Dumpsites are hard to locate globally. Here the authors apply deep networks to satellite images to provide an effective and low-cost way to detect dumpsites with the new method saving more than 96.8% of the manual time with a strong sensitivity to dumpsites.

Abstract Background Fundamental motor skills (FMSs) are crucial for children’s health and comprehensive development. Obese children often encounter a considerable challenge in the development of FMSs. School-family blended PA programs are considered a potentially effective approach to improve FMSs and health-related outcomes among obese children, however, empirical evidence is still limited. Therefore, this paper aims to describe the development, implementation, and evaluation of a 24-week school-family blended multi-component PA intervention program for promoting FMSs and health among Chinese obese children, namely the Fundamental Motor Skills Promotion Program for Obese Children (FMSPPOC) employing behavioral change techniques (BCTs) and building on the Multi-Process Action Control (M-PAC) framework as well as using the Reach, Effectiveness, Adoption, Implementation, and Maintenance (RE-AIM) framework for improving and evaluating the program. Methods Using a cluster randomized controlled trial (CRCT), 168 Chinese obese children (8–12 years) from 24 classes of six primary schools will be recruited and randomly assigned to one of two groups by a cluster randomization, including a 24-week FMSPPOC intervention group and a non-treatment waiting-list control group. The FMSPPOC program includes a 12-week initiation phase and a 12-week maintenance phase. School-based PA training sessions (2 sessions/week, 90 min each session) and family-based PA assignments (at least three times per week, 30 min each time) will be implemented in the initiation phase (semester time), while three 60-min offline workshops and three 60-min online webinars will be conducted in the maintenance phase (summer holiday). The implementation evaluation will be undertaken according to the RE-AIM framework. For intervention effectiveness evaluation, primary outcome (FMSs: gross motor skills, manual dexterity and balance) and secondary outcomes (health behaviors, physical fitness, perceived motor competence, perceived well-being, M-PAC components, anthropometric and body composition measures) will be collected at four time-points: at baseline, 12-week mid-intervention, 24-week post-intervention, and 6-month follow-up occasions. Discussion The FMSPPOC program will provide new insights into the design, implementation, and evaluation of FMSs promotion among obese children. The research findings will also supplement empirical evidence, understanding of potential mechanisms, and practical experience for future research, health services, and policymaking. The effect of hydrogen content on the deformation and fracture behavior of 27Cr−4Mo−2Ni super ferritic stainless steel (SFSS) was investigated in this study. It was shown that the plasticity and yield strength of SFSS were very susceptible to hydrogen content. The introduction of hydrogen led to a significant decrease in elongation and a concurrent increase in yield strength. Nevertheless, a critical threshold was identified in the elongation reduction, after which the elongation remained approximately constant even with more hydrogen introduced, while the yield strength exhibited a monotonic increase with increasing hydrogen content within the experimental range, attributed to the pinning effect of the hydrogen Cottrell atmosphere on dislocations. Furthermore, the hydrogen-charged SFSS shows an apparent drop in flow stress after upper yielding and a reduced work hardening rate during the subsequent plastic deformation. The more hydrogen is charged, the more the flow stress drops, and the lower the work hardening rate becomes.

Cisplatin nephrotoxicity is a critical limitation of solid cancer treatment. Until now, the complex interplay of various pathophysiological mechanisms leading to proximal tubular cell apoptosis after cisplatin exposure has not been fully understood. In our study, we assessed the role of the autophagy-related protein BECLIN1 (ATG6) in cisplatin-induced acute renal injury (AKI)—a candidate protein involved in autophagy and with putative impact on apoptosis by harboring a B-cell lymphoma 2 (BCL2) interaction site of unknown significance. By using mice with heterozygous deletion of Becn1, we demonstrate that reduced intracellular content of BECLIN1 does not impact renal function or autophagy within 12 months. However, these mice were significantly sensitized towards cisplatin-induced AKI, and by using Becn1+/−;Sglt2-Cre;Tomato/EGFP mice with subsequent primary cell analysis, we confirmed that nephrotoxicity depends on proximal tubular BECLIN1 content. Mechanistically, BECLIN1 did not impact autophagy or primarily the apoptotic pathway. In fact, a lack of BECLIN1 sensitized mice towards cisplatin-induced ER stress. Accordingly, the ER stress inhibitor tauroursodeoxycholic acid (TUDCA) blunted cisplatin-induced cell death in Becn1 heterozygosity. In conclusion, our data first highlight a novel role of BECLIN1 in protecting against cellular ER stress independent from autophagy. These novel findings open new therapeutic avenues to intervene in this important intracellular stress response pathway with a promising impact on future AKI management.

The mechanical properties of various Ti-6Al-4V alloys are influenced by their respective microstructures. This study generated an ultrafine-grain (UFG) Ti-6Al-4V alloy featuring bimodal grain distribution characteristics achieved through initial heat treatment, multi-axial forging (MF), and annealing. The study also extensively examined the evolution process of the alloy’s microstructure. By subjecting the materials to heat treatments at 900 °C with air cooling and 950 °C with air cooling, both materials were found to be consisted of primary α (αp) and transformed β (αs+β) regions with different proportions. Following MF, the sample treated at 900 °C displays a microstructure featuring UFGs of α+β surrounding larger micron-sized αp grains. On the other hand, the sample treated at 950 °C displays a microstructure distinguished by twisted αs lamellar and fragmented β grains surrounding larger micron-sized αp grains. Following annealing, no significant grain growth was observed in the sample. The geometrically necessary dislocations (GNDs) within the UFGs were eliminated, though some GNDs persisted within the αp grains. The samples undergoing the 900 °C heat treatment, MF, and subsequent annealing exhibited elevated strength (1280 MPa) and total elongation (10.7%). This investigation introduces a novel method for designing the microstructure of the Ti-6Al-4V alloy to achieve superior performance.

BackgroundColon cancer (CC) ranks as one of the leading causes of cancer-related mortality globally. Single-cell transcriptome sequencing (scRNA-seq) offers precise gene expression data for distinct cell types. This study aimed to utilize scRNA-seq and bulk transcriptome sequencing (bulk RNA-seq) data from CC samples to develop a novel prognostic model.MethodsscRNA-seq data was downloaded from the GSE161277 database. R packages including “Seurat”, “Harmony”, and “singleR” were employed to categorize eight major cell types within normal and tumor tissues. By comparing tumor and normal samples, differentially expressed genes (DEGs) across these major cell types were identified. Gene Ontology (GO) enrichment analyses of DEGs for each cell type were conducted using “Metascape”. DEGs-based signature construction involved Cox regression and least absolute shrinkage operator (LASSO) analyses, performed on The Cancer Genome Atlas (TCGA) training cohort. Validation occurred in the GSE39582 and GSE33382 datasets. The expression pattern of prognostic genes was verified using spatial transcriptome sequencing (ST-seq) data. Ultimately, an established prognostic nomogram based on the gene signature and age was established and calibrated. Sensitivity to chemotherapeutic drugs was predicted with the “oncoPredict” R package.ResultsUsing scRNA-Seq data, we examined 33,213 cells, categorizing them into eight cell types within normal and tumor samples. GO enrichment analysis revealed various cancer-related pathways across DEGs in these cell types. Among the 55 DEGs identified via univariate Cox regression, four independent prognostic genes emerged: PTPN6, CXCL13, SPINK4, and NPDC1. Expression validation through ST-seq confirmed PTPN6 and CXCL13 predominance in immune cells, while SPINK4 and NPDC1 were relatively epithelial cell-specific. Creating a four-gene prognostic signature, Kaplan-Meier survival analyses emphasized higher risk scores correlating with unfavorable prognoses, confirmed across training and validation cohorts. The risk score emerged as an independent prognostic factor, supported by a reliable nomogram. Intriguingly, drug sensitivity analysis unveiled contrasting anti-cancer drug responses in the two risk groups, suggesting significant clinical implications.ConclusionWe developed a novel prognostic four-gene risk model, and these genes may act as potential therapeutic targets for CC.

