111. Phase characteristics of self-excited chatter in cutting

Abstract

The behavior of the self-excited chatter in cutting was investigated from the space phase point of view; the space phase is defined as that between outer and inner modulation. Records of oscillation for the horizontal motion of the work and those for the horizontal component of the cutting force were stored in a newly developed measurement system. The amplitude of the vibration increases for decreasing cutting speed and vice versa, the amplitude for the cutting force does not increase as the amplitude of the work motion does. Amplified and damped amplitudes for the decreasing and the increasing cutting speed are shown to be closely related to decreasing and increasing space phases, respectively. The mechanism of this phenomenon was analyzed on the complex plane by taking the cutting dynamics and the structural compliance into account. Attempts to suppress the chatter amplitude were conducted on the basis of these phenomena.

Abstract

It is important to analyze the characteristics of magnetic flux and eddy current on the inner part of an electric machine in order to raise the efficiency of the machine. In general, characteristics of those are investigated using numerical analysis. In this study, we present a novel measuring method of eddy currents on the surface of the sheet iron using a search needle. Abstract

Objectives: The aim of this study was to evaluate the applicability of genotyping of the ABCG2 gene using MALDI‐TOF MS and to estimate the allele frequency in the Japanese population. Background: Jr (a−) phenotype has a prevalence of approximately 0·05% among Japanese blood donors; DNA‐based genotyping was conducted to investigate the molecular basis of the Jr (a−) phenotype along with serological typing. To detect all SNPs of the ABCG2 gene, a high‐throughput SNP genotyping platform is needed. Methods: Overall, 1004 Jr (a−) blood samples were collected from blood donors in Japan and pre‐genotyped. To detect the SNPs of the ABCG2 gene using MALDI‐TOF MS, polymerase chain reaction and unextend primer were designed. In total, 205 Jr (a−) samples were genotyped using MALDI‐TOF MS analysis. Results: The SNPs of 1004 Jr (a−) samples were identified using the HRM analysis and DNA sequencing, and 799 of 1004 (80%) Jr (a−) samples had the homozygous for c.376 T. The designed primers for MALDI‐TOF MS perfectly detected the SNPs of the ABCG2 gene. A total of 205 Jr (a−) samples were genotyped using MALDI‐TOF MS. Calling failures occurred in only two samples with the mutations c.736CT to c.376C and c.421C to c.421CA. The concordance rate between the pre‐genotyped and MALDI‐TOF MS‐based genotyping results was very high (99·02%) for all ABCG2 alleles. Conclusions: Jr (a‐) Japanese donors had almost the homozygous for c.376 T. However, detections of more than 20 SNPs of the ABCG2 gene for the JR blood group genotyping are needed. MALDI‐TOF MS‐based genotyping was highly concordant with the pre‐genotyped results for all ABCG2 alleles. [ABSTRACT FROM AUTHOR]

Abstract

Arterial thrombosis in the rabbit was established as a novel model to evaluate the effects of antithrombotic agents. Endothelial injury was produced by applying electrical stimulation to the femoral artery. The process of primary endothelial injury, and subsequent platelet activation and fibrin formation were confirmed by electron microscopy. In this model, vessel occlusion occurred within 30 min after stimulation without changes in heart rate and blood pressure. Using this model, several agents were evaluated for their antithrombotic activities: aspirin (30 mg/kg, p.o.), ticlopidine (10-100 mg/kg, p.o.), heparin (300 unit/kg, i.v.), PPACK (10-33 micrograms/kg/min, i.v.), WEB-2347 (1 mg/kg, p.o.) and nicardipine (10 micrograms/kg, i.v.). Fifty per cent decrease in vessel temperature (T1/2), assessed by a thermic probe, averaged 15.1 +/- 1.2 (n = 11, p.o.) and 15.6 +/- 1.9 min (n = 8, i.v.) in the vehicle groups, and this was significantly prolonged by aspirin (23.0 +/- 2.6 min), ticlopidine at a dose of 100 mg/kg (24.6 +/- 2.5 min), heparin (27.2 +/- 2.8 min) and PPACK at a dose of 33 micrograms/kg (30.0 min). However, WEB-2347 and nicardipine were without effect. The effect of aprosulate, a new class of polyanion with anticoagulant activity, was further examined. Aprosulate (1-30 mg/kg, i.v.) inhibited thrombus formation in a dose-dependent manner. These results show that acute occlusive thrombus can be readily and reproducibly formed in the rabbit femoral artery and suggest that this thrombus formation depends on the activation of both platelets and blood coagulation. The merit of this model lies in its simplicity for evaluating the antithrombotic effects of antiplatelet and anticoagulant agents and is therefore expected to be extensively used in the future.

147. High-resolution melting analysis as an alternative method for human neutrophil antigen genotyping

Abstract

Human neutrophil antigen (HNA)-typed granulocyte panels are widely used to screen for the presence of HNA antibodies and to determine antibody specificity. Many laboratories screen donors for HNA genotypes using low-throughput methods such as allele-specific polymerase chain reaction (PCR), PCR–restriction fragment–length polymorphism, and multiplex PCR. In the present study, we used a high-resolution melting (HRM) analysis to determine HNA genotypes. For the HRM analysis, purified genomic DNA samples were amplified via PCR with HNA-specific primers. Nucleotide substitutions in genes encoding HNAs were differentiated on the basis of the HRM curves, and the results of HRM and DNA sequencing analyses were determined to be in complete agreement. The gene frequency of HNA-1a, -1b, -1c, -3a, -3b, -4a, -4b, -5a, and -5b in the Japanese population was consistent with the previous reports. Our results suggest that HRM analysis can be used for genotyping HNA antigens determined by single nucleotide substitutions. Immunohematology2015;31:7–13.
