The acquired data were subjected to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, and a protein-protein interaction (PPI) network was constructed to screen potential markers. The selected proteins were further verified through ELISAs. The differences between control and sepsis groups and between survivors and non-survivors were analysed. Receiver operating characteristic (ROC) curves were drawn to explore their diagnostic value and prognostic efficacy.Results: A total of 149 DEPs were identified by bioinformatics methods. The analyses showed that these proteins are mainly involved in biological processes such as cell movement, stress response, cell proliferation, and immune response. Functional pathway analysis showed that they are mainly involved in leukocyte transendothelial migration, protein synthesis and processing, and various bacterial infections. LGALS3BP was selected as a potential plasma biomarker and further verified through an ELISA. Its level in septic patients was significantly higher than that in normal controls, and its level in non-survivors was also higher than that in survivors. The ROC curves suggested its great diagnostic efficacy and prognostic ability in sepsis.Conclusion: LGALS3BP levels were significantly different between the normal and sepsis groups; it has good diagnostic value in sepsis, and is related to patient prognosis; thus, it might be a biomarker for sepsis.Keywords: LGALS3BP, sepsis, plasma biomarker, DIA, ELISA

Thus, targeting CHK1 is an efficient strategy for cancer therapy.Purpose: The present study aimed to investigate the potential therapeutic effects of GDC-0575, a CHK1-specific inhibitor, in colitis-associated cancer (CAC) and colitis.Methods: We established a DSS-induced acute colitis model and an azoxymethane/dextran sodium sulfate (DSS)-induced CAC model using mice and tested the effect of GDC-0575 on them. Flow cytometry and immunofluorescence were employed to investigate the infiltration of immune cells, and inflammatory cytokine expression in the colon of mice with CAC or colitis was investigated using ELISA and qPCR. We also investigated the correlation between CHK1 and CCL2/CCR2 in human colorectal cancer (CRC) tissues.Results: Administration of GDC-0575 significantly inhibited CHK1 expression in the colon and dramatically impaired the development of CAC and colitis in mice. Moreover, the inhibition of CHK1 expression resulted in efficient inhibition of infiltration by iNOS-positive macrophages, but had no significant effect on CD4 T cells, CD8 T cells, and myeloid-derived suppressor cells (MDSCs). Significant downregulation of TNF-α, IL-6, and IL-1β and dramatic upregulation of IL-10 were observed in the colons of both mice with CAC and colitis treated with GDC-0575. CCL2 expression was also downregulated by GDC-0575 in both mice with CAC and colitis; this was followed by the inhibition of CCR2+ macrophage infiltration in the colon. Furthermore, we report a positive correlation between CHK1 expression and CCL2/CCR2 expression in the malignant tissues of patients with CRC.Conclusion: Taken together, we infer that GDC-0575 impairs the development of CAC and colitis by regulating cytokine expression and inhibiting CCR2+ macrophage infiltration in mice colon.Keywords: colitis, colitis-associated cancer, CHK1, macrophage

- Abstract
The prognosis for patients with pancreatic ductal adenocarcinoma (PDAC) remains extremely poor. It has been suggested that the adenosine pathway contributes to the ability of PDAC to evade the immune system and its resistance to immunotherapies (Immuno-Oncology Therapy, IOT), by generating extracellular adenosine (eAdo).Using genetically engineered allograft models of PDAC in syngeneic mice with differential immune infiltration and response to IOT, we showed enrichment of the adenosine pathway in tumour-infiltrating immune cells (in particular, myeloid populations). Using MS-Imaging, we showed that extracellular adenosine distribution is heterogeneous in tumours, with high concentrations in hypoxic margins that surround necrotic areas, associated with a rich myeloid infiltration, demonstrated using Imaging Mass Cytometry (IMC). Pro-tumorigenic M2 macrophages express high levels of the Adora2a receptor; particularly in the IOT resistant model. Blocking thein vivoformation and function of eAdo (Adoi), using a combination of anti-CD73 antibody and an Adora2a inhibitor slowed tumour growth and reduced metastatic burden. In addition, blocking the adenosine pathway improved the efficacy of combinations of cytotoxic agents or immunotherapy. Finally, Adoi remodelled the tumour microenvironment (TME), as evidenced by reduced infiltration of M2 macrophages and Tregs. RNAseq analysis showed that genes related to immune modulation, hypoxia and tumour stroma were downregulated following Adoi and a specific adenosine signature derived from this is associated with a poorer prognosis in patients with PDAC.The formation of eAdo appears to promote the development of the immunosuppressive TME in PDAC, contributing to its resistance to conventional and novel therapies. Therefore, inhibition of the adenosine pathway may represent a strategy to modulate the stroma and improve therapy response in patients with PDAC.

