Abstract

A global compilation of in situ data is useful to evaluate the quality of ocean-colour satellite data records. Here we describe the data compiled for the validation of the ocean-colour products from the ESA Ocean Colour Climate Change Initiative (OC-CCI). The data were acquired from several sources (including, inter alia, MOBY, BOUSSOLE, AERONET-OC, SeaBASS, NOMAD, MERMAID, AMT, ICES, HOT and GeP&CO) and span the period from 1997 to 2018. Observations of the following variables were compiled: spectral remote-sensing reflectances, concentrations of chlorophyll a, spectral inherent optical properties, spectral diffuse attenuation coefficients and total suspended matter. The data were from multi-project archives acquired via open internet services or from individual projects, acquired directly from data providers. Methodologies were implemented for homogenization, quality control and merging of all data. No changes were made to the original data, other than averaging of observations that were close in time and space, elimination of some points after quality control and conversion to a standard format. The final result is a merged table designed for validation of satellite-derived ocean-colour products and available in text format. Metadata of each in situ measurement (original source, cruise or experiment, principal investigator) was propagated throughout the work and made available in the final table. By making the metadata available, provenance is better documented, and it is also possible to analyse each set of data separately. In late antique and early Christian studies, not only scholars of women and gender but also scholars working on any men of the empire who wrote about, corresponded with, or had sexual relations with young women should read this book to gain a fuller context for their work.

Abstract

The Industry 4.0 concept embodies a topic widely discussed during specialized meetings and tutorials. Originally, this approach was interpreted as an attempt to shift industrial processes towards establishing the fully digitized smart factory; at present, however, it already exerts an influence on many other fields and disciplines. In this article, the concept is described from the perspective of discrete manufacturing. The implementation possibilities are outlined, with options such as the Asset Administrative Shell or Consumer-Provider model introduced in greater detail. We also mention some drawbacks to implementing job shop scheduling as provided by the latter option; these disadvantages include, for instance, the deadlock situation. Some methods to solve the problem are demonstrated to facilitate well-balanced presentation. A job shop scheduling algorithm using the AAS is also addressed, and a new evaluation function is proposed.

Abstract

BACKGROUND: Allergic inflammation is a common feature of asthma and may contribute to both development and perpetuation of disease. The interaction of antigen presenting cells (APC) with sensitized helper T lymphocytes (TC) producing Th2 cytokines may determine the inflammatory response. Recruitment of APC and TC to the lung during allergic responses has been demonstrated, but functional studies in humans have been limited. OBJECTIVE: This study examined the function of APC and TC accumulating at sites of inflammation after segmental allergen challenge (SAC). METHODS: 15 allergic patients underwent SAC, and cells from bronchoalveolar lavage (BAL) were collected after 24 hours. APC and TC from the blood and BAL were purified based on expression of the monocyte marker, CD14; the plasmacytoid dendritic cell (pDC) marker, BDCA4, identifying neuropilin 1 (NRP1); and the helper T cell marker, CD4. Functional activity was assessed using allergen-induced T cell proliferation. Flow-cytometry identified cells expressing CD14 and NRP1. RESULTS: SAC resulted in a 12-fold increase of mononuclear cells having the morphologic appearance of blood monocytes. Most of these cells co-expressed CD14 and NRP1. After saline challenge, BAL mononuclear cells demonstrated little APC function Following SAC, BAL mononuclear cells showed function equal to pDC from blood and greater than blood monocytes. Purified NRP1(+) cells from BAL had even greater function than pDC cells from blood (p = .008). Using consistent sources of APC, enhanced proliferation of TC from lung compared to blood was also demonstrated (p = .002). CONCLUSIONS: The marked increase in APC function for allergen-specific TC proliferation during allergic inflammation is largely due to the recruitment of monocytes and dendritic cells. There is also an enhanced response in the lung TC population, consistent with recruitment of allergen-specific T cells. Interactions between recruited APC and TC may occur as an early event promoting allergic airway inflammation.

Abstract

Background Group B Streptococcus (GBS) infection causes inflammatory co-morbidities in newborns. While the mechanisms remain unclear, evidence suggests that impaired innate-adaptive immune interactions may be contributory. We hypothesized that GBS-stimulated neonatal neutrophils provide a milieu that may drive pro-inflammatory T helper cell programming. Methods Neutrophils were stimulated with Type III GBS (COH1); supernatants or intact neutrophils were co-cultured with CD4+ T cells or Treg cells. Resulting intracellular cytokines and nuclear transcription factors were determined by multicolor flow cytometry. Results GBS-stimulated neutrophils released soluble mediators that induced greater IL-17 responses in neonatal vs. adult CD4+ T cells in the absence of added polarizing cytokines. GBS-stimulated neonatal neutrophils also induced robust expression of the canonical nuclear transcription factors for Th1 (Tbet) and Th17 (IL-17) cells in CD4+ T cells. Following GBS stimulation, both intact neutrophils and neutrophil-derived mediators promoted the generation of Tregs with Th1 and Th17 characteristics. Conclusion GBS-stimulated neonatal neutrophils bias the in vitro Th differentiation program of neonatal CD4+ T cells and promote pro-inflammatory Th1 and Th17 phenotypes in Tregs. Our data suggest that developmental modifications of innate-adaptive immune cross-talk mechanisms may contribute to the inflammatory complications associated with neonatal GBS infection.

