Abstract

The interest towards the so-called ancoient wheat, einkorn, emmer, oriental wheat or khorasan and spelt is constantly increased in the last decades. Oriental wheat (Triticum turanicum Jakubz) is a neglected and underutilised tetraploid species which survived over the time in some marginal areas of Near East and North Africa. Scarce information is available in literature on the gliadin variation within the oriental wheat germplasm. In this study the gliadin profile of 23 oriental wheat accessions with different origin, were analysed by capillary electrophoresis.

Abstract

Vista la scarsa o nulla utilizzazione dei frumenti minori Kamut (Triticum turanicum Jacubz.) e spelta (Triticum spelta L.) nella produzione di pane, in questo studio ci si è proposti di valutarne l'attitudine panificatoria. Gli sfarinati di questi due cereali, sia raffinati, sia integrali, sono stati impiegati in prove di panificazione eseguite con varie modalità comprendenti metodo diretto ed indiretto, valutando quindi direttamente la qualità del prodotto finale. I risultati ottenuti hanno dimostrato, pur se con forti variazioni nel contenuto di glutine e di proteine a seconda dell'annata agraria, la panificabilità sia del Kamut, sia dello spelta grazie alla non eccessiva tenacità del glutine.

Abstract

It has been happened a significant increase of critically severe pneumonia cases in northern Italy by the end of February 2020 with consequent overcrowding of intensive care units (ICU). Due to the COVID-19 fast outbreak, a strict quarantine was imposed in the areas with the higher number of affected cases. The WHO declared COVID-19 a pandemic, with over 50 countries on the 10th of March. By the 10th of March Italy was the second most affected country after China, with more than nine thousand confirmed cases and more than 460 deaths, becoming the most affected in April.

127. Salvaguardia, caratterizzazione e valorizzazione dell'antico agro-ecotipo pugliese 'Lenticchia di Altamura'

Abstract

Since the '70s, the Institute of Plant Genetics (IGV) has been working on the safeguard of autochthonous Italian lentil agro-ecotypes in collaboration with other Italian and international institutions. This activity has led to the establishment of a rich germplasm collection of Italian lentils. Some years ago, a number of studies has been started on the characterisation of specific agro-ecotypes, particularly interesting because élite germplasm or because at risk of genetic erosion or extinction. Recently, a project on the safeguard, characterisation and reintroduction of the disappeared agroecotype from Apulia region 'Lenticchia di Altamura' (Lens culinaris Medik. ssp. macrosperma Barul.) was funded by some entrepreneurs. This ecotype is characterised by large, flat and pale green seeds. The only 10 lentil accessions collected in Altamura countryside in the past were retrieved from the germplasm bank present at the IGV. From the year 2002/2003, these accessions were evaluated for morpho-agronomic traits in two localities for two years. The highest yields were observed for populations ALT9 (5,1 q/ha) and ALT8 (4,5 q/ha), similar to the modern commercial cultivars Laird, Eston and Regular, used as controls. A low phenotypic intra-population variation was observed. The other most important agronomic descriptors detected were: time to flowering, plant height and height of lowest pod. Moreover, in the year 2003/2004, an agronomic trial with two sowing time, autumn and spring, was performed. For the spring sowing, a yield decrease of 45% was observed, whereas the harvesting time was no significantly different. Cluster analysis derived from morpho-agronomic data grouped all ecotypes but ALT3 into three clusters: (a) ALT1, ALT2 e ALT5, (b) ALT4, ALT7 e ALT9 e (c) ALT6, ALT8 e ALT10. In order to determine the level of genetic variation within and between populations, AFLP analysis was performed on 15 individuals per each of the six populations which showed bigger seeds; cultivars Laird and Eston were used as controls. Three primer combinations were tested (E-ACT+M-CAT, E-AAG+M-CTC, E-AGC+M-CAT), producing a total of 204 fragments; in the Altamura populations 197 fragments were present, 84.31% of which were polymorphic. The presence or absence of each fragment was detected and a binary matrix was constructed. These data were used to calculate some indices of genetic diversity: number of observed alleles, Nei's index of gene diversity, Nei's total genetic diversity, genetic diversity within and among populations. Results showed that, at the molecular level, most of the variation was present within populations, indicating that the six Altamura populations analysed are not well differentiated among them and could represent sub-populations recently derived from a single population. Another possible interpretation could be that there has been a certain level of geneflow among the sub-populations, due to a continuous seed exchange among farmers.

