Sphingosine-1-phosphate (S1P) is a bioactive sphingolipid that acts either as an intracellular messenger or as a ligand for its membrane receptors. S1P is a normal constituent of blood, where it is found both in plasma and blood cells. Compared with other cell types, sphingolipid metabolism in erythrocytes and platelets has unique features that allow the erythrocytes and platelets to accumulate S1P. In plasma, S1P is bound mainly to HDLs and albumin. Of note, metabolism and biological activity of S1P is to a large extent affected by the type of its carrier. Plasma S1P is characterized by a short half-life, indicating rapid clearance by degradative enzymes and the presence of high-capacity sources involved in maintaining its high concentration. These sources include blood cells, vascular endothelium, and hepatocytes. However, the extent to which each of these contributes to the plasma pool of S1P is a matter of debate. Circulating S1P plays a significant physiological role. It was found to be the key regulator of lymphocyte trafficking, endothelial barrier function, and vascular tone. The purpose of this review is to summarize the present state of knowledge on the metabolism, transport, and origin of plasma S1P, and to discuss the mechanisms regulating its homeostasis in blood.

Aim. To find out the strength of blood cells aggregation properties and specifics of antiaggregation vascular control in persons with arterial hypertension and dyslipidemia.Material and methods. The study was done on 380 patients with arterial hypertension of 1 -2 grade, risk 4 with dyslipidemia lib type, of middle age. Controls were 26 healthy same aged people. Studies were done with biochemical, hematological and statistical methods.Results. Among the patients studied with arterial hypertension and dyslipidemia we found a decrease of antioxidant plasma protection with activation of lipid oxidation, that significantly stimulated blood cells and caused alteration of vessel wall. In the persons studied there was significant decrease of the control of vessel wall over the increased cells aggregability.Conclusion. In comorbidity of arterial hypertension with dyslipidemia there is excessive erythrocyte, thrombocyte and neutrophils aggregation with a decrease of antiaggregation control by vessel wall.

Intraflagellar transport (IFT) proteins are vital for the genesis and maintenance of cilia. Our identification of ift122 transcripts in zebrafish thrombocytes that lack primary cilia was unexpected. IFT proteins serve transport in cilia, whose narrow dimensions may have necessitated the evolution of IFT from vesicular transport in ancestral eukaryotes. We hypothesized that IFTs might also facilitate transport within the filopodia that form when thrombocytes are activated. To test this possibility, we knocked down ift122 expression by injecting antisense Morpholino oligonucleotides (MOs) into zebrafish embryos. Laser-induced arterial thrombosis showed prolonged time to occlusion (TTO) of the vessel, as would be expected with defective thrombocyte function. Acute effects in adult zebrafish were evaluated by Vivo-Morpholino (Vivo-MO) knockdown of ift122. Vivo-MO morphants showed a prolonged time to thrombocyte aggregation (TTA) in the plate tilt assay after thrombocyte activation by the following agonists: ADP, collagen, PAR1 peptide, and epinephrine. A luminescence assay for ATP revealed that ATP secretion by thrombocytes was reduced in collagen-activated blood of Vivo-MO ift122 morphants. Moreover, DiI-C18 labeled morphant thrombocytes exposed to collagen showed reductions in filopodia number and length. Analysis of ift mutants, in which cilia defects have been noted, also showed prolongation of TTO in our arterial laser thrombosis assay. Additionally, collagen activation of wild-type thrombocytes led to a concentration of IFT122 both within and at the base of filopodia. Taken together these results, suggest that IFT proteins are involved in both the extension of filopodia and secretion of ATP, which are critical in thrombocyte function. [ABSTRACT FROM AUTHOR]

Objective: To examine granulocytic and non-granulocytic cells in children with severe congenital neutropenia (SCN) and their nonneutropenic parents. Materials and Methods: Fifteen patients with SCN and 21 nonneutropenic parents were evaluated for a) CD95, CD95 ligand, annexin V, propidium iodide, cell cycle, and lymphocyte subsets by flow cytometry; b) rapid cell senescence (of leukocytes) by senescenceassociated β-galactosidase stain; c) aggregation tests by aggregometer; d) in vitro bleeding time by PFA-100 instrument; e) mepacrine-labeled dense granule number of thrombocytes by fluorescence microscope; and f) hematomorphology by light and electron microscope. HAX1, ELANE, G6PC3, CSF3R, and JAGN1 mutations associated with SCN were studied in patients and several parents. Results: Significant increase in apoptosis and secondary necrosis in monocytes, lymphocytes, and granulocytes of the patients and parents was detected, irrespective of the mutation type. CD95 and CD95 ligand results implied that apoptosis was non-CD95- mediated. Leukocytes of 25%, 12.5%, and 0% of patients, parents, and controls showed rapid cell senescence. The cell cycle analysis testable in four cases showed G1 arrest and apoptosis in lymphocytes of three. The patients had HAX1 (n=6), ELANE (n=2), G6PC3 (n=2), and unidentified (n=5) mutations. The CD3, CD4, and NK lymphocytes were below normal levels in 16.6%, 8.3%, and 36.4% of the patients and in 0%, 0%, and 15.4% of the parents (controls: 0%, 0%, 5.6%). The thrombocytes aggregated at low rates, dense granule number/ thrombocyte ratio was low, and in vitro bleeding time was prolonged in 37.5%-66.6% of patients and 33.3%-63.2% of parents (vs. 0% in controls). Under electron and/or light microscope, the neutrophils, monocytes, lymphocytes, and thrombocytes in the peripheral blood of both patients and parents were dysplastic and the bone marrow of patients revealed increased phagocytic activity, dysmegakaryopoiesis, and necrotic and apoptotic cells. Ultrastructurally, thrombocyte adhesion, aggregation, and release were inadequate. Conclusion: In cases of SCN, patients' pluripotent hematopoietic stem cells and their non-neutropenic parents are both affected irrespective of the genetic defect. [ABSTRACT FROM AUTHOR]