IntroductionAs relatively little is known about self-efficacy and social support in individuals aged 65 years and older and whether they are facing a decline in life due to multimorbidity and previous COVID-19 infection, this study investigated hypotheses based on Social Cognitive Theory.MethodsIt was tested whether depressive symptoms in multimorbid patients who were hospitalized for COVID-19 infection, and recover post infection during their hospital stay, do not differ from those of multimorbid patients hospitalized for other conditions. Furthermore, we tested whether depressive symptoms are associated with increased loneliness scores, low self-efficacy beliefs, and poorly perceived social support. Additionally, it was investigated whether self-efficacy is a mediator variable, and social support is a moderator variable between loneliness and depression. N = 135 patients with or without previous COVID-19 infection (mean age 64.76) were recruited. Paper questionnaires were collected at the time of inpatient hospital admission in the year 2021 and in a cross-sectional study design. The study compared n = 45 multimorbid patients who survived COVID-19 infection with those n = 90 who were not infected before.ResultsNo significant difference in depressive symptomology between these two groups revealed [t(133) = 130, p = 0.90, d = 0.024); F(3, 122) = 0.255, p = 0.86]. The study found a positive correlation between loneliness and anxiety and depression in both groups (rdepression = 0.419 and ranxiety = 0.496). Self-efficacy mediated the relation between loneliness and depression. The completely standardized indirect effect was β = 0.111, percentile Bootstrap 95% CI 0.027–0.201.DiscussionThe research findings suggest the importance of self-efficacy, and loneliness in the development of depressive symptoms, and have several practical implications for improving the mental health of multimorbid patients: Prospectively, treatment should not only focus on physical and cognitive health, but also on promoting self-efficacy and perceived social support, as well as address loneliness with psychoeducational interventions. Replication of the findings and conducting interventional research also employing lifestyle components should follow up, as this study tested associations but no causal relationships.

Abstract Comparative studies of egg recognition and rejection between various sympatric hosts provide insight into the coevolutionary history of the hosts and parasites, as well as the degree of antagonism between the species. Although buntings are widely considered to be a suitable host taxon for cuckoos, there has been relatively little research on this example of parasitism and host antiparasitic behaviour. Here we provided the first report on brood parasitism and egg recognition in three sympatric ground‐nesting bunting hosts of the common cuckoo (Cuculus canorus), namely the yellow‐throated bunting (Emberiza elegans), south rock bunting (E. yunnanensis), and crested bunting (E. lathami). The results show that for the five breeding seasons during 2018–2022, the parasitism rate by common cuckoos was 0.87% and 0.45% in yellow‐throated buntings and south rock buntings, respectively, whereas the parasitism rate by an unidentified parasite was 4% during 2018–2023 in the crested bunting. The rejection rates of the three bunting hosts for blue non‐mimetic eggs were 89.3%, 88.9%, and 100% for yellow‐throated buntings, south rock buntings, and crested buntings, respectively. The rejection rates for red non‐mimetic eggs by yellow‐throated buntings and south rock buntings were lower at 76.9% and 82.4%, respectively. All three sympatric bunting hosts examined had high levels of egg recognition and egg rejection, suggesting that it may have been subjected to high parasitic history and that egg recognition ability was retained after the loss of parasitism, which needs to be further verified by future experiments.

To grasp the phase change characteristics of the filling slurry from fluid to solid state in time, conducted a study on the setting characteristics of the filling slurry with different aggregate combinations, and analyzed the law of the initial and final setting time with the change of the amount of cementation powder, filling plant tailings, tailing pond tailings, and water. The results show that the initial and final setting time of the filling slurry prepared from the tailings of the filling plant is proportional to the concentration, and is positively correlated with the slope decline of the sand-cement ratio. The filling slurry prepared from the tailings of the tailing pond shows roughly the same change trend as the above slurry, but the change law is not obvious, which cannot be expressed by one kind of functional model, and the initial setting time and final setting time are less than the above slurry, which is because the tailings of the tailing pond contain more fine particles, which play a role in delaying setting in the slurry. For example, for five kinds of filling materials with different aggregates of 4:1, 6:1, 8:1, 12:1, and 20:1 sand-to-ash ratios and concentrations of 66%–72%, the initial setting times of filling slurry prepared from tailings of filling plant and tailings of tailing pond were 75.3 h–112.3 h, 77.6 h–122.2 h, and the final setting times were 82.8 h–123.4 h, 85.5 h–133.9 h. Therefore, it shows that the use of filling plant tailings containing fewer fine particles to prepare filling slurry can obtain a better treatment effect of the mining area, and the shortening of the solidification and hardening process of filling slurry can accelerate the mining cycle operation, and increase the amount of solid waste disposal per unit time, thus creating the necessary technical conditions for the enterprise to improve production efficiency and sustainable development.

Underground mining in Karst Plateau landform area may cause the loss of support for the upper rock stratum, resulting in rock collapse and large-scale subsidence of the ground surface. Also, the formation of a large-scale goafs may further lead to geo-hazards such as collapse, water gushing, slope instability and so on in the area. To reduce the impact of goaf settlement on local strata stability, this paper established a standardized safe mining detection model for goafs based on the geological safety characteristics of mining goafs. With reference to the statistical analysis of the geological conditions in the mining area, a numerical model with 358 goafs and the proposed mining area was established using FLAC3D numerical software. The surface subsidence and variations of plastic zone in the mining area were comprehensively analyzed. The results indicated that there was a correlation between the stability of the mining area and the geological occurrence conditions of the goafs. By quantitatively taking the values from standardized safety mining detection models, the standardized safety mining detection and warning technique was finally established. The findings can provide technical guidance for safety detection and early warning in the whole process of underground goaf mining in Karst Plateau karst development zone.

