Abstract

We show that the geometric deformation of shearing yields an improved decay rate for the heat semigroup associated with the Dirichlet Laplacian in an unbounded strip. The proof is based on the Hardy inequality due to the shearing established in [2] and the method of self-similar variables and weighted Sobolev spaces for the heat equation.

Abstract

We continue our study of the binary neutron star parameter space by investigating the effect of the spin orientation on the dynamics, gravitational wave emission, and mass ejection during the binary neutron star coalescence. We simulate seven different configurations using multiple resolutions to allow a reasonable error assessment. Due to the particular choice of the setups, five configurations show precession effects, from which two show a precession ("wobbling") of the orbital plane, while three show a "bobbing" motion, i.e., the orbital angular momentum does not precess, while the orbital plane moves along the orbital angular momentum axis. Considering the ejection of mass, we find that precessing systems can have an anisotropic mass ejection, which could lead to a final remnant kick of $\sim 40 \rm km/s$ for the studied systems. Furthermore, for the chosen configurations, antialigned spins lead to larger mass ejecta than aligned spins, so that brighter electromagnetic counterparts could be expected for these configurations. Finally, we compare our simulations with the precessing, tidal waveform approximant IMRPhenomPv2_NRTidalv2 and find good agreement between the approximant and our numerical relativity waveforms with phase differences below 1.2 rad accumulated over the last $\sim$ 16 gravitational wave cycles., Comment: 18 pages, 16 figures

125. Evaluation of the Tibial Plateau–Patella Angle (TPPA) in Dogs

Abstract

Estimating a dog’s patellar position involves various methods, which categorize it as norma, alta (high), or baja (low). However, they require various calculations. We aimed to evaluate the clinical applicability of a new method, the tibial plateau–patella angle (TPPA). This could aid in planning patella luxation surgery, estimating the patella position after TPLO and various osteotomies. We conducted a two-step study: first, on 15 stifles without pathologies from nine canine cadavers, and second, using 100 patient X-rays from the archive. Three stifle angle positions (45 ± 5°, 90 ± 5°, and 135 ± 5°) and three weight groups (S, M, and L) were evaluated in the first part of this study. Based on these results, the second part of this study was conducted using 100 pathology-free radiographs at the optimal stifle angle (90 ± 5°) from the archive. All radiographs were measured by three observers with varying levels of experience. Our results indicate that the stifle angle significantly impacted the TPPA, whereby lower values were detected with higher stifle angles, which remained consistent within the weight groups. High inter- and intra-observer agreement was achieved. The physiological TPPA values ranged from 26.7° to 48.8°, remaining consistent within the various weight groups. Observer 3 in Group S exhibited a 20% (insignificant) deviation, possibly due to challenges in determining the caudal point of the tibial plateau. In contrast with humans, TPPA values in dogs are negatively correlated with stifle angles, independent of weight. Our reliable and reproducible protocol suggests the potential benefits of training on small-breed dogs stifles.

Abstract

Stress responses play a vital role in cellular survival against environmental challenges, often exploited by cancer cells to proliferate, counteract genomic instability, and resist therapeutic stress. Heat shock factor protein 1 (HSF1), a central transcription factor in stress response pathways, exhibits markedly elevated activity in cancer. Despite extensive research into the transcriptional role of HSF1, the mechanisms underlying its activation remain elusive. Upon exposure to conditions that induce protein damage, monomeric HSF1 undergoes rapid conformational changes and assembles into trimers, a key step for DNA binding and transactivation of target genes. This study investigates the role of HSF1 as a sensor of proteotoxic stress conditions. Our findings reveal that purified HSF1 maintains a stable monomeric conformation independent of molecular chaperones in vitro. Moreover, while it is known that heat stress triggers HSF1 trimerization, a notable increase in trimerization and DNA binding was observed in the presence of protein-based crowders. Conditions inducing protein misfolding and increased protein crowding in cells directly trigger HSF1 trimerization. In contrast, proteosynthesis inhibition, by reducing denatured proteins in the cell, prevents HSF1 activation. Surprisingly, HSF1 remains activated under proteotoxic stress conditions even when bound to Hsp70 and Hsp90. This finding suggests that the negative feedback regulation between HSF1 and chaperones is not directly driven by their interaction but is realized indirectly through chaperone-mediated restoration of cytoplasmic proteostasis. In summary, our study suggests that HSF1 serves as a molecular crowding sensor, trimerizing to initiate protective responses that enhance chaperone activities to restore homeostasis.

Abstract

We deal with the numerical solution of linear partial differential equations (PDEs) with focus on the goal-oriented error estimates including algebraic errors arising by an inaccurate solution of the corresponding algebraic systems. The goal-oriented error estimates require the solution of the primal as well as dual algebraic systems. We solve both systems simultaneously using the bi-conjugate gradient method which allows to control the algebraic errors of both systems. We develop a stopping criterion which is cheap to evaluate and guarantees that the estimation of the algebraic error is smaller than the estimation of the discretization error. Using this criterion and an adaptive mesh refinement technique, we obtain an efficient and robust method for the numerical solution of PDEs, which is demonstrated by several numerical experiments., Comment: 30 pages, 6 figures