Abstract

The energetic economy of running benefits from tendon and other tissues that store and return elastic energy, thus saving muscles from costly mechanical work. The classic “Spring-mass” computational model successfully explains the forces, displacements and mechanical power of running, as the outcome of dynamical interactions between the body center of mass and a purely elastic spring for the leg. However, the Spring-mass model does not include active muscles and cannot explain the metabolic energy cost of running, whether on level ground or on a slope. Here we add explicit actuation and dissipation to the Spring-mass model, and show how they explain substantial active (and thus costly) work during human running, and much of the associated energetic cost. Dissipation is modeled as modest energy losses (5% of total mechanical energy for running at 3 m s-1) from hysteresis and foot-ground collisions, that must be restored by active work each step. Even with substantial elastic energy return (59% of positive work, comparable to empirical observations), the active work could account for most of the metabolic cost of human running (about 68%, assuming human-like muscle efficiency). We also introduce a previously unappreciated energetic cost for rapid production of force, that helps explain the relatively smooth ground reaction forces of running, and why muscles might also actively perform negative work. With both work and rapid force costs, the model reproduces the energetics of human running at a range of speeds on level ground and on slopes. Although elastic return is key to energy savings, there are still losses that require restorative muscle work, which can cost substantial energy during running., Author summary Running is an energetically economical gait whereby the legs bounce like pogo sticks. Leg tendons act elastically to store and return energy to the body, thus saving the muscles from costly work with each running step. Although elasticity is known to save energy, it does not explain why running still requires considerable effort, and why the muscles still do substantial work. We use a simple computational model to demonstrate two possible reasons why. One is that small amounts of energy are lost when the leg collides with the ground and when the tendons are stretched, and muscles must restore that energy during steady running. A second reason is that muscles may perform work to avoid turning on and off rapidly, which may be even more energetically costly. The resulting muscle work, while small in quantity, may still explain most of the energetic cost of running. Economy may be gained from elasticity, but running nonetheless requires muscles to do active work.

Abstract

This is the first book-length study of children in one of the birthplaces of early Christian monasticism, Egypt. Although comprised of men and women who had renounced sex and family, the monasteries of late antiquity raised children, educated them, and expected them to carry on their monastic lineage and legacies into the future. Children within monasteries existed in a liminal space, simultaneously vulnerable to the whims and abuses of adults and also cherished as potential future monastic prodigies. Caroline T. Schroeder examines diverse sources - letters, rules, saints'lives, art, and documentary evidence - to probe these paradoxes. In doing so, she demonstrates how early Egyptian monasteries provided an intergenerational continuity of social, cultural, and economic capital while also contesting the traditional family's claims to these forms of social continuity.

Abstract

Background The mechanisms underlying disease pathogenesis in chronic spontaneous urticaria (CSU) and improvement with omalizumab are incompletely understood. Objectives This study sought to examine whether the rate of clinical remission is concordant with baseline basophil features or the rate of change of IgE-dependent functions of basophils and/or plasmacytoid dendritic cells during omalizumab therapy. Methods Adults (n = 18) with refractory CSU were treated with omalizumab 300 mg monthly for 90 days. Subjects recorded daily urticaria activity scores, and clinical assessments with blood sampling occurred at baseline and on days 1, 3, 6, 10, 20, 30, 60, and 90 following omalizumab. At baseline, subjects were categorized by basophil functional phenotypes, determined by in vitro histamine release (HR) responses to anti-IgE antibody, as CSU-responder (CSU-R) or CSU-non-responder (CSU-NR), as well as basopenic (B) or nonbasopenic (NB). Results CSU-R/NB subjects demonstrated the most rapid and complete symptom improvement. By day 6, CSU-R/NB and CSU-NR/NB had increased anti-IgE–mediated basophil HR relative to baseline, and these shifts did not correlate with symptom improvement. In contrast, CSU-NR/B basophil HR did not change during therapy. The kinetics of the decrease in surface IgE/FceRI was similar in all 3 phenotypic groups and independent of the timing of the clinical response. Likewise, plasmacytoid dendritic cells' surface IgE/FceRI decline and TLR9-induced IFN-α responses did not reflect clinical change. Conclusions Changes in basophil IgE-based HR, surface IgE, or FceRI bear no relationship to the kinetics in the change in clinical symptoms. Baseline basophil count and basophil functional phenotype, as determined by HR, may be predictive of responsiveness to omalizumab.

