Bacteriol. 89:1291-1298. 1965.-The highly immunogenic particulate fraction obtained from mechanically ruptured cells of the H37Ra strain of Mycobacterium tuberculosis was suspended and centrifuged at 20,360 x g. The supernatant liquid from this centrifugation was centrifuged at 56,550 x g to remove the larger particles, and the supernatant liquid from this was centrifuged at 144,000 x g to obtain a ribosomal fraction. The sediments from the first two centrifugations were highly immunogenic, but the ribosomal fraction showed only slight capacity to immunize mice. However, when the ribosomal fraction was mixed with Freund's incomplete adjuvant, the immunogenic activity was equivalent to the particulate fraction from which it was prepared. To test the hypothesis that some membranous substance in the particulate fraction was acting as an adjuvant for the smaller particles in the ribosomal fraction, portions of the particulate fraction were treated separately with each of the membrane-disrupting agents, sodium deoxycholate, sodium lauryl sulfate, and 1 m sodium chloride. The treated materials were then centrifuged at 144,000 x g, and the sediments were tested for immunogenicity both with and without the addition of Freund's incomplete adjuvant. Without the adjuvant, the immunizing activities were very weak or absent; with the adjuvant, they were equivalent to that of the particulate fraction from which they were prepared. Other factors which have been found to damage or destroy membranes, such as freezing and thawing, and heat, also significantly decreased the immunogenic activity of the particulate fraction unless it was incorporated into Freund's incomplete adjuvant. The larger particles which sedimented at 56,550 x g were also treated with sodium lauryl sulfate and sodium chloride. Again, immunogenicity was greatly reduced but was fully restored by use of Freund's incomplete adjuvant. The data suggest, then, that the immunizing component of the particulate fraction is a substance (ribosomal?) which sediments at 144,000 x g, but for maximal immunizing activity a labile, possibly membranous, moiety of the mycobacterial cell, which has the properties of an adjuvant, is required.

When mice immunized with Listeria monocytogenes were given a second injection of listeria, they showed an anamnestic immune response to intravenous challenge with listeria, as measured by enumeration of the viable infecting organisms in the spleens of the infected animals. This response was independent of the effects of the challenge dose. When mice immunized with living or heat-killed attenuated mycobacterial cells were boosted with living H37Ra, there was also an accelerated response to listeria challenge. The response was greater in the mice given the primary immunization with living cells than in those immunized with heat-killed cells. The response to listeria challenge in mice immunized and boosted with mycobacteria was of less magnitude than that in the mice immunized and boosted with listeria. Growth of listeria in the mice immunized and boosted with mycobacteria was retarded only during the first 2 days of the infection, whereas the infecting listeria in mice immunized and boosted with listeria were permanently inactivated. Mice immunized with mycobacterial ribosomal fraction and restimulated with living mycobacterial cells showed no accelerated response to listeria challenge. It is evident from these results that resistance to these organisms is specifically evoked, but that once evoked it is not completely nonspecific in action. Also, the resistance produced by the mycobacterial ribosomal fraction to challenge with mycobacteria is completely specific in action. Therefore, it has been shown that there are two mechanisms involved in acquired immunity to facultative, intracellular parasites. One is nonspecific and mediated by activated macrophages. The other is specific and mediated by a mechanism as yet unknown.

We have shown that splenic lymphocytes from mice immunized with viable attenuated mycobacterial cells (H37Ra) or ribonucleic acid prepared from these cells will bring about inhibition of multiplication of virulent tubercle bacilli (H37Rv) within normal mouse peritoneal macrophages in tissue culture. Apparently these lymphocytes, when stimulated with viable virulent tubercle bacilli, elaborate a filterable substance(s) which is responsible for this intracellular growth inhibitory effect.

Youmans, Guy P. (Northwestern University Medical School, Chicago, Ill.), and Anne S. Youmans. Nonspecific factors in resistance of mice to experimental tuberculosis. J. Bacteriol. 90:1675-1681. 1965.-In contrast to viable attenuated mycobacterial cells, Escherichia coli lipopolysaccharide (LPS) did not produce an acute pulmonary granulomatous response in mice, did not decrease the tolerance of mice to early subsequent intravenous injections of viable attenuated mycobacterial cells, nor did it increase susceptibility to tuberculous infection when injected simultaneously with virulent mycobacterial cells. When the injection of E. coli LPS was followed by the intravenous injection of virulent mycobacterial cells, there was a moderate increase in resistance to tuberculous infection which was maximal 7 to 14 days after the LPS injection. The degree of increased resistance to tuberculous infection was approximately the same as that produced by nearly maximal tolerated doses of heat-killed attenuated mycobacterial cells, and to that produced by a trichloroacetic acid extract of heat-killed attenuated mycobacterial cells. It is suggested that the major, if not entire, immunizing component of heat-killed attenuated mycobacterial cells resides in a heat-stable "nonspecific" component. A "multiple response" theory of immunity to tuberculosis is proposed.

