The development of bacterial resistance to chemical therapy poses a severe danger to efficacy of treating bacterial infections. One of the key factors for resistance to antimicrobial medications is growth of bacteria in biofilm. Quorum sensing (QS) inhibition was created as an alternative treatment by developing novel anti-biofilm medicines. Cell-cell communication is impeded by QS inhibition, which targets QS signaling pathway. The goal of this work is to develop newer drugs that are effective against Pseudomonas aeruginosa by decreasing QS and acting as anti-biofilm agents. In this investigation, N-(benzo[d]thiazol-2-yl)benzamide/N-(thiazol-2-yl)benzamide derivatives 3a-h were designed and synthesized in good yields. Further, molecular docking analyses revealed that binding affinity values were founded -11.2 to -7.6 kcal/mol that were moderate to good. The physicochemical properties of these prepared compounds were investigated through in-silico method. Molecular dynamic simulation was also used to know better understanding of stability of the protein and ligand complex. Comparing N-(benzo[d]thiazol-2-yl)benzamide 3a to salicylic acid (4.40±0.10) that was utilised as standard for quorum sensing inhibitor, the anti-QS action was found greater for N-(benzo[d]thiazol-2-yl)benzamide 3a (4.67±0.45) than salicylic acid (4.40±0.10). Plants respond to water shortage by regulating biochemical pathways which result in the biosynthesis of osmotic compounds. Active metabolites and compatible osmolytes control the inhibition of oxygen free radicals and dehydration. The physiological response of scrophularia striata to drought stress, a factorial completely randomized design (FCRD) experiment was conducted in three replication. Drought stress was induced at two levels (100% and 50% field capacity), and salicylic acid (SA) and silicon (Si) and Ecotype were also used at two levels of (0 and 100 PPM), (0 and 1 g/L) and (Ilam and Abdanan) respectively. Data analysis results indicated that the H 2 O 2 content, Malondialdehyde (MDA), glycine betaine (GB) and the activity of the enzyme glutathione reductase (GR; EC 1.6.4.2) of aerial parts increased during the entire stress exposure period. Although the SA + Si + stress + ecotype interaction increased the content of soluble carbohydrate s and the GR activity in aerial parts of Ilam and Abdanan ecotypes, this interaction led to a decrease in MDA, H 2 O 2 in Ilam ecotypes. The interaction between the stress + SA + Si + ecotype led to an increase in the phenylalanine ammonialyase (PAL; EC 4.3.1.5) activity in the Abdanan ecotype, but no important difference was observed. As compared to the control treatment, the content of Polyphenol increased, The interaction between ecotype + stress + Si caused to increased the of proline content in the Abadanan ecotype. Salicylic acid (SA) is an essential plant hormone that plays critical roles in basal defence and amplification of local immune responses and establishes resistance against various pathogens. However, the comprehensive knowledge of the salicylic acid 5-hydroxylase (S5H) in rice-pathogen interaction is still elusive. Here, we reported that three OsS5H homologues displayed salicylic acid 5-hydroxylase activity, converting SA into 2,5-dihydroxybenzoic acid (2,5-DHBA). OsS5H1, OsS5H2, and OsS5H3 were preferentially expressed in rice leaves at heading stage and responded quickly to exogenous SA treatment. We found that bacterial pathogen Xanthomonas oryzae pv. oryzae (Xoo) strongly induced the expression of OsS5H1, OsS5H2, and OsS5H3. Rice plants overexpressing OsS5H1, OsS5H2, and OsS5H3 showed significantly decreased SA contents and increased 2,5-DHBA levels, and were more susceptible to bacterial blight and rice blast. A simple single guide RNA (sgRNA) was designed to create oss5h1oss5h2oss5h3 triple mutants through CRISPR/Cas9-mediated gene mutagenesis. The oss5h1oss5h2oss5h3 exhibited stronger resistance to Xoo than single oss5h mutants. And oss5h1oss5h2oss5h3 plants displayed enhanced rice blast resistance. The conferred pathogen resistance in oss5h1oss5h2oss5h3 was attributed to the significantly upregulation of OsWRKY45 and pathogenesis-related (PR) genes. Besides, flg22-induced reactive oxygen species (ROS) burst was enhanced in oss5h1oss5h2oss5h3. Collectively, our study provides a fast and effective approach to generate rice varieties with broad-spectrum disease resistance through OsS5H gene editing., (© 2023 The Authors. Isatis spp. are well-known for their industrial significance due to natural sources of indigotin and indirubin, important indole alkaloids, used in the dye and pharmaceutical industries. In this study, silver nanoparticles (AgNP) and salicylic acid-chitosan nanoparticles (SA-CNP) were synthesized and applied to enhance the production of indigotin and indirubin in shoot and root cultures of Isatis tinctoria and Isatis ermenekensis. Different doses of AgNP and SA-CNP were administered to three-week-old shoot and root cultures, and the effects were assessed at 12, 24, and 48 h. The harvested samples were analyzed to quantify indigotin and indirubin levels. Furthermore, the expression levels of It-TSA and CYP79B2 genes, known to be involved in indole alkaloid biosynthesis, were determined. In I. tinctoria roots, the highest levels of indigotin and indirubin were observed after applying 150 mg L -1 of SA-CNP for 48 h while in I. ermenekensis shoots, indigotin and indirubin reached the maximum levels with the application of 8 mg L -1 AgNP for 48 h. NP application had no remarkable effects on the accumulation of indigotin and indirubin in I. tinctoria shoots and I. ermenekensis roots compared to controls. Additionally, shoot cultures demonstrated superior indirubin production, which significantly increased with AgNP applications. The gene expression analysis also exhibited significant correlations with the changes in indigotin and indirubin levels. Exposure of plants to adverse environmental conditions reduces their growth and productivity. Currently, seed priming with phytohormones is considered one of the most reliable and cost-effective approaches that can help alleviate the toxic effects of environmental stress. In this context, the present study aims to investigate the effect of priming alfalfa seeds with salicylic acid (SA) on oxidative stress markers, including malonyldialdehyde, protein content, activities of antioxidant enzymes, and expression of genes encoding these enzymes in leaves and roots of alfalfa (Gabes ecotype) grown under saline stress, iron deficiency, or both. Our results showed that the application of salt stress and iron deficiency separately or simultaneously induces changes in the activities of antioxidant enzymes, but these are organ- and stress-dependent. The Gabes ecotype was able to increase the activities of these enzymes under salt stress to alleviate oxidative damage. Indeed, priming seeds with 100 μM SA significantly increases the enzymatic activities of APX, GPX, CAT, and SOD. Therefore, this concentration can be considered optimal for the induction of iron deficiency tolerance. Our results showed not only that Gabes ecotype was able to tolerate salt stress by maintaining high expression of the Fe-SOD isoform, but also that the pretreatment of seeds with 100 μM SA improved the tolerance of this ecotype to iron deficiency by stimulating Fe-SOD expression and inhibiting CAT and APXc., (© 2023 Scandinavian Plant Physiology Society.)

An experiment was performed to investigate the effect of mycorrhizal symbiosis and foliar application of salicylic acid on quantitative and qualitative traits of maize during 2018 and 2019 in the research farm of Islamic Azad University, Chalous Branch. Split plot in a randomized complete block design with three replications was used. Experimental factors included mycorrhiza species of (G. mosseae), (G. geosporum) and (G. intraradices) at two levels (no consumption and consumption of mycorrhiza) and salicylic acid at two levels (no consumption and consumption of 1 mμ of salicylic acid). Results of interaction effects of mycorrhiza and salicylic acid on the measured traits revealed that the maximum 1000-grain weight, grain yield, biological yield, phosphorus, potassium, nitrogen percentage and yield of maize grain protein were observed in G. mosseae treatment under foliar application of salicylic acid. Foliar application of salicylic acid increases the root length and provides the necessary conditions for increasing water and nutrient uptake alongwith increase in photosynthesis and thus allocates more photosynthetic substance for development of reproductive organs. Hence, it increases maize grain weight and accordingly grain yield. In general, the results revealed that mycorrhiza and foliar application of salicylic acid increase growth indicators, yield and yield components. It also improved the quality traits of the maize plant. Based on results, the interaction effect of G. mosseae treatment and foliar application of salicylic acid yielded better results than other treatments. Mycorrhiza increases the number of grain in the ear, the number of rows in the ear, increases the plant's ability to absorb phosphorus, and the increase of mycorrhiza along with salicylic acid shows the maximum grain yield in maize. Finally, it can be concluded that the use of mycorrhiza and salicylic acid can be effective in increasing grain in the plant.

