The microbiota and its metabolites play an important role in regulating the host metabolism and immunity. However, the underlying mechanism is still not well studied. Thus, we conducted the LC-MS/MS analysis and RNA-seq analysis on Equus przewalskii with and without horse botfly infestation to determine the metabolites produced by intestinal microbiota in feces and differentially expressed genes (DEGs) related to the immune response in blood and attempted to link them together. The results showed that parasite infection could change the composition of microbial metabolites. These identified metabolites could be divided into six categories, including compounds with biological roles, bioactive peptides, endocrine-disrupting compounds, pesticides, phytochemical compounds, and lipids. The three pathways involving most metabolites were lipid metabolism, amino acid metabolism, and biosynthesis of other secondary metabolites. The significant differences between the host with and without parasites were shown in 31 metabolites with known functions, which were related to physiological activities of the host. For the gene analysis, we found that parasite infection could alarm the host immune response. The gene of “cathepsin W” involved in innate and adaptive immune responses was upregulated. The two genes of the following functions were downregulated: “protein S100-A8” and “protein S100-A9-like isoform X2” involved in chemokine and cytokine production, the toll-like receptor signaling pathway, and immune and inflammatory responses. GO and KEGG analyses showed that immune-related functions of defense response and Th17 cell differentiation had significant differences between the host with and without parasites, respectively. Last, the relationship between metabolites and genes was determined in this study. The purine metabolism and pyrimidine metabolism contained the most altered metabolites and DEGs, which mainly influenced the conversion of ATP, ADP, AMP, GTP, GMP, GDP, UTP, UDP, UMP, dTTP, dTDP, dTMP, and RNA. Thus, it could be concluded that parasitic infection can change the intestinal microbial metabolic activity and enhance immune response of the host through the pathway of purine and pyrimidine metabolism. This results will be a valuable contribution to understanding the bidirectional association of the parasite, intestinal microbiota, and host.

Naturally occurring polysaccharide-based polyelectrolyte complex (PEC) physical hydrogels have broad applications in biomedicine. Yet, obtaining high-strength, strong-adhesion, and antibacterial PEC hydrogels via physically crosslinking of natural polysaccharides still remains a challenge. In this work, a chitosan (CS)/sodium alginate (SA) PEC physical hydrogel film was developed by using two oppositely charged natural polysaccharides, CS and SA, of equivalently high molecular weight (Mw, approx. 105 Da), the tensile stress of which was achieved as high as 12.3 MPa due to CS-CS intermolecular hydrogen bonding and CS-SA electrostatic interaction. In addition, the hydrogel films, upon protonation at acidic conditions (e.g. pH 2.6), exhibited excellent antibacterial activity against Escherichia coli (E. coli) and the CS/SA hydrogel films also demonstrated strong adhesion to biological tissue. The high Mw-matching, two-component natural polysaccharide approach presented in this work to preparing high-strength, strong-adhesion, and antibacterial PEC hydrogels may shed some lights on converting renewable biomass to value-added biomaterials.

Even frequently used in wheat breeding, we still have an insufficient understanding of the biology of the products via distant hybridization. In this study, a transcriptomic analysis was performed for six Triticum aestivum-Thinopyrum elongatum substitution lines in comparison with the host plants. All the six disomic substitution lines showed much stronger “transcriptomic-shock” occurred on alien genomes with 57.43–69.22% genes changed expression level but less on the recipient genome (2.19–8.97%). Genome-wide suppression of alien genes along chromosomes was observed with a high proportion of downregulated genes (39.69–48.21%). Oppositely, the wheat recipient showed genome-wide compensation with more upregulated genes, occurring on all chromosomes but not limited to the homeologous groups. Moreover, strong co-upregulation of the orthologs between wheat and Thinopyrum sub-genomes was enriched in photosynthesis with predicted chloroplastic localization, which indicates that the compensation happened not only on wheat host genomes but also on alien genomes.

The objective of this study was to assess the effect of harvesting time on the fermentation characteristics, chemical composition, and microbial community of king grass silage. King grass was harvested at three growth periods of 90 days (KN90S), 110 days (KN110S), and 130 days (KN130S); chopped into 2–3-cm particle size; and ensiled in polyethylene bags (20 × 30 cm). The fermentation quality and chemical composition of silages were analyzed after 1, 3, 7, 14, 30, and 60 days of ensiling. Bacterial community of silage ensiled for 60 days was profiled using next generation sequencing (NGS) technology. The KN110S showed the most extensive lactic acid (LA) fermentation during 7 days of fermentation compared to KN90S and KN130S. After 60 days of fermentation, the KN110S showed the lowest pH and the highest lactic acid content among the three treatments. The butyric acid and ammonia nitrogen contents of KN90S and KN130S were significantly greater than those of KN110S (p < 0.05). After a timespan of 60 days of ensiling, the bacterial community of king grass silage was predominantly populated by Proteobacteria in phylum level, whereas unclassified Enterobacteriaceae genus remained dominant in all silages. A higher relative abundance of Clostridium was observed in KN90S and KN130S, but not in KN110S, and greater abundance of Lactococcus appeared in KN110S and KN130S silages than KN90S. It is concluded that harvesting time had an important effect on the fermentation quality and microbial community of king grass silage.

The outbreak of COVID-19 and the uncertainty it brings have created enormous pressure on governments to control the global pandemic and restore economic growth. It is an inevitable choice for governments of various countries to seek to control the pandemic and to provide support such as subsidies to people who lose their jobs or cannot work. However, governments should evaluate their pandemic policies to determine their effectiveness. To maintain social stability and help vulnerable groups, governments also must determine when subsidies are needed and when these support policies should be withdrawn. This research demonstrates that the administration of vaccines and the wearing of masks have a relatively limited impact on preventing the spread of the COVID-19 virus. By contrast, strict school closure policies combined with personal movement restrictions are more helpful in mitigating the spread of the virus. Compared with vaccine policies and wearing masks, controlling internal movement is the most effective way to manage the pandemic in schools. Additionally, economic support such as subsidies for the unemployed and underemployed is not only conducive to prevention of the virus' spread but also to economic recovery and social stability. When the pandemic is brought under control, economic support for vulnerable groups can be gradually reduced or even withdrawn.

Abstract Background Candida is the common conditionally pathogenic fungus that infected human and animal clinically. C. tropicalis had been isolated from the skin and hair of healthy pigs, but with no report of fatal infection in gastrointestinal diseases. Case presentation In a pig farm in Henan Province of China, about 20 % of pregnant and postpartum sows suffered from severe gastrointestinal diseases, with a mortality rate higher than 60 % in the diseased animals. The sows had gastrointestinal symptoms such as blood in stool and vomiting. Necropsy revealed obvious gastric ulcers, gastrointestinal perforation, and intestinal hemorrhage in the gastrointestinal tract, but no lesions in other organs. The microbial species in gastric samples collected from gastric ulcer of the diseased sows then was initially identified as Candida by using routine systems of microscopic examination, culture characteristics on the medium Sabouraud dextrose agar medium. The fungus was further identified as C. tropicalis by species-specific PCR and sequencing. This study revealed an infection of C. tropicalis in sows through gastrointestinal mucosa could cause fatal digestive system disease and septicemia. Conclusions For the first time, a strain of C. tropicalis was isolated and identified from the gastric tissue of sows with severe gastrointestinal diseases. PCR and sequencing of ITS-rDNA combined with morphology and histopathological assay were reliable for the identification of Candida clinically.

