With the rapid development of the economy, China’s economic form has also undergone signifcant changes. In this case, the requirements for talents are getting higher and higher. In the process of cultivating talents, schools must constantly change their educational concepts and cultivate more application oriented talents for the society. In this context, the reform of university and college English is of great necessity. Based on the importance of application-oriented talent training, this paper analyzes the current situation of English teaching in universities and colleges under the application-oriented talent training mode, and puts forward some specifc measures for the reform of English teaching in universities and colleges.

The microstructure evolution of Mg96−xGd3Zn1Zrx (at%) (x=0, 0.1, 0.2, 0.4) alloys with long period stacking ordered (LPSO) structures prepared by conventional permanent mold casting were investigated by XRD, EDS, SEM and TEM. Also, the deformation behavior and microstructure evolution of Mg95.8Gd3Zn1Zr0.2 (at%) alloy subjected to equal channel angular pressing (ECAP) were examined. The results show that the LPSO structure phases observed in as-cast Mg96Gd3Zn1 alloy did not notably improve the mechanical properties of the alloy owing to their low volume fraction. In contrast, the mechanical properties of the alloy after heat treatment were raised significantly because of the transformation from eutectic structures to LPSO structure phases. Furthermore, moderate Zr addition into Mg–Gd–Zn alloys refined and homogenized α-Mg grains, changed the morphology and distribution of LPSO structure phases and promoted the precipitation of strengthening phases, and hence strengthened the alloys. Besides, tensile test results showed that aging-treated Mg95.8Gd3Zn1Zr0.2 alloy subjected to 2-pass ECAP processing exhibited high ultimate tensile strength (UTS) of 418 MPa, yield strength (YS) of 330 MPa and elongation of 7.5%. Such excellent mechanical properties could be attributed to complex strengthening of kinking LPSO phases and the dispersion strengthening of tiny spherical phases.

The effect of lanthanum content in the range of 0–0.011 wt.%, on the inclusion size distribution, microstructure, texture and magnetic properties of three non-oriented electrical steels was studied. After final annealing, lanthanum effectively inhibited the precipitation of MnS precipitates in steel, the formations of La2O2S and LaS inclusions not only acted as nuclei of AlN precipitates, but also combined with Al2O3 and formed composite inclusions with larger size. Grain size firstly increased and then decreased with lanthanum content increasing. Steel containing 0.0066 wt.% lanthanum obtained the largest grain size, the strongest {110} texture and the weakest {112} texture among all the tested steels. Magnetic flux density firstly increased and then decreased, core loss firstly dramatically decreased and then slightly decreased with lanthanum content increasing. Among the three tested steels, steel with 0.0066 wt.% lanthanum demonstrated the best comprehensive magnetic properties mainly through the development of favorable texture and appropriate final grain size.

BACKGROUND: The prognosis of patients with relapsed B-lineage acute lymphocyte leukemia (B-ALL) after allogeneic hematopoietic stem cells transplantation(HSCT) is poor. It is difficult to obtain disease-free remission and long-term survival in these patients based on chemotherapy, donor leukocyte infusion (DLI) or molecular targeted therapy. Donor-derived anti-CD19 CAR-T(CAR 19) cells may obtain remission in these patients, but many still encounter relapse, especially CD19-negative relapse. CD123 is a surface marker not only associated with the progenitors of acute myeloid leukemia, but also found in progenitors of acute lymphoblastic leukemia. Recent studies have shown that CD123 is highly expressed in the patients with CD19-negative relapsed after CAR19 cells therapy. we developed a novel therapeutic strategy for to administer pooled donor-derived fourth generation CART cells targeting CD19 and CD123 respectively, to patients who relapsed following allogeneic HSCT to avoid CD19-negative relapse. In this study, we report three patients who received combination CARTs therapy achieved disease-free survival that at least more than 6 months (www.clinicaltrials.gov; #NCT03125577). PATIENTS AND METHODS: Three patients with relapsed B-ALL after HLA-matched sibling HSCT have been enrolled in the study to date, and all of their leukemia cells highly expressed CD19 and CD123 antigens. The first and second patients experienced relapse 6 months and one year after all-HSCT respectively; their B-ALLs were p190-positive and carried T315I mutation, and resisted to bonatinib, chemotherapy and DLI. The third patient was also refractory to chemotherapy and DLI. All patients received fludarabine (FLU) and cyclophosphamide (CTX) conditioning chemotherapy (FLU 30mg/m2, d1-3; CTX 300mg/m2, d1-3) before CART infusions. Donor T cells were apheresis collected and transduced with an apoptosis-inducible, safety-engineered lentiviral CD19 or CD123 scFv CAR fused with intracellular signaling domains: CD28/CD27/CD3ζ-iCasp9 (4SCAR19 and 4SCAR123). CAR-T cells were infused at dose range of 0.26-1.38x106 cells/kg. The quality of apheresis cells, gene transfer and T cell proliferation efficiencies, and effective CAR T infusion dose were quantitatively scored and documented. RESULTS: All three patients achieved minimal residual disease (MRD) negative remission within 1 month after CAR-T infusions. Monthly follow-ups of the first and second patients indicated that they achieved stable MRD-negative and p190-negative remission, and remained disease-free for 7 months and 11 months, respectively. The third patient was MRD-positive but achieved bone marrow morphological remission at 7 month follow-up time. Flow cytometry analysis of the MRD cells detected CD19 positive and CD123 partial positive ALL clones, and note that the third patient received the lowest dose of CART infusion, 0.26x10e6/kg. The first patient developed grade 1 cytokine release syndrome (CRS) after CAR-T cell infusions. The second patient developed grade 1 oral acute graft-versus-host disease (aGVHD) and pulmonary infection. The third patient developed grade 2 CRS, with hypoxemia and unilateral massive pleural effusion. We detected high IL-6 in his pleural fluid >5000 IU/L, but serum level IL-6 is normal. There was no significant absorption of pleural effusion after treatment with anti-interleukin 6 receptor monoclonal antibody. The patient improved after 5 days of treatment with chest drainage and dexamethasone 10mg/qd. None of the three patients developed central nervous system toxicity, and there was no greater than grade 2 CRS and severe myelosuppression, consistent with the safety profile of the 4SCAR design. CONCLUSIONS: We have successfully treated three relapsed allo-HSCT B-ALL patients using donor-derived 4SCAR19 and 4SCAR123 T cells. All three patients achieved long-term disease-free survival without severe CRS and GVHD. Thus, the administration of double 4SCAR19/4SCAR123 T cells may overcome CD19 escape and prolong disease-free survival. Disclosures No relevant conflicts of interest to declare.

