The relationship between aluminium (Al) and plants has been investigated for a long time. Al is typically harmful to plants, however Qualea grandiflora Mart., a compulsory accumulator of this metal, may benefit from it. To understand this peculiarity, proteomic and metabolite profiling analyses were carried out to investigate, at physiological, metabolic, and molecular levels, the relevance of Al for this species. Qualea grandiflora plants were grown with MS medium with or without Al-supplementation for 120 days. Metabolite profiling using GC-MS and growth analysis were conducted to evaluate the biochemical and physiological effects of Al. Concomitantly, root proteins were extracted, identified and quantified using label-free LC-MS/MS. Al-starved Q. grandiflora plants had shorter shoots, roots, lower biomass, and chlorotic leaves. Furthermore, there was a high coherence between proteomic and metabolomic data. Al was critical for cell wall and lignin syntheses, genetic information processing, and organic acid metabolism. Also, Al might help P uptake in this plant. Qualea grandiflora plants needed Al to grow and develop properly. Moreover, there was a good match between the data from physiological/metabolic analyses and the upregulated metabolic pathways indicated by proteome investigation. Hence, we propose that Q. grandiflora root metabolism is highly dependent on Al.

This paper proposes a novel approach for detection of stator short-circuit faults in three-phase induction motors. The method is based on two stages: feature extraction and classification by intelligent systems. First, mutual information is estimated from delayed stator current signals, which are used as inputs of C4.5 decision trees and multilayer perceptron neural networks in the second step. Several offline and online experimental tests are presented considering voltage unbalance, load torque variations, and 1% to 10% short-circuit levels. The obtained results corroborate the effectiveness of this new diagnostic approach.

In patients with severe forms of COVID-19, thromboelastometry has been reported to display a hypercoagulant pattern. However, an algorithm to differentiate severe COVID-19 patients from nonsevere patients and healthy controls based on thromboelastometry parameters has not been developed. Forty-one patients over 18 years of age with positive qRT-PCR for SARS-CoV-2 were classified according to the severity of the disease: nonsevere (NS, n=20) or severe (S, n=21). A healthy control (HC, n=9) group was also examined. Blood samples from all participants were tested by extrinsic (EXTEM), intrinsic (INTEM), non-activated (NATEM) and functional assessment of fibrinogen (FIBTEM) assays of thromboelastometry. The thrombodynamic potential index (TPI) was also calculated. Severe COVID-19 patients exhibited a thromboelastometry profile with clear hypercoagulability, which was significantly different from the NS and HC groups. Nonsevere COVID-19 cases showed a trend to thrombotic pole. The NATEM test suggested that nonsevere and severe COVID-19 patients presented endogenous coagulation activation (reduced clotting time and clot formation time). TPI data were significantly different between the NS and S groups. The maximum clot firmness profile obtained by FIBTEM showed moderate/elevated accuracy to differentiate severe patients from NS and HC. A decision tree algorithm based on the FIBTEM-MCF profile was proposed to differentiate S from HC and NS. Thromboelastometric parameters are a useful tool to differentiate the coagulation profile of nonsevere and severe COVID-19 patients for therapeutic intervention purposes.

People with disabilities living in Brazil face great difficulties in the tasks of daily life mainly due to the lack of accessibility in public spaces, products and services. In this context, we noticed a lack of a computing tool that embraces both people with visual and hearing impairment. This work presents TATU -- a mobile application for both Android and iOS platforms aimed at supporting people with visual or hearing impairment to enjoy Brazilian tourist attractions, including both open-air and indoor spaces. TATU application has an adaptive interface exclusively designed for each of the impairment user profiles, it can work on guided tour mode by indoor navigation using BLE beacons and outdoor navigation using GPS. Our solution was evaluated by three experiments, one of which was carried out with blind volunteers and TATU application obtained satisfactory results for both spacious attractions with the lowest density of collection items and for the smallest spaces.

Background Since the beginning of the COVID-19 pandemic, the world’s attention has been focused on better understanding the relation between the human host and the SARS-CoV-2 virus, as its action has led to hundreds of thousands of deaths. Objective In this context, we decided to study certain consequences of the abundant cytokine release over the innate and adaptive immune systems, inflammation, and hemostasis, comparing mild and severe forms of COVID-19. Methods To accomplish these aims, we will analyze demographic characteristics, biochemical tests, immune biomarkers, leukocyte phenotyping, immunoglobulin profile, hormonal release (cortisol and prolactin), gene expression, thromboelastometry, neutralizing antibodies, metabolic profile, and neutrophil function (reactive oxygen species production, neutrophil extracellular trap production, phagocytosis, migration, gene expression, and proteomics). A total of 200 reverse transcription polymerase chain reaction–confirmed patients will be enrolled and divided into two groups: mild/moderate or severe/critical forms of COVID-19. Blood samples will be collected at different times: at inclusion and after 9 and 18 days, with an additional 3-day sample for severe patients. We believe that this information will provide more knowledge for future studies that will provide more robust and useful clinical information that may allow for better decisions at the front lines of health care. Results The recruitment began in June 2020 and is still in progress. It is expected to continue until February 2021. Data analysis is scheduled to start after all data have been collected. The coagulation study branch is complete and is already in the analysis phase. Conclusions This study is original in terms of the different parameters analyzed in the same sample of patients with COVID-19. The project, which is currently in the data collection phase, was approved by the Brazilian Committee of Ethics in Human Research (CAAE 30846920.7.0000.0008). Black rot is a severe disease caused by the bacterium Xanthomonas campestris pv. campestris (Xcc), which can lead to substantial losses in cruciferous vegetable production worldwide. Although the use of resistant cultivars is the main strategy to control this disease, there are limited sources of resistance. In this study, we used the LC-MS/MS technique to analyze young cabbage leaves and chloroplast-enriched samples at 24 h after infection by Xcc, using both susceptible (Veloce) and resistant (Astrus) cultivars. A comparison between susceptible Xcc-inoculated plants and the control condition, as well as between resistant Xcc-inoculated plants with the control was performed and more than 300 differentially abundant proteins were identified in each comparison. The chloroplast enriched samples contributed with the identification of 600 additional protein species in the resistant interaction and 900 in the susceptible one, which were not detected in total leaf sample. We further determined the expression levels for 30 genes encoding the identified differential proteins by qRT-PCR. CHI-B4 like gene, encoding an endochitinase showing a high increased abundance in resistant Xcc-inoculated leaves, was selected for functional validation by overexpression in Arabidopsis thaliana. Compared to the wild type (Col-0), transgenic plants were highly resistant to Xcc indicating that CHI-B4 like gene could be an interesting candidate to be used in genetic breeding programs aiming at black rot resistance.

