108. Presenilin-dependent γ-secretase processing of β-amyloid precursor protein at a site corresponding to the S3 cleavage of Notch

Abstract

The presenilin (PS)-dependent site 3 (S3) cleavage of Notch liberates its intracellular domain (NICD), which is required for Notch signaling. The similar γ-secretase cleavage of the β-amyloid precursor protein (βAPP) results in the secretion of amyloid β-peptide (Aβ). However, little is known about the corresponding C-terminal cleavage product (CTFγ). We have now identified CTFγ in brain tissue, in living cells, as well as in an in vitro system. Generation of CTFγ is facilitated by PSs, since a dominant-negative mutation of PS as well as a PS gene knock out prevents its production. Moreover, γ-secretase inhibitors, including one that is known to bind to PS, also block CTFγ generation. Sequence analysis revealed that CTFγ is produced by a novel γ-secretase cut, which occurs at a site corresponding to the S3 cleavage of Notch.

Abstract

Ontogenetic changes in dry weight, elemental composition (CHN) and activities of digestive enzymes (total protease, amylase) were studied through the moulting cycles of all larval stages (zoea I-II, megalopa; ZI, ZII, M) of the spider crab Maja brachydactyla Balss 1922 reared in the laboratory. All analyses were performed in single larvae to evaluate individual variability. Digestive enzyme activities were analysed by fluorometry. Changes in dry weight were only in premoult M statistically significant, whereas no differences were observed among Z and postmoult M (Fig. 1). No clear pattern was observed in the elemental composition (CHN in % of DW). An increase in enzyme activities throughout development was observed, although no statistically significant differences were found. Amylase (Fig. 2) and total protease activities were different among M moulting stages, being highest at premoult and lowest at intermoult. It seems that high individual variability (high standard errors) masked ontogenical changes in digestive enzyme activities, since previous data obtained from larval pools using spectophotometry analyses indicated that total protease and amylase activities were significantly higher between ZI and M. Despite lacking significance, the present study shows that different moulting stages presented different physiological conditions, and therefore, nutritional requirements may undergo marked changes during larval moult cycles. Hence, moulting stages must be taken into account in future studies to evaluate the nutritional condition of crustacean larvae.

Abstract

Al Ilarg de totes les èpoques, el ball ha estat un aspecte important de totes les cultures. Entre les cultures primitives, el ball va ser una de les formes principals d'expressió social i ritual religiós. Els homes i les dones ballaven per guanyar-se els favors dels déus, els quals proporcionaven aliment, refugi, salut i seguretat. Ballaven per expressar l'alegria en els naixements, la felicitat en l'amor, el valor en la guerra i la tristesa davant la mort. Els seus moviments s'inspiraven en la natura, els moviments dels animals, el vol dels ocells i la bellesa de la sortida o la posta del sol. Les diverses formes de dansar o ballar impressionen de manera diferent cada persona. Pot ser que sigui simplement una activitat social agradable realitzada en el centre comunitari; pot ser un art que exigeixi elevada qualitat; pot ser una expressió cultural, molt colorista i altament competitiva alhora. Tota persona té una necessitat bàsica d'expressar-se a si mateix amb el moviment. El ball proporciona moltes oportunitats artístiques i d'esbarjo per a tots. La dansa, que molts historiadors han assenyalat corn la més antiga de les arts, és, en la seva forma culta, la d'aparició més recent entre nosaltres.

A lo largo de todas las épocas, el baile ha sido un aspecto importante de todas las culturas. Entre las culturas primitivas, el baile fue una de las formas principales de expresión social y ritual religioso. Los hombres y las mujeres bailaban para ganarse los favores de los dioses, quienes proporcionaban el alimento, el refugio, la salud y la seguridad. Bailaban para expresar su alegria en los nacimientos, su felicidad en el amor, su valor en la guerra y su tristeza frente a la muerte. Sus movimientos se inspiraban en la naturaleza, los movimientos de los animales, el vuelo de los pájaros y la belleza del amanecer o de la puesta del sol. Las diversas formas de danzar o bailar impresionan de distinta manera a cada persona. Puede que sea simplemente una actividad social agradable realizada en el centro comunitario; puede ser un arte que exija una elevada calidad; puede ser una expresión cultural, muy colorista y altamente competitiva al mismo tiempo. Toda persona tiene una necesidad básica de expresarse a sí misma con el movimiento. El baile proporciona muchas oportunidades artísticas y de recreo para todos. La danza, que muchos historiadores han señalado como la más antigua de las artes, es en su forma culta, la de más reciente aparición entre nosotros.

