PurposeVitreous vitamin C, as an anti-oxidant, is responsible for regulating oxygen tension and oxidative stress in the eye. Oxidative stress and retinal ischemia are implicated in the development of proliferative diabetic retinopathy (PDR). In this study, we aimed to determine whether vitreous level of vitamin C is compromised in patients with PDR and to investigate the association of diabetic macular ischemia and vitamin C.MethodsThis prospective study enrolled forty patients who underwent pars plana vitrectomy for the treatment of PDR (PDR group, n = 20) and idiopathic epiretinal membrane (control group, n = 20). Serum, aqueous humor, and the vitreous were collected for the analysis of vitamin C level by HPLC. Diabetic macular ischemia (DMI) in PDR group was evaluated with fluorescein angiography (FA).ResultsPDR patients (60.4 ± 2.1 y) were younger than non-diabetic control patients (67.4 ± 1.2 y). Serum, aqueous, and vitreous levels of vitamin C in PDR were 38.7%, 22.5%, and 11.1% of non-diabetic control group, respectively. All PDR patients had DMI (grade 1: 25%, grade 2: 30%, grade 3: 30%, grade 4: 15%). DMI grade was inversely correlated with the level of vitreous vitamin C (r = -0.546, P = 0.019), not with HbA1C, serum, or aqueous vitamin C level. In addition, the level of vitreous vitamin C (4.5 ± 2.6 μg/ml) in high DMI group (Gr 3 &4) was lower than that (31.0 ± 9.1 μg/ml) in low DMI group (Gr 1&2) (P = 0.015).ConclusionsVitreous level of vitamin C in PDR patients showed a tenfold decrease, which was associated with the degree of macular ischemia. This suggests that vitreous vitamin C depletion may cause macula ischemia in PDR patients.

The cellular macromolecular contents and energy value of phytoplankton as primary food source determine the growth of higher trophic levels, affecting the balance and sustainability of oceanic food webs. Especially, proteins are more directly linked with basic functions of phytoplankton biosynthesis and cell division and transferred through the food chains. In recent years, the East/Japan Sea (EJS) has been changed dramatically in environmental conditions, such as physical and chemical characteristics, as well as biological properties. Therefore, developing an algorithm to estimate the protein concentration of phytoplankton and monitor their spatiotemporal variations on a broad scale would be invaluable. To derive the protein concentration of phytoplankton in EJS, the new regional algorithm was developed by using multiple linear regression analyses based on field-measured data which were obtained from 2012 to 2018 in the southwestern EJS. The major factors for the protein concentration were identified as chlorophyll-a (Chl-a) and sea surface nitrate (SSN) in the southwestern EJS. The coefficient of determination (r2) between field-measured and algorithm-derived protein concentrations was 0.55, which is rather low but reliable. The satellite-derived estimation generally follows the 1:1 line with the field-measured data, with Pearson’s correlation coefficient, which was 0.40 (p-value < 0.01, n = 135). No remarkable trend in the long-term annual protein concentration of phytoplankton was found in the study area during our observation period. However, some seasonal difference was observed in winter protein concentration between the 2003–2005 and 2017–2019 periods. The algorithm is developed for the regional East/Japan Sea (EJS) and could contribute to long-term monitoring for climate-associated ecosystem changes. For a better understanding of spatiotemporal variation in the protein concentration of phytoplankton in the EJS, this algorithm should be further improved with continuous field surveys.

We aimed to evaluate the preclinical efficacy of GC1118, a novel anti-epidermal growth factor receptor (EGFR) monoclonal antibody (mAb), against glioblastoma (GBM) tumors using patient-derived xenograft (PDX) models. A total of 15 distinct GBM PDX models were used to evaluate the therapeutic efficacy of GC1118. Genomic data derived from PDX models were analyzed to identify potential biomarkers associated with the anti-tumor efficacy of GC1118. A patient-derived cell-based high-throughput drug screening assay was performed to further validate the efficacy of GC1118. Compared to cetuximab, GC1118 exerted comparable growth inhibitory effects on the GBM tumors in the PDX models. We confirmed that GC1118 accumulated within the tumor by crossing the blood–brain barrier in in vivo specimens and observed the survival benefit in GC1118-treated intracranial models. Genomic analysis revealed high EGFR amplification as a potent biomarker for predicting the therapeutic efficacy of GC1118 in GBM tumors. In summary, GC1118 exerted a potent anti-tumor effect on GBM tumors in PDX models, and its therapeutic efficacy was especially pronounced in the tumors with high EGFR amplification. Our study supports the importance of patient stratification based on EGFR copy number variation in clinical trials for GBM. The superiority of GC1118 over other EGFR mAbs in GBM tumors should be assessed in future studies.

