102. LOCATION DISCOVERY OF VECTOR DATA UPDATE DRIVEN BY MAP IMAGE TILES FOR NATIONAL GEO-INFORMATION SERVICE PLATFORM

Abstract

With the popularization of geographic information data applications, new requirements are put forward for the rapid update of vector data. The overall update of vector data is expensive and time-consuming. Therefore, we need to use various technical means to intelligently sense the changes of geographical entities and realize the active monitoring of the changes of vector data. In this paper, the building layers that are closely related to human beings and gradually become active geographic entities with the urbanization process are selected to monitor the location of the vector data to be updated. This paper first trains the model using single building layer tiles and image tiles. Then, based on the trained model, the location where the building layer tiles are inconsistent with the image tiles is found in the area to be detected. According to different situations, we set different thresholds to find the position to be updated in the vector data. After manual discrimination, the overall accuracy of the method proposed in this paper is 89%. This paper provides new insights into the update discovery of vector data. In addition, by further improving the boundary accuracy of extracted buildings, the extracted building results can be directly applied to the fusion update of vector data.

106. The chloroplast genome of Cephalanthera nanchuanica (Orchidaceae): comparative and phylogenetic analysis with other Neottieae species

Abstract

Abstract Background Cephalanthera nanchuanica is a terrestrial orchid species and has been red listed as a second-grade protected plant due to its limited distributions in China. Initially classified within a monotypic genus Tangtsinia, this species was later reassigned to Cephalanthera based on morphological and molecular data. However, previous phylogenetic analyses of Cephalanthera using several segment sequences exhibited a low discriminatory power in delineating its relationships. Results In this study, we characterized and comparatively analyzed the complete chloroplast (cp) genome of C. nanchuanica with those of six previously reported Cephalanthera species. Our findings revealed that the cp genome of C. nanchuanica had the typical quadripartite structure, with a size of 161,365 bp and a GC content of 37.27%. A total of 113 unique genes were annotated, among which nearly half of protein-encoding genes (RSCU > 1) showed a preference in codon usage. No structural rearrangements were observed among the cp genomes of Cephalanthera species, except for C. humilis, which displayed structural alterations due to gene loss, relocation, and inverted repeat (IR) expansion/contraction. The cp genomes of Cephalanthera species were highly conserved, with only a small number of SSRs detected, most of which preferred A/T bases. Comparative analysis of cp genomes indicated that IR and coding regions were less divergent than single copy and non-coding regions and eight mutational hotspots were identified. Phylogenetic analysis suggested that the tribe Neottieae was a monophyletic group, divided into five clades. Palmorchis was the earliest-diverging lineage, followed by Cephalanthera. Diplandrorchis was deeply nested within Neottia, forming a clade. Aphyllorchis and Limodorum formed another clade, sister to Epipactis. Within the Cephalanthera clade, C. nanchuanica was sister to C. falcata with a strong support. Conclusions This study demonstrated that the cp genome characters of C. nanchuanica are highly similar to those of other Cephalanthera species, except for the mycoheterotrophic species C. humilis. Although the cp genomes of Cephalanthera species (excluding C. humilis) exhibited conservation in genome structure and sequence, SSR repeats and mutational hotspots were identified, which could potentially serve as as molecular markers for distinguishing Cephalanthera species. The phylogenetic analysis based on the protein-coding genes provided high-resolution support for the infrageneric classification. Therefore, cp genome data will be instrumental in resolving the phylogeny of the genus Cephalanthera.

