Recently, broccoli, a vegetable of the Brassica family, has been found to protect the myocardium from ischaemic reperfusion injury through the redox signalling of sulphoraphane, which is being formed from glucosinolate present in this vegetable. Since cooked broccoli loses most of its glucosinolate, we assumed that fresh broccoli could be a superior cardioprotective agent compared to cooked broccoli. To test this, two groups of rats were fed with fresh (steamed) broccoli or cooked broccoli for 30 d, while a third group was given vehicle only for the same period of time. After 30 d, all the rats were sacrificed, and the isolated working hearts were subjected to 30 min ischaemia followed by 2 h of reperfusion. Both cooked and steamed broccolis displayed significantly improved post-ischaemic ventricular function and reduced myocardial infarction and cardiomyocyte apoptosis compared to control, but steamed broccoli showed superior cardioprotective abilities compared with the cooked broccoli. Corroborating with these results, both cooked and steamed broccolis demonstrated significantly enhanced induction of the survival signalling proteins including Bcl2, Akt, extracellular signal-regulated kinase 1/2, haemoxygenase-1, NFE2 related factor 2, superoxide dismutase (SOD1) and SOD2 and down-regulation of the proteins (e.g. Bax, c-Jun N-terminal kinase, p38 mitogen-activated protein kinase) of the death signalling pathway, steamed broccoli displaying superior results over its cooked counterpart. The expressions of proteins of the thioredoxin (Trx) superfamily including Trx1 and its precursor sulphoraphane, Trx2 and Trx reductase, were enhanced only in the steamed broccoli group. The results of the present study documented superior cardioprotective properties of the steamed broccoli over cooked broccoli because of the ability of fresh broccoli to perform redox signalling of Trx.

A kinetic spectrophotometric method for the determination of thiocyanate, based on its inhibitory effect on silver(I) catalyzed substitution of cyanide ion, by phenylhydrazine in hexacyanoferrate(II) is described. Thiocyanate ions form strong complexes with silver(I) catalyst which is used as the basis for its determination at trace level. The progress of reaction was monitored, spectrophotometrically, at 488 nm (lambda(max) of [Fe(CN)(5)PhNHNH(2)](3-), complex) under the optimum reaction conditions at: 2.5x10(-3)M [Fe(CN)(6)](4-), 1.0x10(-3)M [PhNHNH(2)], 8.0x10(-7)M [Ag(+)], pH 2.8+/-0.02, ionic strength (mu) 0.02 M (KNO(3)) and temperature 30+/-0.1 degrees C. A linear relationship obtained between absorbance (measured at 488 nm at different times) and inhibitor concentration, under specified conditions, has been used for the determination of [thiocyanate] in the range of 0.8-8.0x10(-8)M with a detection limit of 2x10(-9)M. The standard deviation and percentage error have been calculated and reported with each datum. A most plausible mechanistic scheme has been proposed for the reaction. The values of equilibrium constants for complex formation between catalyst-inhibitor (K(CI)), catalyst-substrate (K(s)) and Michaelis-Menten constant (K(m)) have been computed from the kinetic data. 4-Hydroxyphenylacetic acid (HPA) and nitrite are present in human mixed whole saliva, and HPA can be nitrated by peroxidase/hydrogen peroxide (H(2)O(2))/nitrite systems in the oral cavity. Thus, the objectives of the present study were to estimate the concentrations of HPA, nitrated HPA [4-hydroxy-3-nitrophenylacetic acid (NO(2)HPA)], nitrite, and thiocyanate (SCN(-)) in saliva from 73 patients with periodontal diseases and to elucidate the conditions necessary to induce nitration of HPA. High concentrations of HPA, nitrite, and SCN(-) were found in the saliva of patients older than 50 yr of age. NO(2)HPA was detected in seven patients who were older than 60 yr of age. Nitrite-dependent formation of NO(2)HPA by a bacterial fraction prepared from mixed whole saliva was faster at pH 5.3 than at pH 7, and increased as the rate of H(2)O(2) formation increased. The formation of NO(2)HPA was inhibited by SCN(-) and by salivary antioxidants such as uric acid, ascorbic acid, and glutathione. These results suggest that nitration can proceed at an acidic site in the oral cavity where H(2)O(2) is produced under conditions of decreased concentrations of SCN(-) and of antioxidants.

A poly(vinyl chloride) matrix membrane sensor for the selective determination of thiocyanate has been developed based on the use of copper(II)-2-(5-bromo-2-pyridylazo)-5-diethylaminophenol complex (Cu-PADAP) as a novel charged carrier, and o-nitrophenyloctyl ether (o-NPOE) as a solvent mediator. The sensor displays a significantly enhanced response towards SCN(-) ions over the concentration range 7.0 x 10(-6) to 1.0 x 10(-2) mol L(-1) with a detection limit of 5.6 x 10(-6) mol L(-1) and a calibration slope of -57.5 +/- 0.5 mV decade(-1). The sensor exhibits a long life-span, long-term stability, high reproducibility, and a fast response time. The selectivity coefficients of some anions were calculated using the separate solutions method, and found to be in the following order: SCN(-) > ClO(4)(-) > I(-) > Sal(-) > NO(2)(-) > Br(-) > NO(3)(-) = CH(3)COO(-) > Cl(-) > SO(4)(2-) = PO(4)(3-). The effects of the pH and ionic membrane additives (e.g. tridodecylmethylammonium chloride, TDMAC and potassium tetrakis[bis(3,5-trifluoromethyl)phenyl] borate, KTFPB) were examined. The sensor was used for the determination of SCN(-) ions in saliva and urine samples collected from some smoker and non-smoker donors. The developed sensor was also applied to determine the cyanide content in electroplating waste water samples after its conversion into thiocyanate. The application of the sensor to monitor the potentiometric titration of Ag(+) and Hg(2+) using SCN(-) resulted in sharp inflection breaks at the equivalent points. The data obtained using the proposed sensor correlate very well with results collected using the standard methods of thiocyanate, cyanide and metal analysis.

Erucin (ER) is a dietary isothiocyanate present in cruciferous vegetables, such as rocket salads (Erucasativa Mill., Diplotaxis sp.), that has been recently considered a promising cancer chemopreventive phytochemical. Biological activity of ER was investigated on human lung adenocarcinoma A549 cells, analyzing its effects on molecular pathways involved in apoptosis and cell cycle arrest, such as PARP-1 cleavage, p53 and p21 protein expression. Our results show that ER affects the A549 cell proliferation, enhancing significantly p53 and p21 protein expression in a dose-dependent manner (p<0.001). PARP-1 cleavage occurs only after exposure to high concentrations of ER (50 microM), in accordance to previous studies showing similar bioactivity of other isothiocyanates (ITCs). Our study reports for the first time that the induction of p53, p21 and PARP-1 cleavage may participate in the anti-proliferative activity of ER in human lung adenocarcinoma A549 cells. Comparison of data with those obtained with the isothiocyanate sulforaphane (SF), structurally related to ER, underlines the strong relationship between structural analogy of ITCs and their biological activity. The ability of dietary compounds to modulate molecular mechanisms that affect cancer cell proliferation is certainly a key point of the cancer prevention potential by functional foods.

