102. Thiol efflux mediated by an ABCC-like transporter participates for Trypanosoma cruzi adaptation to environmental and chemotherapeutic stresses

The protozoan Trypanosoma cruzi is the etiologic agent for Chagas disease, which affects 6-7 million people worldwide. The parasite presents high biological diversity, reflecting on the inefficiency of benznidazole in chronic or older patients. ABC superfamily proteins contain active transporters involved in the xenobiotic and endobiotic efflux and overexpressed in MDR cells. An ABCC-like transport was identified in the T. cruzi Y strain, being able to extrude thiol-conjugated compounds. As non-protein thiols represent prime line of defense towards reactive species, ABCC-like activity could participate in the regulation of mediators implicated in responses to cellular stress arising from a variety of stimuli, as environmental or chemotherapeutic. This study shows that T. cruzi ABCC-like protein transports GSH, GSSG and ceramides, all implicated in cellular stress. Hemin, representative from the hematophagous feeding of the vector, was transported as well, suggesting a role for ABCC as a metal-thiol transporter. In addition, all strains evaluated showed ABCC-like activity, while no ABCB1-like activity was detected. Also, results suggest that ABCC-like does not associate to natural resistance to benznidazole, considering that the sensitive strains CL Brener and Berenice showed higher ABCC-like activity than the resistant strains Y and Colombiana. Instead, ABCC-like efflux increased after continuous exposure of Y strain to benznidazole. Moreover, ABCC does not perform direct efflux of drug and its participation in the machinery of protection against stress depends on the efflux of metabolites in conjugation to or in cotransport with thiol.

Oltre alla presentazione dello stato dell’arte sull’Interim Design quale strumento di attuazione del progetto urbano tattico, si illustra un riuscito esperimento di riqualificazione temporanea con community design e costruzione partecipata dello spazio, una volta sottoutilizzato, del Playland alla 43rd Avenue di S. Francisco, precisando metodologia, fasi, attori, finanziatori e budget impegnati. Aspetto originale è il coinvolgimento dei giovani dall’inizio del progetto nella valutazione dei bisogni del quartiere, nella generazione dell’idea, nella costruzione, nella manutenzione e creazione di un luogo di svago molto gradito da utenti di tutte le fasce di età. La centralità degli aspetti di processo è determinante nella riuscita delle iniziative, potenziando esiti e prospettive evolutive. This paper illustrates the state of the art of temporary design as a tool of Tactical Urbanism with a specific focus on a successful regeneration project in San Francisco. Playland at 43rd Avenue is a community-driven, participatory project on an under-utilized lot. The paper illustrates the community design process, methodology, design and construction phases, stakeholders involved, sponsor and budget required for the project’s realization. Interesting and unique aspect of the project is the involvement of youth in the whole planning processfrom neighborhood’s existing needs assessment to the idea generation, design, construction and maintenance of this project that has been enjoyed by all ages. Focusing on the specifics of the design and implementation process of the project it is important to explain its success and to offer a blueprint for its replicability.

- Abstract
The NA62 experiment at the CERN SPS reports the first search for K+ -> pi(+) nu(nu) over bar using the decay-in-flight technique, based on a sample of 1.21 x10(11) K+ decays collected in 2016. The single event sensitivity is 3.15 x10(-10), corresponding to 0.267 Standard Model events. One signal candidate is observed while the expected background is 0.152 events. This leads to an upper limit of 14 x10(-10) on the K+ -> pi(+) nu(nu) over bar branching ratio at 95% CL., The cost of the experiment and of its auxiliary systems were supported by the funding agencies of the Collaboration Institutes. We review the physics opportunities of the Future Circular Collider, covering its e+e−, pp, ep and heavy ion programmes. We describe the measurement capabilities of each FCC component, addressing the study of electroweak, Higgs and strong interactions, the top quark and flavour, as well as phenomena beyond the Standard Model. We highlight the synergy and complementarity of the different colliders, which will contribute to a uniquely coherent and ambitious research programme, providing an unmatchable combination of precision and sensitivity to new physics.

