Abstract

Cardif, also known as IPS-1, VISA, and MAVS, is an intracellular adaptor protein that functions downstream of the retinoic acid-inducible gene I family of pattern recognition receptors. Cardif is required for the production of type I IFNs and other inflammatory cytokines after retinoic acid-inducible gene I-like receptors recognize intracellular antigenic RNA. Studies have recently shown that Cardif may have other roles in the immune system in addition to its role in viral immunity. In this study, we find that the absence of Cardif alters normal NK cell development and maturation. Cardif(-/-) mice have a 35% loss of mature CD27(-)CD11b(+) NK cells in the periphery. In addition, Cardif(-/-) NK cells have altered surface marker expression, lower cytotoxicity, decreased intracellular STAT1 levels, increased apoptosis, and decreased proliferation compared with wild-type NK cells. Mixed chimeric mice revealed that the defective maturation and increased apoptotic rate of peripheral Cardif(-/-) NK cells is cell intrinsic. However, Cardif(-/-) mice showed enhanced control of mouse CMV (a DNA β-herpesvirus) by NK cells, commensurate with increased activation and IFN-γ production by these immature NK cell subsets. These results indicate that the skewed differentiation and altered STAT expression of Cardif(-/-) NK cells can result in their hyperresponsiveness in some settings and support recent findings that Cardif-dependent signaling can regulate aspects of immune cell development and/or function distinct from its well-characterized role in mediating cell-intrinsic defense to RNA viruses.

115. Anti-IL8/AuNPs-rGO/ITO as an Immunosensing Platform for Noninvasive Electrochemical Detection of Oral Cancer

Abstract

An efficient electrochemical transducer matrix for biosensing devices requires specific characteristics, such as fast electron transfer, stability, high surface area, biocompatibility, and presence of specific functional groups, to facilitate biomolecule attachment. We demonstrate the fabrication of an electrochemical immunosensor based on a highly stable gold nanoparticles-reduced graphene oxide (AuNPs-rGO) composite material as a transducer matrix for label-free and noninvasive detection of salivary oral cancer biomarker interleukin-8 (IL8). The synergy between rGO and AuNPs allowed the immunosensor to exhibit fast response and high sensitivity due to the improved electron transfer behavior of the composite. The immunosensor shows very fast detection (9 min) of IL8 and high sensitivity with an experimental linear dynamic range of 500 fg mL–1to 4 ng mL–1and a detection limit of 72.73 ± 0.18 pg mL–1. The fabricated immunosensor exhibits excellent specificity toward the detection of IL8 in human saliva samples. Furthermore, the reusability and stability up to 3 months of the immunosensor demonstrates the commercial potential of this nanoplatform for the detection of other biomarkers of clinical relevance.

Leishmania organisms are susceptible to microbicidal responses generated in response to phagocytosis. Assuming that both phagocyte and parasite populations are heterogeneous, it is advantageous to examine the response of individual cells phagocytosing living parasites. Because Leishmania spp. lose virulence during the raising of transfectants, we developed a method to label live Leishmania chagasi short-term with fluorescent dyes. Up to six parasite divisions were detected by flow cytometry after labeling with carboxyfluorescein diacetate succinimidyl ester (CFSE), dioctadecyl-tetramethylindo carbocyanine perchlorate, or chloromethyl tetramethylrhodamine. Labeled parasites entered mononuclear phagocytes as determined by confocal and time-lapse microscopy. Dihydroethidium (DHE) was used to detect macrophage-derived oxidants generated during phagocytosis. Presumably Leishmania organisms are opsonized with host serum/tissue components such as complement prior to phagocytosis. Therefore, we investigated the effects of opsonization and found that this increased the efficiency of CFSE-labeled parasite entry into monocytes (84.6% ± 8.8% versus 20.2% ± 3.8% monocytes infected; P < 0.001). Opsonization also increased the percentage of phagocytes undergoing a respiratory burst (66.0% ± 6.3% versus 41.0% ± 8.3% of monocytes containing CFSE-labeled parasites; P < 0.001) and the magnitude of oxidant generation by each infected monocyte. Inhibitor data indicated that DHE was oxidized by products of the NADPH oxidase. These data suggest that opsonized serum components such as complement lead to more efficient entry of Leishmania into their target cells but at the same time activate the phagocyte oxidase to generate microbicidal products in infected cells. The parasite must balance these positive and negative survival effects in order to initiate a viable infection.