Abstract Fungal hyphae constitute a special challenge for the immune system, since they are too large to be phagocytosed. This review summarizes our current knowledge on those immune cells that are able to attack and eliminate hyphae and we discuss the different means that are employed by these cells in order to kill hyphae. Platelets, or thrombocytes, are important players in inflammation, wound healing, initiation of immune response and regeneration in the organism. Disruption of the blood-brain barrier occurs during certain neurological disorders, such as brain trauma, Alzheimer’s disease or stroke, when blood cells including platelets can invade nervous tissue. However, the role of platelets in the context of neuroinflammation remains understudied. Recent studies have shown that activated platelets release a wide set of coagulative and vascular factors during neurovascular pathologies in the central nervous system. Moreover, platelets stimulate immunity and regulate inflammation in the central nervous system. Trophic and growth factors stored in platelets may play a role in neuronal regeneration. Activated platelets release neurotransmitters, serotonin, dopamine, histamine, and glutamate, and can modify neuronal cell activity in neuropathologies. This review focuses on the major aspects and mechanisms of platelet functions in neuroinflammation, and therapeutic potential of platelets for treatment of neurodegenerative disorders. Recently, ketamine has been investigated as a potential antidepressant option for treatment resistant depression. Unlike traditional drugs, it yields immediate effects, most likely via increased glutamatergic transmission and synaptic plasticity. However, ketamine administration in humans is systemic and its long–term impact on blood parameters has not yet been described in clinical studies. Here we investigated potential sustained effects of ketamine administration (0.5 mg/kg ketamine racemate) on hematological and biochemical values in plasma and serum in a randomized double–blinded study. 80 healthy young participants were included and whole blood samples were collected 5 days before, and 14 days after the infusion. To assess the group effect, repeated measure analyses of co–variance (rmANCOVA) were conducted for the following blood parameters: levels of sodium, potassium, calcium, hemoglobin and number of erythrocytes, lymphocytes , and thrombocytes. RmANCOVA revealed a significant time by treatment effect on thrombocyte levels ( F 1, 74 = 13.54, p < 0.001, eta = 0.155), driven by an increase in the ketamine group (paired t -test, t = −3.51, df = 38, p = 0.001). Specificity of thrombocyte effect was confirmed by logistic regression, and in addition, no other coagulation parameters showed significant interaction. Moreover, the relative increase in the ketamine group was stable across sexes and not predicted by age, BMI, smoking, alcohol or drug use, and contraception. Our results describe aftereffects of sub–anesthetic ketamine administration on blood coagulation parameters, which should be considered especially when targeting psychiatric populations with relevant clinical comorbidities. Objective: There are currently 2 representative diagnostic criteria for essential thrombocythemia (ET), the 2014 British Committee for Standards in Hematology Guidelines (BCSH) criteria and the 2016 World Health Organization (WHO) criteria. We compare and discuss the advantages and disadvantages of the 2 criteria. Method: We applied the 2 criteria to 403 patients with thrombocytosis and suspected myeloproliferative neoplasms (MPN) and compared patient populations. Results: The BCSH criteria diagnosed ET in 279 patients (BCSH-ET) whereas the WHO criteria diagnosed ET in 203 patients (WHO-ET). There were 83 patients diagnosable only by the BCSH criteria (BCSH-only-ET), of which under the WHO classification, 69 patients fell under the category of MPN, unclassifiable (MPN-u), 12 patients were PMF, prefibrotic/early stage (pre-PMF), and 2 patients were polycythemia vera. Patient characteristics such as age, hemoglobin, hematocrit, platelet counts, lactate dehydrogenase levels, JAK2V617F allele burdens, prevalence of myelofibrosis and splenomegaly, and frequencies of thrombotic events and treatment did not differ between WHO-ET and BCSH-only-ET, but BCSH-only-ET patients showed higher WBC counts and higher JAK2V617F mutation frequencies. Conclusion: The BCSH criteria diagnosed ET in a broader range of patients encompassing a significant number of patients who would otherwise be diagnosed as pre-PMF or MPN-u. [ABSTRACT FROM AUTHOR]

