Peripartum nutrition is crucial for developing the immune system of neonates. We hypothesized that maternal short-chain fructooligosaccharide (scFOS) supplementation could accelerate the development of intestinal immunity in offspring. Thirty-four sows received a standard or a scFOS supplemented diet (10 g scFOS/d) for the last 4 weeks of gestation and the 4 weeks of lactation. Colostrum and milk immunoglobulins (Ig) and TGFβ1 concentrations were evaluated on the day of delivery and at d 6 and d 21 postpartum. Piglet intestinal structure, the immunologic features of jejunal and ileal Peyer's patches, and mesenteric lymph node cells were analysed at postnatal d 21. Short-chain fatty acid concentrations were measured over time in the intestinal contents of suckling and weaned piglets. Colostral IgA (P<0.05) significantly increased because of scFOS and TGFβ1 concentrations tended to improve (P<0.1). IFNγ secretion by stimulated Peyer's patch and mesenteric lymph node cells, and secretory IgA production by unstimulated Peyer's patch cells were increased (P<0.05) in postnatal d 21 scFOS piglets. These differences were associated with a higher proportion of activated CD25+CD4α+ T cells among the CD4+ helper T lymphocytes (P<0.05) as assessed by flow cytometry. IFNγ secretion was positively correlated with the population of activated T lymphocytes (P<0.05). Total short-chain fatty acids were unchanged between groups during lactation but were higher in caecal contents of d 90 scFOS piglets (P<0.05); specifically propionate, butyrate and valerate. In conclusion, we demonstrated that maternal scFOS supplementation modified the intestinal immune functions in piglets in association with increased colostral immunity. Such results underline the key role of maternal nutrition in supporting the postnatal development of mucosal immunity.

Numerous xenobiotics have been shown to be harmful for the kidney. Thus, to improve our knowledge of the cellular processing of these nephrotoxic compounds, we evaluated, by real-time PCR, the mRNA expression level of 377 genes encoding xenobiotic-metabolizing enzymes (XMEs), transporters, as well as nuclear receptors and transcription factors that coordinate their expression in eight normal human renal cortical tissues. Additionally, since several renal in vitro models are commonly used in pharmacological and toxicological studies, we investigated their metabolic capacities and compared them with those of renal tissues. The same set of genes was thus investigated in HEK293 and HK2 immortalized cell lines in commercial primary cultures of epithelial renal cells and in proximal tubular cell primary cultures. Altogether, our data offers a comprehensive description of kidney ability to process xenobiotics. Moreover, by hierarchical clustering, we observed large variations in gene expression profiles between renal cell lines and renal tissues. Primary cultures of proximal tubular epithelial cells exhibited the highest similarities with renal tissue in terms of transcript profiling. Moreover, compared to other renal cell models, Tacrolimus dose dependent toxic effects were lower in proximal tubular cell primary cultures that display the highest metabolism and disposition capacity. Therefore, primary cultures appear to be the most relevant in vitro model for investigating the metabolism and bioactivation of nephrotoxic compounds and for toxicological and pharmacological studies., (Copyright © 2014 Elsevier Inc. Renal proximal tubular epithelial cells play a central role in renal physiology and are among the cell types most sensitive to ischemia and xenobiotic nephrotoxicity. In order to investigate the molecular and cellular mechanisms underlying the pathophysiology of kidney injuries, a stable and well-characterized primary culture model of proximal tubular cells is required. An existing model of proximal tubular cells is hampered by the cellular heterogeneity of kidney; a method based on cell sorting for specific markers must therefore be developed. In this study, we present a primary culture model based on the mechanical and enzymatic dissociation of healthy tissue obtained from nephrectomy specimens. Renal epithelial cells were sorted using co-labeling for CD10 and CD13, two renal proximal tubular epithelial markers, by flow cytometry. Their purity, phenotypic stability and functional properties were evaluated over several passages. Our results demonstrate that CD10/CD13 double-positive cells constitute a pure, functional and stable proximal tubular epithelial cell population that displays proximal tubule markers and epithelial characteristics over the long term, whereas cells positive for either CD10 or CD13 alone appear to be heterogeneous. In conclusion, this study describes a method for establishing a robust renal proximal tubular epithelial cell model suitable for further experimentation.

