Abstract Background Flowering is a critical physiological change that interferes with not only biomass yield but also secondary metabolism, such as the biosynthesis of flavonoids, in rhizome/root plants. The continuous inflorescence removal (CIR) treatment is frequently conducted to weaken this effect. Fagopyrum dibotrys (D.Don) H.Hara (Golden buckwheat) is a kind of rhizome medicinal plant rich in flavonoids and is widely used for the treatment of lung diseases. The CIR treatment is usually conducted in F. dibotrys because of its excessive reproductive growth. To uncover the molecular mechanisms, comprehensive analysis was performed using metabolome and transcriptome data obtained from normally bloomed and the CIR treated plants. Results Metabolome results demonstrated that in the rhizomes of F. dibotrys, its bioactive compound called epicatechin has higher amount than most of the detected precursors. Compared with the normally bloomed plants, the level of epicatechin in the rhizomes of the CIR group increased by 25% at the withering stage. Based on 96 samples of the control and the CIR groups at 4 flowering stages for 4 tissues, RNA-Seq results revealed a 3 ~ 5 times upregulations of all the key enzyme genes involved in the biosynthesis of epicatechin in both time (from the bud stage to the withering stage) and spatial dimensions (from the top of branch to rhizome) under the CIR treatment compared to normal flowering. Integrated analysis of LC–MS/MS and transcriptome revealed the key roles of several key enzyme genes besides anthocyanidin reductase (ANR). A total of 93 transcription factors were identified to co-expressed with the genes in epicatechin biosynthetic pathway. The flowering activator SQUAMOSA promoter-binding protein like (SPLs) exhibited opposite spatiotemporal expression patterns to that of the epicatechin pathway genes; SPL3 could significantly co-express with all the key enzyme genes rather than the flowering repressor DELLA. Weighted gene co-expression network analysis (WGCNA) further confirmed the correlations among chalcone synthases (CHSs), chalcone isomerases (CHIs), ANRs, SPLs and other transcription factors. Conclusions SPL3 might dominantly mediate the effect of normal flowering and the CIR treatment on the biosynthesis of epicatechin in rhizomes mainly through the negative regulations of its key enzyme genes including CHS, CHI and ANR.

IntroductionStroke-associated pneumonia (SAP) is a common complication of stroke that can increase the mortality rate of patients and the burden on their families. In contrast to prior clinical scoring models that rely on baseline data, we propose constructing models based on brain CT scans due to their accessibility and clinical universality.MethodsOur study aims to explore the mechanism behind the distribution and lesion areas of intracerebral hemorrhage (ICH) in relation to pneumonia, we utilized an MRI atlas that could present brain structures and a registration method in our program to extract features that may represent this relationship. We developed three machine learning models to predict the occurrence of SAP using these features. Ten-fold cross-validation was applied to evaluate the performance of models. Additionally, we constructed a probability map through statistical analysis that could display which brain regions are more frequently impacted by hematoma in patients with SAP based on four types of pneumonia.ResultsOur study included a cohort of 244 patients, and we extracted 35 features that captured the invasion of ICH to different brain regions for model development. We evaluated the performance of three machine learning models, namely, logistic regression, support vector machine, and random forest, in predicting SAP, and the AUCs for these models ranged from 0.77 to 0.82. The probability map revealed that the distribution of ICH varied between the left and right brain hemispheres in patients with moderate and severe SAP, and we identified several brain structures, including the left-choroid-plexus, right-choroid-plexus, right-hippocampus, and left-hippocampus, that were more closely related to SAP based on feature selection. Additionally, we observed that some statistical indicators of ICH volume, such as mean and maximum values, were proportional to the severity of SAP.DiscussionOur findings suggest that our method is effective in classifying the development of pneumonia based on brain CT scans. Furthermore, we identified distinct characteristics, such as volume and distribution, of ICH in four different types of SAP.

