Keteleeria davidiana (Bertrand) Beissner 1891 (Pinaceae) is a rare tertiary relict plant endemic to China. However, since the main morphological characteristics used for identifying K. davidiana are variable, some taxonomic treatments within the species are still controversial. Here a complete chloroplast genome of K. davidiana representing a special genotype was assembled, which could provide more information for the taxonomic study of this species. The assembled genome was 117,642 bp in length with a large single-copy (LSC) region (74,825 bp), a small single-copy (SSC) region (40,247 bp), and two incomplete inverted repeats (IRs) regions (1285 bp each). In total, 118 genes were predicted, including 4 rRNAs, 34 tRNAs, and 80 protein-coding genes. The overall GC content of the assembled genome was 38.5%. Phylogenetic analysis showed that different accessions of K. davidiana formed a clade with relatively low support (bootstrap value = 71), which indicated a high level of sequences variation within the species.

A layer of cuticular wax is deposited on the surface of terrestrial plants, which reduces the damage caused by environmental stress and maintains growth in a relatively stable internal environment. Apple cuticular wax is an important part of the fruit epidermis that plays an essential role in apple development, storage, and adaptation to environmental stress. The formation of cuticular wax has been described at the transcriptional, post-transcriptional, and translational levels in Arabidopsis, whereas less research has been performed on apple cuticular wax. Here, we provide a brief overview of how apple cuticular wax is formed, as well as its structure, composition, and function. An association among the environment, genes, and apple cuticular wax deposition was revealed. Cuticular wax prevents fruit rust from occurring on apple. Taken together, a detailed understanding of apple cuticular wax is discussed. The results will act as a reference for extending the storage period and increasing the commodity value of apple.

Objective: To analyze the etiology, triggering factors, clinical characteristics and therapeutic outcomes of erythroderma. Methods: The clinical data of 165 inpatients with erythroderma treated in our department from January 2017 to January 2019 were retrospectively analyzed. Results: These patients included 116 males and 49 females, with mean age of (53.46±18.58) years. Erythroderma secondary to skin disorders, drug allergy and tumors accounted 93.33%, 3.03%, and 1.21%, respectively. The etiologies of the rest cases were unknown. The most common triggering factor was non-standardized use of glucocorticoids. Etiology-based treatment regimens yielded effective rate of 72.73%. Conclusions: Erythroderma occurs mainly in aged males. Psoriasis is the most common cause of erythroderma, followed by eczema. The treatment and prognosis of erythroderma are associated with the primary disease. Etiology-based therapy and symptomatic support can improve the survival rate.

BackgroundGuanXinNing tablet (GXNT), a traditional Chinese patent medicine, has been found to have remarkable antithrombotic effects and can effectively inhibit pro-thrombotic factors in previous studies. However, the mechanism of its antithrombotic effects remains little known.MethodsIn this study, we first determined and identified the sources of each main compound in GXNT using liquid chromatography-mass spectrometry (LC-MS). Through the approach of network pharmacology, we predicted the action targets of the active components, mapped the target genes related to thrombus, and obtained potential antithrombotic targets for active ingredients. We then performed gene ontology (GO) enrichment analyses and KEGG signaling pathway analyses for the action targets, and constructed networks of active component–target and active component–target–pathway for GXNT. Additionally, we evaluated the pharmacodynamic effects of GXNT on thrombus using the rat thrombus model induced by FeCl3, observed the effects of antiplatelet aggregation via platelet assay, and further verified the results predicted by network pharmacology via Western blot.ResultsIn total, 14 active ingredients were identified in GXNT, and 83 action targets were predicted, 17 of which are antithrombotic targets that potentially participate in processes including response to oxidative stress and positive regulation of blood vessel endothelial cell migration. KEGG pathway analyses revealed that the predicted action targets were involved in multiple signal pathways, such as MAPK, IL-17, and platelet activation. Pharmacodynamics study found that GXNT could significantly reduce the thrombus length and weight, lower platelet aggregation function, and decrease the levels of Fbg and PAI-1. In addition, GXNT could significantly increase 6-keto-PGF1α content and regulate the ratio of TXB2/6-keto-PGF1α, while not having dramatic effects on TXB2. GXNT was also observed to visibly inhibit maximum platelet aggregation. Herein, we further studied the thrombus-related MAPKs signaling pathway and found that GXNT could significantly reduce the phosphorylation levels of p38MAPK, ERK, and JNK proteins in platelet.ConclusionsThis study revealed the pharmacodynamic material basis of GXNT and its potential multicomponent–multitarget–multipath pharmacological effects, confirmed the antithrombotic effects of GXNT, and showed that its mechanism may be related to inhibiting phosphorylation of p38, ERK, and JNK proteins in MAPKs signaling pathway, partially verifying the results from network pharmacology. The results from this study could provide a theoretical basis for the development and clinical application of GXNT.

Insulin resistance has been implicated in alcoholic liver disease. A previous study has shown that microRNAs (miRNAs) play a major role in the production, secretion, and function of insulin. MiRNAs are capable of repressing multiple target genes that in turn negatively regulate various physiological and pathological activities. However, current information on the biological function of miRNAs in insulin resistance is limited. The goal of the present study was to elucidate the role of miR-378b in alcohol-induced hepatic insulin resistance and its underlying mechanism. This study has observed that miR-378b is up-regulated in National Institute on Alcohol Abuse and Alcoholism (NIAAA) alcoholic mouse models as well as in ethanol-induced L-02 cells in vitro. Furthermore, miR-378b overexpression impaired the insulin signaling pathway, and inhibition of miR-378b improved insulin sensitivity in vivo and in vitro. A mechanistic study revealed that IR and p110α are direct targets of miR-378b. Together, these results suggest that miR-378b controls insulin sensitivity by targeting the insulin receptor (IR) as well as p110α and possibly play an inhibitory role in the development of insulin resistance, thereby providing insights into the development of novel diagnostic and treatment methods.

Digestive cancers-including gastric cancer (GC), colorectal cancer, hepatocellular carcinoma, esophageal cancer, and pancreatic cancer-accounted for 26% of cancer cases and 35% of cancer deaths worldwide in 2018. It is crucial and urgent to develop biomarkers for the diagnosis, prognosis, and therapeutic benefits of digestive cancers, especially for GC, since the incidence of GC is lower only than lung cancer in China, is hard to detect at an early stage, and is associated with poor prognosis. Mucins, glycoproteins encoded by MUC family genes, act as a part of a physical barrier in the digestive tract and participate in various signaling pathways. Some mucins have been used or proposed as biomarkers for carcinomas, such as MUC16 (CA125) and MUC4. However, there are no systematic investigations on the association of MUC family members with diagnoses and clinical outcomes even though relevant data have been largely accumulated in the past decade. By analyzing transcriptomic and clinical data of digestive cancer samples from TCGA involving colon adenocarcinoma (COAD), esophageal carcinoma (ESCA), liver hepatocellular carcinoma (LIHC), stomach adenocarcinoma (STAD), and pancreatic adenocarcinoma (PAAD), it was found that expressions levels of MUC15, MUC13, and MUC21 were individually associated with survival for digestive cancers, and high expressions of EMCN (MUC14) and MUC15 were correlated with poor survival for STAD. Cox regression analysis indicated the predictive power of an EMCN/MUC15 combination for overall survival (OS) of GC patients, which was validated on an independent dataset from GEO. EMCN/MUC15 correlated genes were identified to be enriched in cancer-related processes, such as vasculature development, mitosis, and immunity. Therefore, we propose that an EMCN/MUC15 combination could be a potential prognostic signature for gastric cancer.

