Purpose: A breast dedicated positron emission tomography (PET) scanner has been developed based on monolithic LYSO crystals coupled to position sensitive photomultiplier tubes (PSPMTs). In this study, we describe the design of the PET system and report on its performance evaluation. Methods: MAMMI is a breast PET scanner based on monolithic LYSO crystals. It consists of 12 compact modules with a transaxial field of view (FOV) of 170 mm in diameter and 40 mm axial FOV that translates to cover up to 170 mm. The patient lies down in a prone position that facilitates maximum breast elongation. Quantitative performance analysis of the calculated method for the attenuation correction specifically developed for MAMMI, and based on PET image segmentation, has also been conducted in this evaluation. In order to fully determine the MAMMI prototype's performance, we have adapted the measurements suggested for National Electrical Manufacturers Association (NEMA) NU 2-2007 and NU 4-2008 protocol tests, as they are defined for whole-body and small animal PET scanners, respectively. Results: Spatial resolutions of 1.6, 1.8, and 1.9 mm were measured in the axial, radial, and tangential directions, respectively. A scatter fraction of 20.8% was obtained and the maximum NEC was determined to be 25 kcps at 44 MBq. The average sensitivity of the system was observed to be 1% for an energy window of (250 keV-750 keV) and a maximum absolute sensitivity of 1.8% was measured at the FOV center. Conclusions: The overall performance of the MAMMI reported on this evaluation quantifies its ability to produce high quality PET images. Spatial resolution values below 3 mm were measured in most of the FOV. Only the radial component of spatial resolution exceeds the 3 mm at radial positions larger than 60 mm. This study emphasizes the need for standardized testing methodologies for dedicated breast PET systems similar to NEMA standards for whole-body and small animal PET scanners. [ABSTRACT FROM AUTHOR]

Abstract: Aim of the study: This study aimed to evaluate the sensitivity and specificity of a new system (Cica-Beta-Test, Kanto Chemical, Japan) for rapid detection of AmpC-derepressed, extended-spectrum β-lactamases (ESβL) and metallo-β-lactamases (MβL). Methods: Two hundred Multi-Drug Resistant (MDR)-Gram-negative bacilli were studied: 170 Enterobacteriaceae and 30 Gram-negative non-fermentative bacteria. One hundred and eighteen strains produced an ESβL, seven MβL and 75 derepressed cephalosporinases. One drop of substrate was dispensed onto the filter pad of the Cica-Beta-Test strip. The bacterial colonies were spread on the filter pad of strip. The reading was performed after 2 to 15min: turning chromogenic indicated the positive test. Three tests were used: Cica-Beta I for detection of MDR bacteria; Cica-Beta MβL for detection of MβL-producing bacteria and Cica-Beta CVA, which distinguish ESβL and AmpC-derepressed producers. Results were compared with molecular assays. Results: Cica-Beta-Test I has detected 194 MDR (sensitivity 97%), Cica-Beta-Test MβL has shown the presence of six MβL tested (sensitivity 85.7%). Five strains were non-MβL false positive (specificity 97.3%). Cica-Beta-Test CVA allowed the differentiation of ESβL-producing strains (109/115) and AmpC-derepressed strains (56/67) (sensitivity 94.8%, specificity 83.8%). Conclusions: Because of their epidemic nature, the MDR strains are screened and require strict hygienic measures patients. The simultaneous use of three strips can quickly determine the presence of MDR including ESβL and MβL. Rapid screening of MDR avoids transmission and limits the use of broad-spectrum antibiotics. Objective: To survey the opinion of critical care providers in Argentina about abortion.Methods: An anonymous questionnaire was distributed to critical care providers attending the 20th National Critical Care Conference in Argentina.Results: 149 of 1800 attendees completed the questionnaire, 69 (46.3%) of whom were members of the Argentine Society of Critical Care (ASCC). 122 (81.9%) supported abortion decriminalization in situations excluded from the current law; 142 (95.3%) in cases of congenital defects; 133 (89.3%) in cases of rape; 115 (77.2%) when women's mental health is at risk; 71 (47.7%) when pregnancy is unintended; and 61 (40.9%) for economic reasons. 