The East China Sea (ECS) is the largest marginal sea in the northern western Pacific Ocean. In comparison to various physical studies, little information on the seasonal patterns in community structure of phytoplankton is currently available. Based on high performance liquid chromatography (HPLC) pigment analysis, spatiotemporal variations in phytoplankton community compositions were investigated in the northern ECS. Water temperature and salinity generally decreased toward the western part of the study area but warmer conditions in August led to strong vertical stratification of the water column. In general, major inorganic nutrient concentrations were considerably higher in the western part with a shallow water depth, and consistent with previous results, had no discernable vertical pattern during our observation period except in August. This study also revealed PO4-limited environmental conditions in May and August. The monthly averaged integral chlorophyll-a concentration varied seasonally, highest (35.2 ± 20.22 mg m−2) in May and lowest (5.2 ± 2.54 mg m−2) in February. No distinct vertical differences in phytoplankton community compositions were observed for all the sampling seasons except in August when cyanobacteria predominated in the nutrient-deficient surface layer and diatoms prevailed at deep layer. Canonical correlation analysis results revealed that nutrient distribution and the water temperature were the major drivers of the vertical distribution of phytoplankton communities in August. Spatially, a noticeable difference in phytoplankton community structure between the eastern and western parts was observed in November with diatom domination in the western part and cyanobacteria domination in the eastern part, which were significantly (p < 0.01) correlated with water temperature, salinity, light conditions, and nutrient concentrations. Overall, the two major phytoplankton groups were diatoms (32.0%) and cyanobacteria (20.6%) in the northern ECS and the two groups were negatively correlated, which holds a significant ecological meaning under expected warming ocean conditions.

Prostate cancer is the fourth most common cancer affecting South Korean males, and the biochemical recurrence (BCR) of prostate cancer occurs in approximately 25% of patients five years after radical prostatectomy. The ability to predict BCR would help clinicians and patients to make better treatment decisions. Therefore, in this study, we have proposed a web-based clinical decision support system that predicts the BCR of prostate cancer in Korean patients. The data were obtained from the Korean Prostate Cancer Registry (KPCR) database, which contained information about 7394 patients with prostate cancer who were treated at one of the six major medical institutions in South Korea between May 2001 and December 2014. We tested 13 prediction models and selected the gradient boosting classifier because it demonstrated excellent prediction performance. Using this model, we were able to create a web application and once clinical data from patients were entered, the three- and five-year post-surgery BCR predictions could be extracted. We developed a clinical decision support system to provide a prostate cancer BCR predictive function to facilitate postoperative follow-up and clinical management. This system will help clinicians develop a strategic approach for prostate cancer treatment by predicting the likelihood of prostate cancer recurrence.

In this paper, we introduce a novel system for physics-based vehicle simulation from input trajectory. The proposed system approximates the physical movements of a real vehicle using a proportional derivative (PD) servo which estimates proper torques for wheels and controls a vehicle’s acceleration based on the conditions of the given trajectory. To avoid expensive simulation calculation, the input trajectory is segmented and compared to the optimized trajectories stored in a path library. Based on the similarity of the curve shape between the input and simulated trajectories, an iterative search method is introduced to generate a physically derivable trajectory for convincing simulation results. For an interaction with other objects in the virtual environment, the surface of the vehicle is subdivided into several parts and deformed individually from external forces. As demonstrated in the experimental results, the proposed system can create diverse traffic scenes with multiple vehicles in a fully automated way.