The filament stretching properties of various polysaccharides (including xanthan, carob, hydroxypropyl guar, hydroxypropylmethyl and hydroxyethyl celluloses) were investigated and compared to synthetic polymers generally used as texturing agents in cosmetic emulsions. The stretchability was examined by sensory evaluation as "the amount of sample that strings rather than breaks when fingers are separated". Different behaviors were evidenced: the xanthan emulsion showed the highest stretchability, followed by the hydroxypropyl guar and hydroxyethyl cellulose emulsions while the synthetic polymers presented stretching properties to a much lesser extent. The instrumental characterization of the stretchability was conducted at a controlled speed and recorded with a camera using a texture analyzer. The maximum stretchable length at 40mm/s was highly significantly correlated to the sensory Stringiness, thus allowing a good predictability of this attribute. Finally, this method was applied to aqueous solutions to better understand the role of the polymers in emulsion and to validate the measurement on a wider range of products., (Copyright © 2012 Elsevier Ltd. This study deals with the impact of emollients on the spreading properties of cosmetic products using a combined sensory-instrumental approach. To that purpose, three esters and one silicone were selected and incorporated separately into an oil phase. Different cosmetic o/w emulsions were then prepared with these different oil phases. Both of them were analyzed by instrumental techniques and in vivo sensory analyses. A significant effect of the emollient used was established in emulsions and in oil phases as well. Concerning emulsions, results reveal a clear correlation between in vivo spreading evaluation and friction coefficient parameters measured by texture analyzer, despite a fairly low correlation coefficient (Pearson coefficient=-0.78). Concerning oil phases, characterization of spreading was done by monitoring the contact angle relaxation of a drop of solution after deposition on a flat PMMA surface whereas sensory procedure was based on spontaneous spreading of oil phases onto the skin. Finally, good correlations between in vivo sensory analysis and instrumental measurements of both oils and emulsions were found, thus promising the possible development of predictive tools to evaluate spreadability., (Copyright © 2012 Elsevier B.V. MicroRNAs (miRNAs) are a class of noncoding RNA acting at a post-transcriptional level to control the expression of large sets of target mRNAs. While there is evidence that miRNAs deregulation plays a causative role in various complex disorders, their role in fibrotic kidney diseases is largely unexplored. Here, we found a strong up-regulation of miR-21 in the kidneys of mice with unilateral ureteral obstruction and also in the kidneys of patients with severe kidney fibrosis. In addition, mouse primary fibroblasts derived from fibrotic kidneys exhibited higher miR-21 expression level compared to those derived from normal kidneys. Expression of miR-21 in normal primary kidney fibroblasts was induced upon TGFβ exposure, a key growth factor involved in fibrogenesis. Finally, ectopic expression of miR-21 in primary kidney fibroblasts was sufficient to promote myofibroblast differentiation. As circulating miRNAs have been suggested as promising non-invasive biomarkers, we further assess whether circulating miR-21 levels are associated with renal fibrosis using sera from 42 renal transplant recipients, categorized according to their renal fibrosis severity, evaluated on allograft biopsies (Interstitial Fibrosis/Tubular Atrophy (IF/TA). Circulating miR-21 levels are significantly increased in patients with severe IF/TA grade (IF/TA grade 3: 3.0±1.0 vs lower grade of fibrosis: 1.5±1.2; p = 0.001). By contrast, circulating miR-21 levels were not correlated with other renal histological lesions. In a multivariate linear regression model including IF/TA grade and estimated GFR, independent associations were found between circulating miR-21 levels and IF/TA score (ß = 0.307, p = 0.03), and between miR-21 levels and aMDRD (ß = -0.398, p = 0.006). Altogether, these data suggest miR-21 has a key pathogenic role in kidney fibrosis and may represent a novel, predictive and reliable blood marker of kidney fibrosis.

Protein level in the maternal diet plays a crucial role in fetal programming during pregnancy. Low or high protein level increases the risk of intrauterine growth retardation (IUGR). The aim of this study was to investigate the structural and functional development of the small intestine in piglets from sows fed a control (C, 12.1% protein), a high protein (HP, 30% protein), or a low protein (LP, 6.5% protein) diet during pregnancy. Newborns were classified as IUGR (birth weight ≤1.18 kg) and non-IUGR (birth weight >1.18 kg). The piglets were euthanized on postnatal day (PD)1, PD28 and PD188. The LP diet in non-IUGR neonates resulted in decreased body weight on PD1. The LP and HP diets resulted in both decreased body weight and delayed catch-up growth in the IUGR piglets. The HP and LP-diets increased the length of villi on PD1 in non-IUGRs but not in IUGRs. At birth, the expressions of Ki67 and active caspase 3 in mid-jejunum epithelium of HP and LP non-IUGR neonates were significantly lower as compared to C non-IUGRs whilst in IUGRs the respective expressions were as high as in C non-IUGRs. The postnatal dynamics of brush border enzyme activities and vacuolated enterocytes disappearance showed significant drop in enterocyte maturation in IUGR as compared to non-IUGR neonates. In conclusion, both HP and LP diets led to retarded development of non-IUGR piglets. In IUGR piglets both HP and LP diets resulted in delayed catch-up growth, without adaptive changes in brush border digestive enzymes.

