103. Myofibroblasts acquire retinoic acid–producing ability during fibroblast-to-myofibroblast transition following kidney injury

Abstract

Tubular injury and interstitial fibrosis are the hallmarks of chronic kidney disease. While recent studies have verified that proximal tubular injury triggers interstitial fibrosis, the impact of fibrosis on tubular injury and regeneration remains poorly understood. We generated a novel mouse model expressing diphtheria toxin receptor on renal fibroblasts to allow for the selective disruption of renal fibroblast function. Administration of diphtheria toxin induced upregulation of the tubular injury marker Ngal and caused tubular proliferation in healthy kidneys, whereas administration of diphtheria toxin attenuated tubular regeneration in fibrotic kidneys. Microarray analysis revealed down-regulation of the retinol biosynthesis pathway in diphtheria toxin-treated kidneys. Healthy proximal tubules expressed retinaldehyde dehydrogenase 2 (RALDH2), a rate-limiting enzyme in retinoic acid biosynthesis. After injury, proximal tubules lost RALDH2 expression, whereas renal fibroblasts acquired strong expression of RALDH2 during the transition to myofibroblasts in several models of kidney injury. The retinoic acid receptor (RAR) RARγ was expressed in proximal tubules both with and without injury, and αB-crystallin, the product of an RAR target gene, was strongly expressed in proximal tubules after injury. Furthermore, BMS493, an inverse agonist of RARs, significantly attenuated tubular proliferation in vitro. In human biopsy tissue from patients with IgA nephropathy, detection of RALDH2 in the interstitium correlated with older age and lower kidney function. These results suggest a role of retinoic acid signaling and cross-talk between fibroblasts and tubular epithelial cells during tubular injury and regeneration, and may suggest a beneficial effect of fibrosis in the early response to injury., 腎障害における線維化の正の側面の発見 --線維化が腎臓を修復する--. 京都大学プレスリリース. 2019-01-18.

Abstract

This paper reports loading tests conducted to investigate the effect of tri-directional seismic force on the shear capacity of reinforced concrete (RC) shear walls. The experimental parameters were the magnitude of out-of-plane deformation and axial load condition. All specimens reached the in-plane maximum load capacity at approximately Rx = 0.50%. The maximum value of the average shear stress divided by the tensile strength of concrete decreased by 12.1 to 18.2% with increasing difference between axial compressive force and tensile force. The minimum safety margin was 1.07 attained for the specimen subjected to out-of-plane lateral load and varying axial load acting simultaneously, while that of the specimen subjected to only in-plane lateral load under constant axial load was 1.50. The safety margin was defined as the ratio of the maximum load capacity obtained experimentally to the calculated capacity. [ABSTRACT FROM AUTHOR]

114. Ostwald ripening under temperature gradient: A phase-field study

Abstract

Ostwald ripening under a temperature gradient in binary model alloys is investigated using a quantitative phase-field model. The simulations show that a cube of average radius of a second-phase particle is proportional to time, and the particle size distribution shows self-similarity in a steady state, as with a uniform temperature field. It is found that the growth rate of particles under a temperature gradient is faster than that in the isothermal case, and the steady-state particle size distribution depends on the magnitude of the temperature gradient. Furthermore, the second-phase particles migrate from low temperature regions to high temperature regions when a non-uniform temperature field is applied. The migration velocity of particles, averaged over the whole system, increases with the magnitude of the temperature gradient. On the other hand, the velocity of each particle is not relevant to particle size. Hence, the particle migration is entirely ascribed to the diffusion flux driven by the concentration gradient originating from the temperature dependence of solute solubility.

Abstract

High ZT value and large Seebeck coefficient have been reported in the nanostructured Fe-doped Si-Ge alloys. In this work, the large Seebeck coefficient in Fe-doped Si-Ge systems was qualitatively reproduced from the computed electronic density of states, where a hybrid functional, HSE06, was used for an exchange-correlation functional, as well as a special quasi-random structure (SQS) for a disordered atomic configuration. Furthermore, by replacing Fe with other transition metals, such as Mn, Co, Ni, Cu, Zn, and Au, a dopant that produces a large Seebeck coefficient in Si-Ge alloy systems was explored. It was found that the Mn-doped system produces a large Seebeck coefficient comparable with the Fe-doped system.

116. Comparison of the prognosis of the remaining teeth between implant-supported fixed prostheses and removable partial dentures in partially edentulous patients : A retrospective study

Abstract

Background There have been several reports about the prognosis of teeth adjacent to edentulous spaces for implant-supported fixed prostheses (ISFPs) and removable partial dentures (RPDs). However, there are few reports about the prognosis of the other remaining teeth comparing ISFPs with RPDs. Purpose The aim of this study was to evaluate and compare the prognosis of the remaining teeth for ISFPs and RPDs in terms of survival and complication-free rates. Methods Subjects were partially edentulous patients with ISFPs or RPDs inserted in 2003-2016. Teeth adjacent to edentulous spaces (A-teeth), teeth not adjacent to edentulous spaces (R-teeth), and teeth opposing edentulous spaces (O-teeth) were investigated. The endpoints were tooth extraction and complications. A multivariate cox regression model was used to estimate the risk factors for survival of the investigated teeth. Results A total of 233 (ISFP: 89, RPD: 144) patients were included in the statistical analyses. An IFSP prosthesis, when compared to an RPD prosthesis did not significantly decrease the tooth loss rate for A-teeth (hazard ratio [HR]: 0.76; 95% confidence interval [CI]: 0.30-1.92), for R-teeth (HR: 0.54; 95% CI: 0.28-1.05), or for O-teeth (HR: 0.45; 95% CI: 0.10-2.09). Conclusions In partially edentulous spaces, the difference between ISFPs and RPDs does not affect the prognosis of teeth adjacent to edentulous spaces, teeth not adjacent to edentulous spaces, and teeth opposing edentulous spaces. Namely, our findings suggest that it depends largely on the tooth type, jaw, endodontic therapy performed, not on the type of prostheses.