Several studies have been conducted to evaluate the efficacy of statins in Korean and Asian patients. However, most previous studies only observed the percent reduction in low-density lipoprotein cholesterol (LDL-C) and did not consider the effects of various patient conditions simultaneously, such as abnormal test results, patient demographics, and prescribed drugs before taking a statin. Moreover, the characteristics of the patients whose percent reduction in LDL-C was higher than expected were not provided. Therefore, in this study, we aimed to derive meaningful phenotypes by using tensor factorization to observe the characteristics of the patients whose percent reduction in LDL-C was higher than expected among patients taking moderate-intensity statins. In addition, we used the derived phenotypes to predict how much the LDL-C levels of new patients decreased. We consequently identified eight phenotypes that represented the characteristics of the patients whose percent reduction in LDL-C was higher than expected. Moreover, the latent representations of the derived phenotypes achieved prediction performance similar to that obtained using the raw data. These results demonstrate that the derived phenotypes and latent representations are useful tools for observing the characteristics of patients and predicting LDL-C levels. Additionally, our findings provide direction on how to conduct clinical studies in the future.

Dynamic human movements such as dance are difficult to capture without using external markers due to the high complexity of a dancer’s body. This article introduces a marker-free motion capture and composition system for dance motion that uses multiple RGB and depth sensors. Our motion capture system utilizes a set of high-speed RGB and depth sensors to generate skeletal motion data from an expert dancer. During the motion acquisition process, a skeleton tracking method based on a particle filter is provided to estimate the motion parameters for each frame from a sequence of color images and depth features retrieved from the sensors. The expert motion data become archived in a database. The authoring methods in our composition system automate most of the motion editing processes for general users by providing an online motion search with an input posture and then performing motion synthesis on an arbitrary motion path. Using the proposed system, we demonstrate that various dance performances can be composed in an intuitive and efficient way on client devices such as tablets and kiosk PCs.

Recent studies show that IL-22, a cytokine produced by activated CD4+ T cells and NK cells, plays a pathogenic role in acute and chronic skin diseases. While IL-22 is produced by immune cells, the expression of IL-22Rα, the functional subunit of IL-22R, is mostly restricted to non-hematopoietic cells in organs such as the skin and pancreas. Although it is well known that ultraviolet B (UVB) radiation induces skin inflammation, there have been no reports regarding the effect of UVB on the expression of IL-22Rα. This study investigated IL-22Rα expression and IL-22-mediated proliferation and pro-inflammatory cytokine production by UVB-irradiated keratinocytes. IL-22Rα was increased in HaCaT and primary human keratinocytes after UVB irradiation through the translocation of IL-22Rα from the cytosol to the membrane. This increase in the expression of IL-22Rα was mediated by the PI3K/Akt pathway. Moreover, the suppression of keratinocyte proliferation by UVB irradiation was inhibited by treatment with IL-22. At the same time, IL-22 increased the production of IL-1α, IL-6, and IL-18 in UVB-irradiated HaCaT cells and primary human keratinocytes. Finally, IL-22Rα expression was increased in UVB-irradiated human and mouse skin by immunohistochemistry. The increased expression of IL-22Rα therefore promotes keratinocyte proliferation and pro-inflammatory cytokine production during UVB-induced skin inflammation, suggesting that UVB facilitates skin inflammation by increasing the responsiveness of keratinocytes to IL-22. This study provides a new insight into UVB-induced skin inflammation and the regulation of related inflammatory skin diseases.

Excessive smartphone use causes personal and social problems. To address this issue, we sought to derive usage patterns that were directly correlated with smartphone dependence based on usage data. This study attempted to classify smartphone dependence using a data-driven prediction algorithm. We developed a mobile application to collect smartphone usage data. A total of 41,683 logs of 48 smartphone users were collected from March 8, 2015, to January 8, 2016. The participants were classified into the control group (SUC) or the addiction group (SUD) using the Korean Smartphone Addiction Proneness Scale for Adults (S-Scale) and a face-to-face offline interview by a psychiatrist and a clinical psychologist (SUC = 23 and SUD = 25). We derived usage patterns using tensor factorization and found the following six optimal usage patterns: 1) social networking services (SNS) during daytime, 2) web surfing, 3) SNS at night, 4) mobile shopping, 5) entertainment, and 6) gaming at night. The membership vectors of the six patterns obtained a significantly better prediction performance than the raw data. For all patterns, the usage times of the SUD were much longer than those of the SUC. From our findings, we concluded that usage patterns and membership vectors were effective tools to assess and predict smartphone dependence and could provide an intervention guideline to predict and treat smartphone dependence based on usage data.