Background: Physiology of the exocrine pancreas has been well studied in domestic and in laboratory animals as well as in humans. However, it remains quite unknown in wildlife mammals. Roe deer and cattle (including calf) belong to different families but have a common ancestor. This work aimed to evaluate in the Roe deer, the adaptation to diet of the exocrine pancreatic functions and regulations related to animal evolution and domestication., Results: Forty bovine were distributed into 2 groups of animals either fed exclusively with a milk formula (monogastric) or fed a dry feed which allowed for rumen function to develop, they were slaughtered at 150 days of age. The 35 Roe deer were wild animals living in the temperate broadleaf and mixed forests, shot during the hunting season and classified in two groups adult and young. Immediately after death, the pancreas was removed for tissue sample collection and then analyzed. When expressed in relation to body weight, pancreas, pancreatic protein weights and enzyme activities measured were higher in Roe deer than in calf. The 1st original feature is that in Roe deer, the very high content in pancreatic enzymes seems to be related to specific digestive products observed (proline-rich proteins largely secreted in saliva) which bind tannins, reducing their deleterious effects on protein digestion. The high chymotrypsin and elastase II quantities could allow recycling of proline-rich proteins. In contrast, domestication and rearing cattle resulted in simplified diet with well digestible components. The 2nd feature is that in wild animal, both receptor subtypes of the CCK/gastrin family peptides were present in the pancreas as in calf, although CCK-2 receptor subtype was previously identified in higher mammals., Conclusions: Bovine species could have lost some digestive capabilities (no ingestion of great amounts of tannin-rich plants, capabilities to secrete high amounts of proline-rich proteins) compared with Roe deer species. CCK and gastrin could play an important role in the regulation of pancreatic secretion in Roe deer as in calf. This work, to the best of our knowledge is the first study which compared the Roe deer adaptation to diet with a domesticated animal largely studied.

Objectives: Exocrine pancreatic secretion contributes to limit pathogenic bacteria-associated diarrhea. Bovine colostrum, used in the treatment of diarrhea, reduces symptoms originating from gut pathogenic bacteria overgrowth. We hypothesized that bovine colostrum may stimulate the exocrine pancreatic secretion., Methods: Eighteen piglets fitted with 2 permanent catheters (for pancreatic juice collection and reintroduction) were allocated to 1 of the following 2 dietary treatments for 5 days: a control diet or a diet supplemented with defatted bovine colostrum. Pancreatic juice was collected daily, and digestive enzyme activities and antibacterial activity were determined., Results: The prandial pancreatic juice outflow, the basal and prandial lipase output, and the basal secretion of the antibacterial activity were, respectively, 60% (P = 0.08), 154% (P = 0.08), 92% (P = 0.06), and 72% (P < 0.05) higher in piglets fed a diet supplemented with defatted bovine colostrum., Conclusions: With defatted bovine colostrum, the increased antibacterial activity secretion against Escherichia coli may limit pathogenic bacteria overgrowth of the gut and reduce diarrheal episodes. The role of secretin in the increased pancreatic juice flow and lipase secretion was considered.

The release of a strawberry aroma from different composite gels taken as models of fruit preparations and from a sucrose solution was investigated. The composition of the model systems differed with regard to the gelling agent, either pectin or carrageenan, and to the rigidity of the gel. With the use of atmospheric pressure chemical ionization-mass spectrometry, the release profiles of the aroma compounds were determined under stirring. At the same time, purge and trap measurements were performed to determine the release profiles of the aroma compounds without stirring. The comparison of the patterns obtained using these two complementary methods made it possible to determine how the structure of the matrix, the mechanical treatment, and the properties of the aroma compound affect aroma release. A far greater proportion of the aroma compounds was retained in the fruit preparation systems than in the sucrose solution. The different release profiles could be interpreted in terms of the volatility of the aroma compounds and of their diffusion through the gels.

