NADH-quinone oxidoreductase 1 (NQO1) modulates cellular NAD(+)/NADH ratio which has been associated with the aging and anti-aging mechanisms of calorie restriction (CR). Here, we demonstrate that the facilitation of NQO1 activity by feeding beta-lapachone (beta L), an exogenous NQO1 co-substrate, prevented age-dependent decline of motor and cognitive function in aged mice. beta L-fed mice did not alter their food-intake or locomotor activity but did increase their energy expenditure as measured by oxygen consumption and heat generation. Mitochondrial structure and numbers were disorganized and decreased in the muscles of control diet group but those defects were less severe in beta L-fed aged mice. Furthermore, for a subset of genes associated with energy metabolism, mice fed the beta L-diet showed similar changes in gene expression to the CR group (fed 70% of the control diet). These results support the potentiation of NQO1 activity by a beta L diet and could be an option for preventing age-related decline of muscle and brain functions.

The six-party process aimed at North Korea's denuclearization has long been stalled since the Six-Party Talks (SPT) failed to agree on a verification protocol in early December 2008. North Korea now wants to be recognized as a nuclear state, and is unlikely to abandon its nuclear weapons without security guarantees provided by the U.S. Some scholars argue that the Six- Party Talks have outlived their usefulness, while others maintain that the SPT are still useful. The author has argued over the years that the six-party process remains alive and useful as the best means of resolving North Korea's nuclear issue to achieve North Korea's denuclearization, which is a pre-requisite for building a peace regime on the Korean peninsula and regional peace in Northeast Asia. The author discusses the current status of the Six-Party Talks, which have been stalled for over two years, analyzing the major events on the Korean peninsula as key obstacles to the restoration of the six-party process. A Korean peninsula peace regime has not been established yet over a half century by replacing the Korean armistice agreement with a peace treaty. The two approaches to a Korean peninsula peace regime building are required: inter-Korean and international approaches. The two Koreas at the inter-Korean and international levels should play key roles in building inter-Korean confidence-building measures by reducing tension through inter-Korean reconciliation and economic cooperation and in a Korean peace forum involving the U.S., China and two Koreas under the Sept.19/Feb.13 joint agreements by concluding a Korean peninsula peace treaty to replace the armistice agreement. A peace regime can be institutionalized by implementing the inter-Korean basic agreement (1992) through inter-Korean cooperation and by concluding a Korean peninsula peace treaty at the forthcoming four-party Korean peace forum.The author makes three major arguments. First, the two Koreas and the four powers need to agree on a comprehensive roadmap for the Korean peninsula peace regime building. The inter-Korean and international approaches to peace regime building should be considered in parallel. Second, the North Korean nuclear issue should be first resolved peacefully through the six-party talks. Third, the two Koreas need to work together to find an alternative to their respective proposals. The author proposes a Korean peninsula peace treaty signed by the ROK, the DPRK, the U. S. and China as an alternative. Unless the two Koreas demonstrate their desire to cooperate through sincere deeds by sincerely implementing inter-Korean agreements and are willing to make concessions by working together for establishing a peace regime in Korea, there is little chance of achieving this goal. ..PAT.-Unpublished Manuscript [ABSTRACT FROM AUTHOR]