Alzheimer disease (AD) represents an oncoming epidemic that without an effective treatment promises to exact extraordinary human and financial burdens. Studies of pathogenesis are essential for defining targets for discovering disease-modifying treatments. Past studies of AD neuropathology provided valuable, albeit limited, insights. Nevertheless, building on these findings, recent studies have provided an increasingly rich harvest of genetic, molecular and cellular data that are creating unprecedented opportunities to both understand and treat AD. Among the most significant are those documenting the presence within the AD brain of toxic oligomeric species of Aβ and tau. Existing data support the view that such species can propagate and spread within neural circuits. To place these findings in context we first review the genetics and neuropathology of AD, including AD in Down syndrome (AD-DS). We detail studies that support the existence of toxic oligomeric species while noting the significant unanswered questions concerning their precise structures, the means by which they spread and undergo amplification and how they induce neuronal dysfunction and degeneration. We conclude by offering a speculative synthesis for how oligomers of Aβ and tau initiate and drive pathogenesis. While 100 years after Alzheimer's first report there is much still to learn about pathogenesis and the discovery of disease-modifying treatments, the application of new concepts and sophisticated new tools are poised to deliver important advances for combatting AD.

Alzheimer disease (AD) represents an oncoming epidemic that without an effective treatment promises to exact extraordinary financial and emotional burdens (Apostolova, 2016). Studies of pathogenesis are essential for defining critical molecular and cellular events and for discovering therapies to prevent or mitigate their effects. Through studies of neuropathology, genetic and cellular, and molecular biology recent decades have provided many important insights. Several hypotheses have been suggested. Documentation in the 1980s of selective loss of cholinergic neurons of the basal forebrain, followed by clinical improvement in those treated with inhibitors of acetylycholinesterase, supported the "cholinergic hypothesis of age-related cognitive dysfunction" (Bartus et al., 1982). A second hypothesis, prompted by the selective loss of cholinergic neurons and the discovery of central nervous system (CNS) neurotrophic factors, including nerve growth factor (NGF), prompted the "deficient neurotrophic hypothesis" (Chen et al., 2018). The most persuasive hypothesis, the amyloid cascade hypothesis first proposed more than 25 years ago (Selkoe and Hardy, 2016), is supported by a wealth of observations. Genetic studies were exceptionally important, pointing to increased dose of the gene for the amyloid precursor protein (APP) in Down syndrome (DS) and a familial AD (FAD) due to duplication of APP and to mutations in APP and in the genes for Presenilin 1 and 2 (PSEN1, 2), which encode the γ-secretase enzyme that processes APP (Dorszewska et al., 2016). The "tau hypothesis" noted the prominence of tau-related pathology and its correlation with dementia (Kametani and Hasegawa, 2018). Recent interest in induction of microglial activation in the AD brain, as well as other manifestations of inflammation, supports the "inflammatory hypothesis" (Mcgeer et al., 2016). We place these findings in the context of the selective, but by no means unique, involvement of BFCNs and their trophic dependence on NGF signaling and speculate as to how pathogenesis in these neurons is initiated, amplified and ultimately results in their dysfunction and death. In so doing we attempt to show how the different hypotheses for AD may interact and reinforce one another. Finally, we address current attempts to prevent and/or treat AD in light of advances in understanding pathogenetic mechanisms and suggest that studies in the DS population may provide unique insights into AD pathogenesis and treatment.

Porous liquids are potentially useful materials for the identification and separation of non-gaseous compounds. Herein, the authors report a type II porous ionic liquid with permanent porosity and high selectivity towards l-tryptophan (l-Trp) over other aromatic amino acids.

The cytosolic chaperonin T-complex protein (TCP) 1-ring complex (TRiC) has been shown to exert neuroprotective effects on axonal transport through clearance of mutant Huntingtin (mHTT) in Huntington's disease. However, it is presently unknown if TRiC also has any effect on axonal transport in wild-type neurons. Here, we examined how TRiC impacted the retrograde axonal transport of brain-derived neurotrophic factor (BDNF). We found that expression of a single TRiC subunit significantly enhanced axonal transport of BDNF, leading to an increase in instantaneous velocity with a concomitant decrease in pauses for retrograde BDNF transport. The transport enhancing effect by TRiC was dependent on endogenous tau expression because no effect was seen in neurons from tau knockout mice. We showed that TRiC regulated the level of cyclin-dependent kinase 5 (CDK5)/p35 positively, contributing to TRiC-mediated tau phosphorylation (ptau). Expression of a single TRiC subunit increased the level of ptau while downregulation of the TRiC complex decreased ptau. We further demonstrated that TRiC-mediated increase in ptau induced detachment of tau from microtubules. Our study has thus revealed that TRiC-mediated increase in tau phosphorylation impacts retrograde axonal transport.