Background and Objectives: Lactoperoxidase system contains Lactoperoxidase, Hydrogen peroxide and Thiocyanate ions, which have inhibitory action against cariogenic oral microflora. The present study was undertaken to assess the effect of lactoperoxidase system containing toothpaste on cariogenic microflora in children with early childhood caries., Methods: Study group included 30 children with Early Childhood Caries. 15 were considered as test group who used the test product Biotene toothpaste and other 15 as control group who used Colgate Active as control product. Salivary samples were analyzed for mutans streptococci (MS) and Lactobacilli, and for the levels of Thiocyanate ions., Results: Showed significant increase in the levels of Thiocyanate ion in saliva during experimental period. Compared to the control group test group showed significant increase in the levels of thiocyanate ions during experimental and washout period, whereas the number of colonies of MS and Lactobacilli were significantly reduced in test group during experimental period., Conclusion: The levels of thiocyanate ions can be increased in vivo by supplementing the saliva with natural enzymes like lactoperoxidase. This increased concentration of thiocyanate will reduce the number of cariogenic microflora in children with Early Childhood Caries.

A new optode membrane for the sensitive and selective determination of thiocyanate ion, based on a change in the absorption spectrum of a polymer film, is proposed. A membrane composed of plasticized poly vinyl chloride (PVC), an Fe(III) Schiff-base complex as a chromoionophore and hexadecyl trimethyl ammonium bromide (HDTMABr) as a cationic additive was prepared. The influence of different plasticizers was studied concerning the sensitivity, linear range and selectivity of the membrane film. Satisfactory analytical sensing characteristics for determining thiocyanate ion were obtained in terms of the selectivity, reversibility and reproducibility with a good detecting range. In addition, the optical film responds to thiocyanate ion reversibly over a wide dynamic range 1.0 x 10(-8) to 1.0 x 10(-3) M with fast response and recovery times. The optode membrane has been applied to determine the thiocyanate ion in urine samples.

Sulforaphane (1-isothiocyanato-4-(methylsulfinyl)-butane) content in cruciferous vegetables. Sulforaphane is an isothiocyanate which has antimicrobial and anticarcinogenic properties, this compound is found in a wide variety of plants from genus Brassica oleracea, being the most important broccoli and cabbage. The objective of this research was to quantify sulforaphane in the edible parts of broccoli and cabbage leaves by high-performance liquid chromatography (HPLC). Sample preparation for the quantification of sulforaphane include the conversion of glucoraphanin to sulforaphane (45 +/- 2 degrees C for 2.5 h), extracted with dichloromethane, purification of the extract in columns of solid phase extraction and detection by HPLC- UV. Sulforaphane concentration in broccoli is in the range of 214 microg/g DW (stems) to 499 microg/g DW (inflorescences). The purple cabbage (101.99 microg/g DW) has values greater than the green cabbage (7.58 microg/g DW). The inflorescences of broccoli and red cabbage leaves are rich in sulforaphane.

Environmental monitoring of biological wastewater treatment plants (BWWTP) treating industrial effluents produces large amount of data. Frequent sampling is done in the influent and effluent but also in intermediate points. Samples are analyzed for classical and specific contaminants and physical-chemical parameters are monitored. In this paper data from a BWWTP treating the effluents of a coke and steel-processing factory are analyzed. Due to a complex situation, this BWWTP gave poor performances that did not match environmental regulations, meanwhile upgrading proved to be uneasy. Data analysis using principal component analyses (PCA) or kinetic modeling with a Haldane model was unsuccessful in handling these data, which was attributed to undetermined toxic effects. A new methodology is reported, that allowed to identify a kinetics for thiocyanate degradation and a relation between pH and toxic effects. This analysis of the plant data allowed to make hypothesis on the process control parameters and to recommend management modifications, allowing a further increase of the performances.

Glucosinolate, sulfur-containing organic anions bonded to glucose, is found in plants including the family of Brassicaceae. Glucosinolate is hydrolyzed by myrosinase and produces various by-products that possess biological activities. Among the decomposed products, thiocyanate is known for its adverse effects on thyroid metabolism due to competition with iodine. This is of concern in that Korean dietary habits consist of consumption of large amounts of Brassicaceae vegetables such as cabbage and radishes, which may be correlated with high incidence of thyroid dysfunction. Thus, quantification of thiocyanate in Brassicaceae vegetables was performed by hydrolysis and spectrophotometrical detection. Average daily intake of Brassicaceae vegetables was obtained from the Third Korea National Health and Nutrition Examination Survey (KNHANES III), 2005-Nutrition Survey and from The Vegetable Production Statistics, 2007. Average daily intake of thiocyanate through Brassicaceae vegetables in Korea was estimated to be 16.3 micromol SCN(-)/d/person. When this was compared to published animal studies, average thiocyanate intake per person was lower than doses required to produce adverse effects. However, further studies may be warranted to ensure safety with sufficient margins of safety.

Ion-exclusion/cation-exchange chromatography with an eluent containing the bile salt-type zwitterionic surfactant CHAPS was performed in order to evaluate variations in anion (SO(4)(2-), NO(3)(-), and SCN(-)) and cation (Na(+), K(+), NH(4)(+), Mg(2+), and Ca(2+)) concentrations in human saliva. CHAPS prevents the adsorption of proteins to the stationary phase, i.e., weakly acidic cation-exchange resin, since it aggregates proteins without denaturing them. Addition of 1mM CHAPS to the eluent comprising 6mM tartaric acid and 7 mM 18-crown-6 yielded reproducible separations of anions and cations in protein-containing saliva. The resolutions of anions and cations were not significantly affected by the addition of CHAPS to the eluent. The concentrations of Na(+) and K(+) varied before and after meals; or that of SCN(-), upon smoking. The relative standard deviations of peak areas ranged from 0.3 to 5.1% in 1 day (n=20) and from 1.4 to 5.8% over 6 days (n=6).

Tropaeolum tuberosum Ruiz & Pavon, along with other several species, is an edible-tuber crop that grows in the Andean region. Folk medicine describes the use of mashua to reduce reproductive function in men. The present study aimed to evaluate the effect of mashua (1 g kg(-1)) on sperm production in rats during 7, 12, 21 and 42 days of treatment. The following parameters were assessed: reproductive organ weights, spermatid count and daily sperm production (DSP), sperm count in epididymis and sperm transit and serum testosterone levels. Freeze-dried extract of mashua had 3.7 g 100 g(-1) of benzyl glucosinolate. Mashua-treated rats showed a reduction in testicular spermatid number and DSP from day 12 to day 42; meanwhile, the effect of mashua was noted in epididymal sperm count after 12 and 42 days of treatment. In addition, epididymal sperm transit time was delayed at day 7 and it was accelerated on days 12 and 21 of treatment. No differences in serum testosterone levels were found between rats treated with vehicle and mashua after 42 days of treatment. Finally, mashua reduces testicular function after one spermatogenic cycle by reducing spermatid and sperm number, DSP and epididymal sperm transit time.