Background. Soil contamination by hexavalent chromium is becoming a main environmental concern in China. This study developed a sewage sludge biochar modified by FeSO4 (CHBC) as a new reductant for Cr(VI)-contaminated soil. The effectiveness of CHBC-stabilized Cr(VI)-contaminated soil was investigated. Methods. Typical industrial Cr(VI)-contaminated soil in China was chosen as the medium. The total and Cr(VI) contents of the contaminated soil were 1014.6 and 973.5 mg/kg, respectively. The effectiveness of the Cr(VI)-contaminated soil stabilized by CHBC was investigated by the leaching test (US EPA method 1312), the simplified bioaccessibility extraction test (US EPA 2007 protocol and British Geological Survey), alkaline digestion (US EPA method 3060A), sequential extraction (BCR sequential extraction procedure), X-ray diffraction, and the risk assessment code test. Results. Results show that CHBC substantially reduced the leachability and Cr(VI) content of the contaminated soil. The leachability and content of Cr(VI) were lower than the thresholds of the Environmental Quality Standards of Soil in China for civil reuse and the China Environmental Quality Standards of surface water for civil use when the soil was stabilized with 10% dosage of CHBC. Conclusion. CHBC is highly efficient in stabilizing Cr(VI) and can effectively reduce the leachability and bioavailability of Cr in contaminated soil and thus feasible for stabilizing Cr(VI)-contaminated soil and shows potential for application in the field.

Background: Experimental studies have demonstrated that cadmium exposure induces alterations on immune function, but epidemiological evidence is lacking. Objective: To examine the associations between prenatal and postnatal cadmium exposure and cellular immune responses among pre-school children. Methods: Pre-school aged children (n = 407) were followed from a prospective birth cohort study in Wuhan, China. Maternal urinary and children’s plasma cadmium concentrations were measured as biomarkers of prenatal and postnatal cadmium exposure, respectively. Children’s cellular immune responses were assessed by peripheral blood T lymphocyte subsets and plasma cytokines. Multivariable adjusted models were applied to estimate the associations of prenatal and postnatal cadmium exposure with T lymphocyte subsets and cytokines, and the effect modification by child gender were also examined. Results: Maternal urinary cadmium was associated with reduced absolute counts of CD3+CD4+ cells (−12.45%; 95% CI: −23.74%, 0.40% for the highest vs. lowest quartile; p for trend = 0.045). Inverse associations of maternal urinary cadmium with %CD3+CD4+ cells and CD4+/CD8+ ratio were only observed among females (both p-interaction

P0 proteins encoded by poleroviruses Brassica yellows virus (BrYV) and Potato leafroll virus (PLRV) are viral suppressors of RNA silencing (VSR) involved in abolishing host RNA silencing to assist viral infection. However, other roles that P0 proteins play in virus infection remain unclear. Here, we found that C-terminal truncation of P0 resulted in compromised systemic infection of BrYV and PLRV. C-terminal truncation affected systemic but not local VSR activities of P0 proteins, but neither transient nor ectopic stably expressed VSR proteins could rescue the systemic infection of BrYV and PLRV mutants. Moreover, BrYV mutant failed to establish systemic infection in DCL2/4 RNAi or RDR6 RNAi plants, indicating that systemic infection might be independent of the VSR activity of P0. Partially rescued infection of BrYV mutant by the co-infected PLRV implied the functional conservation of P0 proteins within genus. However, although C-terminal truncation mutant of BrYV P0 showed weaker interaction with its movement protein (MP) when compared to wild-type P0, wild-type and mutant PLRV P0 showed similar interaction with its MP. In sum, our findings revealed the role of P0 in virus systemic infection and the requirement of P0 carboxyl terminal region for the infection.

Whether there is an association between dietary quality and sleep disorder in American adults is unclear. We conducted this study to analyze whether dietary quality, using the Healthy Eating Index-2015 (HEI-2015) scores as the measure, was associated with self-reported sleep disorders. Data came from the National Health and Nutrition Examination Survey (2005–2014). Step-weighted logistic regression models were performed to explore the relationships between the HEI-2015 scores and sleep disorder. Weighted quantile sum regression model was used to identify the HEI-2015 components most strongly associated with sleep disorders. According to quartiles, HEI scores were categorized into inadequate (75%). Compared to inadequate HEI status, average HEI status (OR: 0.961, 95%CI: 0.959–0.962) and optimal HEI status (OR: 0.913, 95% CI: 0.912–0.915) were associated with reduced risk of sleep disorder after multivariable adjustments. Greens and beans, added sugars, saturated fats, total vegetables and total protein foods were the top five important components for sleep disorders. Our results suggest that there is a statistically significant association between better dietary quality and reduced risk of sleep disorder among United States adults.

Background/Aims: Gastric cancer is considered as a common malignancy with a poor prognosis as well as unsatisfactory treatment. Neutrophil gelatinase-associated lipocalin (NGAL) has been reported to affect multiple aspects of human tumor, including gastric cancer. This study aims to explore the effects of NGAL gene silencing on the proliferation as well as apoptosis of human gastric cancer MGC-803 cells. Methods: This study included 87 patients with gastric cancer. MGC-803 cells were collected and mainly treated with siRNA against NGAL and recombinant NGAL plasmid. The expression of NGAL mRNA and the expressions of NGAL protein and apoptosis-related proteins were determined by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and western blot analysis, respectively. Cell cycle and apoptosis were tested by flow cytometry, and cell proliferation was detected by water soluble tetrazolium-1 (WST-1) assay. The effect of NGAL gene silencing on tumorigenicity of MGC-803 cells in vivo was detected through establishment of xenograft in nude mice. Results: NGAL was highly expressed in gastric cancer tissues. The protein and mRNA expressions of NGAL gene in MGC-803 cells treated with NGAL-siRNA were obviously reduced, and the amount of cells in G0/G1 phase was increased. Moreover, MGC-803 cells treated with NGAL-siRNA exhibited inhibited proliferation, enhanced apoptosis, decreased expressions of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) as well as B-cell lymphoma-2 (Bcl-2) and increased expressions of cysteine-aspartic acid specific protease-9 (caspase-9) and Bcl2-associated X (Bax), as well as repressed tumorigenicity in vivo. Conclusion: NGAL gene silencing inhibits proliferation and promotes apoptosis of MGC-803 cells, which can provide a novel theory for treatment of gastric cancer.

Loop-mediated isothermal amplification (LAMP) technology has been applied in a wide range of fields such as detection of foodborne bacteria and clinical pathogens due to its simplicity and efficiency. However, existing LAMP primer designing systems require a conserved gene or a short genome region as input, and they can’t design group-specific primers. With the growing number of whole genomes available, it is possible to design better primers to target a set of genomes with high specificity based on whole genomes. We present here a whole Genome based LAMP primer designer (GLAPD), a new system to design LAMP primer for a set of target genomes using whole genomes. Candidate single primer regions are identified genome wide and then combined into LAMP primer sets. For a given set of target genomes, only primer sets amplifying them and only these genomes will be output. In order to accelerate the primer designing, a GPU version is provided as well. The effectiveness of primers designed by GLAPD has been assessed for a wide range of foodborne bacteria. Abstract Background With increases in global travel and trade, the spread of arboviruses is undoubtedly alarming. Pathogen detection in field-caught mosquitoes can provide the earliest possible warning of transmission. Insect-specific flavivirus (ISFV) has been first detected in 1991 and documented worldwide in the latest ten years. Although infection with ISFVs is apparently limited to insects, an increase in the infection rate of mosquito-borne flaviviruses may be able to induce cytopathic effects in vertebrate cells during co-infection with other human pathogens. However, little is known whether ISFVs persist in most regions of China. Methods During the mosquito activity season in 2016, a surveillance program was carried out to detect ISFVs in mosquitoes in metropolitan Shanghai, China. The presence of ISFVs was randomly tested in different species of mosquitoes using RT-PCR-based and hemi-nested PCR assays, following by the sequencing of PCR products. Sequences from positive pooled samples were compared with those deposited in GenBank. Thereafter, sequences of representative insect flaviviruses were used for further phylogenetic and molecular evolutionary analyses. Results Our investigations showed: (1) the presence of Aedes flavivirus (AEFV) in 11/161 pooled samples (nine pools in Songjiang District, one pool in Huangpu District, and one pool in Qingpu District) of Aedes albopictus, (2) the presence of Quang Binh virus (QBV) in 10/195 pooled samples (all in Chongming District) of Culex tritaeniorhynchus; and (3) the presence of Culex flavivirus (CxFV) in 9/228 pooled samples (six pools in Pudong New Area, two pools in Huangpu District, and one pool in Chongming District) of Cx. pipiens. Furthermore, phylogenetic analyses of the gene sequences of envelope proteins indicated that Shanghai CxFV strains belonged to the Asia/USA genotype. The overall maximum likelihood estimation values (and 95% confidence interval) for CxFV, QBV, and AEFV in mosquitoes collected in Shanghai in 2016 were 1.34 (0.66–2.45), 1.65 (0.87–2.85), and 1.51 (0.77–2.70) per 1000, respectively. Conclusions This study reveals the presence and the geographical distribution of ISFVs, and determines the genetic variation and the infection rate of ISFVs in Shanghai, China. At least, three insect flaviviruses including ISFVs, AEFV, CxFV, and QBV, co-circulate in this area. To our knowledge, this is the first report of AEFV in China.