I primi due hanno ormai una consolidata posizione di nicchia, mentre poche e contraddittorie sono le informazioni sui caratteri agronomici, nutrizinali e tecnologici del Kamut. Scopo di questo studio è stato valutare la possibilità di creare per il Kamut una filiera di panificaizone nel sud Italia, dove vi è una lunga tradizione di panificaizone col grano duro.

Abstract

Twenty-two lines of emmer (T. dicoccon Schrank) and ten of spelt (T. spelta L.) were analysed, using capillary electrophoresis, for their gliadins. These proteins were separated on a 30 cm long (22 cm to detector, 50?m ID) uncoated fused-silica capillary using the isoelectric buffer 40mM aspartic acid, 4M urea, 0.5% (w/v) HEC and 20% (v/v) acetonitrile. Samples were run for 20 min at 22kV and 42°C. By using these conditions, gliadins were separated into 21-30 components (peaks and shoulders). The major peaks eluted between 4.5 and 8.5 min. Electrophoregrams of tested lines showed qualitative and quantitative differences, including number of peaks, presence or absence of some major peaks and areas of peaks. Lines belonging to the same species can be discriminated mainly on the bases of beta- and omega-gliadin patterns. The gamma- and omega-gliadins seems to be more useful in the differentiation of emmer from spelt. The comparison of electrophoregrams relative to hulled and unhulled species evidenced the high similarity between species with the same genome composition (durum wheat/emmer and common wheat/spelt).

Abstract

Emmer and spelt are two ancient cultivated species of hulled wheat that have been replaced with unhulled wheat. However, in Italy and elsewhere, they are becoming promising niche crops. Their success is due to their sensorial charactersistics and remarkable potential as alternative crops for marginal areas. Twenty-nine accessions of emmer and twenty-four accessions of spelt were grown in 6 different environments of two regions in southern Italy (Molise and Basilicata) over a period of 6 years. This was done to choose the best accessions in terms of yield and stability.

Abstract

PurposeTo determine the differences in the presentation of ophthalmic giant cell arteritis between African-Americans and Caucasians.MethodsThis was a multicenter retrospective case series comparing African-American patients with ophthalmic GCA to a previously published Caucasian cohort. Neuro-ophthalmic centers across the United States were contacted to provide data on African-American patients with biopsy-proven ophthalmic giant cell arteritis. The differences between African-American and Caucasian patients with respect to multiple variables, including age, sex, systemic and ophthalmic signs and symptoms, ocular ischemic lesions, and laboratory results were studied.ResultsThe Caucasian cohort was slightly older (mean=76.1 years) than the African-American cohort (mean=72.6 years, P=0.03), and there was no difference in sex distribution between the two cohorts. Headache, neck pain, and anemia were more frequent, while jaw claudication was less frequent in African-Americans (P<0.01, <0.001, 0.02, and 0.03 respectively). Acute vision loss was the most common presentation of giant cell arteritis in both groups, though it was less common in African-Americans (78 vs 98% of Caucasians, P<0.001). Eye pain was more common in African-Americans (28 vs 8% of Caucasians, P<0.01).ConclusionsThe presenting features of ophthalmic giant cell arteritis in African-Americans and Caucasians are not markedly different, although a few significant differences exist, including higher rates of headache, neck pain, anemia, and eye pain, and lower rates of jaw claudication and acute vision loss in African-Americans. Persons presenting with suspicious signs and symptoms should undergo evaluation for giant cell arteritis regardless of race.
