardiac hypertrophy often leads to heart failure (HF). Understanding the regulation of cardiomyocyte growth is crucial for the treatment of adverse ventricular remodeling and HF. Cell division cycle 20 (CDC20) is an anaphase-promoting complex activator that is essential for cell division and tumorigenesis, but the role of CDC20 in cardiac hypertrophy is unknown. We aimed to test whether CDC20 participates in the regulation of pathological cardiac hypertrophy and investigate the underlying mechanism in vitro and in vivo. Methods: Male C57BL/6 mice were administered a recombinant adeno-associated virus serotype 9 (rAAV9) vector expressing CDC20 or a siRNA targeting CDC20 and their respective controls by tail intravenous injection. Results: Microarray analysis showed that CDC20 was significantly upregulated in the heart after angiotensin II infusion. Knockdown of CDC20 in cardiomyocytes and in the heart reduced cardiac hypertrophy upon agonist stimulation or transverse aortic constriction (TAC). Conversely, enforced expression of CDC20 in cardiomyocytes and in the heart aggravated the hypertrophic response. Furthermore, we found that CDC20 directly targeted LC3, a key regulator of autophagy, and promoted LC3 ubiquitination and degradation by the proteasome, which inhibited autophagy leading to hypertrophy. Moreover, knockdown of LC3 or inhibition of autophagy attenuated Ang II-induced cardiomyocyte hypertrophy after deletion of CDC20 in vitro. Conclusions: Our study reveals a novel cardiac hypertrophy regulatory mechanism that involves CDC20, LC3 and autophagy, and suggests that CDC20 could be a new therapeutic target for patients with hypertrophic heart diseases.

A series of giant metallofullerenes Y

We demonstrate that a finite-length (10,0) carbon nanotube (CNT) with two fullerene caps, namely D5(450)-C100, is an ideal prototype to study the mechanical responses of small CNTs upon endohedral metal doping. Encapsulation of a large La2C2 cluster inside D5(450)-C100 induces a 5% axial compression of the cage, as compared with the structure of La2@D5(450)-C100. Detailed crystallographic analyses reveal quantitively the flexibility of the [10]cyclacene-sidewall segment and the rigidity of the pentagon-dominating caps for the first time. The internal C2-unit acts as a molecular spring that attracts the surrounding cage carbon atoms through strong interactions with the two moving lanthanum ions. This is the first crystallographic observation of the axial compression of CNTs caused by the internal stress, which enhances our knowledge about the structural deformation of novel carbon allotropes at the atomic level.

We have made a systematic investigation of the regioselective reaction between triethylamine and C60 under thermal conditions, presenting X-ray crystallographic results of the pentane-fused adduct for the first time. In addition to finding the reaction conditions which gave the highest yield and the best regioselectivity, we captured with GC–MS the acetaldehyde and diethylamine intermediates that were involved in the reaction, thus clarifying the reaction mechanism.

Successful placentation depends on the proper invasion of extravillous trophoblast (EVT) cells into maternal tissues. Previous reports have demonstrated that FoxM1 is oncogenic and plays important roles in angiogenesis, invasion, and metastasis. However, little is known about the roles of FoxM1 in the invasion of EVT cells. EGF, as a growth factor (epidermal growth factor), has been studied extensively in reproduction. JAR cells are a reliable model for studying early invasive trophoblast regulation. We have observed the relationship between EGF and FoxM1 in JAR cells by using specific inhibitors for the intervention in and study of potential signal pathways. We have also tested the ability of JAR cells to be influenced by the expression of FoxM1. Our data indicate that EGF promotes FoxM1 expression through the ERK signal pathway. Over-FoxM1 expression upregulates the ability of JAR cells to migrate and invade and vice versa. Our investigation of FoxM1 should provide new insights into the molecular mechanisms of EVT invasion.