ABSTRACT This study describes the hematological and biometric characteristics of male and female Gymnotus species from the Pantanal, Mato Grosso do Sul state, Brazil. Fifty adult specimens of Gymnotus inaequilabiatus were weighed, measured, and then euthanized. Blood was collected by puncturing the celiac mesenteric vein to determine the hematocrit, hemoglobin content, number of erythrocytes, mean corpuscular volume, mean corpuscular hemoglobin concentration, glucose level, absolute value of leukocytes, and relative value of leukocytes and thrombocytes. Body weight and relative condition factor did not differ (P > 0.05) between the sexes, as well as erythrogram and the blood glucose values. Hematocrit ranged from 18.0% to 54.0%; hemoglobin from 1.1 to 14.7 g dL-1; number of erythrocytes from 0.2 × 106 to 3.8 ×106 µL-1; MCV from 24.2 to 321.7 fL; and MCHC from 4.2 to 44.5 g dL-1. In the differential count were identified thrombocytes, lymphocytes, neutrophils, monocytes, basophils, immature leukocytes, and PAS-positive granular leukocyte (PAS-GL). Females had a higher percentage of immature leukocytes (P < 0.05) than males. Glucose levels, erythrogram, leukogram, and the morphology of defense cells are comparable to other fish species of the Pantanal. Thrombocytes were the most frequent defense cells, followed by lymphocytes and neutrophils.

The exceptionally high cellular uptake of gold nanorods (GNRs) bearing cationic surfactants makes them a promising tool for biomedical applications. Given the known specific toxic and stress effects of some preparations of cationic nanoparticles, the purpose of this study was to evaluate, in an in vitro and in vivo in mouse, the potential harmful effects of GNRs coated with (16-mercaptohexadecyl)trimethylammonium bromide ( MTAB GNRs). Interestingly, even after cellular accumulation of high amounts of MTAB GNRs sufficient for induction of photothermal effect, no genotoxicity (even after longer-term accumulation), induction of autophagy, destabilization of lysosomes (dominant organelles of their cellular destination), alterations of actin cytoskeleton, or in cell migration could be detected in vitro . In vivo , after intravenous administration, the majority of GNRs accumulated in mouse spleen followed by lungs and liver. Microscopic examination of the blood and spleen showed that GNRs interacted with white blood cells (mononuclear and polymorphonuclear leukocytes) and thrombocytes, and were delivered to the spleen red pulp mainly as GNR-thrombocyte complexes. Importantly, no acute toxic effects of MTAB GNRs administered as 10 or 50 μg of gold per mice, as well as no pathological changes after their high accumulation in the spleen were observed, indicating good tolerance of MTAB GNRs by living systems. Sphingosine‐1‐phosphate (S1P) is a bioactive lysosphingolipid that is found in relatively high concentration in human plasma. Erythrocytes, endothelial cells, and activated platelets are the main sources of circulating S1P. The majority of plasma S1P is transported bound to high‐density lipoprotein (HDL) and albumin. In recent years, HDL‐bound S1P attracted much attention due to its cardioprotective and anti‐atherogenic properties. We have previously found that endurance‐trained athletes are characterized by higher plasma S1P concentration compared to untrained individuals. This finding prompted us to examine the effect of endurance training on S1P metabolism in blood. Thirteen healthy, untrained, male subjects completed an 8‐week training program on a rowing ergometer. Three days before the first, and 3 days after the last training session, blood samples were drawn from an antecubital vein. We found that total plasma S1P concentration was increased after the training. Further analysis of different plasma fractions showed that the training selectively elevated HDL‐bound S1P. This effect was associated with activation of sphingosine kinase in erythrocytes and platelets and enhanced S1P release from red blood cells. We postulate that increase in HDL‐bound S1P level is one of the mechanisms underlying beneficial effects of regular physical activity on cardiovascular diseases. [ABSTRACT FROM AUTHOR]