Objective: This study aimed to investigate effective components of dried ginger (DG) in warming lung to reduce watery phlegm and in vivo tissue distribution on the syndrome of cold fluid retained in lung of rats with chronic obstructive pulmonary disease (COPD) by means of the “syndrome-efficacy-biological sample analysis” method and then to explore its meridian tropism. Methods: Wistar rats were given nasal drops of 200-μL lipopolysaccharide and smoke 30 min two times a day, then put the appropriate dose of ice water, and freeze for an hour to build model rats. On the 16th day, the drug group was orally administered of DG (500 mg/mL) until the 30th day. Blood samples and biological tissues were collected from the orbital venous plexus into heparinized hemostasis tubes at 5, 10, 15, 30, 45, 60, 90, 120, 180, 240, and 360 min after the last administration. Using ultraviolet-high-performance liquid chromatography (Waters, USA) method, the effective components were tested, and DAS 3.0 software (Mathematical Pharmacology Professional Committee of China, Shanghai, China) was used to analyze the results. Results: The compounds of DG entering into blood were 6-gingerol, 6-shogaol, and 8-gingerol. Tissue distribution analysis indicates that three active ingredients are widely present in the lung, spleen, kidney, liver, heart, large intestine, stomach, small intestine, and other organs of rats with COPD. Conclusions: 6-gingerol, 6-shogaol, and 8-gingerol belong to effective components of DG in curing the syndrome of cold fluid retained in lung of rats with COPD and mainly distributed in organs including the spleen, stomach, lung, kidney, liver, and heart.

Brassica yellows virus (BrYV) is a tentative species of the genus Polerovirus, which has at least three genotypes (A, B, and C) in China. The P0 protein of BrYV-A (P0BrA) has been identified as a viral suppressor of RNA silencing (VSR), which can also induce cell death in infiltrated Nicotiana benthamiana leaves. In this study, we demonstrated that the cell death induced by P0BrA was accompanied by the accumulation of reactive oxygen species (ROS) and increased Pathogenesis-related protein genes-1 (PR1) expression. Meanwhile, this cell death phenotype was delayed by salicylic acid (SA) pretreatment. Biological function comparison of the three P0 proteins showed that transiently expressed P0BrB or P0BrC induced a significantly delayed and milder cell death response compared with P0BrA. However, like P0BrA, they also suppressed local and systemic RNA silencing. Six residues of P0BrA essential for inducing cell death were identified by comparative analysis and amino acid substitution assay. We also show that all three BrYV genotypes have synergistic interactions with pea enation mosaic virus 2 (PEMV 2) in N. benthamiana. This study provides theoretical guidance for controlling the viral disease caused by poleroviruses in the future.

Gardenia jasminoides Ellis is a traditional aromatic and medicinal plant in China. Here, the complete chloroplast genome of a wild-type gardenia adapted to island climate was assembled. The assembled genome was 155,247 bp in length, with four typical regions, i.e., a large single-copy (LSC) region (85,414 bp), a small single-copy (SSC) region (18,235 bp) and two inverted repeats (IRs) regions (25,799 bp each). In total, 138 genes were predicted, including 90 protein-coding genes, 40 tRNA genes and eight rRNA genes. The overall GC content of the chloroplast genome was 37.5%. The chloroplast genome would provide more information for the phylogeography and phylogeny study of G. jasminoides.

Abstract Macrosclereid cells, which are a layer in the seed coat of Medicago truncatula, accumulate large amounts of phytochemicals during their development. But little is known about the complex and dynamic changes during macrosclereid cell development. To characterize the phytochemicals and the related gene expression during the development of M. truncatula macrosclereid cells, a high performance liquid chromatography-mass spectrometry (HPLC-MS) assay and microarray study were conducted on transcriptome changes from macrosclereid cell during seed development. A total of 16 flavonoids by HPLC-MS and 4861 genes exhibited significant differences at transcript levels by microarray analysis were identified for macrosclerid cells at six different time points during seed development. 815 abiotic and biotic stress genes, 223 transcriptional factors (TFs), and 155 annotated transporter proteins exhibited differential expression during the development of macrosclereid cells. A total of 102 genes were identified as involved in flavonoid biosynthesis, phenypropanoid biosynthesis, and flavone and flavonol biosynthesis. We performed a weighted gene co-regulatory network (WGCNA) to analyze the gene-flavonoid association and rebuilt the gene regulatory network during macrosclereid cell development. Our studies revealed that macrosclereid cells are, beside as the first barrier of defense against diseases, an excellent model system to investigate the regulatory network that governs flavonoid biosynthesis.

Abstract Background Yunnan Province is located in southwestern China and neighbors the Southeast Asian countries, all of which are dengue-endemic areas. In 2000–2013, sporadic imported cases of dengue fever (DF) were reported almost annually in Yunnan Province. During 2013–2015, we confirmed that a large-scale indigenous DF outbreak emerged in cities of Yunnan Province near the China-Myanmar-Laos border. Methods Epidemiological characteristics of DF in Yunnan Province during 2013–2015 were evaluated by retrospective analysis. A total of 232 dengue virus (DENV)-positive sera were randomly collected for sequence analysis of the capsid/premembrane region of DENV from patients with DF in Yunnan Province. The envelope gene of DENV isolates was also amplified and sequenced. Phylogenetic analyses were performed using the neighbor-joining method with the Tajima-Nei model. Results Phylogenetically, all DENV-positive samples could be classified into DENV-1 genotype I and DENV-2 Asian I genotype during 2013–2015 and DENV-4 genotype I in 2015 from Ruili City; and DENV-3 genotype II in 2013 and DENV-2 Cosmopolitan genotype in 2015 from Xishuangbanna Prefecture. Conclusions Our results indicated that imported DF from patients from Laos and Myanmar was the primary cause of the DF epidemic in Yunnan Province. Additionally, DENV strains of all four serotypes were identified in indigenous cases in Yunnan Province during the same time period, while the dengue epidemic pattern observed in southwestern Yunnan showed characteristics of a hypoendemic nature: circulation of DENV-1 and DENV-2 over consecutive years.

Abstract Background Malignant pleural mesothelioma (MPM) is a difficult-to-treat global disease. Pegylated arginase (BCT-100) has recently shown anti-tumor effects in hepatocellular carcinoma, acute myeloid leukemia and melanoma. This study aims to investigate the effects of PEG-BCT-100 in MPM. Methods A panel of 5 mesothelioma cell lines (H28, 211H, H226, H2052 and H2452) was used to study the in vitro effects of BCT-100 by crystal violet staining. The in vivo effects of BCT-100 were studied using 211H and H226 nude mice xenografts. Protein expression (argininosuccinate synthetase, ornithine transcarbamylase, cleaved PARP, cleaved caspase 3, cyclins (A2, D3, E1 and H), CDK4 and Ki67) and arginine concentration were evaluated by Western blot and ELISA respectively. Cellular localization of BCT-100 was detected by immunohistochemistry and immunoflorescence. TUNEL assay was used to identify cellular apoptotic events. Results Argininosuccinate synthetase was expressed in H28, H226, and H2452 cells as well as 211H and H266 xenografts. Ornithine transcarbamylase was undetectable in all cell lines and xenograft models. BCT-100 reduced in vitro cell viability (IC50 values at 13–24 mU/ml, 72 h) across different cell lines and suppressed tumor growth in both 211H and H226 xenograft models. Background: Chronic intermittent hypoxia is the most remarkable feature of obstructive sleep apnea/hypopnea syndrome and it can induce the change of hypoxia-inducible factor-1α (HIF-1α) expression and contractile properties in the genioglossus. To clarify the role of HIF-1α in contractile properties of the genioglossus, this study generated and compared high-throughput RNA-sequencing data from genioglossus between HIF-1α conditional knockout (KO) mice and littermate wild-type (WT) mice. Methods: KO mice were generated with cre-loxP strategy. Gene expression profile analysis was performed using gene enrichment analysis. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses of differently expressed messenger RNAs were performed to identify the related pathways and biological functions. Six differentially expressed genes (DEGs) were validated by qualitative reverse transcription polymerase chain reaction. Results: A total of 142 (77 upregulated and 65 downregulated) transcripts were found to exhibit statistically significant difference between the HIF-1α-KO and WT mice. GO and KEGG analyses indicated that DEGs included genes involved in “skeletal muscle cell differentiation,” “muscle organ development,” “glucose metabolic process,” “glycogen biosynthetic and metabolic process,” etc. Conclusion: This study might provide evidence that HIF-1α affects the expression of multiple genes involved in the myogenesis, muscle development, and carbohydrate metabolism through transcriptome analysis in conditional HIF-1α-KO mice.