The amount of ribonucleic acid (RNA), protein, and deoxyribonucleic acid (DNA) was determined in pellicle cultures of different ages of the H37Ra strain of Mycobacterium tuberculosis, grown on a synthetic medium. We found that the highest content of RNA and protein was present in 2-week-old cultures, indicating that these cells were in the logarithmic phase of growth. DNA content was highest at 1 and 2 weeks. The amount of all three compounds then decreased about 50% during the following 6 weeks. Two-week-old cells should therefore be used for preparation of the immunogenic ribosomal fraction. The optimal concentration of zinc chloride increased RNA and protein synthesis, and also improved the appearance of the pellicle growth. Two-week-old cells, which contained the largest amount of RNA and protein, immunized mice significantly better than older cells. Since protein and DNA are not involved in the production of immunity, a correlation could be made between amount of RNA and the capacity of viable H37Ra cells to immunize mice. The immunizing capacity of these cells was not affected by ribonuclease, probably because the ribonuclease did not penetrate into the whole cells.

By following careful procedures, mycobacterial ribosomal fractions and ribonucleic acid (RNA) prepared by ethyl alcohol precipitation were obtained which have immunogenic activities similar to the viable attenuated H37Ra cells of Mycobacterium tuberculosis from which they were obtained. This comparison was based on the amount of ribonucleic acid (RNA) present. These preparations consisted of approximately 63% RNA and 37% protein; no deoxyribonucleic acid or polysaccharide was detected by chemical tests. A high correlation was found between the immunogenic activity of a preparation and the per cent increase in hyperchromicity at 260 nm of a ribonuclease-hydrolyzed portion. Final concentrations of sodium dodecyl sulfate higher than 0.25% when used for the preparation of the ribosomal fractions and RNA resulted in significantly lower immune responses and greater variation between experiments. This was not related to the amount of protein present. The stability of the ribosomal and RNA preparations was tested under a variety of conditions. The need for a good protective adjuvant again was shown since mouse serum readily hydrolyzed the RNA. Equal immunity was obtained after immunization by the intraperitoneal and subcutaneous routes; however, no immune response was obtained when the intravenous route was used. Preliminary results with RNA prepared with phenol showed that it was more easily degraded during preparation. This resulted in a lower immune response than was obtained with the RNA prepared with ethyl alcohol.

Youmans, Anne S. (Northwestern University Medical School, Chicago, Ill.), and Guy P. Youmans. Further studies on a labile immunogenic particulate substance isolated from Mycobacterium tuberculosis. J. Bacteriol. 87:278-285. 1964.-A particulate fraction which was highly immunogenic for mice was collected by ultracentrifugation from mycobacteria disrupted in 0.25 m sucrose buffer. The active immunogenic material was present in the gelatinous pellet obtained after centrifugation at 40,000 rev/min (144,000 x g) for 3 hr. This active material could be prepared free from whole cells and cell walls. This was done either by several centrifugations at lower speeds, or by filtering the supernatant fluid from the 10,000 rev/min centrifugation through a Millipore filter (porosity 0.5 mu). No microorganisms were found on slides or in cultures made from these filtrates. The immunogenic moiety in the particulate fraction was found to be very labile. Temperatures higher than 0 to 4 C inactivated the immunogenic activity. There was an irreversible linear decrease in activity as the temperature increased. If fractions were frozen or lyophilized, the activity remained as high as the original material for 4 weeks, and then rapidly decreased. The immunogenic material also was very sensitive to the hydrogen-ion concentration; the optimal activity was found at pH 6.8 to 7.0. The activity decreased rapidly at more acid or alkaline pH values. Also, particulate fraction prepared in sucrose buffer at pH 7.3 and 7.6 was much less active than that prepared in sucrose buffer at pH 7.0. Immunogenic activity was decreased if the particulate fraction was dialyzed overnight against 0.01 m phosphate buffer or distilled water at 4 C. The detergent sodium lauryl sulfate inactivated immunogenic activity. Moreover, the use of a Waring Blendor to blend the ruptured cell mass before centrifugation decreased the activity. Finally, a markedly lower activity resulted if both the 20,000 and 40,000 rev/min centrifugations were done the day after the rupture of the cells. Some refinements in technique which are used now in the preparation of the particulate fraction are detailed.