Plants release a mixture of volatile compounds when subjects to environmental stress, allowing them to transmit information to neighboring plants. Here, we find that Nicotiana benthamiana plants infected with tobacco mosaic virus (TMV) induces defense responses in neighboring congeners. Analytical screening of volatiles from N. benthamiana at 7 days post inoculation (dpi) using an optimized SPME-GC-MS method showed that TMV triggers the release of several volatiles, such as (E)-2-octenal, 6-methyl-5-hepten-2-one, and geranylacetone. Exposure to (E)-2-octenal enhances the resistance of N. benthamiana plants to TMV and triggers the immune system with upregulation of pathogenesis-related genes, such as NbPR1a, NbPR1b, NbPR2, and NbNPR1, which are related to TMV resistance. Furthermore, (E)-2-octenal upregulates jasmonic acid (JA) that levels up to 400-fold in recipient N. benthamiana plants and significantly affects the expression pattern of key genes in the JA/ET signaling pathway, such as NbMYC2, NbERF1, and NbPDF1.2, while the salicylic acid (SA) level is not significantly affected. Our results show for the first time that the volatile (E)-2-octenal primes the JA/ET pathway and then activates immune responses, ultimately leading to enhanced TMV resistance in adjacent N. benthamiana plants. Caffeine (CAF) and salicylic acid (SA) are frequently detected in waterbody, though information on their biological impact is poor. This work assesses the effects of CAF (5 ng/L to 10 µg/L) and SA (0.05 µg/L to 100 µg/L) alone and combined as CAF+SA (5 ng/L+0.05 µg/L to 10 µg/L+100 µg/L) on mussel Mytilus galloprovincialis under 12-days exposure by histomorphology of digestive gland and oxidative stress defense at molecular and biochemical levels. Besides evaluating tissue accumulation, absence of histomorphological damage and haemocyte infiltration highlighted activation of defensive mechanisms. Up-regulation of Cu/Zn-sod, Mn-sod, cat and gst combined with increased catalase and glutathione S-transferase activity were found in CAF-exposed mussels, while SA reduced ROS production and mitochondrial activity. CAF+SA exposure induced differential responses, and the integrated biomarker response (IBR) revealed more pronounced effects of SA than CAF. In plant immunity, the mutually antagonistic hormones salicylic acid (SA) and jasmonic acid (JA) are implicated in resistance to biotrophic and necrotrophic pathogens, respectively. Promoters that can respond to both SA and JA signals are urgently needed to engineer plants with enhanced resistance to a broad spectrum of pathogens. However, few natural pathogen-inducible promoters are available for this purpose. To address this problem, we have developed a strategy to synthesize dual SA- and JA-responsive promoters by combining SA- and JA-responsive cis elements based on the interaction between their cognate trans-acting factors. The resulting promoters respond rapidly and strongly to both SA and Methyl Jasmonate (MeJA), as well as different types of phytopathogens. When such a synthetic promoter was used to control expression of an antimicrobial peptide, transgenic plants displayed enhanced resistance to a diverse range of biotrophic, necrotrophic, and hemi-biotrophic pathogens. A dual-inducible promoter responsive to the antagonistic signals auxin and cytokinin was generated in a similar manner, confirming that our strategy can be used for the design of other biotically or abiotically inducible systems., (Copyright © 2023 The Author(s). Salt and osmotic stress seriously restrict the growth, development, and productivity of horticultural crops in the greenhouse. The papain-like cysteine proteases (PLCPs) participate in multi-stress responses in plants. We previously demonstrated that salt and osmotic stress affect cysteine protease 15 of pepper ( Capsicum annuum L.) ( CaCP15 ); however, the role of CaCP15 in salt and osmotic stress responses is unknown. Here, the function of CaCP15 in regulating pepper salt and osmotic stress resistance was explored. Pepper plants were subjected to abiotic (sodium chloride, mannitol, salicylic acid, ethrel, methyl jasmonate, etc.) and biotic stress ( Phytophthora capsici inoculation). The CaCP15 was silenced through the virus-induced gene silencing (VIGS) and transiently overexpressed in pepper plants. The full-length CaCP15 fragment is 1568 bp, with an open reading frame of 1032 bp, encoding a 343 amino acid protein. CaCP15 is a senescence-associated gene 12 (SAG12) subfamily member containing two highly conserved domains, Inhibitor 129 and Peptidase_C1. CaCP15 expression was the highest in the stems of pepper plants. The expression was induced by salicylic acid, ethrel, methyl jasmonate, and was infected by Phytophthora capsici inoculation. Furthermore, CaCP15 was upregulated under salt and osmotic stress, and CaCP15 silencing in pepper enhanced salt and mannitol stress resistance. Conversely, transient overexpression of CaCP15 increased the sensitivity to salt and osmotic stress by reducing the antioxidant enzyme activities and negatively regulating the stress-related genes. This study indicates that CaCP15 negatively regulates salt and osmotic stress resistance in pepper via the ROS-scavenging.

Origanum vulgare L., a medicinal and aromatic herb, has been used for hundreds of years. This plant contains valuable chemical compounds that can be used as medicine for treatment. On the other hand, a gradual increase in the planet's average temperature could negatively affect the growth and the composition of the O. vulgare. For this reason, in this study, the effect of two protective compounds, namely salicylic acid (SA) and gamma-aminobutyric acid (GABA), on temperature and salinity stress conditions was investigated. Oregano plants were grown at 23/12°C temperature as control and 27/16°C heat stress in the greenhouse (16/8 h photoperiod) for one months. The plants were treated with GABA and SA and subjected to salt stress for 30 days. Subsequently, the plant's physiological, biochemical, and phytochemical characteristics were examined. The results showed that all studied traits (in control and treated samples) were significantly different at 27°C, from 23°C. In addition, the highest amount of thymol and carvacrol were detected from plants grown at 27°C. In regards to salinity, stressed- plants had less damage to membrane stability and H2O2 level, when treated with GABA or SA. This study revealed that both SA and GABA compounds had an exellent protective effect on temperature and salt stress on O. vulgare. Based on enzyme-pigment evaluations and secondary metabolites, SA showed a better protective effect on temperature effects and GABA in a saline environment. In general, using these compounds can provide better conditions for the growth and preservation of O. vulgare chemical compounds. The identification of several fructan exohydrolases (FEHs, EC 3.2.1.80) in non-fructan accumulating plants raised the question of their roles. FEHs may be defense-related proteins involved in the interactions with fructan-accumulating microorganisms. Since known defense-related proteins are upregulated by defense-related phytohormones, we tested the hypothesis that FEHs of non-fructan accumulating plants are upregulated by salicylic acid (SA), jasmonic acid (JA) and ethylene (ET) using the model plant Arabidopsis thaliana and the agronomically relevant and genetically related species Brassica napus. By sequence homologies with the two known FEH genes of A. thaliana, At6-FEH, and At6&1-FEH, the genes coding for the putative B. napus FEHs, Bn6-FEH and Bn6&1-FEH, were identified. Plants were treated at root level with SA, methyl jasmonate (MeJA) or 1-aminocyclopropane-1-carboxylic acid (ACC). The transcript levels of defense-related and FEH genes were measured after treatments. MeJA and ACC did not upregulate FEHs, while HEL (HEVEIN-LIKE PREPROTEIN) expression was enhanced by both phytohormones. In both species, the expression of AOS, encoding a JA biosynthesis enzyme, was enhanced by MeJA and that of the defensine PDF1.2 and the ET signaling transcription factor ERF1/2 by ACC. In contrast, SA not only increased the expression of genes encoding antimicrobial proteins (PR1 and HEL) and the defense-related transcription factor WRKY70 but also that of FEH genes, in particular 6&1-FEH genes. This result supports the putative role of FEHs as defense-related proteins. The water-soluble coordination polymer of formula {[Pb(Sal) 2 (H 2 O)] n } (SaLead), was obtained from the reaction between Pb(NO 3 ) 2 and the potassium salt of salicylic acid (SalH), an anti-inflammatory drug, which is also use as food preservation, in cosmetics etc. The compound was characterized by melting point, Attenuated Total Reflectance-Fourier Transform Infra-Red (ATR-FTIR) spectroscopy and X-ray diffraction crystallography (XRD) in solid state and in solution by Ultra Violet (UV) and 1 H NMR spectroscopies. The binding affinity of SalK to Pb(II) ions towards SaLead was determined in order to examine its possible implementation in lead detoxification. The in vitro non-toxic behaviour of SalK and its complex SaLead was evaluated against normal human fetal lung fibroblast cells (MRC-5). The corresponding IC 50 values are 260 ± 13 and > 1600 μM respectively. GDSL esterases/lipases are a subclass of lipolytic enzymes that play critical roles in plant growth and development, stress response, and pathogen defense. However, the GDSL esterase/lipase genes involved in the pathogen response of apple remain to be identified and characterized. Thus, in this study, we aimed to analyze the phenotypic difference between the resistant variety, Fuji, and susceptible variety, Gala, during infection with C. gloeosporioides , screen for anti-disease-associated proteins in Fuji leaves, and elucidate the underlying mechanisms. The results showed that GDSL esterase/lipase protein GELP1 contributed to C. gloeosporioides infection defense in apple. During C. gloeosporioides infection, GELP1 expression was significantly upregulated in Fuji. Fuji leaves exhibited a highly resistant phenotype compared with Gala leaves. The formation of infection hyphae of C. gloeosporioides was inhibited in Fuji. Moreover, recombinant His:GELP1 protein suppressed hyphal formation during infection in vitro. Transient expression in Nicotiana benthamiana showed that GELP1-eGFP localized to the endoplasmic reticulum and chloroplasts. GELP1 overexpression in GL-3 plants increased resistance to C. gloeosporioides . MdWRKY15 expression was upregulated in the transgenic lines. Notably, GELP1 transcript levels were elevated in GL-3 after salicylic acid treatment. These results suggest that GELP1 increases apple resistance to C. gloeosporioides by indirectly regulating salicylic acid biosynthesis.