Objective To analyze clinical and pathological characteristics of mitochondrial myopathy (MM) in 15 patients, and to study the value of simplified serum lactic acid exercise test in the screening of mitochondrial myopathy. Methods A total of 15 patients with mitochondrial myopathy diagnosed clinically and pathologically, 11 patients with other muscular diseases (OM), and 21 normal controls were collected. All subjects went up and down stairs for 5 min with medium effort. Blood samples for serum lactic acid detection were collected from all subjects before exercise, immediately after exercise and 10 min after exercise. Serum lactic acid levels were compared among 3 groups and among 3 time points. Results Patients with mitochondrial myopathy mainly presented as paroxysmally progressive muscular soreness and weakness. Histopathological examination showed there were 8 cases with the proportion of ragged red fibers (RRF) more than 5%. Serum lactic acid level before exercise, immediately after exercise and 10 min after exercise were (3.57 ± 1.88), (10.98 ± 4.84) and (7.87 ± 4.38) mmol/L in MM group, (1.89 ± 0.98), (6.05 ± 4.07) and (4.13 ± 3.14) mmol/L in OM group, (1.91 ± 0.53), (3.37 ± 1.22) and (2.52 ± 0.89) mmol/L in control group. Serum lactic acid level in MM group was significantly higher than that in control and OM groups before exercise (P = 0.000, 0.001), immediately after exercise (P = 0.000, 0.001), and 10 min after exercise (P = 0.000, 0.003). Serum lactic acid level in OM group was significantly higher than that in control group immediately after exercise (P = 0.042). Serum lactic acid level in 3 groups immediately after exercise (P = 0.000, 0.000, 0.003) and 10 min after exercise (P = 0.000, 0.000, 0.013) was significantly higher than that before exercise. Serum lactic acid level immediately after exercise was significantly higher than that 10 min after exercise in 3 groups (P = 0.000, 0.000, 0.003). Serum lactic acid level had most obvious elevation in MM group, while had flattest elevation in control group. Conclusions Simplified serum lactic acid exercise test is useful in the creening of mitochondrial myopathy, especially in primary hospitals. DOI:10.3969/j.issn.1672-6731.2016.12.009

Synechococcus is one of the most important contributors to global primary productivity, and ocean warming is predicted to increase abundance and distribution of Synechococcus in the ocean. Here, we investigated molecular response of an oceanic Synechococcus strain WH8102 grown in two nitrogen sources (nitrate and urea) under present (25°C) and predicted future (28°C) temperature conditions using an isobaric tag (IBT)-based quantitative proteomic approach. Rising temperature decreased growth rate, contents of chlorophyll a, protein and sugar in the nitrate-grown cells, but only decreased protein content and significantly increased zeaxanthin content of the urea-grown cells. Expressions of CsoS2 protein involved in carboxysome formation and ribosomal subunits in both nitrate- and urea-grown cells were significantly decreased in rising temperature, whereas carbohydrate selective porin and sucrose-phosphate synthase (SPS) were remarkably up-regulated, and carbohydrate degradation associated proteins, i.e., glycogen phosphorylase kinase, fructokinase and glucose-6-phosphate dehydrogenase, were down-regulated in the urea-grown cells. Rising temperature also increased expressions of three redox-sensitive enzymes (peroxiredoxin, thioredoxin, and CP12) in both nitrate- and urea-grown cells. Our results indicated that rising temperature did not enhance cell growth of Synechococcus; on the contrary, it impaired cell functions, and this might influence cell abundance and distribution of Synechococcus in a future ocean.

Natural bear bile has been used for liver disease in East Asia for thousands of years. However, its use has restrictions. In the current study, the therapeutic effects and potential mechanisms of cultured bear bile powder (CBBP) against hepatic fibrosis were evaluated in a dimethylnitrosamine (DMN)-induced rat model. CBBP treatment significantly improved DMN-induced hepatic necrosis and inflammatory infiltration. Additionally, CBBP remarkably alleviated the increased hepatic collagen content and expression of alpha-smooth muscle actin. Serum metabolomics revealed that 14 serum metabolites, including docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were decreased in DMN-treated rats, which was reversed by CBBP. Pathway analyses revealed that the main metabolic pathways affected by CBBP were related to fatty acid biosynthesis and metabolism, and biosynthesis of unsaturated fatty acids. EPA and DHA are ligands of peroxisome proliferator activated receptors (PPARs). CBBP treatment significantly stimulated liver mRNA and protein expression of PPARα and PPARγ. CBBP also markedly increased liver expression of PPARα target genes, which are involved in fatty acid β-oxidation, and down-regulated IL-6, a downstream inflammatory gene of PPARγ. In conclusion, CBBP has the potential to attenuate liver fibrosis and its mechanism involves the promotion of the liver expression of PPARα and PPARγ. Our results may help in the development of a novel substitute for bear bile and therapeutic strategies for fibrotic liver diseases.

Ordinary Portland cement (OPC) is widely used in the solidification/stabilization of Pb-contaminated soils. However, many studies have suggested that the high content of Pb would degrade the mechanical properties of OPC-solidified/stabilized soils. This paper presents a new binder, geopolymer fine aggregate (GFA), composed of ground granulated blast furnace slag, fly ash, CaO, and Na2SiO3. For comparison, OPC was used as a conventional binder. Mechanical properties and leaching characteristics are typically used to evaluate the effects of binders on solidified/stabilized soils. Nevertheless, limited information on the mechanical properties and leaching characteristics of the GFA-solidified/stabilized soils is available. This study thus investigated the mechanical properties and leaching characteristics of geopolymer-solidified/stabilized Pb-contaminated soil. Unconfined compressive strength test, permeability test, synthetic precipitation leaching procedure, simplified bioaccessibility extraction, phytoavailability extraction (with diethylene-triamine penta-acetic acid), sequential extraction procedure, mercury intrusion porosimetry, and scanning electron microscopy (SEM) were performed on OPC- and GFA-solidified/stabilized soil. The results showed that the GFA presented a better effect on the mechanical properties and leachability of the solidified/stabilized soils than the OPC-solidified/stabilized soils. The GFA-solidified/stabilized soil displayed considerably lower leachability, bioaccessibility, and phytoavailability of Pb and higher mechanical properties and chemical stability than the OPC counterpart. This study demonstrated that GFA had a better effect than OPC on the solidification/stabilization of Pb-contaminated soils.