The chemical properties of carbide-cluster metallofullerenes (CCMFs) remain largely unexplored, although several new members of CCMFs have been discovered recently. Herein, we report the reaction between Sc2 C2 @C3v (8)-C82 , which is viewed as a prototypical CCMF because of its high abundance, and 3-triphenylmethyl-5-oxazolidinone (1) to afford the corresponding pyrrolidino derivative Sc2 C2 @C3v (8)-C82 (CH2 )2 NTrt (2; Trt=triphenylmethyl). Single-crystal X-ray crystallography studies of 2 revealed that the reaction takes place at a [6,6]-bond junction, which is directly over the encapsulated C2 unit and is far from either of the two scandium atoms. On the basis of theoretical calculations and by considering previously reports, we have found that a hexagonal carbon ring on the cage of Sc2 C2 @C3v (8)-C82 is highly reactive toward different reagents due to the overlap of high p-orbital axis vector (POAV) angles and large LUMO coefficients. We propose that this highly concentrated area of reactivity is generated by the encapsulation of the Sc2 C2 cluster because this region is absent from the empty fullerene C3v (8)-C82 . Moreover, the absorption and electrochemical results confirm that derivative 2 is more stable than pristine Sc2 C2 @C3v (8)-C82 , thus illuminating its potential applications.

Background/Aims: Osteopontin (OPN) is an Extracellular Matrix (ECM) molecule and is involved in many physiologic and pathologic processes, including cell adhesion, angiogenesis and tumor metastasis. OPN is a well-known multifunctional factor involved in various aspects of cancer progression, including endometrial cancer. In this study, we examined the significance of OPN in endometrial cancer. Methods: The proliferation, migration and invasion ability of HEC-1A cells were detected by Cell Counting Kit-8 (CCK-8), Wound scratch assay and transwell. Western blots were employed to detect the expression of Matrix metalloproteinase-2 (MMP-2) and epithelial-mesenchymal transition (EMT)-related factors in HEC-1A cells treated with rhOPN. Results: rhOPN promotes cell proliferation, migration and invasion in HEC-1A cells. rhOPN influenced EMT-related factors and MMP-2 expression in HEC-1A cells. rhOPN promoted HEC-1A cells migration, invasion and EMT through protein kinase B (PKB/AKT) and Extracellular regulated protein kinases (ERK1/2) signaling pathway. Conclusions: These results may open up a novel therapeutic strategy for endometrial cancer: namely, rhOPN have important roles in controlling growth of endometrial of cancer cells and suggest a novel target pathway for treatment of this cancer.

Epithelial-mesenchymal transition (EMT) contributes to the occurrence and development of tumors, particularly to the promotion of tumor invasion and metastasis. As a newly discovered ubiquitin hydrolase family member, USP22 plays a key role in the malignant transformation of tumors and the regulation of the cell cycle. However, recent studies on USP22 have primarily focused on its role in cell cycle regulation, and the potential mechanism underlying the promotion of tumor invasion and metastasis by abnormal USP22 expression has not been reported. Our studies revealed that the overexpression of USP22 in PANC-1 cells promoted Ezrin redistribution and phosphorylation and cytoskeletal remodeling, upregulated expression of the transcription factors Snail and ZEB1 to promote EMT, and increased cellular invasion and migration. In contrast, blockade of USP22 expression resulted in the opposite effects. In addition, the focal adhesion kinase (FAK) signaling pathway was shown to play a key role in the process of EMT induction in PANC-1 cells by USP22. Thus, the present study suggests that USP22 acts as a regulatory protein for EMT in pancreatic cancer, which may provide a new approach for the targeted therapy of pancreatic cancer.

