113. K-PLUG: Knowledge-injected Pre-trained Language Model for Natural Language Understanding and Generation in E-Commerce

Abstract

Existing pre-trained language models (PLMs) have demonstrated the effectiveness of self-supervised learning for a broad range of natural language processing (NLP) tasks. However, most of them are not explicitly aware of domain-specific knowledge, which is essential for downstream tasks in many domains, such as tasks in e-commerce scenarios. In this paper, we propose K-PLUG, a knowledge-injected pre-trained language model based on the encoder-decoder transformer that can be transferred to both natural language understanding and generation tasks. We verify our method in a diverse range of e-commerce scenarios that require domain-specific knowledge. Specifically, we propose five knowledge-aware self-supervised pre-training objectives to formulate the learning of domain-specific knowledge, including e-commerce domain-specific knowledge-bases, aspects of product entities, categories of product entities, and unique selling propositions of product entities. K-PLUG achieves new state-of-the-art results on a suite of domain-specific NLP tasks, including product knowledge base completion, abstractive product summarization, and multi-turn dialogue, significantly outperforms baselines across the board, which demonstrates that the proposed method effectively learns a diverse set of domain-specific knowledge for both language understanding and generation tasks., Comment: Accepted by Findings of EMNLP 2021

134. Surgical treatment of specific Unified Classification System B fractures: potentially destabilising lesser trochanter periprosthetic fractures

Abstract

Abstract To investigate the clinical effects of specific Unified Classification System B (UCS B)-lesser trochanter periprosthetic fractures and determine whether they occur only with non-cemented stems. A retrospective analysis of 28 patients with specific UCS B2 fractures who underwent two surgical treatments, longer stem revision and internal fixation (LSRIF) and open reduction and internal fixation (ORIF), was performed. The patients were assessed at 1, 3, 6, 12, and 24 months and annually thereafter. Fracture healing, complications, Harris Hip Score (HHS), and the Short Form Health Survey questionnaire (SF-36) quality of life score were assessed at each follow-up. At the time of the last follow-up, seven patients had been lost: three were lost to contact, two died, and two were hospitalised elsewhere and unavailable for follow-up. The remaining 21 patients were followed for an average of 49.3 ± 15.4 (range: 24–74.4) months. Their average fracture healing time was 13.5 ± 1.1 (12–15.4) weeks. Complications included three cases (10.71%) of thrombus, one (3.57%) of heart failure, and one (3.57%) of pulmonary infection. There were no revisions due to prosthesis loosening, subsidence, or infection. At the last follow-up, the HHS, SF-36 mental score, and SF-36 physical score were recorded, LSRIF vs. ORIF (82.9 ± 6.6 vs. 74.7 ± 3.9, p = 0.059; 50.9 ± 7.6 vs. 38 ± 1.4, p = 0.012, and 51.7 ± 8.4 vs. 39.7 ± 3.4, p = 0.032, respectively). Specific UCS B2 fractures mostly occur with non-cemented stems. LSRIF with cables is the main treatment, while ORIF is an option for those elderly in poor condition.

Abstract

Abstract The Santos Basin, located in the southeastern waters of Brazil, is a passive continental margin basin with the most abundant deepwater petroleum resources in the world discovered to date. However, few studies have been conducted on the present geothermal fields of the Santos Basin, which severely restricts the oil and gas resource evaluation of the basin. This study first utilizes 35 temperature data from 16 post-salt drilling wells and 370 temperature data from 31 pre-salt drilling wells to calculate the post-salt and pre-salt geothermal gradients and terrestrial heat flows in the Santos Basin. Then, the basin simulation software BasinMod 1D is used to quantitatively evaluate the impacts of salt rock sedimentation on the present geothermal fields and the maturity of pre-salt hydrocarbon source rocks. The results demonstrate that the present post-salt geothermal gradient in the Santos Basin is 2.20–3.97 °C/100 m, with an average value of 2.99 °C /100 m, and the post-salt terrestrial heat flow is 54.00–97.32 mW/m2, with an average value of 73.36 mW/m2, while the present pre-salt geothermal gradient is 2.21–2.95 °C/100 m, with an average value of 2.53 °C/100 m, and the pre-salt terrestrial heat flow is 61.85–82.59 mW/m2, with an average value of 70.69 mW/m2. These values are characteristic of a low-temperature geothermal field in a zone with a stable structure. The sedimentation of the salt rock causes a decrease in the temperature of the pre-salt strata, which inhibits pre-salt hydrocarbon source rock maturity, with an inhibition rate of up to 1.32%. The inhibition degree decreases with increasing salt rock thickness. At the same time, the salt rock thickness is positively correlated with the present surface heat flow. The unique distribution of the salt rock and related salt structures lead to present terrestrial heat flow differences among different structural units in the basin. This study is of great significance for evaluating and exploring the pre-salt oil and gas resources in the Santos Basin.