Abstract

A study of ageing effects on the magnetic properties of Single-Molecule-Magnets (SMMs) of the Mn 12 based Acetate, ([Mn 12 O 12 (CH 3 COO) 16 (H 2 O) 4 ]·2CH 3 COOH·4H 2 O ( 1) and Stearate, [Mn 12 O 12 (CH 3 (CH 2 ) 16 COO) 11 (CH 3 COO) 5 (H 2 O) 4 ] ( 2) complexes has been carried out. Detailed magnetization (M) measurements as a function of temperature (T ∼ 1.8–10 K), magnetic field (H ∼ 0 to ±40 kOe) and time (t) have been performed on relatively fresh samples ( 1A and 2A ) and samples aged for ∼4 weeks ( 1B and 2B ) of both Mn 12 -Acetate and Mn 12 -Stearate. The blocking temperatures ( T B ) extracted from the measured M(T) lie between ∼3.0 and 3.4 K for all the four samples. In all cases, below T B , the M-H loops exhibit hysteresis with periodic steps. Interestingly, the ageing process leads to significant changes in the magnetic response of both the complexes. With ageing the Mn 12 -Acetate exhibits a large increase in the magnetization drop near zero-field, but the estimated anisotropy energy barrier ( U ) remains unchanged ∼71 K. Whereas, in the case of Mn 12 -Stearate ageing results in a change of U from ∼52 K ( 2A ) to ∼35 K ( 2B ). Abstract

The impact of single-molecule magnet (SMM), Mn12-acetate, on the dielectric properties of ferroelectric liquid crystal (FLC) has been investigated by dielectric spectroscopy and electro-optical techniques in a dual electrode sample cell (DESC). The temperature-dependent dielectric studies on Mn12-acetate/FLC composite have revealed the enhancement in the ferroelectric (smectic C ) to paraelectric (smectic A ) phase transition temperature by 3.5°C. The relaxation process corresponding to Goldstone mode in the ferroelectric phase of the composite is found to be slower compared to pure FLC sample. The electrical response for an input triangular wave shows the existence of one extra polarisation peak in Mn12-acetate/FLC composite which is ascribed to the induced dipole moment in Mn12-acetate molecule. The electro-optical texture of Mn12-acetate/FLC composite revealed that the incorporation of SMM in FLC significantly improve the memory effect. [ABSTRACT FROM AUTHOR]

Abstract

Purified terephthalic acid (PTA) plant of a petrochemical unit generates wastewater having high pollution load. Acid treatment of this wastewater reduces the chemical oxygen demand (COD) load by more than 50%, still leaving substantial COD load (>1500 mg/L) which should be removed. The present study reports on the use of a bio-waste-adsorbent bagasse fly ash (BFA) for the reduction of COD and other recalcitrant acids from this wastewater. The BFA showed basic character and was mesoporous with a BET specific surface area of 82.4 m/g. Optimum conditions for the adsorptive treatment of acid-pretreated PTA wastewater were found to be as follows: initial pH (pH) = 4, BFA dosage = 15 g/L, and contact time = 3 h. Adsorption treatment resulted in 58.2% removal of COD, 96.3% removal of terephthalic acid (TA), and 99.9% removal of benzoic acid (BA). TA and BA were removed from the pretreated PTA wastewater through precipitation and sedimentation of un-dissociated acid molecules inside the mesopores of the BFA. The results showed that the COD removed by the BFA followed pseudo-second-order kinetics. Equilibrium sorption data were best correlated by the Freundlich isotherm. The process of adsorptive removal of COD was found to be exothermic. The change in the Gibbs free energy was found to be negative, suggesting that the adsorption process is spontaneous and feasible for the treatment of PTA wastewater. [ABSTRACT FROM AUTHOR]

Abstract

Background: The transverse abdominis plane (TAP) block, a regional block provides effective analgesia after lower abdominal surgeries if used as part of multimodal analgesia. In this prospective, randomized double-blind study, we determined the efficacy of TAP block in patients undergoing cesarean section. Materials and Methods: Totally, 62 parturients undergoing cesarean section were randomized in a double-blind manner to receive either bilateral TAP block at the end of surgery with 20 ml of 0.25% bupivacaine or no TAP block, in addition to standard analgesic comprising 75 mg diclofenac 8 hourly and intravenous patient-controlled analgesia (PCA) tramadol. Each patient was assessed at 0, 4, 8, 12, 24, 36, and 48 h after surgery by an independent observer for pain at rest and on movement using numeric rating scale of 0-10, time of 1st demand for tramadol, total consumption of PCA tramadol, satisfaction with pain management and side effects. Results: Use of tramadol was reduced in patients given TAP block by 50% compared to patients given no block during 48 h after surgery (P < 0.001). Pain scores were lower both on rest and activity at each time point for 24 h in study group (P < 0.001), time of first analgesia was significantly longer, satisfaction was higher, and side effects were less in study group compared to control group. Conclusion: Transverse abdominis plane block was effective in providing analgesia with a substantial reduction in tramadol use during 48 h after cesarean section when used as adjunctive to standard analgesia.
