Abstract Kluyveromyces marxianus is a food‐safe yeast with great potential for producing heterologous proteins. Improving the yield in K. marxianus remains a challenge and incorporating large‐scale functional modules poses a technical obstacle in engineering. To address these issues, linear and circular yeast artificial chromosomes of K. marxianus (KmYACs) were constructed and loaded with disulfide bond formation modules from Pichia pastoris or K. marxianus. These modules contained up to seven genes with a maximum size of 15 kb. KmYACs carried telomeres either from K. marxianus or Tetrahymena. KmYACs were transferred successfully into K. marxianus and stably propagated without affecting the normal growth of the host, regardless of the type of telomeres and configurations of KmYACs. KmYACs increased the overall expression levels of disulfide bond formation genes and significantly enhanced the yield of various heterologous proteins. In high‐density fermentation, the use of KmYACs resulted in a glucoamylase yield of 16.8 g/l, the highest reported level to date in K. marxianus. Transcriptomic and metabolomic analysis of cells containing KmYACs suggested increased flavin adenine dinucleotide biosynthesis, enhanced flux entering the tricarboxylic acid cycle, and a preferred demand for lysine and arginine as features of cells overexpressing heterologous proteins. Consistently, supplementing lysine or arginine further improved the yield. Therefore, KmYAC provides a powerful platform for manipulating large modules with enormous potential for industrial applications and fundamental research. Transferring the disulfide bond formation module via YACs proves to be an efficient strategy for improving the yield of heterologous proteins, and this strategy may be applied to optimize other microbial cell factories.

The purpose of this paper is to present a study of $ \alpha $-$ \eta $-type generalized $ F $-proximal contraction mappings in the framework of modular metric spaces and to prove some best proximity point theorems for these types of mappings. Some examples are given to show the validity of our results. We also apply our results to establish the existence of solutions for a certain type of non-linear integral equation.

In this paper, we investigated several new fixed points theorems for multivalued mappings in the framework $ b $-metric spaces. We first generalized $ S $-iterative schemes for multivalued mappings to above spaces by means of a convex structure and then we developed the existence of fixed points and approximate endpoints of the multivalued contraction mappings using iteration techniques. Furthermore, we introduced the modified $ S $-iteration process for approximating a common endpoint of a multivalued $ \alpha_{s} $-nonexpansive mapping and a multivalued mapping satisfying conditon $ (E^{'}) $. We also showed that this new iteration process converges faster than the $ S $-iteration process in the sense of Berinde. Some convergence results for this iterative procedure to a common endpoint under some certain additional hypotheses were proved. As an application, we applied the $ S $-iteration process in finding the solution to a class of nonlinear quadratic integral equations.

Abstract Immune checkpoint inhibitor (ICI) rechallenge in non‐small cell lung cancer (NSCLC) is a promising therapeutic strategy. The situation for ICI rechallenge can be divided into three categories: adverse events (AEs); resistance to ICIs, and rechallenge becomes compulsive because of tumor relapse while the patients had completed a 2 year course of immunotherapy. However, these categories are still controversial and should be explored further. Through voting at the 6th Straits Summit Forum on Lung Cancer, in this study we summarize the consensus of 147 experts in ICI rechallenges. A total of 97.74% experts agreed to rechallenge; 48.87% experts rechallenge with the original drug, and the others rechallenge with a different drug; 40.3% agreed to rechallenge directly after progression; 88.06% experts agreed to ICI rechallenge with a combination regimen; and factors such as previous performance status score, PD‐1 expression, and age should also be considered. Understanding the the clinical studies in ICI rechallenge could bring us one step closer to understanding the consensus. In patients with advanced NSCLC who have suffered recurrent or distant metastasis after immunotherapy, the option of rechallenge with ICIs is a promising treatment option.