A significant number of studies have demonstrated that paternal exercise modulates future generations via effects on the sperm epigenome. However, comprehensive information regarding the effects of exercise performed by the father on different tissues and their clinical relevance has not yet been explored in detail. This narrative review is focused on the effects of paternal exercise training on various physiological systems of offspring. A detailed mechanistic understanding of these effects could provide crucial clues for the exercise physiology field and aid the development of therapeutic approaches to mitigate disorders in future generations. Non-coding RNA and DNA methylation are major routes for transmitting epigenetic information from parents to offspring. Resistance and treadmill exercise are the most frequently used modalities of planned and structured exercise in controlled experiments. Paternal exercise orchestrated protective effects over changes in fetus development and placenta inflammatory status. Moreover paternal exercise promoted modifications in the ncRNA profiles, gene and protein expression in the hippocampus, left ventricle, skeletal muscle, tendon, liver and pancreas in the offspring, while the transgenerational effects are unknown. Paternal exercise demonstrates clinical benefits to the offspring and provides a warning on the harmful effects of a paternal unhealthy lifestyle. Exercise in fathers is presented as one of the most logical and cost-effective ways of restoring health in the offspring and, consequently, modifying the phenotype. It is important to consider that paternal programming might have unique significance in the developmental origins of offspring diseases.

The present study was conducted to decipher the proteome of in vivo-produced pre-implantation ovine embryos. Ten locally adapted Morana Nova ewes received hormonal treatment and were inseminated 12 hr after ovulation. Six days later, 54 embryos (morula and blastocyst developmental state) were recovered from eight ewes and pooled to obtain sufficient protein for proteomic analysis. Extracted embryo proteins were analysed by LC-MS/MS, followed by identification based on four database searches (PEAKS, Proteome Discoverer software, SearchGUI software, PepExplorer). Identified proteins were analysed for gene ontology terms, protein clusters and interactions. Genes associated with the ovine embryo proteome were screened for miRNA targets using data sets of TargetScan (http://www.targetscan.org) and mIRBase (http://www.mirbase.org) servers. There were 667 proteins identified in the ovine embryos. Biological processes of such proteins were mainly related to cellular process and regulation, and molecular functions, to binding and catalytic activity. Analysis of the embryo proteins revealed 49 enriched functional clusters, linked to energy metabolism (TCA cycle, pyruvate and glycolysis metabolism), zona pellucida (ZP), MAPK signalling pathway, tight junction, binding of sperm to ZP, translation, proteasome, cell cycle and calcium/phospholipid binding. Sixteen miRNAs were related to 25 pre-implantation ovine embryo genes, all conserved in human, bovine and ovine species. The interaction network generated by miRNet showed four key miRNAs (hsa-mir-106b-5p; hsa-mir-30-5p; hsa-mir-103a-5p and hsa-mir-106a-5p) with potential interactions with embryo-expressed genes. Functional analysis of the network indicated that miRNAs modulate genes related to cell cycle, regulation of stem cell and embryonic cell differentiation, among others. Retrieved miRNAs also modulate the expression of genes involved in cell signalling pathways, such as MAPK, Wnt, TGF-beta, p53 and Toll-like receptor. The current study describes the first major proteomic profile of 6-day-old ovine embryos produced in vivo, setting a comprehensive foundation for our understanding of embryo physiology in the ovine species.

Herein the descriptive label-free mass spectrometry (LC-MS/MS) proteome of Erythroxylum pungens (roots, barks, leaves) harvested from its natural habitat is described. Processes related to defense were most important in roots. Main protective proteins differed among organs, suggesting part-specific strategies of environmental adaptation.

Scorpion venoms are formed by toxins harmful to various organisms, including humans. Several techniques have been developed to understand the role of proteins in animal venoms, including proteomics approach. Rhopalurus agamemnon (Koch, 1839) is the largest scorpion in the Buthidae family in the Brazilian Cerrado, measuring up to 110 mm in total length. The accident with R. agamemnon is painful and causes some systemic reactions, but the specie's venom remains uninvestigated. We explore the venom protein composition using a proteomic and a biological-directed approach identifying 230 protein compounds including enzymes like Hyaluronidase, metalloproteinase, L-amino acid oxidase and amylase, the last two are first reported for scorpion venoms. Some of those new reports are important to demonstrate how distant we are from a total comprehension of the diversity about venoms in general, due to their diversity in composition and function. Biological significance In this study, we explored the composition of venom proteins from the scorpion Rhopalurus agamemnon. We identified 230 proteins from the venom including new enzyme reports. These data highlight the unique diversity of the venom proteins from the scorpion R. agamemnon, provide insights into new mechanisms of envenomation and enlarge the protein database of scorpion venoms. The discovery of new proteins provides a new scenario for the development of new drugs and suggests molecular targets to venom components.