The endohedral fullerene once erroneously identified as Sc3@C82 was recently shown to be Sc3C2@Ih-C80, the first example of an open-shell cluster metallofullerene. We herein report that benzyl bromide (1) reacts with Sc3C2@ Ih-C80 via a regioselective radical addition that affords only one isomer of the adduct Sc3C2@Ih-C80(CH2C6H5) (2) in high yield. An X-ray crystallographic study of 2 demonstrated that the benzyl moiety is singly bonded to the fullerene cage, which eliminates the paramagnetism of the endohedral in agreement with the ESR results. Interestingly, X-ray results further reveal that the 3-fold disordered Sc3C2 cluster adopts two different configurations inside the cage. These configurations represent the so-called "planar" form and the computationally predicted, but not crystallographically characterized, "trifoliate" form. It is noteworthy that this is the first crystallographic observation of the "trifoliate" form for the Sc3C2 cluster. In contrast, crystallographic investigation of a Sc3C2@Ih-C80/Ni(OEP) cocrystal, in which the endohedral persists in an open-shell structure with paramagnetism, indicates that only the former form occurs in pristine Sc3C2@ Ih-C80. These results demonstrate that the cluster configuration in EMFs is highly sensitive to the electronic structure, which is tunable by exohedral modification. In addition, the electrochemical behavior of Sc3C2@Ih-C80 has been markedly changed by the radical addition, but the absorption spectra of the pristine and the derivative are both featureless. These results suggest that the unpaired electron of Sc3C2@Ih-C80 is buried in the Sc3C2 cluster and does not affect the electronic configuration of the cage.

Ubiquitin-specific protease 22 (USP22), a newly discovered member of ubiquitin hydrolase family, exhibits a critical function in cell cycle progression and tumorigenesis. The forkhead box M1 (FoxM1) transcription factor plays a crucial role in cell proliferation, differentiation and transformation. However, the expression and functions of USP22 in pancreatic ductal adenocarcinoma (PDA) and whether FoxM1 is involved in USP22-mediated cell cycle regulation have not been studied. We examined the expression of USP22 and FoxM1 in 136 stage II PDA tissues by immunohistochemistry. Clinical significance was analyzed by multivariate Cox regression analysis, Kaplan-Meier curves and log-rank test. RT-PCR, western blot analysis, luciferase and immunofluorescence assays were used to investigate the molecular function of USP22 and FoxM1 in PDA fresh tissues and cell lines. USP22 and FoxM1 were significantly upregulated in PDA tissues compared with the paired normal carcinoma-adjacent tissues. A statistical correlation was observed between USP22 and FoxM1 expression. The expression of USP/FoxM1 and co-expression of both factors correlated with tumor size, lymph node metastasis and overall survival. Multivariate Cox regression analysis revealed that the expression of USP22/FoxM1, especially the co-expression of both factors, is an independent, unfavorable prognostic factor. USP22 overexpression is accompanied by an increase in FoxM1 expression and USP22 increases FoxM1 expression to promote G1/S transition and cell proliferation through promoting β-catenin nuclear translocation in PDA cell lines. USP22 promotes the G1/S phase transition by upregulating FoxM1 expression via promoting β-catenin nuclear localization. USP22 and FoxM1 may act as prognostic markers and potential targets for PDA.

Formation of a very stable, unprecedented pyrazole-ring fused derivative of endohedral metallofullerenes was achieved by the first 1,3-dipolar cycloaddition reaction of Sc3N@C80 with diphenylnitrilimine in a highly regioselective manner.

Endohedral metallofullerenes (EMFs) encapsulating divalent metal ions have rarely been studied because of their low production yields. We report here for the first time the chemical understanding of two typical divalent EMFs, i.e. Yb@D3h-C74 and Yb@C2(13)-C84 with a disilirane reagent (1), which has shown great success in probing the chemical reactivity of EMFs encapsulating a variety of metallic cores. It was surprising to find that Yb@D3h(1)-C74 reacted with 1 neither thermally nor photochemically under normal conditions, showing an unusually low reactivity. However, monoadduct formation was achieved by laser-irradiation of their mixtures during the mass spectroscopic measurement. Accordingly, the thermal and photochemical reactions of Yb@C2(13)-C84 with 1 were conducted under harsher conditions and monoadduct formation was confirmed by both chromatographic and mass spectroscopic studies. Background/Aims: Bufalin can induce apoptosis in certain human cancer cell lines, but bufalin has not yet been thoroughly evaluated in colorectal cancer cells. Cleavage and polyadenylation specific factor 4 (CPSF4) and human telomerase reverse transcriptase (hTERT) play important roles in colorectal cancer growth. The aim of this study was to investigate the roles and interactions of bufalin, CPSF4 and hTERT and the effects of bufalin in human colorectal cancer. Methods: We treated LoVo and SW620 cells with bufalin to investigate the effect of bufalin on proliferation, apoptosis and migration. We verified the relationship between CPSF4 and hTERT using pulldown assays, luciferase reporter assays and chromatin immunoprecipitation (ChIP) assays. Results: Bufalin inhibited the proliferation and migration of and induced apoptosis in LoVo and SW620 cells. We identified CPSF4 as an hTERT promoter-binding protein in colorectal cancer cells. Conclusion: Our study identified bufalin as a potential small molecule inhibitor for cancer therapy.