With the shortage of water resources in China, water-saving agriculture has been highly addressed and developed by the government. This paper studies the channel structure of a helical micro pressure drip irrigation emitter, focusing on the tilt and tooth chamfer’s angles within eight different combinations of structure parameters. By comparison of hydraulic performances, it show a small discrepancies between CFD software simulating results and testing results on 6 selected real trial prototype samples of flaky emitters and inlaid drip pipes. A conclude is made that under various flow coefficient K and pressure sensitive coefficient X, in the section of minimum flow, the smaller angle θ the better hydraulic performance, the bigger angle r the better anti clogging ability.

This study was primarily involved with the experimental determination of the dynamic mechanical properties of five different sheet steels. The nominal yield strengths of these five types of sheet steels ranged from 172 to 690 MPa (25–100 ksi) and the range of strain rates varied from 10 −4 to 1.0 mm/mm/s. All these five materials were studied in both longitudinal and transverse directions under various strain rates, and some materials were also tested under different strain rates and different amounts of prior cold stretching. In addition, two types of steels were tested to estimate the aging effect on the mechanical properties of sheet steels. The test results showed that the proportional limit, yield strength, and ultimate tensile strength increase with increasing strain rate. In general, the amount of increase was found to be dependent on the static yield stress of sheet steel, the amounts of prior cold stretching, and the strain rates used in the tests, and independent on the test directions (longitudinal or transverse), test types (tension or compression), and material aging conditions (aged or nonaged). General equations were also derived to predict the tensile and compressive yield stresses for strain rates ranging from 10 −4 to 10 2 mm/mm/s.

Fluctuating light intensity had a more significant impact on growth of gametophytes of transgenic Laminaria japonica in a 2500 ml bubble-column bioreactor than constant light intensity. A fluctuating light intensity between 10 and 110 microE m(-2) s(-1), with a photoperiod of 14 h:10 h light:dark, was the best regime for growth giving 1430 mg biomass l(-1).

To investigate the injuries of intestinal mitochondria induced by different doses of whole-body radiation in Tibet minipigs.Eighteen Tibet minipigs were randomized into 5 radiation groups (n=3) and a control group (n=3). The minipigs in the radiation groups were subject to a total body X-ray radiation at 2, 5, 8, 11, or 14 Gy, and 72 h after the exposure, the mRNA expressions of the intestinal mitochondrial genes were examined using RT-PCR. The changes in the respiratory chain complexes I-IV and the respiratory functions of succinate and NADH were assayed, and the intestinal ultrastructures were observed using transmission electron microscopy (TEM) following the exposures.Compared with those in the control group, the expression levels of the related mitochondrial genes, the activities of the respiratory chain complexes and the function of the respiratory chain were significantly lowered in the radiation groups. At the doses below 8 Gy, the exposures caused significant reduction in the measurements as the radiation doses increased, but at higher doses, these measurements showed no further reductions. Ultrastructurally, exposures at 2 and 5 Gy caused mitochondrial expansion and mild reduction of the density, whereas radiation at 8 Gy or greater resulted in vacuolar changes and obvious expansion of the mitochondria with damages of the mitochondrial cristae and membranes.Below the doses of 8 Gy, intestinal mitochondrial damages in the minipigs increase with the radiation dose, but at higher doses, the damages do not further increase with the radiation dose.