Dental pulp is a specialized tissue able to respond to infectious processes. Nevertheless, infection progress and root canal colonization trigger an immune-inflammatory response in tooth-surrounding tissues, leading to apical periodontitis and bone tissue destruction, further contributing to tooth loss. In order to shed some light on the effects of IL-4 on periradicular pathology development modulation, microtomographic, histological and proteomic analyses were performed using 60 mice, 30 wild type and 30 IL-4−/−. For that, 5 animals were used for microtomographic and histological analysis, and another 5 for proteomic analysis for 0, 7 and 21 days with/without pulp exposure. The periapical lesions were established in WT and IL-4−/− mice without statistical differences in their volume, and the value of p Significance: IL-4 might play a protective role in diseases involving bone destruction and its activity may contribute to host protection, mainly due to its antiosteoclastogenic action.

Amphibian skin secretions are abundant in bioactive compounds, especially antimicrobial peptides. These molecules are generally cationic and rich in hydrophobic amino acids, have an amphipathic structure and adopt an &alpha, helical conformation when in contact with microorganisms membranes. In this work, we purified and characterized Figainin 1, a novel antimicrobial and antiproliferative peptide from the cutaneous secretion of the frog Boana raniceps. Figainin 1 is a cationic peptide with eighteen amino acid residues&mdash, rich in leucine and isoleucine, with an amidated C-terminus&mdash, and adopts an &alpha, helical conformation in the presence of trifluoroethanol (TFE). It displayed activity against Gram-negative and especially Gram-positive bacteria, with MIC values ranging from 2 to 16 &micro, M, and showed an IC50 value of 15.9 &micro, M against epimastigote forms of T. cruzi, however, Figanin 1 did not show activity against Candida species. This peptide also showed cytolytic effects against human erythrocytes with an HC50 of 10 &micro, M, in addition to antiproliferative activity against cancer cells and murine fibroblasts, with IC50 values ranging from 10.5 to 13.7 &micro, M. Despite its adverse effects on noncancerous cells, Figainin 1 exhibits interesting properties for the development of new anticancer agents and anti-infective drugs against pathogenic microorganisms.

Intestinal ischemia reperfusion injury (iIRI) is a severe clinical condition presenting high morbidity and mortality worldwide. Some of the systemic consequences of IRI can be prevented by applying ischemic preconditioning (IPC), a series of short ischemia/reperfusion events preceding the major ischemia. Although neutrophils are key players in the pathophysiology of ischemic injuries, neither the dysregulation presented by these cells in iIRI nor the protective effect of iIPC have their regulation mechanisms fully understood. Protein phosphorylation, as well as the regulation of the respective phosphatases and kinases are responsible for regulating a large number of cellular functions in the inflammatory response. Moreover, in previous work we found hydrolases and transferases to be modulated in iIR and iIPC, suggesting the possible involvement of phosphatases and kinases in the process. Therefore, in the present study, we analyzed the phosphoproteome of neutrophils from rats submitted to mesenteric ischemia and reperfusion, either submitted or not to IPC, compared to quiescent controls and sham laparotomy. Proteomic analysis was performed by multi-step enrichment of phosphopeptides, isobaric labeling, and LC-MS/MS analysis. Bioinformatics was used to determine phosphosite and phosphopeptide abundance and clustering, as well as kinases and phosphatases sites and domains. We found that most of the phosphorylation-regulated proteins are involved in apoptosis and migration, and most of the regulatory kinases belong to CAMK and CMGC families. An interesting finding revealed groups of proteins that are modulated by iIR, but such modulation can be prevented by iIPC. Among the regulated proteins related to the iIPC protective effect, Vamp8 and Inpp5d/Ship are discussed as possible candidates for control of the iIR damage.

Aerobic organisms require oxygen for energy. In the course of the infection, adaptation to hypoxia is crucial for survival of human pathogenic fungi. Members of the Paracoccidioides complex face decreased oxygen tensions during the life cycle stages. In Paracoccidioides brasiliensis proteomic responses to hypoxia have not been investigated and the regulation of the adaptive process is still unknown, and this approach allowed the identification of 216 differentially expressed proteins in hypoxia using iTRAQ-labelling. Data suggest that P. brasiliensis reprograms its metabolism when submitted to hypoxia. The fungus reduces its basal metabolism and general transport proteins. Energy and general metabolism were more representative and up regulated. Glucose is apparently directed towards glycolysis or the production of cell wall polymers. Plasma membrane/cell wall are modulated by increasing ergosterol and glucan, respectively. In addition, molecules such as ethanol and acetate are produced by this fungus indicating that alternative carbon sources probably are activated to obtain energy. Also, detoxification mechanisms are activated. The results were compared with label free proteomics data from Paracoccidioides lutzii. Biochemical pathways involved with acetyl-CoA, pyruvate and ergosterol synthesis were up-regulated in both fungi. On the other hand, proteins from TCA, transcription, protein fate/degradation, cellular transport, signal transduction and cell defense/virulence processes presented different profiles between species. Particularly, proteins related to methylcitrate cycle and those involved with acetate and ethanol synthesis were increased in P. brasiliensis proteome, whereas GABA shunt were accumulated only in P. lutzii. The results emphasize metabolic adaptation processes for distinct Paracoccidioides species.