113. Activation of Human Basophils by A549 Lung Epithelial Cells Reveals a Novel IgE-Dependent Response Independent of Allergen

Abstract

Evidence for epithelial cell (EC)–derived cytokines (e.g., thymic stromal lymphopoietin [TSLP]) activating human basophils remains controversial. We therefore hypothesize that ECs can directly activate basophils via cell-to-cell interaction. Basophils in medium alone or with IL-3 ± anti-IgE were coincubated with TSLP, IL-33, or IL-25. Analogous experiments cocultured basophils (1–72 h) directly with EC lines. Supernatants were tested for mediators and cytokines. Abs targeting receptors were tested for neutralizing effects. Lactic acid (pH 3.9) treatment combined with passive sensitization tested the role of IgE. Overall, IL-33 augmented IL-13 secretion from basophils cotreated with IL-3, with minimal effects on histamine and IL-4. Conversely, basophils (but not mast cells) released histamine and marked levels of IL-4/IL-13 (10-fold) when cocultured with A549 EC and IL-3, without exogenous allergen or IgE cross-linking stimuli. The inability to detect IL-33 or TSLP, or to neutralize their activity, suggested a unique mode of basophil activation by A549 EC. Half-maximal rates for histamine (4 h) and IL-4 (5 h) secretion were slower than observed with standard IgE-dependent activation. Ig stripping combined with passive sensitization ± omalizumab showed a dependency for basophil-bound IgE, substantiated by a requirement for cell-to-cell contact, aggregation, and FcεRI-dependent signaling. A yet unidentified IgE-binding lectin associated with A549 EC is implicated after discovering that LacNAc suppressed basophil activation in cocultures. These findings point to a lectin-dependent activation of basophil requiring IgE but independent of allergen or secreted cytokine. Pending further investigation, we predict this unique mode of activation is linked to inflammatory conditions whereby IgE-dependent activation of basophils occurs despite the absence of any known allergen.

Abstract

Introduction Cockroach allergen exposure elicits cockroach sensitization and poses an increased risk for asthma. However, the major components in cockroach allergen and the mechanisms underlying the induction of cockroach allergen-induced allergy and asthma remain largely elusive. We sought to examine the role of cockroach-associated glycan in regulating human basophil function. Methods N-linked glycans from naturally purified cockroach allergen Bla g 2 were characterized by MALDI-TOF mass spectrometry. Binding of cockroach allergen to serum IgE from cockroach allergic subjects was determined by solid-phase binding immunoassays. Role of cockroach associated glycan in histamine release and IL-4 production from human basophils was examined. Expression of C-type lectin receptors (CLRs) and their role in mediating glycan-uptake in the basophils was also investigated. Results MALDI-TOF mass spectrometric analysis of N-glycan from Bla g 2 showed complex hybrid-types of glycans that terminated with mannose, galactose, and/or N-acetyl glucosamine (GlcNAc). Deglycosylated Bla g 2 showed reduced binding to IgE and was less capable of inducing histamine release from human basophils. In contrast, N-glycan derived from Bla g 2 significantly inhibited histamine release and IL-4 production from basophils passively sensitized with serum from cockroach allergic subjects. An analysis of CLRs revealed the expression of DC-SIGN and DCIR, but not MRC1 and dectin-1, in human basophils. Neutralizing antibody to DCIR, but not DC-SIGN, significantly inhibited Bla g 2 uptake by human basophils. A dose-dependent bindings of cockroach allergen to DCIR was also observed. Conclusions These observations indicate a previously unrecognized role for cockroach allergen-associated glycans in allergen-induced immune reactions, and DCIR may play a role in mediating the regulation of glycan on basophil function.

Abstract

Human locomotion has been well described but is still not well understood. This is largely true because the observable aspects of locomotion-neuromuscular activity that generates forces and motions-relate to both the task solution and the problem being solved. Identifying the fundamental task achieved in locomotion makes it possible to critically evaluate the motor control strategy used to accomplish the task goal. We contend that the readily observed movements and activities of locomotion should be considered mechanism(s). Our proposal is that the fundamental task of walking and running is analogous to flight, and should be defined in terms of the interaction of the individual's mass with the medium in which it moves: a low-density fluid for flight, or the supporting substrate for legged locomotion. A rigorous definition of the fundamental task can help identify the constraints and opportunities that influence its solution and guide the selection of appropriate mechanisms to accomplish the task effectively. The results from robotics-based modeling studies have demonstrated how the interaction of the mass and substrate can be optimized, making the goal of movement a defined trajectory of the individual's mass. We assessed these concepts by evaluating the ground reaction forces generated by an optimization model that satisfies the task but uses none of the mechanisms that are available to the human leg. Then we compared this model to normal human walking. Although it is obvious that the specific task of locomotion changes with a variety of movement challenges, clearly identifying the fundamental task of locomotion puts all other features in an interpretable context.

Abstract

Although the study of the Bible was central to early Humanities Computing efforts, now Biblical Studies and Religious Studies are marginal disciplines in the emerging field known as Digital Humanities (English, History, Library Science, for example, are much more influential in DH.) This paper explores two questions: First, what does it mean for Biblical Studies to be marginal to the Digital Humanities when DH is increasingly seen as the locus of as transformation in the humanities? Second, how can our expertise in Biblical Studies influence and shape Digital Humanities for the better? Digital Humanities, I argue, constitutes a powerful emerging field with which Biblical Studies and Religious Studies must engage as critical participants or analysts. Moreover, our own field’s expertise on the history of canon, orthodoxy, and commentary can contribute to shaping a more inclusive and self-critical Digital Humanities.