In cold regions, traditional air source heat pumps are easily frosted in heating state. The frosting problem can be effectively overcome by the direct-expansion solar/air-assisted heat pump (DX-SASIHP). One of the most critical components in the DX-SASIHP system is the collector/evaporator, whose performance hinges on its structure and material. To improve the heating performance of DX-SASIHP, this paper attempts to optimize the structure and material of the collector/evaporator by the Computational Fluid Dynamics (CFD) method. Specifically, key parameters like tube diameters, rib height, rib thicknesses, and tube material were examined under the same weather conditions. On this basis, the heat collection capacity and heat collection efficiency of the collector/evaporator were evaluated. Finally, the collector/evaporator structure was optimized in the light of heat collection capacity, heat collection efficiency and manufacturing cost. The key parameters were optimized as follows: 23 mm for tube diameter, 45 mm for rib height, 4 mm for rib thickness, and aluminum for tube material. The research results lay a theoretical basis for the collector/evaporator design of DX-SASIHP in heating state.

The development of integrated electricity and heat system (IEHS) promotes the renewable energy penetration and the complementary coupling between the electric power system (EPS) and the district heat system (DHS). The efficient energy flow calculation (EFC) in the IEHS has become a hotspot as the system scale and complexity increase. This paper firstly constructed the dynamic DHS model using Euler difference scheme. On this basis, the fast decomposed method is proposed to decompose the system with various scales and topologies, which improves the computation efficiency. Moreover, the modification mode is then developed to modify the EFC results in the decomposed systems and ensure the accuracy. Numerical simulation verifies the accuracy of the proposed method and its superiority in computation efficiency.

In modern industry, due to the poor working environment and the complex working conditions of mechanical equipment, the characteristics of the impact signals caused by faults are often submerged in strong background signals and noises. Therefore, it is difficult to effectivelyextract the fault features. In this paper, a fault feature extraction method based on improved VMD multi-scale dispersion entropy and TVD-CYCBD is proposed. First, the marine predator algorithm (MPA) is used to optimize the modal components and penalty factors in VMD. Second, the optimized VMD is used to model and decompose the fault signal, and then the optimal signal components are filtered according to the combined weight index criteria. Third, TVD is used to denoise the optimal signal components. Finally, CYCBD filters the de-noised signal and then envelope demodulation analysis is carried out. Through the simulation signal experiment and the actual fault signal experiment, the results verified that multiple frequency doubling peaks can be seen from the envelope spectrum, and there is little interference near the peak, which shows the good performance of the method.

Stimuli-responsive hydrogel actuators have attracted tremendous interest in switches and microrobots. Based on N-isopropylacrylamide (NIPAM) monomers with LCST phase separation and photochromic molecule spiropyran which can respond to ultraviolet light and H+, we develop a novel multistimuli-responsive co-polymer anisotropic bilayer hydrogel, which can undergo complex deformation behavior under environmental stimuli. Diverse bending angles were achieved based on inhomogeneous swelling. By controlling the environmental temperature, the bilayer hydrogels achieved bending angles of 83.4° and −162.4° below and above the critical temperature of PNIPAM. Stimulated by ultraviolet light and H+, the bilayer hydrogels showed bending angles of −19.4° and −17.3°, respectively. In addition, we designed a strategy to enhance the mechanical properties of the hydrogel via double network (DN). The mechanical properties and microscopic Fourier transform infrared (micro-FTIR) spectrum showed that the bilayer hydrogel can be well bonded at the interfaces of such bilayers. This work will inspire the design and fabrication of novel soft actuators with synergistic functions.

As a special road-building material widely distributed in desert areas, critical state soil mechanics is used to study the mechanical properties of sand and make up for the lack of research on its engineering characteristics. A series of drained and undrained triaxial compression tests with a loading rate of 0.12 mm/min medium-density aeolian sands taken from Tengger Desert in the northwest of China was carried out to obtain the three-dimensional state boundary surface. The test results reveal that the strength gained from drained and undrained tests increased, respectively, linearly and non-linearly with the increase of the effective confining pressure. Affected by the variation of pore pressure and shear rate, the undrained strength was higher than the drained strength at low effective confining pressures, and the two types of strengths tend to be consistent when the effective confining pressure becomes greater than 800 kPa. The volumetric changes of the aeolian specimens transition from dilatation to contraction when the effective confining pressures increase. The investigation of the strength, deformation and failure characteristics gives rise to the shape parameters of its state boundary surface, which provides not only a basis for the constitutive modelling of the aeolian sand, but also a reference for roadbed construction and other foundation engineering in desert areas.

Abstract Background Each rare disease only affects a small number of population. However, a total of 7000 rare diseases may affect 10% of the population. Due to the severity and lack of rare disease awareness, rare disease represents a huge challenge for the healthcare system. In Western countries, patient organizations have been playing an integral role in raising awareness, advocating legislation, and supporting drug development. This study aims to assess the unmet needs of rare disease patient organizations in China, and identify their unmet needs, providing essential information for the government and legislators. Results A total of 28 individuals representing 28 patient organizations in China were interviewed. Most organizations do not have official registration, employees, written standard operation protocol, or reliable financial resources. Misdiagnosis or delayed diagnosis is common, and treatment is often lacking. Due to the lack of financial resources, no organizations have been able to sponsor academic research, unlike their counterparts in Western countries. As to challenges, 71.4% of interviewees listed lack of rare disease awareness among the general public, while 67.9% selected lack of financial resources. Further, only 7.3% of these organizations received support from the government, and 28.6% received support from the general public. As to recommendations to the government, 82.1% of interviewees selected special insurance programs for rare diseases because rare diseases have been generally excluded from the national medical insurance programs. In addition, 78.6% of interviewees recommended to stimulate rare disease research, 75% recommended to import orphan drugs, and 71.4% recommended legislation of an orphan drug act, highlighting the urgent need of therapies. Conclusions Due to lack of support and rare disease awareness, patient organizations in China are still in the early phase. To empower these patient organizations, the interviewees’ recommendations, including legislating orphan drug act and releasing official definition of rare diseases, should be considered by the government and legislators.

Rice–crab co-culture is a high-benefit eco-breeding pattern that has been extensively developed in many regions of China. However, little attention has been paid to the safety of pesticides used to control rice pests in the crab–rice co-culture system. This study monitored the actual residue levels of propiconazole in water and soil of rice–crab co-culture fields and evaluated the acute toxicity, subchronic toxicity, and bioaccumulation of propiconazole to Eriocheir sinensis. We observed that the residue level of propiconazole in paddy soil was higher than that in paddy water within 42-day field monitoring. Propiconazole demonstrated a low acute toxicity (96 h-LC50 > 100 mg/L) to E. sinensis and exhibited no obvious adverse impact on the growth of E. sinensis after exposure to 500 μg/L, which was 10 times the actual residual concentration of propiconazole in the crab–rice co-culture field. The highest bioaccumulation of propiconazole was obtained from gills, followed by the hepatopancreas and meat. These results will contribute to the guidance of scientific utilization of pesticides in the crab–rice co-culture field.