The simulation of terrestrial stellar radiation information suffers from distortions due to limited precision of stellar spectrum and magnitude simulations. Hence, the effective verification and evaluation of experimental conditions for new techniques such as astronomical spectral velocity measurement and navigation are hindered. This study proposes a multi-color temperature and multi-magnitude simulation approach for astronomical spectral velocity measurement and navigation. First, we analyzed the composition and operational principles of the multi-color temperature and multi-magnitude simulation system. Furthermore, we established a mathematical modulation model for multi-color temperature and multi-magnitude simulation. Second, we designed a fuzzy proportional–integral–derivative (PID) controller with dual input and three outputs, as well as a multi-magnitude feedback control simulation algorithm based on the principles of spectral modulation and illuminance conversion. Finally, we constructed an experimental platform to validate the accuracy of spectral simulation and proposed a method to extend the simulation range for multi-magnitude simulation measurements based on existing experimental conditions. Moreover, we simulated 0–8 stars in the color temperature spectrum ranging from 3000 K to 11000 K. Experimental results demonstrated that the maximum spectral simulation error for color temperature between 3000 K and 11000 K color temperature was within ±4.996%, while the maximum simulation error for magnitude 0–8 stars was within ±0.1 Mv. The proposed method enables simultaneous simulation of multi-color temperature and multi-magnitude, enhancing the accuracy of spectral and magnitude simulations. Additionally, it resolved the issue of a limited magnitude simulation range caused by detection limits, providing effective verification and evaluation of experimental conditions for the functionality and performance of the new autonomous navigation method for astronomical spectral velocity measurement.

Mountain bike (MTB) racing is a highly intensive physical activity and requires a high degree of technical ability to perform at the elite athlete level, which might compromise mental well-being, increasing symptoms of anxiety and depression through overtraining, injury, and burnout. Yoga Pranayama is the key to bringing about psychosomatic integration and harmony. This study aimed to explore the effects of yoga pranayama practices (YPP) on elite mountain bikers' burnout. This is a single-arm pilot study. Twenty-seven subjects practiced 30 sessions of YPP seven times a week for 1 month. The outcomes measured were blood biochemical parameters accompanied by complete blood count and athlete burnout score. Cubital vein blood test and burnout questionnaire were conducted at baseline and after 1 month. Test results showed a significant decrease in cortisol (CO) (P = 0.001) and urea nitrogen (P < 0.001) and an increase in testosterone: CO ratio (P = 0.001). This study indicates that YPP might improve burnout in elite mountain bikers.

Understanding the distinct functions of human muscles could not only help professionals obtain insights into the underlying mechanisms that we accommodate compromised neuromuscular system, but also assist engineers in developing rehabilitation devices. This study aims to determine the contribution of major muscle and the energy flow in the human musculoskeletal system at four sub-phases (collision, rebound, preload, push-off) during the stance of walking at different speeds. Gait experiments were performed with three self-selected speeds: slow, normal, and fast. Muscle forces and mechanical work were calculated by using a subject-specified musculoskeletal model. The functions of individual muscles were characterized as four functional behaviors (strut, spring, motor, damper), which were determined based on the mechanical energy. The results showed that during collision, hip flexors (iliacus and psoas major) and ankle dorsiflexors (anterior tibialis) were the most dominant muscles in buffering the stride with energy absorption; during rebound, the posterior muscles (gluteus maximus, gastrocnemius, posterior tibialis, soleus) contributed the most to energy generation; during preload, energy for preparing push-off was mainly absorbed by the muscles surrounding knee (vastus, semimembranosus, semitendinosus); during push-off, ankle plantar flexors (gastrocnemius, soleus, posterior tibialis, peroneus muscles, flexor digitorum, flexor hallucis) mainly behaved to generate energy for forward propulsion. With increased walking speed, additional energy (almost 400%) from harder stride was mainly absorbed by the flexor muscles. Hip extensors and adductors transferred more energy (around 150%) to the distal segments during rebound. Soleus and gastrocnemius muscles generated more energy (about 75%) to the proximal segments for propulsion. Along with our previous study of joint-level energy analysis, these findings could assist better understanding of human musculoskeletal behaviors during locomotion and provide principles for the bio-design of related assistive devices from motors performance enhancement to rehabilitation such as exoskeleton and prosthesis.

Efficient intersection planning is one of the most challenging tasks for an autonomous vehicle at present. The degree of response to other traffic participants and whether the path tracking effect is good are the key aspects that determine the performance of planning algorithm for an autonomous vehicle. In order to deal with these problems, in this paper, the frame structure of POMDP (Partially Observable Markov Decision Processes) and the time distance coordinate system are combined to carry out the speed planning for the self-propelled vehicle. The speed planning designs a novel POMDP model to ensure effective prediction of behavioral intentions and decision-making at intersections with no traffic signs. Specifically, the method is considered both the driving position and the time range of arrival of the vehicle at the intersection in real-world situations. At the same time, the algorithm is projected the state information into the time distance coordinate system in the form of probability, and used the value iteration method to solve the optimal solution, so as to improve the adaptability of the algorithm to real-world scenarios. Furthermore, MPC control is added on this basis. Rational use of dynamic MPC and kinematic MPC can effectively reduce the tracking error, improve the stability of tracking, and carry out finer control when the speed frequently fluctuates.

Abstract Objectives Pulmonary lesion is frequently seen in ANCA-associated vasculitis (AAV) patients primarily due to AAV lung involvement or infection, which are hard to differentiate due to their high similarity in clinical manifestations. We aimed to analyze the clinical features of pulmonary lesions consequent to AAV involvement or infection in AAV patients and further identify the markers for differential diagnosis. Methods 140 AAV patients who admitted to the Renmin Hospital of Wuhan University from January 2016 to July 2021 were included in this study. According to the nature of lung conditions, these patients were divided into the non-pulmonary lesion group, the lung infection group and the non-pulmonary infection group, and their demographics, clinical symptoms, imaging features, as well as laboratory findings were compared. A receiver operating characteristic (ROC) curve was drawn, and the diagnostic efficacy of single biomarker and composite biomarkers on pulmonary infection was then evaluated. Results The patients in the lung infection group were significantly older than those in the no lesion group (63.19 ± 14.55 vs 54.82 ± 15.08, p = 0.022). Patients in the lung infection group presented more frequent symptoms and more obvious pulmonary image findings. Compared with patients in the non-pulmonary infection group, patients in the lung infection group showed a higher symptom incidence of fever, chest tightness, cough and expectoration, and hemoptysis (52.94% vs 16.00%, 61.76% vs 40.00%, 72.06% vs 46.00%, 27.94% vs 8.00%, p

To expand the practical application of rare earth magnesium alloys, rare earth magnesium alloy sheets with no cracks and good plasticity were prepared by cross rolling. The plastic deformation behavior and fracture mechanism of cross rolling and conventional rolling were investigated, and the results showed that the twins after cross rolling have a certain effect on stimulating the activation of the slip systems with lower Schmid factor. The basal slip was dominant in conventional-rolled sheets. However, the plastic deformation process of cross-rolled magnesium alloys is dominated by both basal slip and non-basal slip, especially the second-order pyramidal slip. This is because cross-rolling reduces the critical shear stress of non-basal slip. Although the slip transfer between ordinary rolling grains is good, the activated slip cannot coordinate the c-axis strain. At the same time, the slender precipitation hinders the movement of dislocations and aggravates the strain concentration, which is likely to cause the generation of ledges and further develop into cracks. In the cross-rolling process, the short and flocculent precipitation decreases the resistance of dislocation movement, and the activated non-basal slip can coordinate the strain of the c-axis, which can better coordinate the deformation and reduce the possibility of cracking.