126 (84.6%) supported abortion in public and private institutions, and 121 (81.2%) before 12 weeks of pregnancy. Variables independently associated with abortion support among female versus male attendees were abortion to preserve women's mental health (OR 4.47; 95% CI, 1.61-12.42; P=0.004) and abortion before 12 weeks of pregnancy (OR 3.93; 95% CI, 1.29-11.94; P=0.015). Abortion at request was independently associated with ASCC membership (OR 2.63; 95% CI, 1.07-6.45; P=0.034).Conclusion: Critical care providers would support abortion in situations excluded from the current abortion law and before 12 weeks of pregnancy, in both public and private hospitals. Abstract: Submicron- and, in a low quantity nano-sized carbide particles were successfully prepared through a dynamic process of energetic material detonation. For that, a commercial micrometric SiC powder, homogeneously coated by a polymer layer, was dispersed in an energetic 2,4,6-trinitrotoluene (TNT) matrix and then detonated. After detonation, the resulting carbide material was characterized by several techniques such as X-ray diffraction, infrared spectroscopy, electron microscopies, and nitrogen adsorption. The results showed a decrease in the initial micrometric SiC size with particle sizes reaching, in some cases, less than 20nm. The specific surface area was also considerably increased since an approximate value of 7.8m2 g−1 was obtained for a detonated SiC sample as opposed to 0.12m2 g−1 for the initial micrometric SiC powder. Background: This phase Ib trial assessed safety, tolerability, and maximum tolerated dose (MTD) of figitumumab (CP-751,871), a fully human monoclonal antibody targeting the insulin-like growth factor type 1 receptor (IGF-IR), in combination with docetaxel.Methods: Patients with advanced solid tumours were treated with escalating dose levels of figitumumab plus 75 mg m(-2) docetaxel every 21 days. Safety, efficacy, pharmacokinetics (PKs), and biomarker responses were evaluated.Results: In 46 patients, no dose-limiting toxicities were attributable to the treatment combination. Grade 3 and 4 toxicities included neutropaenia (n=28), febrile neutropaenia (n=11), fatigue (n=10), leukopaenia (n=7), diarrhoea (n=5), hyperglycaemia, lymphopaenia, cellulitis, DVT, and pain (all n=1). The MTD was not reached. Four partial responses were observed; 12 patients had disease stabilisation of > or =6 months. Pharmacokinetic and biomarker analyses showed a dose-dependent increase in plasma exposure, and complete sIGF-IR downregulation at doses of >or =3 mg kg(-1). Pharmacokinetics of docetaxel in combination was similar to when given alone. Out of 18 castration-resistant prostate cancer patients, 10 (56%) had > or =5 circulating tumour cells (CTCs) per 7.5 ml of blood at baseline: 6 out of 10 (60%) had a decline from > or =5 to <5 CTCs and 9 out of 10 (90%) had a > or =30% decline in CTCs after therapy.Conclusions: Figitumumab and docetaxel in combination are well tolerated. Background: This phase I study assessed the maximum tolerated dose, dose-limiting toxicity (DLT) and pharmacokinetics of belinostat with carboplatin and paclitaxel and the anti-tumour activity of the combination in solid tumours.Methods: Cohorts of three to six patients were treated with escalating doses of belinostat administered intravenously once daily, days 1-5 q21 days; on day 3, carboplatin (area under the curve (AUC) 5) and/or paclitaxel (175 mg m(-2)) were administered 2-3 h after the end of the belinostat infusion.Results: In all 23 patients received 600-1000 mg m(-2) per day of belinostat with carboplatin and/or paclitaxel. No DLT was observed. The maximal administered dose of belinostat was 1000 mg m(-2) per day for days 1-5, with paclitaxel (175 mg m(-2)) and carboplatin AUC 5 administered on day 3. Grade III/IV adverse events were (n; %): leucopenia (5; 22%), neutropenia (7; 30%), thrombocytopenia (3; 13%) anaemia (1; 4%), peripheral sensory neuropathy (2; 9%), fatigue (1; 4%), vomiting (1; 4%) and myalgia (1; 4%). The pharmacokinetics of belinostat, paclitaxel and carboplatin were unaltered by the concurrent administration. There were two partial responses (one rectal cancer and one pancreatic cancer). A third patient (mixed mullerian tumour of ovarian origin) showed a complete CA-125 response. In addition, six patients showed a stable disease lasting > or =6 months.Conclusion: The combination was well tolerated, with no evidence of pharmacokinetic interaction. Abstract: Carbon/Carbon disks worn by a reduced scale braking test were characterised by an experimental set-up allowing the analysis of the whole disk. In this way, the structure and the surface chemistry of the wear debris formed during the braking and trapped at the sliding interface were determined. The evolution of the surface chemistry and the structure during rubbing at different braking energies were studied by temperature programmed desorption with mass spectrometry analysis and by measuring the active surface area. The analysis of the composites before and after rubbing highlights the formation of highly disordered and oxygen-rich carbon wear debris at the sliding surface. The surface chemistry of the carbon dust trapped on the disk surface was found to be very similar to that of the wear debris ejected during braking. From this similarity a rough estimation of the amounts of carbon dust lying on the worn composites surface was made. [Copyright &y& Elsevier]

