The recent ban on the use of antibiotics as a feed additive has led to the search for alternative sources of antibiotics in the feed industry. Presently, probiotics are considered as a potential substitute for antibiotic as a live biotherapeutic agent to improve animal health and performance. Accordingly, study was focused on evaluating the effect of Saccharomyces boulardii (Sb) and Bacillus subtilis B10 (Bs) on ultrastructure modulation and mucosal immunity development in broiler chickens. A total of three hundred 1-d-old Sanhuang broilers (a Chinese cross breed) were randomized into 3 groups, each group with 5 replications (n = 20). The control group (Ctr) was fed a basal diet containing an antibiotic (virginiamycin, 20 mg/kg). Meanwhile, broilers in experimental groups received Sb and Bs (1 × 10(8) cfu/kg of feed) in addition to the basal diet for 72 d. The results of the experimental groups revealed a significant improvement in live BW and relative weight of bursa of Fabricius and thymus. Also, intestinal villus height, width, and number of goblet cells increased in the Sb and Bs groups. Meanwhile, modulation in the intestinal ultrastructure and increased mRNA expression levels of occluding, cloudin2, and cloudin3 (P < 0.05) were observed in the Sb and Bs groups. Moreover, IgA-positive cells significantly increased in the jejunum of Sb- and Bs-supplemented groups (P < 0.05). Intestinal cytokines interleukin-6, tumor necrosis factor-α, interleukin-10, transforming growth factor-β, and secretory IgA concentrations were (P < 0.05) improved in the probiotic groups; however, Sb induced inflammatory and antiinflammatory cytokines (P < 0.05) in comparison with the Ctr group. The present findings conclusively revealed that Sb and Bs increased IgA-positive cells in the lumen of the intestinal villus and revealed that Sb and Bs could modulate intestinal ultrastructure through increasing occluding, cloudin2, and cloudin3 mRNA expression levels in broiler intestine.

The transcription factors E2A and HEB (members of the E protein family) have been shown to play essential roles in lymphocyte development, while their negative regulators, the Id proteins, have been implicated in both lymphocyte development and in the CD8(+) T-cell immune response. Here, we show that E proteins also influence CD8(+) T cells responding to infection. E protein expression was upregulated by CD8(+) T cells during the early stages of infection and increased E protein DNA-binding activity could be detected upon TCR stimulation. Deficiency in the E proteins, E2A and HEB, led to increased frequency of terminally differentiated effector KLRG1(hi) CD8(+) T cells in mice during infection, and decreased generation of longer-lived memory-precursor cells during the immune response. These data suggest a model whereby E protein transcription factor activity favors rapid memory-precursor T-cell formation while their negative regulators, Id2 and Id3, are both required for robust effector CD8(+) T-cell response during infection., (© 2012 WILEY-VCH Verlag GmbH & Co. Microorganisms in nitrogen cycle serve as an important part of the ecological function of soil. The aim of this research was to monitor the abundance of nitrogen-fixing, denitrifying and nitrifying bacteria during bioaugmentation of petroleum-contaminated soil using real-time polymerase chain reaction (real-time PCR) of nifH, narG and amoA genes which encode the key enzymes in nitrogen fixation, nitrification and ammoniation respectively. Three different kinds of soils, which are petroleum-contaminated soil, normal soil, and remediated soil, were monitored. It was shown that the amounts of functional microorganisms in petroleum-contaminated soil were far less than those in normal soil, while the amounts in remediated soil and normal soil were comparable. Results of this experiment demonstrate that nitrogen circular functional bacteria are inhibited in petroleum-contaminated soil and can be recovered through bioremediation. Furthermore, copies of the three functional genes as well as total petroleum hydrocarbons (TPH) for soils with six different treatments were monitored. Among all treatments, the one, into which both E. cloacae as an inoculant and wheat straw as an additive were added, obtained the maximum copies of 2.68 x 10(6), 1.71 x 10(6) and 8.54 x 10(4) per gram dry soil for nifH, narG and amoA genes respectively, companying with the highest degradation rate (48% in 40 days) of TPH. The recovery of functional genes and removal of TPH were better in soil inoculated with E cloacae and C echinulata collectively than soil inoculated with E cloacae only. All above results suggest that the nitrogen circular functional genes could be applied to monitor and assess the bioremediation of petroleum-contaminated soil.

