The origin and differentiation of Tetrahymena pyriformis food vacuolar membranes has been studied by freeze-fracture electron microscopy. By measuring the temperature needed to induce the onset of lipid phase separation (as inferred by the appearance of particle-free regions in replicas) and calculating the changes in average intramembrane particle distribution, a distinct modification of the vacuolar membrane could be observed from the time of its formation from disk-shaped vesicles to its maturation before egestion of its indigestible contents. Whereas the nascent vacuolar membrane first showed signs of phase separation at 9 degrees C, this temperature rose to 14 degrees C in the completed vacuole and then, after lysosomal fusion, eventually declined to 12 degrees C. The average membrane particle density on the PF face increased from 761 +/- 219 to 1,625 +/- 350 per micron 2 during membrane differentiation. Like other membranes of the cell, the vacuolar membrane underwent adaptive changes in its physical properties in cells maintained for several hours at low temperature. This exposure to low temperature caused an equal effect in vacuoles formed before, during, or after the temperature shift-down. Normal changes in the properties of the vacuolar membrane may have some bearing on its programmed sequence of fusion reactions.

The level of calmodulin (CaM), a ubiquitous calcium-binding protein of eukaryotic cells was determined at different phases of the cell cycle in a synchronized Tetrahymena population. It was found that the concentration of CaM at G1 was approximately half of the concentration of S and this 2 x G1 level of CaM was maintained through the G2 and M stages of the cell cycle. To ascertain the role of CaM in the initiation of DNA synthesis, the cells were treated with trifluoperazine (TFP), a CaM antagonist, and EGTA (Ca2+-chelator) at the G1/S boundary. It was found that DNA synthesis was inhibited in these drug-treated cells. The uptake of the nucleotide precursor was not affected in TFP and EGTA treated cells, thus excluding the possibility of alteration in the membrane transport properties. Treatment with TFP failed to inhibit the synchronous mitotic division in Tetrahymena. The existence of a variable content of CaM through the cell cycle of Tetrahymena was demonstrated, suggesting the possible involvement of this Ca2+-binding protein in the nuclear DNA replication process.

Guanylate cyclase activity decreased during the division phase of heat-shock synchronized Tetrahymena pyriformis, strain GL. However, when Ca2+ was removed by EGTA to negate the effects of the Ca2+-binding protein (calmodulin), which is required for the full activity of guanylate cyclase in this organism, no significant change in the enzymatic activity was observed throughout the cell cycle. On the other hand, the reduced guanylate cyclase activity at division phase was associated with a decreased level of calmodulin content. These results suggest that fluctuations in guanylate cyclase activity during the cell cycle would be dependent on the concentration of calmodulin.

Cell volume and doubling time have been determined for exponentially growing Tetrahymena pyriformis cells in broth medium with and without glucose and in media made from these media by dilution with water. The cells tolerate media with dry weights from 105 down to 0.06 g/L. In the diluted media the cells have small volumes and the doubling time is increased. When the cell volume increase per time per cell in a given medium is expressed as a function of the cell volume in this same medium, a direct proportionality is found. From this equation the minimum cell volume of division competence (MVDC) can be found. It is 2,100 microns 3 for T. pyriformis at 28 degrees C. The lag period resulting from an upshift of exponentially growing cells from diluted media to more concentrated media is a function of the initial and resulting cell volumes and MVDC. The increase in cell volume per unit of time for a given cell depends on the dry weight of the medium. This parameter can be transformed to mass increase per cell surface area per time, which represents rate of nutrient uptake. When plotted against the dry weight of the media, a Michaelis-Menten-like curve is obtained with two Km values of 3.8 and 0.08 g/L with corresponding Vmax values of 20 and 4 ng/cm2.s. The low Km value (0.08 g/L) indicates that Tetrahymena is able to take up nutrients from highly diluted media. The high value of Vmax (20 ng/cm2.s) increases the ability of growth in more concentrated media.(ABSTRACT TRUNCATED AT 250 WORDS)

