102. Selective therapeutic efficacy of tyrosine kinase inhibitor sorafenib on the restoration of methylglyoxal-induced peritoneal fibrosis

Abstract

Peritoneal fibrosis, a common complication observed in long-term peritoneal dialysis patients, can gradually lead to ultrafiltration failure and the development of encapsulating peritoneal sclerosis. Although mechanisms of peritoneal fibrosis have been proposed, effective therapeutic options are unsatisfactory. Recently, several tyrosine kinase inhibitors have proven to be anti-fibrosis in rodent models. To assess the potential therapeutic effects of tyrosine kinase inhibitors on peritoneal fibrosis in the larger animal model, a novel porcine model of peritoneal fibrosis induced by 40 mM methylglyoxal in 2.5 % dialysate was established, and two different doses (20 mg/kg and 30 mg/kg) of sorafenib were given orally to evaluate their therapeutic efficacy in this study. Our results showed that sorafenib effectively reduced adhesions between peritoneal organs and significantly diminished the thickening of both the parietal and visceral peritoneum. Angiogenesis, vascular endothelial growth factor A production, myofibroblast infiltration, and decreased endothelial glycocalyx resulting from dialysate and methylglyoxal stimulations were also alleviated with sorafenib. However, therapeutic efficacy in ameliorating loss of mesothelial cells, restoring decreased ultrafiltration volume, and improving elevated small solutes transport rates was limited. In conclusion, this study demonstrated that sorafenib could potentially be used for peritoneal fibrosis treatment, but applying sorafenib alone might not be sufficient to fully rescue methylglyoxal-induced peritoneal defects.

Abstract

Goal-oriented dialogue systems face a trade-off between fluent language generation and task-specific control. While supervised learning with large language models is capable of producing realistic text, how to steer such responses towards completing a specific task without sacrificing language quality remains an open question. In this work, we formulate goal-oriented dialogue as a partially observed Markov decision process, interpreting the language model as a representation of both the dynamics and the policy. This view allows us to extend techniques from learning-based control, such as task relabeling, to derive a simple and effective method to finetune language models in a goal-aware way, leading to significantly improved task performance. We additionally introduce a number of training strategies that serve to better focus the model on the task at hand. We evaluate our method, Context-Aware Language Models (CALM), on a practical flight-booking task using AirDialogue. Empirically, CALM outperforms the state-of-the-art method by 7% in terms of task success, matching human-level task performance.

Abstract

Renewable energy has weak feedforward and controlled characteristics, so the traditional industrial frequency protection does not work when renewable energy is sent out through AC transmission line. On the contrary, the traveling wave protection methods can judge the fault before the control system responds, which are one of the effective ways to solve the problem encountered in renewable power system. However, the existing traveling wave protection schemes are limited by the sampling rate, which makes it difficult to accurately identify the internal near-end fault and the internal remote-end fault. To solve this problem, the grid diagram of traveling wave in case of internal and external faults is analyzed at first. Then, the time difference between zero-mode and line-mode is used to judge the fault location. For the near-end fault and remote-end fault which are difficult to be accurately identified based on the time difference, the first reverse polarity traveling wave is used to identify the near-end fault and the polarity of the first two line-mode traveling waves is used to identify the remote-end fault. Simulation results show that the proposed scheme can quickly identify internal and external faults. At the same time, the proposed scheme has good ability to withstand transient resistance.