Purpose: We identified the profile of lower urinary tract changes in ketamine users in the community. In addition, we identified the relative risks of dose, frequency of ingestion and duration of ketamine use for changes in lower urinary tract function., Materials and Methods: A mobile medical assessment service was established at specific youth centers, and subjects who were known to social workers and who had a history of ketamine use were invited to participate in health screening. Lower urinary tract function was evaluated using the Pelvic Pain, Urgency and Frequency questionnaire, and uroflowmetry and ultrasonography., Results: Use of ketamine more than 3 times weekly was significantly associated with lower voided volumes. Pelvic Pain, Urgency and Frequency questionnaire scores were significantly higher for ketamine use for more than 24 months compared to use for short durations (7.82 vs 6.00). The scores on the symptom and bother subscales of the Pelvic Pain, Urgency and Frequency questionnaire decreased progressively with increased duration of abstinence. For individuals after 1 year of abstinence the Pelvic Pain, Urgency and Frequency questionnaire scores were significantly lower and voided volumes were higher than those for active users., Conclusions: Ketamine users with at least a 2-year habit of 3 or more hits per week have altered bladder function that can be recognized and that causes bother. These early functional changes have the potential to normalize after 1 year of ketamine abstinence. Background: On September 21, 2009, China began administering vaccines, obtained from 10 different manufacturers, against 2009 pandemic influenza A (H1N1) virus infection in priority populations. We aimed to assess the safety of this vaccination program., Methods: We designed a plan for passive surveillance for adverse events after immunization with the influenza A (H1N1) vaccine. Physicians or vaccination providers were required to report the numbers of vaccinees and all adverse events to their local Center for Disease Control and Prevention (CDC), which then reported the data to the Chinese CDC through the online National Immunization Information System's National Adverse Event Following Immunization Surveillance System. Data were collected through March 21, 2010, and were verified and analyzed by the Chinese CDC., Results: A total of 89.6 million doses of vaccine were administered from September 21, 2009, through March 21, 2010, and 8067 vaccinees reported having an adverse event, for a rate of 90.0 per 1 million doses. The age-specific rates of adverse events ranged from 31.4 per 1 million doses among persons 60 years of age or older to 130.6 per 1 million doses among persons 9 years of age or younger, and the manufacturer-specific rates ranged from 4.6 to 185.4 per 1 million doses. A total of 6552 of the 8067 adverse events (81.2%; rate, 73.1 per 1 million doses) were verified as vaccine reactions; 1083 of the 8067 (13.4%; rate, 12.1 per 1 million doses) were rare and more serious (vs. common, minor events), most of which (1050) were allergic reactions. Eleven cases of the Guillain-Barré syndrome were reported, for a rate of 0.1 per 1 million doses, which is lower than the background rate in China., Conclusions: No pattern of adverse events that would be of concern was observed after the administration of influenza A (H1N1) vaccine, nor was there evidence of an increased risk of the Guillain-Barré syndrome.

Objective: To investigate the serum level of free fatty acid (FFA) and explore its relationship with cytokines and atherosclerosis (AS) in chronic kidney disease (CKD)., Methods: The serum level of FFA was determined with enzymatic colorimetry. IL-1β, IL-6 and TNFα were determined with ELISA. High-sensitivity C-reactive protein (hsCRP) was measured with immunoturbidimetry. Prevalence of atherosclerosis was detected with carotid ultrasonography. We evaluated the relationship between serum levels of FFA and IL-1β, IL-6, TNFα, hsCRP as well as the renal function in 130 adult patients with CKD, stratified according to the GFR (based on the National Kidney Foundation/Kidney Dialysis Outcomes Quality Initiatives) and in 58 hemodialytic (HD) patients. The relationship between FFA level and cardiac geometry incidence in CKD patients was analyzed with logistic regression model., Results: The serum level of FFA was significantly higher in CKD patients as compared with that in the healthy controls [(492.63±143.59) vs (302.65±142.18) µmol/L, P<0.01], even in the early stage of CKD. The level of FFA increased with the progression of renal dysfunction. In the non-dialytic CKD group, the level of FFA was negatively related to GFR and positively related to the proteinuria (P<0.05), while in the HD group, it was positively correlated with dialysis duration (P<0.05). The serum levels of FFA were higher in CKD patients with carotid artery atherosclerosis than those in patients without (P<0.05or<0.01). However, in both groups with impairment of renal function, the levels of FFA were positively correlated with hsCRP, IL-1β, IL-6, TNFα and TG (all P<0.05). A positive correlation between the level of FFA and the clinical manifestations such as carotid intimal medial thickness (IMT) and AS was also found. A negative correlation was found between the level of FFA and the serum level of albumin and GFR (P<0.05)., Conclusion: Serum levels of FFA are significantly higher either in non-dialytic CKD or in HD patients and it is related with hsCRP, IL-1β, IL-6, TNFα as well as carotid artery atherosclerosis, indicating that FFA is an independent risk factor of AS in CKD.