Abstract When it comes to running and managing modern supply chains, 6G Internet of things (IoT) is of utmost importance. To provide IoT with security and automation, blockchain and machine learning are two upper-layer technology that can help. First, we propose to utilize blockchain in modern supply chains to ensure efficient collaboration between all parties. Second, we adopt multi-head attention (MHA)-based gated recurrent unit (GRU) to do inbound logistics task prediction. Finally, numerical results justify that multi-head attention-based GRU model has better fitting efficiency and prediction accuracy than its counterparts.

In order to produce natural gas hydrate safely and efficiently, it is quite crucial to research the stability of hydrate reservoir in the process of its exploitation. The coupled thermal-hydrodynamic-mechanical (THM) numerical simulation can clearly present the mechanical response laws in the process of hydrate exploitation by horizontal well, but the model is often simplified as a profile vertical to the production well, so the evolution characteristics of real formation deformation space cannot be described sufficiently. To this end, this paper develops a coupled THM simulator applicable to the large-scale hydrate exploitation by horizontal well by introducing interactive interface to the FLAC mechanics program based on the hydrate exploitation program TOUGH + HYDRATE. Then, based on the field data of second hydrate production test in South China Sea, the gas and water production and formation deformation laws under the middle- and long-term development conditions of horizontal wells are studied by taking the actual gas production of horizontal well during the production test as the constraint of coupled THM model. And the following research results are obtained. First, when the production pressure is 7 MPa and the horizontal well length is 300 m, the gas production of horizontal well can reach 1190 × 104 m3, the pressure drop has a larger influence range in the free gas layer, the produced gas mainly comes from the free gas layer and the hydrate layer and the hydrate dissociation range is not large. Second, it is predicted that the seafloor settlement is about 0.16 m after 60 days' production test and 0.52 m after osne year's production. In conclusion, hydrate exploitation by horizontal well can cause a large range of seafloor settlement. The seafloor surface settles linearly and continuously and the spatial settlement distribute is in the shape of ellipse. What's more, the greatest deformation (vertical displacement) occurs at the horizontal well and above it, where formation damage happens the easiest. It is recommended to study seafloor deformation and landslip risk under the conditions of middle and long-term production based on seafloor topography, valley distribution and nonuniform spatial thickness of mineral ores.

Abstract Solid-state cooling technology based on electrocaloric effect (ECE) has been advanced as an alternative to replace the vapour-compression approach to overcome the releasing of the global warming gases. However, the development in high ECE materials is still a challenge. In this work, polarization merging strategy was proposed to achieve a large ECE in xBa(Sn0.07Ti0.93)O3-(1−x)Ba(Hf0.1Ti0.9)O3 ferroelectric ceramics, where x = 0, 0.2, 0.4, 0.6, 0.8, and 1. Ba(Sn0.07Ti0.93)O3 with an orthorhombic phase and Ba(Hf0.1Ti0.9)O3 with a rhombohedral phase at room temperature were prepared beforehand as precursors, and phase-coexisted xBSnT-(1−x)BHfT ceramics were formed via a solid-state reaction approach. Phase coexisting structures were confirmed using the X-ray diffraction. The merged polarization was confirmed by the dielectric and ferroelectric properties. Optimal ECEs were obtained for 0.2BSnT-0.8BHfT ceramics, i.e., adiabatic temperature change ΔT = 2.16±0.08 K at 80 °C and 5 MV/m, and ΔT = 3.35±0.09 K at 80 °C and 7 MV/m.

Abstract Activation of TNFR1 by TNFα induces the formation of a membrane-associated, intracellular complex termed complex I. Complex I orchestrates a complex pattern of modifications on key regulators of TNF signaling that collectively determines the cell fate by activating pro-survival or executing cell death programs. However, the regulatory mechanism of complex I in cell-fate decision is not fully understood. Here we identify protein phosphatase-6 (PP6) as a previously unidentified component of complex I. Loss of PP6 protects cells from TNFα-mediated cell death. The role of PP6 in regulating cell death requires its phosphatase activity and regulatory subunits. Further mechanistic studies show that PP6 modulates LUBAC-mediated M1-ubiquitination of RIPK1 and c-FLIPL to promote RIPK1 activation and c-FLIPL degradation. We also show that melanoma-associated PP6 inactivating mutants offer resistance to cell death due to the loss of sensitivity to TNFα. Thus, our study provides a potential mechanism by which melanoma-related PP6 inactivating mutations promote cancer progression.

In order to ensure the dimensional accuracy during welding assembly, external restraints are often used to control welding-induced distortion. The objective of this study is to investigate the effectiveness of external restraints on the reduction of welding distortion in low alloy high strength (LAHS) steel thin-plate panel structures with different plate thickness by means of an integrated computational approach. In the current study, the total deformation of LAHS steel thin-plate welded structure was computed by a two-step integrated computational approach which combines the thermal elastic plastic finite element method (FEM) with the elastic FEM based on inherent strain theory. In the integrated computational approach, the thermal elastic plastic FEM was employed to obtain the inherent deformations of each typical joint involved in the thin-plate welded structure at the first step, and then the elastic FEM based on the inherent strain theory was used to estimate the total deformation of the whole thin-plate welded structure. Firstly, the integrated computational approach was used to simulate the buckling distortion in a thin-plate panel structure, and the corresponding experiment was carried out to verify its computational accuracy. Then, two finite element models with plate thickness of 5 and 12 mm were established to clarify the effectiveness of external restraint on the reduction of welding distortion of the thin-plate welded structures. The simulated results suggest that external restraint could not effectively prevent the occurrence of buckling distortion for the thin-plate structure with 5 mm thickness, while it can reduce the out-of-plane deformation to a large extent for the thin-plate welded structure with 12 mm thickness.

To suppress the edge crack of the magnesium alloy sheet during the ordinary rolling process, a new rolling process named width-limited rolling was proposed in this paper. Width-limited rolling is a rolling method in which the width of the alloy sheet is limited by modifying the shape of the rollers, allowing a compressive stress field to form at the edge portion of the alloy sheet during rolling, resulting in the reduction of edge cracks. At present work, magnesium alloy sheets were separately subjected to ordinary rolling and width-limited rolling. The microstructure evolution and mechanical properties of the rolled sheets were investigated by EBSD, TEM, and XRD. The results exhibited that under the same rolling conditions, the sheet after ordinary rolling exhibited obvious edge cracks while no crack was found at the edge of the sheet after width-limited rolling. The edge crack suppressing effect was attributed to the reduction of the tensile stress along rolling direction during WLR, promoting the synchronous extension of the edge and center regions to suppress edge crack tendency. Microstructure observation showed that the compressive twins formed in the sheet after ordinary rolling usually exhibited as thin plates and cannot continue to fully develop due to the premature generation of the edge cracks. However, the compressive twins developed maturely in some of which double twins formed and various slip systems with different dislocation Burgers vectors occurred in the rolled sheet after WLR. More twin intersections and shear bands, providing more potential recrystallization nucleation sites, which are beneficial to weaken basal texture. With the cooperation of twinning and dislocation slip, the texture of the sheet after the width-limited rolling is weakened and the mechanical properties are improved.