Due to the surge in type 2 diabetes mellitus (T2DM), treatments for chronic metabolic dysregulations with fewer side-effects are sought. Lycii Cortex (LyC), a traditional Chinese Medicine (TCM) herb has a long history of being widely prescribed to treat T2DM as alternative medicine; however, the bioactive molecules and working mechanism remained unknown. Previous studies revealed kukoamine B (KB) as a major and featured compound for LyC with bioactivities for anti-oxidation and acute inflammation, which may be related to anti-diabetes properties. This study aims to understand the efficacy and the mode of action of KB in the diabetic (db/db) mouse model using a metabolomics approach. Parallel comparison was conducted using the first-line anti-diabetic drugs, metformin and rosligtazone, as positive controls. The db/db mice were treated with KB (50 mg kg−1 day−1) for 9 weeks. Bodyweight and fasting blood glucose were monitored every 5 and 7 days, respectively. Metabolomics and high-throughput molecular approaches, including lipidomics, targeted metabolomics (Biocrates p180), and cytokine profiling were applied to measure the alteration of serum metabolites and inflammatory biomarkers between different treatments vs. control (db/db mice treated with vehicle). After 9 weeks of treatment, KB lowered blood glucose, without the adverse effects of bodyweight gain and hepatomegaly shown after rosiglitazone treatment. Lipidomics analysis revealed that KB reduced levels of circulating triglycerides, cholesterol, phosphatidylethanolamine, and increased levels of phosphatidylcholines. KB also increased acylcarnitines, and reduced systemic inflammation (cytokine array). Pathway analysis suggested that KB may regulate nuclear transcription factors (e.g., NF-κB and/or PPAR) to reduce inflammation and facilitate a shift toward metabolic and inflammatory homeostasis. Comparison of KB with first-line drugs suggests that rosiglitazone may over-regulate lipid metabolism and anti-inflammatory responses, which may be associated with adverse side effects, while metformin had less impact on lipid and anti-inflammation profiles. Our research from holistic and systemic views supports the conclusion that KB is the bioactive compound of LyC for managing T2DM, and suggests KB as a nutraceutical or a pharmaceutical candidate for T2D treatment. In addition, our research provides insights related to metformin and rosiglitazone action, beyond lowering blood glucose.

This study was conducted to evaluate the properties of insulation boards made from bamboo paper sludge and fly ash floating beads. An orthogonal test design was applied to study the effects of certain factors on the properties of the insulation boards. The results indicated that the sequence of significant effect of factors on the properties of the composite boards was particle size of bamboo paper sludge, thickness of boards, and weight ratio of sludge to fly ash floating beads. The verified optimal conditions were confirmed to be 20 to 40 mesh, 80:20, and 14 mm, representing the particle size of bamboo paper sludge, weight ratio of sludge:fly ash floating beads, and thickness of boards, respectively. The thermal conductivity of the bamboo paper sludge and fly ash floating beads insulation boards was measured and suggested that both insulation boards had thermal conductivity values ranging from 0.12 to 0.165 W/mK, which is close to the conventional insulation material lightweight concrete.

Objective: The aim of our study was to evaluate the association between cytochrome P450 2C19 (CYP2C19) 681G>A polymorphisms and the age of development of coronary heart disease. Additionally, the study might find some biological indicators at the gene level that could help to predict and evaluate the risk of coronary heart disease in time. Methods: This study included 352 individuals with coronary heart disease. Polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) was used to identify CYP2C19 681G>A. The objects were divided into wild type (GG or homozygous CYP2C19*1 wild-type) and mutant type (GA/AA or the mutant CYP2C19*2 allele) on the basis of genotype. The association between CYP2C19 gene polymorphism and the age of onset of coronary heart disease was assessed by multivariate linear regression analysis. Results: There was a significant association in the age of onset between the two groups (t=3.398, P=0.001), which was 58.51+12.72 years in the wild type and 53.95+11.63 years in the mutant group. The frequency of CYP2C19 681A (CYP2C19*2 allele) was 3.32, 0.268, 0.227 in different age groups, which was significantly different (χ2=10.745, P=0.005) in different groups, as well as in the genotype. The result, assessed by multiple linear regression, showed that genotype, smoking, obesity, and hyperlipidemia affect the age of onset of coronary heart disease (β was -0.167, 0.156, 0.155, and 0.112, PA gene polymorphism may be one of the risk factors in susceptibility to early onset of coronary heart disease, but not an independent factor because other factors may play a synergistic role.

Background Many stroke patients receive traditional medical care in Korea and China. Stroke patients comprise the largest proportion of inpatients in traditional Korean medicine and traditional Chinese medicine hospitals. The purpose of this study is to identify the types and effects of Chinese medicine widely used in China and Korea, and to apply them to future studies of stroke. Methods Nine Korean and Chinese databases will be surveyed for clinical studies of herbal medicines for stroke, published between the inception of the database and August 2017. Clinical studies of decoctions or modified decoctions will be included without restriction on study type. The frequencies and patterns of formulas or single herb usage and the any type of herbal medicine utilization will be analyzed. Dissemination The results of the systematic review will be published in a peer-reviewed journal and disseminated electronically and in print. Updates of the review will be conducted to inform and guide healthcare practices.

Macronutrients such as nitrogen (N), phosphorus (P), and silicon (Si) are essential for the productivity and distribution of diatoms in the ocean. Responses of diatoms to a particular macronutrient deficiency have been investigated, however, we know little about their common or specific responses to different macronutrients. Here, we investigated the physiology and quantitative proteomics of a diatom Thalassiosira pseudonana grown in nutrient-replete, N-, P-, and Si-deficient conditions. Cell growth was ceased in all macronutrient deficient conditions while cell volume and cellular C content under P- and Si-deficiencies increased. Contents of chlorophyll a, protein and cellular N decreased in both N- and P-deficient cells but chlorophyll a and cellular N increased in the Si-deficient cells. Cellular P content increased under N- and Si-deficiencies. Proteins involved in carbon fixation and photorespiration were down-regulated under all macronutrient deficiencies while neutral lipid synthesis and carbohydrate accumulation were enhanced. Photosynthesis, chlorophyll biosynthesis, and protein biosynthesis were down-regulated in both N- and P-deficient cells, while Si transporters, light-harvesting complex proteins, chloroplastic ATP synthase, plastid transcription and protein synthesis were up-regulated in the Si-deficient cells. Our results provided insights into the common and specific responses of T. pseudonana to different macronutrient deficiencies and identified specific proteins potentially indicating a particular macronutrient deficiency.

The thymic stromal lymphopoietin (TSLP)/TSLP receptor (TSLPR) axis is involved in multiple inflammatory immune diseases, including coronary artery disease (CAD). To explore the causal relationship between this axis and CAD, we performed a three-stage case-control association analysis with 3,628 CAD cases and 3,776 controls using common variants in the genes TSLP, interleukin 7 receptor (IL7R), and TSLPR. Three common variants in the TSLP/TSLPR axis were significantly associated with CAD in a Chinese Han population [rs3806933T in TSLP, Padj = 4.35 × 10−5, odds ratio (OR) = 1.18; rs6897932T in IL7R, Padj = 1.13 × 10−7, OR = 1.31; g.19646A>GA in TSLPR, Padj = 2.04 × 10−6, OR = 1.20]. Reporter gene analysis demonstrated that rs3806933 and rs6897932 could influence TSLP and IL7R expression, respectively. Scaled Subprofile Model of Principal Component Analysis (SSM-PCA) is a multivariate statistical method and has been widely used in Positron Emission Tomography (PET). Recently, SSM-PCA has been applied to discriminate patients with Parkinson's disease and healthy controls with Amplitude of Low Frequency Fluctuation (ALFF) from Resting-State Functional Magnetic Resonance Imaging (RS-fMRI). As RS-fMRI scans are more readily available than PET scans, it is important to investigate the intra- and inter-scanner reliability of SSM-PCA in RS-fMRI. A RS-fMRI dataset with Eyes Open (EO) and Eyes Closed (EC) conditions was obtained in 21 healthy subjects (21.8 ± 1.8 years old, 11 females) on 3 visits (V1, V2, and V3), with V1 and V2 (mean interval of 14 days apart) on one scanner and V3 (about 8 months from V2) on a different scanner. To simulate between-group analysis in conventional SSM-PCA studies, 21 subjects were randomly divided into two groups, i.e., EC-EO group (EC ALFF map minus EO ALFF map, n = 11) and EO-EC group (n = 10). A series of covariance patterns and their expressions were derived for each visit. Only the expression of the first pattern showed significant differences between the two groups for all the visits (p = 0.012, 0.0044, and 0.00062 for V1, V2, and V3, respectively). This pattern, referred to as EOEC-pattern, mainly involved the sensorimotor cortex, superior temporal gyrus, frontal pole, and visual cortex. EOEC-pattern's expression showed fair intra-scanner reliability (ICC = 0.49) and good inter-scanner reliability (ICC = 0.65 for V1 vs. V2 and ICC = 0.66 for V2 vs. V3). While the EOEC-pattern was similar with the pattern of conventional unpaired T-test map, the two patterns also showed method-specific regions, indicating that SSM-PCA and conventional T-test are complementary for neuroimaging studies.

