Abstract

China’s first Mars rover, Zhurong, has successfully touched down on the southern Utopia Planitia of Mars at 109.925° E, 25.066° N, and since performed cooperative multiscale investigations with the Tianwen-1 orbiter. Here we present primary localization and surface characterization results based on complementary data of the first 60 sols. The Zhurong rover has traversed 450.9 m southwards over a flat surface with mild wheel slippage (less than 0.2 in slip ratio). The encountered crescent-shaped sand dune indicates a NE-SW local wind direction, consistent with larger-range remote-sensing observations. Soil parameter analysis based on terramechanics indicates that the topsoil has high bearing strength and cohesion, and its equivalent stiffness and internal friction angle are ~1390-5872 kPa∙m-n and ~21°-34° respectively. Rocks observed strewn with dense pits, or showing layered and flaky structures, are presumed to be involved in physical weathering like severe wind erosion and potential chemical weathering processes. These preliminary observations suggest great potential of in-situ investigations by the scientific payload suite of the Zhurong rover in obtaining new clues of the region’s aeolian and aqueous history. Cooperative investigations using the related payloads on both the rover and the obiter could peek into the habitability evolution of the northern lowlands on Mars.

Abstract

To survey the virulence gene carrying status, drug sensitivity, and genetic characteristics of Vibrio parahaemolyticus, 26 seafood samples from three canteens in the Beidaihe area, as well as self-collected samples, were analyzed in August 2021. Four major virulence genes (tdh, trh, tlh, and toxR) were screened from ten strains of V. parahaemolyticus using PCR and the BD automatic microbial identification and drug susceptibility analysis system. Next-generation genome sequencing was performed on the isolates. A phylogenetic tree, drug resistance genes, and virulence genes were constructed based on core SNP sites. The gene carrying rates of tlh, toxR, were 100.00%, 100.00%; none of the isolates carried tdh or trh. Ten isolates were resistant to aztreonam and moderately sensitive to ampicillin. All the isolates contained the β-lactam resistance gene (CARB-18), corresponding to the resistance phenotype. One isolate was found to contain mexE, a virulence gene rarely reported in similar studies. Nine sequence types were identified. Several V . parahaemolyticus isolates exhibited high homology with local and foreign marine organism isolates. Local V. parahaemolyticus may pose a threat to human health, and its sources are complex and highly genetically diverse. Abstract

Background: With the rapid emergence of plasmid mediated polymyxin resistance gene mcr-1, multidrug-resistant Salmonella caused great troubles in clinical treatment and attracted extensive attention. Here we report multidrug-resistant Salmonella strains harboring mcr-1 in China, which are resistant to both polymyxin, traditional antibiotics and even clinically wide-used antibiotics. Methods: We screened 1454 strains of Salmonella collected in our laboratory from 2006 to 2018 from 3 provinces or regions for mcr-1 by PCR. Antimicrobial susceptibility testing was determined. Plasmid conjugation assays were carried out to analysis the transferability of polymyxin resistance. Genetic polymorphism analysis of Salmonella was performed using the PFGE, and the plasmid profiles were characterized by S1-PFGE and southern blotting. The plasmids harboring mcr-1 were sequenced and compared. Results: Eleven S. Typhimurium isolates harboring mcr-1 with polymyxin resistance (MICs 4μg/ml) were identified from intestinal infections and foods in China. All S. Typhimurium isolates were multidrug-resistant to traditional antibiotics and even clinically wide-used antibiotics. Three types of plasmids harboring mcr-1 were recovered (IncHI2, IncX4 and IncI2). Compared with the reference plasmid, IncX4 and IncI2 plasmids had extremely similar typical backbone, and contain only mcr-1 resistance gene. However, IncHI2 were the most diverse type of plasmid due to containing a large MDR region, including blaCTX-M, oqxB, sul, aph, aadA and blaTEM. IncHI2 plasmids were observed to contain only one or no insertion sequence ISApl1 around mcr-1, without forming a circular intermediate. Conclusion: With the horizontal transfer of different types of plasmids, mcr-1 is widely spread worldwide. These prevalent plasmids are responsible for resistance to polymyxin, traditional antibiotics and even clinically wide-used antibiotics resulting from transmission of mcr-1 and other resistance genes. Our studying emphasizes the necessity to jointly monitor its international epidemic and preemptive further upgrade.