Spider venoms are composed of a complex mixture of bioactive molecules. The structural and functional characterization of these molecules in the venom of the Brazilian spider Acanthoscurria natalensis, has been little explored. The venom was fractionated using reversed-phase liquid chromatography. The fraction with hyaluronidase activity was named AnHyal. The partial sequencing of AnHyal revealed the presence of a CRISP-like protein, in addition to hyaluronidase, comprising 67% coverage for hyaluronidase from Brachypelma vagans and 82% for CRISP-like protein from Grammostola rosea. 1D BN-PAGE zymogram assays of AnHyal confirmed the presence of enzymatically active 53 kDa monomer and 124 and 178 kDa oligomers. The decomposition of the complexes by 2D BN/SDS-PAGE zymogram assays showed two subunits, 53 (AnHyalH) and 44 kDa (AnHyalC), with sequence similarity to hyaluronidase and CRISP proteins, respectively. The secondary structure of AnHyal is composed by 36% of α-helix. AnHyal presented maximum activity at pH between 4.0 and 6.0 and 30 and 60 °C, showed specificity to hyaluronic acid substrate and presented a KM of 617.9 μg/mL. Our results showed that hyaluronidase and CRISP proteins can form a complex and the CRISP protein may contribute to the enzymatic activity of AnHyalH.

In recent years, the number of new antimicrobial drugs launched on the market has decreased considerably even though there has been an increase in the number of resistant microbial strains. Thus, antimicrobial resistance has become a serious public health problem. Amphibian skin secretions are a rich source of host defense peptides, which generally are cationic and hydrophobic molecules, with a broad-spectrum of activity. In this study, one novel multifunctional defense peptide was isolated from the skin secretion of the Chaco tree frog, Boana raniceps. Figainin 2 (1FLGAILKIGHALAKTVLPMVTNAFKPKQ28) is cationic and hydrophobic, adopts an &alpha, helical structure in 50% (v/v) trifluoroethanol (TFE), and is thermally stable. This peptide exhibited activity against Gram-negative and Gram-positive pathogenic bacteria arboviruses, T. cruzi epimastigotes, however, it did not show activity against yeasts. Figainin 2 also showed antiproliferative activity on cancer cells, is moderately active on human erythrocytes, and activates the oxidative burst in human neutrophils.

Fever is a brain-mediated increase in body temperature mainly during inflammatory or infectious challenges. Although there is considerable data regarding the inflammation pathways involved in fever, metabolic alterations necessary to orchestrate the complex inflammatory response are not totally understood. We performed proteomic analysis of rat hypothalamus using label-free LC–MS/MS in a model of fever induced by lipopolysaccharide (LPS) or prostaglandin E2 (PGE2). In total, 7021 proteins were identified. As far as we know, this is the largest rat hypothalamus proteome dataset available to date. Pathway analysis showed proteins from both stimuli associated with inflammatory and metabolic pathways. Concerning metabolic pathways, rats exposed to LPS or PGE2 presented lower relative abundance of proteins involved in glycolysis, pentose phosphate pathway and tricarboxylic acid cycle. Mitochondrial function may also be altered by both stimuli because significant downregulation of several proteins was found, mainly in complexes I and IV. LPS was able to induce downregulation of important proteins in the enzymatic antioxidant system, thereby contributing to oxidative stress. The results offered comprehensive information about fever responses and helped to reveal new insights into proteins potentially involved in inflammatory signaling and metabolic changes in the hypothalamus during systemic LPS and central PGE2 administration. Significance The evolutionary persistence of fever, despite the elevated cost for maintenance of this response, suggests that elevation in core temperature may represent an interesting strategy for survival. Fever response is achieved through the integrated behavioral, physiological, immunological and biochemical processes that determine the balance between heat generation and elimination. The development of such complex response arouses interest in studying how the cell metabolism responds or even contributes to promote fever. Our results offered comprehensive information about fever responses, including metabolic and inflammatory pathways, providing new insights into candidate proteins potentially involved in inflammatory signaling and metabolic changes in the hypothalamus during fever induced by systemic LPS and central PGE2 perturbation.

The calcaneal tendon (CT) is the most commonly injured tendon in the human body. Moreover, with advancing age, the amount of damage increases further. Resistance training (RT) could be used to minimize such damages. The aim of the present study was to obtain the identification, detailed protein cataloging and biochemical characterization based on the effects of the aging process and the RT in CT of rats. The analysis by liquid chromatography tandem-mass spectrometry showed 142 distinct proteins, however, only 29 proteins met the inclusion criteria and were analyzed. Aging causes a reduction in the abundance of seven proteins related to extracellular matrix organization, biological regulation and cellular processes. However, RT promoted the positive regulation of proteins important for the maintenance of healthy tendons: seven proteins in young trained and two in older trained group. This study contributes to a better understanding of molecular aspects of the tendon. The down regulation of proteins linked to mechanical function and extracellular matrix remodeling of the tendon during aging can contribute to the increase of injury and weakness in the tendon. Nevertheless, RT proved to be a tool to prevent these adverse effects during aging by increasing proteins involved in the functionality of the tendon.