Fourteen nitrosoguanidine-induced mutations that bring about temperature-sensitive morphological abnormalities resulting from a specific effect on cell division have been isolated as heterozygous phenotypic assortants in Tetrahymena pyriformis syngen 1. Genetic analysis revealed all to be single-gene recessives. Detailed analysis of the kinetics of assortment for one of the mutated alleles revealed a rate (0.0104 pure lines per fission) consistent with that previously observed at other loci in this organism. The mutations fall into six complementation groups (mo1, mo2, mo3, mo6, mo8, and mo12). Homozygotes of mo2 are unconditionally expresed, while all alleles of mo1, mo6, mo8, and mo12 are heat sensitive for division arrest. At the mo3 locus two alleles are heat senstivie, one is primarily cold sensitive, while two are sensitive to both heat and cold. Two out of three combinations of different mo3 alleles show conventional Mendelian segregation of conditions of expression. Different alleles of mo1, mo3, mo8, and mo12 also manifest differences in penetrance at the restrictive temperature. Despite these differences involving expression, the abnormal phenotypes themselves are locus-specific and distinctive; in the one case (mo1a and mo1b) in which two alleles manifest somewhat different phenotypes, the F1 between them is intermediate. One additional recessive mutation (fat1) brings about a nonconditional lengthening of the cell cycle, with some arrest of cell division at the restrictive temperature. These findings demonstrate that selection of heterozygotes undergoing phenotypic assortment can be an effective method for obtaining substantial numbers of a desired class of temperature-sensitive mutations in T. pyriformis.

The effects of cytochalasin B on oral apparatus morphogenesis and cell division were studied in synchronized Tetrahymena pyriformis, strain WH-6 syngen 1. Cytochalasin B brought about the rapid arrest of oral apparatus primordium development when added prior to the completion of oral apparatus membranelle differentiation. Cells arrested in development did not divide. When cytochalasin B was added after this transition point, oral apparatus morphogenesis and cell division were completed. The effects of cytochalasin B could be reversed by washing it from the medium. Even though cytochalasin B (at 400 mug/ml) reduced protein synthesis by 30%, the data are consistent with the interpretation that cytochalasin B prevents an assembly process during the membranelle differentiation phase of oral apparatus development.

A single cell isolated from the sexual progeny of mutagenized parents gave rise to a clone of cells with an abnormal, conical shape. Breeding analysis revealed that this shape results from the action of a single recessive gene, co (conical). Homozygous mutant cells are shorter and wider than wild type cells, and have their widest point at a more posterior position. Nonetheless, cortical parameters such as number of ciliary rows, number of ciliary units within these rows, and positions of contractile vacuole pores remain essentially unchanged in conical cells, suggesting a considerable degree of mutual independence of pattern and form. Shape changes prior to cell division bring about some convergence in form of dividing conical and wild type cells. However, in conical cells the new oral apparatus and fission line form well posterior to the cell equator, so the opisthes are invariably smaller than proters. Macronuclei nonetheless undergo constriction at the normal central location, and the characteristic inequality in the DNA content of the two macronuclear division products is not increased by the conical condition. Generation times are, on the average, nearly the same in the two wild type daughter cells and in conical proters, while the small conical opisthes have generation times averaging one-third longer. This prolongation explains why population doubling times are always somewhat longer in cultures of conical cells than in parallel cultures of wild type cells. The unusually long generation times of conical opisthes allows for the compensation of initial size differences. Observations on shape changes in conjugating cells of various genotypic combinations suggest that the wild type gene product is not freely exchangeable across the conjugation bridge. The implications of the conical phenotype for problems of cellular patterning and positioning of organelle systems are considered in the discussion.

Ansamitocins inhibited cell division in Tetrahymena pyriformis W at low concentrations and caused cells to become more rounded. Exposure to ansamitocins for 5 h or more resulted in a burst of synchronous division at 100 min after removal of the antibiotics with a maximum division index of 70%. A second burst of synchronous division occurred 300 min after removal with a peak division index of 55%. The rounding of the cell shape was restored by the completion of the first division. When RNA or protein synthesis was blocked by chromomycin A3 or cycloheximide, the first division was suppressed. When DNA synthesis was blocked by methotrexate plus uridine, synchronous division still occurred; furthermore, the DNA content of T. pyriformis cells did not increase before the first division. These findings suggests that the first synchronous division requires RNA and protein synthesis but does not require DNA synthesis. Interference by ansamitocins with the fuction of microtubule systems in T. pyriformis is discussed.

Crocidolite asbestos fibers are rapidly ingested in large amounts by Tetrahymena. This varies little with incubation time of fibers in the culture medium or with dilution of the medium. The ingested fibers form large, oblong bundles in the cell rather than the normal spherical food vacuoles. In addition, crocidolite results in a rapid dose dependent induction of various minor morphological abnormalities. At 0.1-3 mg/ml these are observed in approximately one third of the population. Crocidolite is, however, not cytotoxic to Tetrahymena even in these high concentrations as measured by cell death and population doubling time. Phagocytosis and cytotoxicity are thus not related in this cell. Preliminary studies indicate that all the U.I.C.C. standard reference asbestos samples have largely similar effects but that their ability to induce abnormalities depends on the species of Tetrahymena used. For studies of the effects of asbestos Tetrahymena should be a suitable model system because the massive ingestion of fibers and its short generation time should facilitate rapid detection of adverse effects. Among such effects is the induction of heterogenous cell lines. This may involve modification of non-nucleic acid carried hereditary (cytotactic) information.