Perchlorate is an inhibitor of iodide uptake that is found widely in the environment. Given the potential for perchlorate accumulation during egg formation and the widespread consumption of eggs, it is important to examine eggs as a source of exposure to perchlorate and other potential inhibitors of iodide uptake (nitrate and thiocyanate). This study was conducted to determine potential human exposure to perchlorate from eggs produced by chicken flocks consuming differing amounts of perchlorate. The mean concentrations of perchlorate (7.16 ( 1.99 microg/kg of dry weight), nitrate (2820 ( 2100 microg/kg of dry weight), thiocyanate (574 +/- 433 microg/kg of dry weight), and iodide (2980 ( 1490 microg/kg of dry weight) in eggs (n = 180) from 15 chicken houses on 3 U.S. farms were determined. Chickens secreted into eggs an average of 23% of the perchlorate ingested from feed and water. Perchlorate levels in eggs were positively correlated with perchlorate intake (p < 0.001). Increased intake of perchlorate, nitrate, and thiocyanate was associated with decreased iodide levels in eggs, possibly indicating a competitive transport mechanism, such as sodium-iodide symporter. It was estimated that egg consumption contributes minimal perchlorate (approximately 0.040 microg) compared to the average total intake of approximately 10.5 microg for U.S. adults. Additionally, it was found that egg consumption was not associated with increased perchlorate exposure in 2820 individuals from the National Health and Nutrition Examination Survey (p value for the difference of least-squares means, pDiff = 0.225). From these findings it was concluded that, although chickens secrete perchlorate in eggs, eggs do not appear to be a significant source of perchlorate exposure for adults in the United States.

Environmental tobacco smoke is a major factor influencing the indoor air quality. Various toxic compounds emitted during tobacco smoking into the environment have a significant influence on the chemical composition of human biological fluids. The thiocyanate concentration in saliva is a biochemical measure, frequently used as an objective indicator of tobacco consumption. The goal of this study was to find significant relationships between salivary thiocyanates and other inorganic ions, which are constituents of natural saliva (Na(+), K(+), Mg(2+), Ca(2+), Cl(-), PO(4)(3-)) and to present the effectiveness of the proposed sample preparation procedure combined with ion chromatography technique for the determination of inorganic ions in human saliva samples collected from passive, moderate and heavy smokers.

Papaya is a climacteric fruit that has high amounts of benzylglucosinolates (BG) and benzylisothiocyanates (BITC), but information regarding levels of BG or BITC during fruit development and ripening is limited. Because BG and BITC are compounds of importance from both a nutritional and a crop yield standpoint, the aim of this work was to access data on the distribution and changes of BG and BITC levels during fruit development and ripening. BG and BITC levels were quantified in peel, pulp, and seeds of papaya fruit. Volatile BITC was also verified in the internal cavity of the fruit during ripening. The influence of the ethylene in BG and BITC levels and mirosinase activity was tested by exposing mature green fruits to ethylene and 1-methylcyclopropene (1-MCP). The highest BG levels were detected in seeds, followed by the peel and pulp being decreased in all tissues during fruit development. Similarly, the levels of BITC were much higher in the seeds than the peel and pulp. The levels of BG for control and ethylene-treated fruit were very similar, increasing in the pulp and peel during late ripening but not changing significantly in seeds. On the other hand, fruit exposed to 1-MCP showed a decrease in BG amount in the pulp and accumulation in seed. The treatments did not result in clear differences regarding the amount of BITC in the pulp and peel of the fruit. According to the results, ethylene does not have a clear effect on BITC accumulation in ripening papaya fruit. The fact that BG levels in the pulp did not decrease during ripening, regardless of the treatment employed, and that papaya is consumed mainly as fresh fruit, speaks in favor of this fruit as a good dietary source for glucosinolate and isothiocyanates.

Thiocyanate ranks after perchlorate as a potent inhibitor of iodide uptake by the thyroid but may be more concentrated in some food items such as milk products as to supersede perchlorate as the goitrogen of concern. A column-switching anion-exchange chromatographic method with UV spectral detection was developed to measure and confirm iodide and thiocyanate in powders of dry milk and infant formula. An aqueous solution was subjected to centrifugal ultrafiltration, the ultrafiltrate was cleaned up on a carbon solid-phase extraction column, and an aliquot was transferred to a precolumn for enrichment and subsequent injection onto an analytical column. In infant formula samples, thiocyanate was found at 2.0-5.1 mg/kg in five of seven milk-based products and was not found in the other two nor in three soy-based products tested (0.2 mg/kg LOQ); iodide was found at 0.3-1.3mg/kg (0.04 mg/kg LOQ). In 13 dry milk samples, thiocyanate was found at 27-38 mg/kg (1 mg/kg LOQ), and iodide was found at 1.8-3.2 mg/kg (0.2 mg/kg LOQ).

Perchlorate has the potential to cause thyroid dysfunction by inhibiting iodide uptake by the sodium iodide symporter. Perchlorate-contaminated waters may lead to human exposure through drinking water and food chain transfer in crops by way of irrigation water. Perchlorate has been found in dairy milk collected nationally and internationally. This study was conducted to evaluate perchlorate in the feed-dairy continuum in the southwestern United States. All feed products collected at dairies in this study had detectable levels of perchlorate as analyzed by ion chromatography-tandem mass spectrometry. The calculated total perchlorate intake across dairies ranged from 1.9 to 12.7 mg/cow per day. The variation in total perchlorate intake across dairies was largely associated with variation in forage and silage products. Alfalfa products were the single most important source of perchlorate intake variability among dairies. The estimated perchlorate intake from drinking water ranged from 0.01 mg per cow per day and was generally less than 2% of the total perchlorate intake. The perchlorate content of milk ranged from 0.9 to 10.3 microg/L and was similar to levels reported by the Food and Drug Administration's Total Diet Study. The perchlorate content of milk was significantly related to the presence of perchlorate in feed but the variation of perchlorate in milk could not be explained by feed intake alone.

In recent years, rocket plant (Eruca sativa) has gained greater importance as a vegetable and spice, especially among Europeans. E. sativa is a member of the Brassicaceae, which is considered to be an important chemopreventive plant family. In the present study, we assessed the chemopreventive potency and underlying mechanisms of extracts of E. sativa in HepG2 cells. No genotoxic effect could be observed in HepG2 cells treated with up to 50 microl/ml plant juice for 24 h when using the comet assay. In antigenotoxicity experiments, E. sativa extract reduced the benzo(a)pyrene-induced genotoxicity in a U-shaped manner. This effect was accompanied by a significant induction of glutathione S-transferase. No significant suppression of B(a)P-induced CYP1A1 protein expression or enzyme activity could be observed. Chemical analysis of the plant material by gas chromatography identified the isothiocyanates erucin, sulforaphane, erysolin and phenylethyl isothiocyanate. Results derived with the single ITC compounds support the assumption that their synergistic interaction is responsible for the strong antigenotoxicity of the plant material. The present study provided an assessment of the bioactive effects of rocket plant extract in a human cell culture system. This could help to evaluate the balance between beneficial vs. possible adverse effects of rocket plant consumption.