Three-phase induction motors are the primary means of transformation of electrical energy into mechanical energy in industry, since they are robust and present low cost. However, despite being robust, these machines are subject to electrical or mechanical faults. Thus, identifying a defect in a running motor may decrease the risk of possible damage. This paper proposes an alternative approach to identify defects in the stator of these motors, by analyzing current signals in the time domain. In addition, it presents the determination of the consequent fault severity by means of two proposals: 1) a multiagent system with a classifier behavior; and 2) a neural estimator. The faults observed are related to short circuits between turns in the stator coil of 1% to 10%. Experimental results are observed with motors of different powers, under various adverse situations of electrical feed and a wide range of load conditions on the machine shaft.

We have investigated the potential for analysis of the complex peptide mixtures secreted from frog skin using off-line LC-MALDI MS/MS. Since only limited information about the sequence of such peptides is available, de novo sequencing followed by Blast search was needed. An automated workflow has been applied based on automated collection of the chromatographic fractions on the MALDI target followed by automated recording of the MS/MS spectra of the 10 most intense peaks in each fraction. The MS/MS spectra were submitted to de novo sequencing by the Peaks software and searched for potential matches in an amphibian database extracted from UniProt. A number of known skin secreted peptides as well as precursor proteins for these were identified. However, most of the recorded MS/MS data and deduced sequences did not result in matches with the databases. These sequences might serve as a reference for future studies of peptides secreted from frogs.

Ancestral obesogenic exposure is able to trigger harmful effects in the offspring left ventricle (LV) which could lead to cardiovascular diseases. However, the impact of the father’s lifestyle on the offspring LV is largely unexplored. The aim of this study was to investigate the effects of 8 weeks of paternal resistance training (RT) on the offspring left ventricle (LV) proteome exposed to control or high-fat (HF) diet. Wistar rats were randomly divided into two groups: sedentary fathers and trained fathers (8 weeks, 3 times per week with weights secured to the animals’ tails). The offspring were obtained by mating with sedentary females. Upon weaning, male offspring were divided into 4 groups (5 animals per group): offspring from sedentary fathers, exposed to control diet (SFO-C); offspring from trained fathers, exposed to control diet (TFO-C); offspring from sedentary fathers, exposed to high-fat diet (SFO-HF); and offspring from trained fathers, exposed to high-fat diet (TFO-HF). The LC-MS/MS analysis revealed 537 regulated proteins among groups. Offspring exposure to HF diet caused reduction in the abundance levels of proteins related to cell component organization, metabolic processes, and transport. Proteins related to antioxidant activity, transport, and transcription regulation were increased in TFO-C and TFO-HF as compared with the SFO-C and SFO-HF groups. Paternal RT demonstrated to be an important intervention capable of inducing significant effects on the LV proteome regardless of offspring diet due to the increase of proteins involved into LV homeostasis maintenance. This study contributes to a better understanding of the molecular aspects involved in transgenerational inheritance.

This dataset brief is about the descriptive proteome of Qualea grandiflora plants by label free mass spectrometry (LC-MS/MS). Q. grandiflora is a plant that accumulates aluminum (Al) in high quantities and requires it for growth and development. Although quite relevant for the understanding of Al effects on plants, the proteome of Q. grandiflora has not been studied yet. Therefore, the current proteome analysis identifies a total of 2010 proteins. Furthermore, the identified Q. grandiflora root proteins are associated with several crucial molecular functions, biological processes, and cellular sites. Hence, the proteome analysis of Q. grandiflora will contribute to unravel how plants evolved to cope with high levels of Al in soils. Holocellulase production by Aspergillus niger using raw sugarcane bagasse (rSCB) as the enzyme-inducing substrate is hampered by the intrinsic recalcitrance of this material. Here we report that mild hydrothermal pretreatment of rSCB increases holocellulase secretion by A. niger. Quantitative proteomic analysis revealed that pretreated solids (PS) induced a pronounced up-regulation of endoglucanases and cellobiohydrolases compared to rSCB, which resulted in a 10.1-fold increase in glucose release during SCB saccharification. The combined use of PS and pretreatment liquor (PL), referred to as whole pretreated slurry (WPS), as carbon source induced a more balanced up-regulation of cellulases, hemicellulases and pectinases and resulted in the highest increase (4.8-fold) in the release of total reducing sugars from SCB. The use of PL as the sole carbon source induced the modulation of A. niger’s secretome towards hemicellulose degradation. Mild pretreatment allowed the use of PL in downstream biological operations without the need for undesirable detoxification steps.

A better understanding of the proteome profile after bipolar disorder (BD) and schizophrenia (SCZ) treatment, besides monitoring disease progression, may assist on the development of novel therapeutic strategies with the ability to reduce or control possible side effects. In this pilot study, proteomics analysis employing nano liquid chromatography coupled to mass spectrometry (nLC-MS) and bioinformatic tools were applied to identify differentially abundant proteins in serum of treated BD and SCZ patients. In total, 10 BD patients, 10 SCZ patients, and 14 healthy controls (HC) were included in this study. 24 serum proteins were significantly altered (p 0.58, 8 proteins presented lower abundance in the BD group, while 7 proteins presented higher abundance and 2 lower abundance in SCZ group when compared against HC. Bioinformatics analysis based on these 24 proteins indicated two main altered pathways previously described in the literature; furthermore, it revealed that opposite abundances of the complement and coagulation cascades were the most significant biological processes involved in these pathologies. Moreover, we describe disease-related proteins and pathways associations suggesting the necessity of clinical follow-up improvement besides treatment, as a precaution or safety measure, along with the disease progression. Further biological validation and investigations are required to define whether there is a correlation between complement and coagulation cascade expression for BD and SCZ and cardiovascular diseases.