Progress in ciliatology and in allied fields may demystify ciliate phylogenetics. Concentration on hymenostomes (mainly Tetrahymena and Paramecium) may have obscured directional features of ciliate physiology in phylogenetic problems. Therefore, means are suggested for "domesticating" the presumptively primitive, predominantly marine, sand-dwelling gymnostomes having nondividing macronuclei. The prize quarry is the marine psammophile Stephanopogon whose homokaryotic condition may mark it as a living fossil. Eventual axenic cultivation of these "primitive" ciliates may be aided by use as food of easily grown photosynthetic prokaryotes, some isolated from the marine sulfuretum or adjacent aerobic muds and sands where "karyorelictid" ciliates flourish.

The rate of uridine uptake in Tetrahymena declines by an order of magnitude by two hours after shiftdown to a non-nutrient buffer. This alteration in uptake properties cannot be accounted for by an increase in the intracellular pool of uridine, an increase in apparent Km for uptake or a decline in the rate in which uridine is processed intracellularly. It is argued that the decrease in uridine uptake is due to a reduction in numbers of functional transport molecules exposed at the cell surface and is a reflection of a developmentally related cell surface transformation. In addition, the putative decline in functional transport molecules cannot be entirely explained by metabolic turnover of these molecules in the absence of replacement, nor does it require the synthesis of new protein. We discuss the possibility that a shift in equilibrium between accessible and inaccessible transporters is operating. Finally, a close correlation between conditions which elicit the transport alteration and those which allow the development of mating competency suggests that the two phenomena may be coordinately regulated.

Homopolar doublets of syngen 1, T. pyriformis, may be induced by treatment of conjugating pairs with immobilizing antiserum. These doublets have geometric properties and basal body populations generally indicative of separate autonomous integrative systems in the two halves. The duplex system, though metastable, is transformed through a process of "simplification" back to the simplex state. The transformation is not a single event, but a series involving changes at different times for different structures and processes; for the micronuclei and the macronuclei; for the capacity to generate two oral apparatuses through stomatogenesis and the capacity to develop them through oral replacement; for the structures at the anterior and posterior ends of the cell; for the numbers of ciliary rows and for the numbers of basal bodies which make up the rows. Although the two semicells composing a doublet are in important respects independent of each other, they are coordinated in significant ways. The positions of the contractile vacuole pores and their numbers depend not only on the number of ciliary rows in a semicell, but also on the number of ciliary rows in the opposing twin. Most notably, the probability for dual stomatogenesis, and hence the perpetuation of the doubled oral apparatus, depends on the symmetry of the semicells. The cell as a whole is maintained as an integrated unit over a prolonged interval as various aspects of duplex structure and function are progressively consolidated.

Schizopyrenus russelli, a free-living soil ameba, grows and encysts in the presence of bacteria. The encystment occurs with decline in the division rate. This is accompanied by incorporation of [U-14C] glucose into cyst cellulose. The degree of multiplication (but not of encystment) is a function of bacterial concentration. Berenil, a trypanocidal drug, while allowing excystment, completely inhibited multiplication of emerged amebae and their encystment. Addition of this drug after 24 hr, when amebae had gone into a phase of active division failed to check encystment, although it still inhibited further multiplication of the amebae. The findings suggest that a phase of cell division may be a prerequisite for encystment.

Cells of the ciliate Tetrahymena pyriformis GL overproduce and accumulate massive quantities of the heme intermediate, protoporphyrin IX. Protoporphyrin is localized intracellularly in discrete membranous compartments. The amount of porphyrin stored in the cell changes dramatically as cells progress through the growth cycle. Porphyrin overproduction is stimulated by delta-aminolevulinic acid, but only during the mid-stationary phase. Overproduction of protoporphyrin IX apparently results from an increase, late in the growth cycle, of activities subsequent to delta-aminolevulinic acid synthetase. Feedback inhibition in the pathway by accumulated protoporphyrin IX does not occur. The presence of Co2+ completely inhibits accumulation of protoporphyrin IX in a manner reversed by delta-aminolevulinic acid. Sn4+ stimulates protoporphyrin IX accumulation in the culture.

