Colorectal cancer (CRC) is the third most common malignancy worldwide. It is well known that lipid metabolism reprogramming contributes to the tumor progression. However, the lipid metabolic alterations and potential remodeling mechanism underlying the chemoresistance of CRC remain largely unclear. In this study, we compared the gene expression profiles of chemoresistant versus control CRC cells from the GEO database and identified a key factor, Glycerol‐3‐phosphate acyltransferase 3 (GPAT3), that promotes lipid droplet (LD) production and confers chemoresistance of CRC. With applying of HPLC–MS and molecular dynamics simulation, we also demonstrated that the activity of lysophosphatidic acid synthesis by GPAT3 was dependent on its acetylation at K316 site. In particular, GPAT3‐mediated LD accumulation inhibited immunogenic cell death of tumor, and thus facilitated CD8+ T‐cell exhaustion and malignant progression in mouse xenografts and hepatic‐metastasis tumors in CRC patients. High GPAT3 expression turned CRC cells into nonimmunogenic cells after (Oxaliplatin) Oxa treatment, which was supported by a decrease in cytotoxic IFN‐γ release and CD8+ T‐cell exhaustion. In conclusion, these findings revealed the role of GPAT3‐associated LD accumulation, which conferred a malignant phenotype (chemoresistance) and regulated the tumor microenvironment of CRC. These results suggest that GPAT3 is a potential target to enhance CRC chemosensitivity and develop novel therapeutic interventions. [ABSTRACT FROM AUTHOR]

Tuning the coordination structures of metal sites is intensively studied to improve the performances of single‐atom site catalysts (SASC). However, the pore structure of SASC, which is highly related to the accessibility of active sites, has received little attention. In this work, single‐atom ZnN4 sites embedded in P‐functionalized carbon with hollow‐wall and 3D ordered macroporous structure (denoted as H‐3DOM‐ZnN4/P‐C) are constructed. The creation of hollow walls in ordered macroporous structures can largely increase the external surface area to expose more active sites. The introduction of adjacent P atoms can optimize the electronic structure of ZnN4 sites through long‐rang regulation to enhance the intrinsic activity and selectivity. In the electrochemical CO2 reduction reaction, H‐3DOM‐ZnN4/P‐C exhibits high CO Faradaic efficiency over 90% in a wide potential window (500 mV) and a large turnover frequency up to 7.8 × 104 h−1 at −1.0 V versus reversible hydrogen electrode, much higher than its counterparts without the hierarchically ordered structure or P‐functionalization. This paper proposes an ultra high frequency fatigue experimental approach based on electrodynamic shaker. The ultra high frequency fatigue specimen of TA11 titanium alloy sheet was designed after FEM calculation. The actual testing frequency of the designed specimen could be reached as high as 1756 Hz. The fatigue tests of both the ultra high frequency specimen and standard vibration fatigue specimen were carried out under the same stress level, and the consistency of test results was statistically verified. The results show that under the same stress condition,the testing results of the high frequency fatigue specimen proposed in this study are in good agreement with the experimental results of standard vibration fatigue specimen. Furthermore, the comparison of the present experimental results with the axial-loading fatigue and rotating bending fatigue results for the same material is conducted,and also a good consistency is obtained.

This paper studies the thermoelastic dynamic response of a simply supported beam under movable temporally non-Gaussian laser pulse. The heat conduction equation of the problem was solved by using the method of separation of variables. The Green's function for the forth-order partial differential equation was derived and was used to solve the vibration equation of a heated beam. The temperature, deflection, and strain of the beam were derived analytically and their variations with time and space were illustrated. The influences of moving speed and duration time of the laser pulse on the dynamic responses were also analyzed. It was interesting to observe the propagation of strain wave along the beam axis during the irradiation period of the laser pulse. Nous etudions ici la reponse dynamique thermo-elastique d'une poutre cantilever (a support simple) sous l'effet d'une impulsion laser deplacable temporellement non gaussienne. Nous solutionnons par separation des variables l'equation de conduction de la chaleur du probleme. Nous derivons la fonction de Green pour l'equation differentielle du quatrieme ordre et l'utilisons pour solutionner l'equation de vibration du cantilever chauffe. La temperature, la deflexion et la deformation du cantilever sont determinees analytiquement et nous illustrons leurs variations spatiale et temporelle. Nous analysons egalement l'influence de la vitesse de deplacement et de la duree de l'impulsion sur les reponses dynamiques de la poutre cantilever. Il est interessant d'observer la propagation de l'onde de deformation le long de l'axe de la poutre pendant l'irradiation par l'impulsion laser.