Little is known about whether the application of biogas residues in rice fields will affect the degradation of pesticides. This study investigated the dissipation behavior of the fungicide difenoconazole in paddy water and sediment after the application of a chemical fertilizer or biogas residues. The results showed that the application of biogas residues changes the dissipation of difenoconazole in both paddy water and sediment. The half-lives of difenoconazole in paddy water and sediment with biogas residues were 0.50 and 10.09 days, respectively, while the half-lives of difenoconazole in paddy water and sediment with chemical fertilizer were only 0.22 and 4.64 days, respectively. After biogas residues were applied in a paddy field, no significant changes in pH value of paddy water and sediment and soil microorganisms were observed, but soil organic matter decreased by 30%. The above studies suggest that biogas residues may affect the dissipation of pesticides in paddy field water and sediments, which provides a new focus on the scientific and rational use of biogas residues as organic fertilizer in rice fields.

Circular RNAs (circRNAs) are a type of long non-coding RNA with covalently closed loops that are naturally resistant to exoribonuclease. With the rapid development of high-throughput sequencing technologies and bioinformatics, increasing data suggest that circRNAs are abnormally expressed in renal cell carcinoma (RCC) and act as important regulators of RCC carcinogenesis and progression. CircRNAs play important biological roles in modulating cell proliferation, migration, invasion, apoptosis, and gemcitabine chemoresistance in RCC. Most of the circRNAs studied in RCC have been reported to be significantly associated with many clinicopathologic characteristics and survival parameters of RCC. The stability and specificity of circRNAs enable them potential molecular markers for RCC diagnosis and prognosis. Moreover, circRNAs have emerged as targets for developing new therapies, because they can regulate various signaling pathways associated with RCC initiation and progression. In this review, we briefly summarize the biogenesis, degradation, and biological functions of circRNAs as well as the potential clinical applications of these molecules for RCC diagnosis, prognosis, and targeted therapy.

Due to the transfer of Lannigou gold mining from shallow to deep, a series of stability problems of surrounding rock have been caused. The drilling pressure relief technology has unique advantages in the control of mine pressure in high-stress roadways. In order to explore the damage effect of borehole pressure relief technology on rock, uniaxial compression and acoustic emission tests were carried out on siltstone specimens with borehole diameters of 8 mm, 12 mm, and 16 mm, respectively, and the acoustic emission signals of the whole process were collected simultaneously to explore the uniaxial compression of siltstone specimens with prefabricated holes in this paper. According to the statistical characteristics of acoustic emission, the damage law of siltstone specimens with prefabricated holes was explored from the microscopic point of view and the damage effect of drilling on siltstone specimens reflected by the acoustic emission phenomenon was revealed. The research results show that there are multiple stress drops before the deformation and failure of the rock sample with prefabricated drill holes; there is a positive correlation between the diameter of the drill hole and the power law index of the stress drop distribution; the acoustic emission activity of the rock during the deformation and failure process can be indirectly reflected the evolution of microfractures; the energy probability density function under different borehole diameters conforms to the power law distribution; the critical exponent obtained by the maximum likelihood estimation has an optimal plateau value, which can accurately characterize the power exponent of the energy distribution; the launch waiting time and aftershock sequence have a good power-law distribution in logarithmic coordinates. The research results provide a certain theoretical basis for the application of drilling pressure relief technology in southwestern Guizhou.

In this paper, the particle shape factor of the stone in the soil-rock mixture was used as the specific research object of the geogrid pull-out test, combining the random generation method and evaluation criteria of rock blocks in the soil-rock mixture, and a feasible method for classifying stones according to their shape factors is presented. By comparing the mechanical behavior of soil-rock mixture composed of rock blocks with different rock material shape factors in the geogrid pull-out tests under different working conditions, we studied its changing laws. The research shows that with the increase of the shape factor of the stone, the pull-out resistance of the geogrid gradually decreases. When the stone paved in the model box changes from a single layer to a double layer, the pull-out resistance of the geogrid increases, and there is a disturbance range in the contact interface between the geogrid and the stone; stone blocks 5 cm away from the geogrid surface can still have an indirect effect on the pull-out resistance of geogrids; the pull-out resistance of geogrid in clay is greater than that in the sand; and an empirical expression for predicting the pull-out resistance of geogrids is obtained. This empirical expression helps to explain the influence mechanism of stone shape on drawing resistance of geogrid.

Soil organic carbon (SOC) is significant for soil quality and global carbon cycles. SOC was observed to be related to soil geochemistry, and soils originating from different bedrocks have different geochemical properties, but the effect of bedrock on SOC is still undefined. Soils overlying different bedrocks in Zhenxiong County and Weixin County were sampled. Specifically, soils in the mineral horizon, which are less affected by the external environment than surface soils, are focused on to reveal the effect of bedrock on SOC. Al/Ti, Fe/Ti, and Al/Fe indicate a soil–rock successive relationship. SOC contents in the mineral horizon are 0.19–2.74% (1.24% on average), and those in the surface horizon are 1.26–4.01% (2.63% on average). SOC contents in the surface and mineral horizons of the same bedrock are significantly positively correlated, implying that the bedrock is an important factor affecting SOC. SOC in the mineral horizon is related to the first transition metal ions. Significantly, positive correlations of SOC (p < 0.01) with Co, Cu, Ti, V, and Zn, and a positive correlation (p < 0.05) with Ni were observed in the mineral horizon. Organic transition metal complexation seems to play an important role in governing SOC in the mineral horizon. That is, the complexation maintains organic carbon stability, slows down its decomposition rate, and accumulates organic carbon. The Ca–SOC positive correlation in the mineral horizon exits because Ca also can complex with organic carbon. Co, Cu, and V–SOC positive correlations (p < 0.05) were also observed, but there were no significant positive correlations (p < 0.01) in the surface horizon because surface SOC had diversified sources. An SOC evolution model influenced by the bedrock was forwarded. Thus, the different soil geochemistry originating from different bedrocks should be noticed when SOC and global carbon cycles are discussed.