It has been fifty-five years since the conclusion of the 1953 Korean Armistice Agreement (KAA), but a peace regime on the Korean peninsula replacing the KAA with a peace treaty has not yet been achieved. For the last fifty-five years, the two Koreas have attempted to build a peace regime on the Korean peninsula. But their conflicting approaches to a peace mechanism are major obstacles to the Korean peace regime building process. The DPRK has insisted on concluding a peace treaty between the United States and itself to replace the KAA since 1974, while the ROK has equally insisted on a peace treaty between the two Koreas. The six-party talks have been stalled because of North Korea's delayed nuclear declaration of all of its nuclear activities and stockpiles under the October 3, 2007 deal among six nations. When the declaration problem is resolved, the six-party talks will discuss the Phase III (dismantlement of North Korea's nuclear facilities) and a Korean peninsula peace forum. Thus, it is significant to review North Korea's peace regime-building initiative and to make a preparation for a Korean peace forum within the framework of the six-party talks.This paper first will briefly review North Korean peace regime proposals, including re-evaluation of the four-party talks and the six-party talks dealing with a Korean peace regime building, and will look into the major problems of transforming the 1953 Korean armistice agreement into a Korean peninsula peace treaty before it will make policy suggestions for establishing a peace system on the Korean peninsula. In the South Korean society, there are ongoing debates over North Korea¡¯s change. The conservatives and the progressives in South Korean society have conflicting views on this issue. The progressives hold that North Korea has changed significantly, whereas the conservatives argue that it has not changed fundamentally. To end futile and divisive debates over North Korea¡¯s change and to build a national consensus on the Republic of Korea¡¯s engagement policy of national reconciliation and cooperation with the DPRK, it is important to operationally define the concept of North Korea¡¯s change and establish objective criteria for its measurement. The authors of this paper have three specific objectives: (1) to analyze ROK’s engagement policy toward North Korea as a catalyst for North Korea’s change; (2) to set up objective criteria for evaluating North Korea¡¯s change; and (3) to make policy recommendations for constructive inter-Korean relations. The authors attempt to operationalize the concept of North Korea’s change to avoid confusion about North Korea’s change. A clear distinction needs to be made between changes in the North Korean-style socialist system and changes in North Korean leadership¡¯s thinking and policy. We do not expect North Korea will change its socialist system in the short run because North Korean leadership fears a sudden systemic change, which would threaten the very survival of its own system. In this paper we attempt to analyze North Korea’s changes in terms of thinking and policy. We argue that North Korea’s changes in thinking and policy would eventually bring about its systemic change. Age-related hearing loss (ARHL), a degenerative disorder characterized by age-dependent progressive increase in the threshold of auditory sensitivity, affects 40% of people over the age of 65, and it has emerged as an important social and public health problem. Various factors, including genetic and environmental components, are known to affect both the onset and severity of ARHL. In particular, age-dependent changes in cellular oxidative stress and inflammatory responses accompanied by altered cellular signaling and gene expression progressively affect the function of the auditory system and eventually lead to hearing impairment. Recent findings suggest that a disturbance of intracellular NAD
+ levels is clinically related to the progression of age-associated disorders. Therefore, maintenance of optimal intracellular NAD+ levels may be a critical factor for cellular senescence, and thus, understanding its molecular signaling pathways would provide critical insights into the prevention and treatment of ARHL as well as other age-related diseases. In this review, we describe the role of NAD+ metabolism in aging and age-related diseases, including ARHL, and discuss a potential strategy for prevention or treatment of ARHL with a particular interest in NAD+ -dependent cellular pathways. The article focuses on the study which investigates the potential effects of β-lapachone (βL) on vascular smooth muscle cells (VSMCs) on proliferation. Materials such as platelet-derived growth factor (PDGF), dicoumarol, compound C, and adenoviral expression vector were observed. Results reveal that βL does inhibit VSMCs proliferation via the NAD(P)H:quinone oxidoreductase (NQO1) and LKB1-dependent activation of AMP-activated protein kinase (AMPK).

On two-dimensional gel electrophoresis, proteasomes (multicatalytic proteinase complexes) from the yeast Saccharomyces cerevisiae were separated into a characteristic set of approximately 20 components with molecular weights of 21,000 to 31,000 and isoelectric points of 3.5 to 7.5. The main components were isolated by reverse-phase high performance liquid chromatography on a TSK gel phenyl-5PW RP column and named YC1 to YC11, in order of their elution. Immuno-blot analysis showed that two components (YC1-alpha and YC1-beta) with molecular weights of 30,800 and 28,300 strongly cross-reacted with antibody against the P-component of ATP-dependent protease Ti from Escherichia coli, but no components were found to react with antibodies against the A-component of protease Ti or another ATP-dependent protease La (the Ion gene product) of Escherichia coli. These results indicate a structural relationship between eukaryotic proteasomes and bacterial ATP-dependent protease Ti.

The security policy of the ROK since 1950 has been based upon US commitment to South Korea`s defense by providing military and economic aid to the ROK, and for the last 38 years keeping US troops in Korea against another Northern attack. However, in view of new conditions and variables confronting the Sixth Republic of Korea, it has become an imperative to take new perceptions and attitudes with regard to the US troop withdrawal issue and to pursue a new security policy towards North Korea. In the broad context of ROK security thinking as far as the US troop issue is concerned, there appear to be three schools of thought-a status quo policy, anti-status quo or revolutionary, and a gradualist view. For those who support a status quo policy, the presence of the US troops in Korea is a prerequisite for peace and stability on the Korean peninsula. Pyongyang has maintained an anti-status quo policy by demanding that US troops be withdrawn from South Korea. In the North`s view, the presence of US troops is the basic obstacle to inter-Korean dialogue and Korean unification. The gradualist view seeks to break the current stalemate in inter-Korean dialogue. This view holds that the Seoul government should seriously consider the long-term strategic planning about US troop withdrawal issues and have serious discussions on this issue with the US Government. At the same time, the US should seriously consider transferring its operational rights to the ROK. Seoul may then consider convening South-North Korean military talks as suggested by North Korea. In short, the ROK should seek its security by improving and normalizing relations with North Korea in the 1990s. The ROK should be prepared for US ground troop withdrawal in the future, to be used as a bargaining chip in negotiations with the North Korean Government. The rational for this argument is based upon: first. that the ROK will not and should not expect US troops to stay permanently on the Korean peninsula. Second, should the US troops in Korea be pulled out abruptly in the future, the South Korean Government would lose an opportunity to use the US troop withdrawal issue as a political leverage in dealing with North Korea. Third, various conditions seem to be right for such an initiative, Lastly, it is unlikely that North Korea would attempt to use military force against the South.