The PANDA (anti-Proton ANnihiliation at DArmstadt) experiment will be one ofthe four flagship experiments at the new international accelerator complex FAIR(Facility for Antiproton and Ion Research) in Darmstadt, Germany. PANDA willaddress fundamental questions of hadron physics and quantum chromodynamicsusing high-intensity cooled antiproton beams with momenta between 1.5 and15 GeV/c and a design luminosity of up to 2× 10^32 cm−2 s−1. Excellent particleidentification (PID) is crucial to the success of the PANDA physics program.Hadronic PID in the barrel region of the target spectrometer will be performed by afast and compact Cherenkov counter using the detection of internally reflectedCherenkov light (DIRC) technology. It is designed to cover the polar angle rangefrom 22° to 140° and will provide at least 3 standard deviations (s.d.) π/Kseparation up to 3.5GeV/c, matching the expected upper limit of the final statekaon momentum distribution from simulation. This documents describes thetechnical design and the expected performance of the PANDA Barrel DIRCdetector. The design is based on the successful BaBar DIRC with several keyimprovements. The performance and system cost were optimized in detaileddetector simulations and validated with full system prototypes using particle beamsat GSI and CERN. The final design meets or exceeds the PID goal of clean π/Kseparation with at least 3 s.d. over the entire phase space of charged kaons in theBarrel DIRC.

- Abstract
The NA62 experiment at CERN reports a search for the lepton number violating decays K+ -> pi(-)e(+)e(+) and K+ -> pi(-)mu(+)mu(+) using a data sample collected in 2017. No signals are observed, and upper limits on the branching fractions of these decays of 2.2 x 10(-10) and 4.2 x 10(-11) are obtained, respectively, at 90% confidence level. These upper limits improve on previously reported measurements by factors of 3 and 2, respectively.

- Abstract
The NA62 experiment at the CERN SPS reports the first search for K+ -> pi(+) nu(nu) over bar using the decay-in-flight technique, based on a sample of 1.21 x10(11) K+ decays collected in 2016. The single event sensitivity is 3.15 x10(-10), corresponding to 0.267 Standard Model events. One signal candidate is observed while the expected background is 0.152 events. This leads to an upper limit of 14 x10(-10) on the K+ -> pi(+) nu(nu) over bar branching ratio at 95% CL.

The need to develop armour systems to protect against attacks from various sources is increasingly a matter of personal, social and national security. To develop innovative armour systems it is necessary to monitor developments being made on the type, technology and performance of the threats (weapons, projectiles, explosives, etc.) Specifically, the use of high protection level helmets on the battlefield is essential. The development of evaluation methods that can predict injuries and trauma is therefore of major importance. However, the risk of injuries or trauma that can arise from induced accelerations is an additional consideration. To develop new materials and layouts for helmets it is necessary to study the effects caused by ballistic impacts in the human head on various scenarios. The use of numerical simulation is a fundamental tool in this process. The work here presented focuses on the use of numerical simulation (finite elements analysis) to predict the consequences of bullet impacts on military helmets on human injuries. The main objectives are to assess the level and probability of head trauma using the Head Injury Criterion, caused by the impact of a 9 mm NATO projectile on a PASGT helmet and to quantify the relevance of projectile plasticity on the whole modelling process. The accelerations derived from the impact phenomenon and the deformations caused on the helmet are evaluated using fully three-dimensional models of the helmet, head, neck and projectile. Impact studies are done at impact angles ranging from 0 to 75°. Results are presented and discussed in terms of HIC and probability of acceleration induced trauma levels. Thorough comparison analyses are done using a rigid and a deformable projectile and it is observed that plastic deformation of the projectile is a significant energy dissipation mechanism in the whole impact process. The paper reports about the opportunity to apply the principles of sustainable stormwater management in urban open spaces, as realized in some countries all over the world. Focusing on the design criteria of green streets and emphasizing their multiple benefits, the main technical solutions of best management practices are presented in a glossary proposal. Having selected nine North American mayor cities, to study possible connections between facilities typologies and geographical positions, their climatic features are associated with the technical solutions suggested in the related stormwaters sustainable design manuals and toolkits. The discussion of presented comparisons indicates directions of development for critical reflections and research on the subject, useful for cities still lacking in these technical indications.