Brassicaceae seed meals produce ionic thiocyanate (SCN (-)), a bioherbicidal compound. This study determined the fate of SCN (-) in a field soil amended with seed meals of Sinapis alba, Brassica juncea, and Brassica napus and quantified crop phytotoxicity by monitoring carrot ( Daucus carota) emergence. Meals were applied at 1 or 2 t ha (-1), and soils were sampled to 35 cm for SCN (-). Maximum SCN (-) (211 micromol kg (-1) of soil) was measured at 5 days in 0-5 cm samples from plots amended with S. alba meal at 2 t ha (-1). Less than 30 micromol of SCN (-) kg (-1) of soil was measured at soil depths below 15 cm. At 44 days, SCN (-) was <15 micromol kg (-1) of soil in all treatments. Emergence inhibition of carrots seeded 15-36 days after meal amendment was found only in S. alba treatments. The rapid decrease of SCN (-) concentrations in Brassicaceae meal-amended soil indicates limited potential for off-site environmental impacts.

A self-aspirating, liquid microjunction surface sampling probe/electrospray emitter mass spectrometry system was demonstrated for use in the direct analysis of spotted and dosed drugs and their metabolites in thin tissue sections. Proof-of-principle sampling and analysis directly from tissue without the need for sample preparation was demonstrated first by raster scanning a region on a section of rat liver onto which reserpine was spotted. The mass spectral signal from selected reaction monitoring was used to develop a chemical image of the spotted drug on the tissue. The probe was also used to selectively spot sample areas of sagittal whole-body tissue from a mouse that had been dosed orally (90 mg/kg) with R,S-sulforaphane 3 h prior to sacrifice. Sulforaphane and its glutathione and N-acetyl cysteine conjugates were monitored with selected reaction monitoring and detected in the stomach and various other tissues from the dosed mouse. No signal for these species was observed in the tissue from a control mouse. The same dosed-tissue section was used to illustrate the possibility of obtaining a lane scan across the whole-body section. In total, these results illustrate the potential for rapid screening of the distribution of drugs and metabolites in thin tissue sections with the liquid micro-junction surface sampling probe/electrospray mass spectrometry approach. The lugworm Arenicola marina inhabits marine sediments in which sulfide concentrations can reach up to 2 mM. Although sulfide is a potent toxin for humans and most animals, because it inhibits mitochondrial cytochrome c oxidase at micromolar concentrations, A. marina can use electrons from sulfide for mitochondrial ATP production. In bacteria, electron transfer from sulfide to quinone is catalyzed by the membrane-bound flavoprotein sulfide : quinone oxidoreductase (SQR). A cDNA from A. marina was isolated and expressed in Saccharomyces cerevisiae, which lacks endogenous SQR. The heterologous enzyme was active in mitochondrial membranes. After affinity purification, Arenicola SQR isolated from yeast mitochondria reduced decyl-ubiquinone (K(m) = 6.4 microm) after the addition of sulfide (K(m) = 23 microm) only in the presence of cyanide (K(m) = 2.6 mM). The end product of the reaction was thiocyanate. When cyanide was substituted by Escherichia coli thioredoxin and sulfite, SQR exhibited one-tenth of the cyanide-dependent activity. Six amino acids known to be essential for bacterial SQR were exchanged by site-directed mutagenesis. None of the mutant enzymes was active after expression in yeast, implicating these amino acids in the catalytic mechanism of the eukaryotic enzyme.

An online technique for stacking based on the generation of a stationary isotachophoretic (sITP) boundary is presented. By balancing the anodic migration of an ITP boundary with a cathodic EOF, a stationary boundary is formed that can be used to indefinitely concentrate analytes according to ITP principles during electrokinetic injection. The ITP boundary is created by using an electrolyte containing a leading ion (chloride) and a suitable terminating ion added to the sample (2-morpholinoethanesulphonic acid, MES). Destacking and separation are achieved simply by replacement of the sample vial with electrolyte. The formation and stabilisation of the sITP boundary were evaluated through computer simulation which revealed that the pH had little impact upon the formation of the sITP boundary, but did govern the position at which it becomes stationary. Simulations also demonstrated that similar results were obtained when the capillary was initially filled with sample/terminator or leader/electrolyte, which was also supported by experimental results. Using 100 mM Cl(-), 200 mM Tris, pH 8.05 as the leader/electrolyte and adding 100 mM MES, 200 mM Tris, pH 8.05 to the sample, the sITP boundary was established after 5 min at -20 kV and was stable for at least 60 min. This provided detection limits for NO(2) (-), NO(3) (-) and SCN(-) of 0.05-0.66 ppb, which are 10,000 times lower than hydrodynamic injection and 10-50 times lower than other stacking approaches used for these inorganic ions.

The Colorado River is contaminated with low levels of perchlorate. Perchlorate has the potential to disrupt thyroid function by inhibiting the uptake of iodide. Brassica are rich sources of thiocyanate and nitrate, also inhibitors of iodide uptake. This study was conducted to estimate potential human exposure to perchlorate, thiocyanate, and nitrate from Brassica sp. irrigated with Colorado River water. Results indicate that Brassica sp. irrigated with Colorado River water do accumulate trace levels of perchlorate. However, the levels of perchlorate observed are low relative to the nitrate and thiocyanate naturally present in these species and low relative to the reference dose recommended by the NAS and the USEPA.

Thiocyanate (SCN) compounds in photo-processing wastewater (PPWW) could be treated by an SBR system without any release of thiocyanate to the atmosphere during the aeration step. An SCN loading greater than 84 g m(-3)d(-1) showed negative effects on the growth of bio-sludge and removal efficiencies of the system. The acclimatization period of the system was increased with an increase in SCN concentration or loading. The COD, BOD(5), TKN, and SCN removal efficiencies were 96.0 +/- 1.6%, 72 +/- 2%, 49 +/- 5%, and 82 +/- 3%, respectively, under an SCN loading of up to 84 g m(-3)d(-1). The removal efficiency of the system was repressed by SCN due to the repressed growth rate of nitrification bacteria. However, the removal efficiency could be increased with an increase in HRT or a decrease in SCN loading. Also, increases in HRT or decreases in SCN loading led to increased sludge age or solid retention time (SRT) and decreased the sludge volume index (SVI) value. The SRT and SVI of the system with synthetic wastewater containing 840 mg l(-1) SCN under an HRT of 3 days (SCN loading of 280 g m(-3)d(-1)) were 3.9 +/- 0.7 days and 65 +/- 4 ml g(-1), respectively, while they were 11.2+/-0.8 days and 55 +/- 6 ml g(-1), respectively under an HRT of 10 days (SCN loading of 84 g m(-3)d(-1)).