Abstract

Abstract Lower limb arterial occlusive disease is treated with intraluminal devices, such as paclitaxel (PTX)-coated balloons (PCBs); however, post-procedural restenosis remains a significant challenge. NLRP3 activation is known to play a significant role in atherosclerosis, but its involvement in restenosis following PCB intervention remains to be investigated. We identified that NLRP3 was differentially expressed in lower-limb arterial tissues sourced from healthy controls and patients with arterial occlusive disease. Through cell experiments, we confirmed that PTX is involved in the activation of NLRP3. Subsequently, we demonstrated that NLRP3 activation promotes the proliferation and migration of vascular smooth muscle cell (VSMC), thereby reducing their sensitivity to PTX. NLRP3 activation also stimulates the secretion of the inflammatory cytokine interleukin IL-1β. RNA sequencing of IL-1β-treated VSMC revealed the upregulation of BRD4 and LIN9. Further mechanistic investigations confirmed that IL-1β facilitates BRD4 recruitment, leading to enhanced LIN9 expression. The transcription factor LIN9 binds to the promoter region of the cell-cycle regulator AURKA, thereby promoting its transcription and subsequently upregulating the expression of the cell proliferation-associated molecule FOXM1. These processes ultimately mediate the proliferation, migration, and PTX resistance of VSMC. Additionally, we discovered that JQ1 inhibited the overexpression of the above molecules, and exhibited a synergistic effect with PTX. Our conclusions were validated through in vivo experiments in Sprague-Dawley rats. Collectively, our findings provide insights into the molecular mechanisms underlying restenosis following PCB therapy, and suggest that the combined use of JQ1 and PTX devices may represent a promising therapeutic strategy. Abstract

In this paper, the enthalpy change of 1035 steel during phase transformation at different cooling rates was measured by differential scanning calorimetry (DSC). The activation energy of the phase transformation process was calculated by the equal conversion method, and the mechanism function of the process was also determined. The results shown that the value of the activation energy of phase transformation process varied with the transformation fraction, and the mechanism functions of transformation process are different in different temperature ranges, which are –ln(1–α)1/3 for ≥ 645 °C –ln(1–α) 1/2 for 660-655 °C and –ln(1–α) for 650-645 °C respectively.

Abstract

Background: The Omicron wave of COVID-19 in England peaked in January 2022 resulting from the rapid transmission of the Omicron BA.1 variant. We investigate the spread and dynamics of the SARS-CoV-2 epidemic in the population of England during February 2022, by region, age and main SARS-CoV-2 sub-lineage. Methods: In the REal-time Assessment of Community Transmission-1 (REACT-1) study we obtained data from a random sample of 94,950 participants with valid throat and nose swab results by RT-PCR during round 18 (8 February to 1 March 2022). Findings: We estimated a weighted mean SARS-CoV-2 prevalence of 2.88% (95% credible interval [CrI] 2.76–3.00), with a within-round effective reproduction number (R) overall of 0.94 (0·91–0.96). While within-round weighted prevalence fell among children (aged 5 to 17 years) and adults aged 18 to 54 years, we observed a level or increasing weighted prevalence among those aged 55 years and older with an R of 1.04 (1.00–1.09). Among 1,616 positive samples with sublineages determined, one (0.1% [0.0–0.3]) corresponded to XE BA.1/BA.2 recombinant and the remainder were Omicron: N=1,047, 64.8% (62.4–67.2) were BA.1; N=568, 35.2% (32.8–37.6) were BA.2. We estimated an R additive advantage for BA.2 (vs BA.1) of 0.38 (0.34–0.41). The highest proportion of BA.2 among positives was found in London. Interpretation: In February 2022, infection prevalence in England remained high with level or increasing rates of infection in older people and an uptick in hospitalisations. Abstract

Trichloroacetic acid is a common nonvolatile byproduct of drinking water disinfection and poses carcinogenic risks to the human body. In this study, four types of nano-oxide@microcrystalline-cellulose-based adsorbents (P25@microcrystalline cellulose, SiO₂@microcrystalline cellulose (MCC-g-GMA@SiO2), Fe3O4@microcrystalline cellulose, and Fe2O3@microcrystalline cellulose) were successfully prepared by the pre-radiation grafting-embedding method. Subsequently, their ability to remove trichloroacetic acid from drinking water was investigated. The micro zonation morphology and surface properties of the materials were characterized and tested using Fourier transform infrared spectroscopy (FTIR), X-ray diffraction spectroscopy (XRD), Thermogravimetric (TG) analysis, and Scanning electron microscopy (SEM) characterization methods. The effects of monomer concentration, embedding concentration, and absorbed dose on the weight gain rate were systematically investigated. A complete static adsorption equilibrium curve was obtained on the basis of the results of adsorption experiments of four buried nano-oxides. The performance of SiO2@microcrystalline cellulose was found to be significantly higher than that of the other three adsorbents. When the volum percentage of monomer concentration was 30%, the mass percentage of embedding concentration was 4%, and the absorbed dose was 60 kGy, the removal rate of trichloroacetic acid in drinking water reached 83.27%. This series of adsorbent materials present significant potential for practical application in drinking water purification.