Abstract

Three new transition metal complexes, [Zn(L1)2(OTs)(CH3OH)](TsO)·CH3OH (1), {[Zn(L2)(H2O)4](SiF6)·H2O}n (2) and {[Cd(L2)2(OTs)2]·2DMF}n (3), were prepared from pyridine thiazolone derivatives (where L1 = 2-[4-(2-pyridinyl)-2-thiazolyl]hydrazone, L2 = 2-[4-(3-pyridinyl)-2-thiazolyl]hydrazone) and characterized by 1H-NMR, elemental analysis (EA), and infrared spectra (IR). The crystalline behavior of L1 and 1–3 were analyzed by X-ray diffraction. In the structure of 1, L1 acts as a bidentate agent coordinating the Zn2+ ion through the pyridine nitrogen and thiazole nitrogen, while L2 bonded in a 2D structure by coordinating Zn2+ through the nitrogen atoms of two pyridine rings in the structure of 2. Additionally, with Cd2+ ions as connection points and the spatial extension of L2, 3 possesses an intricate 3D metal–organic framework. The luminescent properties for the ligands and complexes were investigated. The in vitro antimicrobial and antitumor activities were evaluated against ten pathogenic bacterial strains and four human cancer cell lines.

125. Feasibility of Wrist-Worn, Cancelable, Real-Time Biometric Authentication via HD-sEMG and Dynamic Gestures

Abstract

In recent years, high-density surface electromyography (HD-sEMG) has been applied for cancelable authentication of wearable devices used in healthcare applications. However, previous studies focused on the forearm or hand, whereas the wrist is more comfortable for practical use. Covering a larger area with fewer electrodes (16 channels on the wrist versus 256 channels on the forearm) also reduces the chance of electrode corruption and improves the stability of the authentication system. In addition, fast dynamic gestures are more desirable than static gestures for real-time authentication. Shorter gesture execution time also makes dynamic gesture codes harder to record by imposters. We propose a cancelable real-time biometric system via wrist-worn HD-sEMG and dynamic gestures. A wearable wristband was designed to collect HD-sEMG signals. Based on a dataset with 12 dynamic gestures, we realized a fast real-time authentication. It achieved an equal error rate (EER) of 0.0039 even when imposters entered a correct gesture code. The cancelability was confirmed with an EER of 0.0018. The real-time authentication with an authentication accuracy of 96.7% and a low latency of 64 ms makes it practical in healthcare applications. These results demonstrate the feasibility of wrist-worn biometrics, which allows secure healthcare applications without compromising the user experience.

Abstract

In multiregional randomized clinical trials (MRCTs), determining the regional treatment effect of a new treatment over an existing one is important to both the sponsor and related regulatory agencies. Also of particular interest is to test the null hypothesis that the treatment benefit is the same among all the regions. Existing methods are mainly for continuous endpoint and use parametric models, which are not robust. MRCTs are known for facing increased variation and heterogeneity and a robust model for its design and analysis would be desirable. We consider clinical trials with a binary primary endpoint and propose a robust semiparametric logistic model which has a known parametric and an unknown nonparametric component. The parametric component represents our prior knowledge about the model, and the nonparametric part reflects uncertainty. Compared to the classic logistic model for this problem, the proposed model has the following advantages: robust to model assumption, more flexible and accurate to model the relationship between the response and covariates, and possibly more accurate parameter estimates. The model parameters are estimated by profile maximum likelihood approach, and the null hypothesis of regional treatment difference being the same is tested by the profile likelihood ratio statistic. Asymptotic properties of the estimates are derived. Simulation studies are conducted to evaluate the performance of the proposed model, which demonstrated clear advantages over the classic logistic model. Abstract

Right ventricular (RV) structure and function influence the morbidity and mortality from coronary artery disease (CAD), dilated cardiomyopathy (DCM), pulmonary hypertension and heart failure. Little is known about the genetic basis of RV measurements. Here we perform genome-wide association analyses of four clinically relevant RV phenotypes (RV end-diastolic volume, RV end-systolic volume, RV stroke volume, RV ejection fraction) from cardiovascular magnetic resonance images, using a state-of-the-art deep learning algorithm in 29,506 UK Biobank participants. We identify 25 unique loci associated with at least one RV phenotype at P< 2.27 ×10−8, 17 of which are validated in a combined meta-analysis (n= 41,830). Several candidate genes overlap with Mendelian cardiomyopathy genes and are involved in cardiac muscle contraction and cellular adhesion. The RV polygenic risk scores (PRSs) are associated with DCM and CAD. The findings substantially advance our understanding of the genetic underpinning of RV measurements.