Background and objective In regenerative dentistry, platelet preparations are applied to stimulate bone healing and periodontal regeneration. Here, we pursue a strategy where bone substitutes are used as carriers for platelet-released supernatants. The mitogenic capacity and release kinetics of loaded bone substitutes were assessed. Material and Methods Platelet-released supernatants of washed platelets (washed PRS) and platelet-released supernatants of unwashed platelets (unwashed PRS) were lyophilized onto the bone substitutes deproteinized bovine bone mineral, hydroxyapatite and β-tricalcium phosphate. Scanning electron microscopy images were taken. Supernatants of bone substitutes were collected at hours 1, 3, 6, 24, and 48 and medium was replaced. We evaluated the protein content with the bicinchoninic acid assay and the effect on proliferation using bioassays with human periodontal fibroblasts. Release of growth factors from the loaded bone substitutes was measured based on the platelet-derived growth factor isoform ( PDGF- BB) and thrombin immunoassays. Furthermore, we assessed DNA and RNA content of washed PRS and unwashed PRS. Results Unwashed PRS showed higher total protein concentrations than washed PRS, while the concentration of PDGF- BB, thrombin, DNA, RNA and their mitogenic effect was not significantly different. The bone substitute materials adsorbed protein over time but no significant changes in overall appearance was found. Supernatants collected from unwashed PRS-loaded bone substitute after 1 h induced a potent mitogenic response in periodontal fibroblasts. This pro-mitogenic capacity of the supernatants decreased over the observation period. Supernatants of washed PRS-loaded bone substitutes did not induce a substantial mitogenic effect. Levels of PDGF- BB, thrombin and protein were higher in supernatants of unwashed PRS-loaded bone substitutes than of washed PRS-loaded bone substitutes. Conclusion Bone substitutes loaded with unwashed PRS, but not bone substitutes loaded with washed PRS show continuously declining release kinetics. Objectives To evaluate the efficacy of a salvage treatment with rituximab (RTX) in adults with primary immune thrombocytopenia (ITP), in terms of short-term response and long-term response (LTR, i.e., probability to achieve and maintain response) and to identify biological and clinical predictors of response. Methods We retrospectively evaluated the outcome of patients with primary ITP treated with standard dosage RTX (375 mg/m2 × 4) as salvage therapy in five Italian centers. One hundred and three patients, median age of 46 yr, were included. The median period of observation was 59 months. Results Response (R) and complete response (CR) were documented in 57 (55%) and 37 (36%) patients, respectively. Patients younger than 40 yr had a higher probability to achieve CR ( P = 0.025). Younger women (age < 40 yr) had a significantly higher probability to achieve R and CR ( P = 0.039 and P = 0.009, respectively). The estimated LTR rate was 36% and 31% after 48 and 72 months, respectively; female sex ( P = 0.033) and younger age ( P = 0.021) were associated with better LTR. Younger women had the highest LTR rate ( P = 0.006). Response duration was associated with the obtainment of CR after RTX (CR vs. partial response, P = 0.002). Conclusions The effect of RTX salvage treatment appears higher in younger women, with LTR rate possibly approaching that of splenectomy. [ABSTRACT FROM AUTHOR]

Background: Platelet preparations can stimulate the healing process and have mitogenic properties. We hypothesized that collagen barrier membranes (CBM), clinically used in guided bone regeneration and guided tissue regeneration, can serve as carriers for platelet secretome. Methods: Secretome was generated from washed platelets and unwashed platelets (washed/unwashed PSEC) and lyophilized onto CBM. Overall appearance of CBM was evaluated by scanning electron microscopy. The impact of PSEC on cell attachment was measured based on fluorescence microscopy with DiI-labeled cells. To assess the release kinetics, supernatants of CBM were collected and medium was replaced at hour 1-48. The mitogenic effect was evaluated with periodontal fibroblasts. Furthermore, the release of total protein, platelet-derived growth factor (PDGF)-BB, and transforming growth factor (TGF) β1 was measured. Results: CBM overall appearance and cell attachment was not modulated by PSEC. Supernatants taken after one hour induced a mitogenic response in fibroblasts and showed the highest levels of total protein, TGFβ1 and PDGF-BB. These effects decreased rapidly in subsequent supernatants. While supernatants of CBM loaded with unwashed PSEC induced a stronger mitogenic response than supernatants of CBM loaded with washed PSEC this difference between the PSEC preparations was not observed when cells were seeded on 48-hours-washed CBM. Balance of components' activity of thrombocyte-vessel hemostasis is the basis of the whole hemostasis process functioning in conditions of 'in vivo' and, thus, optimum of fl uidic blood features and adequacy of blood supply of all the tissues of an animal during whole its life. The aim is – to fi nd out physiological peculiarities of thrombocyte-vessel hemostasis of healthy cows during the normal process of pregnancy. The investigation was fulfilled with the help of 47 healthy cows of black-variegated breed which were examined and investigated 7 times during pregnancy: on the day of insemination, on the 45th, 90th, 135th, 180th, 230th and 280th day of pregnancy. The received results of state evaluation of thrombocyte-vessel hemostasis of cows after successful insemination testify to the existence of physiological regularity in the dynamics of its separate components' activity. It was found that during pregnancy the cows were noted to have weakening of thrombocyte hemostasis component's activity and strengthening of vessel hemostasis component. It provides them with necessary hemostatic and fl uidic blood features, thus promoting the formation of optimal conditions for placenta blood supply and anabolism in fetus' tissues. Plenova O.O.Bohomolets National Medical University Abstract Disorders in the hemostasis system have a major impact on the disease course and complication in the hypertensive disease and chronic obstructive pulmonary disease patients. To assess the platelet hemostasis we examined 62 persons: 15 apparently healthy (Control Group), 15 HD patients without COPD, 17 HD patients with COPD associated, and 15 COPD patients of Degree II and III. We have found out that HD, COPD and the mix patients demonstrate an increased thrombocyte functional activity expressed in a higher degree of spontaneous aggregation, and significant activation of arachidonic acid induced thrombocyte aggregation. The HD patients feature a specific increase of adrenalin induced aggregation, contrary to other examined groups, that may be explained by a role of sympathetic nervous system in the HD pathogenesis. Chronic inflammation in the COPD patients is accompanied with a significant increase of arachidonic acid thrombocyte reaction (the highest among all examined groups) that should be taken into account when establishing a treatment policy with anti-inflammatory agents included. 150 cadets ages 16 to 25years were under the obsrevation of adaptation of intravascular microcirculation component during the course of their military professional study. The research data proved that active erythrocyte aggregation, the size of thrombocyte aggregates and the speed of thrombocyte aggregation are higher in the first-, second- and third-year cadets than in senior students. These findings are to be used in the assessment ofcadets' adaptation to the military service and the prognosis of their professional capacity.

