ABSTRACT The abundances and diversity of dinoflagellate cysts (including non-indigenous or harmful species) in ballast sediments were examined for the east and west coasts and the Great Lakes, to assess the potential invasion risk for these organisms and to determine similarity across regions., In total, 147 ships were sampled, distributed among three categories: (1) transoceanic, with mandatory ballast water exchange (BWE); (2) coastal with BWE; and (3) coastal without BWE. Factors that could account for variability in results were examined including ship routes, BWE, ballast water age and sediment volume in ballast tanks., The pattern of potential invasion risk differs according to the region examined. The east coast had greater concentrations of viable cysts per tank, particularly for coastal ships (with a maximum close to 2 × 106 cysts per tank in coastal exchanged ships compared with 1 × 105 for the west coast). When considering viable cysts g-1 dry sediment, maximum concentrations were found in unexchanged coastal ships from the east coast (28 cysts g-1 compared with 5 for the west coast), but these ships contained less sediment, decreasing the overall abundance of cysts per tank. These results are in the lower range of values reported for Scotland or Australia, which reached a maximum of 2.3 × 104 cysts cm−3 of sediment., Ballast water exchange had a different influence according to the region: on the east coast, the invasion risk was reduced by BWE while it had no significant effect in the other two regions. Diatoms are important contributors to aquatic primary production, and can dominate phytoplankton communities under variable light regimes. We grew two marine diatoms, the small Thalassiosira pseudonana and the large Coscinodiscus radiatus, across a range of temperatures and treated them with a light challenge to understand their exploitation of variable light environments. In the smaller T. pseudonana, photosystem II (PSII) photoinactivation outran the clearance of PSII protein subunits, particularly in cells grown at sub- or supraoptimal temperatures. In turn the absorption cross section serving PSII photochemistry was down-regulated in T. pseudonana through induction of a sustained phase of nonphotochemical quenching that relaxed only slowly over 30 min of subsequent low-light incubation. In contrast, in the larger diatom C. radiatus, PSII subunit turnover was sufficient to counteract a lower intrinsic susceptibility to photoinactivation, and C. radiatus thus did not need to induce sustained nonphotochemical quenching under the high-light treatment. T. pseudonana thus incurs an opportunity cost of sustained photosynthetic down-regulation after the end of an upward light shift, whereas the larger C. radiatus can maintain a balanced PSII repair cycle under comparable conditions. Long-term growth response to natural solar radiation with enhanced ultraviolet-B (UVB) exposure was examined in two species of dinoflagellates [ Alexandrium tamarense (M. Lebour) Balech, At, and Heterocapsa triquetra (Ehrenb.) F. Stein, Ht], including two strains of A. tamarense, one from Spain and another from UK, and one diatom species ( Thalassiosira pseudonana Hasle et Heimdal). We examined whether variable photoprotection (mycosporine-like amino acids [MAAs] and xanthophyll-cycle pigments) affected photosynthetic performance, phytoplankton light absorption, and growth. Growth rate was significantly reduced under enhanced UVB for the UK strain of At and for Ht (both grew very little) as well as for the diatom (that maintained high growth rates), but there was no effect for the Spanish strain of At. MAA concentration was high in the dinoflagellates, but undetectable in the diatom, which instead used the xanthophyll cycle for photoprotection. The highest cell concentrations of MAAs and photoprotective pigments were observed in the UK strain of At, along with lowest growth rates and Fv/ Fm, indicating high stress levels. In contrast, the Spanish strain showed progressive acclimation to the experimental conditions, with no significant difference in growth between treatments. Increase in total MAAs followed linearly the cumulative UVB of the preceding day, and both total and primary MAAs were maintained at higher constitutive levels in this strain. Acclimation to enhanced UVB in the diatom resulted in an increase in PSII activity and reduction in nonphotochemical quenching, indicating an increased resistance to photoinhibition after a few weeks. All four species showed increased phytoplankton light absorption under enhanced UVB. Large intrastrain differences suggest a need to consider more closely intraspecific variability in UV studies. The dinoflagellateAlexandrium tamarense(Lebour) Balech 1985 is responsible for recurrent outbreaks of paralytic shellfish poisoning in the St. Lawrence Estuary. In July 1998, anA. tamarensered tide developed in the estuary with maximum cell concentrations reaching 2.3× 106 cells·L−1 in brackish surface waters. To estimate the growth rate of these cells, surface water samples from different locations and days during the bloom were incubated for 5 to 9 days underin situtemperature