A new, simple and sensitive kinetic spectrophotometric method with no need for removing of interfering substances is proposed for the determination of thiocyanate ion in biological and water samples. The procedure is based on the inhibiting effect of thiocyanate on the sodium periodate-potassium bromide-meta cresol purple (MCP) system in acidic media. The induction period of the reaction is proportional to the SCN- concentration. The decolorization of meta cresol purple by the reaction products was used to monitor the reaction spectrophotometrically at 525 nm. Under optimum conditions, thiocyanate can be determined in the range of 0.02-0.8 microg ml(-1) with a 3sigma detection limit of 5 ng ml(-1). The relative standard deviations for 10 replicate determinations of 0.060, 0.10 and 0.50 microg ml(-1) thiocyanate are 3.7, 2.4 and 1.0%, respectively. This method has been successfully used to the determination of thiocyanate content in smokers and non-smokers saliva and spiked water sample.

An accurate, inexpensive and less laborious liquid-liquid extractive spectrophotometric procedure for the determination of thiocyanate ions in aqueous media has been developed. The method has been based upon the formation of a yellow colored complex ion associate of the ion-pairing reagent 1-(3, 5-diamino-6-chloropyrazinecarboxyl) guanidine hydrochloride monohydrate, namely amiloride hydrochloride, DPG+.Cl- and the thiocyanate ions in aqueous media containing HNO3 (0.5 mol L(-1)) and subsequent extraction with 4-methyl-2-pentanone. The absorption electronic spectrum of the ion associate showed one well-defined peak at lambda(max) 366 nm. The stoichiometric mole ratio of DPG+.Cl- to the thiocyanate ions is 1:1. The effective molar absorptivity (epsilon) of the ion associate at lambda(max) 366 nm is 1.1+/-0.1x10(4) L mol(-1) cm(-1). The extraction constants (K(d), K(ex), and beta) enabled a simple and convenient use of the developed binary ion associate for the extractive spectrophotometric determination of traces of thiocyanate ions in the aqueous media. Beer's law and Ringbom's plots are obeyed in the concentration range 0.05-10 and 0.1-7 microg mL(-1) of the thiocyanate ions, respectively with a relative standard deviation of +/-2.3%. The calculated lower limits of detection (LOD) and quantitation (LOQ) of the developed procedure for the thiocyanate ions were found equal to 0.02 and 0.066 microg mL(-1), respectively. The developed method has been applied for the determination of trace amounts of thiocyanate ions in tap-, waste- and natural water samples and compared successfully with the reported methods at the 95% confidence level. The proposed method was also applied successfully for the determination of thiocyanate ions in saliva samples.

Novel nano-silver coated multi-walled carbon nanotube composites were prepared and used to fabricate a modified electrode. The application of the nano-silver coated multi-walled carbon nanotube composites modified electrode for determination of trace thiocyanate is demonstrated for the first time. The influence of substrate, pH and interference of coexisting substances was investigated for response properties of the electrode. There was a linear relationship at the range 2.5 x 10(-9) to 5 x 10(-8) mol L(-1) and 5 x 10(-8) to 1 x 10(-6) mol L(-1) of thiocyanate with the decrement of anodic DPV peak currents. The limit of detection was 1 x 10(-9) mol L(-1)(S/N=3). The constructed electrode showed excellent reproducibility and stability. Actual urine and saliva samples of smoker and non-smoker were analyzed and satisfactory results were obtained. This method provides a new way to construct any electrode for biological and environmental analysis.

Aim: To determine the effect of two different extracts of red maca in male rats., Methods: Prostatic hyperplasia was induced in male rats with testosterone enanthate (TE). The study comprised six groups: one control group (group 1), one group treated with TE (group 2), two groups treated with TE and aqueous extract of red maca (groups 3 and 4), one group treated with hydroalcoholic extract of red maca (group 5) and one group treated with finasteride (0.1 mg, group 6). Differences in the aqueous extract dependent on the length of time of boiling, whether for 2 or 3 hours, for groups 3 and 4 was assessed. Extracts of red maca contained 0.1 mg of benzylglucosinolate. Thereafter, a dose-response effect of different doses of benzylglucosinolates (0.02-0.08 mg) in red maca extracts was assessed., Results: Prostate weight was similar in rats treated with freeze-dried aqueous extract of red maca prepared after 2 and 3 hours of boiling. Freeze-dried aqueous extract of red maca, hydroalcoholic extract of red maca and finasteride reduced prostate weight in rats with prostatic hyperplasia. No difference was observed between the data obtained from aqueous extract or hydroalcoholic extract of red maca. A dose dependent reduction of prostate weight was observed with the increase of the dose of benzylglucosinolates in red maca extracts., Conclusion: The present study showed that hydroalcoholic or aqueous extract of red maca containing 0.1 mg of benzylglucosinolate can reduce prostate size in male rats in which prostatic hyperplasia had been induced by TE.

In this work the effects of common household practices such as chilling, freezing, and cooking on vitamin C retention in broccoli (Marathon cv.), as well as their influence on the release of sulforaphane upon enzymatic hydrolysis of glucoraphanin by the endogenous enzyme myrosinase, were investigated. When chilled at 6 degrees C and 95% R.H. for 35 d, broccoli showed a vitamin C and sulforaphane loss of about 39% and 29%, respectively, while storage at -18 degrees C for 60 d resulted in similar losses, but mainly due to the blanching step. Boiling, steaming, microwaving, pressure-cooking, and the combined use of pressure and microwaves were the cooking methods investigated. Boiling and steaming caused significant vitamin C losses, 34% and 22%, respectively, while with the other treatments more than 90% retention was observed. Sulforaphane was no more detectable after boiling or steaming, while pressure/microwave cooking did not cause any significant loss. The quantitative distribution of these moieties in the florets and stems of fresh and chilled broccoli was also investigated.

Background: Perchlorate and thiocyanate interfere with iodide uptake at the sodium-iodide symporter and are potential disruptors of thyroid hormone synthesis. Perchlorate is a common contaminant of water, food, and human milk. Although it is known that iodide undergoes significant diurnal variations in serum and urinary excretion, less is known about diurnal variations of milk iodide levels., Objectives: Variability in perchlorate and thiocyanate excretion in human milk has not been examined. Our objective was to determine variability of perchlorate, thiocyanate, and iodide in serially collected samples of human milk., Methods: Ten lactating women were asked to collect six milk samples on each of 3 days. As an alternative, subjects were asked to collect as many milk samples as comfortably possible over 3 days. Samples were analyzed for perchlorate, iodide, and thiocyanate by ion chromatography coupled with mass spectrometry., Results: Individual perchlorate, iodide, and thiocyanate levels varied significantly over time; there was also considerable variation among individuals. The iodide range, mean +/- SD, and median for all samples (n = 108) were 3.1-334 microg/L, 87.9 +/- 80.9 microg/L, and 55.2 microg/L, respectively. The range, mean +/- SD, and median of perchlorate in all samples (n = 147) were 0.5-39.5 microg/L, 5.8 +/- 6.2 microg/L, and 4.0 microg/L. The range, mean +/- SD, and median of thiocyanate in all samples (n = 117) were 0.4 -228.3 microg/L, 35.6 +/- 57.9 microg/L, and 5.6 microg/L. The data are not symmetrically distributed; the mean is higher than the median in all cases., Conclusions: Iodine intake may be inadequate in a significant fraction of this study population. Perchlorate and thiocyanate appear to be common in human milk. The role of these chemicals in reducing breast milk iodide is in need of further investigation.