In this study we evaluated the acute (immobility/mortality) and chronic (survival and reproduction) effects of the drugs caffeine, diclofenac sodium salt, ketoprofen, paracetamol and salicylic acid on the cladoceran Ceriodaphnia silvestrii. The environmental risks of these substances for tropical freshwaters were estimated from the risk quotient MEC/PNEC. Sensitivity in acute exposures varied up on the drug as follows: salicylic acid (EC 50  = 69.15 mg L - 1 ) < caffeine (EC 50  = 45.94 mg L - 1 ) < paracetamol (EC 50  = 34.49 mg L - 1 ) < ketoprofen (EC 50  = 24.84 mg L - 1 ) < diclofenac sodium salt (EC 50  = 14.59 mg L - 1 ). Chronic toxicity data showed negative effects of the drugs on reproduction. Paracetamol and salicylic acid caused reduction in fecundity in concentrations starting from 10 mg L - 1 and 35 mg L - 1 , respectively. Ketoprofen caused total inhibition at 5 mg L - 1 . Two significant soil degradation processes that pose a hazard to our ecosystems are soil salinization and sodification. The information on potential of salicylic acid (SA) and trehalose (Tre) to induce abiotic stress signaling and triggers physio-biochemical responses in crop plants is limited. Therefore, the present study was aimed to investigate the efficacy of 5 μM SA and/or 10 mM Tre in improving the growth, photosynthesis, ion homeostasis, nutrient acquisition, antioxidant defense system and yield of mustard plants growing under sodium chloride (NaCl) stress (0, 50, 100 and 150 mM NaCl). The data showed that increasing NaCl stress concentration decreased growth, photosynthesis, membrane permeability, ion homeostasis and yield in a dose-dependent manner while increasing considerably enzymatic antioxidant enzyme activities, compatible solute accumulation, sodium ion and oxidative stress biomarkers linearly with increasing NaCl stress concentration. The spray of SA, Tre, and SA + Tre played diversified roles in enhancing NaCl stress tolerance in mustard at morpho-physiological and biochemical levels. The combined SA + Tre application proved best and completely neutralized the NaCl stress-induced suppression in growth, photosynthesis, ion homeostasis, nutrient acquisition and yield by significantly enhancing the activities of enzymatic antioxidants, compatible solutes accumulation, water status and membrane permeability, while reducing considerably osmotic stress, reactive oxygen species generation, lipid peroxidation, cell death and sodium uptake in mustard. The SA + Tre application enhanced relative water content by 23%, net photosynthetic rate by 48%, superoxide dismutase activity by 51% and seed yield per plant by 64%, while decreased superoxide anion content by 26%, sodium ion content by 36% and malondialdehyde content by 25% over 0 mM NaCl treatment. Ralstonia solanacearum, the causal agent of the devastating bacterial wilt disease, is of particular interest to the scientific community. The repertoire of type III effectors plays an important role in the evasion of plant immunity, but tolerance to plant immunity is also crucial for the survival and virulence of R. solanacearum. Nevertheless, a systematic study of R. solanacearum tolerance to plant immunity is lacking. In this study, we used exogenous salicylic acid (SA) to improve the immunity of tomato plants, followed by transposon insertion sequencing (Tn-seq) analysis and the identification of R. solanacearum genes associated with tolerance to plant immunity. Target gene deletion revealed that the lipopolysaccharide (LPS) production genes RS&#95;RS02830, RS&#95;RS03460, and RS&#95;RS03465 are essential for R. solanacearum tolerance to plant immunity, and their expression is induced by plant immunity, thereby expanding our knowledge of the pathogenic function of R. solanacearum LPS. SA treatment increased the relative abundance of transposon insertion mutants of four genes, including two genes with unknown function, RS&#95;RS11975 and RS&#95;RS07760. Further verification revealed that deletion of RS&#95;RS11975 or RS&#95;RS07760 resulted in reduced in vivo competitive indexes but increased tolerance to plant immunity induced by SA treatment, suggesting that these two genes contribute to the trade-off between tolerance to plant immunity and fitness cost. In conclusion, this work identified and validated R. solanacearum genes required for tolerance to plant immunity and provided essential information for a more complete view of the interaction between R. solanacearum and the host plant., (© 2023 The Authors. Hemidesmus indicus (L.) R. Br. ex Schult. and Tylophora indica (Burm. F.) Merrill shoot cultures were treated with different concentrations of yeast extract (YE; 25-200 mg/L) and salicylic acid (SA; 50-200 µM), and their effect on lupeol production was assessed. The maximum dry weight (DW) biomass was recorded when H. indicus shoots were treated with SA (50 µM) and T. indica shoots with YE (200 mg/L). Highest lupeol yield (335.40 ± 0.04 µg/g DW) was obtained in H. indicus shoots after treatment with 50 µM of SA for 3 weeks. Whereas in T. indica , maximum lupeol content (584.26 ± 8.14 µg/g DW) was recorded by giving treatment with 25 μM of SA for 6 weeks.

Afgekia mahidolae is a rare plant species that possesses antioxidant, antimicrobial, and wound healing properties. This study aimed to establish the in vitro culture of A. mahidolae and investigate the effects of elicitors on their phenolic and flavonoid production, including the antioxidant activities. The established callus was prepared in the form of cell suspension cultures to determine the effect of elicitors. After elicitation for 3 days, A. mahidolae cell suspension cultures treated by 5 µM salicylic acid or 100 mg/L yeast extract exhibited significantly higher levels of total phenolic and total flavonoid content than untreated cultures, which correlated to the antioxidant activities. In addition, the profiles of phenolic and flavonoid compounds in the callus and intact leaves of A. mahidolae were determined by LC-MS, which showed different phytochemicals. The findings of this study may encourage more sustainable development of A. mahidolae cultivation.

