Methods ①Collecting the active ingredients of Shengji Yuhong ointment and their target genes We retrieved data from TCMSP, BATMAN-TCM and SuperPred databases to collect the active ingredients of Shengji Yuhong ointment and their target genes. ②Collecting target genes for the onset of DFU We searched and collected target genes for the onset of DFU in GeneCards, OMIM, PharmGkb, and TTD databases. ③Screening of target genes in the treatment of DFU by Shengji Yuhong ointment and analysis of their biological functions The target genes of the active ingredients of Shengji Yuhong ointment and the target genes for the onset of DFU was intersected to screen the target genes of Shengji Yuhong ointment in the treatment of DFU by Cytoscape3. 8. 0 software. Totally 1 762 target genes that had targeted binding relationships with the active ingredients of the ointment, mainly including epidermal growth factor receptor (EGFR), signal transducer and activator of transcription 3 (STAT3), and SRC proto-oncogene (SRC), transforming growth factor- β1 (TGF-β1), and serine/threonine protein kinase 1 (AKT1). ② There were 2 690 target genes for the onset of DFU, including EGFR, STAT3, SRC, TGF-β1, AKT1, etc. ③ There were 39 target genes of Shengji Yuhong ointment in the treatment of DFU, including EGFR, STAT3, AKT1, etc. The target genes of Shengji Yuhong ointment in the treatment of DFU were mainly involved in regulating epithelial cell proliferation, muscle cell proliferation, microRNA (miRNA) transcription, and other biological processes. The target genes were mainly involved in phosphatidylinositol-3-kinase/protein kinase B (PI3K/AKT), diabetic complications advanced glycation end products-receptor for advanced glycation end products (AGE-RAGE), and hypoxia-inducible factor-1 (HIF-1) signaling pathway, etc. ④ EGFR, STAT3, TGF-β1, SRC, and AKT1 were the core target genes of Shengji Yuhong ointment in the treatment of DFU. The active ingredients of Shengji Yuhong ointment with targeted binding relationships mainly included Gancao (Glycyrrhizae Radix et Rhizoma), Danggui (Angelicae Sinensis Radix), and Baizhi (Angelicae Dahuricae Radix). The docking score of quercetin with EG-FR was–7. 9 kcal/moL, the docking score of 1-methyl-2-dodecyl-4- (1H)-quinolone with EGFR was–5. 9 kcal/moL, and the docking scores of ursolic acid and licochalcone A with STAT3 were–8. 0 and–6. 3 kcal/moL, respectively. Conclusions The active ingredients of Shengji Yuhong ointment in the treatment of DFU mainly include quercetin, ursolic acid, licochalcone A, etc. The core target genes of Shengji Yuhong ointment in the treatment of DFU mainly include EGFR, STAT3, AKT1, etc. The main active ingredients of Shengji Yuhong ointment have good targeted binding abilities with the key target genes. The key target genes of Shengji Yuhong ointment in the treatment of DFU can participate in the process of promoting epithelial cell proliferation and inhibiting the production of advanced glycation end products by affecting PI3K/AKT, AGE-RAGE and HIF-1 signaling pathways, so as to exert its therapeutic effect on DFU.