For this study, we applied activated biochar (AB) and its composition with magnetite (AB-Fe3O4) as adsorbents for the removal of polychlorophenols in model wastewater. We comprehensively characterized these adsorbents and performed adsorption tests under several experimental parameters. Using FTIR, we confirmed successful synthesis of AB-Fe3O4 composite through cetrimonium bromide surfactant. We conducted adsorption tests using AB and AB-Fe3O4 to treat model wastewater containing polychlorophenols, such as 2,3,4,6-Tetrachlorophenol (TeCP), 2,4,6-Trichlorophenol (TCP), and 2,4-Dichlorophenol (DCP). Results of the isotherm and the kinetic experiments were well adapted to Freundlich’s isotherm model and the pseudo-second-order kinetic model, respectively. Main adsorption mechanisms in this study were attributed to non-covalent, π-electron acceptor−donor interactions and hydrophobic interactions judging from the number of chloride elements in each chlorophenol and its hydrophobic characteristics. We also considered the electrostatic repulsion effect between TeCP and AB, because adsorption performance of TeCP at basic condition was slightly worse than at weak acidic condition. Lastly, AB-Fe3O4 showed high adsorption selectivity of TeCP compared to other persistent organic pollutants (i.e., bisphenol A and sulfamethoxazole) due to hydrophobic interactions. We concluded that AB-Fe3O4 may be used as novel adsorbent for wastewater treatment including toxic and hydrophobic organic pollutants (e.g., TeCP).

In this paper, we introduce a novel acoustic source localization in a three-dimensional (3D) space, based on a direction estimation technique. Assuming an acoustic source at a distance from adjacent microphones, its waves spread in a planar form called a planar wavefront. In our system, the directions and steering angles between the acoustic source and the microphone array are estimated based on a planar wavefront model using a delay and sum beamforming (DSBF) system and an array of two-dimensional (2D) microelectromechanical system (MEMS) microphones. The proposed system is designed with parallel processing hardware for real-time performance and implemented using a cost-effective field programmable gate array (FPGA) and a micro control unit (MCU). As shown in the experimental results, the localization errors of the proposed system were less than 3 cm when an impulsive acoustic source was generated over 1 m away from the microphone array, which is comparable to a position-based system with reduced computational complexity. On the basis of the high accuracy and real-time performance of localizing an impulsive acoustic source, such as striking a ball, the proposed system can be applied to screen-based sports simulation.

As glioblastomas are mostly localized infiltrative lesions, gene therapy based on the retroviral replicating vector (RRV) system is considered an attractive strategy. Combinations of multiple suicide genes can circumvent the limitations associated with each gene, achieving direct and synergistic cytotoxic effects, along with bystander cell killing. In this study, we constructed a semi-and pseudotyped-RRV (sp-RRV) system harboring two suicide genes—herpes simplex virus type 1 thymidine kinase (TK) and yeast cytosine deaminase (CD)—to verify the dissemination and antitumor efficacy of our sp-RRV system (spRRVe-sEF1α-TK/sRRVgp-sEF1α-CD) in seven patient-derived glioblastoma stem-like cells (GSCs). Flow cytometry and high-content analysis revealed a wide range of transduction efficiency and good correlation between the delivery of therapeutic genes and susceptibility to the prodrugs ganciclovir and 5-fluorocytosine in patient-derived GSCs in vitro. Intra-tumoral delivery of spRRVe-sEF1α-TK/sRRVgp-sEF1α-CD, combined with prodrug treatment, synergistically inhibited cell proliferation and angiogenesis while increasing apoptosis and the depletion of tumor-associated macrophages in orthotopic glioblastoma xenografts. Genomic profiling of patient-derived GSCs revealed that the key genes preventing sp-RRV infection and transmission were associated with cell adhesion, migration, development, differentiation, and proliferation. This is the first report demonstrating that a novel sp-RRV-mediated TK/CD double suicide gene transfer system has high oncolytic power against extremely heterogeneous and treatment-refractory glioblastomas.