ORF3a, a newly identified non-AUG-initiated ORF encoded by members of genera Polerovirus and Luteovirus, is required for long-distance movement in plants. However, the mechanism of action of P3a in viral systemic movement is still not clear. In this study, sequencing of a brassica yellows virus (BrYV) mutant defective in systemic infection revealed two-nucleotide variation at positions 3406 and 3467 in the genome. Subsequent nucleotide substitution analysis proved that only the non-synonymous substitution (C→U) at position 3406, resulting in P3aP18L, abolished the systemic infection of BrYV. Preliminary investigation showed that wild type BrYV was able to load into the petiole of the agroinfiltrated Nicotiana benthamiana leaves, whereas the mutant displayed very low efficiency. Further experiments revealed that the P3a and its mutant P3aP18L localized to the Golgi apparatus and near plasmodesmata, as well as the endoplasmic reticulum. Both P3a and P3aP18L were able to self-interact in vivo, however, the mutant P3aP18L seemed to form more stable dimer than wild type. More interestingly, we confirmed firstly that the ectopic expression of P3a of other poleroviruses and luteoviruses, as well as co-infection with Pea enation mosaic virus 2 (PEMV 2), restored the ability of systemic movement of BrYV P3a defective mutant, indicating that the P3a is functionally conserved in poleroviruses and luteoviruses and is redundant when BrYV co-infects with PEMV 2. These observations provide a novel insight into the conserved function of P3a and its underlying mechanism in the systemic infection.

Nitrogen (N) is a primary limiting nutrient for bacterial growth and productivity in the ocean. To better understand bacterial community and their N utilization strategy in different N regimes of the ocean, we examined bacterial diversity, diazotrophic diversity, and N utilization gene expressions in the northwestern Pacific Ocean (NWPO) using a combination of high-throughput sequencing and real-time qPCR methods. 521 and 204 different operational taxonomic units (OTUs) were identified in the 16s rRNA and nifH libraries from nine surface samples. Of the 16s rRNA gene OTUs, 11.9% were observed in all samples while 3.5 and 15.9% were detected only in N-sufficient and N-deficient samples. Proteobacteria, Cyanobacteria and Bacteroidetes dominated the bacterial community. Prochlorococcus and Pseudoalteromonas were the most abundant at the genus level in N-deficient regimes, while SAR86, Synechococcus and SAR92 were predominant in the Kuroshio-Oyashio confluence region. The distribution of the nifH gene presented great divergence among sampling stations: Cyanobacterium_UCYN-A dominated the N-deficient stations, while clusters related to the Alpha-, Beta-, and Gamma-Proteobacteria were abundant in other stations. Temperature was the main factor that determined bacterial community structure and diversity while concentration of NOX-N was significantly correlated with structure and distribution of N2-fixing microorganisms. Expression of the ammonium transporter was much higher than that of urea transporter subunit A (urtA) and ferredoxin-nitrate reductase, while urtA had an increased expression in N-deficient surface water. The predicted ammonium transporter and ammonium assimilation enzymes were most abundant in surface samples while urease and nitrogenase were more abundant in the N-deficient regions. These findings underscore the fact that marine bacteria have evolved diverse N utilization strategies to adapt to different N habitats, and that urea metabolism is of vital ecological importance in N-deficient regimes.

Hydroxyproline-rich glycoproteins (HRGPs) are abundant cell wall components involved in mycorrhizal symbiosis, but little is known about their function in orchid mycorrhizal association. To gain further insight into the role of HRGPs in orchid symbiosis, the location and function of HRGPs were investigated during symbiotic germination of Dendrobium officinale. The presence of JIM11 epitope in developing protocorms was determined using immunodot blots and immunohistochemical staining procedures. Real-time PCR was also employed to verify the expression patterns of genes coding for extensin-like genes selected from the transcriptomic database. The importance of HRGPs in symbiotic germination was further investigated using 3,4-dehydro-L-proline (3,4-DHP), an inhibitor of HRGP biosynthesis. In symbiotic cultures, immunodot blots of JIM11 signals were moderate in mature seeds, and the signals became stronger in swollen embryos. After germination, signal intensities decreased in developing protocorms. In contrast, in asymbiotic cultures, JIM11 signals were much lower as compared with those stages in symbiotic cultures. Immunofluorescence staining enabled the visualization of JIM11 epitope in mature embryo and protocorm cells. Positive signals were initially localized in the larger cells near the basal (suspensor) end of uninfected embryos, marking the future colonization site of fungal hyphae. After 1 week of inoculation, the basal end of embryos had been colonized, and a strong signal was detected mostly at the mid- and basal regions of the enlarging protocorm. As protocorm development progressed, the signal was concentrated in the colonized cells at the basal end. In colonized cells, signals were present in the walls and intracellularly associated with hyphae and the pelotons. The precise localization of JIM11 epitope is further examined by immunogold labeling. In the colonized cells, gold particles were found mainly in the cell wall and the interfacial matrix near the fungal cell wall. Four extensin-like genes were verified to be highly up-regulated in symbiotically germinated protocorms as compared to asymbiotically germinated ones. The 3,4-DHP treatment inhibited the accumulation of HRGPs and symbiotic seed germination. In these protocorms, fungal hyphae could be found throughout the protocorms. Our results indicate that HRGPs play an important role in symbiotic germination. They can serve as markers for fungal colonization, establishing a symbiotic compartment and constraining fungal colonization inside the basal cells of protocorms.

Intrahepatic cholestasis is a serious symptom of liver disorders with limited therapies. In this study, we investigated the efficacy of Huangqi decoction (HQD), a two-herb classic traditional Chinese medicine (TCM), in the treatment of alpha-naphthylisothiocyanate (ANIT)-induced intrahepatic cholestasis in mice. HQD treatment ameliorated impaired hepatic function and tissue damage. A metabolomics study revealed that the endogenous metabolites significantly affected by HQD were related to bile acid (BA) biosynthesis and glutathione metabolism pathways. HQD treatment decreased the intrahepatic accumulation of cytotoxic BAs, normalized serum BA levels, and increased biliary and urinary BA excretion. Additionally, HQD restored the hepatic glutathione content and suppressed reactive oxygen species (ROS) in cholestatic mice. Protein and gene analysis revealed that HQD increased the expression of the hepatic metabolizing enzymes cytochrome P450 (CYP) 2B10 and UDP glucuronosyltransferase family 1 member A1 (UGT1A1), as well as multidrug resistance-associated protein 2 (Mrp2), Mrp3, and Mrp4, which play crucial roles in BA homeostasis. Further, HQD increased the protein expression of glutamate-cysteine ligase, which is involved in the synthesis of glutathione. Importantly, HQD increased the nuclear expression of nuclear factor-E2-related factor-2 (Nrf2). In conclusion, HQD protects against intrahepatic cholestasis by reversing the disordered homeostasis of BAs and glutathione.

To make dynamic traffic grooming faster and more efficient,and achieve an intelligent differentiated protection,a differentiated protection strategy with dynamic traffic grooming based on clustering(DPS-DTGC)was proposed.The whole network topology was allocated some clusters based on maximal independent set,in order to reduce the routing time consumption.Meanwhile,by the cooperation of layered auxiliary graph,residual capacity matrix and cluster aggregation layer,the traffic in inter- and intra- clusters would been groomed to realize the reasonable planning of resources and the higher efficiency of grooming.Furthermore,according to the proportion of different priority traffic in one wavelength ,the link importance was evaluated and a smart P-cycle was designed to give differentiated protection to the link.The simulation results show this strategy can make a better utilization of network resource.And with the increase of network load,it will gain a good performance in blocking rate.