Abstract

Optogenetics has been harnessed to shed new mechanistic light on current and future therapeutic strategies. This has been to date achieved by the regulation of ion flow and electrical signals in neuronal cells and neural circuits that are known to be affected by disease. In contrast, the optogenetic delivery of trophic biochemical signals, which support cell survival and are implicated in degenerative disorders, has never been demonstrated in an animal model of disease. Here, we reengineered the human and Drosophila melanogaster REarranged during Transfection (hRET and dRET) receptors to be activated by light, creating one-component optogenetic tools termed Opto-hRET and Opto-dRET. Upon blue light stimulation, these receptors robustly induced the MAPK/ERK proliferative signaling pathway in cultured cells. In PINK1B9 flies that exhibit loss of PTEN-induced putative kinase 1 (PINK1), a kinase associated with familial Parkinson's disease (PD), light activation of Opto-dRET suppressed mitochondrial defects, tissue degeneration and behavioral deficits. In human cells with PINK1 loss-of-function, mitochondrial fragmentation was rescued using Opto-dRET via the PI3K/NF-кB pathway. Our results demonstrate that a light-activated receptor can ameliorate disease hallmarks in a genetic model of PD. The optogenetic delivery of trophic signals is cell type-specific and reversible and thus has the potential to inspire novel strategies towards a spatio-temporal regulation of tissue repair.

We analyze the convergence of the (algebraic) multiplicative Schwarz method applied to linear algebraic systems with matrices having a special block structure that arises, for example, when a (partial) differential equation is posed and discretized on a domain that consists of two subdomains with an overlap. This is a basic situation in the context of domain decomposition methods. Our analysis is based on the algebraic structure of the Schwarz iteration matrices, and we derive error bounds that are based on the block diagonal dominance of the given system matrix. Our analysis does not assume that the system matrix is symmetric (positive definite), or has the $M$- or $H$-matrix property. Abstract

The construction of accurate and consistent initial data for various binary parameters is a critical ingredient for numerical relativity simulations of the compact binary coalescence. In this article, we present an upgrade of the pseudospectral SGRID code, which enables us to access even larger regions of the binary neutron star parameter space. As a proof of principle, we present a selected set of first simulations based on initial configurations computed with the new code version. In particular, we simulate two millisecond pulsars close to their breakup spin, highly compact neutron stars with masses at about $98\%$ of the maximum supported mass of the employed equation of state, and an unequal mass systems with mass ratios even outside the range predicted by population synthesis models ($q = 2.03$). The discussed code extension will help us to simulate previously unexplored binary configurations. This is a necessary step to construct and test new gravitational wave approximants and to interpret upcoming binary neutron star merger observations. When we construct initial data, one has to specify various parameters, such as a rotation parameter for each star. Some of these parameters do not have direct physical meaning, which makes comparisons with other methods or models difficult. Abstract

Accurate and fast gravitational waveform (GW) models are essential to extract information about the properties of compact binary systems that generate GWs. Building on previous work, we present an extension of the NRTidal model for binary neutron star (BNS) waveforms. The upgrades are: (i) a new closed-form expression for the tidal contribution to the GW phase which includes further analytical knowledge and is calibrated to more accurate numerical relativity data than previously available; (ii) a tidal correction to the GW amplitude; (iii) an extension of the spin-sector incorporating equation-of-state-dependent finite size effects at quadrupolar and octupolar order; these appear in the spin-spin tail terms and cubic-in-spin terms, both at 3.5PN. We add the new description to the precessing binary black hole waveform model IMRPhenomPv2 to obtain a frequency-domain precessing binary neutron star model. In addition, we extend the SEOBNRv4_ROM and IMRPhenomD aligned-spin binary black hole waveform models with the improved tidal phase corrections. Focusing on the new IMRPhenomPv2_NRTidalv2 approximant, we test the model by comparing with numerical relativity waveforms as well as hybrid waveforms combining tidal effective-one-body and numerical relativity data. Abstract

CU Vir has been the first main sequence star that showed regular radio pulses that persist for decades, resembling the radio lighthouse of pulsars and interpreted as auroral radio emission similar to that found in planets. The star belongs to a rare group of magnetic chemically peculiar stars with variable rotational period. We study the ultraviolet (UV) spectrum of CU Vir obtained using STIS spectrograph onboard the Hubble Space Telescope (HST) to search for the source of radio emission and to test the model of the rotational period evolution. We used our own far-UV and visual photometric observations supplemented with the archival data to improve the parameters of the quasisinusoidal long-term variations of the rotational period. We predict the flux variations of CU Vir from surface abundance maps and compare these variations with UV flux distribution. We searched for wind, auroral, and interstellar lines in the spectra. The UV and visual light curves display the same long-term period variations supporting their common origin. New updated abundance maps provide better agreement with the observed flux distribution. The upper limit of the wind mass-loss rate is about $10^{-12}\,M_\odot\,\rm{yr}^{-1}$. We do not find any auroral lines. We find rotationally modulated variability of interstellar lines, which is most likely of instrumental origin. Our analysis supports the flux redistribution from far-UV to near-UV and visual domains originating in surface abundance spots as the main cause of the flux variability in chemically peculiar stars. Therefore, UV and optical variations are related and the structures leading to these variations are rigidly confined to the stellar surface. An even number of fermions can behave in a bosonic way. The simplest scenario involves two fermions which can form a single boson. But four fermions can either behave as two bipartite bosons or further assemble into a single four-partite bosonic molecule. In general, for 2N fermions there are many possible arrangements into composite bosons. The question is: what determines which fermionic arrangement is going to be realized in a given situation and can such arrangement be considered truly bosonic? This work aims to find the answer to the above question.
