In order to explore the chemical nature of red pigment in Nuodeng ham, the red pigment was extracted with 75% acetone aqueous solution and purified using a C18 solid phase extraction column, and its chemical structure was characterized by using ultraviolet-visible (UV-Vis) spectroscopy, fluorescence spectroscopy and ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), Fourier transform infrared (FTIR) spectroscopy and nuclear magnetic resonance (NMR) spectroscopy. The UV-Vis spectrum showed a strong absorption peak at 416 nm and two weakly symmetrical absorption peaks at 546 and 584 nm, which are characteristic of metalloporphyrins. When it was excited at 420 nm, the red pigment exhibited a strong fluorescence emission peak at 590 nm and a weak fluorescence emission peak at 644 nm, which highly coincided with those of Zn protoporphyrin IX (ZnPP) standard, indicating that the metal ions in the porphyrin ring were zinc ions. ZnPP was detected by UPLC-MS/MS in both positive (m/z 625.177 9 [M + H]+) and negative ion (m/z 623.161 4 [M-H]–) modes, and the stretching vibrations of characteristic functional groups on the porphyrin ring such as –CH3, –CH2–, C=O, –C–OH in carboxyl groups, C–H on olefin and porphyrin ring C=N were observed in the FTIR spectrum. In addition, the 1H-NMR results of the red pigment were consistent with the structure of the target compound ZnPP. Therefore, the red pigment in Nuodeng ham was identified as ZnPP. The results of this study provide a theoretical basis for the color regulation of Nuodeng ham.

Neurotrophic factors, particularly nerve growth factor, enhance neuronal regeneration. However, the in vivo applications of nerve growth factor are largely limited by its intrinsic disadvantages, such as its short biological half-life, its contribution to pain response, and its inability to cross the blood-brain barrier. Considering that let-7 (human miRNA) targets and regulates nerve growth factor, and that let-7 is a core regulator in peripheral nerve regeneration, we evaluated the possibilities of let-7 application in nerve repair. In this study, anti-let-7a was identified as the most suitable let-7 family molecule by analyses of endogenous expression and regulatory relationship, and functional screening. Let-7a antagomir demonstrated biosafety based on the results of in vivo safety assessments and it entered into the main cell types of the sciatic nerve, including Schwann cells, fibroblasts and macrophages. Use of hydrogel effectively achieved controlled, localized, and sustained delivery of let-7a antagomir. Finally, let-7a antagomir was integrated into chitosan conduit to construct a chitosan-hydrogel scaffold tissue-engineered nerve graft, which promoted nerve regeneration and functional recovery in a rat model of sciatic nerve transection. Our study provides an experimental basis for potential in vivo application of let-7a.

Objective: This study focuses on improving the comprehensive utilization rate of Polygonatum cyrtonema. Methods: The contents of total flavonoids, total saponins and total phenols from roots, stems, leaves, flowers and buds, of P. cyrtonema were determined. The scavenging rates of DPPH·, ·OH and nitrites in extracts of above tissue were measured and compared as well. Results: The contents of total flavonoids and total phenols five different kinds of non-medicinal tissue were in the same order, in which from highest to lowest was, leaves, buds, flowers, stems and roots, and the content of total saponins in flower was the highest In terms of antioxidant activity, the DPPH ·, ·OH and nitrite scavenging rate of leaf extract were the highest, followed by buds and flowers. Correlation analysis showed that the clearance of DPPH ·, · OH and nitrite by the extract from different tissue showed a certain positive correlation with the contents of total flavonoids, total saponins and total phenols. Conclusion: The extract of non-medicinal parts of P. cyrtonema has good biological activity.

Abstract The nucleus accumbens (NAc) is considered a hub of reward processing and a growing body of evidence has suggested its crucial role in the pathophysiology of major depressive disorder (MDD). However, inconsistent results have been reported by studies on reward network-focused resting-state functional MRI (rs-fMRI). In this study, we examined functional alterations of the NAc-based reward circuits in patients with MDD via meta- and mega-analysis. First, we performed a coordinated-based meta-analysis with a new SDM-PSI method for all up-to-date rs-fMRI studies that focused on the reward circuits of patients with MDD. Then, we tested the meta-analysis results in the REST-meta-MDD database which provided anonymous rs-fMRI data from 186 recurrent MDDs and 465 healthy controls. Decreased functional connectivity (FC) within the reward system in patients with recurrent MDD was the most robust finding in this study. We also found disrupted NAc FCs in the DMN in patients with recurrent MDD compared with healthy controls. Specifically, the combination of disrupted NAc FCs within the reward network could discriminate patients with recurrent MDD from healthy controls with an optimal accuracy of 74.7%. This study confirmed the critical role of decreased FC in the reward network in the neuropathology of MDD. Disrupted inter-network connectivity between the reward network and DMN may also have contributed to the neural mechanisms of MDD. These abnormalities have potential to serve as brain-based biomarkers for individual diagnosis to differentiate patients with recurrent MDD from healthy controls.