135. MODIFICATION OF A RAPID STARCH METHOD FOR CANE JUICE.

Abstract

Rapid methods of starch analysis in raw juice avoid the initial precipitation of starch with ethanol, and so are useful for routine monitoring of cane quality or starch removal in clarification. Detailed evaluation of two such published procedures, however revealed inconsistent results and / or low starch recovery. Our tests indicated that a part of cane starch is removed from solution while boiling the juice sample, presumably by entrainment or co-precipitation during heat-induced denaturing and coagulation of cane proteins. The entrained fraction of starch is then removed by centrifugation or filtration, leading to underestimates of starch in the juice. A modification was tested where prior to the standard iodometric reaction, insoluble cane starch gets completely solubilized by heating in 15% CaCl2 solution. At this concentration, CaCl2 appears to prevent any loss of starch during initial heating of the juice sample. This modified procedure preserves the simplicity and ease of the previous SPRI rapid method; the method is suited to simultaneously analyze a large number of samples, is robust (not subject to cane matrix interferences) and gives 100% starch recovery. 143. PPARγ co-activator-1α (PGC-1α) reduces amyloid-β generation through a PPARγ-dependent mechanism.

Abstract

We have previously reported that the nuclear receptor peroxisome proliferator activated receptor-γ (PPARγ) regulates the transcription of β-secretase (BACE1), a key enzyme involved in amyloid-β (Aβ) generation. Here, we aimed to investigate the role of PPARγ coactivator-1α (PGC-1α), which controls major metabolic functions through the co-activation of PPARγ and other transcription factors. Western blotting experiments with nuclear extracts from brain cortex of AD cases and controls showed a reduction in the levels of PGC-1α in AD patients. PGC-1α overexpression in N2a neuroblastoma cells induced a decrease in the levels of secreted Aβ and an increase in the levels of non-amyloidogenic soluble AβPPα. The decrease in Aβ after exogenous expression of PGC-1α was a consequence of reduced BACE1 expression and transcription, together with a decrease in BACE1 promoter activity. In addition, we detected a significant reduction in β-secretase activity by measuring the levels of β-carboxy terminus fragment (β-CTFs) and by using a commercial assay for β-secretase. In contrast, down-regulation of PGC-1α levels by transfection with PGC-1α siRNA increased BACE1 expression. These effects appeared to be dependent on PPARγ, because PGC-1α did not affect Aβ and BACE1 levels in N2a cells transfected with PPARγ siRNA or in PPARγ knockout fibroblasts. In conclusion, since PGC-1α appears to decrease Aβ generation, therapeutic modulation of PGC-1α could have real potential as a treatment for AD. [ABSTRACT FROM AUTHOR]

149. New layered double hydroxides with the hydrotalcite structure containing Ni(II) and V(III)

Abstract

Layered double hydroxides (LDH) with the hydrotalcite-like structure, but containing Ni(II) and V(III) cations in the layers, have been prepared by the constant pH method. Characterization by elemental chemical analysis, powder X-ray diffraction, differential thermal analysis, thermogravimetric analysis, FT-IR spectroscopy, temperature-programmed reduction and specific surface area assessment indicates the formation of well crystallized materials after hydrothermal treatment, without significant oxidation of V(III) species, and with interlayer, flat-oriented, carbonate anions. Reduction in H2 takes place in a single step at ca. 400 °C [Ni(II)→Ni(0)]; V(III) is not reduced. On calcination in air, Ni2V3O8 is formed, together with NiO, but crystallization is only evident after calcination at 750 °C. No major difference was found between samples with Ni:V molar ratios close to 2 or 4.

150. Density of alpha-2A adrenoceptors and Gi proteins in the human brain: ratio of high-affinity agonist sites to antagonist sites and effect of age.

Abstract

In the postmortem human brain, the specific binding of the agonists [3H]clonidine and [3H]bromoxidine ([3H]UK14034) and of the antagonist [3H]2-methoxyidazoxan (RX821002) and inhibition by l-epinephrine of [3H]RX821002 binding were determined in parallel to quantify the fractions of high (RH)- and low-affinity (RL) agonist sites of alpha-2A adrenoceptors and to investigate the association of these inhibitory receptors with various regulatory Gi proteins and the effect of aging. In the frontal cortex, the density of RH with [3H]clonidine was not different from that with [3H]UK14304 and the ratio of Bmax values in the same subjects was 0.88 +/- 0.05 (n = 39). The inhibition of [3H] RX821002 binding by l-epinephrine allowed the resolution and quantitation of the RH and RL with similar values to those obtained in saturation experiments. The simultaneous analysis and computer modeling of competition and saturation experiments performed in the same subjects indicated that a high proportion of alpha-2A adrenoceptors exist in the RH conformation (70-80%). The density of alpha-2A adrenoceptors (Bmax for [3H]agonist and [3H]antagonist binding) and age (range, 4-89 years) were negatively correlated (r = -.60, n = 39, P < .001). However, only the density of RH declined with aging (r = -.68, n = 38, P < .001); that of RL did not appear to be age related (r = -.26, n = 37, P > .05).(ABSTRACT TRUNCATED AT 250 WORDS)