Abstract Background A new C2 transpedicular lag screw designed by our team has been used in human cadaver spines for biomechanical testing, and the results showed that the biomechanical properties of the new C2 transpedicular lag screw were better than ordinary screws. The objective of this study is to analyze the clinical efficacy and safety of the new C2 transpedicular lag screw fixation for the treatment of unstable Hangman’s fracture. Methods From March 2013 to June 2017, 25 patients who had unstable Hangman’s fractures were operated on with a new C2 transpedicular lag screw fixation. The patients included 18 males and 7 females whose ages ranged from 31 to 62 years (average 45.4 ± 9.3 years). The cause of the injury was a traffic accident in 17 patients and a fall from height in 8 patients. Other associated lesions included rupture of the spleen (1 patient) and rib fractures (2 patients). According to the Levine-Edwards classification, 17 patients were type II and 8 patients were type IIA, and according to the Frankel Neurological Performance scale, 8 cases and 17 cases were graded as spinal cord injury D and E, respectively. Twenty-three cases received bilateral screw fixation, and 2 cases had unilateral screw fixation because another pedicle was chipped. The whole procedure was accomplished with monitoring by “C”-arm fluoroscopy. Results The mean follow-up time was 36 ± 12 months and ranged from 24 to 60 months. No obvious symptomatic or radiologic postoperative complications were found during the follow-up period. Six cases were restored from D to E while 2 cases remained D according to the American Spinal Injury Association (ASIA) grade. Pre- and postoperative visual analogue scale (VAS) and Neck Disability Index (NDI) were statistically different (P < 0.001). Osseous union was achieved in all cases, and the range of cervical motion recovered to the normal level up to the last follow-up. Conclusions The primary clinical and radiographic efficacies of a new C2 transpedicular lag screw fixation for the treatment of unstable Hangman’s fracture were satisfactory. This approach could be considered a simple, effective, reliable, and economic surgical method for managing unstable Hangman’s fractures.

Abstract Background Hepatocellular carcinoma is the third top cause of cancer-related mortalities worldwide. The prognosis of HCC patients remains poor due to rapid progression and high incidence of tumor recurrence. Nicastrin (NCSTN), a core subunit of γ-Secretase, has been reported to play a vital role in tumor progression. However, no study till now has revealed its role in HCC. Methods The expression of NCSTN was evaluated by immunohistochemical staining, Western blot, and quantitative real-time PCR. Cell counting kit-8, colony formation and cell cycle assays were used for evaluating cell growth in vitro. Transwell and wound-healing assays were used for evaluating cell migration and invasion capacity. Immunofluorescence, subcellular protein fractionation and co-immunoprecipitation were used for location analysis of β-catenin. The in vivo functions of NCSTN were illustrated by xenograft tumor models. Results NCSTN was dramatically overexpressed in HCC compared to normal liver tissues. Elevated NCSTN expression level was significantly correlated to worse overall and recurrence-free survival of HCC patients. Enhanced NCSTN expression promoted HCC cell growth, migration and invasion in vitro and in vivo. Mechanistic investigations showed that NCSTN induced epithelial-mesenchymal transition (EMT) process via upregulation of Zeb1. Subsequently, we revealed that NCSTN facilitated nuclear translocation of β-catenin, a positive transcriptional regulator of Zeb1. Using Notch and AKT inhibitors, we revealed that NCSTN promoted β-catenin activation through Notch1 and AKT signaling pathway. NCSTN increased AKT and GSK-3β phosphorylation by cleavage of Notch1, which decreased GSK-3β/β-catenin complex. The inactivation of GSK-3β inhibited the β-catenin degradation and promoted nuclear translocation of β-catenin to initiate transcription of Zeb1, resulting in malignant phenotype. Conclusions Our results demonstrated that NCSTN promoted HCC cell growth and metastasis via β-catenin-mediated upregulation of Zeb1 in a Notch1/AKT dependent manner, suggesting that NCSTN might serve as a potential prognostic marker and therapeutic target for HCC.

Abstract Background Angelica root is the dry root of the Umbelliferae plant Angelica sinensis (oliv) Diels. Angelica organic acid (OA) is the main active ingredient in Angelica sinensis, and it exerts potential anti-atherosclerotic effects by preventing Oxidized low-density lipoprotein (Ox-LDL) induced endothelial injury. To study the protective effects of OA on ox-LDL-induced HUVECs autophagic flux dysfunction and inflammatory injury. Methods OA were isolated by water extraction and alcohol precipitation, and then the content of ferulic acid (FA) in the OA was determined by high performance liquid chromatography. The ox-LDL-induced endothelial injury model was established. The effect of ferulic acid on the survival of Human umbilical vein endothelial cells (HVUECs) was detected by CCK-8 assay. HUVECs were pretreated with different concentrations of OA (20 μmol/L, 40 μmol/L, and 80 μmol/L), and Western Blot was used to detect the expressions of LC3II, p62, MCP-1, VCAM-1 and LOX-1. The autophagosomes in HUVECs were observed by transmission electron microscopy (TEM). Results 20 μmol/L OA could increase the expression of LC3II and decrease the expression of p62, MCP-1, VCAM-1 and LOX-1. The results of TEM showed that angelica organic acids promoted cell organelle degradation in autolysosomes. Conclusion OA could reduce inflammation, protect endothelial cells and play an anti-atherosclerotic role by enhancing the autophagy flux of damaged endothelial cells, in which FA the major active ingredient of OA played a major role.

The present study reviewed the relevant recent literature regarding the development and application of oblique lumbar interbody fusion (OLIF), with a particular focus on its application and associated complications. The study evaluated the rationality of this technique and demonstrated the direction of future research by collecting data on previous operative outcomes and complications. A literature search was performed in Pubmed and Web of Science, including the following keywords and abbreviations: anterior lumbar interbody fusion (ALIF), lateral lumbar interbody fusion (LLIF), direct lateral interbody fusion (DLIF), extreme lateral interbody fusion (XLIF), oblique lateral interbody fusion (OLIF), adjacent segment disease (ASD), and adult degenerative scoliosis (ADS). A search of literature published from January 2005 to January 2019 was conducted and all studies evaluating development and application of OLIF were included in the review. According to the literature, the indications for OLIF are various. OLIF has excellent orthopaedic effects in degenerative scoliosis patients and the incidence of bony fusion is higher than for other approaches. It also provides a better choice for revision surgery. It has various advantages in many aspects, but the complications cannot be ignored. As a new minimally invasive technique, the advantages of OLIF are obvious, but further evaluation is needed to compare its operation‐related data with that of traditional open surgery. In addition, more prospective studies are required to compare minimally invasive and open spinal surgery to confirm its specific efficacy, risk, advantages, learning curve, and ultimate clinical efficacy.

Curcumin (CM) is a natural polyphenolic compound with multiple biomedical functions. However, clinical applications face more challenges due to its low dissolution rate and poor bioavailability. Micronization is an effective strategy to overcome these drawbacks. Herein, CM nanoparticles (CM NPs, ∼300 nm) were fabricated using solution enhanced dispersion by supercritical CO2 (SEDS). The solubility of CM NPs was remarkably enhanced. Aim to study the effects of micronization on the biological functions of CM, we investigated the antibacterial activity of original CM and CM NPs upon Pseudomonas aeruginosa. In vitro, the minimal inhibitory concentrations (MIC) assay, solid-medium spot assay, growth kinetics assay and morphologic observation using atomic force microscopy (AFM) confirmed that the anti-P. aeruginosa activity of CM NPs was enhanced compared to original CM. Moreover, CM NPs also showed stronger inhibition for adhesion and biofilm formation of P. aeruginosa compared to original CM. Experiments on mice infected with P. aeruginosa showed that CM NPs have a better therapeutic effect than the original CM in vivo. In summary, CM NPs may be a novel and promising therapeutic candidate for bacterial infection.