A comparative proteomic analysis between two near-isogenic rice lines, displaying a resistant and susceptible phenotype upon infection with Magnaporthe oryzae was performed. We identified and validated factors associated with rice disease susceptibility, representing a flourishing source toward a more resolute rice-blast resistance. Proteome profiles were remarkably different during early infection (12 h post-inoculation), revealing several proteins with increased abundance in the compatible interaction. Potential players of rice susceptibility were selected and gene expression was evaluated by RT-qPCR. Gene Ontology analysis disclosed susceptibility gene-encoded proteins claimed to be involved in fungus sustenance and suppression of plant immunity, such as sucrose synthase 4-like, serpin-ZXA-like, nudix hydrolase15, and DjA2 chaperone protein. Two other candidate genes, picked from a previous transcriptome study, were added into our downstream analysis including pyrabactin resistant-like 5 (OsPYL5), and rice ethylene-responsive factor 104 (OsERF104). Further, we validated their role in susceptibility by Transient-Induced Gene Silencing (TIGS) using short antisense oligodeoxyribonucleotides that resulted in a remarkable reduction of foliar disease symptoms in the compatible interaction. Therefore, we successfully employed shotgun proteomics and antisense-based gene silencing to prospect and functionally validate rice potential susceptibility factors, which could be further explored to build rice-blast resistance. Significance R gene-mediated disease resistance is race-specific and often not durable in the field. More recently, advancements in new breeding techniques (NBTs) have made plant disease susceptibility genes (S-genes) a new target to build a broad spectrum and more durable resistance, hence an alternative source to R-genes in breeding programs. We successfully coupled shotgun proteomics and gene silencing tools to prospect and validate new rice-bast susceptibility genes that can be further exploited toward a more resolute blast disease resistance.

The protozoan Trypanosoma cruzi is the causative agent of the neglected infectious illness Chagas disease. During its life cycle it differentiates into replicative and non-replicative life stages. So far, T. cruzi cell division has been investigated by transcriptomics but not by proteomics approaches. Here we show the first quantitative proteome analysis of T. cruzi cell division. T. cruzi epimastigote cultures were subject to synchronization with hydroxyurea and harvested at different time points. Analysis by flow cytometry, bright field and fluorescence microscopy indicated that samples collected at 0 h, 2 h, 6 h and 14 h overrepresented G1, G1-S, S and M cell cycle phases, respectively. After trypsin digestion of these samples, the resulting peptides were labelled with iTRAQ and subjected to LC-MS/MS. Also, iTRAQ-labelled phosphopeptides were enriched with TiO2 to access the phosphoproteome. Overall, 597 protein groups and 94 phosphopeptides presented regulation with the most remarkable variation in abundance at 6 h (S-phase). Comparison of our proteomic data to previous transcriptome-wise analysis of epimastigote cell cycle showed 16 sequence entries in common, with the highest mRNA/protein correlation observed in transcripts with peak abundance in G1-phase. Our data revealed regulated proteins and phosphopeptides which play important roles in the control of cell division in other organisms and some of them were previously detected in the nucleus or associated with T. cruzi chromatin.

Extracellular vesicles (EVs), including exosomes, play a key role in inter and intracellular communication, promoting the proliferation and invasion of recipient cells to support tumor growth and metastasis. Metastasis comprises multiple steps that first include the detachment of tumor cells through epithelial to mesenchymal transition (EMT), allowing the physical dissemination to distant organs. Thereafter, cancer-derived exosomes are still critical components for preparing the tumor microenvironment by (i) enabling tumor cells to escape from the immunological surveillance and (ii) arranging the pre-metastatic site for the engraftment of detached cancer cells. In this review, we discuss the multifaceted role of EVs in the multiple steps of metastasis. Future research directions draw attention to EVs as biological targets for cancer diagnosis, prognosis and therapy. However, due to their significant role in cell communication, they may become a valuable drug delivery system.

The three-phase induction motors are considered one of the most important elements of the industrial process. However, in this environment, these machines are subject to electrical and mechanical faults, which may cause significant financial losses. Thus, the purpose of this paper is to present a pattern recognition method for the detection of stator windings short circuits based on measures of mutual information between the phase current signals. In order to validate the proposed patterns, feature vectors obtained from normal and faulty motors are applied to two topologies of artificial neural networks. The classification results presented accuracies over 93% even when the motors were subject to several conditions of load torque and power supply voltage unbalance.

Depsipeptides are a group of biologically active peptides that have at least one of the amide bonds replaced by an ester bond. These peptides sometimes present additional chemical modifications, including unusual amino acid residues in their structures. Depsipeptides are known to exhibit a large array of bioactivities, such as anticancer, antiproliferative, antimicrobial, antiviral and antiplasmodial properties. They are commonly found in marine organisms: bacteria, tunicates, mollusks, sponges, and others. Herein, we summarize the latest insights about marine depsipeptides, their mechanisms of action and potential as therapeutic agents.

This review discusses the importance and properties of antimicrobial peptides from frogs and their synthetic analogues as potential therapeutic alternatives in fighting not only bacterial infections, but also protozoans involved with the major neglected diseases, which afflict human populations (e.g., Chagas disease, African sleeping sickness, Leishmaniasis and malaria). Here, we emphasize their multifunctional properties such as promising broad-spectrum drugs that target protozoan parasites too.