Abstract

An angular analysis of the decay B[over ¯]→D^{*}l^{-}ν[over ¯]_{l}, l∈{e,μ}, is reported using the full e^{+}e^{-} collision data set collected by the BABAR experiment at the ϒ(4S) resonance. One B meson from the ϒ(4S)→BB[over ¯] decay is fully reconstructed in a hadronic decay mode, which constrains the kinematics and provides a determination of the neutrino momentum vector. The kinematics of the semileptonic decay is described by the dilepton mass squared, q^{2}, and three angles. The first unbinned fit to the full four-dimensional decay rate in the standard model is performed in the so-called Boyd-Grinstein-Lebed approach, which employs a generic q^{2} parametrization of the underlying form factors based on crossing symmetry, analyticity, and QCD dispersion relations for the amplitudes. A fit using the more model-dependent Caprini-Lellouch-Neubert (CLN) approach is performed as well. Our form factor shapes show deviations from previous fits based on the CLN parametrization. The latest form factors also provide an updated prediction for the branching fraction ratio R(D^{*})≡B(B[over ¯]→D^{*}τ^{-}ν[over ¯]_{τ})/B(B[over ¯]→D^{*}l^{-}ν[over ¯]_{l})=0.253±0.005. Finally, using the well-measured branching fraction for the B[over ¯]→D^{*}l^{-}ν[over ¯]_{l} decay, a value of |V_{cb}|=(38.36±0.90)×10^{-3} is obtained that is consistent with the current world average for exclusive B[over ¯]→D^{(*)}l^{-}ν[over ¯]_{l} decays and remains in tension with the determination from inclusive semileptonic B decays to final states with charm.

Abstract

Background:Psychological distress is presumed to be an important factor that can adversely impact the outcome of orthopaedic procedures. The Distress and Risk Assessment Method (DRAM) is an evaluation tool which assesses psychological distress in patients with low back pain. The purpose of this prospective study was to assess the influence of preoperative psychological distress, as determined by the DRAM score, on the functional outcomes of total joint arthroplasty (TJA).Materials and methods:A prospective study of 61 TJAs was performed at a single institution. The DRAM questionnaire and a variety of functional measures (12-Item Short Form Health Survey [SF-12], visual analogue scale [VAS], Oxford Hip Score [OHS], Oxford Knee Score [OKS], and Oswestry Disability Questionnaire [ODQ]) were administered to the patient at baseline, 1 month, and 6 months postoperatively. Mixed model regressions and Mann-Whitney tests were utilised to evaluate the relationship of the DRAM score with functional outcomes.Results:The summed quantitative DRAM score was predictive of functional outcomes. With each 1 point increase in psychological distress, VAS pain increased by 0.023 ( p = 0.015), OKS decreased by 0.34 ( p = 0.01), ODQ increased by 0.065 ( p = 0.02), and MCS decreased by 0.14 ( p = 0.015). In addition, patients with lower preoperative distress scores had higher rates of improvement than patients with higher preoperative distress scores for VAS pain ( p = 0.034).Discussion:Psychological distress was associated with decreased baseline mental health and function in the early postoperative period, which has important implications for bundled payments. However, patients with psychological distress still demonstrated functional improvements and TJA should thus not be contraindicated in these patients.

Abstract

In Asia, flexible bamboo poles are routinely used to carry substantial loads on the shoulder. Various advantages have been attributed to this load-carrying strategy (e.g. reduced energy consumption), but experimental evidence remains inconsistent – possibly because carriers in previous studies were inexperienced. Theoretical models typically neglect the individual's capacity to optimize interactions with the oscillating load, leaving the complete dynamics underexplored. This study used a trajectory optimization model to predict gait adaptations that minimize work-based costs associated with carrying compliant loads and compared the outcomes with naturally selected gait adaptations of experienced pole carriers. Gait parameters and load interactions (e.g. relative amplitude and frequency, phase) were measured in rural farmworkers in Vietnam. Participants carried a range of loads with compliant and rigid poles and the energetic consequences of step frequency adjustments were evaluated using the model. When carrying large loads, the empirical step frequency changes associated with pole type (compliant versus rigid) were largely consistent with model predictions, in terms of direction (increase or decrease) and magnitude (by how much). Work-minimizing strategies explain changes in leg compliance, harmonic frequency oscillations and fluctuations in energetic cost associated with carrying loads on a compliant bamboo pole.