Abstract Philadelphia chromosome‐positive acute myeloid leukemia (Ph + AML) is a rare type of AML with a low survival rate and poor prognosis. We first report a Ph + AML patient who remained in long‐term remission after the combination of flumatinib and venetoclax, which could provide corresponding treatment ideas for clinical practice.

The CO2 sequestration capacity evaluation of reservoirs is a critical procedure for carbon capture, utilization, and storage (CCUS) techniques. However, calculating the sequestration amount for CO2 flooding in low-permeability reservoirs is challenging. Herein, a method combining numerical simulation technology with artificial intelligence is proposed. Based on the typical geological and fluid characteristics of low-permeability oil reservoirs in the Liaohe oilfield, the CMG 2020 version software GEM module is used to establish a model for CO2 flooding and sequestration. Meanwhile, a calculation method for the effective sequestration coefficient of CO2 is established. We systematically study the sequestration rules in low-permeability reservoirs under varying conditions of permeability, reservoir temperature, and initial reservoir pressure. The results indicate that, as the permeability and sequestration pressure of the reservoir increase, oil recovery gradually increases. The proportion of structurally bound sequestration volume increases from 55% to 60%. Reservoir temperature has minimal impact on both the recovery rate and the improvement in sequestration efficiency. Sequestration pressure primarily improves sequestration efficiency by increasing the dissolution of CO2 in the remaining oil and water. The calculation chart for the effective sequestration coefficient, developed using artificial intelligence algorithms under multi-factor conditions, enables accurate and rapid evaluation of the sequestration potential and the identification of favorable sequestration areas in low-permeability reservoirs. The role that visual discriminative ability plays among giant pandas in social communication and individual discrimination has received less attention than olfactory and auditory modalities. Here, we used an eye-tracker technology to investigate pupil fixation patterns for 8 captive male giant pandas Ailuropoda melanoleuca. We paired images (N =  26) of conspecifics against: 1) sympatric predators (gray wolves and tigers), and non-threatening sympatric species (golden pheasant, golden snub-nosed monkey, takin, and red panda), 2) conspecifics with atypical fur coloration (albino and brown), and 3) zookeepers/non-zookeepers wearing either work uniform or plain clothing. For each session, we tracked the panda's pupil movements and measured pupil first fixation point (FFP), fixation latency, total fixation count (TFC), and duration (TFD) of attention to each image. Overall, pandas exhibited similar attention (FFPs and TFCs) to images of predators and non-threatening sympatric species. Images of golden pheasant, snub-nosed monkey, and tiger received less attention (TFD) than images of conspecifics, whereas images of takin and red panda received more attention, suggesting a greater alertness to habitat or food competitors than to potential predators. Pandas' TFCs were greater for images of black-white conspecifics than for albino or brown phenotypes, implying that familiar color elicited more interest. Pandas reacted differently to images of men versus women. For images of women only, pandas gave more attention (TFC) to familiar combinations (uniformed zookeepers and plain-clothed non-zookeepers), consistent with the familiarity hypothesis. That pandas can use visual perception to discriminate intra-specifically and inter-specifically, including details of human appearance, has applications for panda conservation and captive husbandry. The ways in which learning environments are spatially conceived have undergone a significant transformation from space as “a realm without meaning” into place as “a meaningful location.” In this context, the relevance of online interactions and the significance of online space and place in facilitating positive learning are worth exploration. Drawing on a nationwide survey involving 1010 international students at 41 Chinese HEIs in the COVID‐19 pandemic, this research applied k‐means cluster analyses which produced a typology of international students' perceived online educational environments and another typology of their online interpersonal interactions. The logistic regression results indicated the predictive power of both typologies on ability development. The discussion highlights the importance of considering spatial dimensions of international students' online learning. Promoting international students' online interactions and supporting inclusive, engaging learning experiences require both space for hosting and place enabling intercultural learning. The watermelon (Citrullus lanatus L.) holds substantial economic value as a globally cultivated horticultural crop. However, the genetic architecture of watermelon fruit weight (FW) remains poorly understood. In this study, we used sh14-11 with small fruit and N14 with big fruit to construct 100 recombinant inbred lines (RILs). Based on whole-genome resequencing (WGR), 218,127 single nucleotide polymorphisms (SNPs) were detected to construct a high-quality genetic map. After quantitative trait loci (QTL) mapping, a candidate interval of 31–38 Mb on chromosome 2 was identified for FW. Simultaneously, the bulked segregant analysis (BSA) in the F2 population corroborated the identification of the same interval, encompassing the homologous gene linked to the known FW-related gene fas. Additionally, RNA-seq was carried out across 11 tissues from sh14-11 and N14, revealing expression profiles that identified 1695 new genes and corrected the annotation of 2941 genes. Subsequent differential expression analysis unveiled 8969 differentially expressed genes (DEGs), with 354 of these genes exhibiting significant differences across four key developmental stages. The integration of QTL mapping and differential expression analysis facilitated the identification of 14 FW-related genes, including annotated TGA and NAC transcription factors implicated in fruit development. This combined approach offers valuable insights into the genetic basis of FW, providing crucial resources for enhancing watermelon cultivation. [ABSTRACT FROM AUTHOR]