A simple, selective and rapid method for solid phase extraction and spectrophotometric determination of thiocyanate using a manganese (III) tetrakis (p-sulfonatophenyl) porphyrin, [Mn (TPPS) OAC] bound to Amberlite IR-400 has been developed. The influence of pH, amount of solid phase, sample matrix, type and amount of eluting agent and flow rates i.e. variables affecting the efficiency of the extraction system were evaluated and conditions of the sample, eluting solution and active phase were optimized. The maximal capacity was found to be as 1.16 microg mL(-1) for 1200 mL. Thiocyanate ions can be eluted quantitatively with 8 mL 0.3 M ferric chloride. The enrichment factor was 150. The linear range of the determination is between 0.4-2.0 microg mL(-1) for preconcentration method with a limit of detection of 2.8 ng mL(-1). The method has been successfully applied for determination of trace amounts of thiocyanate in tap water, saliva sample and a synthetic mixture.

A modified principle component artificial neural network (PC-ANN) model is developed for simultaneous determination of thiocyanate and salycilate concentration after passing through the bulk of a liquid membrane by tri-phenyl benzyl phosphonium chloride. All calibration, and test samples data were obtained using UV-Vis spectrophotometer. In this way, a modified PC-ANN consisting of three layers of nodes was trained by combination of Bayesian-Levenberg-Marquardt as training rule. Sigmoid and liner transfer functions were used in the hidden and output layers respectively to facilitate nonlinear calibration. The model could accurately estimate the concentration of components with acceptable precision and accuracy, for mixtures. The PC-ANN model exhibits a good ability for the simultaneous determination of the thiocyanate and salycilate in concentration range 0.5 x 10(-4) mol.l(-1) up to 5.0 x 10(-4) mol.l(-1) with Root Mean square error (2.22% and 2.20%, for thiocyanate and salycilate, respectively) and high correlation coefficients (R2= 0.998 or greater). Results obtained with modified trained PC-ANN were compared with stepwise linear regression (SMLR) model. Validation of the two models shows a better ability in estimation of the modified PC-ANN as compared with the SMLR model (MSRE given are 3.12%, 6.31%.).

The determination of biomarkers in human body fluids is a useful tool, which allows the quantitative assessment of the exposure to chemicals or complex mixtures of chemicals and of early biological effects as a result of the exposure. Biomarkers require validation before their successful application in human studies. This review describes some general purposes of human biomonitoring and biomarkers including the requirements for validation. Risk assessment and harm reduction of smoking and tobacco products, respectively, is a very suitable field for the application of biomarkers. A brief historical review shows that the application of biomarkers of exposure and effect in human smoking goes back more than 150 years. Two 'classical' biomarkers of exposure to tobacco, namely carboxyhemoglobin (COHb and its equivalent carbon monoxide in exhalate, COex) and thiocyanate (SCN) in body fluids are discussed in terms of sources of exposure, metabolism, disposition kinetics and influencing host factors. Data on COHb/COex and SCN in nonsmokers and smokers as well as the power to discriminate between smokers and nonsmokers are presented. Both biomarkers are significantly correlated with the daily cigarette consumption. Smoking machine-derived yields of the precursors carbon monoxide and hydrogen cyanide were not correlated with COHb/COex and SCN, respectively. It is concluded that, while COHb/COex is a useful biomarker for assessing the smoke inhalation, preferably in controlled studies, the application of SCN in body fluids as a biomarker for smoking is limited, mainly due to the abundance of other sources for SCN.

A method is described for simultaneous determination of cyanide (CN) and thiocyanate (SCN) in human saliva, or oral fluid. SCN concentrations in body fluids appeared to be important in classifying patients as smokers or nonsmokers, in determining some clinical conditions, and in specimen validity testing in forensic drug testing. The human saliva samples were diluted and the anions were separated by an extractive alkylation technique. Tetrabutylammonium sulfate was used as phase-transfer catalyst and pentafluorobenzyl bromide as the derivatizing agent. The products were analyzed by a gas chromatography-mass spectrometry (GC-MS) with selected ion monitoring method. 2,5-Dibromotoluene was used as internal standard for quantitation of CN and SCN in saliva. The calibration plot was linear over the concentration range from 1 to 100 micromol/L (0.026-2.60 microg/mL) for CN (R=0.9978) and 5 to 200 micromol/L (0.29-11.6 microg/mL) for SCN (R=0.9996). The method was used to examine 10 saliva specimens. The concentration ranged from 4.8 to 29 micromol/L (0.13-0.75 microg/mL) for CN and 293 to 1029 micromol/L (17-59.7 microg/mL) for SCN. The SCN results were similar to those obtained from a method using oxidation of SCN to CN with colorimetric detection (R=0.9882). The proposed GC-MS confirmatory method was found useful when the concentrations of CN and SCN in saliva needed to be accurately determined.

The aim of this work was to investigate the association between recurrent aphthous stomatitis (RAS) and salivary thiocyanate levels. The sample comprised men and women of age ranging from 15 to 55 years, who were allocated to four groups: 28 patients in RAS active phase (group 1); 28 patients in RAS remission phase (group 2); 29 smokers (group 3); 26 non-smokers without RAS (group 4). Samples of whole saliva mechanically stimulated were collected, and thiocyanate levels were measured. The results were analyzed by ANOVA and paired t-test. Mean salivary thiocyanate values were 0.55 mM, 0.64 mM, 2.36 mM and 0.96 mM in groups 1 (active RAS), 2 (remission RAS), 3 (smokers) and 4 (control), respectively. There was no significant difference in thiocyanate levels when groups 1 and 2 were compared with group 4. Group 3 showed a significantly higher thiocyanate concentration when compared with groups 1, 2 and 4 (P < 0.05). There was no significant difference in thiocyanate levels between groups 1 and 2 (P > 0.05). It is therefore suggested that there is no association between RAS and salivary thiocyanate levels.