The exclusive charmonium production process in pp¯ annihilation with an associated π0 meson pp¯ → J=ψπ0 is studied in the framework of QCD collinear factorization. The feasibility of measuring this reaction through the J=ψ → eþe− decay channel with the AntiProton ANnihilation at DArmstadt (PANDA ¯ ) experiment is investigated. Simulations on signal reconstruction efficiency as well as the background rejection from various sources including the pp¯ → πþπ−π0 and pp¯ → J=ψπ0π0 reactions are performed with PANDAROOT, the simulation and analysis software framework of the PANDA ¯ experiment. It is shown that the measurement can be done at PANDA ¯ with significant constraining power under the assumption of an integrated luminosity attainable in four to five months of data taking at the maximum design luminosity

- Abstract
Evidence-based guidelines on the management of pancreatic cystic neoplasms (PCN) are lacking. This guideline is a joint initiative of the European Study Group on Cystic Tumours of the Pancreas, United European Gastroenterology, European Pancreatic Club, European-African Hepato-Pancreato-Biliary Association, European Digestive Surgery, and the European Society of Gastrointestinal Endoscopy. It replaces the 2013 European consensus statement guidelines on PCN. European and non-European experts performed systematic reviews and used GRADE methodology to answer relevant clinical questions on nine topics (biomarkers, radiology, endoscopy, intraductal papillary mucinous neoplasm (IPMN), mucinous cystic neoplasm (MCN), serous cystic neoplasm, rare cysts, (neo)adjuvant treatment, and pathology). Recommendations include conservative management, relative and absolute indications for surgery. A conservative approach is recommended for asymptomatic MCN and IPMN measuring < 40 mm without an enhancing nodule. Relative indications for surgery in IPMN include a main pancreatic duct (MPD) diameter between 5 and 9.9 mm or a cyst diameter >= 40 mm. Absolute indications for surgery in IPMN, due to the high-risk of malignant transformation, include jaundice, an enhancing mural nodule > 5 mm, and MPD diameter > 10 mm. Lifelong follow-up of IPMN is recommended in patients who are fit for surgery. The European evidence-based guidelines on PCN aim to improve the diagnosis and management of PCN.

Pancreatic ductal adenocarcinoma (PDAC) is a very aggressive tumor with a five-year survival of less than 6%. Chronic pancreatitis (CP), an inflammatory process in of the pancreas, is a strong risk factor for PDAC. Several genetic polymorphisms have been discovered as susceptibility loci for both CP and PDAC. Since CP and PDAC share a consistent number of epidemiologic risk factors, the aim of this study was to investigate whether specific CP risk loci also contribute to PDAC susceptibility. We selected five common SNPs (rs11988997, rs379742, rs10273639, rs2995271 and rs12688220) that were identified as susceptibility markers for CP and analyzed them in 2,914 PDAC cases, 356 CP cases and 5,596 controls retrospectively collected in the context of the international PANDoRA consortium. We found a weak association between the minor allele of the PRSS1-PRSS2-rs10273639 and an increased risk of developing PDAC (ORhomozygous = 1.19, 95% CI 1.02–1.38, p = 0.023). Additionally all the SNPs confirmed statistically significant associations with risk of developing CP, the strongest being PRSS1-PRSS2-rs10273639 (ORheterozygous = 0.51, 95% CI 0.39–0.67, p = 1.10 × 10−6) and MORC4-rs 12837024 (ORhomozygous = 2.07 (1.55–2.77, ptrend = 0.7 × 10−11). Taken together, the results from our study do not support variants rs11988997, rs379742, rs10273639, rs2995271 and rs12688220 as strong predictors of PDAC risk, but further support the role of these SNPs in CP susceptibility. Our study suggests that CP and PDAC probably do not share genetic susceptibility, at least in terms of high frequency variants. © 2017 UICC