The aim of this paper is to compare three different methods for counting white blood cells [WBC] (Natt and Herrick method, estimation with 1,000 and 2,000 erythrocytes) and three methods for counting total thrombocytes [TT] (Wojtaszek method, estimation with 1,000 and 2,000 erythrocytes) in a South American freshwater turtle species, Podocnemis expansa, Schweigger 1812 (Reptilia, Pelomedusidae). Direct WBC counts using the Natt and Herrick method showed limitations, which are discussed here. The WBC and TT counts using 1,000 erythrocytes from blood smears are not recommended for Amazon turtles nor other reptilian species, since wide variation in counts can be observed. Estimation methods for determining WBC and TT based on 2,000 erythrocytes of blood smears were most acceptable because they allow a differentiation between leukocytes and thrombocytes and also had a smaller variation. The methods investigated here for the Amazon turtle, which have been widely used in other reptile species, provided evidence that the most acceptable method is not that of using diluted stains and a hemocytometer.O objetivo deste estudo foi comparar três diferentes métodos para contar leucócitos totais [LT] (método de Natt & Herrick, de estimação em 1000 e 2000 eritrócitos) e três métodos para contar trombócitos totais [TT] ( método de Wojtaszek, de estimação em 1000 e 2000 eritrócitos) em uma espécie de tartaruga de água doce da América do Sul, Podocnemis expansa, Schweigger 1812 (Reptilia, Pelomedusidae). As contagens diretas de LT usando o método de Natt & Herrick mostraram limitações que são aqui discutidas. As contagens de LT e TT usando estimativa em 1000 eritrócitos na extensão sanguínea não são recomendadas para tartaruga-da-Amazônia nem para outras espécies de répteis, pois houve ampla variação nestas contagens. Os métodos para determinar LT e TT baseados em 2000 eritrócitos nas extensões sanguíneas foram mais aceitáveis porque eles permitem uma diferenciação entre leucócitos e trombócitos, além disso, teve uma variação menor. Os métodos aqui investigados para tartaruga-da-Amazônia, os quais são amplamente usados em outras espécies de répteis, proveram evidências de que o método mais aceitável não é o que usa corantes diluentes e um hemocitômetro.

Toxoplasma (T.) gondii is able to infect various cell types in different hosts. The replication of this parasite within different peripheral mononuclear blood cell populations in chicken has not yet been fully understood. Aim of the present study was to investigate the impact of chicken erythrocytes and thrombocytes as potential host cells for T. gondii. Cultures of primary avian erythrocytes and thrombocytes were inoculated with tachyzoites of T. gondii type II strain ME49. Parasite replication was detected by a quantitative real-time PCR at different times postinoculation until 24 or 48 h, respectively, displaying long-term investigations for the chosen cultures. The parasite replication curve showed a continuous decrease of parasite stages in erythrocytes and thrombocytes. Observations by light microscopy showed massive destruction for both cell populations. Few macrophages in between the infected thrombocytes were viable during the investigation period and showed internalised tachyzoites by confocal laser scanning microscopy. These findings show that T. gondii is not capable of replication in chicken erythrocytes and thrombocytes; therefore, both cannot be considered as potential host cells. In further consequence, monocyte-derived macrophages seem to be the key to the dissemination mechanisms for T. gondii in chicken. [ABSTRACT FROM AUTHOR]

Background The storage time of platelet products negatively affects bacterial safety and platelet function. However, low maximum storage time increases outdating of valuable products. Thus, to quantify the effect of platelet storage time on platelet measurements after platelet transfusion, a systematic review and meta-analyses were performed. Methods Reports and meeting abstracts of randomized trials and observational studies, performed in humans, reporting platelet measurements after transfusion of platelet products of different storage times, were selected until February 2016. Meta-analyses were performed for four different storage time contrasts, each answering a different question. Random-effects models were used to account for substantial heterogeneity and the weighted mean differences calculated. Results Our search strategy yielded 4234 studies of which 46 papers satisfied the inclusion criteria. As judged by the 1-h corrected count increment, transfusions of fresher platelets compared to stored platelets showed better increment. The weighed mean difference varied from 2·11 (95% CI: 1·51-2·71) to 2·68 (95% CI: 1·92-3·45). For the 24-h corrected count increment, the weighted mean difference varied from 1·36 (95% CI: 0·12-2·60) to 1·68 (95% CI: 1·07-2·28) depending on the contrast. Recovery and survival of old platelets as percentage of fresh platelets were 81% and 73% for the original definition contrast. For the extended storage contrast, recovery and survival were 75% and 68%. Conclusions Fresh platelets were superior to old platelets for all platelet measurements and for all storage time contrasts meta-analysed. [ABSTRACT FROM AUTHOR]