Objective To study the mechanism of miR-30b by regulating fragile histidine triad gene(FHIT) on cell activity diffuse large B-cell lymphoma(DLBCL). Methods The tumor paraffin specimens of DLBCL patients and the paraffin markers of lymph node hyperplasia at the same time were selected from the Shanghai Minhang District Tumor Hospital (February 2017-June 2019) and divided them into the DLBCL group and the control group. DLBCL cells were divided into the WW group (DLBCL non transfected group), WZ group (DLBCL transfected NC group )and ZR group (DLBCL transfected miR-30b group). RT-qPCR method was used to detect miR-30b expression; Western blot was used to detect FHIT protein expression; colorimetric method was used to detect caspase-3 activity; Flow cytometry was used to detect DLBCL cell apoptosi and Transwell method was applied to detect cell invasion. Results The expression of miR-30b and FHIT in the control group was significantly higher than those in the DLBCL group (P＜0.05); the expression of miR-30b, FHIT and caspase-3 in the ZR group was higher than those in the WW and WZ groups (P＜0.05) The apoptosis rate of ZR group was time-dependently higher than that of WW and WZ groups(P＜0.05); the counting of DLBCL cell invasion in ZR group was significantly less than that of WW and WZ groups (P＜0.05). Conclusions After miR-30b transfection, the apoptosis of diffuse large B-cell lymphoma cells increases and the counting of invasion cells decreases. The mechanism of this result may be related to the enhancement of miR-30b, FHIT, and caspase-3 level.

Integrating solar thermal conversion with phase change materials (PCMs) offers a promising pathway for continuous thermal energy generation with a zero‐carbon footprint. However, substantial infrared radiation losses at elevated temperatures often hinder the efficiency of such integrated systems. Inspired by the thermoregulation mechanisms of polar bears, this work introduces composite PCMs with spectrally selective absorption to enhance solar thermal energy storage efficiency. These composite phase change materials (CPCMs), featuring densely packed SiC ceramic grains with high porosity, exhibit a thermal conductivity of up to 14 W m−1 K−1 and an energy storage density of 195.1 kJ kg−1. The incorporation of nanoparticle‐coated foil induces a plasmonic effect that increases solar absorptivity to 90.57% and reduces infrared emissivity from 71.94% to 7.47%. Consequently, the solar thermal storage efficiency is significantly increased from 54.56% to 81.65% at 500 K, effectively addressing the challenges associated with high‐temperature solar thermal storage. Additionally, the low infrared emissivity of the c (PNC), combined with the inherent heat absorption properties of PCMs, enables infrared stealth functionality. These multifunctional CPCMs demonstrate considerable potential for advancing high‐temperature solar thermal storage technologies and other heat‐related applications. [ABSTRACT FROM AUTHOR]