The TAMs-like macrophages (TAMs-M&phis;s) were triggered with the help of 25 ng/mL hM-CSF and 50% NBCM. EdU assay wound healing assay, transwell assay, and TUNEL assay, as well as flow cytometry, were carried out to study the effect of HMGA2 or TAMs on the functioning of HCC cells.Results: HCC tumor tissues were detected with upregulated HMGA2 and TAMs markers (CD68, CD163, and CD204); in addition, HMGA2 was positively correlated with TAMs markers. The proliferation, migration, and invasion of HepG2 cells were also observed to be stimulated by HMGA2. Remarkably, cell apoptosis was not affected by upregulated HMGA2, but HMAG2 inhibition was observed to intensify it. Also, the release of CSF1 was observed to be amplified by HMGA2. HMGA2-overexpressed-HepG2 cells promoted the migrating abilities of both M0-M&phis;s and TAMs-M&phis;s but were suppressed by HMGA2 down-regulated HepG2 cells. 135. Twin peaks: the Omicron SARS-CoV-2 BA.1 and BA.2 epidemics in England

Abstract

BACKGROUND Rapid transmission of the SARS-CoV-2 Omicron variant has led to record-breaking incidence rates around the world. Sub-lineages have been detected in many countries with BA.1 replacing Delta and BA.2 replacing BA.1. METHODS The REal-time Assessment of Community Transmission-1 (REACT-1) study has tracked SARS-CoV-2 infection in England using RT-PCR results from self-administered throat and nose swabs from randomly-selected participants aged 5+ years. Rounds of data collection were approximately monthly from May 2020 to March 2022. RESULTS In March 2022, weighted prevalence was 6.37% (N=109,181), more than twice that in February 2022 following an initial Omicron peak in January 2022. Of the lineages determined by viral genome sequencing, 3,382 (99.97%) were Omicron, including 346 (10.2%) BA.1, 3035 (89.7%) BA.2 and one (0.03%) BA.3 sub-lineage; the remainder (1, 0.03%) was Delta AY.4. The BA.2 Omicron sub-lineage had a growth rate advantage (compared to BA.1 and sub-lineages) of 0.11 (95% credible interval [CrI], 0.10, 0.11). Prevalence was increasing overall (reproduction number R=1.07, 95% CrI, 1.06, 1.09), with the greatest increase in those aged 55+ years (R=1.12, 95% CrI, 1.09, 1.14) among whom estimated prevalence on March 31, 2022 was 8.31%, nearly 20-fold the median prevalence since May 1, 2020. CONCLUSIONS We observed unprecedented levels of SARS-CoV-2 infection in England in March 2022 and an almost complete replacement of Omicron BA.1 by BA.2. The high and increasing prevalence in older adults may increase hospitalizations and deaths despite high levels of vaccination. (Funded by the Department of Health and Social Care in England.)