In the human oral cavity, nitrite is reduced to nitric oxide (NO) by certain bacteria. 4,5-Diaminofluorescein (DAF-2) was transformed to a fluorescent component triazolfluorescein (DAF-2T) in a bacterial fraction of saliva in the presence of nitrite. No detectable consumption of DAF-2 and formation of DAF-2T were observed in bacterial fraction in the absence of nitrite. The nitrite-dependent transformation of DAF-2 to DAF-2T was inhibited by catalase, SCN(-), and CN(-) suggesting the participation of peroxidases in saliva in the transformation. The formation of DAF-2T, which was observed by the addition of an NO generating reagent (+/-)-(E)-4-ethyl-2-[(E)-hydroxyimino]-5-nitro-3-hexenamide (NOR 3) to bacterial fraction, was also inhibited by catalase, SCN(-), and CN(-). The degree of the inhibition by SCN(-) decreased as the concentration of nitrite or NOR 3 was increased. Superoxide dismutase (SOD) enhanced nitrite- and NOR 3-induced fluorescence increase in the bacterial fraction, and the degree of the enhancement decreased as the concentrations of nitrite and NOR 3 were increased. In whole saliva filtrate, the inhibitory effects of SCN(-) on the fluorescence increase decreased as the concentration of nitrite was increased, but the enhancement by SOD was not significantly affected by the increase in the concentration of nitrite. As salivary bacteria produce O(2)(-), H(2)O(2), and NO and as peroxidase/H(2)O(2)/nitrite systems in saliva produce NO(2), the effects of SCN(-) are discussed taking SCN(-)-dependent inhibition of NO(2) formation by peroxidases in saliva into consideration and the effects of SOD are discussed taking O(2)(-)-dependent consumption of NO into consideration. It is concluded that when the rate of the formation of NO is high, SCN(-) is not effective enough to inhibit the formation of N(2)O(3) in the oral cavity.

This paper describes a method for the determination of cyanide using in-capillary enzymatic reaction with rhodanese. Poorly absorbing cyanide is in rhodanese reaction transformed into highly absorbing thiocyanate that is further separated by capillary electrophoresis (CE) and determined spectrophotometrically at 200 nm. Cyanide is thus estimated indirectly from the result of thiocyanate quantification and moreover, it can be easily determined with sufficient sensitivity by means of CE apparatus equipped with common UV detector. The linear detection range for concentration versus peak area for the assay is from 15 to 500 microM (correlation coefficient 0.997) with a detection limit of 3 microM and a limit of quantitation 9 microM. The inter-day reproducibility of the peak area was below 3.2% and the inter-day reproducibility of the migration time below 0.1%. The method is relatively rapid, simple and can be easily automated. Moreover, only limited amount of the sample is required.

Aims: To study the bactericidal properties of the lactoperoxidase (LPER)-thiocyanate and soybean peroxidase (SBP)-thiocyanate systems at low pH, their efficiency for inactivation of Escherichia coli and Shigella in acidic fruit and vegetable juices, their effect on colour stability of the juices and interaction with ascorbic acid., Methods and Results: Three-strain cocktails of E. coli and Shigella spp. in selected juices were supplemented with the LPER or SBP system. Within 24 h at 20 degrees C, the LPER system inactivated both cocktails by > or = 5 log10 units in apple, 2-5 log10 units in orange and < or = 1 log10 unit in tomato juices. In the presence of SBP, browning was significant in apple juice and white grape juice, slight in pink grape juice and absent in orange or tomato juice. Ascorbic acid protected E. coli and Shigella against inactivation by the LPER system, and peroxidase systems significantly reduced the ascorbic acid content of juices., Conclusions: Our results suggest a different specificity of LPER and SBP for SCN-, phenolic substrates of browning and ascorbic acid in acidic juices. The LPER system appeared a more appropriate candidate than the SBP system for biopreservation of juices., Significance and Impact of the Study: This work may open perspectives towards the development of LPER or other peroxidases as biopreservatives in acidic foods.

Broccoli sulforaphane has received attention as a possible anticarcinogen. Sulforaphane analysis is difficult due to the lack of a chromophore for spectrometric detection. Hence, we developed a method for determining sulforaphane by using high-performance liquid chromatography (HPLC) coupled with an evaporative light-scattering detector (ELSD). Sulforaphane was extracted from acid-hydrolyzed broccoli samples, followed by solid-phase extraction and reversed-phase HPLC. Sulforaphane was detected by ELSD and concurrently identified by electrospray ionization time-of-flight mass spectrometry. The recovery of sulforaphane from broccoli samples was above 95%. The detection limit was 0.5 mug. The present method was sensitive enough to determine sulforaphane in mature broccoli, broccoli sprouts, and commercial broccoli products. Sulforaphane concentration in broccoli sprout (1153 mg/100 g dry weight) was about 10 times higher than that of mature broccoli (44-171 mg/100 g dry weight). Therefore, the broccoli sprout is recommended as a source of sulforaphane-rich products. In contrast, we found that sulforaphane could not be detected in most of broccoli products, suggesting present commercial broccoli products having low quality.

A pneumatic flow injection-tandem spectrometer system, without a delivery pump was used for the speciation of iron. In this system, the suction force of a pneumatic nebulizer of a flame absorption spectrometer was used for solution delivery through the manifold. The Fe(III) and total Fe concentrations were determined using thiocyanate ion in a UV-Vis spectrometer and a FAAS, respectively. The Fe(II) was determined by the difference. The calibration curves were linear up to 18 microg mL(-1) and 25 microg mL(-1) with detection limits of 0.09 microg mL(-1) and 0.07 microg mL(-1) for Fe(III) and Fe(II), respectively. The mid-range precision and accuracy were <2.5% and +/-3% for the two species, respectively, at a sampling rate of 120 h(-1). This system was applied for the determination of Fe(III) and Fe(II) in industrial water, natural water and spiked samples.

This study reports a sensitive kinetic spectrophotometric method for the determination of trace amounts of thiocyanate. In acidic solution, Methylene Blue (MB) is oxidized by bromate to form a colorless compound. The reaction is accelerated by trace amounts of thiocyanate and can be followed by measuring the absorbance at 664 nm. The absorbance of the reaction decreased with an increase in the reaction time. Under the optimum experimental conditions (0.56 M of sulfuric acid, 3.9 x 10(-5) M of MB, 3.0 x 10(-3) M of bromate, 180 s, 25 degrees C), thiocyanate can be determined in the range 5.0 - 180 ng/ml. The relative standard deviations (n = 8) are 2.81 and 1.43% for 10.0 and 150 ng/ml thiocyanate, respectively. The detection limit of this method is (3sigma) 3.8 ng/ml. This method was successfully applied to the determination of thiocyanate in real samples.

The adsorption of thiocyanate onto ZnCl2 activated carbon developed from coir pith was investigated to assess the possible use of this adsorbent. The influence of various parameters such as agitation time, thiocyanate concentration, adsorbent dose, pH and temperature has been studied. Adsorption followed second-order rate kinetics. Two theoretical adsorption isotherms, namely, Langmuir and Freundlich were used to describe the experimental results. The Langmuir adsorption capacity (Q0) was found to be 16.2 mg g(-1) of the adsorbent. The per cent adsorption was maximum in the pH range 3.0-7.0. pH effect and desorption studies showed that ion exchange and chemisorption mechanism are involved in the adsorption process. Thermodynamic parameters such as DeltaG0, DeltaH0 and DeltaS0 for the adsorption were evaluated. The negative values of DeltaH0 confirm the exothermic nature of adsorption. Effects of foreign ions on the adsorption of thiocyanate have been investigated. Removal of thiocyanate from ground water was also tested.