MicroRNAs (miRNAs) are a class of small non-coding RNAs that are closely associated with carcinogenesis. Accumulating data indicate that miR-196b participates in the development of various types of cancers. However, the role of miR-196b in the formation of hydatidiform mole (HM) is still unclear. Our previous studies have demonstrated that miR-196b levels were decreased in JAR and BeWo cells and in HM tissue samples, as demonstrated by RT-PCR analysis. Furthermore, we discovered that overexpression of miR-196b in JAR and BeWo cells inhibited cellular proliferation, migration and invasion, as shown by Cell counting kit-8 (CCK-8) and transwell assays, respectively. Subsequently, we explored the interaction of miR-196b with its target gene in human choriocarcinoma cell lines. MAP3K1 is a target gene predicted by bioinformatic analysis that was previously shown to exhibit reduced expression levels following treatment with miR-196b in JAR and BeWo cells. We demonstrated that MAP3K1 was a direct target of miR-196b using the dual-luciferase reporter assay in Hela cells. In summary, the present study demonstrated that miR-196b suppressed proliferation, migration and invasion of human choriocarcinoma cells by inhibiting its transcriptional target MAP3K1. miR-196b and MAP3K1 may be considered potential targets for the clinical treatment of HM.

A large family of crystalline organometallic compounds with silver ethynide (R–C≡CAg) as a component has been synthesized and characterized. In most cases the R–C≡C− species coordinate to 3–5 silver(I) atoms, which may be symbolized as R–C≡C⊃Ag n (n = 3, 4, 5; R = alkyl, aryl, heteroaryl) by leaving out the charges. In the present review, emphasis is laid on systematic synthetic and structural studies of high-nuclearity silver ethynide clusters, of which the size and shape can be controlled to a certain extent by the judicious choice of templating anions and auxiliary ligands.

The cage-like structure of fullerenes enables a new strategy for derivatization, that is, endohedral doping with metallic species. Distinctly different from exohedral metal-fullerene complexes, endohedral metallofullerenes (EMFs) show several new features in metal–carbon interactions which result from the electron transfer from the internal metallic species to the outer carbon cage. Taking advantage of recent achievements in the X-ray crystallographic characterization of EMFs, it was revealed that the single metal cation in mono-EMFs are always apart from the cage center, coordinating with the cage from inside, dominantly via electrostatic interactions, whereas the metal–cage interactions are much weaker in di-EMFs, featuring a moving dimetallic cluster in most cases. Moreover, formation of various metallic cluster species inside the fullerene cage suggest that the strong coordination ability of rare earth metals which always requests a maximal coordinated stereo environments, is a critical factor controlling the formation of stable EMF compounds. Finally, the structural features of the fullerene cage, such as the symmetry, the shape and the size, are also found to exert fundamental influences on the metal location or motion.

The organometallic chemistry of endohedral metallofullerenes has lagged far behind that of empty fullerenes. Herein, we report a highly regioselective coordination reaction involving two Sc3N@C80 isomers and W(CO)4(Ph2PC2H4PPh2) (1), which affords only one mononuclear complex for each Sc3N@C80 isomer (2a for Sc3N@Ih-C80 and 2b for Sc3N@D5h-C80). X-ray results demonstrate that the tungsten center coordinates with the cages in an η(2) fashion on a [6,6]-bond in both cases, forming the first derivatives with a closed three membered ring for Sc3N@C80. Although IR and absorption spectroscopic results reveal relatively weak exohedral-metal to ligand π back-donation in both 2a and 2b, they show remarkable air-stability which promises their potential uses in related fields. Moreover, the coordination has narrowed down the electrochemical gaps of the parent Sc3N@C80 isomers, making the complexes better electron donors but worse electron acceptors than the corresponding precursors.