Abstract

Human adenovirus (HAdV) infection is a major cause of respiratory disease, yet no antiviral drugs have been approved for its treatment. Herein, we evaluated the antiviral and anti-inflammatory effects of cyclin-dependent protein kinase (CDK) inhibitor indirubin-3′-monoxime (IM) against HAdV infection in cells and a transgenic mouse model. After evaluating its cytotoxicity, cytopathic effect reduction, antiviral replication kinetics, and viral yield reduction assays were performed to assess the anti-HAdV activity of IM. Quantitative real-time polymerase chain reaction (qPCR), quantitative reverse transcription PCR (qRT-PCR), and western blotting were used to assess the effects of IM on HAdV DNA replication, transcription, and protein expression, respectively. IM significantly inhibited HAdV DNA replication as well as E1A and Hexon transcription, in addition to significantly suppressing the phosphorylation of the RNA polymerase II C-terminal domain (CTD). IM mitigated body weight loss, reduced viral burden, and lung injury, decreasing cytokine and chemokine secretion to a greater extent than cidofovir. Altogether, IM inhibits HAdV replication by downregulating CTD phosphorylation to suppress viral infection and corresponding innate immune reactions as a promising therapeutic agent. Abstract

Abstract Background Enterovirus 71 (EV71), one of the most important neurotropic EVs, has caused death and long-term neurological sequelae in hundreds of thousands of young children in the Asia-Pacific region in the past decade. The neurological diseases are attributed to infection by EV71 inducing an extensive peripheral and central nervous system (CNS) inflammatory response with abnormal cytokine production and lymphocyte depletion induced by EV71 infection. In the absence of specific antiviral agents or vaccines, an effective immunosuppressive strategy would be valuable to alleviate the severity of the local inflammation induced by EV71 infection. Presentation of the hypothesis The complement system plays a pivotal role in the inflammatory response. Inappropriate or excessive activation of the complement system results in a severe inflammatory reaction or numerous pathological injuries. Previous studies have revealed that EV71 infection can induce complement activation and an inflammatory response of the CNS. CR2-targeted complement inhibition has been proved to be a potential therapeutic strategy for many diseases, such as influenza virus-induced lung tissue injury, postischemic cerebral injury and spinal cord injury. In this paper, a mouse model is proposed to test whether a recombinant fusion protein consisting of CR2 and a region of Crry (CR2-Crry) is able to specifically inhibit the local complement activation induced by EV71 infection, and to observe whether this treatment strategy can alleviate or even cure the neurogenic inflammation. Testing the hypothesis CR2-Crry is expressed in CHO cells, and its biological activity is determined by complement inhibition assays. 7-day-old ICR mice are inoculated intracranially with EV71 to duplicate the neurological symptoms. The mice are then divided into two groups, in one of which the mice are treated with CR2-Crry targeted complement inhibitor, and in the other with phosphate-buffered saline. A group of mice deficient in complement C3, the breakdown products of which bind to CR2, are also infected with EV71 virus. The potential bioavailability and efficacy of the targeted complement inhibitor are evaluated by histology, immunofluorescence staining and radiolabeling. Implications of the hypothesis CR2-Crry-mediated targeting complement inhibition will alleviate the local inflammation and provide an effective treatment for the severe neurological diseases associated with EV71 infection.

150. Rapid Identification and Source Tracing of a SalmonellaTyphimurium Outbreak in China by Metagenomic and Whole-Genome Sequencing

Abstract

Salmonellaspp. are among the most prevalent foodborne pathogens. Rapid identification of etiologic agents during foodborne outbreaks is of great importance. In this study, we report a traceback investigation of a Salmonellaoutbreak in China. Metagenomic sequencing of suspected food samples was performed on MinION and MiSeq platforms. Real-time nanopore sequencing analysis identified reads belonging to the Enterobacteriaceaefamily. MiSeq sequencing identified 63 reads specifically mapped to Salmonella. Conventional methods including quantitative-PCR and culture-based isolation confirmed as Salmonella entericaserovar Typhimurium. The foodborne outbreak of SalmonellaTyphimurium was further recognized by whole-genome sequencing and pulsed-field gel electrophoresis analysis. Our study demonstrates the ability of metagenomic sequencing to rapidly identify enteric pathogens directly from food samples. These results highlight the capacity of metagenomic sequencing to deliver actionable information rapidly and to expedite the tracing and identification of etiologic agents during foodborne outbreaks.