A comprehensive study of intelligent tools used to classify broken rotor bars in induction motors, which operate with three different types of frequency inverters, is presented. The diagnosis of defective rotor bars is a critical issue for the predictive maintenance of induction motors. A proper classification of these defects in their early stages of evolution is necessary for preventing major machine failures and production downtime. The proposed approach is performed by analysing the amplitude of the stator current signal in the time domain, using a dynamic acquisition rate based on machine frequency supply. To assess classification accuracy under the various severity levels of the faults, the performance of four different learning machine techniques is investigated: (i) fuzzy ARTMAP network; (ii) support vector machine (sequential minimal optimisation); (iii) k-nearest neighbour; and (iv) multilayer perceptron network. Results obtained from 1274 experimental tests are presented in order to validate the study, which considers a wide range of load conditions and operating frequencies. Experimental results presented in this study validate the robustness and efficacy of the proposed approach.

Nanoscience and Nanotechnology enable innovations for Medicine and for Biomedical Science. Among the current nanoparticles, maghemite associated with rhodium citrate (Magh-RhCit) has been shown to be promising, because it reduces side effects of drugs while maintaining cytotoxicity for tumor cells. However, NPs in contact with biological fluids are immediately coated by proteins (protein corona) that are unique to each nanomaterial. In this study, the adsorption of the most abundant binding proteins was studied in vitro using a three-step analysis: (1) characterization of the magnetic fluid (Magh-RhCit) before and after incubation with serum; (2) identification and physical and biochemical analysis of protein corona; and (3) the cellular internalization of these nanoparticles in human macrophages. Magh-RhCit was initially obtained in the magnetite phase (Fe3O4) via alkaline coprecipitation of Fe2+ and Fe3+ ions and subsequently oxidized to maghemite by the bubbling of oxygen gas in the suspension. Later, rhodium citrate was associated with nanoparticles. Dynamic Light Scattering data were used for characterization of the hydrodynamic diameter and zeta potential. The morphological characterization and measurement of the particles were obtained from Scanning and Transmission Electron Microscopy, and the X-ray Diffraction technique. The identification of the proteins was performed by Liquid Chromatograph coupled to Mass Spectrometry. Human blood serum altered the characteristics of the nanoparticle, making it less polydisperse, larger and with less negative zeta potential, indicating the formation of the protein corona. Forty-nine proteins (which mostly promote opsonization, phagocytosis and endocytosis in cells of the immune system) were identified and characterized: albumin, IgGs, apoliproteins, serpins, complement (C5), kinases, haptoglobin, glycoproteins and transferrin. Nanoparticle characterization and mass spectrometric data of the digested protein corona suggest improved biocompatibility. Moreover, results regarding nanoparticles’ interaction with macrophages suggest that the corona may have profoun implications for in vitro and in vivo extrapolations and will require some consideration in the future.

Clonostachys byssicola is a filamentous fungus with biotechnological potential for holocellulases production. The present work describes a qualitative and quantitative proteome analysis of C. byssicola secretome cultivated in media supplemented with commercial (Avicel, carboxymethylcellulose or oat spelt xylan) or agricultural (corn cob, soybean hulls, banana stem or sugarcane bagasse) carbon sources. Proteomic data revealed that the fungus produces a common core of lignocellulolytic enzymes when cultivated in different carbon sources. Most of the Carbohydrate-Active Enzymes (CAZymes) identified in the secretomes belong to the families AA9, GH 5, 6, 7, 10 and 43, CE 1, 10 and 12, and PL 1 and 3. Secretomes with the highest specific activity, obtained in corncob, banana stem and sugarcane bagasse, were applied for saccharification of partially delignified corn cob or sugarcane bagasse, and promoted extensive hemicellulose solubilization, with the release of mainly xylobiose. Additionally, the secretomes displayed remarkable differences in the abundance of identified proteins. Considered together, both qualitative and quantitative proteomic characterization presented in this study suggest that C. byssicola produces a similar core of carbohydrases, apparently suitable for the action over a wide range of different lignocellulosic substrates. These findings demonstrate the potential of C. byssicola as a novel source of holocellulases.

RESUMO A avaliação proteica do humor aquoso (HA) pode ser utilizada como método diagnóstico nas uveítes. Entretanto, estudos sobre as proteínas nesse fluido, em equinos hígidos, são escassos e apresentam variações conforme a metodologia empregada. Dessa forma, objetivou-se realizar a análise proteica e citológica do HA nessa espécie, bem como verificar sua correlação com as proteínas plasmáticas. Foram avaliados 13 equinos adultos (26 olhos), sem raça definida, machos ou fêmeas. Mediante aqueocentese, foi coletado 0,5 mL de humor aquoso de cada olho. Cada amostra foi encaminhada para quantificação proteica pelo método de Bradford modificado e pela eletroforese em gel de poliacrilamida - dodecil sulfato de sódio (SDS-PAGE), bem como para avaliação citológica. Por meio de venopunção, coletou-se sangue para determinação da concentração de proteínas séricas. Treze olhos (50% das amostras) apresentaram valor proteico médio de 40,3 mg/dL±6,45 e a eletroforese demonstrou presença de proteínas de massas mais elevadas que 43 KDa. Houve ausência de células em 96,15% das amostras (25 olhos). Equinos hígidos apresentaram baixa concentração de proteínas no HA. Já a correlação entre proteína no humor aquoso/proteína plasmática total foi de 0,56%. ABSTRACT Evaluation of equine aqueous humor (AH) proteins can help the diagnosis of uveitis. However, studies on proteins in this fluid in healthy horses are scarce and present variations according to the methodology employed. This study aimed to perform protein analysis and cytology of equine aqueous humor of healthy horses and verify its correlation with plasmatic proteins. Thirteen adult horses (26 eyes), mixed breed, male or female were evaluated. A volume of 0.5 mL of aqueous humor was collected through aqueocentesis from both eyes. The samples were submitted to protein quantification by modified Bradford method and to sodium dodecyl sulphate - polyacrylamide gel electrophoresis (SDS-PAGE), and to cytological evaluation. Blood was collected for determination of plasmatic protein concentration. Thirteen eyes (50% of the samples) had values larger than zero by the Bradford method, with an average of 40.3 mg/dl±6.45. Electrophoresis showed presence of higher masses of proteins (43 KDa). There were no cells in 96.15% of the samples (25 eyes). Healthy equines presented low protein concentration in the HA. The ratio between protein concentration in the aqueous humor / total plasma protein of 0.56%.

