Abstract

BACKGROUND AND PURPOSE: Regulating P2X7 receptor‐mediated activation of NLRP3 inflammasomes could be a therapeutic strategy to treat alcoholic hepatosteatosis. We investigated whether this process was modulated by gentiopicroside, the main active secoiridoid glycoside from Gentiana manshurica Kitagawa. EXPERIMENTAL APPROACH: In vivo models of acute and chronic alcoholic hepatosteatosis were established by intragastrically administered ethanol or using chronic plus binge ethanol feeding of Lieber‐DeCarli liquid diet to male C57BL/6 mice. In vitro, HepG2 cells were treated with ethanol. RAW 264.7 macrophages and murine bone marrow‐derived macrophages (BMDMs) were stimulated with LPS and ATP. KEY RESULTS: In both the acute and chronic alcohol‐induced mouse hepatosteatosis models, gentiopicroside decreased serum aminotransferases and triglyceride accumulation. Up‐regulated SREBP1, down‐regulated PPARα and phosphorylated acetyl‐CoA carboxylase caused by acute and chronic alcohol feeding were modulated by gentiopicroside, through the elevation of LKB1 and AMPK. Suppression of P2X7 receptor‐NLRP3 activation by gentiopicroside inhibited IL‐1β production. In ethanol‐exposed HepG2 cells, gentiopicroside reduced lipogenesis and promoted lipid oxidation via activation of P2X7 receptor‐NLRP3 inflammasomes. Genetic or pharmacological blockade of P2X7 receptors enhanced AMPK activity and reduced SREBP1 expression in ethanol‐treated HepG2 cells. Gentiopicroside down‐regulated P2X7 receptor‐mediated inflammatory responses in LPS/ATP‐stimulated RAW 264.7 macrophages and BMDMs. IL‐1β from macrophages accelerated lipid accumulation in hepatocytes. Depleting macrophages by clodronate liposomes ameliorated alcoholic hepatosteatosis, and it was further alleviated by gentiopicroside. CONCLUSIONS AND IMPLICATIONS: Activation of LKB1/AMPK signalling by gentiopicroside was mediated by the P2X7 receptor‐NLRP3 inflammasome, suggesting the therapeutic value of blocking P2X7 receptors in the treatment of alcoholic hepatosteatosis.

Abstract

Background: Increased use of thin-section computed tomography (CT) scans has revealed that small lung nodules, termed ground-glass nodules, are frequent in primary breast cancer patients and are associated with pre-invasive or invasive pulmonary adenocarcinomas. However, little is known of the incidence and fate of ground-glass nodules. Purpose: To elucidate the incidence and natural course of CT-detected pulmonary ground-glass nodules in Chinese women with breast cancer. Material and Methods: We retrospectively reviewed data from female breast cancer patients who underwent lung CT scans and who were followed for ≥3 months after the initial scan to identify the incidence of ground-glass nodules and any changes in them during the follow-up period. Results: Between January 2008 and April 2018, 693 out of 4682 breast cancer patients (14.8%) had persistent lung ground-glass nodules as detected by CT scan. The median age was 52 years (interquartile range [IQR] = 45–62 years). Median nodule size was 4.9 mm in diameter on initial CT scan. Frequency of growth was 7.5% (52/693 patients). Median volume doubling time was 1092 days (IQR = 719–1808 days) for 39 growing in size nodules. Initial nodule size, nodule type, and follow-up period were independent predictors of nodule growth. Conclusion: Most pulmonary ground-glass nodules in breast cancer patients were stable during long-term follow-up; most growing nodules had an indolent clinical course, suggesting that nodules should be monitored until growth is detected. Abstract

Using rainfall data from the Global Precipita- tion Climatology Project (GPCP), NOAA extended re- construction sea surface temperature (ERSST), and NCEP/NCAR reanalysis, this study investigates the in- terannual variation of summer rainfall southwest of the Indian Peninsula and the northeastern Bay of Bengal as- sociated with ENSO. The composite study indicates a decreased summer rainfall southwest of the Indian Penin- sula and an increase in the northeastern Bay of Bengal during the developing phase, but vice versa during the decay phase of El Nino. Further regression analysis dem- onstrates that abnormal rainfall in the above two regions is controlled by different mechanisms. Southwest of the Indian Peninsula, the precipitation anomaly is related to local convection and water vapor flux in the decay phase of El Nino. The anomalous cyclone circulation at the lower troposphere helps strengthen rainfall. In the north- eastern Bay of Bengal, the anomalous rainfall depends on the strength of the Indian southwest summer monsoon (ISSM). A strong/weak ISSM in the developing/decay phase of El Nino can bring more/less water vapor to strengthen/weaken the local summer precipitation.