Abstract

Investigating large-scale spatial patterns of ecosystem services (ESs) and their underlying drivers can greatly contribute to policies-making and regional sustainability development. With water yield (WY), soil conservation (SC), and carbon sequestration (CS) as representative ESs, we aim to quantify their spatial patterns in the Yangtze River Economic Belt, China, and to identify their driving factors, and to formulate sound environmental management strategies. Spatial geography and socioeconomic data from 2000 to 2020 were mined and a range of research methods, including multiscale geographic weighted regression, self-organizing maps, and linear discriminant analysis, were employed for such a purpose. Annual average WY, SC, and CS were 403 mm, 9897 t·km−1, and 1071 g·CO2·m−2. The three ESs examined exhibit spatial heterogeneity. WY exhibited significant patterns of variation along the north–south gradient, while SC and CS exhibited significant variation along the topographic gradient. In the context of high correlation of driving factors among ESs, WY and SC exhibited a greater sensitivity to natural factors (such as precipitation), while CS demonstrated a height sensitivity to human activities in addition to vegetation cover. Spatial heterogeneity is pronounced among the main driving factors of ESs. Three threshold equations were established to describe the manner in which driving factors of different regional ecosystem services undergo transformations, equations possessed a high level of credibility in this study (coincidence > 80 %). This study reveals spatial variations in ecosystem services and their natural and socio-economic drivers. More specifically, we quantitatively validated the threshold in the expression of ecosystem service drivers, establishing a strong scientific foundation for regional ecosystem conservation and management.

137. PEDV-spike-protein-expressing mRNA vaccine protects piglets against PEDV challenge

Abstract

ABSTRACT Porcine epidemic diarrhea virus (PEDV), a swine enteropathogenic coronavirus, causes severe diarrhea in neonatal piglets, which is associated with a high mortality rate. Thus, developing effective and safe vaccines remains a top priority for controlling PEDV infection. Here, we designed two lipid nanoparticle (LNP)-encapsulated mRNA (mRNA-LNP) vaccines encoding either the full-length PEDV spike (S) protein or a multiepitope chimeric spike (Sm) protein. We found that the S mRNA-LNP vaccine was superior to the Sm mRNA-LNP vaccine at inducing antibody and cellular immune responses in mice. Evaluation of the immunogenicity and efficacy of the S mRNA vaccine in piglets confirmed that it induced robust PEDV-specific humoral and cellular immune responses in vivo. Importantly, the S mRNA-LNP vaccine not only protected actively immunized piglets against PEDV but also equipped neonatal piglets with effective passive anti-PEDV immunity in the form of colostrum-derived antibodies after the immunization of sows. Our findings suggest that the PEDV-S mRNA-LNP vaccine is a promising candidate for combating PEDV infection.IMPORTANCEPorcine epidemic diarrhea virus (PEDV) continues to harm the global swine industry. It is important to develop a highly effective vaccine to control PEDV infection. Here, we report a PEDV spike (S) mRNA vaccine that primes a potent antibody response and antigen-specific T-cell responses in immunized piglets. Active and passive immunization can protect piglets against PED following the virus challenge. This study highlights the efficiency of the PEDV-S mRNA vaccine and represents a viable approach for developing an efficient PEDV vaccine.

139. Brevicidine acts as an effective sensitizer of outer membrane-impermeable conventional antibiotics for Acinetobacter baumannii treatment

Abstract

The antibiotic resistance of Acinetobacter baumannii poses a significant threat to global public health, especially those strains that are resistant to carbapenems. Therefore, novel strategies are desperately needed for the treatment of infections caused by antibiotic-resistant A. baumannii. In this study, we report that brevicidine, a bacterial non-ribosomally produced cyclic lipopeptide, shows synergistic effects with multiple outer membrane-impermeable conventional antibiotics against A. baumannii. In particular, brevicidine, at a concentration of 1 μM, lowered the minimum inhibitory concentration of erythromycin, azithromycin, and rifampicin against A. baumannii strains by 32–128-fold. Furthermore, mechanistic studies were performed by employing erythromycin as an example of an outer membrane-impermeable conventional antibiotic, which showed the best synergistic effects with brevicidine against the tested A. baumannii strains in the present study. The results demonstrate that brevicidine disrupted the outer membrane of A. baumannii at a concentration range of 0.125–4 μM in a dose-dependent manner. This capacity of brevicidine could help the tested outer membrane-impermeable antibiotics enter A. baumannii cells and thereafter exert their antimicrobial activity. In addition, the results show that brevicidine–erythromycin combination exerted strong A. baumannii killing capacity by the enhanced inhibition of adenosine triphosphate biosynthesis and accumulation of reactive oxygen species, which are the main mechanisms causing the death of bacteria. Interestingly, brevicidine and erythromycin combination showed good therapeutic effects on A. baumannii-induced mouse peritonitis–sepsis models. These findings demonstrate that brevicidine is a promising sensitizer candidate of outer membrane-impermeable conventional antibiotics for treating A. baumannii infections in the post-antibiotic age.