Helices are one of the most common secondary structures found in peptides and proteins. The wheel and net projections have been proposed to represent in two dimensions the tridimensional helical structures and facilitate the observation of their properties, especially in terms of residues polarity and intramolecular bonding. Nevertheless, there are few software options to create these projections. We have developed a web-based application that has several futures to create, customize and export these projections and is freely available athttp://lbqp.unb.br/NetWheels.

This paper proposes a method for classification of broken rotor bars of three-phase induction motors driven with three different models of frequency inverters by using the amplitude of the stator current signal in the time domain. To assess the effectiveness of the method under evaluation in this work, classification accuracy of four different learning machine techniques were compared: Fuzzy ARTMAP network; Support Vector Machine; k-Nearest Neighbour and Multilayer Perceptron Network. Moreover, this paper presents the comparison of an analogue and a digital filter to the line current signal provided by the motor. Results obtained from 2293 experimental tests are presented to validate this comparative study, considering a wide range of speed and load conditions.

Aluminium in acid soils is a hindrance to crop growth. Nonetheless, Brazilian Cerrado possesses many species such asQualea grandiflora, which are adapted to acid soils with large amounts of Al and accumulate this metal in its tissues and organs. Nonetheless, the mechanisms involved in these processes are poorly understood, mainly at molecular level. Thus, a root proteomic analysis was accomplished to identify Al-responsive proteins inQ. grandifloraplants. Concomitantly, a root growth analysis of plants grown with and without Al supplementation was conducted to determine the effects of Al on the whole plant. Subsequently, proteins from both treatments were identified and quantified by LC-MS/MS in a label-free fashion. From the 2,520 identified proteins, 410 were differentially abundant between the two treatments, which were associated with carbohydrate metabolism, redox activity, stress response and catabolism of organic compounds. Furthermore, Al was crucial for the growth and development ofQ. grandiflora. In fact, this species may have an Al-dependent metabolism. Moreover, it was possible to correlate plant growth to Al-upregulated proteins that were directly involved in cell wall synthesis, oxidative phosphorylation, genetic information processing, and amino acid metabolism. Additionally, this work provides an extensive dataset of Al-regulated protein inQ. grandiflora, which will be crucial to understanding the functions of Al in this species.

Oil palm is an oleaginous plant of relevant economic importance since its fruits are rich in vegetable oil. These plants have a single apical meristem and the main method for vegetative propagation is somatic embryogenesis. The aim of this study was to identify differentially abundant proteins from oil palm genotypes contrasting in the capacity of embryogenic competence acquisition, using shotgun proteomics. Oil palm leaves were subjected to callus induction and the material was collected in biological triplicates at 14 and 90 days of callus induction. LC-MS/MS analysis was performed and revealed a total of 4695 proteins. Responsive and non-responsive genotypes were compared at 14 and 90 days of callus induction and 221 differentially abundant proteins were obtained. The data analysis revealed several proteins mainly related to energy metabolism, stress response and regulation of cell cycle, further analyzed by qRT-PCR, which seem important for embryogenic development. We suggest some of these proteins as key factors for the success of callus formation in oil palm including antioxidant and cell division proteins as well as proteins involved in the ubiquitination pathway. These proteins may also be potential biomarkers for the acquisition of embryogenic competence. Significance Antioxidant and cell division proteins as well as proteins involved in the ubiquitination pathway are key factors for the success of callus formation in oil palm. The proteins identified in this study may be potential biomarkers for embryogenic competence acquisition.

Fever is a brain-mediated increase in body temperature mainly during inflammatory or infectious challenges. Although there is considerable data regarding the inflammation pathways involved in fever, metabolic alterations necessary to orchestrate the complex inflammatory response are not totally understood. We performed proteomic analysis of rat hypothalamus using label-free LC-MS/MS in a model of fever induced by lipopolysaccharide (LPS) or prostaglandin EThe evolutionary persistence of fever, despite the elevated cost for maintenance of this response, suggests that elevation in core temperature may represent an interesting strategy for survival. Fever response is achieved through the integrated behavioral, physiological, immunological and biochemical processes that determine the balance between heat generation and elimination. The development of such complex response arouses interest in studying how the cell metabolism responds or even contributes to promote fever. Our results offered comprehensive information about fever responses, including metabolic and inflammatory pathways, providing new insights into candidate proteins potentially involved in inflammatory signaling and metabolic changes in the hypothalamus during fever induced by systemic LPS and central PGE