Microtubule‐targeting agents represent one of the most successful classes of anticancer agents. However, the development of drug resistance and the appearance of adverse effects hamper their clinical implementation. Novel microtubule‐targeting agents without such limitations are urgently needed. By employing a gene expression‐based drug repositioning strategy, this study identifies VU‐0365114, originally synthesized as a positive allosteric modulator of human muscarinic acetylcholine receptor M5 (M5 mAChR), as a novel type of tubulin inhibitor by destabilizing microtubules. VU‐0365114 exhibits a broad‐spectrum in vitro anticancer activity, especially in colorectal cancer cells. A tumor xenograft study in nude mice shows that VU‐0365114 slowed the in vivo colorectal tumor growth. The anticancer activity of VU‐0365114 is not related to its original target, M5 mAChR. In addition, VU‐0365114 does not serve as a substrate of multidrug resistance (MDR) proteins, and thus, it can overcome MDR. Furthermore, a kinome analysis shows that VU‐0365114 did not exhibit other significant off‐target effects. Taken together, our study suggests that VU‐0365114 primarily targets microtubules, offering potential for repurposing in cancer treatment, although more studies are needed before further drug development.

Discontinuous perfobond rib shear (PBL) connectors can be applied to fabricated steel-concrete composite beams. In this paper, six push-out specimens were designed to explore the shear performance of PBL under static and repeated loads. Four specimens were used for static push-out test. The remaining two were used for fatigue push-out test. The static load test mainly considers the influence of concrete at steel plate end and post-cast strip on the shear bearing capacity, initial shear stiffness and load-slip curve of discontinuous PBL shear connectors. On the basis of the test results, the calculating formulas of the shear capacity and shear stiffness of discontinuous PBL connectors considering the influence of end bearing, bond group effect and post-cast strip are proposed. The predicted results of the formula are close to the experiment. From the fatigue test results, it is found that the fatigue performance of above specimens with C50 and UHPC is better than that with studs, but the fatigue resistance of discontinuous PBL connectors with UHPC is not increased in proportion to the shear capacity. In addition, the residual slip and shear stiffness of PBL specimens after cyclic loading are related to fatigue load amplitude.

Abstract Highly multiplexed protein imaging is emerging as a potent technique for analyzing protein distribution within cells and tissues in their native context. However, existing cell annotation methods utilizing high-plex spatial proteomics data are resource intensive and necessitate iterative expert input, thereby constraining their scalability and practicality for extensive datasets. We introduce MAPS (Machine learning for Analysis of Proteomics in Spatial biology), a machine learning approach facilitating rapid and precise cell type identification with human-level accuracy from spatial proteomics data. Validated on multiple in-house and publicly available MIBI and CODEX datasets, MAPS outperforms current annotation techniques in terms of speed and accuracy, achieving pathologist-level precision even for typically challenging cell types, including tumor cells of immune origin. By democratizing rapidly deployable and scalable machine learning annotation, MAPS holds significant potential to expedite advances in tissue biology and disease comprehension.

Abstract Background The 5´ untranslated region (5´ UTR) plays a key role in regulating translation efficiency and mRNA stability, making it a favored target in genetic engineering and synthetic biology. A common feature found in the 5´ UTR is the poly-adenine (poly(A)) tract. However, the effect of 5´ UTR poly(A) on protein production remains controversial. Machine-learning models are powerful tools for explaining the complex contributions of features, but models incorporating features of 5´ UTR poly(A) are currently lacking. Thus, our goal is to construct such a model, using natural 5´ UTRs from Kluyveromyces marxianus, a promising cell factory for producing heterologous proteins. Results We constructed a mini-library consisting of 207 5´ UTRs harboring poly(A) and 34 5´ UTRs without poly(A) from K. marxianus. The effects of each 5´ UTR on the production of a GFP reporter were evaluated individually in vivo, and the resulting protein abundance spanned an approximately 450-fold range throughout. The data were used to train a multi-layer perceptron neural network (MLP-NN) model that incorporated the length and position of poly(A) as features. The model exhibited good performance in predicting protein abundance (average R2 = 0.7290). The model suggests that the length of poly(A) is negatively correlated with protein production, whereas poly(A) located between 10 and 30 nt upstream of the start codon (AUG) exhibits a weak positive effect on protein abundance. Using the model as guidance, the deletion or reduction of poly(A) upstream of 30 nt preceding AUG tended to improve the production of GFP and a feruloyl esterase. Deletions of poly(A) showed inconsistent effects on mRNA levels, suggesting that poly(A) represses protein production either with or without reducing mRNA levels. Conclusion The effects of poly(A) on protein production depend on its length and position. Integrating poly(A) features into machine-learning models improves simulation accuracy. Deleting or reducing poly(A) upstream of 30 nt preceding AUG tends to enhance protein production. This optimization strategy can be applied to enhance the yield of K. marxianus and other microbial cell factories.

