128. The Onset of English Lexical Acquisition among Malaysian Preschoolers

Abstract

English is undoubtedly an important language for educational and socio-economic mobility in numerous countries including Malaysia. Regardless of its importance, studies to document English language acquisition among Malaysian children acquiring English in the local context remain scarce. This normative data is imperative for syllabus-designers, policymakers, teachers and linguists to understand and to develop materials that are developmentally sensitive and contextualised to the local setting. Thus, the objective of this paper is to present the onset of English lexical acquisition among 99 Malaysian preschoolers at the beginning of formal instruction. The 99 children were pupils in 3 public preschools in Klang Valley, Malaysia. The children's language production was elicited in individual communicative tasks. Their lexical output, both in English and Malay were analysed based on the children's language choice during the session as well as the frequencies of types and tokens. The results indicate that Malaysian preschoolers displayed a limited range of productive abilities and age-sensitive vocabulary in English at the beginning of formal instruction. However, at that point, the children already possess receptive skills greater than their productive skills. Based on the findings, implications for pedagogy are also discussed.

129. Play and Responsibility: Clean-Up Time for Japanese Preschoolers

Abstract

Japanese teachers consider clean-up time to be the group's responsibility, and they rely on the use of music and encouragement to make clean-up time an appealing activity. The children's positive feelings also may be partially explained by the fact that teachers consider clean-up time to be an extension of play, saving toys and activities for the next day so children may enjoy them again. Teachers use the "han" (group) to help children find peer-based rather than teacher-based methods of dealing with breakdowns in cooperative work. In each "han," children are divided into small groups of 4 or 5 children, and members of each "han" are responsible for their peers in the group. If "han" members fail to work together, teachers will encourage children to find the solution to problems by directing them to suggest a pragmatic strategy for eliciting cooperative behavior instead of using criticism or peer pressure. According to the author, to minimize the impression of their control, teachers delegate authority to children and encourage them to manage their own problems without teacher intervention. Teachers use both music and encouragement to promote children's involvement in various activities and tasks. All preschool teachers are required to be skilled in playing the piano, and they use music rather than spoken words to signal any transition time. These children's positive feelings about cleaning appear to be associated with teacher guidance. The teachers do not force children to engage in clean-up time, yet simply perceive it to be the group's responsibility and make it appealing.

Abstract

Cry toxins are insecticidal proteins produced by Bacillus thuringiensis (Bt). They are used commercially to control insect pests since they are very active in specific insects and are harmless to the environment and human health. The gene encoding ATP-binding cassette subfamily A member 2 (ABCA2) was identified in an analysis of Cry2A toxin resistance genes. However, we do not have direct evidence for the role of ABCA2 for Cry2A toxins or why Cry2A toxin resistance does not cross to other Cry toxins. Therefore, we performed two experiments. First, we edited the ABCA2 sequence in Bombyx mori using transcription activator-like effector-nucleases (TALENs) and confirmed the susceptibility-determining ability in a diet overlay bioassay. Strains with C-terminal half-deleted BmABCA2 showed strong and specific resistance to Cry2A toxins; even strains carrying a deletion of 1 to 3 amino acids showed resistance. However, the C-terminal half-deleted strains did not show cross-resistance to other toxins. Second, we conducted a cell swelling assay and confirmed the specific ability of BmABCA2 to Cry2A toxins in HEK239T cells. Those demonstrated that BmABCA2 is a functional receptor for Cry2A toxins and that BmABCA2 deficiency-dependent Cry2A resistance does not confer cross-resistance to Cry1A, Cry1Ca, Cry1Da, Cry1Fa or Cry9Aa toxins.

Abstract

Chronic obstructive pulmonary disease (COPD) mainly develops after long-term exposure to cigarette or biomass fuel smoke, but also occurs in non-smokers with or without a history of asthma. We investigated the proportion and clinical characteristics of non-smokers among middle-aged to elderly subjects with airflow obstruction.We retrospectively analyzed 1,892 subjects aged 40-89 years who underwent routine preoperative spirometry at a tertiary university hospital in Japan. Airflow obstruction was defined as a forced expiratory volume in 1 second (FEV1)/forced vital capacity < 0.7 or as the lower limit of the normal.Among 323 patients presenting with FEV1/forced vital capacity < 0.7, 43 had asthma and 280 did not. Among the non-asthmatic patients with airflow obstruction, 94 (34%) were non-smokers. A larger number of women than men with airflow obstruction had asthma (26% vs. 7.6%, p < 0.001), or were non-smokers among non-asthmatics (72% vs. 20%, p < 0.001). Non-asthmatic non-smokers, rather than non-asthmatic smokers, asthmatic non-smokers, and asthmatic smokers, exhibited better pulmonary function (median FEV1: 79% of predicted FEV1 vs. 73%, 69%, and 66%, respectively, p = 0.005) and less dyspnea on exertion (1% vs. 12%, 12%, and 28%, respectively, p = 0.001). Pulmonary emphysema on thoracic computed tomography was less common in non-smokers (p < 0.001). Using the lower limit of the normal to define airflow obstruction yielded similar results.There are a substantial number of non-smokers with airflow obstruction compatible with COPD in Japan. In this study, airflow obstruction in non-smokers was more common in women and likelier to result in mild functional and pathological abnormalities than in smokers. Further studies are warranted to investigate the long-term prognosis and appropriate management of this population in developed countries, especially in women.

Abstract

The fractions and polysaccharides from Sasa veitchii were evaluated for their preventive effects on influenza A virus (IFV-A) infection. In IFV-A infected mice, the high-molecular portion (SH) of hot-water extract from S. veitchii significantly reduced virus yields in lung and bronchoalveolar lavage fluid (BALF) and increased in IFV-A-specific secretory IgA in BALF and intestine samples, and neutralizing antibody titers in BALFs and sera. Separation of SH by combination with ion-exchange column and gel filtration yielded six purified polysaccharides. Among them, SH2A1B1 with molecular weight of 29.7 kDa was found to reduce virus yields in lung as well as BALF in IFV-A-infected mice. A significant increase in the production of neutralizing antibody was also observed in BALFs on day 15 post-infection. Furthermore, significant enhancement of nitrite oxide production in RAW 264.7 cells was observed. These results suggest that the fractions from S. veitchii and SH2A1B1 could provide useful materials for prevention of IFV-A infection.

Abstract

When ABC transporter family C2 (ABCC2) and ABC transporter family B1 (ABCB1) were heterologously expressed in non-susceptible cultured cells, the cells swelled in response to Cry1A and Cry3 toxins, respectively. Consistent with the notion that 3D-Cry toxins form cation-permeable pores, Bombyx mori ABCC2 (BmABCC2) facilitated cation-permeable pore formation by Cry1A when expressed in Xenopus oocytes. Furthermore, BmABCC2 had a high binding affinity (KD) to Cry1Aa of 3.1 × 10−10 M. These findings suggest that ABC transporters, including ABCC2 and ABCB1, are functional receptors for 3D-Cry toxins. In addition, the Cry2 toxins most distant from Cry1A toxins on the phylogenetic tree used ABC transporter A2 as a receptor. These data suggest that 3D-Cry toxins use ABC transporters as receptors. In terms of inducing cell swelling, ABCC2 has greater activity than cadherin-like receptor. The pore opening of ABC transporters was hypothesized to be linked to their receptor function, but this was repudiated by experiments using mutants deficient in export activity. The synergistic relationship between ABCC2 and cadherin-like receptor explains their ability to cause resistance in one species of insect.