Objective: To explore the effects of Cordyceps extract in regulating the imbalance of Th1/Th2 ratio and inhibiting the inflammatory reaction, and to find the theoretical basis of Cordyceps extract for treating asthma in remission stage., Methods: A total of 20 peripheral venous blood samples (3 mL) were collected from 20 asthmatic children during remission stage, and peripheral blood mononuclear cells (PBMCs) were separated by Ficoll method. PBMCs were separated into three groups (blank group, low-dose group and high-dose group). The PBMCs were incubated in vitro for 48 hours in the absence (blank group) or presence (low-dose group and high-dose group) of Cordyceps extract at different concentrations (10, 20 microg/mL). The expressions of interferon-gamma (IFN-gamma), interleukin-4 (IL-4), IL-10, T-box expressed in T cells (T-bet), GATA-binding protein-3 (GATA-3) and forkhead/winged-helix transcription factor-3 (Foxp3) mRNAs in PBMCs were measured by real-time fluorescent quantitative polymerase chain reaction, and the contents of IFN-gamma, IL-4 and IL-10 in supernatants were measured by enzyme-linked immunosorbent assay., Results: The expressions of IFN-gamma mRNA showed no significant differences among the three groups. The expressions of the IL-4 mRNA in the high-dose group and the low-dose group were lower than that in the blank group (P=0.014, P=0.011). The expression of IL-10 mRNA in the high-dose group was higher than that in the blank group (P=0.034). And the differences of the IFN-gamma mRNA/IL-4 mRNA ratio presented no statistic significance among the three groups. The level of IL-4 content in the high-dose group was lower than that in the blank group (P=0.018), but the level of IL-10 content, and ratio of IFN-gamma/IL-4 in the high-dose group were higher than those in the blank group (P=0.011, P=0.045). The differences of the IFN-gamma presented no statistic significance among the three groups. The T-bet/GATA-3 ratio and Foxp3 mRNA expression in the high-dose group were higher than those in the blank group (P=0.001, P=0.015). There was significant difference in expression of GATA-3 mRNA between the high-dose group and the blank group (P=0.028), and between the low-dose group and the blank group (P=0.019). The expression differences of T-bet mRNA were insignificant between any two groups., Conclusion: Cordyceps extract can inhibit the proliferation and differentiation of Th2 cells and reduce the expression of related cytokines by down-regulating the expression of GATA-3 mRNA and up-regulating the expression of Foxp3 mRNA in PBMCs. Meanwhile, it can alleviate the chronic allergic inflammation by increasing the content of IL-10.

Objective: To know the characteristic of AEFI clusters, and provide reference for preventing and handling of AEFI clusters., Methods: The literature were collected from CNKD and WAN FANG datebases 1999-2009. 4. The related information were analysed from the qualified literatures., Results: 75 clusters from 83 literatures of 41 kinds of journal were identified. Among them, 64% occurred in rural area, 62.67% occurred at school. 10 kinds of vaccines were identified. 13 clusters of Japanese encephalitis vaccine (JEV), 12 of Bacillus Calmette-Guerin (BCG), 10 of meningococcal polysaccharide vaccine (MPV), and 10 of live attenuated measles vaccine (MV), totally accounted for 60%. Common, minor reactions accounted for 5.33%, Rare, serious reactions was 17.33%, Coincidental illnesses was 12%, Psychogenic reactions was 46.67%, practice error events was 46.67%. 4 465 cases were reported, and 95.52% of them were school students. 82.22% were psychogenic reactions and 9.14% were coincidental illnesses. Common, minor vaccine reactions were fever; rare, serious vaccine reactions mainly were anaphylaxis, which accounted for 57.14%; psychogenic reactions mainly were hysteria, which accounted for 97.14%, 57.58% of them occurred after Vaccination of JEV, MPV and MV. Practice error events mainly were local redness and swelling, induration abscess and diabrosis. Clusters occurred after BCG vaccination accounted for 71.43%., Conclusion: AEFI clusters occurred mainly in rural areas and after immunization campaigns at school. The incidence rate of common, minor vaccine reactions and rare, serious vaccine reactions were low. Psychogenic reactions and practice error events, especially after BCG vaccination, had a higher incident rate. The total occurrence of clusters and the average cases of each events were larger. It is important to improve the quality of vaccination service for preventing and handling AEFI clusters.