Abstract

BACKGROUND Mechanical testing of implant constructs designed to treat distal femur fractures has been hampered by a lack of clinical data on the biomechanical properties of the distal femur in patients who sustain these fractures. Therefore, the purpose of this study was to use quantitative computed tomography (qCT) to investigate the mechanical characteristics of fractured distal femurs to inform the selection of synthetic materials for biomechanical testing. METHODS Distal femur fractures treated at a Level I trauma center were retrospectively reviewed and 43 cases with preoperative CT scans were identified for analysis. Scans were segmented and each bone fragment was reconstructed as a 3D model. The Young's modulus of the distal femur was determined from voxel-based radiodensity. We report the observation of the rare charm decay D^{0}→K^{-}π^{+}e^{+}e^{-}, based on 468 fb^{-1} of e^{+}e^{-} annihilation data collected at or close to the center-of-mass energy of the ϒ(4S) resonance with the BABAR detector at the SLAC National Accelerator Laboratory. Abstract

Recent investigations have suggested that the six-quark combination uuddss could be a deeply bound state (S) that has eluded detection so far, and a potential dark matter candidate. We report the first search for a stable, doubly strange six-quark state in Upsilon -> S anti-Lambda anti-Lambda decays based on a sample of 90 million Upsilon(2S) and 110 million Upsilon(3S) decays collected by the BABAR experiment. No signal is observed, and 90% confidence level limits on the combined Upsilon(2S,3S) -> S anti-Lambda anti-Lambda branching fraction in the range (1.2-1.4)x10^-7 are derived for m_S < 2.05 GeV. Abstract

We study the processes $e^+e^-\to 2(\pi^+\pi^-)\pi^0\pi^0\pi^0\gamma$ and $2(\pi^+\pi^-)\pi^0\pi^0\eta\gamma$ in which an energetic photon is radiated from the initial state. The data were collected with the \babar~ detector at SLAC. About 14\,000 and 4700 events, respectively, are selected from a data sample corresponding to an integrated luminosity of 469 fb^{-1}. The invariant mass of the hadronic final state defines the effective \epem center-of-mass energy. The center-of-mass energies range from threshold to 4.5 GeV. From the mass spectra, the first ever measurements of the $e^+e^-\to 2(\pi^+\pi^-)\pi^0\pi^0\pi^0$ and the $e^+e^-\to2(\pi^+\pi^-)\pi^0\pi^0\eta$ cross sections are performed. The contributions from $\omega\pi^+\pi^-\pi^0\pi^0$, $\eta2(\pi^+\pi^-)$, and other intermediate states are presented. We observe the $J/\psi$ and $\psi(2S)$ in most of these final states and measure the corresponding branching fractions, many of them for the first time.

Abstract

We present first evidence that the cosine of the CP-violating weak phase 2β is positive, and hence exclude trigonometric multifold solutions of the Cabibbo-Kobayashi-Maskawa (CKM) Unitarity Triangle using a time-dependent Dalitz plot analysis of B^{0}→D^{(*)}h^{0} with D→K_{S}^{0}π^{+}π^{-} decays, where h^{0}∈{π^{0},η,ω} denotes a light unflavored and neutral hadron. The measurement is performed combining the final data sets of the BABAR and Belle experiments collected at the ϒ(4S) resonance at the asymmetric-energy B factories PEP-II at SLAC and KEKB at KEK, respectively. The data samples contain (471±3)×10^{6}BB[over ¯] pairs recorded by the BABAR detector and (772±11)×10^{6}BB[over ¯] pairs recorded by the Belle detector. The results of the measurement are sin2β=0.80±0.14(stat)±0.06(syst)±0.03(model) and cos2β=0.91±0.22(stat)±0.09(syst)±0.07(model). The result for the direct measurement of the angle β of the CKM Unitarity Triangle is β=[22.5±4.4(stat)±1.2(syst)±0.6(model)]°. The measurement assumes no direct CP violation in B^{0}→D^{(*)}h^{0} decays. The quoted model uncertainties are due to the composition of the D^{0}→K_{S}^{0}π^{+}π^{-} decay amplitude model, which is newly established by performing a Dalitz plot amplitude analysis using a high-statistics e^{+}e^{-}→cc[over ¯] data sample. CP violation is observed in B^{0}→D^{(*)}h^{0} decays at the level of 5.1 standard deviations. The significance for cos2β>0 is 3.7 standard deviations. The trigonometric multifold solution π/2-β=(68.1±0.7)° is excluded at the level of 7.3 standard deviations. The measurement resolves an ambiguity in the determination of the apex of the CKM Unitarity Triangle.

Abstract

We study the process e^+e^− → e^+e^−η′ in the double-tag mode and measure for the first time the γ⋆γ⋆ → η′ transition form factor F_η′(Q^2_1,Q^2_2) in the momentum-transfer range 2 < Q^2_1, Q^2_2 < 60 GeV^2. The analysis is based on a data sample corresponding to an integrated luminosity of around 469 fb^(-1) collected at the PEP-II e^+e^− collider with the BABAR detector at center-of-mass energies near 10.6 GeV.