- Abstract
Dams and reservoirs are among the most widespread human-made infrastructures on Earth. Despite their societal and environmental significance, spatial inventories of dams and reservoirs, even for the large ones, are insufficient. A dilemma of the existing georeferenced dam datasets is the polarized focus on either dam quantity and spatial coverage (e.g., GlObal geOreferenced Database of Dams, GOODD) or detailed attributes for a limited dam quantity or region (e.g., GRanD (Global Reservoir and Dam database) and national inventories). One of the most comprehensive datasets, the World Register of Dams (WRD), maintained by the International Commission on Large Dams (ICOLD), documents nearly 60g000 dams with an extensive suite of attributes. Unfortunately, the WRD records provide no geographic coordinates, limiting the benefits of their attributes for spatially explicit applications. To bridge the gap between attribute accessibility and spatial explicitness, we introduce the Georeferenced global Dams And Reservoirs (GeoDAR) dataset, created by utilizing the Google Maps geocoding application programming interface (API) and multi-source inventories. We release GeoDAR in two successive versions (v1.0 and v1.1) at 10.5281/zenodo.6163413 (Wang et al., 2022). GeoDAR v1.0 holds 22g560 dam points georeferenced from the WRD, whereas v1.1 consists of (a) 24g783 dam points after a harmonization between GeoDAR v1.0 and GRanD v1.3 and (b) 21g515 reservoir polygons retrieved from high-resolution water masks based on a one-to-one relationship between dams and reservoirs. Due to geocoding challenges, GeoDAR spatially resolved g1/4g40g% of the records in the WRD, which, however, comprise over 90g% of the total reservoir area, catchment area, and reservoir storage capacity. GeoDAR does not release the proprietary WRD attributes, but upon individual user requests we may provide assistance in associating GeoDAR spatial features with the WRD attribute information that users have acquired from

The cytokinin two‐component signal transduction system (TCS) is involved in many biological processes, including hormone signal transduction and plant growth regulation. Although cytokinin TCS has been well characterized in Arabidopsis thaliana, its role in tomato remains elusive.In this study, we characterized the diversity and function of response regulator (RR) genes, a critical component of TCS, in tomato. In total, we identified 31 RR genes in the tomato genome. These SlRR genes were classified into three subgroups (type‐A, type‐B and type‐C).Various stress‐responsive cis‐elements were present in the tomato RR gene promoters. Their expression responses under pesticide treatment were evaluated by transcriptome analysis. Their expression under heat, cold, ABA, salinity and NaHCO3 treatments was further investigated by qRT‐PCR and complemented with the available transcription data under these treatments. Specifically, SlRR13 expression was significantly upregulated under salinity, drought, cold and pesticide stress and was downregulated under ABA treatment. SlRR23 expression was induced under salt treatment, while the transcription level of SlRR1 was increased under cold and decreased under salt stress.We also found that GATA transcription factors played a significant role in the regulation of SlRR genes. Based on our results, tomato SlRR genes are involved in responses to abiotic stress in tomato and could be implemented in molecular breeding approaches to increase resistance of tomato to environmental stresses. Gram-negative bacteria possess an asymmetric outer membrane (OM) composed primarily of lipopolysaccharides (LPSs) on the outer leaflet and phospholipids (PLs) on the inner leaflet. The loss of this asymmetry due to mutations in the LPS biosynthesis or transport pathways causes the externalization of PLs to the outer leaflet of the OM and leads to OM permeability defects. Here, we used metabolic labeling to detect a compromised OM in intact bacteria. Phosphatidylcholine synthase expression in Escherichia coli allowed for the incorporation of exogenous propargylcholine into phosphatidyl(propargyl)choline and exogenous 1-azidoethyl-choline (AECho) into phosphatidyl(azidoethyl)choline (AEPC), as confirmed by LC/MS analyses. A fluorescent copper-free click reagent poorly labeled AEPC in intact wild-type cells but readily labeled AEPC from lysed cells. Fluorescence microscopy and flow cytometry analyses confirmed the absence of significant AEPC labeling from intact wild-type E. coli strains and revealed significant AEPC labeling in an E. coli LPS transport mutant (lptD4213) and an LPS biosynthesis mutant (E. coli lpxC101). Our results suggest that metabolic PL labeling with AECho is a promising tool for detecting a compromised bacterial OM, revealing aberrant PL externalization, and identifying or characterizing novel cell-active inhibitors of LPS biosynthesis or transport.