INTRODUCTION: Referring to current standards the quality of an apheresis procedure is estimated by the quantity of collected cells. Nowadays a new kind of quality measurement could be found in the detection of cell volumina. Recent diagnostics have shown that stem cells and platelets - when separated - are likely to appear in a higher volume inside the cell product. Therefore, in this study the question should be discussed wether platelets of higher volume are more likely to be separated than platelets showing a lesser volume. METHODS: Blood samples of three different apheresis procedures could be observed: allogenic platelet donations (n = 7) (Trima, Terumo), autologous (n = 5) and allogenic stem cell donations (n = 5) (Cobe Spectra, Terumo). To examine the blood samples the Sysmex hematology analyser (XT-2000) has been used. RESULTS: During stem cell apheresis, the volume of the separated platelets was 1.2 fold increased compared to the platelet volume in the peripheral blood before separation. Before apheresis the mean platelet volume in the peripheral blood was found to be 6,21 fl, after apheresis 6,09 fl and inside the platelet concentrate 7,42 fl. The platelet number in the peripheral blood was also significantly decreased (before separation 180.1/nl and after separation 133.5/nl). In the blood products the concentration of platelets was nearly 8 fold higher than in the peripheral blood before separation. CONCLUSION: Overall, the observed apheresis procedures are more likely to separate platelets showing a higher voulme than common in the peripheral blood. This might indicate that not only the amount of separated cells reflects the quality of the apheresis procedure but also that the volume of the separated cells can be used as a parameter for quality assessment. A transcriptome wide analysis of the constructed cDNA library of snakehead murrel Channa striatus revealed a full length cDNA sequence of coagulation factor X. Sequence analysis of C . striatus coagulation factor X ( Cs FX) showed that the cDNA contained 1232 base pairs (bp) comprising 1209 bp open reading frame (ORF). The ORF region encodes 424 amino acids with a molecular mass of 59 kDa. The polypeptide contains γ-carboxyglutamic acid (GLA) rich domain and two epidermal growth factor (EGF) like domains including EGF-CA domain and serine proteases trypsin signature profile. Cs FX exhibited the maximum similarity with fish species such as Stegastes partitus (78%), Poecilia formosa (76%) and Cynoglossus semilaevis (74%). Phylogenetically, Cs FX is clustered together with the fish group belonging to Actinopterygii . Secondary structure of factor X includes alpha helix 28.54%, extended strand 20.75%, beta turn 7.78% and random coil 42.92%. A predicted 3D model of Cs FX revealed a short α-helix and a Ca 2+ (Gla domain) binding site in the coil. Four disulfide bridges were found in serine protease trypsin profile. Obviously, the highest gene expression (P < 0.05) was noticed in blood. Further, the changes in expression of Cs FX was observed after inducing with bacterial ( Aeromonas hydrophila ) and fungal ( Aphanomyces invadans ) infections and other synthetic immune stimulants. Variation in blood clotting time (CT), prothrombin time (PT) and activated prothromboplastin time (APTT) was analyzed and compared between healthy and bacterial infected fishes. Equilibria between the membranes of erythrocytes as well as thrombocytes and solution ions in fatal accidental hypothermia were analyzed using a theoretical four-equilibria model. The model was developed to determinate parameters characterizing cell membrane-surrounding ion interactions: the total surface concentrations of both acidic and basic groups C A, C B, and association constants K AH, K BOH. Knowledge of these parameters was necessary to calculate the theoretical values of surface charge density. The model was validated by curve-fitting the experimental data points to simulated data generated by the model. The process of myeloid cell development (myelopoiesis) in fish has mainly been studied in three cyprinid species: zebrafish ( Danio rerio ), ginbuna carp ( Carassius auratus langsdorfii ) and goldfish ( C. auratus , L.). Our studies on goldfish myelopoiesis have utilized in vitro generated primary kidney macrophage (PKM) cultures and isolated primary kidney neutrophils (PKNs) cultured overnight to study the process of macrophage (monopoiesis) and neutrophil (granulopoiesis) development and the key growth factors, receptors, and transcription factors that govern this process in vitro . The PKM culture system is unique in that all three subpopulations of macrophage development, namely progenitor cells, monocytes, and mature macrophages, are simultaneously present in culture unlike mammalian systems, allowing for the elucidation of the complex mixture of cytokines that regulate progressive and selective macrophage development from progenitor cells to fully functional mature macrophages in vitro . Furthermore, we have been able to extend our investigations to include the development of erythrocytes (erythropoiesis) and thrombocytes (thrombopoiesis) through studies focusing on the progenitor cell population isolated from the goldfish kidney. Herein, we review the in vitro goldfish model systems focusing on the characteristics of cell sub-populations, growth factors and their receptors, and transcription factors that regulate goldfish myelopoiesis. The application of simvastatine in arterial hypertension patients with metabolic syndrom corrects dislipoproteidemia, peroxidation syndrome, thrombocyte formation and optimized the thrombocyte hemostasis. Simvastatine inhibits decreased adhesive and aggregative function of thrombocytes. For stabilization of the correction results of metabolic processes and thrombocyte hemostasis in arterial hypertension patients with metabolic syndrom the application of simvastatine should be prolonged. With the aim of the decrease of body mass and weakening of insulin resistance in arterial hypertension patients with metabolic syndrom to suppose the combination of simvastatine with inmedicamental threatment.