Locating the tiny insulator defect object with complex backgrounds in high-resolution aerial images is a challenging task. In this paper, we propose a novel method which cascades detection and segmentation networks to identify the defect from the global and local two levels: (1) The improved Faster R-CNN is carried out to capture both defects and insulators in the entire image. ResNeXt-101 is adopted as the feature extraction network so as to fully extract features, and Feature Pyramid Network (FPN) is built to enhance the ability of detecting small targets. In addition, the Online Hard Example Mining (OHEM) training strategy is applied to solve the imbalance problem of positive and negative samples. (2) All the detected insulators are extracted and fed into the improved U-Net network to futher inspect at pixel level, we utilize the pre-trained ResNeXt-50 as the encoder of U-Net, incorporate an attention module, Spatial and Channel Squeeze & Excitation Block (SCSE), into the decoding path to highlight the meaningful information. A hybrid loss which merges binary cross entropy (BCE) loss and dice coefficient loss is designed to train our network for figuring out the class imbalance issue. The missed detection can be greatly reduced with the combination of two modified network, which makes comprehensive use of the original map information and local information. On the test set of actual images, the insulator defect recognition precision and recall of the cascade network is 91.9% and 95.7%, exhibiting strong robustness and accuracy.

Because social networks exemplify the phenomenon of homogeneity in complex networks, researchers generally believe that a user's privacy disclosure is closely related to that of the users around them, but we find that the related users studied in previous methods were not correct. That is, the analyzed user groups may have had nothing to do with the privacy disclosure of the target users. Since private information is time-sensitive, information held by users who are no longer in the same environment as the target user may no longer be true and have lost its value. For example, considering students and members of the working class, transfers to another school for further studies or job changes entail dramatic changes to most of their information. This lack of timeliness has an overarching impact on the effectiveness of social network analysis and privacy protection, but this problem has not been addressed by researchers. Therefore, we study and characterize this problem, add the user's behavior trace to solve this problem and measure the user's privacy status more accurately.

The hydrogen-induced plastic loss behavior of titanium alloys is often reported, but there are relatively few studies on high-strength aluminum alloys. In this article, the hydrogen-induced plastic damage behavior of 7085-T7651 high-strength aluminum alloy was investigated using a tensile specimen with pre-charging hydrogen, and the microstructure was characterized by transmission electron microscopy, scanning electron microscopy and time-of-flight secondary ion mass spectrometry. The results showed that 7085-T7651 high-strength aluminum alloy material has certain hydrogen embrittlement sensitivities, and with the increase of hydrogen-charging time, the hydrogen content and sensitivity of the material increases significantly. For the first time, the theoretical analysis and intuitive quantitative characterization of the hydrogen-induced plastic loss behavior mechanism on 7085-T7651 high-strength aluminum alloy is stated as the formation of Mg and H segregation formed at the crystal boundary, which will result in the weakening of the crystal boundary.

Hydrogels with high mechanical strength, good crack resistance, and good adhesion are highly desirable in various areas, such as soft electronics and wound dressing. Yet, these properties are usually mutually exclusive, so achieving such hydrogels is difficult. Herein, we fabricate a series of strong, tough, and adhesive composite hydrogels from polyampholyte (PA) gel reinforced by nonwoven cellulose-based fiber fabric (CF) via a simple composite strategy. In this strategy, CF could form a good interface with the relatively tough PA gel matrix, providing high load-bearing capability and good crack resistance for the composite gels. The relatively soft, sticky PA gel matrix could also provide a large effective contact area to achieve good adhesion. The effect of CF content on the mechanical and adhesion properties of composite gels is systematically studied. The optimized composite gel possesses 35.2 MPa of Young’s modulus, 4.3 MPa of tensile strength, 8.1 kJ m−2 of tearing energy, 943 kPa of self-adhesive strength, and 1.4 kJ m−2 of self-adhesive energy, which is 22.1, 2.3, 1.8, 6.0, and 4.2 times those of the gel matrix, respectively. The samples could also form good adhesion to diverse substrates. This work opens a simple route for fabricating strong, tough, and adhesive hydrogels.

The purpose of this study was to evaluate the effects of dietary coated lysozyme on growth performance, serum biochemical indexes, antioxidant activity, digestive enzyme activity, intestinal permeability, and the cecal microbiota in weaned piglets. In total, 144 weaned Large White × Landrace piglets were divided into six treatment groups, with 3 replicates and 8 piglets per replicate: CN, a basal diet; CL-L, CL-M, and CL-H, basal diet supplemented with 100, 150, 500 mg/kg coated lysozyme; UL, basal diet supplemented with 150 mg/kg lysozyme; and Abs, basal diet supplemented with 150 mg/kg guitaromycin for 6 weeks. Compared with the CN and UL diets, dietary CL-H inclusion increased the average daily gain (ADG) and decreased the feed/gain (F/G) ratio of piglets (p < 0.05). The addition of 500 mg/kg coated lysozyme to the diet significantly increased the total protein (TP) and globulin (Glob) plasma levels of weaned piglets (p < 0.05). Supplementation with 500 mg/kg coated lysozyme significantly increased the serum IgM concentration and increased lipase activity in the duodenum (p < 0.05). The addition of coated lysozyme and lysozyme significantly decreased the malondialdehyde (MDA) content, while the superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and total antioxidant capacity (T-AOC) levels all increased (p < 0.05). High-throughput sequencing results showed that CL-H treatment effectively improved the intestinal microbiome. The relative abundance of Terrisporobacter in the CL-H and CL-M groups was significantly lower than that in the other groups (p < 0.05). LEfSe analysis results showed that the relative abundance of Coprococcus_3 was higher in the CL-M treatment group. The marker species added to the CL-H treatment group was Anaerofilum. In summary, as a potential substitute for feed antibiotics, lysozyme is directly used as a dietary additive, which is inefficient. Therefore, we used palm oil as the main coating material to coat lysozyme. Lysozyme after coating can more effectively improve the growth performance of piglets by improving the intestinal flora, improving the activity of digestive enzymes, reducing the damage to intestinal permeability and oxidative stress in piglets caused by weaning stress, and improving the immunity of piglets.