Abstract

The β common chain (βc) cytokine family includes granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-3 (IL-3) and IL-5, all of which use βc as key signaling receptor subunit. GM-CSF, IL-3 and IL-5 have specific roles as hematopoietic growth factors. IL-3 binds with high affinity to the IL-3 receptor α (IL-3Rα/CD123) and then associates with the βc subunit. IL-3 is mainly synthesized by different subsets of T cells, but is also produced by several other immune [basophils, dendritic cells (DCs), mast cells, etc.] and non-immune cells (microglia and astrocytes). The IL-3Rα is also expressed by immune (basophils, eosinophils, mast cells, DCs, monocytes, and megacaryocytes) and non-immune cells (endothelial cells and neuronal cells). IL-3 is the most important growth and activating factor for human and mouse basophils, primary effector cells of allergic disorders. IL-3-activated basophils and mast cells are also involved in different chronic inflammatory disorders, infections, and several types of cancer. IL-3 induces the release of cytokines (i.e., IL-4, IL-13, CXCL8) from human basophils and preincubation of basophils with IL-3 potentiates the release of proinflammatory mediators and cytokines from IgE- and C5a-activated basophils. IL-3 synergistically potentiates IL-33-induced mediator release from human basophils. IL-3 plays a pathogenic role in several hematologic cancers and may contribute to autoimmune and cardiac disorders. Several IL-3Rα/CD123 targeting molecules have shown some efficacy in the treatment of hematologic malignancies.

Abstract

Androgen deprivation therapy (ADT), a primary treatment for locally advanced or metastatic prostate cancer, is associated with the adverse effects on numerous physiologic parameters, including alterations in cardiometabolic variables that overlap with components of the metabolic syndrome (MetS). As MetS is an established risk factor for cardiovascular mortality and treatment for prostate cancer has been associated with the development of MetS, interventions targeting cardiometabolic factors have been investigated in prostate cancer patients to attenuate the detrimental effects of ADT. Much support exists for exercise interventions in improving MetS variables in insulin-resistant adults, but less evidence is available in men with prostate cancer. Regular exercise, when performed at appropriate intensities and volumes, can elicit improvements in ADT-related adverse effects, including MetS, and contributes to the growing body of literature supporting the role of exercise in cancer survivorship. This review (1) discusses the biologic inter-relationship between prostate cancer, ADT and MetS, (2) evaluates the current literature in support of exercise in targeting MetS and (3) describes the physiological mechanisms by which exercise may favorably alter MetS risk factors in prostate cancer patients on ADT.

Abstract

Four different polymorphs of GdScO 3 are assessed theoretically and experimentally with respect to their suitability as a dielectric. The calculations carried out by density functional theory reveal lattice constants, band gaps and the energies of formation of three crystal phases. Experimentally all three crystal phases and the amorphous phase can be realized as thin films by pulsed laser deposition using various growth templates. Their respective crystal structures are confirmed by X-ray diffraction and transmission electron microscopy reflecting the calculated lattice constants. X-ray photoelectron spectroscopy unveils the band gaps of the different polymorphs of GdScO 3 which are above 5 eV for all films demonstrating good insulating properties. From capacitance voltage measurements, high permittivities of up to 27 for hexagonal GdScO 3 are deduced.

Abstract

Recent experiments demonstrated that the cyclotron cooling rate of an electron plasma in a Penning–Malmberg trap can be increased by placing the plasma in a cavity and adjusting the magnetic field to make the cyclotron motion resonant with a cavity mode. Here this physics is studied with a coupled oscillator model and analyzed both analytically and numerically. Plasma cooling performance is evaluated over a wide range of system parameters, including the number of electrons, the coupling to the local electric field, the magnetic field gradient, and the detuning between the cavity and cyclotron frequencies. Scaling the equations shows that the system is well-described by a few key dimensionless quantities.

Abstract

The decay τ^− → K^−K_Sν_τ has been studied using 430×10^6 e^+e^− → τ^+τ^− events produced at a center-of-mass energy around 10.6 GeV at the PEP-II collider and studied with the BABAR detector. The mass spectrum of the K^−K_S system has been measured and the spectral function has been obtained. The measured branching fraction B(τ^− → K^−K_Sν_τ) = (0.739 ± 0.011(stat) ± 0.020(syst)) × 10^(-3) is found to be in agreement with earlier measurements.

Abstract

Compliant bamboo poles have long been used for load carriage in Asian cultures. Although this custom differs from Western conventions of rigid body attachments (e.g. backpack), potential benefits include reduced peak shoulder forces as well as metabolic transport cost savings. Evidence that carrying a flexible pole benefits locomotion remains mixed, perhaps in part because the properties of pole design (e.g. bamboo material, structural geometry, etc.) have largely been neglected. These properties influence vibrational forces and consequently, the energy required by the user to manage the oscillations. We collected authentic bamboo poles from northern Vietnam and characterized their design parameters. Four poles were extensively studied in the lab (load-deflection testing, resonance testing, and computed tomography scans of three-dimensional geometry), and 10 others were tested at a rural Vietnamese farm site (basic measures of form and resonance). A mass-spring-damper model was used to characterize a relationship between resonant frequency (which affects the energetics of the pole-carrier system) and pole properties concerning stiffness, damping, etc. Model predictions of resonant frequencies agreed well with empirical data. Although measured properties suggest the poles are not optimally designed to reduce peak oscillation forces, resonant frequencies are within range of a typical human walking cadence, and this is likely to have a consequence on locomotion energetics.