Abstract

Dental implants are key components in treating dental diseases and correcting dental defects, and they have gained widespread utilisation in various medical operations. However, traditional screw thread implants have been associated with certain drawbacks, including issues such as poor consistency with the alveolar bone and difficulty in customisation. Root analogue implants (RAI) are considered a promising substitute; however, they are not widely applied until now as their geometry design needs more improvements. The utilisation of porous structures, such as triply periodic minimal surfaces, in dental implants can reduce the elastic modulus, increase interconnectivity, and allow for strictly controllable structural characteristics. To achieve optimal initial and long-term stability, it is essential to have a moderate geometry that promotes a proper fit between the dental implants and the natural bone. Different porous and macro-retention structures of RAIs have been investigated due to the above two demands. However, owing to their complex porosity, such structures can only be effectively manufactured using additive manufacturing techniques. Various additive manufacturing technologies have been employed for the fabrication of dental implants and have provided good surface quality and mechanical properties. In vivo tests have demonstrated that their performance is similar to that of commercial implants fabricated using subtractive manufacturing techniques. In this review, we comprehensively examine the design concepts and processes involved in the development of dental implants, along with the application of additive manufacturing methods for their fabrication, while also offering recommendations for the future advancement of dental implant designs.

148. Study on anti-impact support structure performance of multi-direction energy absorption for roadway

Abstract

Aiming at the problem of insufficient impact resistance of the existing portal support, a kind of anti-impact support structure for multi-direction energy absorption was constructed by adding V-shaped groove to the beam structure and flexible chain support structures on both sides. Parameters such as total energy absorption, plastic strain and vertical displacement were used as analysis indexes to study the supporting performance of the support in the face of vertical impact and lateral impact, and the deformation degree for each structure of the support and the overall most vulnerable position were obtained. Under the vertical impact condition, the equivalent plastic strain position of the new support appeared at the middle joint of the top beam, and then the joint of the top beam should be thickened. In the whole process of vertical impact, the vertical displacement of the three hydraulic columns was not average, which indicated that the support was not very ideal in the face of concentrated impact. Under vertical impact conditions, the V-shaped groove had a small deformation in the face of the impact with large velocity, and a large deformation in the face of the impact with small velocity, which showed that the V-shaped groove had a certain response speed in the face of impact. Under lateral impact conditions, the equivalent plastic strain of the support was mainly concentrated at the connection between the flexible chain and the support when the impact velocity is low, and the annular base should be installed at the connection between the flexible chain and the support. The bearing capacity and total energy absorption of flexible chain support structure under lateral impact were slightly weaker than that of support under vertical impact, but they can play a good role in horizontal support.

149. Mucus-penetrating nonviral gene vaccine processed in the epithelium for inducing advanced vaginal mucosal immune responses

Abstract

Establishment of vaginal immune defenses at the mucosal interface layer through gene vaccines promise to prevent infectious diseases among females. Mucosal barriers composed of a flowing mucus hydrogel and tightly conjugated epithelial cells (ECs), which represent the main technical difficulties for vaccine development, reside in the harsh, acidic human vaginal environment. Different from frequently employed viral vectors, two types of nonviral nanocarriers were designed to concurrently overcome the barriers and induce immune responses. Differing design concepts include the charge-reversal property (DRLS) to mimic a virus that uses any cells as factories, as well as the addition of a hyaluronic acid coating (HA/RLS) to directly target dendritic cells (DCs). With a suitable size and electrostatic neutrality, these two nanoparticles penetrate a mucus hydrogel with similar diffusivity. The DRLS system expressed a higher level of the carried human papillomavirus type 16 L1 gene compared to HA/RLS in vivo. Therefore it induced more robust mucosal, cellular, and humoral immune responses. Moreover, the DLRS applied to intravaginal immunization induced high IgA levels compared with intramuscularly injected DNA (naked), indicating timely protection against pathogens at the mucus layer. These findings also offer important approaches for the design and fabrication of nonviral gene vaccines in other mucosal systems.