Intestinal microbiota play an important role in the survival of the host. However, no study to date has elucidated the adjustment of intestinal microbiota of the host during rewilding. Thus, this study aims to describe the intestinal bacterial community of reintroduced Przewalski’s horse (RPH) after being released into their original habitat for approximately 20 years in comparison with that of captive Przewalski’s horse (CPH), sympatric domestic horse (DH) and Mongolian wild ass (MWA) by sequencing the 16S rRNA gene. The results showed that the prevalent bacterial communities were different among CPHs, RPHs, DHs and MWAs at the family level. NMDS and ANOSIM analysis showed that the pattern of bacterial community composition in captive equines was distinct from that in the wild groups. It is shown that some bacteria had significant differences among different taxa (p < 0.001), such as Firmicutes, Bacteroidetes, Armatimonadetes, Clostrida, Bacteroidia, Clostridiales, Bacteroidales, Rikenellaceae and Bacteroidales_UCG-001. These bacteria were associated with the transition from in captive to in the wild (CPH and RPH), which reflected the change of environmental conditions. Meanwhile, Proteobacteria, Clostridia, Bacilli, Negativicutes, Gammaproteobacteria, Clostridiales, Bacillales, Selenomonadales, Pseudomonadales and Planococcaceae were the changed groups among RPHs, MWAs and DHs, which are related to feeding habits and diseases. Our results clearly showed the differences between intestinal microbiota in reintroduced animals and wild animals and led us to understand the survival state of reintroduced animals in the wild.

In this paper, we study the regularized Huber regression algorithm in a reproducing kernel Hilbert space (RKHS), which is applicable to both fully supervised and semi-supervised learning schemes. Our focus in the work is two-fold: first, we provide the convergence properties of the algorithm with fully supervised data. We establish optimal convergence rates in the minimax sense when the regression function lies in RKHSs. Second, we improve the learning performance of the Huber regression algorithm by a semi-supervised method. We show that, with sufficient unlabeled data, the minimax optimal rates can be retained if the regression function is out of RKHSs.

Sepsis, an infection-induced systemic inflammatory disorder, is often accompanied by multiple organ dysfunction syndromes with high incidence and mortality rates, and those who survive are often left with long-term sequelae, bringing great burden to social economy. Therefore, novel approaches to solve this puzzle are urgently needed. Previous studies revealed that long non-coding RNAs (lncRNAs) have exerted significant influences on the process of sepsis. The aim of this review is to summarize our understanding of lncRNAs as potential sepsis-related diagnostic markers and therapeutic targets, and provide new insights into the diagnosis and treatment for sepsis. In this study, we also introduced the current diagnostic markers of sepsis and discussed their limitations, while review the research advances in lncRNAs as promising biomarkers for diagnosis and prognosis of sepsis. Furthermore, the roles of lncRNAs in sepsis-induced organ dysfunction were illustrated in terms of different organ systems. Nevertheless, further studies should be carried out to elucidate underlying molecular mechanisms and pathological process of sepsis.

Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory disease that causes high rates of disability and mortality worldwide because of severe progressive and irreversible symptoms. During the period of COPD initiation and progression, the immune system triggers the activation of various immune cells, including Regulatory T cells (Tregs), dendritic cells (DCs) and Th17 cells, and also the release of many different cytokines and chemokines, such as IL-17A and TGF-β. In recent years, studies have focused on the role of IL-17A in chronic inflammation process, which was found to play a highly critical role in facilitating COPD. Specially, IL-17A and its downstream regulators are potential therapeutic targets for COPD. We mainly focused on the possibility of IL-17A signaling pathways that involved in the progression of COPD; for instance, how IL-17A promotes airway remodeling in COPD? How IL-17A facilitates neutrophil inflammation in COPD? How IL-17A induces the expression of TSLP to promote the progression of COPD? Whether the mature DCs and Tregs participate in this process and how they cooperate with IL-17A to accelerate the development of COPD? And above associated studies could benefit clinical application of therapeutic targets of the disease. Moreover, four novel efficient therapies targeting IL-17A and other molecules for COPD are also concluded, such as Bufei Yishen formula (BYF), a Traditional Chinese Medicine (TCM), and curcumin, a natural polyphenol extracted from the root of Curcuma longa.

A visual dialog task entails an agent engaging in a multiple round conversation about an image. Notably, one of the main issues is capturing the semantic associations of multiple inputs, such as the questions, dialog history, and image features. Many of the techniques use a token or a sentence granularity semantic representation of the question and dialog history to model semantic associations; however, they do not perform collaborative modeling, which limits their efficacy. To overcome this limitation, we propose a multi-granularity semantic collaborative reasoning network to properly support a visual dialog. It employs different granularity semantic representations of the question and dialog history to collaboratively identify the relevant information from multiple inputs based on attention mechanisms. Specifically, the proposed method collaboratively reasons the question-related information from the dialog history based on its granular semantic representations. Then, it collaboratively locates the question-related visual objects in the image by leveraging refined question representations. The experimental results conducted on the VisDial v.1.0 dataset verify the effectiveness of the proposed method, showing the improvements of the best normalized discounted cumulative gain score from 59.37 to 60.98 with a single model, from 60.92 to 62.25 with ensemble models, and from 63.15 to 64.13 with performing multitask learning.

Studies have shown that illuminance and correlated colour temperature (CCT) are strongly correlated with body responses such as circadian rhythm, alertness, and mood. It is worth noting that these responses show a complex and variable coupling, which needs to be solved using accurate mathematical models for the regulation of indoor light parameters. Therefore, in this study, by weighing the evaluations of visual comfort, alertness, valence, and arousal of mood, a multi-objective optimisation mathematical model was developed with constraints conducive to the healthy rhythm. The problem was solved with the multi-objective evolutionary algorithm based on the decomposition differential evolution (MOEA/D-DE) algorithm. Taking educational space as the analysis goal, a dual-parameter setting strategy for illuminance and CCT covering four modes was proposed: focused learning, comfortable learning, soothing learning, and resting state, which could provide a scientific basis for the regulation of the lighting control system. The alertness during class time reached 3.01 compared to 2.34 during break time, showing a good light facilitation effect. The proposed mathematical model and analysis method also have the potential for application in the lighting design and control in other spaces to meet the era of intelligent, highly flexible, and sustainable buildings.

Prediction of building energy consumption using mathematical modeling is crucial for improving the efficiency of building energy utilization, assisting in building energy consumption planning and scheduling, and further achieving the goal of energy conservation and emission reduction. In consideration of the non-linear and non-smooth characteristics of building energy consumption time series data, a short-term, hybrid building energy consumption prediction model combining variational mode decomposition (VMD), a simulated annealing (SA) algorithm, and a deep belief network (DBN) is proposed in this study. In the proposed VMD-SA-DBN model, the VMD algorithm decomposes the time series into different modes to reduce the fluctuation of the data. The SA-DBN prediction model is built for each mode separately, and the DBN network structure parameters are optimized by the SA algorithm. The prediction results of each model are aggregated and reconstructed to obtain the final prediction output. The validity and prediction performance of the proposed model is evaluated on a publicly available dataset, and the results show that the proposed new model significantly improves the accuracy and stability of building energy consumption prediction compared with several typical machine learning methods. The mean absolute percent error (MAPE) of the VMD-SA-DBN model is 63.7%, 65.5%, 46.83%, 64.82%, 44.1%, 36.3%, and 28.3% lower than that of the long short-term memory (LSTM), gated recurrent unit (GRU), VMD-LSTM, VMD-GRU, DBN, SA-DBN, and VMD-DBN models, respectively. The results will help managers formulate more-favorable low-energy emission reduction plans and improve building energy efficiency.

