120. Load adaptive and fault tolerant framework for energy saving in fiber-wireless access networks.

Abstract

Energy saving (ES) in telecommunication networks is an important criterion when planning access networks. In fiber-wireless (FiWi) access networks the ES potential is high, when compared with other architectures, because different routes and optical access points can be used by routers at the wireless section. Although some proposals to increase energy efficiency in these architectures have been presented, these are not approaches that can adapt to variations in traffic load or distribution of traffic across the network. Here we fill this gap and propose a load adaptive and fault tolerant framework for ES in FiWi access networks. This framework allows optical network units (ONUs) to enter long-standing sleep mode under low traffic conditions, reducing energy waste, permitting fast reaction to ONU or fiber failures, and allowing quality of service (QoS) to be kept at a certain level. Results show that significant ES can be achieved under low to medium traffic loads while maintaining QoS and fault tolerance. [ABSTRACT FROM PUBLISHER]

Abstract

Spin-dependent scattering in Fe, Gd and Tb polycrystalline metallic foils was investigated using circularly polarized synchroton radiation measurements. The results showed that a significant enhancement of the magnetic scattering amplitude compared to the magnetic resonance scattering of linearly polarized light and sample magnetization perpendicular to the scattering plane occured. It was shown that magnetic scattering using circularly polarized synchroton radiation was useful for investigating ferromagnetic materials.

130. Design of QoS-aware energy-efficient fiber-wireless access networks.

Abstract

Energy-efficient network design has recently become a very important topic because of the energy cost increases in service providers' infrastructures. This is of particular importance in access networks because of the growing demand for digital traffic by end users. Here we address the challenge of reducing the energy consumption of fiber-wireless (FiWi) access networks, which use both optical and radio frequency technologies to provide high bandwidth and ubiquity for end-user applications, while keeping delay under a threshold. Our goal is to find optimal sleep mode schedulings that allow energy consumption to be reduced while keeping packet delay acceptable. For this purpose a mathematical formalization and an algorithm are developed. The results show that the proposed approach is able to reduce the average packet delay, with negligible energy cost increases, in many scenarios, besides being computationally efficient and scalable. The proposed approach may, therefore, serve as a basis for planning and design of quality-of-service-aware energy-efficient FiWi access networks. [ABSTRACT FROM PUBLISHER]

132. Synthesis, Characterization, and Application of Cy-Dye- and Alexa-Dye-Labeled Hongotoxin<INF>1</INF> Analogues. The First High Affinity Fluorescence Probes for Voltage-Gated K<SUP>+</SUP> Channels

Abstract

Hongotoxin1 (HgTX1), a 39-residue peptide recently isolated from the venom of Centruroides limbatus, blocks the voltage-gated K+ channels Kv1.1, Kv1.2, and Kv1.3 at picomolar toxin concentrations (Koschak, A., Bugianesi, R. M., Mitterdorfer, J., Kaczorowski, G. J., Garcia, M. L., and Knaus, H. G. (1998) J. Biol. Chem. 273, 2639−2644). In this report, we determine the three-dimensional structure of HgTX1 using NMR spectroscopy (PDB-code:  1HLY). HgTX1 was found to possess a structure similar to previously characterized K+ channel toxins (e.g. margatoxin) consisting of a three-stranded antiparallel β-sheet (residues 2−4, 26−30, and 33−37) and a helical conformation (part 310 helix and part α helix; residues 10−20). Due to the importance of residue Lys-28 for high-affinity interaction with the respective channels, lysine-reactive fluorescence dyes cannot be used to label wild-type HgTX1. On the basis of previous studies (see above) and our NMR data, a HgTX1 mutant (HgTX1-A19C) was engineered, expressed, and purified. HgTX1-A19C-SH was labeled using sulfhydryl-reactive Cy3-, Cy5-, and Alexa-dyes. Pharmacological characterization of fluorescently labeled HgTX1-A19C in radioligand binding studies indicated that these hongotoxin1 analogues retain high-affinity for voltage-gated K+ channels and a respective pharmacological profile. Cy3- and Alexa-dye-labeled hongotoxin1 analogues were used to investigate the localization of K+ channels in brain sections. The distribution of toxin binding closely follows the distribution of Kv1.2 immunoreactivity with the highest expression levels in the cerebellar Purkinje cell layer. Taken together, these results demonstrate that fluorescently labeled HgTX1 analogues comprise novel probes to characterize a subset of voltage-gated K+ channels.

Abstract

The negative symptoms of schizophrenia remain a major clinical challenge. Reboxetine is an antidepressant whose major mechanism of action is as a noradrenergic reuptake inhibitor. This study was a 6-week randomized placebo-controlled trial of reboxetine or placebo add on to haloperidol 5 mg in the treatment of 30 patients with DSM-IV schizophrenia. The trial failed to demonstrate any significant difference between the placebo and reboxetine groups on any of the outcome measures. This trial does not suggest that increased noradreneregic drive mediated by reuptake inhibition in patients taking dopamine antagonists is of therapeutic value in schizophrenia.

147. Rescue of mice homozygous for lethal albino deletions: implications for an animal model for the human liver disease tyrosinemia type 1

Abstract

Mice homozygous for specific deletions around the albino locus on chromosome 7 die within the first few hours of birth. They have a complex phenotype in liver and kidney, which includes multiple changes in gene expression and ultrastructural abnormalities. On the basis of this phenotype, it was proposed that these deletions remove a regulatory locus, alf or hsdr-1. Recently, we and others showed that the gene for fumarylacetoacetate hydrolase (Fah), an enzyme involved in tyrosine catabolism, was disrupted by the lethal albino deletion c14CoS. The finding that the Fah gene in wild-type mice is highly expressed only in cell types that develop a phenotype in mutants, and the fact that Fah deficiency determines the human liver disease hereditary tyrosinemia type 1 (HT1), suggested that disruption of the Fah gene was responsible for the lethal albino phenotype. To test this hypothesis, we have created lines of mice carrying Fah transgenes. We find that c14CoS homozygotes which express transgenic Fah are complemented for all aspects of the complex lethal albino phenotype. Moreover, the degree to which the phenotype is corrected depends on the level of transgenic Fah expression. These results unequivocally establish Fah as the gene mapping at alf/hsdr-1 and prove that the phenotype depends ultimately on the blockage of tyrosine metabolism. Finally, they suggest lethal albino mice as an animal model for HT1.