In the field of nanotechnology, rotary jet spinning technology has garnered attention due to its unique advantages in fabricating composite nanofibers. The composite nanofibers prepared using this technology exhibit exceptional purity, uniformity, and consistency, thereby enhancing reliability and controllability in practical applications. During the process of rotary jet spinning, the spinning solution flows into the nozzle and converges at a micro‐triangle under the combined influence of centrifugal force, viscous force, surface tension, and gravity. Stepped jet refers to the flow phenomenon occurring within a channel with a gradually decreasing cross‐sectional area. In this process, an increase in the flow velocity of the composite spinning solution aids in stabilizing stretching at the micro‐triangle which is crucial for forming continuous jets. This article analyzes cone formation mechanisms for composite spinning solutions and stepped jet principles while establishing corresponding models that reflect relationships between solution velocity magnitude and parameters such as rotational speed, concentration, and nozzle diameter. Finally, polyethylene oxide (PEO)/polyvinylpyrrolidone (PVP) composite nanofibers were prepared using a rotary jet spinning device where experimental investigation focused on examining how parameters such as rotational speed, solution concentration, and nozzle diameter impact motion of spinning solution within micro‐triangle. The rotary jet spinning process involves the joint action of centrifugal force, gravity, temperature, humidity, and flow fields on the spinning solution. The solution is ejected through the nozzle after flowing from the storage container and stretched in the air to form a composite fiber. At present, the investigation of rotary jet composite spinning primarily focused on the dynamics and kinematics of the spinning solution and jet within the container. However, there is still a lack of comprehensive exploration into the motion and slip phenomena exhibited by the composite spinning solution in air. This article investigates the slip phenomenon between the polymer and the gas contact surface after the spinning solution leaves the spinneret aperture. A slip model is established to analyze the motion and force of the polymer in air. Meanwhile, a mathematical model for the evaporation rate of composite spinning solution motion under the influence of multi‐field coupling is established through theoretical analysis. A numerical simulation of the rotary jet spinning process was conducted to obtain clouds of jet exit velocity. The morphology of composite fibers produced by rotary jet spinning was analyzed using scanning electron microscopy, enabling a comparison of diameter distribution and surface quality under different environmental and equipment parameters. Lycium barbarum has been widely planted in arid and semi-arid areas due to its drought-resistant ability, which is of great economic value as a medicinal and edible homology plant. In this study, the metabolome of the L. barbarum variety "Ningqi 7" under different drought stress conditions was compared and analyzed by the non-targeted UPLC-MS (ultra-high performance liquid chromatography with mass spectrometry) technique. The results showed that drought stress significantly decreased the water content of leaves, increased the activity of antioxidant enzymes in plants, and up-regulated the metabolites and pathways involved in osmoregulation, antioxidant stress, energy metabolism, and signal transduction. Under moderate drought (40–45% FC), L. barbarum accumulated osmoregulatory substances mainly through the up-regulation of the arginine metabolism pathway. At the same time, phenylalanine metabolism and cutin, suberine, and wax biosynthesis were enhanced to improve the antioxidant capacity and reduce water loss. However, in severe drought (10–15% FC), L. barbarum shifted to up-regulate purine metabolism and lysine degradation and redistributed energy and nitrogen resources. In addition, vitamin B6 metabolism was significantly upregulated in both groups of stress levels, playing a key role in antioxidant and growth regulation. Neurotrophic factors, including the members of the neurotrophin family, play important roles in the development and maintenance of the nervous system. Trophic factor signals must be transmitted over long distances from axons and dendrites to the cell bodies of neurons. A mode of signaling well suited to the challenge of robust long distance signaling is the signaling endosome. We review the biology of signaling endosomes and the "signaling endosome hypothesis". Evidence for disruption of signaling endosome function in disorders of the nervous system is also reviewed. Changes in endosome structure in Alzheimer disease (AD) and Down syndrome (DS) are present early in these disorders. Data for the APP products responsible are reviewed and the consequent changes in signaling from endosomes discussed. We conclude by pointing to the need for additional studies to explore the biology of signaling endosomes in normal neurons and to elucidate their role in the pathogenesis of neurodegeneration.

Intercropping achieved through agroforestry is increasingly being recognized as a sustainable form of land use. In agroforestry, the roots of trees and crops are intermingled, and their interactions and the production of exudates alter the soil environment and soil microbial community. Although tree–crop interactions vary depending on the stand age of the trees, how stand age affects beneficial microorganisms, including arbuscular mycorrhizal fungi (AMF), and whether changes in soil microorganisms feed back on crop growth in agroforestry systems are unknown. We therefore conducted a long-term field study to compare changes in the soil microbial and AMF communities in a jujube/wheat agroforestry system containing trees of different stand ages: 3-year-old jujube, 8-year-old jujube, and 13-year-old jujube. Our results showed that by changing soil moisture and available phosphorus content, the stand age of the trees had a significant effect on the soil microbial and AMF communities. Soil moisture altered the composition of soil bacteria, in particular the proportions of Gram-positive and Gram-negative species, and available phosphorus had significant effects on the AMF community. A network analysis showed that older stands of trees reduced both AMF diversity and network complexity. An ordinary least squares regression analysis indicated that AMF diversity, network complexity, and stability contributed to wheat yield. Finally, structural equation modeling showed that changes in edaphic factors induced by tree age brought about significant variation in the soil microbial and AMF communities, in turn, affecting crop growth. Our study highlights the crucial roles of soil microorganisms, in particular AMF, in supporting plant growth in agroforestry systems as well as the need to consider stand age in the establishment of these systems.