Lycii Cortex (LyC) is widely used as a traditional anti-ageing food ingredient in soup, congee and tea for preventing degenerative disease. However, its bioactive components and modes of action have never been characterized scientifically. The aims of this study are to: 1) examine the neuroprotective effects of LyC against oxidative stress; 2) identify the bioactive components; 3) elucidate the possible molecular mechanisms of the key components. Phytochemical profiling using LC–MS/MS revealed that 7-(4-amino-butoxy)-6-methoxycoumarin, kukoamines A and B, and lyciumins A and B are the major characteristic compounds in LyC. LyC crude extract and kukoamines were effective in preventing cell damage in the H2O2-induced primary cerebellar granule neurons (CGNs) model, but kukoamines were more efficacious and potent than the extract. Western blot analysis revealed that KB, the most abundant kukoamine, was capable of inhibiting the neuronal apoptosis via activating the PI3-K/Akt/GSK3β pathway. Thus, our explorative results indicated that kukoamine, the major compounds in LyC, acts as a promising neuroprotectant against the H2O2 induced toxicity in CGNs model via activating the PI3-K/Akt/GSK3β pathway. Furthermore, molecular insight to substantiate the application of LyC or kukoamines for prevention and treatment of neurodegenerative diseases in which oxidative stress is involved is demonstrated.

Objective: The aim of our study was to evaluate the association between cytochrome P450 2C19 (CYP2C19) 681G>A polymorphisms and the age of development of coronary heart disease. Additionally, the study might find some biological indicators at the gene level that could help to predict and evaluate the risk of coronary heart disease in time. Methods: This study included 352 individuals with coronary heart disease. Polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) was used to identify CYP2C19 681G>A. The objects were divided into wild type (GG or homozygous CYP2C19*1 wild-type) and mutant type (GA/AA or the mutant CYP2C19*2 allele) on the basis of genotype. The association between CYP2C19 gene polymorphism and the age of onset of coronary heart disease was assessed by multivariate linear regression analysis. Results: There was a significant association in the age of onset between the two groups (t=3.398, P=0.001), which was 58.51±12.72 years in the wild type and 53.95±11.63 years in the mutant group. The frequency of CYP2C19 681A (CYP2C19*2 allele) was 0.32, 0.268, 0.227 in different age groups, which was significantly different (χ2=10.745, P=0.005) in different groups, as well as in the genotype. The result, assessed by multiple linear regression, showed that genotype, smoking, obesity, and hyperlipidemia affect the age of onset of coronary heart disease (β was -0.167, 0.156, 0.155, and 0.112, PA gene polymorphism may be one of the risk factors in susceptibility to early onset of coronary heart disease, but not an independent factor because other factors may play a synergistic role.

Based on the inhomogeneous T−Q relation constructed via the off-diagonal Bethe Ansatz, the Bethe-type eigenstates of the XXZ spin-12 chain with arbitrary boundary fields are constructed. It is found that by employing two sets of gauge transformations, proper generators and reference state for constructing Bethe vectors can be obtained respectively. Given an inhomogeneous T−Q relation for an eigenvalue, it is proven that the resulting Bethe state is an eigenstate of the transfer matrix, provided that the parameters of the generators satisfy the associated Bethe Ansatz equations.

Background: Pyroptosis is the term for caspase-1-dependent cell death associated with pro-inflammatory cytokines. The role of alveolar macrophage (AM) pyroptosis in the pathogenesis of the acute lung injury and acute respiratory distress syndrome (ALI/ARDS) remains unclear. Methods: C57BL/6 wild-type mice were assigned to sham, lipopolysaccharide (LPS) + vehicle, LPS + acetyl-tyrosyl-valyl- alanyl-aspartyl-chloromethylketone (Ac-YVAD-CMK) and LPS + Z-Asp-Glu-Val-Asp-fluoromethylketone groups. Mice were given intraperitoneal (IP) injections of LPS. Drugs were IP injected 1 h before LPS administration. Mice were sacrificed 16 h after LPS administration, and AMs were isolated. Western blot analysis for active caspase-1 and cleaved caspase-3, evaluation of lung injury and a cytokine release analysis were performed. AMs were treated with LPS and adenosine triphosphate (ATP); caspase-1-dependent cell death was evaluated using flow cytometry; the apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) pyroptosomes were examined by immunofluorescence. Results: The expression of activated caspase-1 in AMs was enhanced following LPS challenge compared with the sham group. In the ex vivo study, the caspase-1/propidium iodide-positive cells, caspase-1 specks and ASC pyroptosomes were up-regulated in AMs following LPS/ATP stimulation. The specific caspase-1 inhibitor Ac-YVAD-CMK inhibited the activation of caspase-1 and pyroptotic cell death. Ac-YVAD-CMK also reduced the lung injury, pulmonary edema and total protein in bronchoalveolar lavage fluid (BALF). In addition, Ac-YVAD-CMK significantly inhibited interleukin-α2 (IL-1α2) release both in serum and BALF and reduced the levels of IL-18, tumor necrosis factor-α± (TNF-α±), High Mobility Group Box 1 (HMGB1) in BALF during LPS-induced ALI/ARDS. Conclusions: This study reported AM pyroptosis during LPS-induced ALI/ARDS in mice and has demonstrated that Ac-YVAD-CMK can prevent AM-induced pyroptosis and lung injury. These preliminary findings may form the basis for further studies to evaluate this pathway as a target for prevention or reduction of ALI/ARDS.

Background: DNA hypomethylation of long interspersed nuclear elements-1 (LINEs-1) occurs during carcinogenesis, whereas information addressing LINE-1 methylation in Wilms tumor (WT) is limited. The main purpose of our study was to quantify LINE-1 methylation levels and evaluate their relationship with relative telomere length (TL) in WT. Methods: We investigated LINE-1 methylation and relative TL using bisulfite-polymerase chain reaction (PCR) pyrosequencing and quantitative PCR, respectively, in 20 WT tissues, 10 normal kidney tissues and a WT cell line. Significant changes were analyzed by t-tests. Results: LINE-1 methylation levels were significantly lower (P < 0.05) and relative TLs were significantly shorter (P < 0.05) in WT compared with normal kidney. There was a significant positive relationship between LINE-1 methylation and relative TL in WT (r = 0.671, P = 0.001). LINE-1 Methylation levels were significantly associated with global DNA methylation (r = 0.332, P < 0.01). In addition, relative TL was shortened and LINE-1 methylation was decreased in a WT cell line treated with the hypomethylating agent 5-aza-2′-deoxycytidine compared with untreated WT cell line. Conclusion: These results suggest that LINE-1 hypomethylation is common and may be linked to telomere shortening in WT.

Background: This study characterized the cardiac telocyte (TC) population both in vivo and in vitro, and investigated its telomerase activity related to mitosis. Methods: Using transmission electron microscopy and a phase contrast microscope, the typical morphological features of cardiac TCs were observed; by targeting the cell surface proteins CD117 and CD34, CD117 + CD34 + cardiac TCs were sorted via flow cytometry and validated by immunofluorescence based on the primary cell culture. Then the optimized basal nutrient medium for selected population was examined with the cell counting kit 8. Under this conditioned medium, the process of cell division was captured, and the telomerase activity of CD117 + CD34 + cardiac TCs was detected in comparison with bone mesenchymal stem cells (BMSCs), cardiac fibroblasts (CFBs), cardiomyocytes (CMs). Results: Cardiac TCs projected characteristic telopodes with thin segments (podomers) in alternation with dilation (podoms). In addition, 64% of the primary cultured cardiac TCs were composed of CD117 + CD34 + cardiac TCs; which was verified by immunofluorescence. In a live cell imaging system, CD117 + CD34 + cardiac TCs were observed to enter into cell division in a short time, followed by an significant invagination forming across the middle of the cell body. Using a real-time quantitative telomeric-repeat amplification assay, the telomerase concentration in CD117 + CD34 + cardiac TCs was obviously lower than in BMSCs and CFBs, and significantly higher than in CMs. Conclusions: Cardiac TCs represent a unique cell population and CD117 + CD34 + cardiac TCs have relative low telomerase activity that differs from BMSCs, CFBs and CMs and thus they might play an important role in maintaining cardiac homeostasis.