136. Auditory-motor interactions during speech perceptions and production

Abstract

Speech perception and production are two central aspects of human language. It remains unclear, however, how sensory and motor systems interact with each other during these two processes. The overall aim of this thesis was to investigate auditory-motor interactions during both speech perception and production. There were two lines of research. The first explored sensorimotor adaptation in speech production, which plays an important role in the development and maintenance of fluent speech production. In study 1 (chapter 3), I examined the compensatory responses of speakers when they experienced perturbations to the auditory feedback of their own speech. The results showed feedback perturbation induced changes in subsequent speech production that were evident not only in the shifted formant, but also in the unaltered one. Moreover, such combined changes resulted in moving the produced speech token closer to a known pattern of speech production for an existing vowel category. This study confirmed the utility of this specific experimental paradigm, and that the apparatus used to apply real-time feedback perturbation was effective. It allowed the development of additional research questions to be tackled using the speech feedback perturbation paradigm. Following the successful implementation of the paradigm in study 1, I carried out a brain stimulation study (chapter 4) exploring the neural basis of sensorimotor adaptation in speech using transcranial magnetic stimulation (TMS), which temporarily inhibits function in stimulated brain areas. This allowed testing of causal hypotheses concerning the role of different parts of the motor system in speech motor adaptation. Here, I aimed to determine whether temporary disruption of the articulatory representation in left primary motor cortex (M1) by repetitive transcranial magnetic stimulation (rTMS) impairs sensorimotor adaptation in speech. Results showed that disruption of tongue, but not hand representation in left M1 almost abolished the expected compensatory response for the upwards F1 shift in auditory feedback, namely a decrease in F1 frequency alongside an increase in F2 frequency. This is the first study to address the key question as to whether cortical motor areas that control movements of articulators causally contribute to speech motor adaptation. The second line of research explored in this thesis was focused on the asymmetry of auditory-motor speech processing in tonal and non-tonal language speakers (study 3, chapter 5). To achieve this, I temporarily disrupted activity in either the left or right motor cortex using the same form of rTMS used in Study 2 and measured the impact of these disruptions on auditory processing of phoneme and tone changes in sequences of syllables using electroencephalography (EEG). I found that the effect of motor disruptions on processing of tone changes differed in tonal and nontonal language speakers: disruption of the right speech motor cortex affected processing of tone changes in non-tonal but not tonal language speakers, whereas disruption of the left speech motor cortex significantly impaired processing of tone changes in tonal language speakers. In contrast, disruption of the speech motor cortex in the left, but not the right hemisphere, impaired processing of phoneme changes in both language groups equally. For the first time, I provide causal evidence that auditory-motor speech processing asymmetries are shaped by the functional role of the acoustic cues in the listener’s native language. Overall, the work presented in this thesis emphasizes the importance of understanding the interactions between auditory and motor systems when investigating speech perception and production. It also calls for more comparative studies between speakers of different languages, since ignoring the diversity of spoken languages can to lead to an incomplete understanding of the neurobiology of speech and language.

We estimated an R additive advantage for BA.2 (vs BA.1 or BA.1.1) of 0.40 (0.36–0.43). The highest proportion of BA.2 among positives was found in London. Abstract

Rapid transmission of the SARS-CoV-2 Omicron variant has led to record-breaking case incidence rates around the world. Since May 2020, the REal-time Assessment of Community Transmission-1 (REACT-1) study tracked the spread of SARS-CoV-2 infection in England through RT-PCR of self-administered throat and nose swabs from randomly-selected participants aged 5 years and over. In January 2022, we found an overall weighted prevalence of 4.41% (n = 102,174), three-fold higher than in November to December 2021; we sequenced 2,374 (99.2%) Omicron infections (19 BA.2), and only 19 (0.79%) Delta, with a growth rate advantage for BA.2 compared to BA.1 or BA.1.1. Prevalence was decreasing overall (reproduction number R = 0.95, 95% credible interval [CrI], 0.93, 0.97), but increasing in children aged 5 to 17 years (R = 1.13, 95% CrI, 1.09, 1.18). In England during January 2022, we observed unprecedented levels of SARS-CoV-2 infection, especially among children, driven by almost complete replacement of Delta by Omicron.

Abstract

The SARS-CoV-2 pandemic has been characterised by the regular emergence of genomic variants. With natural and vaccine-induced population immunity at high levels, evolutionary pressure favours variants better able to evade SARS-CoV-2 neutralising antibodies. The Omicron variant (first detected in November 2021) exhibited a high degree of immune evasion, leading to increased infection rates worldwide. However, estimates of the magnitude of this Omicron wave have often relied on routine testing data, which are prone to several biases. Using data from the REal-time Assessment of Community Transmission-1 (REACT-1) study, a series of cross-sectional surveys assessing prevalence of SARS-CoV-2 infection in England, we estimated the dynamics of England's Omicron wave (from 9 September 2021 to 1 March 2022). We estimate an initial peak in national Omicron prevalence of 6.89% (5.34%, 10.61%) during January 2022, followed by a resurgence in SARS-CoV-2 infections as the more transmissible Omicron sub-lineage, BA.2 replaced BA.1 and BA.1.1. Assuming the emergence of further distinct variants, intermittent epidemics of similar magnitudes may become the 'new normal'.