Gut dysbiosis is regarded as a pathogenetic factor of nonalcoholic fatty liver disease (NAFLD), but its role in NAFLD persistence is unknown. We investigated the influence of the gut microbiota on persistent NAFLD. This cohort study included 766 subjects with 16S ribosomal RNA (rRNA) gene sequencing data from fecal samples at baseline who underwent repeated health check-up examinations. Fatty liver was determined using ultrasound at baseline and follow-up. Participants were categorized into four groups: none (control), developed, regressed, or persistent NAFLD. The persistent NAFLD group had lower richness compared with the control group. Significant differences were also found in both non-phylogenic and phylogenic beta diversity measures according to NAFLD persistence. Pairwise comparisons indicated that taxa abundance mainly differed between the control and persistent NAFLD groups. A relative high abundance of Fusobacteria and low abundance of genera Oscillospira and Ruminococcus of the family Ruminococcaceae and genus Coprococcus of the family Lachnospiraceae were found in the persistent NAFLD group. Based on the functional predictions, pathways related to primary and secondary bile acid biosynthesis were highly detected in the persistent NAFLD group compared with the control group. These findings support that the composition of the gut microbiome associated with dysregulation of bile acid biosynthetic pathways may contribute to the persistence of NAFLD. This is the first cohort study to demonstrate the influence of microbiota on persistent NAFLD. Our findings may help identify potential targets for therapeutic intervention in NAFLD.

The impact of incidentally identified diffuse thyroid FDG uptake on 18F-FDG PET/CT scan on the incidence of thyroid dysfunction remains unclear. We examined the association of diffuse thyroid FDG uptake with the development of thyroid dysfunction. This cohort study involved 39,098 Korean adults who were free of malignancy and thyroid disease at baseline and underwent regular health checkup examinations including an 18F-FDG whole body PET/CT scan, thyroid-stimulating hormone and free thyroxine. The participants were annually or biennially followed for up to 5 years. A parametric proportional hazard model was used to estimate the adjusted hazard ratio (HR) and 95% confidence interval (CI). Diffuse thyroid uptake was positively associated with increased risk of thyroid dysfunction in both the cross-sectional and cohort studies. During 104,261.4 person-years of follow-up, 102 incident hypothyroidism cases and 172 hyperthyroidism cases were identified. Multivariable-adjusted HR (95% CI) for incident hypothyroidism or hyperthyroidism comparing diffuse thyroid uptake to no uptake were 15.72 (9.23–26.77) and 7.38 (4.23–12.87), respectively. In this large cohort, incidentally, identified diffuse thyroid uptake on 18F-FDG PET/CT was associated with increased risk of both prevalent and incident thyroid dysfunction. Therefore, baseline and follow-up evaluations in individuals with diffuse thyroid uptake may help identify individuals with thyroid dysfunction.

The removal process of activated sludge in sewage treatment plants is very nonlinear, and removal performance has a complex causal relationship depending on environmental factors, influent load, and operating factors. In this study, how causal relationships are expressed in collected data was identified by structural equation modeling. First, path modeling was attempted as a preliminary step in structural equation model (SEM) construction and, as a result, the nutrient-removal mechanism could not be sufficiently represented as a direct causal relationship between measured variables. However, as a result of the deduced SEMs for effluent total nitrogen (T-N) and total phosphorus (T–P) concentrations, accompanied by exploratory factor analysis to extract latent variables, a causal network was formed that describes the direct or indirect effect of the latent factors and measured variables. Hereby, this study suggests that it is possible to construct an SEM explaining the nutrient-removal mechanism of the activated-sludge process with latent variables. Moreover, nonlinear features embedded in the mechanism could be represented by SEM, which is a model based on linearity, by including causal relations and variables that were not derived by path analysis. This attempt to model the direct and indirect causalities of the process could enhance the understanding of the process, and help decision making such as changing the driving conditions that would be required.