Abstract

The frequency stability of the wind power grid-connected system may be improved by the participation of wind turbines in frequency regulation. However, it is difficult for the existing droop control to coordinate the frequency response characteristics and the operating state of the wind turbine. An adaptive droop control strategy is proposed to make full use of the rotor kinetic energy to participate in frequency regulation and ensure the stable operation of the wind turbine considering the rate of change of frequency (ROCOF) and the rotor kinetic energy. Firstly, a coupling function between the droop coefficient and ROCOF is established by the piecewise function with the intervals of ROCOF according to the system frequency, which can be released to ensure more energy from wind turbines at the initial stage of the disturbance. In this condition, the frequency drop is slowed down due to the support of the wind turbine for frequency regulation. Besides, to avoid the over-deceleration of the wind turbine and secondary frequency drop, an influence factor on the rotor speed is introduced to adjust the droop coefficient according to the operating state of the wind turbine. Finally, a wind-thermal combined system simulation model is built on the MATLAB/Simulink platform to verify the effectiveness of the proposed control strategy. The simulation results show that the proposed strategy can effectively apply the rotor kinetic energy of the wind turbine to improve the frequency response characteristics of the system while ensuring the stability of the wind turbine speed.

Abstract

Zero-knowledge proof (ZKP) is a fundamental cryptographic primitive that allows a prover to convince a verifier of the validity of a statement without leaking any further information. As an efficient variant of ZKP, non-interactive zero-knowledge proof (NIZKP) adopting the Fiat-Shamir heuristic is essential to a wide spectrum of applications, such as federated learning, blockchain and social networks. However, the heuristic is typically built upon the random oracle model making ideal assumptions about hash functions, which does not hold in reality and thus undermines the security of the protocol. Here, we present a quantum resolution to the problem. Instead of resorting to a random oracle model, we implement a quantum randomness service. This service generates random numbers certified by the loophole-free Bell test and delivers them with postquantum cryptography (PQC) authentication. Employing this service, we conceive and implement a NIZKP of the three-colouring problem. By bridging together three prominent research themes, quantum non-locality, PQC and ZKP, we anticipate this work to open a new paradigm of quantum information science., Comment: 20 pages, 9 figures, 6 tables

145. Evaluation of the Music Therapy Program Interventions on Agitated Behavior for People with Dementia in Taiwan Institutional Care

Abstract

Music therapy can help social workers deal with negative behaviors and symptoms of dementia in older adults without drugs or physical restraints, thereby improving the quality of care. This study tested the effectiveness of music therapy activities for improving agitated behavior in older adults with dementia living in long-term care institutions. The study was quasi-experimental. Eighty older adults were recruited from two nursing homes. A purposive sampling strategy was used to select subjects aged 65 years and above with dementia and agitated behavioral tendencies residing in nursing homes. The music therapy program comprised 90 minute sessions 3 times a week for two months. The program evaluation focused on one criterion: the agitated behavior of participants at the beginning and end of the music theory program, which was measured using a structured scale. Ultimately, the music therapy program showed promise for improving agitated behavior in older adults with dementia living in nursing homes. Incorporating music therapy activities into social work may help make long-term care more diverse and innovative.

Abstract

Biomarker-assisted diagnosis and intervention in Alzheimer's disease (AD) may be the key to prevention breakthroughs. One of the hallmarks of AD is the accumulation of tau plaques in the human brain. However, current methods to detect tau pathology are either invasive (lumbar puncture) or quite costly and not widely available (Tau PET). In our previous work, structural MRI-based hippocampal multivariate morphometry statistics (MMS) showed superior performance as an effective neurodegenerative biomarker for preclinical AD and Patch Analysis-based Surface Correntropy-induced Sparse coding and max-pooling (PASCS-MP) has excellent ability to generate low-dimensional representations with strong statistical power for brain amyloid prediction. In this work, we apply this framework together with ridge regression models to predict Tau deposition in Braak12 and Braak34 brain regions separately. We evaluate our framework on 925 subjects from the Alzheimer's Disease Neuroimaging Initiative (ADNI). Each subject has one pair consisting of a PET image and MRI scan which were collected at about the same times. Experimental results suggest that the representations from our MMS and PASCS-MP have stronger predictive power and their predicted Braak12 and Braak34 are closer to the real values compared to the measures derived from other approaches such as hippocampal surface area and volume, and shape morphometry features based on spherical harmonics (SPHARM)., Comment: 10 pages, 5 figures, 17th International Symposium on Medical Information Processing and Analysis