Labelling of surface membrane of living ciliates: Paramecium aurelia and Tetrahymena pyriformis with fluorescent compound--cycloheptaamylose-dansyl chloride complex (CDC) has been achieved. Fluorescence micrographs of the dried samples showed specific localization of CDC on the cell membrane without any intracellular penetration. On the contrary the ciliates which have been dead during labelling revealed a non-specific fluorescence of their whole bodies. Microspectrofluorimetric analysis of labelled Paramecium cells was performed with Leitz microspectrograph. Spectrum of fluorescence emission measured over the cell membrane level had maximum at 450 nm. Strikingly, the emission maximum of the cells dead at the moment of labelling was shifted 10 nm to a longer wavelength. The rate of photofading measured in this case was almost 3-fold higher than for the ciliates labelled as living ones. Fluorescence excitation spectra did not show any difference in the peak position. Thus CDC staining appears to be an useful method of supravital labelling of cell surface enabling also to distinguish--on the basis of spectral characteristics--the ciliates being alive from those dead at the moment of fluorochrome binding.

The concentration of 2-aminoethylphosphonic acid (AEP) in 96 h cultures of Tetrahymena pyriformis W was studied in order to apply it as an indicator in the assay of the relative nutritive value (RNV; protozoa population with test protein:protozoa population with whole-egg powder) of protein. Foodstuffs and food mixtures of different protein contents and qualities were used as test samples. 2. RNV values based on AEP determination (RNVAEP) were compared with corresponding values calculated from protozoa counts (RNVpc), as well as with biological value (BV) and net protein utilization (NPU) of the same proteins assayed on rats. 3. Both for foodstuffs and food mixtures highly significant correlations were found between RNVAEP and RNVpc, RNVAEP and both BV and NPU, and RNVpc and both BV and NPU. 4. AEP content in the protozoal suspension was preferred to cell count as a measure of growth response, since it took into account large differences in cell dimensions that were observed between cultures grown with different test proteins.

Synthesis of RNA in the macronucleus and appearance of RNA in the cytoplasm were studied in heat synchronized Tetrahymena pyriformis GL and compared to those found under conditions of logarithmic growth (28 C) and during heat shocks (34 C). In macronuclei of logarithmically growing cells precursors were processed to 2 rRNA species (25S and 17S). In addition, another RNA (15S), more homogeneous than the RNA (8-15S) in the cytoplasm, was observed in the macronucleus. Both 17S and 25S rRNA species were found in the cytoplasm, 17S rRNA appearing more rapidly than 25S rRNA. Synthesis of rRNA was suppressed at 34 C in cells subjected to heat synchronization; 8-15S RNA synthesis appeared to be inhibited to a lesser extent. During the time preceding the first synchronized division, the synthesis of rRNAs in the macronucleus slowly recovered. Early in the cycle, almost no newly synthesized rRNAs were extracted. By 30 min after the last heat shock (EH), most of the RNA synthesized was not identified as rRNA. By 60 min after EH, the pattern of RNA synthesis had not returned to that observed in logarithmically growing cells.

Histone fraction F1 has been isolated and purified from macronuclei of the ciliated protozoan, Tetrahymena pyriformis. In many respects, Tetrahymena F1 is similar to that of other organisms. It is the only Tetrahymena histone soluble in 5% perchloric acid or 5% trichloroacetic acid, has a higher molecular weight than any other Tetrahymena histone, is the histone most easily dissociated from Tetrahymena chromatin, and is susceptible to specific proteolytic cleavage. However, unlike F1 in all other organisms, Tetrahymena F1 is not the slowest-migrating histone fraction when analyzed by polyacrylamide gel electrophoresis at low pH. Tetrahymena F1 also exhibits unusual behavior in sodium dodecyl sulfate-containing polyacrylamide gels, migrating faster than calf thymus F1 at pH 10, and slower than calf thymus F1 at pH 7.6. Tetrahymena F1 was found to be highly phosphorylated in rapidly growing cells, suggesting that the relationship between cell replication and F1 phosphorylation previously observed in mammalian cells may extend to all eukaryotes. The observation that extensive F1 phosphorylation occurs in macronuclei, which divide amitotically, argues against a unique role for F1 phosphorylation in the process of chromosome condensation at mitosis.

The laser flow microphotometer for rapid cell size analysis is introduced. In contrast to known principles absorption is detected of a part of each cell only. Cell classification is achieved by pulse form analysis of absorption pulses. Cell lengths between 5 and 300 mum have been measured without changing the geometry of the flow system. Other ranges of size can be covered by changing the flow diameters. The principle can be applied to stained or unstained cells. No further preparation of samples is necessary. Examples of high speed measurements on different cell populations of unstained ciliated (Tetrahymena pyriformis), human blood and different pollen grains are given.