In order to promote the development and construction of smart cities, the massive equipment requirements of sensing terminals increased the pressure on urban site resource allocation. The light pole is suitable for carrying various urban functional equipment to form a smart street lighting system, which can provide rich site resources for the large-scale construction of urban functional facilities such as 5G micro base stations. However, the selection and combination of equipment mounted in the smart street lighting system only focus on the functional superposition at the physical level, without considering the relevance of each subsystem in practical application scenarios. Therefore, this study proposed a 5G micro base station location model based on a smart street lighting system. The correlation and cooperativity between 5G micro base stations and mounted devices were fully considered, and a universal system-level location selection index was developed to realize rational utilization of urban space site resources and intelligent linkage between subsystems. The results showed that the model is significantly effective for functional areas with different road network characteristics and provides practical, robust, effective, and accurate help for similar location selection problems.

Flavor is an important index to evaluate the sensory quality of tobacco. The process of fermentation is a key step in the production of aromatic substances in tobacco leaves and an important factor in improving their quality. Worldwide, reams of research show that chromatographic technology plays an irreplaceable role in the tobacco aromatic chemistry. Nevertheless, the degradation mechanism of latent aromatic compounds and the formation mechanism of characteristic aromatic substances have not been fully and systematically elucidated. In this study, the latest progress of basic methods, techniques, and research results of the separation, analysis, and identification of aromatic substances in fermented tobacco leaves were reviewed, and the next research and application directions were prospected. It is expected to provide theoretical reference for the study of molecular mechanism of tobacco flavor, reveal the degradation mechanism of potential aroma compounds, and help improve the quality of tobacco.

Background As one of the most important vegetable crops, pepper has rich nutritional value and high economic value. Increasing heat stress due to the global warming has a negative impact on the growth and yield of pepper. Methods To understand the heat stress response mechanism of pepper, an iTRAQ-based quantitative proteomic analysis was employed to identify possible heat-responsive proteins and metabolic pathways in 17CL30 and 05S180 pepper seedlings under heat stress. Result In the present study, we investigated the changes of phenotype, physiology, and proteome in heat-tolerant (17CL30) and heat-sensitive (05S180) pepper cultivars in response to heat stress. Phenotypic and physiological changes showed that 17CL30 had a stronger ability to resist heat stress compared with 05S180. In proteomic analysis, a total of 3,874 proteins were identified, and 1,591 proteins were considered to participate in the process of heat stress response. According to bioinformatic analysis of heat-responsive proteins, the heat tolerance of 17CL30 might be related to a higher ROS scavenging, photosynthesis, signal transduction, carbohydrate metabolism, and stress defense, compared with 05S180.

Abstract Background Shunxinzufang decoction is tutors, empirical formula and has been used in Chinese patients of HFpEF for several years. The aim of this study was to make into sustained release granules and select the best formula for the preparation of Shunxin sustained release granules and to evaluate its in vivo and in vitro drug release behavior. Methods Response surface methodology and Center composite design were applied to screen the optimal formula of Shunxin sustained release granules. HPLC was used to detect indicative ingredients—paeoniflorin, calycosin-7-glucoside and ferulic acid in Shunxin sustained release granules. The in vitro sustained release character of indicative ingredients was investigated in simulated digestive fluids. In-vivo process of active components was studied through pharmacokinetics. Results The optimal formula of Shunxin sustained release granules consisted of 35% shunxinzufang extract and 65% HPMC/starch (HPMC/starch ratio = 2:1). Three indicative components can be separated well under selected HPLC conditions. Compared with Shunxinzufang extract, the active components of Shunxin sustained release granules have obvious sustained-release character and improved bioavailability. Conclusion Shunxin sustained release granules has obvious sustained-release character and improved bioavailability.

Introduction and aims: Interferon-α (IFN) has shown potential benefits in patients with hepatocellular carcinoma (HCC), and these effects may be mediated by inhibiting cancer cell proliferation. However, the detailed mechanisms underlying the anti-proliferative effects of IFN remain obscure. In this study, we evaluate the role of the novel oncogenic microRNA (miRNA) miR-155 in the anti-proliferative effects of pegylated interferon-α (PEG-IFN) on HCC cells. Methods: The effects of PEG-IFN on HepG2 cell proliferation, migration and invasion were determined using the MTT assay, flow cytometry analysis and the Transwell assay, respectively. Reverse transcription quantitative polymerase chain reaction was used to analyze miR-155 expression. The levels of proteins involved in Wnt/β-catenin signal transduction were determined by western blot analysis and immunofluorescence staining. Mimics of miR-155 were transfected into HepG2 cells to assess the role of miR-155 in the PEG-IFN-induced anti-proliferative effect. Results: PEG-IFN significantly inhibited the proliferation, migration and invasion of HepG2 cells in a dose-dependent manner by inhibiting cell cycle progression. In parallel with reduced cell proliferation, migration and invasion, miR-155 was efficiently downregulated by PEG-IFN in a dose-dependent manner. Moreover, the transfection of miR-155 decreased the inhibitory effect of PEG-IFN on HepG2cell proliferation, migration and invasion, as well as the downregulation of proteins in the Wnt/β-catenin pathway. Conclusions: The anti-proliferative effects of PEG-IFN on HCC are at least partially attributable to the downregulation of miR-155.

Energy consumption predictions for residential buildings play an important role in the energy management and control system, as the supply and demand of energy experience dynamic and seasonal changes. In this paper, monthly electricity consumption ratings are precisely classified based on open data in an entire region, which includes over 16 000 residential buildings. First, data mining techniques are used to discover and summarize the electricity usage patterns hidden in the data. Second, the particle swarm optimization-K-means algorithm is applied to the clustering analysis, and the level of electricity usage is divided by the cluster centers. Finally, an efficient classification model using a support vector machine as the basic optimization framework is proposed, and its feasibility is verified. The results illustrate that the accuracy and F-measure of the new model reach 96.8% and 97.4%, respectively, which vastly exceed those of conventional methods. To the best of our knowledge, the research on predicting the electricity consumption ratings of residential buildings in an entire region has not been publicly released. The method proposed in this paper would assist the power sector in grasping the dynamic behavior of residential electricity for supply and demand management strategies and provide a decision-making reference for the rational allocation of the power supply, which will be valuable in improving the overall power grid quality.

High-strength aluminum alloys are exposed to more and more environmentally-induced cracking failure behaviors during service. However, due to the hard to detect nature of hydrogen, and the special working conditions, failure research has obvious hysteresis and complexity, and it is impossible to truly reflect the material failure phenomenon and mechanism. In this paper, 7085 high-strength aluminum alloy is selected as the research material to simulate and reproduce the environmental failure phenomenon of aircraft under extreme working conditions (temperature 70 °C, humidity 85%). The results proved that high-strength aluminum alloy has environmental cracking failure behavior under extreme working conditions. The failure mode that was determined was due to environment-induced hydrogen and hydrogen-induced cracking, which is the result of the combined action of hydrogen and stress. Meanwhile, we demonstrate that high-strength aluminum alloy’s environmental failure behavior in an environment of high temperature and high humidity is different from traditional stress corrosion cracking behavior.