North Korea's alleged admission of a highly enriched uranium program sparked the second nuclear crisis on the Korean peninsula, accelerating North Korea's nuclear weapons development program. Pyongyang officially announced that it had nuclear weapons on February 10, 2005, and tested its first nuclear weapon on October 9, 2006. It is estimated that North Korea already has six or seven nuclear devices. The North Korean nuclear issue-- a serious international issue as the most important obstacle to the Korean peace process-- needs to be resolved peacefully through the six-party process as the Bush administration insisted. The author of this paper argues that while the six-party process is the best means to resolve the North Korean nuclear issue, U.S.-DPRK bilateral talks are equally important to a peaceful and diplomatic resolution of North Korea's nuclear issue. Both U.S. and North Korea's hard-line policies cannot resolve the nuclear issue peacefully. Therefore, both sides need to be flexible about their respective positions with political will to make a compromise. The Bush administration insisted that the North Korean nuclear crisis be resolved through the six-party process. The author has argued that the six-party process is the best means to resolve the North Korean nuclear dispute. The author maintains that bilateral talks between the U. S. and North Korea are essential to a resolution of North KoreaÂ’s nuclear standoff peacefully and diplomatically. Both need to engage in direct negotiations without preconditions through the six-party talks. Both U.S. and North KoreaÂ’s hard-line policies cannot resolve the nuclear issue peacefully. The viral vector-mediated overexpression of the defined transcription factors, Brn4/Pou3f4, Sox2, Klf4, and c-Myc (BSKM), could induce the direct conversion of somatic fibroblasts into induced neural stem cells (iNSCs). However, viral vectors may be randomly integrated into the host genome thereby increasing the risk for undesired genotoxicity, mutagenesis, and tumor formation. Here we describe the generation of integration-free iNSCs from mouse fibroblasts by non-viral episomal vectors containing BSKM. The episomal vector-derived iNSCs (eiNSCs) closely resemble control NSCs, and iNSCs generated by retrovirus (r-iNSCs) in morphology, gene expression profile, epigenetic status, and self-renewal capacity. The e-iNSCs are functionally mature, as they could differentiate into all the neuronal cell types both in vitro and in vivo. Our study provides a novel concept for generating functional iNSCs using a non-viral, non-integrating, plasmid-based system that could facilitate their biomedical applicability. Cancer-associated thrombosis is the second-leading cause of mortality in patients with cancer and presents a poor prognosis, with a lack of effective treatment strategies. NAD(P)H quinone oxidoreductase 1 (NQO1) increases the cellular nicotinamide adenine dinucleotide (NAD+) levels by accelerating the oxidation of NADH to NAD+, thus playing important roles in cellular homeostasis, energy metabolism, and inflammatory responses. Using a murine orthotopic 4T1 breast cancer model, in which multiple thrombi are generated in the lungs at the late stage of cancer development, we investigated the effects of regulating the cellular NAD+ levels on cancer-associated thrombosis. In this study, we show that dunnione (a strong substrate of NQO1) attenuates the prothrombotic state and lung thrombosis in tumor-bearing mice by inhibiting the expression of tissue factor and formation of neutrophil extracellular traps (NETs). Dunnione increases the cellular NAD+ levels in lung tissues of tumor-bearing mice to restore the declining sirtuin 1 (SIRT1) activity, thus deacetylating nuclear factor-kappa B (NF-κB) and preventing the overexpression of tissue factor in bronchial epithelial and vascular endothelial cells. In addition, we demonstrated that dunnione abolishes the ability of neutrophils to generate NETs by suppressing histone acetylation and NADPH oxidase (NOX) activity. Overall, our results reveal that the regulation of cellular NAD+ levels by pharmacological agents may inhibit pulmonary embolism in tumor-bearing mice, which may potentially be used as a viable therapeutic approach for the treatment of cancer-associated thrombosis.