- Abstract
Eating wildlife has been a long-standing social custom, and its changing pattern reflects the changes of complex and diverse social practices in societies. Leveraging on the past social surveys, this article reports the findings of a 2020 survey, with 826 completed samples on the current situation and characteristics of public consumption of wildlife in China. We find that the public consumption of wildlife has continued to decline, and that it has ceased to be a popular habit by the majority of our sample. On the basis of the comparative analysis of the four factors of gender, occupation, age and education level, the current public consumption of wildlife shows the characteristics of ‘gender differentiation’ and ‘consumption elitism’. At the same time, there may be conflict between internal and external tensions in the cultivation of public good eating behaviour. The article also outlines some future directions of further analysis and research.

To address the challenges in remediating cesium contaminated aqueous environments, a low-cost, magnetically recoverable and superior composite adsorbent was fabricated based on the concept of magnetizing montmorillonite and entrapping potassium copper hexacyanoferrate that offers excellent selectivity for Cs+. The facile, green and scalable synthesis route involved exchanging the interlayer ions of montmorillonite with ferrous ions before oxidizing to form magnetic montmorillonite using a low-temperature hydrothermal method. The composite was then coated with polydopamine to be complexed with copper ions and subsequently reacted with the hexacyanoferrate precursor to in situ grow potassium copper hexacyanoferrate nanoparticles, thus forming the composite, D-Mt-Mag-HCF. The adsorbent exhibited excellent Cs+ sorption capacity (~159.2 mg/g) and Cs+ selectivity greater than 8.2 × 104 mL g−1 in concentrated brine. Moreover, the magnetic properties (17.4 emu/g) of the adsorbent facilitated its separation from contaminated aqueous environments once the adsorbent had removed Cs+. The current study demonstrates a novel and scalable production of a composite adsorbent that can be readily used to remediate contaminated water.

Ageing is associated with increased vulnerability to environmental cold exposure. Previously, we identified the role of the cold-sensitive transient receptor potential (TRP) A1, M8 receptors as vascular cold sensors in mouse skin. We hypothesised that this dynamic cold-sensor system may become dysfunctional in ageing. We show that behavioural and vascular responses to skin local environmental cooling are impaired with even moderate ageing, with reduced TRPM8 gene/protein expression especially. Pharmacological blockade of the residual TRPA1/TRPM8 component substantially diminished the response in aged, compared with young mice. This implies the reliance of the already reduced cold-induced vascular response in ageing mice on remaining TRP receptor activity. Moreover, sympathetic-induced vasoconstriction was reduced with downregulation of the α2c adrenoceptor expression in ageing. The cold-induced vascular response is important for sensing cold and retaining body heat and health. These findings reveal that cold sensors, essential for this neurovascular pathway, decline as ageing onsets.