Considering the relevance of bell pepper and the limitations imposed by the deleterious effects of salt stress, especially in semi-arid regions, it is extremely important to establish strategies that can facilitate the use of saline water in vegetable production. In this scenario, this study aimed to evaluate the effect of the frequency of foliar application of salicylic acid on the morphophysiology and production of the bell pepper cv. "All Big" irrigated with saline water. The study was conducted at a greenhouse in Campina Grande - PB. The treatments were distributed in a completely randomized design and set up in a 4 × 4 factorial arrangement with three replications, corresponding to four application frequencies of salicylic acid (F1- No application of salicylic acid, F2 - Weekly application, F3- fortnightly application, and F4- monthly application) and four levels of electrical conductivity of irrigation water - ECw (0.8, 1.6, 2.4 and 3.2 dS m-1). The fortnightly application of salicylic acid at a concentration of 1.0 mM mitigated the effects of salt stress on the morphophysiology and production components of bell pepper cv. All Big cultivated with ECw of up to 2.4 dS m-1, which reinforces the hypothesis that salicylic acid can act as a signaling molecule and reduce the effects of saline stress in bell pepper, enabling the use of brackish water in agricultural activity, mainly in semi-arid regions of northeastern Brazil, which have a shortage of fresh water.

Background: As a keratolytic, salicylic acid (SA) can be topically applied in various formulations and doses in dermatology. Supramolecular SA hydrogel, a new SA formulation with higher bioavailability, is developed and commercially available nowadays. However, there still remain concerns that the long-term and continual application of SA at low concentrations may jeopardize the cutaneous barrier properties., Aim of the Study: To reveal the long-term effects of 0.5-5% supramolecular SA hydrogel on the skin barrier in normal mice models., Materials and Methods: The 0.5%, 1%, 2%, and 5% supramolecular SA hydrogel or hydrogel vehicle without SA was applied to mice's shaved dorsal skin once per day respectively. Tissue samples of the dorsal skin were harvested on day 14 and 28 of the serial application of SA for histopathological observation and transcriptomic analysis., Results: Following topical supramolecular SA hydrogel therapy with various concentrations of SA (0.5%, 1%, 2%, and 5%) for 14 days and 28 days, there were no obvious macroscopic signs of impaired cutaneous health and no inflammatory or degenerative abnormalities were observed in histological results. Additionally, the transcriptomic analysis revealed that on day 14, SA dramatically altered the expression of genes related to the extracellular matrix structural constituent. And on day 28, SA regulated gene expression profiles of keratinization, cornified envelope, and lipid metabolism remarkably. Furthermore, the expression of skin barrier related genes was significantly elevated after the application of SA based on RNA-seq results, and this is likely to be associated with the PPAR signaling pathway according to the enrichment analysis., Conclusion: Our findings demonstrated that the sustained topical administration of the 0.5-5% supramolecular SA hydrogel for up to 28 days did no harm to normal murine skin and upregulated the expression of genes related to the epidermal barrier., Competing Interests: The authors declare no conflicts of interest in this work., (© 2023 Zhou et al.)

Oplopanax elatus is a valuable medicinal plant, but its plant resource is lacking. Adventitious root (AR) culture of O. elatus is an effective way for the production of plant materials. Salicylic acid (SA) exerts enhancement effect on metabolite synthesis in some plant cell/organ culture systems. To clarify the elicitation effect of SA on fed-batch cultured O. elatus ARs, this study investigated the effects of SA concentration, and elicitation time and duration. Results showed that flavonoid and phenolic contents, and antioxidant enzyme activity obviously increased when the fed-batch cultured ARs were treated with 100 μM SA for 4 days starting on day 35. Under this elicitation condition, total flavonoid and phenolic contents reached 387 rutin mg/g DW and 128 gallic acid mg/g DW, respectively, which were significantly (p < 0.05) higher than those in the SA-untreated control. In addition, DPPH scavenging and ABTS + scavenging rates, and Fe 2+ chelating rate also greatly increased after SA treatment, and their EC 50 values were 0.0117, 0.61, and 3.34 mg/L, respectively, indicating the high antioxidant activity. Systemic acquired resistance (SAR) is a potent innate immunity system in plants that is induced through the salicylic acid (SA)-mediated signaling pathway. Here, we characterized 3-chloro-1-methyl-1 H -pyrazole-5-carboxylic acid (CMPA) as an effective SAR inducer in Arabidopsis . The soil drench application of CMPA enhanced a broad range of disease resistance against the bacterial pathogen Pseudomonas syringae and fungal pathogens Colletotrichum higginsianum and Botrytis cinerea in Arabidopsis , whereas CMPA did not show antibacterial activity. Foliar spraying with CMPA induced the expression of SA-responsible genes such as PR1 , PR2 and PR5 . The effects of CMPA on resistance against the bacterial pathogen and the expression of PR genes were observed in the SA biosynthesis mutant, however, while they were not observed in the SA-receptor-deficient npr1 mutant. Thus, these findings indicate that CMPA induces SAR by triggering the downstream signaling of SA biosynthesis in the SA-mediated signaling pathway.

The hormones salicylic acid (SA) and jasmonic acid (JA) often act antagonistically in controlling plant defense pathways in response to hemibiotrophs/biotrophs (hemi/biotroph) and herbivores/necrotrophs, respectively. Threonine deaminase (TD) converts threonine to α-ketobutyrate and ammonia as the committed step in isoleucine (Ile) biosynthesis and contributes to JA responses by producing the Ile needed to make the bioactive JA-Ile conjugate. Tomato (Solanum lycopersicum) plants have two TD genes: TD1 and TD2. A defensive role for TD2 against herbivores has been characterized in relation to JA-Ile production. However, it remains unknown whether TD2 is also involved in host defense against bacterial hemi/biotrophic and necrotrophic pathogens. Here, we show that in response to the bacterial pathogen-associated molecular pattern (PAMP) flagellin flg22 peptide, an activator of SA-based defense responses, TD2 activity is compromised, possibly through carboxy-terminal cleavage. TD2 knockdown (KD) plants showed increased resistance to the hemibiotrophic bacterial pathogen Pseudomonas syringae but were more susceptible to the necrotrophic fungal pathogen Botrytis cinerea, suggesting TD2 plays opposite roles in response to hemibiotrophic and necrotrophic pathogens. This TD2 KD plant differential response to different pathogens is consistent with SA- and JA-regulated defense gene expression. flg22-treated TD2 KD plants showed high expression levels of SA-responsive genes, whereas TD2 KD plants treated with the fungal PAMP chitin showed low expression levels of JA-responsive genes. Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb) is a leading cause of infectious disease mortality. The salicylic acid derived small molecule siderophores known as mycobactins are essential in vivo for iron acquisition of Mtb where iron is restricted in the host. Herein, we synthesize and explore the mechanism of action of polyfluorinated salicylic acid derivates, which were previously reported to possess potent antimycobacterial activity. We hypothesized fluorinated salicylic acid derivates may inhibit mycobactin biosynthesis through initial bioactivation and conversion to downstream metabolites that block late steps in assembly of the mycobactins. Enzymatic studies demonstrated that some of the fluorinated salicylic acid derivatives compounds were readily activated by the bifunctional adenylating enzyme MbtA, responsible for incorporation of salicylic acid into the mycobactin biosynthetic pathway; however, they did not inhibit mycobactin biosynthesis as confirmed by LS-MS/MS using an authentic synthetic mycobactin standard. Further mechanistic analysis of the most active derivative (Sal-4) using an MbtA-overexpressing Mtb strain as well as complementation studies with iron and salicylic acid revealed Sal-4 cannot be antagonized by overexpression of MbtA or through supplementation with iron or salicylic acid. Gentamicin (GM) is a broadly used antibiotic against severe and life-threatening infections, but its efficacy is restricted by the development of liver toxicity. The present study was designed to evaluate the protective effect of salicylic acid (SA) in gentamicin-induced hepatotoxicity in rabbits. Gentamicin and salicylic acid were given at a dose of 80 mg/kg i.p for twenty days. For this purpose, 24 male albino rabbits were randomly divided into four groups. Group I remained untreated and served as control. Group II was given gentamicin, group III was given gentamicin along with Salicylic acid (SA) and group IV was given only salicylic acid. The degree of hepatoprotection was measured by assessment of body weight, liver weight, absolute liver weight and estimations of plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), total and direct bilirubin, tissue malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) activities. Significant reduction in the elevated liver weight, plasma levels of AST, ALT, bilirubin and tissue MDA and significant elevation in reduced body weight, SOD and CAT activities were found that confirms the protective role of salicylic acid in gentamicin induced hepatotoxicity.