Melanoma is rare in Taiwan. Asian melanoma is distinct from Western melanoma because acral and mucosal melanoma accounts for the majority of melanoma cases, leading to distinct tumor behaviors and genetic profiling. With consideration of the clinical guidelines in Western countries, Taiwanese experts developed a local clinical practice consensus guideline. This consensus includes diagnosis, staging, and surgical and systemic treatment, based only on clinical evidence, local epidemiology, and available resources evaluated by experts in Taiwan. This consensus emphasizes the importance of surgical management, particularly for sentinel lymph node biopsies. In addition, molecular testing for BRAF is mandatory for patients before systemic treatment. Furthermore, immunotherapy and targeted therapy are prioritized for systemic treatment. This consensus aimed to assist clinicians in Taiwan in diagnosing and treating patients according to available evidence.

Abstract The circadian clock regulator Bmal1 modulates tumorigenesis, but its reported effects are inconsistent. Here, we show that Bmal1 has a context-dependent role in mouse melanoma tumor growth. Loss of Bmal1 in YUMM2.1 or B16-F10 melanoma cells eliminates clock function and diminishes hypoxic gene expression and tumorigenesis, which could be rescued by ectopic expression of HIF1α in YUMM2.1 cells. By contrast, over-expressed wild-type or a transcriptionally inactive mutant Bmal1 non-canonically sequester myosin heavy chain 9 (Myh9) to increase MRTF-SRF activity and AP-1 transcriptional signature, and shift YUMM2.1 cells from a Sox10high to a Sox9high immune resistant, mesenchymal cell state that is found in human melanomas. Our work describes a link between Bmal1, Myh9, mouse melanoma cell plasticity, and tumor immunity. This connection may underlie cancer therapeutic resistance and underpin the link between the circadian clock, MRTF-SRF and the cytoskeleton.

In recent years, more and more technical tools have been introduced into various museums, bringing new opportunities and challenges for the research of museum learning. Whether the application of these technologies can effectively improve the effects of museum learning has become a major problem perplexing museum learning researchers. In this paper, a meta-analysis method is used to analyze and evaluate the 42 experimental or quasi-experimental studies on the technology application in the field of museum learning in the past decade. The results show that the technology application has a moderate or higher effect on museum learning, moreover, the age of learners, the type of technology, the type of museum and the visiting pattern of museum will significantly affect the effectiveness of technology on the museum learning. Therefore, the museum institutions can learn from excellent cases to accelerate the application of museum technology, give priority to the construction of digital resources, continue to promote the specialisation of museum technology application, and better play the role of technology in museum education.

Gradient refractive index (GRIN) metamaterial lenses can achieve multi-band fusion infrared imaging and has the characteristics of integration, light weight, and achromaticity, compared with existing curved refractive lenses group. Constructing a three-dimensional (3D) GRIN lens for dual-band (3.0–5.0 and 7.5–9.2 µm) mid-infrared imaging is challenging due to the difficulty of fabricating the desired 3D GRIN materials with continuously changing linewidths. Here, we present a 3D self-focusing GRIN lens with a cylindrical symmetry configuration in the mid-infrared band. Such a 3D GRIN lens is designed with gradient woodpile polymer metamaterials based on effective medium theory and fabricated with high fidelity by grayscale two-photon lithography. Simulated and experimental results simultaneously exhibit a 3D GRIN lens possessing dual-band, achromatic, near-diffraction-limit focusing on the wavelengths of 4.5 and 7.5 µm. The protocol for developing the 3D GRIN lens with dual-band fusion imaging would prompt potential applications in integrated light-coupled devices and lightweight infrared imaging devices.