Current therapies for Fabry disease are based on reversing intracellular accumulation of globotriaosylceramide (Gb3) by enzyme replacement therapy (ERT) or chaperone-mediated stabilization of the defective enzyme, thereby alleviating lysosomal dysfunction. However, their effect in the reversal of end-organ damage, like kidney injury and chronic kidney disease, remains unclear. In this study, ultrastructural analysis of serial human kidney biopsies showed that long-term use of ERT reduced Gb3 accumulation in podocytes but did not reverse podocyte injury. Then, a CRISPR/Cas9–mediated α-galactosidase knockout podocyte cell line confirmed ERT-mediated reversal of Gb3 accumulation without resolution of lysosomal dysfunction. Transcriptome-based connectivity mapping and SILAC-based quantitative proteomics identified α-synuclein (SNCA) accumulation as a key event mediating podocyte injury. Genetic and pharmacological inhibition of SNCA improved lysosomal structure and function in Fabry podocytes, exceeding the benefits of ERT. Together, this work reconceptualizes Fabry-associated cell injury beyond Gb3 accumulation, and introduces SNCA modulation as a potential intervention, especially for patients with Fabry nephropathy.

Nasopharyngeal carcinoma (NPC) is a malignant tumor that occurs in the wall of the nasopharyngeal cavity and is prevalent in Southern China, Southeast Asia, North Africa, and the Middle East. According to studies, NPC is one of the most common malignant tumors in Hainan, China, and it has the highest incidence rate among otorhinolaryngological malignancies. We proposed a new deep learning network model to improve the segmentation accuracy of the target region of nasopharyngeal cancer. Our model is based on the U-Net-based network, to which we add Dilated Convolution Module, Transformer Module, and Residual Module. The new deep learning network model can effectively solve the problem of restricted convolutional fields of perception and achieve global and local multi-scale feature fusion. In our experiments, the proposed network was trained and validated using 10-fold cross-validation based on the records of 300 clinical patients. The results of our network were evaluated using the dice similarity coefficient (DSC) and the average symmetric surface distance (ASSD). The DSC and ASSD values are 0.852 and 0.544 mm, respectively. With the effective combination of the Dilated Convolution Module, Transformer Module, and Residual Module, we significantly improved the segmentation performance of the target region of the NPC.

Abstract Birds select suitable nest sites for breeding to ensure their own and offspring's survival; however, they inevitably suffer some potential predation risk. We studied the breeding ecology of Daurian redstarts (Phoenicurus auroreus) by providing nest boxes for their breeding from March to August of 2022. We recorded the predation of both Daurian redstarts eggs or nestlings by Oriental magpie‐robins (Copsychus saularis) and tree sparrow (Passer montanus). Oriental magpie‐robin were recorded attacking a feeding female adult and depredating nestlings. After the nestling predation event, the Daurian redstarts abandoned the nest. This video evidence provide a better understanding of the potential predators of cavity‐nesting birds.

ObjectiveThe aim of this paper is to analyze the application of machine learning in heart failure-associated diseases using bibliometric methods and to provide a dynamic and longitudinal bibliometric analysis of heart failure–related machine learning publications.Materials and methodsWeb of Science was screened to gather the articles for the study. Based on bibliometric indicators, a search strategy was developed to screen the title for eligibility. Intuitive data analysis was employed to analyze the top-100 cited articles and VOSViewer was used to analyze the relevance and impact of all articles. The two analysis methods were then compared to get conclusions.ResultsThe search identified 3,312 articles. In the end, 2,392 papers were included in the study, which were published between 1985 and 2023. All articles were analyzed using VOSViewer. Key points of the analysis included the co-authorship map of authors, countries and organizations, the citation map of journal and documents and a visualization of keyword co-occurrence analysis. Among these 100 top-cited papers, with a mean of 122.9 citations, the most-cited article had 1,189, and the least cited article had 47. Harvard University and the University of California topped the list among all institutes with 10 papers each. More than one-ninth of the authors of these 100 top-cited papers wrote three or more articles. The 100 articles came from 49 journals. The articles were divided into seven areas according to the type of machine learning approach employed: Support Vector Machines, Convolutional Neural Networks, Logistic Regression, Recurrent Neural Networks, Random Forest, Naive Bayes, and Decision Tree. Support Vector Machines were the most popular method.ConclusionsThis analysis provides a comprehensive overview of the artificial intelligence (AI)-related research conducted in the field of heart failure, which helps healthcare institutions and researchers better understand the prospects of AI in heart failure and formulate more scientific and effective research plans. In addition, our bibliometric evaluation can assist healthcare institutions and researchers in determining the advantages, sustainability, risks, and potential impacts of AI technology in heart failure.

The airborne distributed Position and Orientation System (POS) is a key piece of equipment for providing high-precision motion parameters for aerial remote sensing systems. However, wing deformation degrades the performance of distributed POS, thus, it is urgent to obtain high-precision deformation information to assist distributed POS. In this study, a modeling and calibrating method of fiber Bragg grating (FBG) sensors for wing deformation displacement measurement is proposed. First, based on the cantilever beam theory and piecewise superposition, a modeling and calibrating method for wing deformation displacement measurement is established. The wing is then placed under different deformation conditions, and the changes in the wing deformation displacement and corresponding wavelength variations of the pasted FBG sensors are obtained using theodolite coordinate measurement system and FBG demodulator, respectively. Subsequently, linear least square fitting is deployed to develop the relationship model between the wavelength variations of the FBG sensors and wing deformation displacement. Finally, the wing deformation displacement at the measuring point in the temporal and spatial dimensions is obtained by fitting and interpolation. An experiment is conducted, and the results show that the accuracy of the proposed method can reach 0.721 mm with a wing length of 3 m, which can be used in the motion compensation of an airborne distributed POS.

Fetal hyperthyroidism can occur secondary to maternal autoimmune hyperthyroidism. The thyroid-stimulating hormone receptor antibody (TRAb) transferred from the mother to the fetus stimulates the fetal thyroid and causes fetal thyrotoxicosis. Fetuses with this condition are difficult to detect, especially after maternal Graves disease therapy. Here, we present two cases of fetal hyperthyroidism with maternal hypothyroidism and review the assessment and intrauterine therapy for fetal hyperthyroidism. Both women were referred at 22+ and 23+ weeks of gestation with abnormal ultrasound findings, including fetal heart enlargement, pericardial effusion, and fetal tachycardia. Both women had a history of Graves disease while in a state of hypothyroidism with a high titer of TRAb. A sonographic examination showed a diffusely enlarged fetal thyroid with abundant blood flow. Invasive prenatal testing revealed no significant chromosomal aberration. Low fetal serum TSH and high TRAb levels were detected in the cord blood. Fetal hyperthyroidism was considered, and maternal oral methimazole (MMI) was administered as intrauterine therapy, with the slowing of fetal tachycardia, a reduction in fetal heart enlargement, and thyroid hyperemia. During therapy, maternal thyroid function was monitored, and the dosage of maternal levothyroxine was adjusted accordingly. Both women delivered spontaneously at 36+ weeks of gestation, and neonatal hyperthyroidism was confirmed in both newborns. After methimazole and propranolol drug treatment with levothyroxine for 8 and 12 months, both babies became euthyroid with normal growth and development.