Acmella radicans (Asteraceae) is a plant native to America. Despite it having medicinal attributes, studies on its phytochemical properties are scarce, and biotechnological studies do not exist for this species. In this study, we established an adventitious root culture from A. radicans internodal segments in shake flasks with indole-3-butyric acid (IBA), and then elicited it with jasmonic acid (JA) and salicylic acid (SA). The total phenolic content and antioxidant activity were evaluated, and a comparison was made using in vitro plantlets and wild plants. Internodal segments with 0.1 mg/L IBA showed 100% root induction and exhibited better growth after transfer to shake flasks with MS liquid culture medium. JA had a significant effect on biomass increase compared to unelicited roots, mainly with 50 µM JA (28%), while SA did not show significant results. Root elicited with 100 µM (SA and JA) showed a 0.34- and 3.9-fold increase, respectively, in total phenolic content (TPC) compared to the control. The antioxidant activity was also significant, and a lower half-maximal inhibitory concentration (IC 50 ) was observed as the AJ concentration increased. Roots elicited with AJ (100 µM) exhibited high antioxidant activity with DPPH (IC 50 = 9.4 µg/mL) and ABTS (IC 50 = 3.3 µg/mL) assays; these values were close to those for vitamin C (IC 50 = 2.0 µg/mL). The TPC and antioxidant activity of in vitro plants and root cultured in shake flasks showed the lowest values in most cases; even the root cultures without elicitation were better than those of a wild plant. In this study, we demonstrated that A. radicans root culture is capable of producing secondary metabolites, while its production and antioxidant activity can be enhanced using jasmonic acid.

Flooding can damage the photosynthetic apparatus and initial growth of Schinus terebinthifolia. We aimed this study evaluates the potential of silicon (Si) and salicylic acid (SA) as mitigating agents on the ecophysiological responses and initial growth of S. terebinthifolia subjected to flooding periods. Seedlings were grown under the following conditions: 1) control (non-flooded): daily irrigation, 2) Flooded (F): storage of seedlings in a 500 L plastic pool, keeping the water depth at ± 2.0 cm above the substrate level, 3) F + 1.0 mM Si, 4) F + 2.0 mM Si, 5) F + 1.5 mM SA, and 6) F + 3.0 mM SA, and evaluated to 15 and 30 days. We observed that flooded seedlings formed hypertrophied lenticels on the stem on the 7th day as a stress adjustment strategy. S. terebinthifolia is sensitive to flooding, although it maintains a stable gas exchange for up to 15 days in this condition. The applications of 1.0 mM Si mitigated the pronounced decrease of gas exchange by 30 days. Using 1.0 mM Si and 3.0 mM SA contributed for integrity of the photosynthetic apparatus and to photochemical activities in the reaction centers, in addition favors to higher seedling biomass and quality under flooding. Foliar application of Si and SA is promising practice for photosynthetic metabolic and initial growth of S. terebinthifolia seedlings under flooded stress.

Background: The growth and yield of pepper (Capsicum annuum L.) is often affected by the critical salt stress. Salicylic acid (SA) can improve plants' stress tolerance by promoting growth and regulating ion absorption and transportation., Methods and Results: To uncover the alleviated mechanism of salt stress by SA in pepper, we conducted morphological, physiological, cytological, and transcriptomic analyses under a single SA treatment and NaCl with and without SA pre-treatment for 9 days. Seedlings under NaCl treatment showed yellow shrunken leaves, this tatus were alleviated by NS treatment (NaCl with SA pre-treatment). Compared with plants under NaCl treatment, those in the NS treatment showed reduced lipid peroxidation, and significantly increased contents of chlorophyll and osmotic regulators (proline, soluble sugars). Treatment with SA balanced the Na + /K + ratio. We conducted transcriptome sequencing and identified differentially expressed genes (DEGs) contributing to alleviation of salt stress by SA in pepper. Besides photosynthesis related genes, GO and KEGG analyses revealed that the DEGs were enriched in 'sequence-specific DNA binding', 'transcription regulator activity' and 'DNA binding transcription factor activity' by GO terms. And our results showed that TFs, such as MYB, bZIP, BBX, AP2/ERF, NAC, etc., probably make a great contribution in the alleviation of salt stress by SA., Conclusions: These results reveal that SA can improve plants' stress tolerance by balancing ion absorption, gene expression and transcriptional regulation, which provide new ideas and resources for subsequent research on the mechanism of salt tolerance in pepper., (© 2022. Heat stress (HS) has become a serious threat to crop growth and yield. Sulfur dioxide (SO 2 ) is being verified as a signal molecule in regulating the plant stress response. However, it is unknown whether SO 2 plays a significant role in the plant heat stress response (HSR). Herein, maize seedlings were pretreated with various concentrations of SO 2 and then kept at 45 °C for heat stress treatment, aiming to study the effect of SO 2 pretreatment on HSR in maize by phenotypic, physiological, and biochemical analyses. It was found that SO 2 pretreatment greatly improved the thermotolerance of maize seedlings. The SO 2 -pretreated seedlings showed 30-40% lower ROS accumulation and membrane peroxidation, but 55-110% higher activities of antioxidant enzymes than the distilled water-pretreated seedlings under heat stress. Interestingly, endogenous salicylic acid (SA) levels were increased by ∼85% in SO 2 -pretreated seedlings, as revealed by phytohormone analyses. Furthermore, the SA biosynthesis inhibitor paclobutrazol markedly reduced SA levels and attenuated SO 2 -triggered thermotolerance of maize seedlings. Meanwhile, transcripts of several SA biosynthesis and signaling, and heat stress-responsive genes in SO 2 -pretreated seedlings were significantly elevated under HS. These data have demonstrated that SO 2 pretreatment increased endogenous SA levels, which activated the antioxidant machinery and strengthened the stress defense system, thereby improving the thermotolerance of maize seedlings under HS. Melatonin is a pleiotropic molecule that can influence various aspects of plant performance. Recent studies have exhibited that it mediates plant defensive responses, probably through managing redox homeostasis. We tried to track the regulatory effects of melatonin on the antioxidant machinery of Linum album cell culture. To this, different concentrations of melatonin were applied, and the oxidative status of cells was investigated by measuring the levels of oxidative molecules and antioxidant agents. The results showed that H 2 O 2 content did not change at the low melatonin levels, while it increased at the high concentrations. It can be correlated with the low melatonin dosages capacity to remove excessive amounts of H 2 O 2 , while the high melatonin dosages exhibit toxicity effects. In contrast, the NO enhancement occurred at 50 μM melatonin, proposing its role in triggering melatonin-induced defensive responses. The MDA results stated that NO led to oxidative stress in melatonin-treated cells at 50 μM melatonin. Antioxidant enzyme POD was activated by melatonin treatment, while SOD enzyme behaved reversely which can explain the changes in the H 2 O 2 level. In addition, the analysis of the phenolics profile showed that the contents of phenolic acids, flavonoids, and lignans enhanced following an increase in PAL enzyme activity. The increased level of phenolic hormone SA can indicate that melatonin affects the defensive responses in L. album cells through a SA-dependent pathway. The reproductive stage of plant development has the most critical impact on yield. Flowering is highly sensitive to abiotic stress, and increasing temperatures and drought harm crop yields. Salicylic acid is a phytohormone that regulates flowering and promotes stress resilience in plants. However, the exact molecular mechanisms and the level of protection are far from understood and seem to be species-specific. Here, the effect of salicylic acid was tested in a field experiment with Pisum sativum exposed to heat stress. Salicylic acid was administered at two different stages of flowering, and its effect on the yield and composition of the harvested seeds was followed. Plants treated with salicylic acid produced larger seed pods, and a significant increase in dry weight was found for the plants with a delayed application of salicylic acid. The analyses of the seed proteome, lipidome, and metabolome did not show any negative impact of salicylic treatment on seed composition. Identified processes that could be responsible for the observed improvement in seed yields included an increase in polyamine biosynthesis, accumulation of storage lipids and lysophosphatidylcholines, a higher abundance of components of chromatin regulation, calmodulin-like protein, and threonine synthase, and indicated a decrease in sensitivity to abscisic acid signaling.