Amphotericin B (AMB) is an antifungal drug used for serious fungal infections. However, AMB has adverse reactions such as nephrotoxicity, which limit the clinical application of AMB alone or in combination with other antifungal drugs. Nano or micro drug delivery systems (DDS) have been proven to be effective in reducing the toxic and side effects of drugs. Further, the combination of AMB with other compounds with antifungal activity, such as curcumin (CM), may enhance the synergistic effects. Herein, AMB and CM were co-loaded into porous poly (lactic-co-glycolic acid) (PLGA) microparticles (MPs) to prepare AMB/CM-PLGA MPs. The AMB/CM-PLGA MPs showed a remarkably reduced hemolysis (62.2 ± 0.6%) compared to AMB (80.9 ± 1.1%). The nephrotoxicity of AMB/CM-PLGA MPs is significantly lower than that of AMB. In vitro, AMB/CM-PLGA MPs had better inhibitory effects on the adhesion and biofilm formation of Candida albicans compared with AMB. Experiments on mice infected with C. albicans showed that AMB/CM-PLGA MPs have a better therapeutic effect than AMB in vivo. In summary, AMB/CM-PLGA MPs may be a novel and promising therapeutic candidate for fungal infection.

Descurainia sophia L. (flixweeds) is a noxious broad-leaf weed infesting winter wheat fields in China that has evolved high resistance to tribenuron-methyl. In this work, a brand new gene CYP77B34 was cloned from tribenuron-methyl-resistant (TR) D. sophia and transferred into Arabidopsis thaliana, and the sensitivities of Arabidopsis with or without the CYP77B34 transgene to herbicides with a different mode of actions (MoAs) were tested. Compared to Arabidopsis expressing pCAMBIA1302-GFP (empty plasmid), Arabidopsis transferring pCAMBIA1302-CYP77B34 (recombinant plasmid) became resistant to acetolactate synthase (ALS)-inhibiting herbicide tribenuron-methyl, protoporphyrinogen oxidase (PPO)-inhibiting herbicides carfentrazone-ethyl and oxyfluorfen. Cytochrome P450 inhibitor malathion could reverse the resistance to tribenuron-methyl, carfentrazone-ethyl and oxyfluorfen in transgenic Arabidopsis plants. In addition, the metabolic rates of tribenuron-methyl in Arabidopsis expressing CYP77B34 were significantly higher than those in Arabidopsis expressing pCAMBIA1302-GFP. Other than that, the transgenic plants showed some tolerance to very-long-chain fatty acid synthesis (VLCFAs)-inhibiting herbicide pretilachlor and photosystem (PS) II-inhibiting herbicide bromoxynil. Subcellular localization revealed that the CYP77B34 protein was located in the endoplasmic reticulum (ER). These results clearly indicated that CYP77B34 mediated D. sophia resistance to tribenuron-methyl and may have been involved in D. sophia cross-resistance to carfentrazone-ethyl, oxyfluorfen, pretilachlor and bromoxynil.

Drainage of freshwater wetlands is common in coastal regions, although the effects on microbial extracellular enzyme activity (a key mediator of soil organic matter decomposition) in relation to spatial variability (microtopography and soil depth) are poorly understood. Soils were collected from organic (Oi, Oe, Oa) and mineral (A, AB, B) horizons from a natural and drained coastal forested wetland in North Carolina, USA. Activity of seven enzymes were measured: α-glucosidase (AG), β-glucosidase (BG), cellobiohydrolase (CBH), xylosidase (XYL), phenol oxidase (POX), peroxidase (PER) and N-acetyl glucosamide (NAG). Enzyme activity rates were normalized by soil weight, soil organic C (SOC), and microbial biomass C (MBC). Specific enzyme activity (per SOC or MBC) was more sensitive to drainage and soil depth compared to normalization by soil weight. In Oi and Oa horizons, specific enzyme activity (per MBC) (AG, BG, XYL, POX, PER) was higher in the natural compared to drained wetland but lower (AG, CBH, XYL, POX, PER, NAG) in the AB or B mineral soils. Results from this study indicate that organic soil horizons of natural freshwater wetlands contain a highly active microbial community driven by inputs of plant-derived C, while deeper soils of the drained wetland exhibit higher microbial metabolic activity, which likely plays a role in SOC storage of these systems.

The structure, morphology, fracture toughness and flaw sensitivity length scale of chain-extended poly(butylene succinate) with various pre-stretch ratios were studied. PBS modification adopted from a multifunctional, commercially available chain-extension containing nine epoxy groups (ADR9) as the first step chain extension and hydroxyl addition modified dioxazoline (BOZ) as the second step. Time-temperature superposition (TTS) studies show that the viscosity increased sharply and the degree of molecular branching increased. Fourier transform infrared spectroscopy (FT-IR) confirm successful chain extension reactions. The orientation of the polymer in the pre-stretch state is such that spherulites deformation along the stretching direction was observed by polarized light optical microscopy (PLOM). The fracture toughness of sample (λfix = 5) is Γ ≈ 106 J m-2 and its critical flaw sensitivity length scale is Γ/Wc ≈ 0.01 m, approximately 5 times higher than PBS without chain-extension (Γ ≈ 2 × 105 J m-2 and Γ/Wc ≈ 0.002 m, respectively). The notch sensitivity of chain-extended PBS is significantly reduced, which is due to the orientation of spherulites more effectively preventing crack propagation. The principle can be generalized to other high toughness material systems.

With the emergence and rapid spread of new pandemic variants, especially variants of concern (VOCs), the development of next-generation vaccines with broad-spectrum neutralizing activities is of great importance. In this study, SCTV01C, a clinical stage bivalent vaccine based on trimeric spike extracellular domain (S-ECD) of SARS-CoV-2 variants Alpha (B.1.1.7) and Beta (B.1.351) with a squalene-based oil-in-water adjuvant was evaluated in comparison to its two corresponding (Alpha and Beta) monovalent vaccines in mouse immunogenicity studies. The two monovalent vaccines induced potent neutralizing antibody responses against the antigen-matched variants, but drastic reductions in neutralizing antibody titers against antigen-mismatched variants were observed. In comparison, the bivalent vaccine SCTV01C induced relatively higher and broad-spectrum cross-neutralizing activities against various SARS-CoV-2 variants, including the D614G variant, VOCs (B.1.1.7, B.1.351, P.1, B.1.617.2, B.1.1.529), variants of interest (VOIs) (C.37, B.1.621), variants under monitoring (VUMs) (B.1.526, B.1.617.1, B.1.429, C.36.3) and other variants (B.1.618, 20I/484Q). All three vaccines elicited potent Th1-biased T-cell immune responses. These results provide direct evidence that variant-based multivalent vaccines could play important roles in addressing the critical issue of reduced protective efficacy against the existing and emerging SARS-CoV-2 variants.