Growth differentiation factor-15 (GDF-15) is proposed to be strongly associated with several cardiovascular diseases, such as heart failure and atherosclerosis. Moreover, some recent studies have reported an association between GDF-15 and platelet activation. In this study, we isolated peripheral blood platelets from healthy volunteers and evaluated the effect of GDF-15 on adenosine diphosphate (ADP)-induced platelet activation using the platelet aggregation assay. Subsequently, we detected the expression of GDF-15-related receptors on platelets, including the epidermal growth factor receptor (EGFR), human epidermal growth factor receptor 2 (HER2), human epidermal growth factor receptor 3 (HER3), transforming growth factor-beta receptor I (TGF-βRI), transforming growth factor-beta receptor II (TGF-βRII), glial-cell-line-derived neurotrophic factor family receptor α-like (GFRAL), and those rearranged during transfection (RET). Then, we screened for GDF-15 receptors using the GDF-15-related receptor microarray comprising these recombinant proteins. We also performed the immunoprecipitation assay to investigate the interaction between GDF-15 and the receptors on platelets. For the further exploration of signaling pathways, we investigated the effects of GDF-15 on the extracellular signal-regulated kinase (ERK), protein kinase B (AKT), and Janus kinase 2 (JAK2) pathways. We also investigated the effects of GDF-15 on the ERK and AKT pathways and platelet aggregation in the presence or absence of RET agonists or inhibition. Our study revealed that GDF-15 can dose-independently inhibit ADP-induced human platelet aggregation and that the binding partner of GDF-15 on platelets is GFRAL. We also found that GDF-15 inhibits ADP-induced AKT and ERK activation in platelets. Meanwhile, our results revealed that the inhibitory effects of GDF-15 can be mediated by the GFRAL/RET complex. These findings reveal the novel inhibitory mechanism of ADP-induced platelet activation by GDF-15.

Dynamic characteristics play a crucial role in evaluating the performance of weight sensors and are essential for achieving fast and accurate weight measurements. This study focuses on a weight sensor based on optical coherence displacement. Using finite element analysis, the sensor was numerically simulated. Frequency domain and time domain dynamic response characteristics were explored through harmonic response analysis and transient dynamic analysis. The superior dynamic performance and reduced conditioning time of the non-contact optical coherence-based displacement weight sensor were confirmed via a negative step response experiment that compared the proposed sensing method to strain sensing. Moreover, dynamic performance metrics for the optical coherence displacement-type weight sensor were determined. Ultimately, the sensor’s dynamic performance was enhanced using the pole-zero placement method, decreasing the overshoot to 4.72% and reducing the response time to 0.0132 s. These enhancements broaden the sensor’s operational bandwidth and amplify its dynamic response capabilities.

Cultural artifacts found underwater are located in complex environments with poor imaging conditions. In addition, the artifacts themselves present challenges for automated object detection owing to variations in their shape and texture caused by breakage, stacking, and burial. To solve these problems, this paper proposes an underwater cultural object detection algorithm based on the deformable deep aggregation network model for autonomous underwater vehicle (AUV) exploration. To fully extract the object feature information of underwater objects in complex environments, this paper designs a multi-scale deep aggregation network with deformable convolutional layers. In addition, the approach also incorporates a BAM module for feature optimization, which enhances the potential feature information of the object while weakening the background interference. Finally, the object prediction is achieved through feature fusion at different scales. The proposed algorithm has been extensively validated and analyzed on the collected underwater artifact datasets, and the precision, recall, and mAP of the algorithm have reached 93.1%, 91.4%, and 92.8%, respectively. In addition, our method has been practically deployed on an AUV. In the field testing over a shipwreck site, the artifact detection frame rate reached up to 18 fps, which satisfies the real-time object detection requirement.

In this study, an ultrasound-assisted green extraction method was applied for the extraction of polysaccharides from Radix Peucedani based on deep eutectic solvents (DESs), and the result showed that a DES system composed of betaine and 1,2-propylene glycol with a molar ratio of 1:2 possessed the optimal extraction efficiency for polysaccharides. Single-factor and Box–Behnken designs were used to determine the optimum extraction conditions for the maximum yields of polysaccharides from Radix Peucedani by using DESs. The maximum yields of polysaccharides attained 11.372% within a DES water content of 19%, an extraction time of 36 min, an extraction temperature of 54 °C, a solid–liquid ratio of 1:30 and an ultrasonic irradiation power of 420 W. The physicochemical properties of polysaccharides were analyzed using ICS and FT-IR, and the structure morphology was observed by SEM. The polysaccharides extracted from Radix Peucedani exhibited general antioxidant activities in vitro including DPPH, Hydroxyl and ABTS+ radical-scavenging activity. The antioxidant mechanism of Radix Peucedani polysaccharides was investigated using network pharmacology and molecular docking methods. The result showed that the high binding activity of glucose and IL1B, galactose and CASP3 was recognized as a potential mechanism for the antioxidant effects of Radix Peucedani polysaccharides.

Leucogranites in the Lalong Dome are composed of two-mica granite, muscovite granite, albite granite, and pegmatite from core to rim. Albite granite-type Be–Nb–Ta rare metal ore bodies are hosted by albite granite and pegmatite. Based on field and petrographic observations and whole-rock geochemical data, highly differentiated leucogranites have been identified in the Lalong Dome. Two-mica granites, albite granites, and pegmatites yielded monazite ages of 23.6 Ma, 21.9 Ma, and 20.6 Ma, respectively. The timing of rare metal mineralization is 20.9 Ma using U–Pb columbite dating. Leucogranites have the following characteristics: high SiO2 content (>73 wt.%); peraluminosity with high Al2O3 content (13.6–15.2 wt.%) and A/CNK (mostly > 1.1); low TiO2, CaO, and MgO content; enrichment of Rb, Th, and U; depletion of Ba, Nb, Zr, Sr, and Ti; strong negative Eu anomalies; low εNd(t) values ranging from −12.7 to −9.77. These features show that the leucogranites are crust-derived high-potassium calc-alkaline and peraluminous S-type granites derived from muscovite dehydration melting under the water-absent condition, which possibly resulted from structural decompression responding to the activity of the South Tibetan detachment system (STDS). Geochemical data imply a continuous magma fractional crystallization process from two-mica granites through muscovite granites to albite granites and pegmatites. The differentiation index (Di) gradually strengthens from two-mica granite, muscovite granite, and albite granite to pegmatite, in which albite granite and pegmatite are highest (Di = 94). The Nb/Ta and Zr/Hf ratios of albite granite and pegmatite were less than 5 and 18, respectively, which suggests that albite granite and pegmatite belong to rare metal granites and have excellent potential for rare metal mineralization.