Objective: The purpose of this study was to assess the virulence potential of Staphylococcus aureus strains isolated from diabetic foot ulcers and to discriminate noninfected from infected ulcers.Research Design and Methods: Diabetic patients hospitalized in a diabetic foot department with a foot ulcer were prospectively enrolled if they had been free of antibiotic treatment over the previous 6 months. At admission, ulcers were classified as infected or noninfected on the basis of clinical examination, according to the International Working Group on the Diabetic Foot system. Only patients carrying S. aureus as the sole pathogen were included. In individuals with a grade 1 ulcer, a second bacterial specimen was obtained 1 month later. Using virulence genotyping markers, clonality tools, and an in vivo Caenorhabditis elegans model, we correlated the virulence of 132 S. aureus strains with grade, time of collection, and ulcer outcome.Results: Among virulence genes, the most relevant combination derived from the logistic regression was the association of cap8, sea, sei, lukE, and hlgv (area under the curve 0.958). These markers were useful to distinguish noninfected (grade 1) from infected (grades 2-4) ulcers and to predict wound status at the follow-up. With use of the nematode model, S. aureus strains isolated from grade 1 ulcers were found to be significantly less virulent than strains from ulcers at or above grade 2 (P < 0.001).Conclusions: This study highlights the coexistence of two S. aureus populations on diabetic foot ulcers. A combination of five genes that may help distinguish colonized grade 1 from infected grade >or=2 wounds, predict ulcer outcome, and contribute to more appropriate use of antibiotics was discovered. Abstract: In recent years, conducting polymers combined with metallic nanoparticles have been paid more attention due to their potential applications in microelectronics, microsystems, optical sensors and photoelectronic chemistry. The work presented in this paper describes the preparation and characterization of a nanocomposite composed by a thin polypyrrole (PPy) film covered with an assembly of magnetic nanoparticles (NPs). The magnetic particles were immobilized on PPy films under appropriate magnetic field in order to control their organization on the PPy film and finally to improve the sensitivity of the system in potential sensing applications. The electrical properties and morphology of the resulting PPy film and the PPy film/NPs composite were characterized with cyclic voltammetry, impedance spectroscopy (IS), scanning electron microscopy (SEM), atomic force microscopy (AFM) and infra-red spectroscopy (IR). By using streptavidin labeled magnetic particles it was possible to functionalize the NPs assembly with biotin-Fab fragment K47 antibody. The designed biosensor had been successfully applied in rapid, simple, and accurate measurements of atrazine concentrations, with a significantly low detection limit of 5ng/ml. Functional barriers are multilayer structures deemed to prevent migration of some chemicals released by food-contact materials into food. In the area of plastics packaging, different migration behaviours of mono- and multilayer structures are assessed in terms of lag time and of their influence of the solubility of the migrants in food simulants. Whereas barriers to oxygen or to aromas must prevent the diffusion of these compounds under conditions of use, a functional barrier must also be efficient under processing conditions, to prevent diffusion of substances when the polymer layers are in contact at high (processing) temperatures. Diffusion in melted polymers at high temperatures is much slower for glassy polymers, than in polymers that are rubbery at ambient temperature. To evaluate the behaviour of functional barriers under conditions of use, a set of reference diffusion coefficients in the 40–60°C range were determined for 14 polymers. Conditions for accelerated migration tests are proposed based on worst-case activation energy in the 40–60°C range. For simulation of migration, numerical models are available. The rules derived from the models can be used both by industry (to optimize a material in terms of migration) or by risk assessors. Abstract: The possible protective effects of glutathione (GSH), cysteine (CYS) and methionine (MET) on the Methylmercury (MeHg)-induced dopamine (DA) release from rat striatum were investigated using in vivo microdialysis coupled to HPLC with electrochemical detection. Intrastriatal infusion