Methane and carbon dioxide were measured with an autonomous and continuous running system on a ferry line crossing the Baltic Sea on a 2–3 day interval from the Mecklenburg Bight to the Gulf of Finland in 2010. Surface methane saturations show great seasonal differences in shallow regions like the Mecklenburg Bight (103–507 %) compared to deeper regions like the Gotland Basin (96–161 %). The influence of controlling parameters like temperature, wind, mixing depth and processes like upwelling, mixing of the water column and sedimentary methane emissions on methane oversaturation and emission to the atmosphere are investigated. Upwelling was found to influence methane surface concentrations in the area of Gotland significantly during the summer period. In February 2010, an event of elevated methane concentrations in the surface water and water column of the Arkona Basin was observed, which could be linked to a wind-derived water level change as a potential triggering mechanism. The Baltic Sea is a source of methane to the atmosphere throughout the year, with highest fluxes occurring during the winter season. Stratification was found to promote the formation of a methane reservoir in deeper regions like Gulf of Finland or Bornholm Basin, which leads to long lasting elevated methane concentrations and enhanced methane fluxes, when mixed to the surface during mixed layer deepening in autumn and winter. Methane concentrations and fluxes from shallow regions like the Mecklenburg Bight are predominantly controlled by sedimentary production and consumption of methane, wind events and the change in temperature-dependent solubility of methane in the surface water. Methane fluxes vary significantly in shallow regions (e.g. Mecklenburg Bight) and regions with a temporal stratification (e.g. Bornholm Basin, Gulf of Finland). On the contrary, areas with a permanent stratification like the Gotland Basin show only small seasonal fluctuations in methane fluxes.

This study highlights the potential of using a low frequency multibeam echosounder for detection and visualization of shallow gas occurring several meters beneath the seafloor. The presence of shallow gas was verified in the Bornholm Basin, Baltic Sea, at 80 m water depth with standard geochemical core analysis and hydroacoustic subbottom profiling. Successively, this area was surveyed with a 95 kHz and a 12 kHz multibeam echosounder (MBES). The bathymetric measurements with 12 kHz provided depth values systematically deeper by several meters compared to 95 kHz data. This observation was attributed to enhanced penetration of the low frequency signal energy into soft sediments. Consequently, the subbottom geoacoustic properties contributed highly to the measured backscattered signals. Those appeared up to 17 dB higher inside the shallow gas area compared to reference measurements outside and could be clearly linked to the shallow gas front depth down to 5 meter below seafloor. No elevated backscatter was visible in 95 kHz MBES data, which in turn highlights the superior potential of low frequency MBES to image shallow sub-seafloor features. Small gas pockets could be resolved even on the outer swath (up to 65°). Strongly elevated backscattering from gassy areas occurred at large incidence angles and a high gas sensitivity of the MBES is further supported by an angular response analysis presented in this study. We conclude that the MBES together with subbottom profiling can be used as an efficient tool for spatial subbottom mapping in soft sediment environments.

Six new Citrullus haplotypes were identified at tRNA-Val – rps12 locus of chloroplast (cpDNA) genome in colocynth (Citrullus colocynthis), citron melon (Citrullus lanatus citroides) and watermelon (Citrullus lanatus lanatus) cultivars. Ancient-DNAs (aDNA) of seed remains excavated from two sites of the Middle Ages (13th-14th Cent. Debrecen and 15th Cent. Budapest, Hungary) and a herbarium samples from the 19th Cent. (Pannonhalma, Hungary) were also investigated to track cpDNA evolution back to the 13th-14th centuries with the final aim of molecular and morphological reconstruction of old varieties. Medieval samples showed molecular similarity to current cultivars ‘Belyj dlinnij’, ‘Csárdaszállás’ ‘Klondike’, and ‘Kecskeméti vörös’ indicating the oldest haplotype of cultivated watermelons. The 170-yr-old herbarium sample showed close molecular similarity to citron melon, which results reflect the former importance of citron melon as fodder in Hungary. Propagule banks are assumed to be able to store considerable genetic variability. Bryophyte populations are expected to rely more heavily on stored propagules than those of seed plants due to the vulnerability of the haploid gametophyte. This reliance has important implications for the genetic structure and evolutionary potential of surface populations. A liverwort, Mannia fragrans, was used to test whether the bryophyte diaspore bank functions as a "genetic memory." If a diaspore bank is capable of conserving genetic variability over generations, the levels of genetic diversity in the soil are expected to be similar or higher than at the surface. Surface and diaspore bank constituents of two populations of M. fragrans were investigated. Genetic structure and diversity measured as unbiased heterozygosity were analyzed using three ISSR markers. Similar genetic diversities were found in the soil (Hs = 0.067) and at the surface (Hs= 0.082). However, more haplotypes and specific haplotype lineages were present in soil samples. The results suggest that the bryophyte diaspore bank has an important role in accumulating genetic variability over generations and seasons. It is postulated that the role of the diaspore bank as a "genetic memory" is especially important in species of temporarily available habitats that have long-lived spores and genetically variable populations.

Background: Revealing the past and present demographic history of populations is of high importance to evaluate the conservation status of species. Demographic data can be obtained by direct monitoring or by analysing data of historical and recent collections. Although these methods provide the most detailed information they are very time consuming. Another alternative way is to make use of the information accumulated in the species' DNA over its history. Recent development of the coalescent theory makes it possible to reconstruct the demographic history of species using nucleotide polymorphism data. To separate the effect of natural selection and demography, multilocus analysis is needed because these two forces can produce similar patterns of polymorphisms. In this study we investigated the amount and pattern of sequence variability of a Europe wide sample set of two peat moss species (Sphagnum fimbriatum and S. squarrosum) with similar distributions and mating systems but presumably contrasting historical demographies using 3 regions of the nuclear genome (appr. 3000 bps). We aimed to draw inferences concerning demographic, and phylogeographic histories of the species. Results: All three nuclear regions supported the presence of an Atlantic and Non-Atlantic clade of S. fimbriatum suggesting glacial survival of the species along the Atlantic coast of Europe. Contrarily, S. squarrosum haplotypes showed three clades but no geographic structure at all. Maximum likelihood, mismatch and Bayesian analyses supported a severe historical bottleneck and a relatively recent demographic expansion of the Non-Atlantic clade of S. fimbriatum, whereas size of S. squarrosum populations has probably decreased in the past.