The studies aimed at monitoring development of phagocytic and bactericidal activity in carps in the course of their ontogenetic development. The studies were performed using the techniques described by Mantur et al. (1986a, b), adapted to fish. Results were expressed in thrombocyte phagocytic index (Ipt), percentage of phagocyting thrombocytes (%tp), percentage of ingested bacteria (%bp) and in the index of intracellular killing by thrombocytes (Ibt). Number of thrombocytes was also examined using the technique of Dessi. Results of testing thrombocyte capacity to ingest the standard Staphylococcus aureus 209P strain showed that Ipt and %tp increased gradually in carps between the age of 3 and 17 months. In older carps, 19 to 29 months of age, as well as in spawners the growth in activity paralelled the ontogenetic development within Ipt values while %tp values remained at a similar level. Phagocytic activity of thrombocytes expressed in Ipt was reflected also by %bp values, which was particularly evident in fish aging 23 months to 5 years. In the case of Ibt, values of the index were not related in any way to stage of ontogeny or sex of the fish. Number of thrombocytes in carps aged 3 months to 5 years was increasing in parallel to their ontogenetic development.

Thrombocytes and their metabolites are involved in numerous physiological and pathophysiological processes, such as blood coagulation at the site of ruptured endothelium. Normal platelet aggregation can be suppressed via platelet antagonists or non-steroidal anti-inflammatory drugs (NSAIDs). That’s why differentiating between resting (“normal”) and inhibited platelets in a quick and precise fashion could be of a great clinical significance. Herein, we present the first step on the way to a novel platelet functionality assay based on Molecularly Imprinted Polymers (MIP) coated onto Quartz Crystal Microbalance (QCM). First results reveal sensor responses that depend on platelet concentration and on incubation of platelets with aspirin: acetyl salicylic acid (AAS).

Aim: Recurrent pregnancy loss (RPL) is known to be associated with increased thrombophilia and oxidative toxicity. However, the mechanism of thrombophilia apoptosis and oxidative toxicity is still unclear. In addition, the treatment of heparin induced regulator roles on intracellular free Ca 2+ ([Ca 2+ ] i ) and cytosolic reactive oxygen species (cytROS) concentrations in several diseases. TRPM2 and TRPV1 channels are activated by different stimuli, including oxidative toxicity. The aim of this study was to investigate the effects of low molecular weight heparin (LMWH) via modulation of TRPM2 and TRPV1 on calcium signaling, oxidative toxicity, and apoptosis in the thrombocytes of RPL patients., Study Design: Thrombocyte and plasma samples collected from 10 patients with RPL and 10 healthy controls were used in the current study., Main Findings: The [Ca 2+ ] i concentration, cytROS (DCFH-DA), mitochondrial membrane potential (JC-1), apoptosis, caspase-3, and caspase-9 levels were high in the plasma and thrombocytes of RPL patients, although they were diminished by the treatments of LMWH, TRPM2 (N-(p-amylcinnamoyl)anthranilic acid) and TRPV1 (capsazepine) channel blockers., Conclusions: The current study results suggest that the treatment of LMWH is useful against apoptotic cell death and oxidative toxicity in the thrombocytes of patients with RPL, which seem to be dependent on increased levels of [Ca 2+ ] i concentration via the activation of TRPM2 and TRPV1., (© 2023 Japan Society of Obstetrics and Gynecology.)

Introduction: Not only are early detection and treatment of diabetic foot ulcers important, but also acknowledging potential risk factors for amputation gives clinicians a considerable advantage in preventing amputations. Amputations impact both healthcare services and the physical and mental health of patients. This study aimed to investigate the risk factors for amputation in patients with diabetic foot ulcers., Methods: The sample for this study was patients with diabetic foot ulcers who were treated by the diabetic foot council at our hospital between 2005 and 2020. A total of 32 risk factors for amputation were identified and investigated among 518 patients., Results: Our univariate analysis showed that 24 of 32 defined risk factors were statistically significant. In the multivariate analysis using the Cox regression model, seven risk factors remained statistically significant. The risk factors most significantly associated with amputation were Wagner grading, abnormal peripheral arteries, hypertension, high thrombocyte levels, low haematocrit levels, hypercholesterolaemia and male sex, respectively. The most common cause of death in patients with diabetes who have undergone amputation is cardiovascular disease, followed by sepsis., Conclusion: To enable optimum treatment of patients with diabetic foot ulcers it is important for physicians to be aware of the amputation risk factors, and thus avoid amputations. Background & Aims Different mechanisms including portal hypertension and hypersplenism have been involved in the development of thrombocytopenia in cirrhosis. However, the relative contribution of each one is unknown. The aim was to evaluate simultaneously different mechanisms that determine platelet count in cirrhosis. Methods Cross-sectional study including cirrhotics ( n = 120) with hepatic venous pressure gradient ( HVPG) measurement. Samples were obtained from peripheral (P) veins to evaluate thrombopoietin ( TPO), stem cell factor, hepatocyte growth factor ( HGF), tumour necrosis factor, interleukin-(IL6) and (IL11) and from hepatic (H) veins to evaluate TPO. A subgroup ( n = 72) had spleen volume estimation. H and P- TPO were also measured in non-cirrhotic patients ( n = 15). Results Patients (Child A: 55, B: 43, C: 22) had a median platelet count of 81 000/mm3 ( IQR 60 500, 110 750), which correlated with spleen volume ( r = −0.38, P < 0.001). Platelets were associated also to HVPG ( r = −0.47, P = 0.004) and P- TPO ( r = 0.31, P = 0.050) only in compensated patients. H- TPO decreased, and the proportion of patients with P-TPO > H-TPO increased, with the presence and the severity of liver disease. H-TPO was correlated with liver function (bilirubin r = −0.350, P < 0.001 and international normalized ratio r = −0.227, P = 0.011). Patients with H-TPO < P-TPO had higher levels of IL-11 and HGF. Conclusion Platelet count in cirrhosis is associated mainly to spleen volume, although portal hypertension as estimated by HVPG and liver function plays a significant role in compensated patients. Raising ostriches became an important economic activity after their products became commodities. The health of farm animals is of paramount importance, so assessing basic immunological responses is necessary to better understand health problems. We developed a method to obtain ostrich thrombocytes and macrophages. The thrombocytes died by apoptosis after 48 h in culture, and the macrophages expanded in size and increased the number of acidic compartments. Macrophages were activated by chicken interferon-γ, producing high levels of nitric oxide. Toxoplasma gondii was able to infect these macrophages, and activation controlled parasitic reproduction. T. gondii, however, persisted in these cells, and infection reduced the production of nitric oxide. These results are important for the future assessment of the basic cellular and immunobiology of ostriches and demonstrate T. gondii suppression of nitric oxide production. [ABSTRACT FROM AUTHOR]