Beta1, 4-Galactosyltransferase-I (β1, 4-GalT-I), which transfers galactose from UDP-Gal to N-acetylglucosamine and N-acetylglucosamine-terminated oligosaccharides of N- and O-linked glycans in a β(1-4) linkage, plays a critical role in cell adhesion, sperm-egg recognition, neurite growth, and tumor cell migration and invasion. Our previously experiments also show that β1, 4-GalT-I was up-regulated by estrogens and some important cytokines of embryo implantation especially Interleukin-1 (IL-1), TGF-α and Leukemia Inhibitory Factor (LIF) in endometrial cells. In the receptive phase human uterus, osteopontin (OPN) is the most highly up-regulated extracellular matrix/adhesion molecule/cytokine. In this study, we demonstrated the correlated expression of OPN and β1, 4-GalT-I in endometrium during early pregnancy, and recombinant human OPN (rhOPN) protein induced the β1, 4-GalT-I up-regulation in RL95-2 cells. Inhibition of MEK/ERK, PI3K/AKT and NF-κB suppressed rhOPN-induced β1, 4-GalT-I expression. In addition, rhOPN promoted the adhesion of blastocysts cells in vitro in β1, 4-GalT-I-dependent manner. Moreover, the adhesion is greatly inhibited when β1, 4-GalT-I was blocked with the specific antibody. Taken together, our data suggest that β1, 4-GalT-I provides a mechanism to bridge embryo to endometrium during implantation.

Covalent porphyrinic cages (CPCs) have been a target of interest for years. In this paper, we report the design and synthesis of two CPCs in which the cofacial porphyrins have a distance of 7.66 and 11.96 A via template-directed imine condensation reactions and through the selective choice of templating linker and diamine length.

A new three-dimensional magnetic network, [Co9(OH)6(C42H24O18P3N3)2(H2O)8] (1), has been successfully prepared by utilizing the flexible hexacarboxylate ligand derived from cyclotriphosphazene, which had been characterized by single-crystal X-ray diffraction and magnetic measurement. This compound consists of one-dimensional (1D) cobalt-hydroxyl chains based on planar nonanuclear clusters, which are located in the b–c plane to form the nearly two-dimensional cobalt layer. Magnetic measurements reveal that 1 shows spin-canted antiferromagnetism with spin-glass behavior. These results suggest that reasonable design and choice of large carboxylate ligand based on a specific scaffold could be effective for the construction of magnetic materials based on a novel 1D magnetic chain.

Lymph node metastasis is recognized as an important mode of liver cancer metastasis. Two hepatocarcinoma cell lines, Hca-F get high (75%) and Hca-P get low (25%) incidences of lymph node metastasis. Forkhead box M1 (FoxM1) is described as a major oncogenic transcription factor in tumor initiation, promotion, and progression. Ezrin is linked to aggressive tumor behavior by involving all stages of tumor metastasis. We firstly compared the expression of FoxM1 and Ezrin between two cells. Then we transiently transfected Hca-P cells with over-FoxM1 plasmid and Hca-F cells with sh-FoxM1 plasmid. We found that both FoxM1 and Ezrin expressed higher in Hca-F than Hca-P. We successfully down-regulated or up-regulated FoxM1 expression in Hca-F or Hca-P cells. And we found that FoxM1 had correlation with proliferation, invasion and migration in mouse hepatocellular carcinoma cell lines.