Abstract

Only 23.6% of adults and 75 years meet physical activity (PA) guidelines (aerobic & muscle-strengthening). Health benefit gym memberships has not improved participation. Therefore, Blue Goji developed a Virtual Reality (VR) treadmill to improve PA through fitness gamification with cognitive involvement in postural control to improve balance. Feasibility testing was done with seven older Veterans and one spouse, mean age of 81.3 years who participate in the VA Gerofit program & were near the 50th percentile (by age & gender) in strength, balance and endurance. Even those with lower levels of fitness, balance or chronic conditions (i.e., kyphosis or vision impairment) strongly supported this for enjoyment, benefit, comfort, safety and strongly recommended this (mean of 4.65 out of 5). The Gamification approach supports anti-ageism and intergenerational fitness activities. Further study on the additive effect on exercise intensity and improvement in fitness, balance and cognition is needed.

Abstract

This article motivates and details the first implementation of a freely available part of speech tag set and tagger for Coptic. Coptic is the last phase of the Egyptian language family and a descendant of the hieroglyphs of ancient Egypt. Unlike classical Greek and Latin, few resources for digital and computational work have existed for ancient Egyptian language and literature until now. We evaluate our tag set in an inter-annotator agreement experiment and examine some of the difficulties in tagging Coptic data. Using an existing digital lexicon and a small training corpus taken from several genres of literary Sahidic Coptic in the first half of the first millennium, we evaluate the performance of a stochastic tagger applying a fine-grained and coarse-grained set of tags within and outside the domain of literary texts. Our results show that a relatively high accuracy of 94–95% correct automatic tag assignment can be reached for literary texts, with substantially worse performance on documentary papyrus data. We also present some preliminary applications of natural language processing to the study of genre, style, and authorship attribution in Coptic and discuss future directions in applying computational linguistics methods to the analysis of Coptic texts.

Abstract

Fluids co-produced with methane from hydraulically fractured organic-rich shales of the Marcellus Formation (USA) are characterized by high total dissolved solids (TDS), including elevated levels of Ba, Sr and Br. To investigate the source and geologic history of these high-TDS fluids and their dissolved constituents, we carried out a series of sequential extraction experiments on dry-drilled cuttings extracted within, below and above the Marcellus Shale from a well in Tioga County, New York State. The experiments were designed to extract (1) water soluble components, (2) exchangeable cations, (3) carbonate minerals, and (4) hydrochloric acid-soluble constituents. The geochemistry of the resultant leachates highlights the different geochemical reservoirs for extractable elements within the shale; notably, Na and Br were largely water-soluble, while Ba was extracted primarily from exchangeable sites, and Ca and Sr were found both in exchangeable sites and carbonate. Strontium isotope ratios measured on the leachates indicate that each of the element reservoirs has a distinct value. Measured 87Sr/86Sr ratios in the water soluble component are similar to those of Marcellus produced water, while the ion exchange reservoir yields lower ratios, and carbonate Sr is lower still, approaching Devonian-Silurian seawater values. Despite the isotopic similarity of water leachates and produced water, the total water chemistry argues against generation of produced water by interaction of hydraulic fracturing fluid with “dry” shale. The high-TDS produced water is most likely trapped formation water (within and/or adjacent to the shale) that is released by hydraulic fracturing. The formation water was affected by multiple processes, possibly including basin scale, tectonically-driven fluid flow. Significant chemical and isotopic differences between Marcellus Shale produced water and overlying Upper Devonian/Lower Mississippian produced waters suggests a hydrologic barrier has been maintained in parts of the Appalachian Basin since the late Paleozoic.

Abstract

Natural gas produced from a zone of thin Upper Devonian/Lower Mississippian sands approximately 1200 m above the hydraulically fractured Middle Devonian Marcellus Shale interval was monitored for evidence of gas migration. Gas samples were collected from seven vertical Upper Devonian/Lower Mississippian gas wells and two vertical Marcellus Shale gas wells 2 months prior to-, during-, and 14 months after the hydraulic fracturing of six horizontal Marcellus Shale gas wells at the study site. The isotopic and molecular compositions of gas from the two producing zones were distinct and remained so during the entire monitoring period. Over the time of monitoring, the molecular/isotopic signatures of gas from the Upper Devonian/Lower Mississippian field did not show any evidence of contamination from deeper Marcellus Shale gas that might have migrated upward from the hydraulically fractured interval. Our results indicate no hydrologic connectivity between the fractured interval and formations 1200 m above, which means that contamination of even shallower drinking water aquifers (∼2200 m above fractured interval) is unlikely at this study site. While localized consideration for geology and site development practices are extremely important, the monitoring methods used in this study are applicable when trying to understand and quantify natural gas mixing and migration trends.