The hematologic system performs a number of essential functions, including oxygen transport, the execution of the immune response against tumor cells and invading pathogens, and hemostasis (blood clotting). These roles are performed by erythrocytes (red blood cells), leukocytes (white blood cells), and thrombocytes (platelets), respectively. Critically, circadian rhythms are evident in the function of all 3 cell types. In this review, we describe these oscillations, explore their mechanistic bases, and highlight their key implications. Since erythrocytes are anucleate, circadian rhythms in these cells testify to the existence of a nontranscriptional circadian clock. From a clinical perspective, leukocyte rhythms could underlie daily variation in the severity of allergic reactions, the symptoms of chronic inflammatory diseases, and the body’s response to infection, while the rhythmic properties of thrombocytes may explain daily fluctuations in the incidence of heart attack and stroke. Consequently, the efficacy of treatments for these conditions is likely to depend on the timing of their administration. Last, we outline preliminary evidence that circadian disruption in the hematologic system could contribute to the deleterious effects of poor diet, shift work, and alcohol abuse on human health. Background: Use of Carica papaya leaf extracts, reported to improve thrombocyte counts in dengue patients, demands further analysis on the underlying mechanism of its thrombopoietic cytokines induction Methods: In vitro cultures of peripheral blood leukocytes (PBL) and stem cells from human exfoliated deciduous teeth (SHED) were treated with unripe papaya pulp juice (UPJ) to evaluate its potential to induce thrombopoietic cytokines (IL-6 and SCF) Results: In vitro scratch gap closure was significantly faster (p < .05) in SHED culture treated with UPJ. IL-6 concentration was significantly increased (p < .05) in SHED and PBL culture supernatant when treated with UPJ. SCF synthesis in SHED culture was also significantly increased (p < .05) when treated with UPJ Conclusion: In vitro upregulated synthesis of IL -6 and SCF both in PBL and SHED reveals the potential mechanism of unripe papaya to induce thrombopoietic cytokines synthesis in cells of hematopoietic and mesenchymal origin. Purpose: Sphingosine-1-phosphate (S1P) regulates cardiovascular function and plays an important role in muscle biology. We have previously reported that cycling exercise increased plasma S1P. Here, we investigated the effect of exercise duration and intensity on plasma and skeletal muscle S1P levels. Methods: In the first experiment, 13 male athletes performed a 60-min exercise at 65 % of VO and a graded exercise until exhaustion on a rowing ergometer. Samples of the venous blood were taken, and plasma, erythrocytes and platelets were isolated. In the second experiment, ten male moderately active subjects performed three consecutive periods of one-leg knee extension exercise (at 25, 55 and 85 % of the maximal workload). Muscle biopsies and blood samples from the radial artery and femoral veins were taken. Results: Under basal conditions, S1P was released from the leg, as its concentration was lower in the arterial than in the venous plasma ( p < 0.01). Exercise until exhaustion increased plasma S1P and sphinganine-1-phosphate (SA1P) concentration ( p < 0.05), whereas moderate-intensity exercise elevated only SA1P ( p < 0.001). Although knee extension increased muscle S1P content ( p < 0.05), it was not released but taken up across the leg during exercise. However, sphingosine was released from both working and resting leg at the highest workload ( p < 0.05). Conclusions: Plasma S1P concentration is elevated only by high-intensity exercise which results, at least in part, from increased availability of sphingosine released by skeletal muscle. In addition, exercise markedly affects S1P dynamics across the leg. We speculate that S1P may play an important role in adaptation of skeletal muscle to exercise. [ABSTRACT FROM AUTHOR]