Abstract

The Northern Indian Ocean (NIO) sea surface temperature (SST) warming, associated with the El Nino/Southern Oscillations (ENSO) and the Indian Ocean Dipole (IOD) mode, is investigated using the International Comprehensive Ocean-Atmosphere Data Set (ICOADS) monthly data for the period 1979-2010. Statistical analyses are used to identify respective contribution from ENSO and IOD. The results indicate that the first NIO SST warming in September-November is associated with an IOD event, while the second NIO SST warming in spring-summer following the mature phase of ENSO is associated with an ENSO event. In the year that IOD co-occurred with ENSO, NIO SST warms twice, rising in the ENSO developing year and decay year. Both short-wave radiation and latent heat flux contribute to the NIO SST variation. The change in shortwave radiation is due to the change in cloudiness. A cloud-SST feedback plays an important role in NIO SST warming. The latent heat flux is related to the change in monsoonal wind. 135. Application of Multiple Genetic Markers in Determination of Full and Half Sibling Relationship: A Case Report.

Abstract

Objective To investigate the application of the comprehensive use of multiple genetic markers in full and half sibling relationship testing through the identification of a case of suspected sibling relationship. Methods Genomic DNA were extracted from bloodstain samples from 4 subjects (ZHANG-1, ZHANG-2, male; ZHANG- 3, ZHANG-4, female). Autosomal STR loci, X- STR, Y- STR loci and polymorphisms of mtDNA HV- I and H were genotyped by EX20 STR kit, X19 kit, Data Y24 STR kit, and Sanger sequencing, respectively. Results According to autosomal STR based IBS scoring results, full sibling relationships were indicated among ZHANG-2, ZHANG-3 and ZHANG-4, but those were not indicated between ZHANG-1 and ZHANG-2 or ZHANG-3 or ZHANG-4. According to autosomal STR based FSI and HSI, with ITO method and discriminant function method, full sibling relationships among ZHANG- 2, ZHANG- 3 and ZHANG- 4 were indicated, and half sibling relationships between ZHANG-1 and ZHANG-2 or ZHANG-3 or ZHANG-4 were also indicated. X-STR and mtDNA sequencing results showed that all the 4 samples came from a same maternal line, and Y-STR results showed that ZHANG-1 and ZHANG- 2 did not come from a same paternal line, which supported the half sibling relationship between ZHANG-1 and ZHANG-2 or ZHANG-3 or ZHANG-4, verified by parental genotype reconstruction based on autosomal STR genotyping. Conclusion For the identification of sibling relationships, it is effective to have reliable results with the mutual verification and support of multiple genetic markers (autosomal STR, sex chromosomal STR and mtDNA sequence) and calculations ( IBS, ITO, discriminant function method and family reconstruction). Recently, the authors proposed the hybrid Local Domain-Free Discretization and Immersed Boundary Method (LDFD-IBM) (Wu et al. 2012), which can be used to solve the flow problem within complex domain with truly 2nd order accuracy (Wu 2017). As a non-conforming mesh method, LDFD-IBM can be extended to handle the moving boundary problems. In this paper, the strategy of LDFD-IBM for incompressible flows problems with moving boundary is presented. Abstract

One novel organic-inorganic hybrid compound based on α-H4(SiW12O40) using 1,10-phenanthroline-5,6-dione (Dione) as neutral organic ligand, namely, [Cu(Dione)2(H2O)]2(SiW12O40), has been synthesized under hydrothermal condition. The organic-inorganic hybrid material, [Cu(Dione)2(H2O)]2(SiW12O40) crystallizes in the orthorhombic space group Pbca with the unit-cell parameters a = 15.5453(10) Å, b = 21.3948(15)Å, c = 22.0731(15)Å, α = 90.00°, β = 90.00° and γ = 90.00°. Single-crystal X-ray diffraction analyses reveal that this compound is zero-dimensional structure. In this compound, intermolecular O-H***O hydrogen bonds play important roles which link the architecture to form 2D supramolecular structure. Abstract

Chinese New Rural Cooperative Medical Insurance System has been implemented for six years, but for various reasons, there is not too much empirical research on the efficiency of the NRCMS. This paper adopts DEA method and makes a comprehensive analysis on static efficiency and dynamic efficiency of the NRCMS, combining with panel data. The result shows that there exists large difference on efficiency of the NRCMS among provinces and efficiency in the main food producing areas is relatively low. At the same time, although all rural population have been involved in the NRCMS basically, benefit that farmers get from the NRCMS is disproportionate with government support and it is urgent to improve farmers' utilization of the NRCMS. At this time, it may be a good starting point to improve technologically or optimize the system itself.