Background/Objectives: The purpose of this study was to compare the differences in enjoyment and affect in response to four weight control intervention protocols over 12 weeks. Methods: Sixty overweight young females were randomised into four intervention groups: repeated sprint training (RST, 6-sec all-out sprint interspersed with 9-sec rest), high-intensity interval training (HIIT) with short interval (HIIT120, 1-min effort at 120% V̇O2peak) and long interval (HIIT90, 4-min effort at 90% V̇O2peak), and moderate-intensity continuous training (MICT, 60% V̇O2peak) by cycling over 12 weeks. The total workload in each training session in HIIT120, HIIT90, and MICT was confined to 200 kJ, while it was lower in RST with 57 ± 4 kJ. Enjoyment (Physical Activity Enjoyment Scale, PACES) and affective valence (Feeling Scale, FS) were measured throughout the intervention. Results: The score of the PACES on average over 12 weeks showed a significant between-group effect that was lower in MICT (80.8 ± 11.8) compared with HIIT120 (92.5 ± 11.4) and HIIT90 (96.8 ± 13.9) (p

Abstract Background Bladder cancer is one of the most lethal malignancy in urological system, and 20–25% of bladder cancer patients are muscle invasive with unfavorable prognosis. However, the role of alternative splicing (AS) in muscle-invasive bladder cancer (MIBC) remains to be elucidated. Methods Percent spliced in (PSI) data obtained from the Cancer Genome Atlas (TCGA) SpliceSeq database (n = 394) were utilized to evaluate the AS events in MIBC. Prognosis-associated AS events were screened out by univariate Cox regression. LASSO Cox regression was used to identify reliable prognostic patterns in a training set and further validated in a test set. Splicing regulatory networks were constructed by correlations between PSI of AS events and RNA expression of splicing factors. Results As a result, a total of 2589 prognosis-related AS events in MIBC were identified. Pathways of spliceosomal complex (FDR = 0.017), DNA-directed RNA polymerase II, core complex (FDR = 0.032), and base excision repair (FDR = 0.038) were observed to be significantly enriched. Additionally, we noticed that most of the prognosis-related AS events were favorable factors. According to the LASSO and multivariate Cox regression analyses, 15-AS-based signature was established with the area under curve (AUC) of 0.709, 0.823, and 0.857 at 1-, 3-, and 5- years, respectively. The MIBC patients were further divided into high- and low-risk groups based on median risk sores. Interestingly, we observed that the prevalence of FGFR3 with mutations and focal amplification was significantly higher in low-risk group. Functional and immune infiltration analysis suggested potential signaling pathways and distinct immune states between these two groups. Moreover, splicing correlation network displayed a regulatory mode of prognostic splicing factors (SF) in MIBC patients. Conclusions This study not only provided novel insights into deciphering the possible mechanism of tumorgenesis and pathogenesis but also help refine risk stratification systems and potential treatment of decision-making for MIBC.

Abstract Purpose The purpose of this review was to synthesize the empirical evidence of relevant studies related to preventive behaviors in response to the COVID-19 pandemic among children and adolescents. Further to this, we aimed to identify the demographic, psychological, and social and environmental correlates of such behaviors. Methods Following PRISMA guidelines, eligible literature was identified by searching seven databases (PsycINFO, PubMed, MEDLINE, EMBASE, Cochrane Library, PROSPERO registry platform, and ClinicalTrials.gov website) and reference list of included studies and relevant review papers from 1st Jan 2020 to 28th Feb 2021. The standardized mean difference and correlation coefficients r were extracted to estimate the effect sizes. Analyses were conducted using R software. Results Of the 35,271 original papers, 23 eligible studies were included in the qualitative synthesis and all these studies were of moderate-to-high quality, of which 17 studies were further included into the quantitative analysis. Children and adolescents (6–20 yrs.) showed a poorer practice of COVID-19 preventive behaviors compared to younger adults (21–59 yrs.) with a small-to-medium effect size (SMD = -.25, 95%CI = -.41 to -.09). For the demographic correlates, children and adolescents’ COVID-19 preventive practice was found to be significantly associated with gender (r = .14, 95%CI = .10 to .18), while not with age (r = -.02, 95%CI = -.14 to .10). Narratively, knowledge was found to be consistently and significantly correlated. For the psychological correlates, small-to-medium overall effects were identified for the association with attitudes (r = .26, 95%CI = .21 to .31) and perceived severity (r = .16, 95%CI = .01 to .30). For the family and social correlates, a non-significant association was identified between family economic status and COVID-19 preventive behaviors (r = .004, 95%CI = -.12 to .12). Conclusions Interventions and relevant policies of promoting children and adolescent’s preventive measures should be a priority. Further, empirical studies identifying the demographic, psychological, and family and social correlates of children and adolescents’ preventive behaviors are needed.

Abstract Insulator icing endangers the safe operation of overhead power lines. The current measures for image monitoring of icing overhead power lines rely on manual observation. There is an urgent need for image intelligent analysis methods with strong generalisation, accuracy, and efficiency to improve power grid decision‐making. Based on the icing monitoring data from China Southern Power Grid, this paper first proposes rules for insulator image data cleaning, classification, and annotation, establishing a dataset of insulator icing monitoring images during 2014–2018. Then, based on Yolo (You only look once) v5, a weakly supervised and phased transfer learning method is proposed to recognise insulators and ice types such as snow, rime, mixed rime, glaze, and the normal. This method reduces the interference of icing background, uneven illumination, and camera occlusion through multidimensional feature fusion. The precision, recall, and mean average precision of the recognition can reach 86.6%, 91.3%, and 90.1%, respectively, and the recognition speed is as fast as 8 ms/image. Using image pseudo‐labelling based on weakly supervised learning enables the intellectualisation of image annotation, which significantly changes the inefficiency of manual labelling.

To investigate the effect of submerged macrophytes on heterotrophic bacterioplankton communities in response to nutrient enrichment, we simulated mesocosms to test two factors, namely, the presence of Ceratophyllum demersum (L.) and the level of nutrients (slight and medium nutrient enrichment) under four possible system combinations for a duration of more than 3 months. The results show that C. demersum can affect the temporal dynamics of heterotrophic bacterioplankton density (HBD) and cause it to decrease. However, the effect of C. demersum on HBD was more pronounced under medium nutrient enrichment. The mean values of HBD in the treatment and control systems under slight nutrient enrichment were 1.30 × 105 cells mL−1 and 1.34 × 105 cells mL−1, respectively; whereas for medium nutrient enrichment, they were 1.78 × 105 cells mL−1 and 2.65 × 105 cells mL−1, respectively. The total nitrogen (TN) and total phosphorus (TP) concentrations were maintained throughout the experiment, and no significant differences were observed in the pH value, chlorophyll a (Chl. a) concentrations or dissolved organic carbon (DOC) levels between the systems with and without macrophytes, regardless of the nutrient level. Furthermore, linear mixed models revealed that environmental variables had a limited impact on HBD and that C. demersum had no significant direct effect on the environmental variables in the systems. A likely explanation is higher predation on bacterioplankton in the mesocosms, although allelopathic effects exerted by C. demersum cannot be excluded.