Diffusion-ordered spectroscopy (DOSY)-pulsed field gradient (PGF) nuclear magnetic resonance (NMR) spectroscopy was used to measure self-diffusion coefficients of aroma molecules in model fruit preparations. The impact of the sucrose content on aroma diffusion was specifically investigated, and the relationship with viscosity, water activity, and dry matter parameters was evidenced. DOSY-PGF NMR spectroscopy was found to be a relevant and accurate technique to follow self-diffusion of aroma compounds at low concentrations in a complex food matrix and to obtain information on diffusion of the sucrose and of the water molecules. We showed that aroma self-diffusion was strongly decreased in fruit preparation because of the high sucrose content, which induces the formation of a network through hydrogen bonds with water. Self-diffusion coefficients were determined for aroma molecules of different natures, and values are related to the physicochemical properties of the molecule.

We investigated the changes in the capacity for synthesis of the exocrine pancreas of piglets during the 2 wk after weaning at 7 d of age (trial 1) by measuring the expression of digestive enzymes at mRNA and activity levels in pancreas homogenates, and the effects of high and low feed intakes during the 1st wk postweaning (trial 2) on these measures. The trypsin mRNA level was transiently decreased 43% 3 d postweaning (P < 0.05). Thereafter, trypsin and lipase mRNAs linearly increased (P < 0.05). During the 1st wk postweaning, trypsin- and lipase-specific activities were reduced 44 and 79% (P < 0.05), respectively, whereas 14 d after weaning, trypsin was at the preweaning value and lipase was at a low level. Amylase-specific activity did not change with weaning. Plasma cholecystokinin (CCK) and gastrin concentrations decreased 1 d postweaning and increased afterward up to 3 and 5 d postweaning, respectively. By 3 d after weaning, the mRNA level of trypsin was twofold higher (P < 0.05) in piglets that consumed more feed than in those that consumed less, whereas 7 d after weaning, the groups did not differ. By 7 d after weaning, the specific activity of amylase was higher, and lipase-specific activity was lower, in piglets that consumed more feed than in those that consumed less. Plasma CCK and gastrin concentrations measured 7 d after weaning were correlated with feed intake (r = +0.56 and r = +0.68, P < 0.05, respectively). In conclusion, by 3 d postweaning, pancreatic exocrine function was adapting to the new diet. Afterward, the expression of specific genes coding digestive enzymes and the levels of pancreatic enzyme activities were restored or stimulated, except for lipase-specific activity. Therefore, the pancreas can adjust to weaning and dry food intake as early as wk 2 of life.

Two trials involving a total of 56 pigs were conducted to examine the effects of weaning at 7 d of age (trial 1) and of energy intake level and length of post-weaning underfeeding period (trial 2) on small intestinal (SI) development and morphometry. At 3 d after weaning, weight of the SI and mucosa (g/kg body weight) and villous height along SI were reduced by 20, 36 and 41%, respectively, compared to the day of weaning. Intestinal morphometrical changes are dependent on SI site and days post-weaning. Villous atrophy on d 3 and recovery on d 14 post-weaning were greater and occurred earlier in the proximal than in the medial and distal SI. Villous height was dependent on the level of energy intake which explains 56% of the variations in proximal SI villous height in weaned pigs and 73% when data of the sow-reared pigs were included in the analysis. Moreover, after 4 d of refeeding, underfed piglets showed similar villous characteristics to piglets fed a continuously high feeding level after weaning stressing that capacities of intestinal restoration were not affected by the length of the post-weaning underfeeding period. Overall, the present results suggest a spatial and temporal effect of weaning on villous atrophy and recovery, and that the level of energy intake is a major factor accounting for the post-weaning villous height.

Purpose: Theoretical differences between ablative and additive surgeries are obvious, but what about final outcome? We decide to compare surface excimer photorefractive ablation (PRK) with Intracorneal segments (Intacs or ICRS) for correction of low myopia., Material: and, Method: Two homogeneous groups of 25 low pure myopic patients were considered: group A including patients operated with Excimer and group B corresponding to Intacs implanted eyes. Retrospective analysis was performed and compared data achieved at two years follow-up, in term of refractive results and qualitative vision., Results: Considering non corrected visual acuity, percentages of eyes who achieved 20/40 or better are comparable in the two series (respectively 97% in group A and 99% in group B). In the adverse, proportion of patients achieving 20/20 or more is superior in Intacs group (77%) versus 62% in PRK group. In all cases, quality of vision is altered in early postoperative period, then returns to normal between 1 and 3 months. Functional complaints appear more frequent and persisting in intrastromal segments group. Based on videotopography and cycloplegic refraction, it seems that risk of induced astigmatism occurrence is significantly increased in case of Intacs. No severe complication is mentioned in both groups., Conclusion: Final outcome are in favor of the efficacy, safety and predictability of the two methods for correction of low myopia. Concept of Intacs presents the interest of potential adjustability and respect of central corneal area; however it appears to be limited by the occurrence of an induced astigmatism.