β1,4-Galactosyltransferase I (β1,4-GalT-I), a key enzyme in glycobiology, mediates several biological mechanisms. However, the correlation between β-1,4-GalT-I expression in the uterine endometrium and embryo implantation remains unclear. This study aims to elucidate the relationship between β1,4-GalT-I and Lewis(Y) (Le(Y)) glycan during embryo implantation. So far, using green fluorescent protein as an indicator, β1,4-GalT-I interference plasmid (pcDNA3.0-siGalT I), overexpression plasmid (pcDNA3.0-HA-GalT I), interference control plasmid (control pcDNA3.0-siGalT I), and empty vector (pcDNA3.0) were transfected into human uterine epithelial RL95-2 cells that imitate the receptive endometrium. Invasive embryos at pre-implantation and treated RL95-2 cells were co-cultured to determine embryo attachment in each of the transfection groups. The results showed that plasmid transfection was successful in all the groups. β1,4-GalT-I and Fucosyltransferase 1 (FUT1) gene expression declined in the interference group, and the synthesis of Le(Y) decreased accordingly, but the expression of this antigen increased in the overexpression group. After co-culturing of the embryos and 36h transfection of RL95-2, the results of these in vitro implantation models showed that the attachment rate was lower in the interference group (30.0 ± 0.2%) than in the untreated group (50.0 ± 0.6%), empty vector group (50.0 ± 0.2%), and interference control group (46.7 ± 0.6%), however, it was highest in the overexpression group (70.0 ± 0.2%). These results indicated that β1,4-galactosyltransferase I possibly regulate mutual uterus-embryo adhesion and embryo implantation by regulating cell surface Le(Y) glycan expression.

To be a successful implantation, endometrial receptivity should be established. Forkhead box M1 (FoxM1) is described as a major oncogenic transcription factor in tumor initiation, promotion, and progression. FoxM1 regulates the expression of lots of targeted genes important to cell differentiation, proliferation and apoptosis; cell-cycle progression; and tumor angiogenesis, migration, invasion, and metastasis. According to these functions, we believe that FoxM1 should also play an essential role in embryo implantation. To test our hypothesis, we observed the expression and distribution of FoxM1 during the early pregnancy of mouse. Then, we used Immunohistochemistry to examine the expression of FoxM1 induced by E2 and/or P4 in the ovariectomized mouse uterus and human endometrium cells. This study further investigated whether FoxM1 was an important factor in the implantation. Our results showed that FoxM1 expressed in the mouse uterus during early pregnancy (Day 1 to 5). The expression of FoxM1 gradually increased along pregnancy process; FoxM1 expression could be increased by E2. On the contrary, FoxM1 expression could be decreased by P4 and E2 plus P4. We also detected the proliferation of human endometrium cells. We found that E2 might promote cells proliferation, while P4 and E2 plus P4 inhibited cells proliferation; Inhibiting FoxM1 could interfere the embryo implantation of mouse. Amplification or inhibiting of FoxM1 in JAR cells can increase or decrease the adhesion rate to Rl95-2 and HEC-1A cells separately. Our data indicate that FoxM1 might play an important role during the process of mouse embryo implantation.

Clinical significance was analyzed by multivariate Cox regression analysis, Kaplan-Meier curves and log-rank test. RT-PCR, western blot analysis, luciferase and immunofluorescence assays were used to investigate the molecular function of USP22 and FoxM1 in PDA fresh tissues and cell lines. USP22 and FoxM1 were significantly upregulated in PDA tissues compared with the paired normal carcinoma-adjacent tissues. A statistical correlation was observed between USP22 and FoxM1 expression. The expression of USP/FoxM1 and co-expression of both factors correlated with tumor size, lymph node metastasis and overall survival. Multivariate Cox regression analysis revealed that the expression of USP22/FoxM1, especially the co-expression of both factors, is an independent, unfavorable prognostic factor. USP22 overexpression is accompanied by an increase in FoxM1 expression and USP22 increases FoxM1 expression to promote G1/S transition and cell proliferation through promoting β-catenin nuclear translocation in PDA cell lines. USP22 promotes the G1/S phase transition by upregulating FoxM1 expression via promoting β-catenin nuclear localization. USP22 and FoxM1 may act as prognostic markers and potential targets for PDA.

A large family of crystalline organometallic compounds with silver ethynide (R–C≡CAg) as a component has been synthesized and characterized. In most cases the R–C≡C− species coordinate to 3–5 silver(I) atoms, which may be symbolized as R–C≡C⊃Ag n (n = 3, 4, 5; R = alkyl, aryl, heteroaryl) by leaving out the charges. In the present review, emphasis is laid on systematic synthetic and structural studies of high-nuclearity silver ethynide clusters, of which the size and shape can be controlled to a certain extent by the judicious choice of templating anions and auxiliary ligands.

