107. Natural killer-like nonspecific tumor cell lysis mediated by specific ligand-activated Valpha14 NKT cells

Abstract

We have recently identified [Alpha]-galactosylceramide ([Alpha]-GalCer) as a specific ligand for an invariant V[Alpha]14/V[Beta]8.2 T cell receptor exclusively expressed on the majority of V[Alpha]14 NKT cells, a novel subset of lymphocytes. Here, we report that [Alpha]-GalCer selectively activates V[Alpha]14 NKT cells resulting in prevention of tumor metastasis. The effector mechanisms of the ligand-activated V[Alpha]14 NKT cells seem to be mediated by natural killer (NK)-like nonspecific cytotoxicity. Indeed, the cytotoxic index obtained by [Alpha]-GalCer-activated V[Alpha]14 NKT cells was reduced by the addition of cold target tumor cells or by treatment with concanamycin A, which inhibits activation and secretion of perforin, but not by mAbs against molecules involved in the NKT cell recognition and conventional cytotoxicity, such as CD1d, V[Beta]8, NK1.1, Ly49C, Fas, or Fas ligand. These results suggest that the ligand-activated V[Alpha]14 NKT cells kill tumor cells directly through a CD1d/V[Alpha]14 T cell receptor-independent, NK-like mechanism.

Abstract

[V.sub.Alpha] 14 NKT cells, originally defined as [NK1.1.sup.+] T cells(1), are a distinct lymphoid lineage expressing several characteristics that demarcate them from T cells, natural killer (NK) cells, and B [...], A lymphocyte subpopulation, the [V.sub.Alpha]14 natural killer T (NKT) cells, expresses both NK1.1 and a single in variant T cell receptor encoded by the [V.sub.Alpha] and [J.sub.Alpha].281 genesegments. Mice with a deletion of the [J.sub.Alpha]281 gene segment were found to exclusively lack this subpopulation. The [V.sub.Alpha] 14 NKT cell-deficient mice could no longer mediate the interieukin-12 (IL-12)-induced rejection of tumors. Although the antitumor effect of IL-12 was thought to be mediated through natural killer cells and T cells, [V.sub.Alpha]14 NKT cells were found to be an essential target of IL-12, and they mediated their cytotoxicity by an NK-like effector mechanism after activation with IL-12.

123. Essential requirement of an invariant Valpha14 T cell antigen receptor expression in the development of natural killer T cells

Abstract

NK[1.1.sup.+] T [natural killer (NK) T] cells express an invariant T cell antigen receptor [Alpha] chain (TCR[Alpha]) encoded by V[Alpha]14 and J[Alpha]281 segments in association with a limited number of V[Beta]s, predominantly V[Beta]8.2. Expression of the invariant V[Alpha]14/J[Alpha]281, but not V[Alpha], TCR in transgenic mice lacking endogenous TCR[Alpha] expression blocks the development of conventional T [Alpha][Beta] cells and leads to the preferential development of V[Alpha]14 NK T cells, suggesting a prerequisite role of invariant V[Alpha]14 TCR in NK T cell development. In V[Beta]8.2 but not V[Beta]3 transgenic mice, two NK T cells with different CD3[Epsilon] expressions, CD3[[Epsilon].sup.dim] and CD3[[Epsilon].sup.high], can be identified. CD3[[Epsilon].sup.high] NK T cells express surface V[Alpha]14/V[Beta]8 TCR, indicating a mature cell type, whereas CD3[[Epsilon].sup.dim] NK T cells express V[Beta]8 without V[Alpha]14 TCR and no significant CD3[Epsilon] expression (CD3[[Epsilon].sup.dim]) on the cell surface. However, the latter are positive for recombination activating gene (RAG-1 and RAG-2) mRNA, which are only expressed in the precursor or immature T cell lineage, and also possess CD3[Epsilon] mRNA in their cytoplasm, suggesting that CD3[[Epsilon].sup.dim] NK T cells are the precursor of V[Alpha]14 NK T cells.

Abstract

The majority of T lymphocytes start to develop at around day 15 of gestation (d15)-d17 in the thymus and comprise the peripheral repertoire characterized by the expression of polymorphic T-cell antigen receptors (TCRs). Contrary to these conventional T cells, a subset of T cells, called natural killer (NK) T cells (most of them expressing an invariant TCR encoded by the V[Alpha]14J[Alpha]281 gene with a 1-nt N-region), preferentially differentiates extrathymically and dominates the peripheral T-cell population at a high frequency (5% in splenic T cells and 40% in bone marrow T cells). Here, we investigated the development of NK T cells and found that the invariant V[Alpha][14.sup.+] TCR transcripts and the circular DNA created by V[Alpha]14 and J[Alpha]281 gene rearrangements can be detected in the embryo body at d9.5 of gestation and in the yolk sac and the fetal liver at d11.5-d13.5 of gestation, but not in the thymus, whereas T cells with V[Alpha][1.sup.+] TCR expression, a major population in the thymus, were not observed at these early stages of gestation. Fluorescence-activated cell sorter analysis also demonstrated that there exist [CD.sup.3+] + [Alpha][[Beta].sup.+] T cells, almost all of which are V[Alpha]14/V/[Beta][8.sup.+] [NK.sup.+] T cells, during early embryogenesis. To our knowledge, this demonstrates for the first time that a T lymphocyte subset develops in extrathymic tissues during the early stages of embryogenesis.

DNA linkage analysis of recombinant mice that are negative for the expression of invariant V-alpha14+ T-cell antigen receptors (TCR) proves that major histocompatibility complex (MHC) class-I-like molecules associated with beta2-microglobulin and delinked with the MHC on chromosome 17 are essential for positive selection of invariant V-alpha14+ T cells. The differentiation of V-alpha14+ T cells and alpha beta TCR+ T cells proceeds through different pathways.

Abstract

Invariant natural killer T (iNKT) cells are a subset of innate-like T cells that act as important mediators of immune responses. In particular, iNKT cells have the ability to immediately produce large amounts of IFN-γ upon activation and thus initiate immune responses in various pathological conditions. However, molecular mechanisms that control IFN-γ production in iNKT cells are not fully understood. Here, we report that basic helix-loop-helix transcription factor family, member e40 (Bhlhe40), is an important regulator for IFN-γ production in iNKT cells. Bhlhe40 is highly expressed in stage 3 thymic iNKT cells and iNKT1 subsets, and the level of Bhlhe40 mRNA expression is correlated with Ifng mRNA expression in the resting state. Although Bhlhe40-deficient mice show normal iNKT cell development, Bhlhe40-deficient iNKT cells show significant impairment of IFN-γ production and antitumor effects. Bhlhe40 alone shows no significant effects on Ifng promoter activities but contributes to enhance T-box transcription factor Tbx21 (T-bet)-mediated Ifng promoter activation. Chromatin immunoprecipitation analysis revealed that Bhlhe40 accumulates in the T-box region of the Ifng locus and contributes to histone H3-lysine 9 acetylation of the Ifng locus, which is impaired without T-bet conditions. These results indicate that Bhlhe40 works as a cofactor of T-bet for enhancing IFN-γ production in iNKT cells.