Background: Cold stress is an effective factor in reducing production and injuring fruit trees. Various materials, such as salicylic acid, ascorbic acid, and putrescine, are used to alleviate the damage of abiotic stress., Results: The effect of different treatments of putrescine, salicylic acid, and ascorbic acid on alleviating the damage of frost stress (- 3 °C) to grapes 'Giziluzum' was investigated. Frost stress increased the amount of H 2 O 2 , MDA, proline, and MSI. On the other hand, it decreased the concentration of chlorophyll and carotenoids in the leaves. Putrescine, salicylic acid and ascorbic acid significantly increased the activities of catalase, guaiacol peroxidase, ascorbate peroxidase, and superoxide dismutase under frost stress. Following frost stress, the grapes treated with putrescine, salicylic acid, and ascorbic acid showed higher levels of DHA, AsA, and AsA/DHA than the untreated grapes. Our results showed that the treatment with ascorbic acid outperformed the other treatments in adjusting frost stress damages., Conclusion: The use of compounds, such as ascorbic ac id, salicylic acid, and putrescine, modulates the effects of frost stress, thereby increasing the antioxidant defense system of cells, reducing its damage, and stabilizing stable cell conditions, so it can be used to reduce frost damage to different grape cultivars., (© 2023. Bakusylan (bakuchiol salicylate) is a bipartite compound obtained by merging two skin-active entities with complementary bioactivities-bakuchiol and salicylic acid-for the purpose of generating a new class of functional retinoids with enhanced skin benefits. Here, we describe its preparation process and report that pure bakusylan exhibits potential for an improved permeation through the stratum corneum, enhances type IV collagen gene expression in organotypic skin substitutes containing both epidermal and dermal layers, and upregulates this protein in adult human dermal fibroblast cultures. The mechanism of action underlying these effects appears to involve the components of the IP3K/Akt signaling pathway selectively implicated in the maintenance of skin integrity, further underlying the suitability of this ester for skin care applications requiring enhanced cutaneous permeation targeting the dermal-epidermal junction.

Hormones play an important role in plant disease resistance and defense. Transcriptome data of late blight-resistant potato genotype SD20 treated by ethylene (ET), jasmonate (JA), salicylic acid (SA), and Phytophthora infestans CN152 was analyzed to assess the role of the ET/JA/SA regulatory network in plant disease resistance and defense and predict key resistant genes. The results show that there was significant crossover of differentially expressed genes among all treatments, and common and specific plant disease interaction genes for the ET, JA, and SA treatments were differentially expressed in the CN152 treatment. The resistance and defense genes of the potato genotype SD20 could be induced to regulate metabolic and hormone signaling pathways by alternative splicing in all treatments. Further analysis found that JA and ET pathways can work together synergistically. JA/ET and SA pathways antagonize each other to initiate the expression of calmodulin-domain protein kinases and calmodulin/calmodulin-like and RPM1-interacting protein 4 genes, and they activate HSP- mediated hypersensitive response and defense-related genes. Meanwhile, nine defense genes, including wound-responsive AP2-like factor, glutathione-s-transferase, serine/threonine-protein kinase BRI1, and Avr9/Cf-9 rapidly elicited protein genes, were obtained using weighted gene coexpression network analysis, which provided reliable targets for functional verification. This study provides a theoretical reference for the comprehensive application of plant hormones to improve resistance to potato late blight disease.

Phytoremediation plays an important role in the treatment of heavy metal pollution in soil. In order to elucidate the mechanism of salicylic acid (SA) on copper absorption, seedlings from Xuzhou (with strong Cu-tolerance) and Weifang Helianthus tuberosus cultivars (with weak Cu-tolerance) were selected for pot culture experiments. 1 mmol/L SA was sprayed upon 300 mg/kg soil copper stress, and the photosynthesis, leaf antioxidant system, several essential mineral nutrients and the changes of root upon copper stress were analyzed to explore the mechanism of copper resistance. The results showed that Pn , Tr , Gs and Ci upon copper stress decreased significantly compared to the control group. Meanwhile, chlorophyll a, chlorophyll b and carotenoid decreased with significant increase in initial fluorescence ( F 0 ), maximum photochemical quantum yield of PSⅡ ( Fv/Fm ), electron transfer rate (ETR) and photochemical quenching coefficient ( qP ) content all decreased. The ascorbic acid (AsA) content was decreased, the glutathione (GSH) value was increased, the superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activity in the leaves were decreased, and the peroxidase (POD) activity was significantly increased. SA increased the Cu content in the ground and root system, and weakened the nutrient uptake capacity of K, Ca, Mg, and Zn in the root stem and leaves. Spray of exogenous SA can maintain the opening of leaf stomata, improve the adverse effect of copper on photosynthetic pigment and PSⅡ reaction center. Mediating the SOD and APX activity started the AsA-GSH cycle process, effectively regulated the antioxidant enzyme system in chrysanthemum taro, significantly reduced the copper content of all parts of the plant, and improved the ion exchange capacity in the body. External SA increased the content of the negative electric group on the root by changing the proportion of components in the root, promoted the absorption of mineral nutrient elements and the accumulation of osmoregulatory substances, strengthened the fixation effect of the root on metal copper, and avoided its massive accumulation in the H . tuberosus body, so as to alleviate the inhibitory effect of copper on plant growth. The study revealed the physiological regulation of SA upon copper stress, and provided a theoretical basis for planting H . tuberosus to repair soil copper pollution.

Pleurotus ostreatus is usually cultivated in horticultural facilities that lack environmental control systems and often suffer heat stress (HS). Salicylic acid (SA) is recognized as a plant defense-related hormone. Here, SA treatment (200 μM) induced fungal resistance to HS of P. ostreatus , with decreased malondialdehyde (MDA) content and HSP expression. Further analysis showed that SA treatment in P. ostreatus increased the cytosolic trehalose content and reduced the intracellular reactive oxygen species (ROS) level. Moreover, H 2 O 2 could restore the MDA content and HSP expression of P. ostreatus treated with SA under HS. In addition, trehalose (25 mM) or CaCl 2 (5 mM) treatment induced fungal resistance to HS, and CaCl 2 treatment increased the cytosolic trehalose content of P. ostreatus under HS. However, inhibiting Ca 2+ levels using Ca 2+ inhibitors or mutants reversed the trehalose content induced by SA in P. ostreatus under HS. In addition, inhibiting trehalose biosynthesis using Tps -silenced strains reversed the MDA content and HSP expression of P. ostreatus treated with SA under HS. Taken together, these results indicate that SA treatment alleviates the HS response of P. ostreatus by reducing the intracellular ROS level and increasing the cytosolic trehalose content. IMPORTANCE Heat stress (HS) is a crucial environmental challenge for edible fungi. Salicylic acid (SA), a plant defense-related hormone, plays key roles in plant responses to biotic and abiotic stresses. In this study, we found that SA treatment increased the cytosolic trehalose content and induced fungal resistance to HS in P. ostreatus . Further analysis showed that SA can alleviate the HS of P. ostreatus by reducing the intracellular ROS level and increasing the cytosolic trehalose content. Our results help to understand the mechanism underlying the responses of P. ostreatus to HS. In addition, this research provides new insights for the cultivation of P. ostreatus .