and light conditions. Growth rates varied both spatially and temporally between 0 and 0.55 day−1, reaching the maximum growth rate reported for this species in culture. High growth rates were measured even during the peak of the red tide, suggesting that the extremely high cell concentrations observed did not solely result from aggregation or physical concentration but also involved active cellular growth.Alexandrium tamarensecells were found over a large range of salinity (20.8–29.5 psu), but high densities and significant growth were only measured when salinity was lower than 24.5 psu. Under these conditions, the number of divisions achieved byA. tamarensewas proportional to the amount of nitrate available at the beginning of the incubations, whereas variations in growth rate were apparently controlled by the availability of phosphate. We hypothesize that the ability ofA. tamarenseto perform vertical migrations and acquire nitrate at night pushes this species toward phosphate limitation in the St. Lawrence Estuary. Ultraviolet radiation effects were examined in natural phytoplankton communities from Rimouski (Canada), Ubatuba (Brazil), and Ushuaia (Argentina). Outdoor pump-mixed mesocosms were submitted to ambient solar radiation (NUVB) and ambient with additional UV-B radiation (UVBR) from lamps (HUVB), corresponding to a local 60% ozone depletion scenario. At all sites, neither algal biomass nor dark-adapted Fv/Fm were significantly affected by additional UVBR, suggesting the presence of active UV protection or repair mechanisms. To examine the role of D1 protein turnover, essential for PSII repair, short-term surface incubations were performed in the presence or absence of lincomycin, a chloroplast protein synthesis inhibitor. Effects on PSII were determined using chl a in vivo fluorescence, whereas the D1 protein was detected immunochemically. In the absence of D1 repair, D1 pools and Fv/Fm decreased to a similar extent under both light treatments. In the presence of D1 repair, D1 pools suffered faster net degradation under HUVB compared with NUVB, whereas Fv/Fm was maintained for both light treatments, suggesting that HUVB exposure in field populations had more effect on D1 synthesis and PSII repair than on D1 degradation. The fewer undamaged reaction centers remaining in phytoplankton under HUVB were able to maintain Fv/Fm or actually recovered during the dark acclimation before Fv/Fm measurements. The D1 pools suffered faster net degradation at the tropical site where high irradiance drove faster D1 degradation and high water temperature enabled fast enzymatic activities. The article discusses the highlights of the second Open Science Meeting of the Global Ecology and Oceanography of Harmful Algal Blooms (GEOHAB) Core Research Project on "HABs in Fjords and Coastal Embayments," which was held at the University of Victoria on May 28-29, 2012. The meeting highlighted the process in interpreting life history and growth dynamics of HABs. An overview of some keynote presentations is provided.

Harmful algal blooms (HABs) are natural phenomena that result from the interplay of biological, chemical, physical, and sedimentary processes occurring at different temporal and spatial scales. This paper provides an integrated description of HAB dynamics occurring at the mesoscale (10-100 km, sensu Haury et al., 1978) in confined and semi-confined coastal environments and under stratified water column conditions in a diversity of habitats where HAB events occur. It also focuses on relevant aspects occurring at the fine scale and even smaller cellular scales that are critical to species interactions with their environments. Examples include the key role of life-history stages in the recurrence of HABs in certain embayments; the physical-biological interactions driving the formation, maintenance, and decline of thin layers of plankton, including harmful microalgae; the fascinating, but poorly understood, domain of small-scale chemical interactions between HAB species and components of the food web; and the potential link between human activities and climate change and the trends in HAB occurrence. Development of new observing and sampling technologies and of new modeling approaches has resulted in greater understanding of these phenomena. We summarize the scientific achievements of two Core Research Projects initiated under the GEOHAB Implementation Strategy, "HABs in Fjords and Coastal Embayments" and "HABs in Stratified Systems," and outline the priorities for future research toward improving the management and mitigation of HAB impacts. Recent empirical and statistical evidence suggest that propagule pressure (i.e., number of individuals introduced per event, and the number and frequency of events) and colonization pressure (i.e., number of species released per event, and the number and frequency of events) are of vital importance to invasion success. To explore possible changes in propagule and colonization pressure during the transport stage of the invasion process, we examine abundance and species richness of virus-like particles, bacteria, diatoms, dinoflagellates, and invertebrates