Abstract

Basophil-derived IL-4 is involved in the alternative activation of mouse monocytes, as recently shown in vivo. Whether this applies to human basophils and monocytes has not been established yet. Here, we sought to characterise the interaction between basophils and monocytes and identify the molecular determinants. A basophil-monocyte co-culture model revealed that IL-3 and basophil-derived IL-4 and IL-13 induced monocyte production of CCL17, a marker of alternative activation. Critically, IL-3 and IL-4 acted directly on monocytes to induce CCL17 production through histone H3 acetylation, but did not increase the recruitment of STAT5 or STAT6. Although freshly isolated monocytes did not express the IL-3 receptor α chain (CD123), and did not respond to IL-3 (as assessed by STAT5 phosphorylation), the overnight incubation with IL-4 (especially if associated with IL-3) upregulated CD123 expression. IL-3-activated JAK2-STAT5 pathway inhibitors reduced the CCL17 production in response to IL-3 and IL-4, but not to IL-4 alone. Interestingly, monocytes isolated from allergen-sensitised asthmatic patients exhibited a higher expression of CD123. Taken together, our data show that the JAK2-STAT5 pathway modulates both basophil and monocyte effector responses. The coordinated activation of STAT5 and STAT6 may have a major impact on monocyte alternative activation in vitro and in vivo.

Abstract

The number of Marcellus Shale gas wells drilled in the Appalachian basin has increased rapidly over the past decade, leading to increased interest in the highly saline water produced with the natural gas which must be recycled, treated, or injected into deep disposal wells. New geochemical and isotopic analyses of produced water for 3 time-series and 13 grab samples from Marcellus Shale gas wells in southwest and north central Pennsylvania (PA) are used to address the origin of the water and solutes produced over the long term (>12 months). The question of whether the produced water originated within the Marcellus Shale, or whether it may have been drawn from adjacent reservoirs via fractures is addressed using measurements of and activity. These parameters indicate that the water originated in the Marcellus Shale, and can be more broadly used to trace water of Marcellus Shale origin. During the first 1–2 weeks of production, rapid increases in salinity and positive shifts in values were observed in the produced water, followed by more gradual changes until a compositional plateau was reached within approximately 1 yr. The values and relationships between Na, Cl, and Br provide evidence that the water produced after compositional stabilization is natural formation water, the salinity for which originated primarily from evaporatively concentrated paleoseawater. The rapid transition from injected water to chemically and isotopically distinct water while of the injected water volume had been recovered, supports the hypothesis that significant volumes of injected water were removed from circulation by imbibition.

Abstract

The energetic cost of transport for walking is highly sensitive to speed but relatively insensitive to changes in gravity level. Conversely, the cost of transport for running is highly sensitive to gravity level but not much to speed. Gait optimization with a minimally constrained bipedal model predicts a similar differential energetic response for walking and running even though the same model parameters and cost function are used for both gaits. This challenges previous assertions that the converse energetic responses are due to fundamentally different energy saving mechanisms in each gait. Our results suggest that energetics of both gaits are highly influenced by dissipative losses occurring as leg forces abruptly alter the center of mass path. The observed difference in energetic consequence of the performance condition in each gait is due to the effect the movement strategy of each gait has on the dissipative loss. The optimization model predictions are tested directly by measuring metabolic cost of human subjects walking and running at different speeds in normal and reduced gravity using a novel reduced gravity simulation apparatus. The optimization model also predicts other, sometimes subtle, aspects of gait such as step length changes. This is also directly tested in order to assess the fidelity of the model’s more nuanced predictions.

Abstract

We search for single-photon events in 53 fb^{-1} of e^{+}e^{-} collision data collected with the BABAR detector at the PEP-II B-Factory. We look for events with a single high-energy photon and a large missing momentum and energy, consistent with production of a spin-1 particle A^{'} through the process e^{+}e^{-}→γA^{'}; A^{'}→invisible. Such particles, referred to as "dark photons," are motivated by theories applying a U(1) gauge symmetry to dark matter. We find no evidence for such processes and set 90% confidence level upper limits on the coupling strength of A^{'} to e^{+}e^{-} in the mass range m_{A^{'}}≤8 GeV. In particular, our limits exclude the values of the A^{'} coupling suggested by the dark-photon interpretation of the muon (g-2)_{μ} anomaly, as well as a broad range of parameters for the dark-sector models.

150. The Discovery of the Papyri from Tura at Dayr al-Qusayr (Dayr Arsaniyus) and Its Legacy

Abstract

This chapter examines the significant papyri find in the ancient St. Arsenius monastery in Tura, which yielded a wealth of writings in Greek from one of the most prominent heads of the Theological School of Alexandria, Didymus the Blind. Didymus has rarely received the scholarly attention one would expect for someone with such a high reputation in antiquity and with such a prodigious output of texts to study. His legacy in historiography has been affected by the perception that he is either an unoriginal thinker or a heretic. In both his commentaries and his treatises, Didymus defends Christianity and his definition of orthodoxy in debates with pagan philosophers and “heretics.” However, the ideas he developed in defending Christianity (such as allegorical exegesis) were the very things by which his ideas were judged heresy after his death by the Greek and Roman churches.