This paper introduces a GPU (graphics processing unit)-based fast motion synthesis algorithm for a large crowd. The main parts of the algorithms were selecting the most appropriate joint model given adaptive screen-space occupancy of each character and synthesizing motions for the joint model with one or two input motion capture data. The different joint models had a character range from fine-detailed and fully-articulated ones to the most simplified ones. The motion synthesizer, running on a GPU, performed a series of motion blending for each joint of the characters in parallel. For better performance of the motion synthesizer, the GPU maintained a novel cache structure for given speed parameters. Using the high computation power of GPUs, the motion synthesizer could generate arbitrary speeds and orientations for the motions of a vast number of characters. Experiments showed that the proposed algorithm could animate more than 5000 characters in real-time on modest graphics acceleration cards.

Nonalcoholic fatty liver disease (NAFLD) and gallstone disease (GD) are often found to coexist but the sequential relationship of NAFLD and GD to each other remains controversial. We prospectively evaluated the bidirectional relationship of NAFLD with GD. A cohort study was performed on Korean adults who underwent a health checkup and were followed annually or biennially for a mean of 6.0 years. Fatty liver and gallstones were diagnosed by ultrasound. NAFLD was defined as hepatic steatosis on ultrasonography in the absence of excessive alcohol use or other identifiable causes. The NAFLD severity was determined by non-invasive fibrosis markers. Among 283,446 participants without either gallstones or cholecystectomy at baseline, 6440 participants developed gallstones. Among 219,641 participants without NAFLD at baseline, 49,301 participants developed NAFLD. The multivariable-adjusted hazard ratio (95% confidence interval) for incident gallstone comparing the NAFLD group vs. the non-NAFLD group was 1.26 (1.17⁻1.35). Increased non-invasive fibrosis markers of NAFLD were positively associated with an increased incidence of gallstones in a graded and dose-responsive manner (p-trend < 0.01). The multivariable-adjusted hazard ratios (95% confidence intervals) for incident NAFLD comparing gallstone and cholecystectomy to no GD were 1.14 (1.07⁻1.22) and 1.17 (1.03⁻1.33), respectively. This large-scale cohort study of young and middle-aged individuals demonstrated a bidirectional association between NAFLD and GD. NAFLD and its severity were independently associated with an increased incidence of gallstones, while GD and cholecystectomy were also associated with incident NAFLD. Our findings indicate that the conditions may affect each other, requiring further studies to elucidate the potential mechanisms underlying this association.

Human poses are difficult to estimate due to the complicated body structure and the self-occlusion problem. In this paper, we introduce a marker-less system for human pose estimation by detecting and tracking key body parts, namely the head, hands, and feet. Given color and depth images captured by multiple red, green, blue, and depth (RGB-D) cameras, our system constructs a graph model with segmented regions from each camera and detects the key body parts as a set of extreme points based on accumulative geodesic distances in the graph. During the search process, local detection using a supervised learning model is utilized to match local body features. A final set of extreme points is selected with a voting scheme and tracked with physical constraints from the unified data received from the multiple cameras. During the tracking process, a Kalman filter-based method is introduced to reduce positional noises and to recover from a failure of tracking extremes. Our system shows an average of 87% accuracy against the commercial system, which outperforms the previous multi-Kinects system, and can be applied to recognize a human action or to synthesize a motion sequence from a few key poses using a small set of extremes as input data.

The purpose of this study was to identify personality factor-associated predictors of smartphone addiction predisposition (SAP). Participants were 2,573 men and 2,281 women (n = 4,854) aged 20-49 years (Mean ± SD: 33.47 ± 7.52); participants completed the following questionnaires: the Korean Smartphone Addiction Proneness Scale (K-SAPS) for adults, the Behavioral Inhibition System/Behavioral Activation System questionnaire (BIS/BAS), the Dickman Dysfunctional Impulsivity Instrument (DDII), and the Brief Self-Control Scale (BSCS). In addition, participants reported their demographic information and smartphone usage pattern (weekday or weekend average usage hours and main use). We analyzed the data in three steps: (1) identifying predictors with logistic regression, (2) deriving causal relationships between SAP and its predictors using a Bayesian belief network (BN), and (3) computing optimal cut-off points for the identified predictors using the Youden index. Identified predictors of SAP were as follows: gender (female), weekend average usage hours, and scores on BAS-Drive, BAS-Reward Responsiveness, DDII, and BSCS. Female gender and scores on BAS-Drive and BSCS directly increased SAP. BAS-Reward Responsiveness and DDII indirectly increased SAP. We found that SAP was defined with maximal sensitivity as follows: weekend average usage hours > 4.45, BAS-Drive > 10.0, BAS-Reward Responsiveness > 13.8, DDII > 4.5, and BSCS > 37.4. This study raises the possibility that personality factors contribute to SAP. And, we calculated cut-off points for key predictors. These findings may assist clinicians screening for SAP using cut-off points, and further the understanding of SA risk factors.