transported in commercial ships--a leading vector for global spread of aquatic nonindigenous species. We collected 154 ballast water samples from ships that had performed or were exempt from ballast water exchange (BWE) prior to arrival at Pacific and Atlantic ports in Canada and Laurentian Great Lakes ports. We found that abundance and species richness varied across taxa and regions, with ships arriving to the Atlantic region carrying the highest abundance of taxa. The highest species richness of invertebrates and diatoms was recorded from ships arriving to the Pacific, whereas the richest communities of dinoflagellates occurred in the Atlantic region. We also found that BWE had no effect on abundance or species richness of most taxa (dinoflagellates, diatoms, bacteria, and virus-like particles), whereas the effect on abundance of invertebrates was not clear. Finally, longer voyages resulted in lower abundance of all taxa except dinoflagellates, and lower species richness of diatoms. Paradoxically, the elevated abundance and species richness of dinoflagellates following BWE suggest that this group could have enhanced invasion potential when ships manage ballast water by exchange [ABSTRACT FROM AUTHOR]

We investigated the photophysiological responses of three ecotypes of the picophytoplankter Ostreococcus and a larger prasinophyte Pyrarninionas obovata to a sudden increase in light irradiance. The deepwater Ostreococcus sp. RCC809 showed very high susceptibility to primary photoinactivation, likely a consequence of high oxidative stress, which may relate to the recently noted plastid terminal oxidase activity in this strain. The three Ostreococcus ecotypes were all capable of deploying modulation of the photosystem II repair cycle in order to cope with the light increase, but the effective clearance of photoinactivated D1 protein appeared to be slower in the deepwater Ostreococcus sp. RCC809, suggesting that this step is rate limiting in the photosystem II repair cycle in this strain. Moreover, the deepwater Ostreococcus accumulated lutein and showed substantial use of the xanthophyll cycle under light stress, demonstrating its high sensitivity to light fluctuations. The sustained component of the nonphotochemical quenching of fluorescence correlated well with the xanthophyll deepoxidation activity. Comparisons with the larger prasinophyte P. obovata suggest that the photophysiology of Ostreococcus ecotypes requires high photosystem II repair rates to counter a high susceptibility to photoinactivation, consistent with low pigment package effects in their minute-sized cells. HPLC is still the technique of choice for the analysis and characterization of phytoplankton pigments. In this chapter we describe procedures for sample preparation and pigment extraction, and the use of octyl silica columns and pyridine-containing mobile phases to separate chlorophylls and carotenoids. The identification of pigments on the basis of their retention times and visible spectra, the preparation of pigment standards, and the quantitative analysis by either external or internal standard procedures are also described.

We report a large cyst bed of the potentially toxic and bloom-forming dinoflagellate Alexandrium tamarense species complex in bottom sediments from the port of Halifax, Nova Scotia, Canada. The average cyst concentrations of that species ranged from 4033±2647 to 220872±148086 cysts g⁻¹ of dry sediments and the highest concentrations were found near ship terminals in Bedford Basin. Although this species is endemic to this region, our work strongly suggests that some of the cysts of A. tamarense species complex found in the port of Halifax were introduced through discharged ballast water and sediments., (Copyright © 2012 Elsevier Ltd. Increasing empirical evidence indicates the number of released individuals (i.e. propagule pressure) and number of released species (i.e. colonization pressure) are key determinants of the number of species that successfully invade new habitats. In view of these relationships, and the possibility that ships transport whole communities of organisms, we collected 333 ballast water and sediment samples to investigate the relationship between propagule and colonization pressure for a variety of diverse taxonomic groups (diatoms, dinoflagellates and invertebrates). We also reviewed the scientific literature to compare the number of species transported by ships to those reported in nature. Here, we show that even though ships transport nearly entire local communities, a strong relationship between propagule and colonization pressure exists only for dinoflagellates. Our study provides evidence that colonization pressure of invertebrates and diatoms may fluctuate widely irrespective of propagule pressure. We suggest that the lack of correspondence is explained by reduced uptake of invertebrates into the transport vector and the sensitivity of invertebrates and diatoms to selective pressures during transportation. Selection during transportation is initially evident through decreases in propagule pressure, followed by decreased colonization pressure in the most sensitive taxa.