Broccoli is a food often consumed for its potential health-promoting properties. The health benefits of broccoli are partly associated with secondary plant compounds that have bioactivity; glucosinolates and phenolic acids are two of the most abundant and important in broccoli. In an effort to determine how variety, stress, and production conditions affect the production of these bioactive components broccoli was grown in the greenhouse with and without selenium (Se) fertilization, and in the field under conventional or organic farming procedures and with or without water stress. High-performance liquid chromatography/mass spectrometry was used to separate and identify 12 primary phenolic compounds. Variety had a major effect: There was a preponderance of flavonoids in the Majestic variety, but hydroxycinnamic esters were relatively more abundant in the Legacy variety. Organic farming and water stress decreased the overall production of phenolics. Se fertilization increased glucosinolates in general, and sulforaphane in particular, up to a point; above that Se fertilization decreased glucosinolate production. Organic farming and water stress also decreased glucosinolate production. These data show environmental and genetic variation in phenolics and glucosinolates in broccoli, and warn that not all broccoli may contain all health-promoting bioactive components. They further show that selection for one bioactive component (Se) may decrease the content of other bioactive components such as phenolics and glucosinolates.

Multiple components of broccoli, such as sulforaphane (Sf) and phenolic acids, may inhibit cancer. Additionally, broccoli can accumulate selenium (Se), and Se has been demonstrated to reduce the risk of cancer. Studies were conducted to determine whether enhancement of broccoli with Se would produce a plant with superior health benefits. Although increasing the concentration of Se in broccoli from <1.0 to >800 microg/g resulted in inhibition of colon cancer in rats, it also decreased the Sf content by >80% and inhibited production of most phenolic acids. The inclusion of Se-enriched broccoli in the diet of rats induced the activity of the selenoprotein thioredoxin reductase beyond the maximum activity induced by Se alone. These results emphasize the complex interactions of bioactive chemicals in a food; attempts to maximize one component may affect accumulation of another, and consumption of high amounts of multiple bioactive compounds may result in unexpected metabolic interactions within the body.

The success of engineered microbiological systems is evident in the global application of activated sludge communities to remediate coking effluent. However, there is a lack of understanding of the microbiology underlying treatment efficiency and stability. In this study, two functionally distinct activated sludge pools, treating the same effluent and operating under the same conditions, were examined to establish a relationship between overall diversity and/or functional diversity with respect to process stability. Molecular profiling, sequencing and RNA-based stable isotope probing were used to examine the bacterial diversity, general composition and functional composition of the most abundant members of the two communities. The inferior process stability in one of the pools could not be explained by reduced total bacterial diversity or evenness. RNA-based stable isotope probing revealed that both pools harboured an abundant phenol-degrading Acidovorax species, and that the pool of inferior stability accommodated an additional closely related phenol-degrading Acidovorax species at high abundance. These results are discussed in the context of deterministic and stochastic models of microbial community assembly.

Simultaneous determination of nitrite (NO2-), nitrate (NO3-), thiocyanate (SCN-) and uric acid in human saliva was performed by capillary zone electrophoresis using a coated capillary with reversed electroosmotic flow (EOF), using a 100 mM sodium phosphate buffer at pH 6.5 as a running buffer. Saliva samples were deproteinized with acetonitrile and filtered through a membrane filter. The important advantages of the reported method are: simple operation, short analysis time, minimal sample pre-treatment and sample dilution. In order to evaluate the daily variations of the anionic components, the concentrations were determined in the human saliva of four healthy volunteers upon waking and at 2qh intervals during a day.

Main Problem: Caries as a multifactorial process is influenced by salivary defense. Cluster analyses should give additional information on the role of salivary variables in relation to caries increment., Methods: Samples of unstimulated and stimulated whole saliva from 28 young adults (mean age 23.5+/-2.1 years) were analyzed for flow rate, pH and buffer variables, lysozyme, lactoferrin, peroxidase, thiocyanate, secretory immunoglobulin A, and total protein. The decayed, missing, and filled surfaces (DMFS) were recorded at baseline and after 4 years. Cluster analyses were executed on the basis of salivary data., Results: The mean caries increment (DeltaDMFS) over 4 years was 6.7+/-4.0 (range 1-16). In two-cluster processing, three out of four volunteers with low caries increments were grouped into one cluster. Only a few variables proved to be important for cluster characteristics., Conclusions: The results suggest that over 4 years (1) the volunteers with very low caries increment (DeltaDMFS=1) are classified always together, (2) these volunteers do not form a separate cluster by themselves, (3) low caries increment was related to higher salivary flow rate and lower levels of lysozyme and lactoferrin for unstimulated saliva and (4) the partial pressure of CO(2) was of importance in stimulated saliva.

Background & Objectives: Consumption of cyanogenic foods has been considered as one of the etiological factors in certain instances for the persistence of endemic goitre. The present study was undertaken to study the cyanogenic glucosides, glucosinolates and thiocyanate content in edible portion of certain selected plant foods of Indian origin. Further in vitro anti-thyroidal activity using raw, boiled and cooked extracts of these plants with and without excess iodide was also studied., Methods: Cyanogenic plant foods generally vegetables were collected from different areas of West Bengal and Tripura. Cassava was obtained from Meghalaya and Kerala and their cyanogenic glucosides, glucosinolates and thiocyanate were estimated. Thyroid peroxidase activity (TPO) of human thyroid was assayed from microsomal fraction following I3- from iodide. The anti-TPO activities of the plants were assayed after adding raw, boiled and cooked extracts in the assay medium with and without extra iodide. Relative antithyroidal potency of the plant extracts was also evaluated in terms of the concentration (IC50) necessary to produce 50 per cent inhibition of TPO activity. PTU equivalence of the plant foods was also determined., Results: Cabbage and cauliflower were rich in glucosinolates, bamboo shoot and cassava were rich in cyanogenic glucosides, mustard, turnip and radish were relatively rich in thiocyanate however all the constituents were present in each plant. Boiled extracts showed maximum inhibition of TPO activity followed by cooked and raw extracts. Excess iodide was found relatively effective for raw extract but less effective for boiled and cooked extracts in reversing anti-TPO activity. Inhibition constant (IC50) was found highest with bamboo shoot and least with cabbage., Interpretation & Conclusion: Raw, boiled and cooked extracts of the plants showed anti-thyroidal activity in vitro. Excess iodide reversed the anti-TPO activity to same extent but could not neutralise it.

Tobacco exposure can be assessed by the measurement of several markers in biological fluids. These markers are more or less specific for tobacco and the different methods to measure them out differ in terms of sensibility, specificity, ease of use and cost. The clinician prescribing a dosage for a patient has to take all these parameters into account to make an accurate choice. In this article, we have analysed the usefulness of the main biological tobacco markers in the follow-up of smokers and compared their methods of dosage. We propose several indications and point out the interest of relevant markers to realize objective measurements of smoking habits.