Endohedral metallofullerenes (EMFs) encapsulating divalent metal ions have received limited attention because of their low production yields. Here, we report the results of structural determination and chemical functionalization of a typical divalent metallofullerene, Yb@C84(II). Single-crystal X-ray crystallographic studies of Yb@C84/Ni(II)(OEP) cocrystals (OEP is the dianion of octaethylporphyrin) unambiguously established the chiral C2(13)-C84 cage structure and revealed multiple sites for Yb(2+), indicating a moving metal ion inside the cage. The chemical property of Yb@C2(13)-C84 was probed with the electrophillic adamantylidene carbene (1). Three monoadduct isomers were isolated and characterized. Crystallographic results of the major isomer (2b) revealed that, although the cycloaddition breaks a [5,6]-bond on the cage, Yb(2+) is localized under a hexagonal ring distant from the sites of addition. Thus, it is proved that the dynamic motion of the divalent metal ion in Yb@C84 has been effectively halted by exohedral functionalization. Spectroscopic results show that the electronic property of Yb@C2(13)-C84 is pertained in the derivatives, although the addend exerts a mild reduction effect on the electrochemical behavior of the EMF. Computational works demonstrated that addition of 1 to Yb@C2(13)-C84 is mainly driven by releasing the local strains of cage carbons rather than charge recombination, which is always prominent to the affinity of typical trivalent EMFs such as M@C2v(9)-C82 (M = Sc, Y, La, Ce, Gd) toward 1. Accordingly, it is speculated that the chemical behaviors of divalent EMFs more likely resemble those of empty fullerenes because both are closed-shell compounds, but they differ from those of trivalent EMFs, which have open-shell electronic configurations instead.

The cage-like structure of fullerenes enables a new strategy for derivatization, that is, endohedral doping with metallic species. Distinctly different from exohedral metal-fullerene complexes, endohedral metallofullerenes (EMFs) show several new features in metal–carbon interactions which result from the electron transfer from the internal metallic species to the outer carbon cage. Taking advantage of recent achievements in the X-ray crystallographic characterization of EMFs, it was revealed that the single metal cation in mono-EMFs are always apart from the cage center, coordinating with the cage from inside, dominantly via electrostatic interactions, whereas the metal–cage interactions are much weaker in di-EMFs, featuring a moving dimetallic cluster in most cases. Moreover, formation of various metallic cluster species inside the fullerene cage suggest that the strong coordination ability of rare earth metals which always requests a maximal coordinated stereo environments, is a critical factor controlling the formation of stable EMF compounds. Finally, the structural features of the fullerene cage, such as the symmetry, the shape and the size, are also found to exert fundamental influences on the metal location or motion.

We present the first chemical modification of a divalent metallofullerene Yb@C80 by carbene addition. Spectroscopic and crystallographic data suggest that its electronic and electrochemical properties have been effectively altered upon chemical functionalization.

Since the first proposal that fullerenes are capable of hosting atoms, ions, or clusters by the late Smalley in 1985, tremendous examples of endohedral metallofullerenes (EMFs) have been reported. Breaking the dogma that monometallofullerenes (mono-EMFs) always exist in the form of M@C2n while clusterfullerenes always require multiple (two to four) metal cations to stabilize a cluster that is unstable as a single moiety, here we show an unprecedented monometallic endohedral clusterfullerene entrapping an yttrium cyanide cluster inside a popular C82 cage--YCN@C(s)(6)-C82. X-ray crystallography and (13)C NMR characterization unambiguously determine the cage symmetry and the endohedal cyanide structure, unexpectedly revealing that the entrapped YCN cluster is triangular. The unprecedented monometallic clusterfullerene structure unveiled by YCN@C(s)(6)-C82 opens up a new avenue for stabilizing a cluster by a single metal cation within a carbon cage, and will surely stimulate further studies on the stability and formation mechanism of EMFs.
