116. Rapid concerted evolution of nuclear ribosomal DNA in two Tragopogon allopolyploids of recent and recurrent origin

Abstract

We investigated concerted evolution of rRNA genes in multiple populations of Tragopogon mirus and T. miscellus, two allotetraploids that formed recurrently within the last 80 years following the introduction of three diploids (T. dubius, T. pratensis, and T. porrifolius) from Europe to North America. Using the earliest herbarium specimens of the allotetraploids (1949 and 1953) to represent the genomic condition near the time of polyploidization, we found that the parental rDNA repeats were inherited in roughly equal numbers. In contrast, in most present-day populations of both tetraploids, the rDNA of T. dubius origin is reduced and may occupy as little as 5% of total rDNA in some individuals. However, in two populations of T. mirus the repeats of T. dubius origin outnumber the repeats of the second diploid parent (T. porrifolius), indicating bidirectional concerted evolution within a single species. In plants of T. miscellus having a low rDNA contribution from T. dubius, the rDNA of T. dubius was nonetheless expressed. We have apparently caught homogenization of rDNA repeats (concerted evolution) in the act, although it has not proceeded to completion in any allopolyploid population yet examined.

Abstract

We demonstrate high repetition-rate deuteron acceleration by irradiating a continuously flowing, ambient temperature liquid heavy water jet with the high-intensity ALEPH laser. The laser delivered up to 5.5 J (120 TW, 1.2 × 1021 W/cm2) laser energy on target at 0.5 Hz. A high repetition-rate Thomson parabola spectrometer measured the deuteron beam energy spectra on each shot for 60 sequential shots (two minutes). Peak fluxes of 5 × 10 10 deuterons/sr/pulse, corresponding to an average flux of 1.5 × 10 12 deuterons/sr/min, were demonstrated with deuteron energies reaching up to 4.4 MeV. High shot-to-shot stability is observed up to 40%–50% of the maximum deuteron energy. These deuteron beams are suited for fast neutron production through deuteron breakup in a converter yielding energies similar to deuteron–deuteron (D–D, 2.45 MeV) fusion reactions of importance for material damage studies. Abstract

A new liquid chromatographic separation method was developed for the speciation of the four main arsenic compounds present in water. Arsenite (As(III)), dimethylarsinic acid (DMA), monomethylarsonic acid (MMA) and arsenate (As(V)) were separated on a recently introduced stationary phase: porous graphitic carbon (PGC). The separation was first obtained under formic acid gradient conditions, but an adsorption phenomenon of As(V) on PGC was observed. To overcome this problem, As(V) was backflushed, and an efficient separation of the four solutes was achieved within 10 min. Extremely low detection limits (ranging from 10 to 70 ng * [L.sup.-1]) were obtained by coupling LC with an ICPMS. The method was successfully applied to different spiked mineral waters and a naturally arsenic-containing freshwater.