Maternal immunity plays a vital role in protecting offspring from pathogen invasion, and it is present in nearly all vertebrates. As the most ancient vertebrates, teleost fish continuously secrete a substantial amount of mucus due to their unique living environment, and often exhibit schooling behavior. Previous studies have highlighted the presence of numerous immune components in the mucus of fish, that plays a crucial role in resisting pathogens. We hypothesize that this represents a distinctive maternal immune response mechanism in fish. In this study, we established a dextran sulfate sodium (DSS)-induced injury model by immersing zebrafish larvae in 0.5% DSS. This elicited robust inflammatory responses and showed DSS-induced alterations in microbial abundance via 16S rRNA sequencing. These changes included the disruption of normal gut structure; an increase in goblet cells; increased expression of inflammatory cytokines; and infiltration of neutrophils and macrophages into the gut, as well as an increase in pathogenic bacteria and a decline in probiotic bacteria. Furthermore, we conducted a mucus protection test by adding adult zebrafish mucus in fish tank water containing 0.5% DSS to test our hypothesis. As suspected, the mucus from adult zebrafish exhibited a protective role in defending zebrafish larvae against DSS-induced enteritis by alleviating excessive inflammatory responses and increasing probiotic abundance. In conclusion, our results confirm a significant role of adult fish mucus in the immune response of teleost fish to DSS-induced inflammation. Collectively, our findings show that the mucus of adult zebrafish may represent a novel form of maternal immunization, playing roles analogous to mammalian milk in immune regulatory functions.

Head and neck squamous cell carcinoma (HNSCC) is a globally prevalent and lethal cancer form, whose precise mechanisms remain incompletely understood. Increasing evidence suggests that N6-methyladenosine (m6A) plays a crucial role in cancer progression. This study aimed to explore the biological function of m6A modification and vir-like m6A methyltransferase associated (VIRMA) in HNSCC. We conducted an analysis of VIRMA expression in HNSCC cells using The Cancer Genome Atlas (TCGA) database and employed reverse transcription quantitative polymerase chain reaction (RT-qPCR) and western blotting to assess its expression levels in HNSCC cell lines. Additionally, m6A levels in HNSCC cells were quantified, and the correlation between VIRMA expression levels and the clinical and pathological features of other genes was analyzed. Upon knocking down VIRMA levels, we assessed HNSCC cell proliferation, migration, and invasion and validated downstream genes using RT-qPCR and western blot. Our findings suggested that VIRMA, as an m6A-related regulator, may significantly influence HNSCC progression by regulating ubiquitin protein ligase E3 component N-recognin 5 (UBR5) through m6A modification. Therefore, VIRMA may serve as a prognostic biomarker.

Study designRetrospective cohort study.ObjectiveThis study aims to report the incidence, risk factors, and recovery rate of neurological complications (NC) in patients with adult cervical deformity (ACD) who underwent corrective surgery.MethodsACD patients undergoing surgery from 2013 to 2015 were enrolled in a prospective, multicenter database. Patients were separated into 2 groups according to the presence of neurological complications (NC vs no-NC groups). The types, timing, recovery patterns, and interventions for NC were recorded. Patients' demographics, surgical details, radiographic parameters, and health-related quality of life (HRQOL) scores were compared.Results106 patients were prospectively included. Average age was 60.8 years with a mean of 18.2 months follow-up. The overall incidence of NC was 18.9%; of these, 68.1% were major complications. Nerve root motor deficit was the most common complication, followed by radiculopathy, sensory deficit, and spinal cord injury. The proportion of complications occurring within 30 days of surgery was 54.5%. The recovery rate from neurological complication was high (90.9%), with most of the recoveries occurring within 6 months and continuing even after 12 months. Only 2 patients (1.9%) had continuous neurological complication. No demographic or preoperative radiographic risk factors could be identified, and anterior corpectomy and posterior foraminotomy were found to be performed less in the NC group. The final HRQOL outcome was not significantly different between the 2 groups.ConclusionsOur data is valuable to surgeons and patients to better understand the neurological complications before performing or undergoing complex cervical deformity surgery.