of MeHg 400 μM increased extracellular DA levels to 1941 ± 199% in terms of basal levels. Infusion of MeHg 400 μM in GSH 400 μM pretreated animals, only increased striatal DA levels to 465 ± 104%, in terms of basal levels, this increase being 76% lower than induced by MeHg alone. Conversely, the infusion of MeHg 400 μM after infusion of GSH 400 μM increased DA levels to 1019 ± 96% in terms of basal levels, this increase being 47.5% lower than that observed in MeHg non-pretreated animals. The infusion of MeHg 400 μM in CYS 400 μM -pretreated animals, increased striatal DA levels to 740 ± 149%, in terms of basal levels, this increase being 62% lower than that induced by MeHg in non-pretreated animals. The infusion of MeHg 400 μM in MET 400 μM pretreated animals increased striatal DA levels to 2011 ± 230% in terms of basal, an increase that was not significantly different from that produced by MeHg 400 μM alone. In summary, the administration of compounds containing free -SH groups prevented the MeHg-induced DA release from rat striatum, probably due to the binding of MeHg to -SH groups. This would result in a lower metal availability to interact with –SH membrane proteins groups, which would decrease MeHg ability to interact with DA transporter. [Copyright &y& Elsevier]

We present here a pollen record from the northern Indian Ocean that goes back to 200 ka, the boundary between marine isotopic stages (MIS) 7 and 6. Pollen, oxygen-isotopic composition and organic carbon have been examined for two sediment cores from the eastern Arabian Sea (15°02′N and 71°41′E, 13°16′N and 71°00′E), to reconstruct the long-term palaeoclimate and palaeovegetation of the Indian subcontinent. Oxygen-isotope data suggest that glacial periods (MIS 2, 4 and 6) are characterised by low precipitation because of a weak southwest monsoon and a strong northeast monsoon. In contrast, interglacial periods (MIS 1, 3 and 5) are marked by high fresh water input resulting from a strong southwest monsoon. During the last glacial–interglacial cycle, sea-surface temperature and surface salinity changed as a result of variations in the evaporation–precipitation (E–P) balance. Throughout the core, the dominant pollen types are Poaceae and Chenopodiaceae and/or Amaranthaceae (>50%). Their dominance during glacial periods (MIS 2, 4 and 6) suggests that the climate was cold and dry. This dominance is also suggestive of salinity-tolerant vegetation colonizing large areas near seashore due to lower sea level or high E–P conditions. During interglacial periods (MIS 1, 3 and 5) when there was high precipitation, the arid taxa were sparse, whereas Poaceae and Piperaceae were abundant. Hence, Chenopodiaceae and/or Amaranthaceae and Artemisia are suggestive of cold and arid/semi-arid climate, and Poaceae and Piperaceae of warm and wet conditions. An ordered mesoporous silica matrix (SBA-15) was infiltrated with carbon using either a liquid or a gas route with various carbon precursors (sucrose, petroleum pitch and propylene). The resulting SiO2/C materials were then chemically treated to dissolve selectively the silica matrix and lead to ordered mesoporous carbon replicas. The influence of the precursors on the physico-chemical properties of the SiO2/C composites and the carbon replicas were studied by X-ray diffraction, nitrogen adsorption measurements, scanning and transmission electron microscopies. It was shown that the content, the route for introducing the carbon into the host matrix, the microporosity and the mesopores order of the carbon replicas are determined by the choice of the precursors. A facile method for the preparation of gold nanoparticles (AuNPs) with various morphologies, including triangles, cubes and nanospheres into a urethane methacrylate polymer matrix, has been developed. The hybrid nanocomposites have been obtained by photoreduction of a gold precursor in the presence of a 30 wt% urethane methacrylate containing quaternary ammonium structure and 70 wt% functional monomer, carboxyl oligo-urethane dimethacrylate. The final materials contain gold nanoparticles with a morphology dependant on the reaction conditions. The influence of photonic and chemical parameters on the shape of the nanoparticles has been examined. Specific interactions between the macromolecular network and the nascent particles play an important role insofar as they control the access of metal atoms to the different crystalline planes of the growing nanoparticles, which is necessary to obtain anisotropic objects. A common mechanism for the formation of the various types of AuNPs is proposed. Coupling the intrinsic characteristics of metal nanoparticles with those of the copolymer substrate could result in materials with innovative properties. [ABSTRACT FROM AUTHOR]