IntroductionThis article aimed to compare the prevalence of dyslipidemia and determine risk factors associated with lipid levels in a cohort of HIV-infected patients receiving two different antiretroviral therapy (ART) regimens, nucleoside reverse transcriptase inhibitor/non-nucleoside reverse transcriptase inhibitor (NRTI/NNRTI) and nucleoside reverse transcriptase inhibitor/integrase strand transfer inhibitor (NRTI/INSTI).MethodsThis longitudinal study analyzed 633 HIV-infected patients with complete blood lipid profile records for at least 1 year at the ART clinic of Zhongnan Hospital of Wuhan University, China, from June 2018 to March 2021. Demographic and clinical data, including age, gender, body weight, height, current/former/non-smoker, current drinker, diabetes mellitus, hypertension, were extracted from electronic medical records. Laboratory tests included hematology, total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), Lipoprotein(a) and CD4 cell count. Abstract As the lowest organisms possessing T cells, fish are instrumental for understanding T cell evolution and immune defense in early vertebrates. This study established in Nile tilapia models suggests that T cells play a critical role in resisting Edwardsiella piscicida infection via cytotoxicity and are essential for IgM+ B cell response. CD3 and CD28 monoclonal antibody crosslinking reveals that full activation of tilapia T cells requires the first and secondary signals, while Ca2+–NFAT, MAPK/ERK, NF‐κB, and mTORC1 pathways and IgM+ B cells collectively regulate T cell activation. Thus, despite the large evolutionary distance, tilapia and mammals such as mice and humans exhibit similar T cell functions. Furthermore, it is speculated that transcriptional networks and metabolic reprogramming, especially c‐Myc‐mediated glutamine metabolism triggered by mTORC1 and MAPK/ERK pathways, underlie the functional similarity of T cells between tilapia and mammals. Notably, tilapia, frogs, chickens, and mice utilize the same mechanisms to facilitate glutaminolysis‐regulated T cell responses, and restoration of the glutaminolysis pathway using tilapia components rescues the immunodeficiency of human Jurkat T cells. Thus, this study provides a comprehensive picture of T cell immunity in tilapia, sheds novel perspectives for understanding T cell evolution, and offers potential avenues for intervening in human immunodeficiency.

Clustered regularly interspaced short palindromic repeats (CRISPR) and their CRISPR-associated proteins (Cas) are an adaptive immune system involved in specific defenses against the invasion of foreign mobile genetic elements, such as plasmids and phages. This study aims to analyze the gene structure and to explore the function of the CRISPR system in the Enterococcus genome, especially with regard to drug resistance. The whole genome information of 110 enterococci was downloaded from the NCBI database to analyze the distribution and the structure of the CRISPR-Cas system including the Cas gene, repeat sequences, and spacer sequence of the CRISPR-Cas system by bioinformatics methods, and to find drug resistance-related genes and analyze the relationship between them and the CRISPR-Cas system. Multilocus sequence typing (MLST) of enterococci was performed against the reference MLST database. Information on the drug resistance of Enterococcus was retrieved from the CARD database, and its relationship to the presence or absence of CRISPR was statistically analyzed. Among the 110 Enterococcus strains, 39 strains (35.45%) contained a complete CRISPR-Cas system, 87 CRISPR arrays were identified, and 62 strains contained Cas gene clusters. The CRISPR system in the Enterococcus genome was mainly type II-A (59.68%), followed by type II-C (33.87%). The phylogenetic analysis of the cas1 gene sequence was basically consistent with the typing of the CRISPR-Cas system. Of the 74 strains included in the study for MLST typing, only 19 (25.68%) were related to CRISPR-Cas typing, while the majority of the strains (74.32%) of MLST typing were associated with the untyped CRISPR system. Additionally, the CRISPR-Cas system may only be related to the carrying rate of some drug-resistant genes and the drug-resistant phenotype. In conclusion, the distribution of the enterococcus CRISPR-Cas system varies greatly among different species and the presence of CRISPR loci reduces the horizontal transfer of some drug resistance genes.

Summary: As the frontline defense against avian brood parasitism, nest defense is important in reducing nest parasitism and increasing host fitness. However, systematic studies on its effectiveness (i.e., on whether it successfully prevents cuckoo parasitism) are scarce. Based on 214 video recordings from 10 years of field observations, we evaluated the effectiveness of nest defenses of the Oriental reed warbler (Acrocephalus orientalis) in deterring common cuckoo (Cuculus canorus) parasitism. Under a breeding pair situation (≤ 2 hosts individuals), warblers were hardly effective in resisting the egg-laying of the cuckoo and many of the cuckoos under attack were able to complete the parasitic process without being harmed. However, when hosts ≥ 3 individuals, the warblers could effectively thwart the cuckoos and cause lethal damage to the cuckoo in partial cases. This indicated that the host group effectively resisted the cuckoo, demonstrating the importance of cooperation among host neighbors in resisting cuckoo parasitism.

Mechanical engineering research focuses on realizing the monitoring and control of the dynamic performance of mechanical systems, as well as realizing the integration of design and system control. OBJECTIVES: In order to improve the disassembly efficiency, reduce the disassembly cost and disassembly energy consumption, it is optimized using social engineering methods to achieve better results and reduce the disassembly cost and energy consumption. METHODS: Aiming at the drive and anti-skid control strategy of four-wheel hub motor, it was simulated using improved social engineering algorithms, and based on this, three road recognition algorithms were selected for low, medium, and high adhesion road verification. RESULTS: Through the study of automobile anti-skid control system, the basic structure of automobile anti-skid control system is summarized and some solution measures are proposed. A new type of drive anti-skid control system is proposed for the problems of high vibration and noise of automobile brake. The drive anti-slip control system is characterized by simple structure, easy maintenance, simple control and reliable operation, and high operation efficiency. CONCLUSION: This study shows that the system not only has excellent drive anti-slip effect, but also has good control performance. In addition, this drive anti-slip system is able to ensure the safe and reliable operation of mechanical brakes in various harsh environments. This new drive anti-slip control system is a new type of drive device that can be widely used for driving force on various mechanical brakes and drive wheels, and the study of this device is of great significance.

PurposeThe aim of this study was to explore the treatment outcomes of a novel single lateral approach via fibular fracture line for patients with posterior pilon fractures.Patients and methodsFrom January 2020 to December 2021, a total of 41 patients with posterior pilon fractures who received surgical treatment in our hospital were retrospectively reviewed. Twenty patients (Group A) were treated with open reduction and internal fixation (ORIF) via posterolateral approach. Twenty-one patients (Group B) were treated with ORIF using a simple single lateral approach via stretching fibular fracture line. Clinical assessments, including operation time, intraoperative blood loss, the American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hindfoot score, visual analogue scale (VAS), and the active range of motion (ROM) of the ankle at the final follow-up visit after surgery, were performed in all patients. Radiographic outcome was evaluated by using the criteria proposed by Burwell and Charnley.ResultsThe mean follow-up time was 21 months (range 12–35). The average operation time and intraoperative blood loss in the Group B were significantly less than those in the Group A. Moreover, the AOFAS score and ankle ROM in the Group B were significantly higher than those in the Group A at the final follow-up visit. Eighteen cases (90%) in Group A and 19 cases (90.5%) in Group B achieved anatomical reduction of the fracture.ConclusionThe single lateral approach via stretching fibular fracture line is a simple and effective technique for reduction and fixation of posterior pilon fractures.
