A good temperature management, such as precooling and cold storage, can delay deterioration of fresh produce. In this study, different forced-air precooling times were applied on Musa AAA Berangan to investigate the influence of forced-air precooling time on the changes of quality attributes and consumer acceptance. The banana was subjected to forced-air precooling treatment (5 ± 1°C) for 0, 14, 50, and 120 min and then stored in a cold room (13 ± 1°C) for 2 weeks. Then, all the fruits were transferred to a ripening room (25 ± 2°C) and initiated to ripen with ethylene gas. Quality attributes analyses and sensory evaluations were conducted when the fruits reached maturity index 5. Quality parameters, such as soluble solids concentration, titratable acidity, pulp firmness, and peel colour, showed no significant differences when fruits were precooled at different times. Blackening of peel as a result of chilling injury occurred in fruits treated with forced-air precooling for 50 and 120 min. This blackening significantly influenced consumer acceptance, although it did not affect the pulp colour and taste. [ABSTRACT FROM AUTHOR]

University-industry collaborative (UIC) in Malaysia has received very little research attention in the literature. Furthermore, worldwide development of UIC project management approaches has been carried out without due consideration of the unique attributes associated neither with this specific environment nor with the high degree of uncertainty intrinsically associated with research projects. This investigation leverages on the available literature, interviews with university-industry research partners and our own works to understand the nature of UIC R&D. This work focuses particular attention on the factors that relate to current project management practices and the methodology applied. It present the findings of qualitative interviews conducted with respondents from universities and industries involved in collaborative R&D projects in Malaysia. Two aspects were investigated; the development process and project management practices in UIC. The study concludes by presenting an outline of the requirements and components required of a project management methodology (PMM) designed specifically for the management of UIC projects. It will provide valuable information and insights to both universities and industries on how to embark on designing a PMM specifically for use in the UIC research environment. [ABSTRACT FROM AUTHOR]