The aim of the manuscript was to demonstrate the efficacy of salicylic acid (SA) in abrogating the fluoride-induced oxidative damages in the susceptible rice cultivar, MTU1010. Prolonged exposure of seedlings to sodium fluoride (25 mg L -1 ) severely impaired growth and overall physiological parameters like germination percentage, biomass and root and shoot length and incited the formation of hydrogen peroxide that enhanced electrolyte leakage, formation of cytotoxic products like malondialdehyde and methylglyoxal and lipoxygenase activity. Exogenous application of SA (0.5 mM) enhanced the endogenous level of SA that restored the chlorophyll content and catalase activity and further escalated the activity of other enzymatic antioxidants (superoxide dismutase, guaiacol peroxidase, ascorbate peroxidase, glutathione peroxidase and glutathione S-transferase), formation of non-enzymatic antioxidants (anthocyanins, carotenoids, flavonoids, phenolics, ascorbate and reduced glutathione) and osmolytes (proline, amino acids and glycine betaine) that cumulatively maintained the integrity of membrane structure and homeostatic balance of the cells by scavenging the accumulated hydrogen peroxide. SA-mediated formation of proline and flavonoids was linked with the enhanced activity of Δ 1 -pyrroline-5-carboxylate synthetase and phenylalanine ammonia lyase. Fluoride stress enhanced the activity of enzymes like glyoxalase I and glyoxalase II which were further aggravated in the seedlings upon treatment with SA, effectively detoxifying the methylglyoxal formed during stress. Overall, the manuscript depicts the pivotal role played by exogenous SA in ameliorating the effects of fluoride-induced damages in the seedlings and proves its potentiality as a protective chemical against fluoride stress when applied exogenously in rice., (© 2021. Accumulated evidences have revealed the critical roles of calcium binding protein (CaBP) in growth and stress responses of plants. However, its function in woody plants is poorly understood. In this study, we cloned the CDS, gDNA and promoter sequences of three tandem-duplicated CaBPs (CsCaBP1, CsCaBP2 and CsCaBP3) from Citrus sinensis, analyzed their sequence characteristics, and investigated their gene expression patterns and promoter activities under treatments of CaCl 2 , several phytohormones and wounding. Results showed that the three CsCaBPs have high sequence similarity. Their expression was strongly induced by CaCl 2 , ethylene, jasmonic acid, salicylic acid and wounding, and the promoting effect of wounding on their expression was found to be partially ethylene-dependent. Consistently, we identified many phytohormone-related cis-acting elements in their promoters, and their promoter activity could be induced significantly by ethylene, jasmonic acid, salicylic acid and wounding. All the three CsCaBPs can interact with WRKY40, whose encoding gene showed a similar expression pattern to CsCaBPs under phytohormone and wounding treatments. In addition, CsERF14, CsERF21, CsERF3 and CsERF2 could bind to their promoters. Key Message: R2R3 MYB transcription factor GhMYB18 is involved in the defense response to cotton aphid by participating in the synthesis of salicylic acid and flavonoids. R2R3 MYB transcription factors (TFs) play crucial roles in plant growth and development as well as response to abiotic and biotic stresses. However, the mechanism of R2R3 MYB TFs in cotton response to aphid infestation remains largely unknown. Here, an R2R3 MYB transcription factor GhMYB18 was identified as a gene up-regulated from upland cotton (Gossypium hirsutum L.) under cotton aphid (Aphis gossypii Glover) infestation. GhMYB18, which has transcription activity, was localized mainly to nucleus and cell membranes. Transient overexpression of GhMYB18 in cotton activates salicylic acid (SA) and phenylpropane signaling pathways and promoted the synthesis of salicylic acid and flavonoids, which leads to enhancing the tolerance to cotton aphid feeding. In contrast, silencing of GhMYB18 increased the susceptibility of G. hirsutum to aphid. Additionally, GhMYB18 significantly promoted the activities of defense-related enzymes including catalase (CAT), peroxidase (POD), polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL). Background: There is a serious global problem of salinization of arable land, causing large reduction in world food production. Use of plant hormones is an effective way to reduce damage caused to crops and salt stress., Results: In this study, PEI-EDA was modified with AM-zein and grafted with plant hormone SA (AM-zein-SA) and used as a nano-pesticide carrier to load emamectin benzoate (EB). The use of AM-zein-SA as a nano-pesticide carrier could reduce the damage caused by salt stress to crops. The structure of AM-zein-SA was characterized by FTIR, UV, fluorescence, Raman, and 1 H NMR spectroscopic techniques. AM-zein-SA could effectively improve the resistance of EB to ultraviolet radiations, resistance of cucumber to salt stress, and the absorption of EB by plants. The experimental results showed that AM-zein-SA could effectively improve the anti-UV property of EB by 0.88 fold. When treated with 120 mmol NaCl, the germination rate of cucumber seeds under salt stress increased by 0.93 fold in presence of 6.25 mg/L carrier concentration. The POD and SOD activities increased by 0.50 and 1.21 fold, whereas the content of MDA decreased by 0.23 fold. In conclusion, AM-zein-SA nano-pesticide carrier could be used to improve the salt resistance of crops and the adhesion of pesticides to leaves., Conclusion: AM-zein-SA, without undergoing any changes in its insecticidal activity, could simultaneously improve the salt stress resistance and salt stress germination rate of cucumber, reduce growth inhibition due to stress under high-concentration salt, and had a good effect on crops. In addition, EB@AM-zein-SA obviously improved the upward transmission rate of EB, as compared with EB. Background: Fusarium wilt and Ascochyta blight are the most important diseases of chickpea. The current study was designed to investigate the individual and combined effect of salicylic acid (SA) with Pseudomonas stutzeri and Pseudomonas putida to suppress Fusarium wilt and promote growth of chickpea varieties: Thal-2006 and Punjab-2008., Methods: At the time of sowing, inoculum of Fusarium oxysporum was applied to the soil and the incidence of Fusarium wilt was recorded after 60 days. The seeds were inoculated with Pseudomonas stutzeri and Pseudomonas putida prior to sowing. Chickpea plants were treated with salicylic acid at seedling stage., Results: The combination of P. stutzeri and SA significantly increased root length (166% and 145%), shoot height (50% and 47%) and shoot biomass (300% and 233%) in cv. Thal-2006 and cv. Punjab-2008, respectively, in infected plants. Similarly, the combined treatment of P. putida + SA, also enhanced the plant growth parameters of chickpea varieties. Maximum reduction in disease severity was observed in both P. stutzeri + SA (90% and 84%) and P. putida + SA (79% and 77%) treatments in cv. Thal-2006 and Punjab-2008, respectively. Both P. putida + SA and P. stutzeri + SA treatments resulted in increased leaf relative water and total protein content, peroxidase, superoxide dismutase, phenylalanine ammonia-lyase and polyphenol oxidase activities in both resistant (cv. Thal-2006) and susceptible (cv. Punjab-2008) cultivars. Both treatments also significantly reduced malondialdehyde (MDA) and proline content in cv. Thal-2006 and Punjab-2008. Cultivar Thal-2006 was more effective than cv. Salinity is one of the most important abiotic stresses that reduce plant growth and performance by changing physiological and biochemical processes. In addition to improving the crop, using nanomaterials in agriculture can reduce the harmful effects of environmental stresses, particularly salinity. A factorial experiment was conducted in the form of a completely randomized design with two factors including salt stress at three levels (0, 50, and 100 mM NaCl) and chitosan-salicylic acid nanocomposite at three levels (0, 0.1, and 0.5 mM). The results showed reductions in chlorophylls (a, b, and total), carotenoids, and nutrient elements (excluding sodium) while proline, hydrogen peroxide, malondialdehyde, total soluble protein, soluble carbohydrate, total antioxidant, and antioxidant enzymes activity increased with treatment chitosan-salicylic acid nanocomposite (CS-SA NCs) under different level NaCl. Salinity stress reduced Fm', Fm, and Fv/Fm by damage to photosynthetic systems, but treatment with CS-SA NCs improved these indices during salinity stress. In stress-free conditions, applying the CS-SA NCs improved the grapes' physiological, biochemical, and nutrient elemental balance traits. CS-SA NCs at 0.5 mM had a better effect on the studied traits of grapes under salinity stress. Salicylic acid is an NSAID with serious side effects on the GIS. The side effects of salicylic acid on the GIS are slightly reduced by acetylating salicylic acid. 12 new ester analogs of salicylic acid were synthesized with high yields in this study. The chemical structures of the synthesized compounds were characterized by 1 H-NMR, 13 C-NMR, and HRMS spectra. The inhibitory potential of the compounds was evaluated on COXs by in vitro and in silico studies. The COX2 inhibitory activity of the most potent inhibitor MEST1 (IC 50 : 0.048 μM) was found to be much higher than the COX2 inhibitory activity of aspirin (IC 50 : 2.60 μM). In docking studies, the strongest inhibitor among the compounds synthesized was predicted to be MEST1, with the lowest binding energy. Background: Tobacco's PR-1a gene is induced by pathogen attack or exogenous application of salicylic acid (SA). Nucleosome mapping and chromatin immunoprecipitation assay were used to delineate the histone modifications on the PR-1a promoter. However, the epigenetic modifications of the inducible promoter of the PR-1a gene are not fully understood yet., Methods and Results: Southern approach was used to scan the promoter of PR-1a to identify presence of nucleosomes, ChIP assays were performed using anti-histones antibodies of repressive chromatin by di- methylated at H3K9 and H4K20 or active chromatin by acetylated H3K9/14 and H4K16 to find epigenetic malleability of nucleosome over core promoter in uninduced or induced state post SA treatment. Class I and II mammalian histone deacetylase (HDAC) inhibitor TSA treatment was used to enhance the expression of PR-1a by facilitating the histone acetylation post SA treatment. Here, we report correlated consequences of the epigenetic modifications correspond to disassembly of the nucleosome (spans from - 102 to + 55 bp, masks TATA and transcription initiation) and repressor complex from core promoter, eventually initiates the transcription of PR-1a gene post SA treatment. While active chromatin marks di and trimethylation of H3K4, acetylation of H3K9 and H4K16 are increased which are associated to the transcription initiation of PR-1a following SA treatment. However, in uninduced state constitutive expression of a negative regulator (SNI1) of AtPR1, suppresses AtPR1 expression by six-fold in Arabidopsis thaliana. Further, we report 50-to-1000-fold increased expression of AtPR1 in uninduced lsd1 mutant plants, up to threefold increased expression of AtPR1 in uninduced histone acetyl transferases (HATs) mutant plants, SNI1 dependent negative regulation of AtPR1, all together our results suggest that inactive state of PR-1a is indeed maintained by a repressive complex., Conclusion: The study aimed to reveal the mechanism of transcription initiation of tobacco PR-1a gene in presence or absence of SA. Quail bush [ Atriplex lentiformis (Torr.) S. Wats] plants were used in removing 2, 4-dinitrophenol (DNP) from wastewater in a hydroponic experiment. The hydroponic system contained three doses of DNP, i.e., 0, 10, and 20 mg L -1 . Quail bush plants were sprayed with 0.1 mM salicylic acid (SA) to study its role in resisting DNP toxicity. DNP significantly ( p  < 0.05) reduced plant growth. Exposure of A. lentiformis plants to 20 mg L -1 of DNP reduced the total chlorophyl and relative water content by 39 and 24%, respectively. SA improved the antioxidant defense in terms of ascorbate peroxidase (APX) and polyphenol oxidase (PPO) activities. SA alleviated DNP toxicity by enhancing the production of osmoprotectants, e.g., proline, phenols, and carbohydrates. SA enhanced the removal efficiency of DNP and the highest removal efficiency (96%) was recorded in the plants sprayed with SA and grown on 10 mg L -1 of DNP. A. lentiformis is a halophytic plant that has good physiological characteristics to resist 2, 4-dinitrophenol toxicity in wastewaters and is qualified to purify water from these harmful compounds. Exogenous application of 0.1 mM SA increased the defense system in A. lentiformis against 2, 4-dinitrophenol toxicity and enhanced the removal efficiency.