AIM: To investigate into the potential involvement of pyrin containing 3 gene (NLRP3), a member of the nucleotide-binding oligomerization domain-like receptors with cytosolic pattern recognition, in the host defense of corneas against viruses. METHODS: The herpes viral keratitis model was utilized in BALB/c mice with inoculation of herpes simplex virus-1 (HSV-1). Corneal tissues removed during therapy of patients with viral keratitis as well as a Simian vacuolating virus 40 (SV40)-immortalized human corneal epithelial cell line were also examined. Immunohistochemistry was used to detect NLRP3 in these subjects, focusing on their distribution in tissue or cells. Western blot was used to measure the level of NLRP3 and another two related molecules in NLPR3 inflammasome, namely caspase-1 and IL-1β. RESULTS: The NLRP3 activation induced by HSV-1 infection in corneas was accompanied with redistribution of NLRP3 from the cytoplasm to the nucleus in both murine and human corneal epithelial cells. Furthermore, in the SV40-immortalized human corneal epithelial cells, NLRP3 was exclusively located in the nucleus, and treatment of the cells with high concentration of extracellular potassium (known as an inhibitor of NLRP3 activation) effectively drove NLRP3 back to the cytoplasm as reflected by both immunohistochemistry and Western blot. CONCLUSION: It is proposed that herpes virus infection activates and causes redistribution of NLRP3 to nuclei. Whether this NLRP3 translocation occurs with other viral infections and in other cell types merit further study.

Apple replant disease (ARD) causes the inhibition of root system development, stunts tree growth and so on. To further investigate the effects of ARD on apple fruits, a 25-year-old apple orchard was remediated to establish a replant orchard between November 2008 and March 2009. A rotational cropping orchard was established on an adjacent wheat field. The cultivar and rootstock-scion combination used in the newly established orchards was Royal Gala/M26/Malus hupehensis Rehd. Ripe fruits were collected in mid-August 2011 and mid-August 2012, meanwhile, the following indices were measured: yield per plant; fruit weight; the fruit shape index; the contents of anthocyanin, carotenoid and chlorophyll; the soluble sugar content in the flesh; titratable acid; the sugar-acid ratio; firmness; and aroma components; apple plant ground diameter, plant height increment and the total length of the current-year shoots. The results showed that compared to rotational cropping, continuous cropping yielded statistically significant reductions in fruit weight and yield per plant of 39.8 and 76.5%, respectively. However, there were no changes in the fruit shape index. The anthocyanin and carotenoid contents decreased by 81.7 and 37.7%, respectively, while the chlorophyll content increased by 251.0%. All of these differences in content were statistically significant. The soluble sugar levels and sugar-acid ratio decreased by 25.4 and 60.9%, respectively, but the titratable acid levels and fruit firmness increased by 90.9 and 42.8%, respectively. Ten of the most important esters contributing to the apple aroma were analyzed, and the following changes were observed: hexyl acetate, butyl acetate, hexyl butyrate, acetate-2-methyl butyl, 2-methyl-hexyl butyrate, amyl acetate, butyl butyrate, 2-methyl-butyl butyrate, hexyl propionate and hexyl hexanoate decreased by 25.5, 78.4, 89.1, 55.5, 79.5, 77.2, 86.8, 69.9, 61.2, and 68.1%, respectively. The contents of three other aroma components, (E)-2-hexenal, hexanal and 1-hexanol, significantly increased. Eight characteristic aroma components were found in the rotational cropping fruits: hexyl acetate, butyl acetate, acetate-2-methyl butyl, 2-methyl-hexyl butyrate, amyl acetate, 2-methyl- butyl butyrate, hexyl acetate and hexyl propionate. There were four characteristic ester components (hexyl acetate, butyl acetate, acetate-2-methyl butyl, 2-methyl-hexyl butyrate) and two characteristic aldehyde aroma components ((E)-2-hexenal and hexanal) in the continuous cropping fruits. Compared with the rotational cropping fruits, four characteristic ester components were declined and two characteristic aldehyde aroma components were increased. Compared with the control, replanted apple plant ground diameter, plant height increment and the total length of the current-year shoots were reduced by 27.6, 40.6 and 72.2%, respectively.

Polerovirus P0 suppressors of host gene silencing contain a consensus F-box-like motif with Leu/Pro (L/P) requirements for suppressor activity. The Inner Mongolian Potato leafroll virus (PLRV) P0 protein (P0PL-IM) has an unusual F-box-like motif that contains a Trp/Gly (W/G) sequence and an additional GW/WG-like motif (G139/W140/G141) that is lacking in other P0 proteins. We used Agrobacterium infiltration-mediated RNA silencing assays to establish that P0PL-IM has a strong suppressor activity. Mutagenesis experiments demonstrated that the P0PL-IM F-box-like motif encompasses amino acids 76-LPRHLHYECLEWGLLCG THP-95, and that the suppressor activity is abolished by L76A, W87A, or G88A substitution. The suppressor activity is also weakened substantially by mutations within the G139/W140/G141 region and is eliminated by a mutation (F220R) in a C-terminal conserved sequence of P0PL-IM. As has been observed with other P0 proteins, P0PL-IM suppression is correlated with reduced accumulation of the host AGO1-silencing complex protein. However, P0PL-IM fails to bind SKP1, which functions in a proteasome pathway that may be involved in AGO1 degradation. These results suggest that P0PL-IM may suppress RNA silencing by using an alternative pathway to target AGO1 for degradation. Our results help improve our understanding of the molecular mechanisms involved in PLRV infection.

Rubber, as a kind of macromolecular material often used in large ships, aviation, aerospace, and other fields, has remarkable viscoelasticity at room temperature. Therefore, it is of great significance to evaluate the viscoelastic properties of polymer composites. In this paper, four kinds of rubber materials are taken as research objects. Based on the principle of ultrasonic detection, the viscoelastic evaluation of the sample materials is carried out through experiments and simulations. On the basis of previous research, the surface reflection method (SRM) and the bottom reflection method (BRM) are compared in depth. First, the spectrum of received signals is analyzed, and the storage elastic modulus, loss elastic modulus, attenuation coefficient and loss tangent value are obtained. Secondly, the results of the BRM and the SRM are compared and analyzed in the frequency domain of −6 dB. The results show that both the SRM and BRM are feasible in the evaluation of the viscoelasticity of the material, and the variation trends observed for the above-mentioned parameters in the effective frequency domain are consistent, especially at the center frequency. Finally, aiming at the mode transformation of the acoustic wave around the ultrasonic sensor, the practical performance of the surface reflection method is optimized by increasing the diameter of the ultrasonic sensor.

The reflected waves received from ultrasonic waves propagating in materials contain information that constitutes the physical properties, material composition, defects, and degradation states. When measuring the dynamic viscoelasticity, the traditional bottom reflection method (BRM) cannot be used to measure the bottom irregular samples. In this paper, the storage modulus, loss modulus, and loss tangent are extracted by the surface reflection method (SRM) to evaluate the elastomer sample viscoelasticity. A theoretical study on the phase change caused by multiple reflections in the case of non-thin layer coupling is conducted. Based on this research, the experimental system is built. The results show that considering the thickness of the coupling layer can optimize the determination of viscoelasticity and reduce the error of the viscoelastic evaluation results of an elastomer with the traditional BRM. Finally, based on the principle of the SRM, the density of the elastomers is measured, and the feasibility and overall efficiency of this method are verified by experiments.

Ten diterpenoid alkaloids, including eight aconitine-type C19-diterpenoid alkaloids and two hetisine-type C20-diterpenoid alkaloids, were isolated from the secondary roots of Aconitum carmichaeli Debx., known as “Fuzi” in Chinese traditional herbal medicine. Their structures were established on the basis of their spectroscopic data and comparison with those of the literature. Among these alkaloids, chasmanine, oxonitine and 15-acetylsongoramine were isolated for the first time from this medicinal plant. The cytotoxic activity of the alkaloids were tested against several cell lines by the MTT method in which aconitine, hypaconitine, mesaconitne and oxonitine were found to strongly inhibit the growth of the HePG2 cell line, which showed that the existence and quantity of the ester groups have a significant influence on the cytotoxicity of the diterpenoid alkaloids.