Aims: Venoms of snakes, scorpions, bees and purified venom phospholipase A2 (PLA2) enzymes were examined to evaluate the antibacterial activity of purified venom enzymes as compared with that of the crude venoms. Methods and Results: Thirty-four crude venoms, nine purified PLA2s and twol-amino acid oxidases (LAAO) were studied for antibacterial activity by disc-diffusion assay (100 μg ml−1). Several snake venoms ( Daboia russelli russelli, Crotalus adamanteus, Naja sumatrana, Pseudechis guttata, Agkistrodon halys, Acanthophis praelongus and Daboia russelli siamensis) showed activity against two to four different pathogenic bacteria. Daboia russelli russelli and Pseudechis australis venoms exhibited the most potent activity against Staphylococcus aureus, while the rest showed only a moderate activity against one or more bacteria. The order of susceptibility of the bacteria against viperidae venoms was – S. aureus > Proteus mirabilis > Proteus vulgaris > Enterobacter aerogenes > Pseudomonas aeruginosa and Escherichia coli. The minimum inhibitory concentrations (MIC) against S. aureus was studied by dilution method (160–1·25 μg ml−1). A stronger effect was noted with the viperidae venoms (20 μg ml−11) as compared with elapidae venoms (40 μg ml−1). The MIC were comparable with those of the standard drugs (chloramphenicol, streptomycin and penicillin). Conclusion: The present findings indicate that viperidae ( D. russelli russelli) and elapidae ( P. australis) venoms have significant antibacterial effects against gram (+) and gram (−) bacteria, which may be the result of the primary antibacterial components oflaao, and in particular, the PLA2 enzymes. The results would be useful for further purification and characterization of antibacterial agents from snake venoms. Objectives: To investigate the effect of δ-aminolevulinic acid dehydratase (ALAD) polymorphisms on the association between blood lead and renal function among Vietnamese and Singaporean workers who were exposed to low to medium levels of inorganic lead, and to study the distribution of ALAD polymorphism among Vietnamese, Chinese, Malays and Indians. Methods: A total of 459 male and female workers were studied. Blood and urine were collected for each worker in order to determine ALAD genotype, blood lead, and urinary δ-aminolevulinic acid (ALAU). Renal function tests included urine albumin (Ualb), urine β2 microglobulin (Uβ2m), urinary α1 microglobulin (Uα1m), N-acelyl-glucosaminidas (NAG), and urine retinol blinding protein (RBP). A multiple regression model with interaction term was applied to fit the entire data and to explore the modifying effect of ALAD polymorphism on the relation of blood lead to each renal function parameter. Results: ALAD1-1 was the predominant genotype for all the ethnic groups while ALAD2-2 was the rarest. The frequency of ALAD2 allele was higher among Malays (8.8%) and Indians (10.6%) compared to the Chinese (5.0%) and Vietnamese (4.3%). The geometric mean of blood lead for all workers was 19.0 μg/dl. The models for U–2m, Uα1m, and NAG showed that the ALAD1-2/2-2 group had higher β coefficients than the ALAD1-1 group. Corresponding to 10 μg/dl blood lead, ALAD1-1 homozygotes had an increment of 1.288 μg/g Cr, 1.175 mg/g Cr, and 1.995 U/g Cr for Uβ2m, Uα1 m, and NAG, respectively. ALAD1-2/2-2 subjects had higher increments of 3.802 μg/g Cr, 2.138 mg/g Cr, and 3.89 U/g Cr for Uβ2m, Uα1m, and NAG, respectively. Conclusion: The frequency of the ALAD2 allele is as low in Vietnamese workers as in Chinese. Background: Burkholderia pseudomallei are the causative agent of melioidosis. Increasing resistance of the disease to antibiotics is a severe problem in treatment regime and has led to intensification of the search for new drugs. Antimicrobial peptides are the most ubiquitous in nature as part of the innate immune system and host defense mechanism. Methods: Here, we investigated a group of venoms (snakes, scorpions and honey bee venoms) for antimicrobial properties against two strains of Gram-negative bacteria Burkholderia pseudomallei by using disc-diffusion assay for in vitro susceptibility testing. The antibacterial activities of the venoms were compared with that of the isolated L-amino acid oxidase (LAAO) and phospholipase A2 (PLA2s) enzymes. MICs were determined using broth dilution method. Bacterial growth was assessed by measurement of optical density at the lowest dilutions (MIC 0.25 mg/ml). The cell viability was measured using tetrazolium salts (XTT) based cytotoxic assay. Results: The studied venoms showed high antimicrobial activity. The venoms of C. adamanteus, Daboia russelli russelli, A. halys, P. australis, B. candidus and P. guttata were equally as effective as Chloramphenicol and Ceftazidime (30 µg/disc). Among those tested, phospholipase A2 enzymes (crotoxin B and daboiatoxin) showed the most potent antibacterial activity against Gram-negative (TES) bacteria. Naturally occurring venom peptides and phospholipase A2 proved to possess highly potent antimicrobial activity against Burkholderia pseudomallei. The XTT-assay results showed that the cell survival decreased with increasing concentrations (0.05-10 mg/mL) of Crotalus adamanteus venom, with no effect on the cell viability evident at 0.5 mg/mL. Conclusion: This antibacterial profile of snake venoms reported herein will be useful in the search for potential antibacterial agents against drug resistant microorganisms like B. pseudomallei. We examined six newly identified polymorphisms in the delta-aminolevulinic acid dehydratase (ALAD) single-nucleotide polymorphisms (SNPs) to determine if these SNPs could modify the relationship between blood lead (PbB) and some renal parameters. This is a cross-sectional study of 276 lead-exposed workers in Vietnam. All workers were measured for PbB, urinary retinol-binding protein (URBP), urinary alpha1-microglobulin (Ualpha1m), urinary beta2-microglobulin (Ubeta 2m), urinary N-acetyl-beta -d-glucosaminidase (NAG), urinary aminolevulinic acid (ALAU), serum alpha1-microglobulin (Salpha1m), serum beta2-microglobulin (Sbeta 2m), and urinary albumin (Ualb). The six SNPs were Msp and Rsa in exon 4, Rsa39488 in exon 5, HpyIV and HpyCH4 in intron 6, and Sau3A in intron 12. Analysis of covariance (ANCOVA) with interaction of PbB times SNPs were applied to examine modifying effect of the SNPs on the association of renal parameters and PbB, adjusting for potential confounders of age, gender, body mass index, and exposure duration. HpyCH4 was found to be associated with certain renal parameters. For HpyCH4 1-1, an increase of 1 microg/dL PbB caused an increase of 1.042 mg/g creatinine (Cr) U alpha1m, 1.069 mg/g Cr Ubeta 2m, 1.038 mg/g Cr URBP, and 1.033 mg/g Cr Ualb, whereas in HpyCH4 1-2, an increase of 1 microg/dL PbB resulted in an increase of only 1.009 mg/g Cr Ualpha1m, 1.012 mg/g Cr Ubeta 2m, 1.009 mg/g Cr URBP, and 1.007 mg/g Cr Ualb. HpyCH4 SNP appeared to modify the lead toxicity to kidney with wild-type allele being more susceptible than variants. The mechanism for this effect is not clear. The protein profiles of Blastocystis hominis, B. lapemi, and B. ratti were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and species could be differentiated by this means as well as by Western-blot analysis with polyclonal antibodies. No intraspecies difference could be distinguished between the two B. hominis isolates or the three B. ratti isolates. Western-blot analysis showed extensive cross-reactivity of B. lapemi and B. hominis antigens with anti- B. ratti serum. The development of cysts of Blastocystis hominis isolated from human feces by the Ficoll-Paque concentration method and cultured in Jones' medium containing 10% horse serum is described. The morphological changes were studied by light and transmission electron microscopy at different intervals for up to 48 h. The cysts developed into a large number of vacuolar forms within 24 h, and binary fission was the only mode of reproduction observed. [ABSTRACT FROM AUTHOR]

The present study investigated the pathogenesis of Blastocystis hominis by intramuscular injection of the organism into experimental mice. A total of 27 naïve BALB/c mice aged 6–8 weeks were injected in the leg muscle with axenic culture isolate B of B. hominis. Histological examination at different times revealed that B. hominis could produce a severe inflammatory reaction and myonecrosis. Most changes were observed at 6 h after injection and for up to 2–3 days. By 2 weeks the muscle had regained normal histology. There was infiltration of polymorphonuclear leukocytes (PML) into the injection site, indicating that B. hominis had a strong chemoattractant activity for PML. [ABSTRACT FROM AUTHOR]