Hormonal imprinting takes place at the primary interaction between target cell and hormone, and alters cellular response to the hormone for lifetime (at the unicellular level in many subsequent generations). Imprinting induced in Tetrahymena cells by diiodotyrosine at the optimum temperature of 25 degrees C took effect on re-exposure to the hormone at 25 degrees C and 15 degrees C, but failed to take effect if the cells were first exposed to the hormone at 15 degrees C or 32 degrees C.

The recessive mp gene in syngen 1 (Species 1) of Tetrahymena pyriformis results in a characteristic set of abnormalities of the adoral membranelles, commonly including some extra membranellar segments (Kaczanowski, '75). The abnormal pattern is produced in about 10% of dividing cells, compared to 80% of cells undergoing oral replacement. Thus the percentage of abnormal cells increases during prolonged stationary phase. Abnormal oral replacement arises primarily for a separation of two kinetosomal fields: the field of kinety no. 1 (the K-1 field), and the anterior field originating from the displaced undulating membrane. Both sites of proliferation appear also during wild type morphogenesis, but they fuse early to form one integrated field (Frankel, '69). The prolonged separation of the subfields in mp/mp cells results in a separate assembly of adoral membranelles; however, the independently developing membranelles of the two subfields are united later to produce a single oral apparatus. A single gene-dependent factor apparently alters some property of the cortical layer, which in turn is responsible for a reduced integrity of the oral replacement primordium.

Tetrahymena pyriformis grown to logarithmic phase and then inoculated into either fresh or conditioned medium exhibited a shorter lag phase than ciliates taken from stationary cultures. The growth inhibiting substance(s), which is probably produced by the growing ciliates, is thermostable and can be adsorbed to activated charcoal.

Cells of the ciliate Tetrahymena pyriformis were suspended with carmine or graphite particles or with Halobacterium gas vesicles, all of which promote bubble formation in aqueous suspensions when tested with 10 atm and above (0.1-0.5 X 10(7) Pa) (carmine and graphite) or 25 atm and above (gas vesicles) of nitrogen supersaturations. All three particles were ingested, but only the gas vesicles promoted intracellular gas bubble formation if the cells containing them were nitrogen or methane saturated in a slow stepwise fashion prior to rapid decompression. Cell rupture did not occur until gas saturation pressures greater than 25 atm were used; this suggests that the ciliate pellicle and cytoplasm cannot resist the mechanical forces of an expanding gas phase induced by decompression from between 25 and 50 atm and thus provides an estimate of the physical strength of these cellular components. The inability of the ingested carmine, graphite, and collapsed gas vesicles to induce intracellular gas bubble formation suggests that the phagocytic process somehow altered them. This procedure may thus provide a tool for the study of early events in the digestive processes of ciliates.

Transformation of Tetrahymena pyriformis to a rapid-swimming (presumably dispersal) form can be induced by washing cells and suspending them in distilled H2O, Dryl's solution or 10 mM Tris. Transformation is possible with high efficiency in mass cultures of axenically grown cells within approximately 5 h at 30 C. The radically different phenotype produced during transformation is characterized by a more elongate body form, increased numbers of somatic basal bodies and cilia, a long caudal cilium and oral membranelles positioned beneath the cell surface. DNA quantities characteristic of G1, S, and G2 cells are found in these transformed ciliates, suggesting that achievement of a particular stage in the DNA-division cycle is not a prerequisite for transformation. Preliminary observations on cells belonging to syngens 2-12 indicate that they also have a capacity to form a caudal cilium, but that the amicronucleate strain GL-C does not. Possible relevance of the transformed phenotype for taxonomy of Tetrahymena is discussed.

The polypeptide hormones insulin, glucagon, thyrotropin (TSH), pregnant mare serum gonadotropin (PMSG) and adrenocorticotropin (ACTH) stimulated the growth of the Tetrahymena, and the non-hormone polypeptides (bovine serum albumin (BSA), protamine) had a similar effect. Re-exposure after 24 h accounted for a greater growth stimulation than pre-exposure alone in cultures treated with TSH and PMSG, and re-exposure after 7 days had such effect in all polypeptide-treated cultures. It follows that the non-hormone polypeptides had a similar imprinting potential to the polypeptide hormone. The non-hormone polypeptides were also able to cross-imprint for one another, i.e. pre-exposure to one enhanced the binding capacity of the cells for the other on re-exposure, and vice versa. A single treatment with a polypeptide hormone or a non-hormone polypeptide did in itself stimulate the growth of the Tetrahymena for as long as 1 week.