IL-22 is a pro- and anti-inflammatory cytokine that is mainly produced by T cells and NK cells. Recent studies have reported the increased number of IL-22 producing T cells in patients with autoimmune noninfectious uveitis; however, the correlation between IL-22 and uveitis remains unclear. In this study, we aimed to determine the specific role of IL-22 and its receptor in the pathogenesis of uveitis. Serum concentration of IL-22 was significantly increased in uveitis patients. IL-22Rα was expressed in the retinal pigment epithelial cell line, ARPE-19. To examine the effect of IL-22, ARPE-19 was treated with recombinant IL-22. The proliferation of ARPE-19 and the production of monocyte chemoattractant protein (MCP)-1 from ARPE-19 were clearly elevated. IL-22 induced MCP-1 which facilitated the migration of inflammatory cells. Moreover, IL-22 increased the IL-22Rα expression in ARPE-19 through the activation of PI3K/Akt. Experimental animal models of uveitis induced by interphotoreceptor retinoid binding protein 1-20 (IRBP1-20) exhibited elevation of hyperplasia RPE and IL-22 production. When CD4+ T cells from the uveitis patients were stimulated with IRBP1-20, the production of IL-22 definitely increased. In addition, we examine the regulatory role of cysteamine, which has an anti-inflammatory role in the cornea, in uveitis through the down-regulation of IL-22Rα expression. Cysteamine effectively suppressed the IRBP1-20-induced IL-22Rα expression and prevented the development of IRBP1-20-induced uveitis in the experimental animal model. These finding suggest that IL-22 and its receptor have a crucial role in the development and pathogenesis of uveitis by facilitating inflammatory cell infiltration, and that cysteamine may be a useful therapeutic drug in treating uveitis by down-regulating IL-22Rα expression in RPE.

Estimation of the motion of ball-shaped objects is essential for the operation of ball sport simulators. In this paper, we propose an estimation system for 3D ball motion, including speed and angle of projection, by using acoustic vector and infrared (IR) scanning sensors. Our system is comprised of three steps to estimate a ball motion: sound-based ball firing detection, sound source localization, and IR scanning for motion analysis. First, an impulsive sound classification based on the mel-frequency cepstrum and feed-forward neural network is introduced to detect the ball launch sound. An impulsive sound source localization using a 2D microelectromechanical system (MEMS) microphones and delay-and-sum beamforming is presented to estimate the firing position. The time and position of a ball in 3D space is determined from a high-speed infrared scanning method. Our experimental results demonstrate that the estimation of ball motion based on sound allows a wider activity area than similar camera-based methods. Thus, it can be practically applied to various simulations in sports such as soccer and baseball.

It has recently been reported that the CD40-CD40 ligand (CD40L) interaction is important in Th17 development. In addition, transforming growth factor-beta (TGF-β) promotes tumorigenesis as an immunosuppressive cytokine and is crucial in the development of Th17 cells. This study investigated the role of CD40 in breast cancer cells and its role in immunosuppressive function and tumor progression. CD40 was highly expressed in the breast cancer cell line MDA-MB231, and its stimulation with CD40 antibodies caused the up-regulation of TGF-β. Direct CD40-CD40L interaction between MDA-MB231 cells and activated T cells also increased TGF-β production and induced the production of IL-17, which accelerated the proliferation of MDA-MB231 cells through the activation of STAT3. Taken together, the direct CD40-CD40L interaction of breast tumor cells and activated T cells increases TGF-β production and the differentiation of Th17 cells, which promotes the proliferation of breast cancer cells.