This experiment was conducted in the poultry field of the College of Agriculture, University of Diyala, and the Biochemistry Laboratory in the Department of Animal Production to show the effect of using tartaric and salicylic acids on the chemical composition of eggs produced by Lohman chickens raised at high temperatures, in which 210 laying hens of brown Lohman strain were used. Sixty weeks were divided into seven treatments: T1, the control treatment, T2 and T3, in which 0.2% of tartaric and salicylic acid were added to drinking water, respectively, and T4 added to it a mixture of organic acids at a rate of 0.4% to the drinking water, and treatments T5 and T6 were added. It contained 0.2% of tartaric and salicylic acid to the ration, respectively, and T7 was added to the ration of 0.4% of the two organic acids to the diet formulation. At the age of 75 weeks, eggs were taken randomly from the treatments and at a rate of 5 eggs/treatment to know the effect of the treatments on the chemical composition of the eggs produced, and the results of the statistical analysis showed It is proved in Appendix 1 that there was no significant effect ( P ≥0.05) of the different treatments on the studied traits, which included the percentage of moisture, protein, fat, carbohydrates and ash in each of the whites and yolks. Freezing severely impacts potato production. Deciphering the pathways and metabolites that regulate the freezing tolerance of potato is useful in cultivation and breeding for hardiness. In the present study, Solanum acaule was identified to be more freezing tolerant than S. tuberosum . Furthermore, the two genotypes before/after exposure to 4 °C for 7 d with additional -1 °C for 12 h were analysed by RNA-seq and metabolomics, and the results were compared with the previous -1 °C for 12 h. The results showed that S. acaule activated numerous genes that differed from those of S. tuberosum . Among the genes, five pathways, such as the hormone signalling pathway, which includes salicylic acid, were enriched. Further metabolomics analysis showed that the content of salicylic acid was improved in S. acaule in response to -1 °C for 12 h. Moreover, exogenous application of 0.1 mM salicylic acid to potato was shown to improve constitutive freezing tolerance and increase the expression of HSFC1 . Following transcriptome and metabolome analyses, it was documented that the content of SA that increased in freezing-tolerant S. acaule after exposure to cold condition, associated with the SA signalling pathway, enhanced potato freezing tolerance, probably through HSFC1 ., Competing Interests: The authors declare no conflict of interest.

Plants deposit photosynthetically-fixed carbon in the rhizosphere, the thin soil layer directly around the root, thereby creating a hospitable environment for microbes. To manage the inhabitants of this nutrient-rich environment, plant roots exude and dynamically adjust microbe-attracting and -repelling compounds to stimulate specific members of the microbiome. Previously, we demonstrated that foliar infection of Arabidopsis thaliana by the biotrophic downy mildew pathogen Hyaloperonospora arabidopsidis (Hpa) leads to a disease-induced modification of the rhizosphere microbiome. Soil conditioned with Hpa-infected plants provided enhanced protection against foliar downy mildew infection in a subsequent population of plants, a phenomenon dubbed the soil-borne legacy (SBL). Here, we show that for the creation of the SBL, plant-produced coumarins play a prominent role as coumarin-deficient myb72 and f6'h1 mutants were defective in creating a Hpa-induced SBL. Root exudation profiles changed significantly in Col-0 upon foliar Hpa infection, and this was accompanied by a compositional shift in the root microbiome that was significantly different from microbial shifts occurring on roots of Hpa-infected coumarin-deficient mutants. Our data further show that the Hpa-induced SBL primes Col-0 plants growing in SBL-conditioned soil for salicylic acid (SA)-dependent defenses. The SA-signaling mutants sid2 and npr1 were unresponsive to the Hpa-induced SBL, suggesting that the protective effect of the Hpa-induced shift in the root microbiome results from an induced systemic resistance that requires SA-signaling in the plant., (© 2022.
