116. Assessment of obesity indices among young adults.

Abstract

Background: Overweight and obesity represent a rapidly growing threat to both physical and psychological health of children. The present study was conducted to assess obesity indices among young adults. Materials &Method: 110 subjects of both genders were enrolled. A thorough physical examination was carried out. Weight in kilograms and height in meters was recorded and BMI was calculated. Results: There were 65 males and 25 females in age group 18-20 years, 40 males and 45 females in age group 20-22 years and 45 males and 30 females in age group 23-25 years. There were 42.5% non-obese (males-47.5%, females-37.5%), 16% overweight (males-13.5%, females-18.5%), 28.5% obese (males- 25%, females- 32%) and 13% morbid obese) males-14%, females-12%). Maximum non-obese (60%) were seen in age group 18-20 years, overweight (20%) in age group 18-20 years, obese (38%) in age group 20-22 years and morbid obese (18%) in age group 23-25 years. A significant difference was observed in both genders (P< 0.05). Conclusion: Age group 23-25 years had maximum obese subjects. Obesity was most prevalent in males as compared to females. Pharmacovigilance refers to the systematic practice of reporting, assessing and preventing such events. Although such practice is vital in any healthcare system, its actual implementation has been found to be very limited in a country like Nepal. With the aim of disseminating information about such events and the role of healthcare professionals in pharmacovigilance, a one-day workshop was organized with international and national speakers on this subject at Nepal Cancer Hospital and Research Center, Lalitpur, Nepal, which also has a regional pharmacovigilance center . The workshop included lectures and hands-on training and had an audience from diverse fields of healthcare such as pharmacy, medical, surgical and radiation oncology, pathology and nursing staff. The feedback from the participants revealed that practical demonstration and hands-on training with extensive topic coverage were the best feature of the workshop, while less interaction during the lectures was a source of dissatisfaction with a recommendation to conduct more events focused on similar topics in the future.Keywords: adverse drug reaction, ADR reporting, Nepal, pharmacovigilance, workshop

Abstract

Chemoreceptor-based signaling pathways are among the major modes of bacterial signal transduction, and Pseudomonas aeruginosa PAO1 is an important model to study their function. Of the 26 chemoreceptors of this strain, PctA has a broad ligand range and responds to most of the proteinogenic amino acids, whereas PctB and PctC have a much narrower range and show strong ligand preference for L-glutamine and -aminobutyrate, respectively. Using several comparative genomics approaches, we show that these receptors are paralogs: pctA gene duplication in the common ancestor of the genus Pseudomonas led to pctC, whereas pctB originated through another, independent pctA duplication in the common ancestor of P. aeruginosa. Thus, the broad-range amino acid chemoreceptor was evolutionarily older, and chemoreceptors that complemented “missing” amino acid sensing abilities arose later in specific Pseudomonas lineages. Using comparative sequence analysis, newly solved crystal structures of PctA, PctB, and PctC ligand-binding domains, and their molecular dynamics simulations, we identified a conserved amino acid recognition motif and changes in the ligand-binding pocket that led to novel ligand specificities. In addition, we determined major forces driving the evolution of this group of chemoreceptors.

Abstract

Conventional immunization strategies will likely be insufficient for the development of a broadly neutralizing antibody (bnAb) vaccine for HIV or other difficult pathogens because of the immunological hurdles posed, including B cell immunodominance and germinal center (GC) quantity and quality. We found that two independent methods of slow delivery immunization of rhesus monkeys (RMs) resulted in more robust T follicular helper (TFH) cell responses and GC B cells with improved Env-binding, tracked by longitudinal fine needle aspirates. Improved GCs correlated with the development of >20-fold higher titers of autologous nAbs. Using a new RM genomic immunoglobulin locus reference, we identified differential IgV gene use between immunization modalities. Ab mapping demonstrated targeting of immunodominant non-neutralizing epitopes by conventional bolus-immunized animals, whereas slow delivery-immunized animals targeted a more diverse set of epitopes. Thus, alternative immunization strategies can enhance nAb development by altering GCs and modulating the immunodominance of non-neutralizing epitopes. Abstract

Empty wagon redistribution, train formation, routing and scheduling are complex problems for large railways, many of which currently have or are planning dedicated freight railway corridors (DFC). DFC operations due to their unique characteristics require research and new models for better operations planning. The rolling-stock, being expensive assets, need to be utilised in an optimal manner while meeting service quality levels. Motivated by Indian DFC, we present an integer programming formulation of the dynamic problem of empty distribution and train scheduling in DFC and discuss associated modelling issues. By unifying the separate problems into a single and dynamic model, we have developed a framework for more effective rolling stock utilisation. Based on this optimization model, an interactive decision support system is proposed for better decision-making on rolling-stock allocation and train scheduling. Extensive experiments and systematic analyses for a case of Indian DFC highlight the potentialities and effectiveness of the proposed DSS for DFC operations planning and management. Abstract

A  major  problem   in   ocular  therapeutics   is   the  attainment   of  optimal   drug concentration at the site of action which is compromised mainly due to precorneal      loss  resulting in  only a  small  fraction  of the drug being ocularly absorbed.  The effective dose administered may be altered by increasing the retention time of medication into the eye by using ocular inserts for treating various ocular diseases. The aim of the present study was to prepare and evaluate novel Ocular inserts of Ketorolac Tromethamine with polymers like HPMC K4M, PVA, Locust Bean Gum and Modified Locust Bean Gum with Glycerol as plasticizer by film casting method. Nine formulations of ocular inserts based on Comaprision Study On the basis of In vitro release studies & physicochemical parameters, the formulation MOG sustained with maximum cumulative of 97.244% for a period 12 hr and it was found to be better than other formulations; hence MOG 2% was selected as optimized formulation. In vitro drug release kinetic data revealed that all formulation followed near to zero order release kinetics, involving in the all formulation drug release by Super Case-II type of diffusion. Optimized formulation (MOG 12%) passed the test for sterility and in stability studies no change of physiological properties.  On the basis  of the present  study,  the ocular  inserts  of Ketorolac Tromethamine  gives  promising  future  for  the  ophthalmic  drug  delivery system. Kyllinga triceps rottb. A monocot perennial herb found in various parts of India is traditionally used in vitiated conditions pitta and vata, hyperdipsia, fever, liver disorders, verminosis, cough, splenopathy, diabetes and dermatitis. The aim of the present study was to evaluate the phytochemical and pharmacognostical study of ignored ayurvedic medicinal herb kyllinga triceps rottb. The plant is monocot grass belongs to the family cyperaceae, commonly used in various ayurvedic preparation’s and called musta. In various ayurvedic texts it is also known as nirvishi. Many species of family cyperaceae resembles the original drug thus the present study will help in identification and collection of original plant. The study includes identification and characterization of chemical component and preliminary phytochemical screening of the plant extract. The generated information of the present study will provide data which are helpful in the correct identification and authentication of medicinal plant kyllinga triceps rottb. and may help in prevention of its adulteration.