To evaluate the risk of sequela after serious vaccine reaction. The rare serious vaccine reactions might result in sequela, the incidence of rare serious vaccine reactions, the estimated incidence of sequela and cases reported were reviewed. Further research should be developed to provide the scientific basis for sequela control and prevention.

Objective: The study is to analyze the occurrence features of Adverse Event Following Immunization (AEFI) in China, and to evaluate the implementation of AEFI surveillance system, the safety of National Immunization Program (NIP) Vaccines and the quality of the immunization services., Methods: The AEFI data of 2007-2008 were collected through the China Information System and the Children Immunization Information System, which reported before march 25, 2009. The descriptive methodology was used in the study., Results: 32120 AEFI cases of 2007-2008 were reported, 95.02% were reported from 10 pilot provinces. The ratio of male and female was 1.41:1.77. 53% cases were < OR =1 years old and the cases were occurred more often between april to october. The first three vaccines are DPT, MPV and JREV. 65.85% happened after the 1st and 2nd dose and 75.05% within 1 d after vaccination. The estimat reported incidence of NIP vaccines were 7.99-322.77 per million doses. In the classification of AEFI cases, 79.93% cases were common, minor reactions, 14.65% were rare, serious reactions and others were <5%. Anaphylaxis, sterile abscess and vaccine reaction of BCG were the most common clinical diagnosis. 91.2% AEFI cases were cured or improved. Only 1.93% were hospitalized, among them 2/3 stayed in hospital < or = 10 d., Conclusions: Comparing with 2005-2006, the completeness and sensitivity of national AEFI surveillance system were improved greatly in 2007-2008, but it still needs to be strengthened. The NIP vaccines were significantly related to the introduced vaccines in Expanded Immunization Program. AEFIs occurrence mostly in young children, first-dose and the early period of vaccination. The pilot study showed that safety of NIP vaccines and the quality of immunization services are satisfied.

We evaluate the security performance of the recently proposed "stealth" approach to covert communications over a public fiber-optical network. We present quantitative security analysis to assess the vulnerability of such systems against different attacks executed by an eavesdropper. We demonstrate the security advantage of the system by examining the BER/SNR performance as a function of the fidelity of the decoder used by an eavesdropper. Effective key length is constructed as a security metric to gauge the level of confidentiality implicit in the secure transmission.

Transcriptional programs that initiate and sustain the proliferation, differentiation and survival of CD8(+) T cells during immune responses are not completely understood. Here we show that inhibitor of DNA binding 2 (Id2), an antagonist of E protein transcription factors, was upregulated in CD8(+) T cells during infection and that expression of Id2 was maintained in memory CD8(+) T cells. Although Id2-deficient naive CD8(+) T cells recognized antigen and proliferated normally early after infection, effector CD8(+) T cells did not accumulate because the cells were highly susceptible to apoptosis. Id2-deficient CD8(+) T cells responding to infection had changes in the expression of genes that influence survival and had altered memory formation. Our data emphasize the importance of Id2 in regulating gene expression by CD8(+) T cells and the magnitude of effector responses, suggesting a mechanism involving Id protein- and E protein-mediated survival and differentiation of mature T cells.

Macrolides have been reported to modify the host immune and inflammatory responses both in vivo and in vitro. We examined the in vitro effect of the macrolides tilmicosin and tylosin, which are only used in the veterinary clinic, on the production of nitric oxide (NO), prostaglandin E(2) (PGE(2)) and cytokines by lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages and mouse peripheral blood mononuclear cells (PBMCs). Compared with 5 microg/mL, tilmicosin and tylosin concentrations of 10 microg/mL and 20 microg/mL significantly decreased the production of 6-keto-prostaglandin F(1alpha) (6-keto-PGF(1alpha)), PGE(2), NO, tumour necrosis factor-alpha (TNF-alpha), interleukin (IL)-1beta and IL-6, and increased IL-10 production. Cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) gene expression were also significantly reduced. These results support the opinion that macrolides may exert an anti-inflammatory effect through modulating the synthesis of several mediators and cytokines involved in the inflammatory process.

We develop a spread-spectrum based approach to secure communications over existing fiber-optical networks. Secure transmission for a dedicated user is achieved by overlaying a covert channel onto a host channel in the existing active fiber link. The covert channel is optically encoded and temporally spread, and has average power below the noise floor in the fiber, making it hidden for a direct detection thus allowing for cryptographic and steganographic security capabilities. The presence for the host channel in the network provides an ad hoc security expansion and increases the difficulty for an eavesdropper to intercept and decode the secure signal.