Abstract

The unprecedented rise in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections during December 2021 was concurrent with rapid spread of the Omicron variant in England and globally. We analyzed the prevalence of SARS-CoV-2 and its dynamics in England from the end of November to mid-December 2021 among almost 100,000 participants in the REACT-1 study. Prevalence was high with rapid growth nationally and particularly in London during December 2021, with an increasing proportion of infections due to Omicron. We observed large decreases in swab positivity among mostly vaccinated older children (12 to 17 years) relative to unvaccinated younger children (5 to 11 years), and in adults who received a third (booster) vaccine dose versus two doses. Our results reinforce the importance of vaccination and booster campaigns, although additional measures have been needed to control the rapid growth of the Omicron variant.

BACKGROUND: The Omicron wave of COVID-19 in England peaked in January 2022 resulting from the rapid transmission of the Omicron BA.1 variant. We investigate the spread and dynamics of the SARS-CoV-2 epidemic in the population of England during February 2022, by region, age and main SARS-CoV-2 sub-lineage. METHODS: In the REal-time Assessment of Community Transmission-1 (REACT-1) study we obtained data from a random sample of 94,950 participants with valid throat and nose swab results by RT-PCR during round 18 (8 February to 1 March 2022). FINDINGS: We estimated a weighted mean SARS-CoV-2 prevalence of 2.88% (95% credible interval [CrI] 2.76-3.00), with a within-round effective reproduction number (R) overall of 0.94 (0·91-0.96). While within-round weighted prevalence fell among children (aged 5 to 17 years) and adults aged 18 to 54 years, we observed a level or increasing weighted prevalence among those aged 55 years and older with an R of 1.04 (1.00-1.09). Among 1,616 positive samples with sublineages determined, one (0.1% [0.0-0.3]) corresponded to XE BA.1/BA.2 recombinant and the remainder were Omicron: N=1047, 64.8% (62.4-67.2) were BA.1; N=568, 35.2% (32.8-37.6) were BA.2. Abstract

Background: Resistant hypertension is associated with increased cardiovascular risk. The endothelin pathway has been implicated in the pathogenesis of hypertension, but it is currently not targeted therapeutically, thereby leaving this relevant pathophysiological pathway unopposed with currently available drugs. The aim of the study was to assess the blood pressure lowering efficacy of the dual endothelin antagonist aprocitentan in patients with resistant hypertension. Methods: PRECISION was a multicentre, blinded, randomised, parallel-group, phase 3 study, which was done in hospitals or research centres in Europe, North America, Asia, and Australia. Patients were eligible for randomisation if their sitting systolic blood pressure was 140 mm Hg or higher despite taking standardised background therapy consisting of three antihypertensive drugs, including a diuretic. The study consisted of three sequential parts: part 1 was the 4-week double-blind, randomised, and placebo-controlled part, in which patients received aprocitentan 12·5 mg, aprocitentan 25 mg, or placebo in a 1:1:1 ratio; part 2 was a 32-week single (patient)-blind part, in which all patients received aprocitentan 25 mg; and part 3 was a 12-week double-blind, randomised, and placebo-controlled withdrawal part, in which patients were re-randomised to aprocitentan 25 mg or placebo in a 1:1 ratio. The primary and key secondary endpoints were changes in unattended office systolic blood pressure from baseline to week 4 and from withdrawal baseline to week 40, respectively. Secondary endpoints included 24-h ambulatory blood pressure changes. The study is registered on ClinicalTrials.gov, NCT03541174. Findings: The PRECISION study was done from June 18, 2018, to April 25, 2022. 1965 individuals were screened and 730 were randomly assigned.
